Description
本发明涉及毛发营养剂及其生产方法。更具体地讲,本发明涉及含有将例如脂肪酶和蛋白酶的混合物或大米酵母加到含植物蛋白,如米糠和植物脂的基质中所得发酵产物的毛发营养剂及其生产方法。
毛发脱落和生长的确切机理有待探索。但脱毛发的主要原因一般认为是血液循环中断,低代谢,过量雄性激素,皮炎和过敏。
毛发包括伸出皮的部分和皮下部分。毛发根的毛发茎包括称为***细胞的组织。有各种毛细管包围发茎,而***细胞从毛细血管中流动的血液吸收营养,重复细胞分化而使毛发生长。
但若细胞中血液供应不足,则细胞分化作用降低,阻碍毛发生长。因此常认为促进皮层中血液循环并激活细胞的作用是必需的。
包围毛发的发囊具有称为5α-还原酶的酶,可与雄性激素结合而转化成5α-二氢睾酮,然后经血管送到***细胞,从而降低其腺苷酸环化酶的活性并导致细胞分化作用降低。结果是发囊逐渐萎缩,使毛发成为绒毛性的,导致秃头等。
如上举例,虽然提出了一系列毛发脱落和生长机理,但还没有一种机理被公认。目前市场上已有各种毛发生长剂,销售者称这些生长剂按毛发生长机理而发挥作用,如促进皮层中血液循环和/或活化***细胞。但市场上还未出现真正对毛发有效的生发剂。
在这种情况下,本发明人进行了长期艰苦的研究,最终提出了本发明对毛发作用优异的毛发营养剂,其中包括将例如脂肪酶和蛋白酶的混合物或大米酵母加到含植物蛋白,如米糠和植物脂的基质上所得的发酵产物。
因此本发明提出毛发营养剂,其中包括将例如脂肪酶和蛋白酶的混合物或大米酶母加到含植物蛋白,如米糠和植物脂的基质上所得的发酶产物。
该基质可由含植物蛋白和植物脂的任何物质构成。发酵酶必须具有分解基质中所含植物蛋白和植物脂的能力,可为脂肪酶和蛋白酶,还可用其它酶,如淀粉酶。
发酵产品经过过滤工艺即得本发明毛发营养剂。该营养剂对人体无害并且对防止毛发脱落和促进毛发生长具有优异的作用效果。
以下详述本发明。
生产本发明毛发营养剂的基质可为含植物蛋白和植物脂的任何物质,如粒状产品,如米糠,日本清酒泥,麦胚芽,麸皮,粗荞麦或粗稗子粉,豆制品,如大豆,***,豆乳,小豆,豆粉或咖啡渣或其它,如薯蓣,芋,茶渣,蜂蜜,芝麻,花生,神食果,明日叶,牛排植物,若布海草或昆布。米糠,日本清酒泥,麸皮,咖啡渣,麦胚芽,芝麻,***,大豆和小豆特别优选,因为其中含有良好平衡量的植物蛋白和植物脂。
用于本发明发酵的酶需能分解基质中的植物蛋白和植物脂。其中可含脂肪酶和蛋白酶且其它酶,如淀粉酶也可用。除脂肪酶和蛋白酶而外,大米酵母也可用,因为具有可在活体内产生各种酶的功能。
本发明所用脂肪酶可为酸性范围内使用的***脂肪酶,含于大量霉菌,酵母,微生物,体液,也可为取自内部器官的酶。更具体地讲,可用胰腺脂肪酶,肝脂肪酶,Tubercle菌脂肪酶,FIB脂肪酶和hima脂肪酶。
用于本发明的蛋白酶优选在最适宜PH下呈酸性。因此,可用胃蛋白酶,胰蛋白酶或胰凝乳蛋白酶,除酶本身的氨基酸残基而外,不具有任何特殊活性碱且不要求任何特殊酶活性制剂。也可用大多数植物酶和动物细胞中找到的酶或组织蛋白酶,如木瓜蛋白酶或无花果蛋白酶,可用还原剂,如氢氰酸酯(盐),抗坏血酸,半胱氨酸或谷胱甘肽活化。更具体地讲,可用胰凝乳蛋白酶,胰蛋白酶,胃蛋白酶,羧基肽酶,组织蛋白酶A,组织蛋白酶B,组织蛋白酶C′,组织蛋白酶Ⅲ,组织蛋白酶Ⅳ,肾酰基转移酶Ⅰ,肾酰基转移酶Ⅱ,亮氨酸氨基肽酶,氨基三肽酶,甘(氨酰)甘(氨酸)二肽酶(glycilglycine dipectitase),氨酰基脯氨酸〔二肽〕酶(prolitase),脯〔氨酸〕肽酶,血纤维蛋白溶酶,thrompin,木瓜蛋白酶,无花果蛋白酶,链球菌属蛋白酶,Cl,historyticum蛋白酶或肽酶。
