免疫***必须在有效应答之间达到平衡以清除致病实体(例如在癌症中),同时保持耐受性以防止自身免疫性疾病。T细胞在维持免疫功能抑制和主动免疫排斥之间的平衡中起关键作用。T调节细胞(Treg)的特征在于CD25+、CD4+、FOXp3+和糖皮质激素诱导的肿瘤坏死因子相关受体(GITR)的表达。Treg抑制病理性免疫应答,且通过调节免疫自体耐受性最终保持免疫体内稳态。Treg的存在抑制负责清除各种病理实体的激活的效应T细胞的活性。
人上皮恶性肿瘤与循环中和肿瘤本身内两者存在的Treg的量增加有关。癌症患者中的抑制性Treg存在的增加导致常规T细胞(包括效应细胞)的抑制,这进而引起IFN-γ产生的减量调节。体内癌症动物模型中Treg的存在或活性的降低导致效应T细胞的量和活性增加,这之后通常是肿瘤大小的缩小和/或其它癌症症状的减轻。

抗体是单价的或二价的。例如抗体是单链抗体。抗体的结合亲和力范围为10-5 M-10-12 M。在某些方面,抗体具有IgG4重链恒定区。在其它方面,抗体具有在氨基酸第234和235位含有突变的Fc区。突变为例如L234A和L235A。
在其它方面,本发明包括含有本发明的人GITR抗体的双特异性抗体和还与肿瘤相关抗原、细胞因子或细胞表面受体结合的抗体。

图1是显示抗GITR抗体的结合亲和力的图。抗GITR抗体针对GITR表达293T细胞的结合亲和力。使表达GITR的293T细胞与不同浓度的抗GITR抗体在4℃下孵育1小时,然后用FITC标记的抗人Fc抗体在4℃下染色又1小时。细胞通过流式细胞术进一步检测,而抗GITR抗体的半最大有效浓度(EC50)通过Prism软件测量。这些数据表明抗GITR抗体显示对GITR的不同结合活性。

图5是Teff和Treg共培养物中被抗GITR抗体刺激的细胞因子概况的系列图。通过MSD V-PLEX Kit,测量以10的Teff/Treg比率在同一培养物中5天后的细胞因子生产。数据显示#1、#3、#10、#14、#15和#17可诱导IFN-γ分泌,#1、#3、#11、#15和#17可诱导IL-10分泌。总之,#1、#3、#15和#17可作为激动剂对GITR具有更好的活性。
发明详述
本发明提供针对糖皮质激素诱导的肿瘤坏死因子受体(亦称GITR)有特异性的人源化单克隆抗体。通过使用GITR作为文库选择靶,通过使用270亿人单链抗体(scFv)噬菌体展示文库鉴定抗体。这些抗体代表针对GITR的一类新的人单克隆抗体。
这些抗GITR人单克隆抗体在本文称为“huGITR抗体”。
有充分证明的证据表明在上皮癌的情况下调节T细胞(Treg)的量增加。还有证据表明GITR在由Treg维持的优势免疫自体耐受性中起关键作用。Treg上GITR表达和癌症期间Treg的增加之间的这种关系,允许靶向作为促进效应T细胞功能的手段的GITR活性的机会。具体地说,这使得靶向GITR,一种癌症治疗的潜在免疫治疗方法。
Treg表达CD28、CD4、FOXP3和GITR。通过FOXP3与活化T细胞核因子NF-AT的二聚化在很大程度上介导效应T细胞活性的抑制,这进而导致IFN-γ、IL-2和IL-4的抑制。已表明通过与其配体结合的GITR连接的增加降低Treg对活化T细胞具有的抑制作用。此外,还显示直接靶向GITR的抗体降低Treg抑制功能。
虽然GITR在Treg和效应T细胞两者中表达,但前者中GITR表达的量显著较大。因此,GITR被视为调节Treg在各种疾病(包括癌症)中的抑制功能的良好的候选靶标。鼠模型表明,GITR的刺激导致Treg抑制活性降低。其它研究还表明,拮抗GITR活性导致恶性细胞的Treg募集减弱。总之,这些数据表明在癌症病理生理学中GITR为决定性受体。
本发明提供特异性结合GITR蛋白的人单克隆抗体。本发明的抗体与GITR结合中断GITR配体与GITR结合的能力。通过不同的机制,huGITR抗体降低Treg对效应细胞具有的抑制功能。给予huGITR抗体可导致Treg耗减、效应T细胞(Teff)增殖增加、抗原特异性T细胞活性提高和效应细胞因子产生增加。在一些情况下,huGITR抗体促进或提高抗原特异性免疫应答。
因此,本发明的huGITR抗体可用于调节T细胞活性。具体地说,huGITR抗体可抑制Treg活性并刺激Teff活性。此外,本发明的huGITR抗体增加NK细胞的细胞毒性和增加IFNγ分泌。
huGITR抗体是单价的或二价的,并包含单链或双链。在功能上,huGITR抗体的结合亲和力在10-5M-10-12 M的范围。例如,huGITR抗体的结合亲和力为10-6 M-10-12 M、10-7 M-10-12 M、10-8 M-10-12 M、10-9 M-10-12 M、10-5 M-10-11 M、10-6 M-10-11 M、10-7 M-10-11 M、10-8M-10-11 M、10-9 M-10-11 M、10-10 M-10-11 M、10-5 M-10-10 M、10-6 M-10-10 M、10-7 M-10-10 M、10-8 M-10-10 M、10-9 M-10-10 M、10-5 M-10-9 M、10-6 M-10-9 M、10-7 M-10-9 M、10-8 M-10-9 M、10-5 M-10-8 M、10-6 M-10-8 M、10-7 M-10-8 M、10-5 M-10-7 M、10-6 M-10-7 M或10-5 M-10-6 M。
此外,本发明的抗体包括治疗剂,包括但不限于毒素、放射性标记、siRNA或细胞因子。
鉴定出11种独特的单克隆huGITR抗体。这些包括mAb #1-81、3-167、#5-139、#7-192、#10-116、#11-126、#12-46、#13-169、#14-182、#15-68和#17-60。可变区核酸序列和氨基酸序列见表1A-11B。与这些抗体的可变区有关的CDR的氨基酸序列见表12。

本文所述huGITR抗体与GITR结合。一方面,huGITR抗体对GITR具有高亲和力和高特异性。另一方面,huGITR抗体可结合GITR受体,并防止、抑制或阻断配体GITR-L结合其受体GITR。

本发明的特征还在于与本文所述huGITR抗体的氨基酸或核苷酸序列具有规定百分比同一性或相似性的抗体。例如,当与本文所述huGITR抗体的任一个的规定区域或全长相比时,抗体可具有60%、70%、75%、80%、85%、90%、91%、92%、93%、94%、95%、96%、97%、98%、99%或更高同一性。与本发明的核酸和蛋白质的序列同一性或相似性可通过本领域已知方法,通过序列比较和/或比对确定。例如,可利用序列比较算法(即BLAST或BLAST 2.0)、手工比对或目测确定本发明的核酸和蛋白质的百分比序列同一性或相似性。
对于氨基酸序列,本领域技术人员应容易认识到,改变、添加、缺失或取代编码序列中的单一氨基酸或少部分氨基酸的核酸、肽、多肽或蛋白质序列的各个取代、缺失或添加在本文统称为“保守修饰变体”。在一些实施方案中,改变导致氨基酸被化学上类似的氨基酸取代。提供在功能上类似的氨基酸的保守取代表是本领域众所周知的。与未修饰的GITR抗体相比,本文公开的huGITR抗体的这类保守修饰变体可显示与GITR的交叉反应性提高。
抗体
本文所用术语“抗体”是指免疫球蛋白分子和免疫球蛋白(Ig)分子的免疫活性部分,即含有与抗原特异性结合(免疫反应)的抗原结合部位的分子。所谓“特异性结合”或“与……免疫反应”意指与所需抗原的一个或多个抗原决定簇反应而不与其它多肽反应的抗体。抗体包括但不限于多克隆、单克隆、嵌合、dAb (结构域抗体)、单链、Fab、Fab’和F(ab')2片段、scFvs和Fab表达文库。
单链Fv (“scFv”)多肽分子是共价连接的VH:VL异二聚体,其可自包括通过肽编码接头连接的VH-和VL-编码基因的基因融合物表达。(参见Huston等(1988) Proc Nat AcadSci USA 85(16):5879-5883)。描述了辨别用于将来自抗体V区的天然聚集的而非化学分离的多肽轻链和重链转化至scFv分子的化学结构的多种方法,所述scFv分子会折叠成与抗原结合部位的结构大致相似的三维结构。参见例如美国专利号5,091,513、5,132,405和4,946,778。
幼稚人scFv文库已非常大且可创建以提供针对大量靶分子的重排抗体基因的极大来源。可从患有感染性疾病的个体构建较小的文库以分离疾病特异性抗体。(参见Barbas等, Proc. Natl. Acad. Sci. USA 89:9339-43 (1992);Zebedee等, Proc. Natl. Acad.Sci. USA 89:3175-79 (1992))。
一般来说,获自人的抗体分子与IgG、IgM、IgA、IgE和IgD类别的任一种有关,所述类别因存在于分子中的重链的性质而彼此不同。某些类别还具有亚类,例如IgG1、IgG2、IgG3和IgG4等。此外,在人中,轻链可为κ链或λ链。术语“抗原结合部位”或“结合部分”是指参与抗原结合的免疫球蛋白分子的一部分。抗原结合部位由重(“H”)和轻(“L”)链的N端可变(“V”)区的氨基酸残基形成。在重链和轻链V区内有3个高度趋异的序列段(称为“超变区”)***在更保守的侧翼序列段(称为“构架区”或“FR”)之间。因此,术语“FR”是指天然存在于免疫球蛋白的超变区之间并与之相邻的氨基酸序列。在抗体分子中,轻链的3个超变区和重链的3个超变区彼此相对配置在三维空间中形成抗原结合表面。抗原结合表面与被结合的抗原的三维表面互补,重链和轻链每个的3个超变区称为“互补决定区”或“CDR”。含有scFv抗体的CDR的VH和VL区见表1A-表11B。
本文所用术语“表位”包括能够与免疫球蛋白、scFv或T细胞受体特异性结合的任何蛋白质决定簇。表位决定簇通常由分子(例如氨基酸或糖侧链)的化学活性表面群聚(grouping)组成,并通常具有特定的三维结构特征以及特定的电荷特征。例如,抗体可针对多肽的N端或C端肽产生。
本文所用术语“免疫结合”和“免疫结合性质”是指免疫球蛋白分子与免疫球蛋白对其有特异性的抗原之间产生的非共价相互作用类型。免疫结合相互作用的强度或亲和力可以相互作用的解离常数(Kd)的方式表示,其中较小的Kd表示较大的亲和力。所选多肽的免疫结合性质可采用本领域众所周知的方法量化。一种这样的方法需要测量抗原结合部位/抗原复合物形成和解离的速率,其中这类速率取决于复合的配偶体的浓度、相互作用的亲和力和同等影响两个方向的速率的几何参数。因此,“结合速率常数(on rate constant)”(Kon)和“解离速率常数(off rate constant)” (Koff)两者可通过计算结合和解离的实际速率及浓度确定。(参见Nature 361:186-87 (1993))。Koff /Kon之比使得能够消除与亲和力无关的所有参数,并等于解离常数Kd。(一般参见Davies等(1990) Annual Rev Biochem 59:439-473)。当平衡结合常数(Kd) ≤10 μM、优选≤ 10 nM、更优选≤ 10 nM、最优选≤ 100pM-约1 pM时,本发明的抗体被称为与GITR表位特异性结合,通过测定法例如放射性配体结合测定法或本领域技术人员已知的类似测定法测量。
本发明的GITR蛋白或其衍生物、片段、类似物、同系物或直向同源物(ortholog),可在产生免疫特异性结合这些蛋白质组分的抗体时用作免疫原。与脂蛋白体偶联的GITR蛋白或其衍生物、片段、类似物、同系物或直向同源物可在产生免疫特异性结合这些蛋白质组分的抗体时用作免疫原。
本领域技术人员应认识到,有可能无需过度实验只是通过确定人单克隆抗体是否防止本发明的人单克隆抗体与GITR结合,便确定前者是否与后者具有相同的特异性。如果受测试的人单克隆抗体与本发明的人单克隆抗体竞争,正如通过本发明的人单克隆抗体的结合降低所示,则很可能2种单克隆抗体与相同的表位或与极相关的表位结合。
测定人单克隆抗体是否具有本发明的人单克隆抗体的特异性的另一方法是使本发明的人单克隆抗体与其通常与之有反应的GITR蛋白一起预孵育,然后加入要测试的人单克隆抗体以确定受测试的人单克隆抗体是否在其结合GITR的能力上受抑制。如果受测试的人单克隆抗体被抑制,则很可能具有与本发明的单克隆抗体相同的或在功能上等同的表位特异性。还可通过使用GITR并确定试验单克隆抗体是否能够中和GITR,来进行本发明的人单克隆抗体的筛选。
本领域已知的各种方法可用于产生针对本发明的蛋白质或针对其衍生物、片段、类似物、同系物或直向同源物的多克隆或单克隆抗体。(参见例如Antibodies: ALaboratory Manual, Harlow E和Lane D, 1988, Cold Spring Harbor LaboratoryPress, Cold Spring Harbor, NY,通过引用并入本文)。
抗体可通过众所周知的技术纯化,例如使用A蛋白或G蛋白(其主要提供免疫血清的IgG部分)的亲和层析法。随后或备选,可将作为所寻找的免疫球蛋白的靶标的特异性抗原或其表位固定在柱上以通过免疫亲和层析法纯化免疫特异性抗体。例如D. Wilkinson论述了免疫球蛋白的纯化(由The Scientist, Inc., Philadelphia PA出版的TheScientist, 第14卷, 第8期(2000年4月17日), 第25-28页)。
本文所用术语“单克隆抗体”或“MAb”或“单克隆抗体组合物”是指只含有由独特的轻链基因产物和独特的重链基因产物组成的抗体分子的一种分子物类的一群抗体分子。具体地说,单克隆抗体的互补决定区(CDR)在该群的所有分子中相同。MAb含有能够与特征在于对其有独特结合亲和力的抗原的特定表位免疫反应的抗原结合部位。
可采用杂交瘤方法,例如Kohler和Milstein,Nature,256:495 (1975)描述的那些方法,制备单克隆抗体。在杂交瘤方法中,小鼠、仓鼠或其它合适的宿主动物通常用免疫剂免疫以诱导产生或能够产生会与免疫剂特异性结合的抗体的淋巴细胞。或者,淋巴细胞可体外免疫。