将酶加入所说基质上即可得发酵产品,HPLC分析发酵产品得以下结果,分析用探测器(LC-9A,Shimadzu Seisakusho,Co.,Ltd.)和柱(3m长),洗脱层用乙腈,柱温保持约30℃,流速约10ml/分钟。
已发现发酵产物含脂肪酸,如肉豆蔻酸,棕榈酸,硬脂酸,花生酸,山萮酸,二十四(碳)烷酸,十二(碳)烯酸,十四(碳)烯酸,十四(碳)二烯酸,十五(碳)烯酸,十六(碳)烯酸,油酸,亚油酸,亚麻酸,二十(碳)烯酸,二十(碳)二烯酸,花生四烯酸,二十(碳)三烯酸,二十二(碳)烯酸(dococenic acid)和二十二(碳)六烯酸(docosahexanic acid)。已发现发酵产品还含氨基酸,如亮氨酸,异亮氨酸,赖氨酸,蛋氨酸,半胱氨酸,苯丙氨酸,酪氨酸,苏氨酸,色氨酸,缬氨酸,组氨酸,精氨酸,天冬氨酸,丙氨酸,谷氨酸,甘氨酸,脯氨酸和celin。
从分析结果可以看出,本发明所得发酵产品含大量氨基酸和脂肪酸。据认为,氨基酸可向***细胞提供营养,而脂肪酸则可活化和扩张毛细血管,使毛发润湿而显现光泽并使毛发囊中所含5α-还原酶纯化而防止毛发软化。据认为,各成分单独或组合还可促进毛发生长。应注意到,发酵产品还包括含硫氨基酸和不饱和脂肪酸,这两者据认为均可对毛发生长起优异作用。
已发现按本发明将大米酵母加入基质中发酵所得产品中,除了用脂肪酶和蛋白酶的混合物所得发酵产品中确定的成分而外,还含少量维生素A,维生素B1,维生素B2,维生素B6,维生素B12,维生素C,维生素E,铜,铁,钙(Calucium),磷,嘌呤,脱氧核糖核酸,核糖核酸,腺嘌呤,鸟嘌呤,尿嘧啶,胞嘧啶和黄嘌呤,因为在大米酵母中,除了脂肪酶和蛋白酶而外,还可生成大量酶,如淀粉酶,谷氨酰胺酶,纤维素酶和果胶酶,不仅分解植物蛋白和植物脂,而且分解基质中的其它成分。据认为,这些其它成分也可与所说氨基酸和脂肪酸协同作用而促进毛发生长。
为保存本发明毛发营养剂,可将其冷藏或煮沸,这不会使毛发营养剂发生功能性变化或作用效果降低。
以下说明本发明毛发营养剂生产方法,步骤包括将酶加入含植物蛋白和植物脂的基质中并过滤所得发酵产品。
发酵在含水条件下进行,可产生其它酶,如淀粉酶的脂肪酶和蛋白酶或大米酵母用作酶。脂肪酶和蛋白酶单独加入或混合加入。该混合物可含其它酶,如淀粉酶,为的是促进分解功能并使基质中其它成分分解。针对指数100的基质,优选将0.1W%的每一种酶加入该混合物中。用大米酵母时,因该酵母分解功能比脂肪酶或蛋白酶弱,所以优选是针对指数100的基质加1W%大米酵母。
发酵温度优选室温或35-45℃,且作为缓冲剂可用PH6-11的盐酸-可尼丁,一代磷酸钾-二代磷酸钾,盐酸-佛罗那钠,盐酸-三氨基甲烷,盐酸-硼砂,硼酸-碳酸钠,盐酸-氨基甲基丙二醇,氯化铵-氨,甘氨酸-氢氧化钠,硼酸-氢氧化钠,盐酸-二甲基甘氨酸钠,碳酸氢钠-碳酸钠,硼砂-氢氧化钠,硼砂-碳酸钠,盐酸一碳酸钠和二代磷酸钾(钠)-氢氧化钠等。任何这些物质任意组合后的混合物亦可用。
发酵时间搅拌条件下优选5-10小时。对于脂肪酶,给出预定色素物质作指示剂并测定酯水解所得放出脂肪酸量或用碱电滴定即可知反应结束。上述操作不易进行时,可用比色法测定水解放出的醇。对于蛋白酶,可用(水合)茚三酮比色法,其中用(水合)茚三酮加热时生成的带蓝色物质进行氨基酸比色试验。在该操作不易进行时,可用S
rensen甲醛滴定法。该法中应用了甲醛存在下α-氨基或亚氨基滴定曲线的发散现象。消毒处理可用蒸压釜消毒法。