免疫剂通常可包括蛋白质抗原、其片段或融合蛋白。一般而言,如果需要人源的细胞,则使用外周血淋巴细胞,或如果需要非人哺乳动物源,则使用脾细胞或***细胞。然后使用合适的融合剂(例如聚乙二醇)使淋巴细胞与永生化细胞系融合,形成杂交瘤细胞(Goding, Monoclonal Antibodies: Principles and Practice, Academic Press,(1986)第59-103页)。永生化细胞系通常是转化的哺乳动物细胞,特别是啮齿动物、牛和人源的骨髓瘤细胞。通常使用大鼠或小鼠骨髓瘤细胞系。可使杂交瘤细胞在合适的培养基中培养,所述培养基优选含有抑制未融合的永生化细胞的生长或存活的一种或多种物质。例如,如果亲代细胞缺乏酶次黄嘌呤鸟嘌呤磷酸核糖基转移酶(HGPRT或HPRT),用于杂交瘤的培养基通常会包括次黄嘌呤、氨基蝶呤和胸苷(“HAT培养基”),所述物质防止HGPRT缺陷型细胞的生长。
优选的永生化细胞系是这样的细胞系,其有效融合、支持所选的产抗体细胞产生稳定的高水平抗体表达且对培养基(例如HAT培养基)敏感。更优选的永生化细胞系是鼠骨髓瘤系,其可获自例如Salk Institute Cell Distribution Center, San Diego,California和American Type Culture Collection, Manassas, Virginia。还描述了用于产生人单克隆抗体的人骨髓瘤和小鼠-人异源骨髓瘤细胞系。(参见Kozbor, J. 然后可就针对抗原的单克隆抗体的存在对杂交瘤细胞培养在其中的培养基进行测定。优选通过免疫沉淀法或通过体外结合测定法,例如放射免疫测定法(RIA)或酶联免疫吸附测定法(ELISA),测定由杂交瘤细胞产生的单克隆抗体的结合特异性。这类技术和测定法是本领域已知的。例如,可通过Munson和Pollard, Anal. Biochem., 107:220 (1980)的Scatchard分析测定单克隆抗体的结合亲和力。此外,在单克隆抗体的治疗应用中,重要的是鉴定对靶抗原具有高特异性程度和高结合亲和力的抗体。
在鉴定出所需的杂交瘤细胞后,克隆可通过有限稀释方法亚克隆,并通过标准方法生长。(参见Goding, Monoclonal Antibodies: Principles and Practice, AcademicPress, (1986), 第59-103页)。用于此目的的合适的培养基包括例如Dulbecco改进的Eagle培养基和RPMI-1640培养基。或者,杂交瘤细胞可在哺乳动物中作为腹水体内生长。
可通过常规免疫球蛋白纯化方法,例如A蛋白-琼脂糖凝胶、羟磷灰石层析法、凝胶电泳、透析或亲和层析法,从培养基或腹水流体中分离并纯化由亚克隆分泌的单克隆抗体。
还可通过例如描述于美国专利号4,816,567的重组DNA方法制备单克隆抗体。可采用常规方法(例如通过使用能够与编码鼠抗体的重链和轻链的基因特异性结合的寡核苷酸探针),容易地分离和测序编码本发明的单克隆抗体的DNA。本发明的杂交瘤细胞用作这类DNA的优选来源。一旦分离,可将DNA置于表达载体中,然后将表达载体转染至不再另产生免疫球蛋白蛋白质的宿主细胞(例如猿猴COS细胞、中华仓鼠卵巢(CHO)细胞或骨髓瘤细胞)中,以在重组宿主细胞中实现单克隆抗体的合成。还可通过例如置换人重链和轻链恒定结构域的编码序列代替同源鼠序列(参见美国专利号4,816,567;Morrison,Nature 368,812-13 (1994))或通过共价连接免疫球蛋白编码序列与非免疫球蛋白多肽的编码序列的全部或部分,来修饰DNA。这类非免疫球蛋白多肽可取代本发明抗体的恒定结构域,或可取代本发明抗体的一个抗原结合部位的可变结构域以产生嵌合二价抗体。
完全人抗体是其中轻链和重链两者的完整序列(包括CDR)产生于人基因的抗体分子。这类抗体在本文被称为“人源化抗体”、“人抗体”或“完全人抗体”。可通过采用trioma技术;人B细胞杂交瘤技术(参见Kozbor等, 1983 Immunol Today 4: 72))和产生人单克隆抗体的EBV杂交瘤技术(参见Cole等, 1985, 载于: Monoclonal Antibodies and CancerTherapy, Alan R. 另外可采用经修饰使得在响应抗原攻击时产生完全人抗体而非动物的内源抗体的转基因非人动物,来产生人抗体。(参见PCT公开文本WO94/02602)。使编码免疫球蛋白重链和轻链的内源基因在非人宿主中失能,并将编码人重链和轻链免疫球蛋白的活性基因座***宿主的基因组中。例如,使用含有必要人DNA区段的酵母人工染色体,掺入人基因中。然后通过使含有少于修饰的完全互补部分的中间转基因动物杂交,获得提供所有所需修饰的动物作为子代。这类非人动物的优选实施方案是小鼠,称为XenomouseTM,正如PCT公开文本WO 96/33735和WO 96/34096所公开的。该动物产生分泌完全人免疫球蛋白的B细胞。抗体可直接获自用目标免疫原免疫的动物,例如多克隆抗体的制备物,或备选获自来源于动物的永生化B细胞,例如产生单克隆抗体的杂交瘤。此外,编码具有人可变区的免疫球蛋白的基因可恢复或表达以直接获得抗体,或可进一步修饰以获得抗体例如单链Fv (scFv)分子的类似物。
产生缺乏内源免疫球蛋白重链表达的非人宿主(以小鼠为例)的方法的实例公开于美国专利号5,939,598。还可通过这样的方法获得,所述方法包括自胚胎干细胞中的至少一个内源重链基因座缺失J区段基因以防止基因座的重排并防止重排免疫球蛋白重链基因座的转录物的形成,所述缺失通过含有编码选择标记的基因的打靶载体实现;并从胚胎干细胞产生其体细胞和生殖细胞含有编码选择标记的基因的转基因小鼠。
用于产生目标抗体(例如人抗体)的一种方法公开于美国专利号5,916,771。该方法包括将含有编码重链的核苷酸序列的表达载体导入培养中的一种哺乳动物宿主细胞中,将含有编码轻链的核苷酸序列的表达载体导入另一种哺乳动物宿主细胞中,并将两种细胞融合形成杂交细胞。杂交细胞表达含有重链和轻链的抗体。
在对该方法的进一步改进中,鉴定免疫原上的临床相关表位的方法和选择以高亲和力与相关表位免疫特异性结合的抗体的相关方法公开于PCT公开文本WO 99/53049。
可通过含有编码上述单链抗体的DNA区段的载体表达抗体。
这些可包括载体、脂质体、裸DNA、佐剂辅助DNA (adjuvant-assisted DNA)、基因枪、导管等。载体包括例如描述于WO 93/64701的化学缀合物,其具有打靶部分(例如细胞表面受体的配体)和核酸结合部分(例如聚赖氨酸))、病毒载体(例如DNA或RNA病毒载体);融合蛋白(例如描述于PCT/US 95/02140 (WO 95/22618),其是含有靶向部分(例如对靶细胞有特异性的抗体)和核酸结合部分(例如鱼精蛋白)的融合蛋白);质粒;噬菌体等。载体可为染色体的、非染色体的或合成的。