本发明毛发营养剂是发酵产品过滤后加水或醇得到的。所得毛发营养剂可如此应用或可与洗发精,发油,洗液,毛发滋补剂,发膏,护发素或染发水混用。
可在本发明毛发营养剂中混入常见毛发生长剂中常用的其它成分,如血液循环促进剂,代谢促进剂,抗雄性激素制剂,消炎剂,抗过敏制剂,润湿剂,抗菌素和/或致冷剂,可为例如生育酚乙酸盐,氯化镱,碳酸气,4-异丙基环庚二烯酚酮,Pantonyl乙基醚苯甲酸酯,甘油酯十五(碳)烷酸酯,维生素H,乙炔***,glythyl蓖麻酸,二苯羟基胺氯化物,透明质酸,Minisasanishiki提取物,单硝基愈创木酚,田芥子染剂,松炬染剂,氢化可的松,联苯羟基胺氯化物,D-泛酰醇,水杨酸,乳酸,乳酸钠和薄荷醇等。
例1
用米糠作基质,将温水1000ml加入米糠500g中并混合好,然后将混合物送入适当容器,如玻璃容器或塑料容器中,向混合物加大米酵母5g。再加碳酸氢钠3g作缓冲剂以控制PH。用加热装置将容器加热到45℃并在该温度下保持。混合物放置一整天,有时搅拌,呈糊状后将该容器中的糊状液用布过滤而得约1000ml半透明头发营养剂。
例2
用豆渣作基质,向该豆渣200g中加水600ml,制成动物内脏器官100g并加水200ml,然后用玻璃均化器研磨制成均化物,再以1500rpm进行离心分离。取出上层清液100ml加入基质。这时还加入碳酸钠-硼酸以将PH调为10.0。混合物放置于约40℃下。用Kletto光电比色计(No.540过滤器)观测酶活性,约8小时后观测到活性终止。
将0.1M(PH10.0)的碳酸钠-硼酸40ml加入所得发酵液中作缓冲液。然后加胰凝乳蛋白酶至0.1M。整个混合物放置在约40℃下。用Conway法测定0.2ml均取自该完全混合物的样品液中氨量确定发酵终止。约10小时后,停止生成氨并完成发酵。发酵液过滤和水洗而得毛发营养剂。
例3
用小豆渣作基质,将水500ml加入该小豆渣200g中并将混合物制成悬浮液。另外,用FIB菌丝体所得脂肪酶浮在蒸馏水100ml中,用Knapp-March Blender搅拌20分钟后室温放置约20分钟。混合物2000rpm下离心分离30分钟后所得顶层液加入小豆渣和水之混合物中。加Britton-Robinson缓冲液(磷酸,乙酸,硼酸,苛性苏打)将PH调为9.0。约35℃下保持振荡约8小时。用0.05M醇溶盐酸滴定而观测发酵。
另外,将市场购得木瓜蛋白酶溶于水中并向混合物中送入硫化氢,历时几小时。之后向混合物中加醇以使木瓜蛋白酶沉降。再向精制木瓜蛋白酶中加水(木瓜蛋白酶1g∶水100ml)并同样加Britton-Robinson缓冲液将PH调为9.0,之后将其加入包括基质的发酵液中。整个发酵液放于约40℃下,用醇滴定0.2ml均取自该全发酵液的样品液可确定发酵终止。约12小时后发酵完成。发酵液过滤后得毛发营养剂。
例4
***用作基质,将水500ml加入该***100g中并将混合物保持在约50℃。另外,将市场上购得的肾脂肪酶溶于蒸馏水中(1%)。将该含肾脂肪酶的蒸馏水(5ml)加入该混合物中。加入磷酸(0.6M,PH7.0),5ml作缓冲液并全混合物约40℃下保持约10小时,间或搅拌。用Warburg流体压力计观测到碳酸气停止生成则确定发酵终止。
此外,将市场上购得的胃蛋白酶(Cudahy,1/10000USP可溶胃蛋白酶)10g溶于20%乙醇液10ml中,再将其加入所说发酵液中。然后加盐酸铵将PH调为7.5。该液体于约35℃放置约20小时。之后用过滤槽(5g)过滤,这时PH为7.0。滤液放置于37℃下。在Conway装置外将0.5ml均取自滤液的样品液取出并将每一样品与已放入该装置的碳酸钾1ml混合即可确定发酵终止。滴定观测挥发性氨量变化而得知发酵终止。约10小时后完成。所得液体以常见方式过滤而得毛发营养剂。