优选的载体包括病毒载体、融合蛋白和化学缀合物。逆转录病毒载体包括莫洛尼鼠白血病毒。优选DNA病毒载体。这些载体包括痘病毒载体(pox vector)例如正痘病毒或禽痘病毒载体(orthopox或avipox vector);疱疹病毒载体例如单纯疱疹I病毒(HSV)载体(参见Geller, A. I.等, J. Neurochem, 64:487 (1995);Lim, F.等, 载于DNA Cloning:Mammalian Systems, D. Glover编辑(Oxford Univ. Press, Oxford England) (1995);Geller, A. I.等, Proc Natl. Acad. Sci.: U.S.A. 90:7603 (1993);Geller, A. 痘病毒载体将基因导入细胞的细胞质中。禽痘病毒载体只引起核酸的短期表达。优选腺病毒载体、腺伴随病毒载体和单纯疱疹病毒(HSV)载体用于将核酸导入神经细胞中。腺病毒载体导致比腺伴随病毒(约4个月)短的表达(约2个月),这进而比HSV载体短。所选的具体载体将取决于靶细胞和待治疗的病况。导入可通过标准技术进行,例如感染、转染、转导或转化。基因转移方式的实例包括例如裸DNA、CaPO4沉淀、DEAE葡聚糖、电穿孔、原生质体融合、脂转染、细胞显微注射和病毒载体。
载体可用来实质上靶向任何所需的靶细胞。例如,可采用立体定位注射以将载体(例如腺病毒、HSV)引导至所需位置。此外,可使用小型泵输注***,例如SynchroMed输注***,通过脑室内(icv)输注递送颗粒。也已证实基于称为对流的总体流动的方法在将大分子递送至脑的延伸区域是有效的,并可用于将载体递送至靶细胞。(参见Bobo等, Proc.Natl. Acad. Sci. USA 91:2076-2080 (1994);Morrison等, Am. J. Physiol. 266:292-305 (1994))。可采用的其它方法包括导管、静脉内、胃肠外、腹膜内和皮下注射及口服或其它已知的给药途径。
可使用这些载体以表达可以多种方式使用的大量抗体。例如,检测样品中GITR的存在。抗体还可用来尝试与GITR结合并破坏GITR活性。
可改编用于产生对本发明的抗原蛋白有特异性的单链抗体的技术(参见例如美国专利号4,946,778)。另外,可改编用于构建Fab表达文库的方法(参见例如Huse等, 1989Science 246: 1275-1281),以允许快速有效地鉴定具有对蛋白质或其衍生物、片段、类似物或同系物有所需特异性的单克隆Fab片段。含有蛋白质抗原的个体遗传型的抗体片段可通过本领域已知技术产生,包括但不限于:(i)通过抗体分子的胃蛋白酶消化产生的F(ab')2片段;(ii)通过还原F(ab')2片段的二硫桥产生的Fab片段;(iii)通过抗体分子用木瓜蛋白酶和还原剂处理产生的Fab片段和(iv) Fv片段。
杂缀合物抗体也在本发明的范围内。杂缀合物抗体由2种共价连接的抗体组合。这类抗体被提议为例如使免疫***细胞靶向不需要的细胞(参见美国专利号4,676,980)和用于治疗HIV感染(参见WO 91/00360、WO 92/200373、EP 03089)。预期可采用合成蛋白质化学的已知方法,包括涉及交联剂的方法,体外制备抗体。例如,免疫毒素可使用二硫化物交换反应或通过形成硫醚键构建。用于该目的的合适的试剂的实例包括亚氨基硫醇盐和甲基-4-mercaptobutyrimidate和公开于例如美国专利号4,676,980中的那些。
对于效应子功能,可能需要修饰本发明的抗体,以提高例如抗体在治疗癌症中的疗效。例如,可将半胱氨酸残基引入Fc区,因此允许这个区中的链间二硫键形成。由此产生的同二聚体抗体可具有改进的内化能力和/或升高的补体介导的细胞杀死和依赖抗体的细胞毒性(ADCC)。(参见Caron等, J. Exp Med., 176: 1191-1195 (1992)和Shopes, J.Immunol., 148: 2918-2922 (1992))。或者,抗体可经工程改造以具有双重Fc区,因此可具有提高的补体溶解和ADCC能力。(参见Stevenson等, Anti-Cancer Drug Design, 3: 219-230 (1989))。
在某些实例方案中,本发明的抗体可包含含有改变抗体的依赖抗原的效应子功能(特别是抗体的循环半寿期)的氨基酸取代的Fc变体。当与缺乏这些取代的抗体相比时,所述抗体显示与FcRn的结合增加或降低,因此,血清中的半寿期分别延长或缩短。预期对FcRn的亲和力改进的Fc变体具有较长的血清半寿期,且所述分子在治疗哺乳动物的方法(其中需要所给予抗体的长的半寿期)中具有有益应用以例如治疗慢性疾病或病症。相比之下,预期FcRn结合亲和力降低的Fc变体具有较短的半寿期,且这类分子还可用于例如给予其中循环时间缩短可能是有益的哺乳动物,例如体内诊断成像或用于其中起始抗体当长期存在于循环中具有毒副作用的情况。FcRn结合亲和力降低的Fc变体也不太可能透过胎盘,因此,还可用于治疗妊娠妇女的疾病或病症。另外,其中可能需要FcRn结合亲和力降低的其它应用包括其中需要定位于脑、肾和/或肝的那些应用。在一个示例性的实施方案中,本发明的改变的抗体显示自血管跨过肾小球上皮的运输减少。在另一个实施方案中,本发明的改变的抗体显示自脑跨过血脑屏障(BBB)进入血管空间的运输减少。在一个实施方案中,FcRn结合改变的抗体包含在Fc结构域的“FcRn结合环”内具有一个或多个氨基酸取代的Fc结构域。FcRn结合环由氨基酸残基280-299 (按照EU编号)组成。改变FcRn结合活性的示例性氨基酸取代公开于国际PCT公开号WO05/047327,其通过引用结合到本文中。在某些示例性的实施方案中,本发明的抗体或其片段包含具有以下取代的一个或多个的Fc结构域:V284E、H285E、N286D、K290E和S304D (EU编号)。
在一些实施方案中,将突变引入mAb的恒定区,使得mAb的依赖抗体的细胞介导的细胞毒性(ADCC)活性改变。例如,突变是CH2结构域中的LALA突变。一方面,bsAb在异二聚体mAb的1个scFv单元上含有突变,其降低ADCC活性。另一方面,mAb在异二聚体mAb的两条链上含有突变,这完全消除ADCC活性。例如,引入mAb的一个或两个scFv单元的突变是CH2结构域中的LALA突变。可优化具有可变ADCC活性的这些mAb,使得mAb对表达被mAb识别的一种抗原的细胞显示最大选择性杀灭,然而对被mAb识别的第二种抗原显示最小杀灭。
在其它实施方案中,用于本文所述诊断和治疗方法的抗体具有恒定区,例如经改变以降低或消除糖基化的IgGl或IgG4重链恒定区。例如,本发明的抗体还可包含Fc变体,其包含改变抗体的糖基化的氨基酸取代。例如,所述Fc变体可能具有减少的糖基化(例如N-或O-联糖基化)。在示例性实施方案中,Fc变体包含正常存在于氨基酸第297位(EU编号)的N-联聚糖的减少的糖基化。在另一个实施方案中,抗体在糖基化基序附近或在糖基化基序内具有氨基酸取代,例如,含有氨基酸序列NXT或NXS的N-联糖基化基序。在一个具体的实施方案中,抗体包含在氨基酸第228或299位(EU编号)具有氨基酸取代的Fc变体。在更具体的实施方案中,抗体包含含有S228P和T299A突变(EU编号)的IgGl或IgG4恒定区。
使糖基化减少或改变的示例性氨基酸取代公开于国际PCT公开号WO05/018572,其通过引用结合到本文中。在优选的实施方案中,本发明的抗体或其片段经修饰以消除糖基化。所述抗体或其片段可称为“agly”抗体或其片段(例如“agly”抗体)。虽然不受理论的束缚,但认为“agly”抗体或其片段体内可具有改进的安全性和稳定性概况。示例性agly抗体或其片段包含IgG4抗体的去糖基化Fc区,其没有Fc-效应子功能,因此消除表达GITR的正常生命器官的Fc介导的毒性的可能性。在另外的其它实施方案中,本发明的抗体或其片段包含改变的聚糖。例如,抗体在Fc区的Asn297的N-聚糖上岩藻糖残基的数目可减少,即去岩藻糖化。在另一个实施方案中,抗体在Fc区Asn297上的N-聚糖的唾液酸残基的数目可改变。iii)共价连接。
本发明还涉及包含与细胞毒性剂例如毒素(例如细菌、真菌、植物或动物来源的酶促活性毒素或其片段)或放射性同位素(即放射性缀合物)缀合的抗体的免疫缀合物。
可使用的酶促活性毒素及其片段包括白喉毒素A链、白喉毒素的非结合活性片段、外毒素A链(来自铜绿假单胞菌(Pseudomonas aeruginosa))、蓖麻毒蛋白A链、相思豆毒蛋白A链、塑莲根毒蛋白A链、α-帚曲霉素、油桐(Aleurites fordii)蛋白、石竹素蛋白、美洲商陆(Phytolaca americana)蛋白(PAPI、PAPII、和PAP-S)、苦瓜(momordica charantia)抑制剂、麻风树毒蛋白、巴豆毒蛋白、肥皂草(sapaonaria officinalis)抑制剂、多花白树毒蛋白、丝林霉素、局限曲菌素、酚霉素、伊诺霉素和单端孢菌霉素。可获得用于产生放射性缀合的抗体的各种放射性核素。实例包括212Bi、131I、131In、90Y和186Re。
使用各种双官能蛋白质偶联剂例如N-琥珀酰亚胺基-3-(2-吡啶基二硫醇)丙酸酯(SPDP)、亚氨基噻烷(IT)、亚氨基酯的双官能衍生物(例如己二酰亚胺二甲酯HCL)、活性酯(例如辛二酸二琥珀酰亚胺)、醛(例如戊二醛)、双叠氮基化合物(例如双(对叠氮基苯甲酰基)己烷二胺)、双-重氮衍生物(例如双-(对重氮苯甲酰基)-乙二胺)、二异氰酸酯类(例如甲苯2,6-二异氰酸酯)和双-活性氟化合物(例如1,5-二氟-2,4-二硝基苯),制备抗体和细胞毒性剂的缀合物。例如,蓖麻毒蛋白免疫毒素可按Vitetta等, Science 238: 1098(1987)所述制备。碳-14标记的1-异硫氰酸基苄基-3-甲基二亚乙基三胺五乙酸(MX-DTPA)是用于放射性核苷酸(radionucleotide)与抗体缀合的示例性螯合剂(参见WO94/11026)。
本领域普通技术人员应认识到,各种各样可能的部分可与所得抗体或本发明的其它分子偶联。(参见例如"Conjugate Vaccines", Contributions to Microbiology andImmunology, J. M. Cruse和R. E. Lewis, Jr (编辑), Carger Press, New York,(1989),其全部内容通过引用并入本文)。
偶联可通过会使两种分子结合的任何化学反应实现,只要抗体和另一部分保留其各自的活性。这种连接可包括许多化学机制,例如共价结合、亲和力结合、***、配位结合和络合。然而优选的结合是共价结合。共价结合可通过现有侧链的直接缩合或通过外部桥接分子的掺入实现。许多二价或多价连接剂可用于将蛋白质分子(例如本发明的抗体)与其它分子偶联。例如,代表性偶联剂可包括有机化合物例如硫酯、碳二亚胺、琥珀酰亚胺、二异氰酸酯、戊二醛、重氮基苯和己二胺。该列表无意穷尽本领域已知的各种类别的偶联剂,而相反只是典型的更常见的偶联剂。(参见Killen和Lindstrom, Jour. 上述接头含有具有不同属性的组分,因此产生具有不同生理化学性质的缀合物。例如,烷基羧酸酯的磺基-NHS酯比芳族羧酸酯的磺基-NHS酯更稳定。与磺基-NHS酯相比,含NHS-酯的接头较不溶解。此外,接头SMPT含有位阻二硫键,并可形成稳定性提高的缀合物。二硫键与其它健相比一般较不稳定,因为二硫键体外被切割,导致可获得较少的缀合物。具体地说,磺基-NHS可提高碳二亚胺偶联物的稳定性。碳二亚胺偶联物(例如EDC)当与磺基-NHS联用时,形成比仅碳二亚胺偶联反应更耐水解的酯。
本文公开的抗体还可作为免疫脂质体配制。含有抗体的脂质体通过描述于例如以下的本领域已知方法制备:Epstein等, Proc. Natl. Acad. Sci. USA, 82: 3688(1985);Hwang等, Proc. Natl Acad. Sci. USA, 77: 4030 (1980)以及美国专利号4,485,045和4,544,545。循环时间延长的脂质体公开于美国专利号5,013,556。
特别有用的脂质体可用包含磷脂酰胆碱、胆固醇和PEG衍生化磷脂酰乙醇胺(PEG-PE)的脂质组合物通过反相蒸发方法产生。使脂质体通过规定孔径的过滤器流出,得到具有所需直径的脂质体。可按Martin等, J. Biol. Chem., 257: 286-288 (1982)所述通过二硫化物交换反应使本发明抗体的Fab’片段与脂质体缀合。
针对GITR的抗体的用途
可给予特异性结合本发明的GITR蛋白或其片段的抗体以治疗GITR相关疾病或病症。“GITR相关疾病或病症”包括疾病状态和/或与疾病状态有关的症状,其中发现GITR的水平升高和/或涉及GITR的细胞信号传导途径的活化。示例性GITR相关疾病或病症包括但不限于癌症和炎性疾病。
许多癌症过量表达GITR,GITR的增量调节与高风险预后因子有关。肿瘤细胞中GITR或其配体GITR-L的过量表达还可表明肿瘤细胞籍以逃避抗肿瘤免疫的机制。这类癌症包括实体瘤和血液肿瘤。使用本发明的抗体抑制或耗减Treg和刺激Teff。另外,本发明的抗体增加NK细胞的毒性并增加IFNγ产量。
本发明的抗体,包括双特异性、多克隆、单克隆、人源化和完全人抗体可用作治疗剂。所述治疗剂一般可用来治疗或预防受试者的癌症,提高疫苗效力或提高天然免疫应答。将抗体制备物、优选对于靶抗原具有高特异性和高亲和力的抗体制备物给予受试者,并且由于其与靶标结合通常会发挥作用。给予抗体可破坏或抑制或干扰GITR蛋白的活性。