本发明例1-4毛发营养液用乙醇稀释后混合物每天早晚各一次涂到男人和鼠上。涂用30天后,两种情况均有出色的毛发生长效果。特别是在鼠上涂用后,长毛效果特别显著。
权利要求1所述的毛发营养剂包括含有各种氨基酸和脂肪酸的产品。酸单独或组合使毛细血管活化并使发囊中所含5α-还原酶钝化。与常见毛发生长剂比较,权利要求1所述毛发营养剂具有更为优异的毛发脱落防治效果以及毛发生长效果。本发明毛发营养剂对人体完全无害。因为是发酵产品且基质包括植物蛋白和植物脂,用的是无害酶。
对于权利要求2的毛发营养剂,用作酶的脂肪酶和蛋白酶可有效地用来使基质中的植物蛋白和植物脂分解而获得高产量的发酵产品/单位重量。
权利要求3的毛发营养剂中除氨基酸和脂肪酸而外,还含有少量维生素E,铜,嘌呤,核酸等,这些其它成分与氨基酸和脂肪酸一起进一步促进毛发生长。
权利要求4的方法提供本发明毛发营养剂,该营养剂可活化毛细血管并使毛发囊中所含的5α-还原酶钝化。因此该营养剂可防止毛发脱落并且促进毛发生长比常见毛发生长剂更快。
权利要求5的方法获得高产量的发酵产品/单位重量,因为脂肪酶和蛋白酶作为酶,使基质中的植物蛋白和植物脂有效地发酵。
权利要求6的方法提供了除氨基酸和脂肪酸而外还含有少量维生素E,铜,嘌呤,核酸等其它成分,用以与氨基酸和脂肪酸一起进一步促进毛发生长。
权利要求7-9的方法使酵达到更快的分解速度,因此达到更高产量的毛发营养剂/单位重量。
Claims (9)
1、毛发营养剂,其中包括将酶加入含有植物蛋白和植物脂的基质中所得的发酵产品。
2、权利要求1的毛发营养剂,其中所说酶为脂肪酶和蛋白酶。
3、权利要求1的毛发营养剂,其中所说酶为大米酵母所得酶。
4、毛发营养剂生产方法,其步骤包括将酶加入含植物蛋白和植物脂的基质中,并过滤得到的发酵产品。
5、权利要求4的毛发营养剂生产方法,其中所说酶为脂肪酶和蛋白蛋白酶的混合物。
6、权利要求4的毛发营养剂生产方法,其中所说酶为大米酵母所得酶。
7、权利要求4的毛发营养剂生产方法,其步骤还包括将选自下列物质的PH6-11任一种或其任意组合作为缓冲剂加入基质中:
盐酸-可尼丁,一代磷酸钾-二代磷酸钾,盐酸-佛罗那钠,盐酸-三氨基甲烷,盐酸-硼砂,硼酸-碳酸钠,盐酸-氨基甲基丙二醇,氯化钠-氨,甘氨酸-氢氧化钠,硼酸-氢氧化钠,盐酸-二甲基甘氨酸钠,碳酸氢钠-碳酸钠,硼酸-氢氧化钠,硼酸-碳酸钠,盐酸-碳酸钠和二代磷酸钾(钠)-氢氧化钠等。
8、权利要求5的毛发营养剂生产方法,其步骤还包括将选自下列物质的PH6-11之任一种或其任意组合作为缓冲剂加入基质中:
盐酸-可尼丁,一代磷酸钾-二代磷酸钾,盐酸-佛罗那钠,盐酸-三氨基甲烷,盐酸-硼砂,硼酸-碳酸钠,盐酸-氨基甲基丙二醇,氯化钠-氨,甘氨酸-氢氧化钠,硼酸-氢氧化钠,盐酸-二甲基甘氨酸钠,碳酸氢钠-碳酸钠,硼砂-氢氧化钠,硼砂-碳酸钠,盐酸-碳酸钠和二代磷酸钾(钠)-氢氧化钠等。
9、权利要求6的毛发营养剂生产方法,其步骤还包括将选自下列物质的PH6-11之任一种或其任意组合作为缓冲剂加入基质中:
盐酸-可尼丁,一代磷酸钾-二代磷酸钾,盐酸-佛罗那钠,盐酸-三氨基甲烷,盐酸-硼砂,硼酸-碳酸钠,盐酸-氨基甲基丙二醇,氯化钠-氨,甘氨酸-氢氧化钠,硼酸-氢氧化钠,盐酸-二甲基甘氨酸钠,碳酸氢钠-碳酸钠,硼砂-氢氧化钠,硼砂-碳酸钠,盐酸-碳酸钠和二代磷酸钾(钠)-氢氧化钠等。