可以药物组合物的形式给予特异性结合本发明的GITR蛋白或其片段的抗体以治疗癌症。例如在Remington: The Science And Practice Of Pharmacy 第19版. (AlfonsoR. Gennaro等编辑) Mack Pub. 治疗有效量的本发明的抗体一般涉及实现治疗目的所需的量。如上所述,这可以是抗体及其靶抗原之间的结合相互作用,这在某些情况下干扰靶标的功能。需要给予的量此外将取决于抗体对其特异性抗原的结合亲和力,并且还将取决于从其所给予的其它受试者的自由体积中耗减所给予的抗体的速率。以非限制性实例来说,治疗有效剂量的本发明的抗体或抗体片段的常见范围可为约0.1 mg/kg体重-约50 mg/kg体重。常见的给药频率的范围可为例如每天两次至一周一次。
当使用抗体片段时,优选与靶蛋白的结合结构域特异性结合的最小抑制片段。例如,根据抗体的可变区序列,可设计肽分子以保持结合靶蛋白序列的能力。这类肽可化学合成和/或通过重组DNA技术产生。(参见例如Marasco等, Proc. Natl. Acad. Sci. USA,90: 7889-7893 (1993))。制剂还可含有待治疗的具体适应症所需的一种以上的活性化合物,优选不会彼此不利影响的具有补充活性的活性化合物。备选或另外,组合物可包含提高其功能的作用剂,例如细胞毒性剂、细胞因子(例如IL-15)、化疗剂或生长抑制剂。这类分子适宜以对预期目的是有效的量组合存在。
还可将活性成分包封在例如分别通过凝聚技术或界面聚合制备的微胶囊(例如羟甲基纤维素或明胶微胶囊和聚甲基丙烯酸甲酯微胶囊);胶态药物递送***(例如脂质体、白蛋白微球体、微乳、纳米粒和纳米胶囊)或粗乳状液中。
待用于体内给药的制剂必须是无菌的。这容易通过经无菌滤过膜过滤实现。
可制备缓释制剂。缓释制剂的合适实例包括含有抗体的固体疏水聚合物的半透性基质,所述基质为成型制品的形式,例如膜或微胶囊。缓释基质的实例包括聚酯、水凝胶(例如聚(2-羟基乙基甲基丙烯酸酯)或聚乙烯醇)、聚丙交酯(美国专利号3,773,919)、L-谷氨酸和γ乙基-L-谷氨酸酯的共聚物、不可降解的乙烯乙酸乙酯、可降解的乳酸-乙醇酸共聚物例如LUPRON DEPOT TM (由乳酸-乙醇酸共聚物和醋酸亮丙瑞林组成的注射用微球体)和聚-D-(-)-3-羟基丁酸。虽然聚合物(例如乙烯乙酸乙酯和乳酸-乙醇酸)使得能够释放分子超过100天,但某些水凝胶释放蛋白质较短的时间。
本发明的抗体可用作检测样品中GITR (或其蛋白质或蛋白质片段)存在的作用剂。优选抗体含有可检测标记。抗体可为多克隆,或更优选为单克隆。可使用完整抗体或其片段(例如Fab、scFv或F(ab)2)。关于探针或抗体,术语“标记的”旨在包括通过使可检测物质与探针或抗体偶联(即物理连接)直接标记探针或抗体,以及通过与直接标记的另一种试剂反应间接标记探针或抗体。间接标记的实例包括如下检测第一抗体:使用荧光标记的第二抗体和用生物素末端标记DNA探针使得它可用荧光标记的链霉抗生物素检出。术语“生物样品”旨在包括自受试者分离的组织、细胞和生物流体,以及存在于受试者内的组织、细胞和流体。因此,术语“生物样品”用法范围内包括血液和血液的部分或组分(包括血清、血浆)或淋巴。也就是说,本发明的检测方法可用来体外以及体内检测生物样品中的分析物mRNA、蛋白质或基因组DNA。例如,用于检测分析物mRNA的体外技术包括RNA杂交和原位杂交。用于检测分析物蛋白质的体外技术包括酶联免疫吸附测定法(ELISA)、蛋白质印迹法、免疫沉淀法和免疫荧光法。用于检测分析物基因组DNA的体外技术包括DNA杂交。用于进行免疫测定法的程序描述于例如“ELISA: Theory and Practice: Methods in Molecular Biology”,第42卷, J. 可将针对GITR蛋白(或其片段)的抗体用于涉及本领域内涉及GITR蛋白的定位和/或定量的已知方法中(例如用于测量合适生理样品内的GITR蛋白的水平、用于诊断方法、用于使蛋白质成像等)。在规定的实施方案中,含有抗体来源的抗原结合结构域、对GITR蛋白或其衍生物、片段、类似物或同系物有特异性的抗体用作药理学活性化合物(下文称为“治疗剂”)。
可通过标准技术,例如免疫亲和层析法或免疫沉淀法,使用对本发明的GITR蛋白有特异性的抗体分离GITR多肽。在诊断上可使用针对GITR蛋白(或其片段)的抗体以监测组织中的蛋白质水平作临床试验程序的一部分,例如以确定规定治疗方案的功效。可通过使抗体与可检测物质偶联(即物理连接)促进检测。可检测物质的实例包括各种酶、辅基、荧光物质、发光物质、生物发光物质和放射性物质。合适酶的实例包括辣根过氧化物酶、碱性磷酸酶、β-半乳糖苷酶或乙酰胆碱酯酶;合适辅基复合物的实例包括链霉抗生物素/生物素和抗生物素蛋白/生物素;合适荧光物质的实例包括伞形酮、荧光素、异硫氰酸荧光素、罗丹明、二氯三嗪基胺荧光素、丹磺酰氯或藻红蛋白;发光物质的实例包括鲁米诺;生物发光物质的实例包括萤光素酶、萤光素和水母发光蛋白,而合适的放射性物质的实例包括125I、131I、35S或3H。
药物组合物
可将本发明抗体或作用剂(本文亦称为“活性化合物”)及其衍生物、片段、类似物和同系物掺入适于给药的药物组合物中。这类组合物通常包含抗体或作用剂和药学上可接受的载体。本文所用术语“药学上可接受的载体”旨在包括与药物给予相容的任何和所有溶剂、分散介质、包衣材料、抗细菌剂和抗真菌剂、等渗剂和吸收延迟剂等。合适的载体描述于本领域的标准参考教材Remington’s Pharmaceutical Sciences的最近版本,其通过引用并入本文。这类载体或稀释剂的优选实例包括但不限于水、盐水、林格氏液、葡萄糖溶液和5%人血清白蛋白。还可使用脂质体和非水性载体例如固定油。用于药用活性物质的这类介质和作用剂的使用是本领域众所周知的。除了任何常规介质或作用剂与活性化合物相容以外,还考虑了其在组合物中的使用。还可将补充活性化合物掺入组合物中。
配制本发明的药物组合物与其既定给药途径相容。给药途径的实例包括胃肠外,例如静脉内、皮内、静脉内、口服(例如口用)、经皮(即局部)、跨黏膜和直肠给药。用于胃肠外、皮内或皮下应用的溶液剂或混悬剂可包括下列组分:无菌稀释剂例如注射用水、盐水溶液、固定油、聚乙二醇、甘油、丙二醇或其它合成溶剂;抗细菌剂例如苄醇或对羟基苯甲酸甲酯;抗氧化剂例如抗坏血酸或亚硫酸氢钠;螯合剂例如乙二胺四乙酸(EDTA);缓冲剂例如乙酸酯、柠檬酯或磷酸酯;和调节张度的物质例如氯化钠或葡萄糖。可用酸或碱(例如盐酸或氢氧化钠)调节pH。可将胃肠外制剂装入安瓿、一次性注射器或用玻璃或塑料制造的多剂量小瓶中。
适于注射用的药物组合物包括无菌水性溶液剂(其中是水溶性的)或用于无菌注射用溶液剂或分散剂的临用时制备的分散剂和无菌粉剂。对于静脉内给药,合适的载体包括生理盐水、注射用抑菌水、Cremophor ELTM (BASF,Parsippany,N.J.)或磷酸缓冲盐水(PBS)。在所有情况下,组合物必须是无菌的,并且就存在流畅注射性而言应是流体。在制造和储存条件下必须是稳定的,并且必须针对微生物(例如细菌和真菌)的污染作用进行防腐。载体可以是溶剂或分散介质,含有例如水、乙醇、多元醇(例如甘油、丙二醇和液体聚乙二醇等)及其合适的混合物。例如,可通过使用包衣材料例如卵磷脂、在分散体的情况下通过保持所需粒径和通过使用表面活性剂,保持适当的流动性。可通过各种抗细菌剂和抗真菌剂,例如对羟基苯甲酸酯、三氯叔丁醇、苯酚、抗坏血酸、硫柳汞等,实现微生物作用的防止。在许多情况下,优选可在组合物中包括等渗剂例如糖、多元醇例如甘露糖醇、山梨糖醇、氯化钠。可在组合物中包括延迟吸收的物质(例如单硬脂酸铝和明胶),来引起注射用组合物的吸收延长。
可通过将所需量的活性化合物与按需要与上列成分的一种或组合一起掺入合适的溶剂中,接着过滤除菌,来制备无菌注射用溶液剂。一般而言,分散剂通过将活性化合物掺入含有基础分散介质和上列所需的其它成分的无菌溶媒中来制备。在用于制备无菌注射用溶液剂的无菌粉剂的情况下,制备方法是真空干燥和冷冻干燥,得到来自其前述除菌过滤溶液的活性成分加任何其它所需成分的粉剂。
口服组合物一般包括惰性稀释剂或可食用载体。可将它们包封在明胶胶囊中或压制成片剂。对于口服治疗性给药的目的,活性化合物可与赋形剂一起掺入,并以片剂、含片或胶囊剂的形式使用。口服组合物还可以使用流体载体制备以用作漱口剂,其中流体载体中的化合物口内使用、漱口并吐出或吞咽。可包括药物上相容的粘合剂和/或辅料作为组合物的部分。片剂、片剂、胶囊剂、含片等可含有下列成分的任一种或类似性质的化合物:粘合剂例如微晶纤维素、西黄蓍胶或明胶;赋形剂例如淀粉或乳糖;崩解剂例如藻酸、Primogel或玉米淀粉;润滑剂例如硬脂酸镁或Sterotes;助流剂例如胶态二氧化硅;甜味剂例如蔗糖或糖精;或矫味剂例如欧薄荷、水杨酸甲酯或橙味调味料。
对于吸入给药,从含有合适抛射剂(例如二氧化碳等气体)的加压容器或分配器中以喷雾剂的形式递送化合物。
全身给药还可通过跨黏膜或经皮手段。对于跨黏膜或经皮给药,在制剂中使用适于通过待渗透的屏障的渗透剂。这类渗透剂通常是本领域已知的,且对于跨黏膜给药包括洗涤剂、胆盐和梭链孢酸衍生物。跨黏膜给药可通过使用鼻腔喷雾剂或栓剂完成。对于经皮给药,按本领域普遍已知的,将活性化合物配制成软膏剂、油膏剂、凝胶剂或乳膏剂。
还可以栓剂(例如用常规栓剂基料例如可可脂和其它甘油酯)或滞留型灌肠剂的形式制备化合物用于直肠递送。
在一个实施方案中,活性化合物用保护化合物免于从身体快速清除的载体制备,例如控释制剂,包括植入剂和微囊化递送***。可使用生物可降解的生物相容的聚合物,例如乙烯乙酸乙酯、聚酐、聚乙醇酸、胶原、聚原酸酯和聚乳酸。用于制备这类制剂的方法对本领域技术人员而言将是显而易见的。原料还可市购获自Alza Corporation和NovaPharmaceuticals,Inc。脂质体混悬液(包括靶向用抗病毒抗原的单克隆抗体感染的细胞的脂质体)也可用作药学上可接受的载体。这些可按照本领域技术人员已知的方法例如美国专利号4,522,811中的描述制备。
尤其有利的是将口服或胃肠外组合物配制在剂量单位形式中以易于给药剂量均匀性。本文所用剂量单位形式是指适于作为待治疗的受试者的单位剂量的物理离散单位;各单位含有经计算产生所需治疗作用的预定量的活性化合物以及所需要的药用载体。本发明的剂量单位形式的规格受制于并直接取决于活性化合物的独特性质和要达到的具体治疗作用及配混用于治疗个体的这类活性化合物领域中的固有限制。
药物组合物可与给药说明一起包括在容器、药包或分配器中。
诊断测定
huGITR抗体在诊断上可用于例如监测免疫细胞病症(例如CLL)的发生或进展作为临床试验程序的一部分以例如确定指定治疗和/或预防方案的功效。
在某些方面,对于诊断目的,本发明的huGITR抗体与可检测部分连接,提供用于检测患有癌症或慢性感染的受试者中的T细胞耗竭的方法。
可检测部分可与抗体或片段直接缀合或通过使用例如荧光第二抗体间接缀合。直接缀合可通过例如荧光团与抗体或抗体片段的标准化学偶联或通过基因工程完成。可构建含有与荧光或生物发光蛋白质偶联的抗体或抗体片段的嵌合体或融合蛋白。例如,Casadei等人描述了制备能够在哺乳动物细胞中表达水母发光蛋白和抗体基因的融合蛋白的载体构建体的方法。
关于探针或抗体的本文所用术语“标记的”旨在包括通过使可检测物质与探针或抗体偶联(即物理连接)直接标记探针或抗体,以及通过与直接标记的另一种试剂反应间接标记探针或抗体。间接标记的实例包括如下检测第一抗体:使用荧光标记的第二抗体和用生物素末端标记DNA探针使得它可用荧光标记的链霉抗生物素检出。术语“生物样品”旨在包括自受试者分离的组织、细胞和生物流体(例如活检样品),以及存在于受试者内的组织、细胞和流体。也就是说,本发明的检测方法可用来体外以及体内检测生物样品中表达GITR的细胞。例如,用于检测GITR的体外技术包括酶联免疫吸附测定法(ELISA)、蛋白质印迹法、免疫沉淀法和免疫荧光法。此外,用于检测GITR的体内技术包括将标记的抗GITR抗体引入受试者中。例如,抗体可用放射性标记进行标记,所述放射性标记在受试者中的存在和位置可通过标准成像技术检测。在“靶向”缀合物的情况下,也就是说含有打靶部分(经设计使缀合物定位于受试者或动物内的一个或多个具体部位的分子或特征)的缀合物的情况下,定位是指当在受试者内在结合的“被定位”实体和未结合的“游离”实体之间基本上达到平衡时的状态。达到所述平衡时的速率取决于给药途径。例如,通过静脉内注射给予以定位于血栓的缀合物可在注射的几分钟内在血栓处定位或蓄积。另一方面,通过口服给予以定位于肠中的感染的缀合物可能花几小时达到定位。或者,定位可能只是指在给予实体后的选定时间受试者或动物内该实体的位置。再举例来说,在给予后当某一部分得以分布时,定位得以实现。
在所有上述情况下,本领域技术人员可对实现定位的时间进行合理估计。此外,可按照本发明的方法,例如用光电探测器,通过使可检测部分(例如发光缀合物)成像,跟踪随时间变化的定位的状态。所用的“光电探测器”应具有足够高的灵敏度使得能够在合理的时间内使哺乳动物内的微光成像,并利用来自该仪器的信号绘制图像。
在其中可能使用极明亮的发光部分和/或检测定位于待成像的受试者或动物表面附近的发光融合蛋白的情况下,可使用一副“夜视”镜或标准高灵敏度摄影机,例如SiliconIntensified Tube (SIT)摄影机(例如来自Hammamatsu Photonic Systems,Bridgewater, N.J.)。然而,更通常需要更灵敏的光检测方法。
在极低的光水平下,每单位面积的光子通量变得如此低,使得待成像的景物不再显得连续。相反地,使得它由在时间和空间两者上彼此不同的光子所代表。在临检器上观察时,这类图像似乎是光闪烁点,每个代表了单个检出的光子。通过累计数字图像处理器中随时间推移的这些检出光子,可获取并绘制图像。与其中给每个图像点指派强度值的常规摄影机形成对比,在光子计数成像中信号的振幅没有意义。目的只是检测信号(光子)的存在并圣信号随时间推移相对于其位置的出现计数。
下述至少两种类型的光电探测器可检测各个光子并产生可通过图像处理器分析的信号。与放大光子信号不同,减噪光检测器(Reduced-Noise Photodetection device)通过降低光子检测器的本底噪声达到灵敏度。噪声主要通过使检测器阵列冷却来降低。器件包括称为“薄背式(backthinned)”冷却CCD摄影机的电荷耦合器件(CCD)摄影机。在更灵敏的仪器中,冷却通过例如液氮(其使CCD阵列的温度达到约-120℃)实现。“薄背式”是指缩短待检测的光子跟踪、从而提高量子效率的光程长度的超薄背板。特别灵敏的薄背式低温CCD摄影机是可获自Photometrics, Ltd. (Tucson, Ariz.)的“TECH 512”,系列200摄影机。
“光子放大器”在它们击中检测屏前放大光子。这个类别包括带有增强器(例如微通道增强器(microchannel intensifier))的CCD摄影机。微通道增强器通常含有与摄影机的检测屏垂直和同延伸的金属通道阵列。微通道阵列置于待成像的样品、受试者或动物和摄影机之间。进入阵列的通道中的大多数光子在离开前接触通道侧面。施加在阵列上的电压导致从各光子碰撞中释放许多电子。来自这类碰撞的电子以“散弹”方式离开其起始通道,并由摄影机检测。
可通过连续引入增强性微通道阵列,使得在第一阶段产生的电子进而引起第二阶段的电子的放大信号,达到甚至更大的灵敏度。然而,灵敏度的增加在损失空间分辨率的情况下达到,所述空间分辨率随每个额外的放大阶段而降低。示例性基于微通道增强器的单光子检测器是可获自Hamamatsu的C2400系列。
图像处理器处理由光电探测器产生的信号,所述光电探测器对光子计数以绘制例如可在监视器上显示或在图像打印机上打印的图像。这类图像处理器通常作为包括上述灵敏的光子计数摄影机的***的一部分出售,因此可获自相同来源。图像处理器常常与个人计算机(例如IBM兼容性PC或Apple Macintosh (Apple Computer, Cupertino, Calif.))连接,所述计算机可作为购买的成像***的一部分包括或不包括在内。一旦图像为数字化文件的形式,它们便可通过各种图像处理程序(例如“ADOBE PHOTOSHOP”、Adobe Systems、Adobe Systems, Mt. View, Calif.)操作并打印。
在一个实施方案中,生物样品含有来自试验受试者的蛋白质分子。一种优选的生物样品是通过常规方法从受试者中分离的外周血白细胞样品。
本发明还包括用于检测生物样品中GITR或表达GITR的细胞的存在的试剂盒。例如,试剂盒可包含:能够检测生物样品中的癌细胞或肿瘤细胞的标记的化合物或作用剂(例如抗GITR scFv或单克隆抗体);用于测定样品中的GITR的量的工具;和用于将样品中的GITR的量与标准物进行比较的工具。在一些实施方案中,标准物是非癌细胞或其细胞提取物。可将化合物或作用剂包括在合适的容器中。试剂盒另可包括使用试剂盒检测样品中的癌症的使用说明。
双特异性抗体
双特异性抗体(bsAb)是包含2个可变结构域或scFv单元的抗体,使得所得抗体识别2种不同的抗原。本发明提供识别GITR和第二抗原的双特异性抗体。示例性第二抗原包括肿瘤相关抗原、细胞因子和细胞表面受体。在一些实施方案中,第二抗原可以是CAIX (碳酸酐酶IX或G250)、IL-10或CCR4。在一些实施方案中,第二抗原可以是细胞表面受体,其中胞表面受体为PD-1、PDL1、CCR4、IL21R、BTLA、HVEM或TIM3。本发明的双特异性抗体包括重链和轻链组合或本文公开的huGITR抗体的scFv。
双特异性抗体的构建
本发明的双特异性抗体可采用本领域已知方法构建。在一些实施方案中,双特异性抗体是单一多肽,其中2个scFv片段通过足够长度的接头多肽连接,以允许2个scFv单元之间的分子内缔合以形成抗体。在其它实施方案中,双特异性抗体是一个以上的通过共价键或非共价键连接的多肽。
在另一个实施方案中,双特异性抗体采用“孔中结(knob into hole)”方法构建(Ridgway等, Protein Eng 7:617-621 (1996))。在该方法中,使2个不同的可变结构域的Ig重链还原以选择性的断开重链配对同时保留重链-轻链配对。将识别2个不同的抗原的2个重链-轻链异二聚体混合以促进异质连接配对,这通过工程改造的CH3结构域的“孔中结”介导。
在另一个实施方案中,可通过来自两个或更多个不同抗体的重链-轻链二聚体交换产生其中第一重链-轻链二聚体识别GITR和第二重链-轻链二聚体识别第二抗原的杂合抗体,来构建双特异性抗体。重链-轻链二聚体的机制类似于人IgG4的形成,其还起双特异性分子的作用。IgG重链的二聚化受分子内力的驱动,例如使各重链的CH3结构域和二硫桥配对。CH3结构域中特定氨基酸的存在(R409)显示促进二聚体交换和IgG4分子的构建。抗体铰链区中的重链间二硫桥还进一步稳定重链配对。具体地说,在IgG4中,铰链区在氨基酸226-230处含有氨基酸序列Cys-Pro-Ser-Cys (与含有序列Cys-Pro-Pro-Cys的稳定的IgG1铰链区相比)。229位处丝氨酸的这种序列差异与IgG4在铰链区中形成新的链内二硫键的趋势有关(Van der Neut Kolfschoten, M.等, 2007, Science 317:1554-1557和Labrijn,A.F.等, 2011, Journal of immunol 187:3238-3246)。
因此,本发明的双特异性抗体可如下产生:通过将R409残基引入CH3结构域中并将Cys-Pro-Ser-Cys序列引入识别GITR或第二抗原的抗体的铰链区,使得重链-轻链二聚体交换产生具有识别GITR的重链-轻链的一个二聚体和识别第二抗原的第二重链-轻链二聚体的抗体分子,其中第二抗原是本文公开的任何抗原。还可改变已知的IgG4分子,使得如本文公开的一样,重链和轻链识别GITR或第二抗原。由于其中Fc区不同于其它IgG亚型的IgG4分子的固有特征所致(因为它与例如由某些白细胞表达的补体和Fc受体等免疫应答的效应器***相互作用不佳),构建本发明的双特异性抗体的该方法的应用可能是有益的。这种特殊性质使这些基于IgG4的双特异性抗体对治疗应用有吸引力,其中需要抗体结合靶标,并且在功能上改变与靶标有关的信号传导途径,但不引发效应子活性。
在一些实施方案中,将突变引入bsAb的恒定区,使得bsAb的依赖抗体的细胞介导的细胞毒性(ADCC)活性改变。例如,突变是CH2结构域中的LALA突变。一方面,bsAb在异二聚体bsAb的一个scFv单元上含有突变,这降低ADCC活性。另一方面,bsAb在异二聚体bsAb的两条链上含有突变,这完全消除ADCC活性。例如,引入bsAb的scFv单元的一个或两个的突变是CH2结构域的LALA突变。可使具有可变的ADCC活性的这些bsAb优化,使得bsAb显示针对表达被bsAb识别的一种抗原的细胞的最大选择性杀灭,然而显示针对被bsAb识别的第二抗原的最小杀灭。
本文公开的双特异性抗体可用于治疗疾病或医学病况,例如癌症。本发明的双特异性抗体可特别用于与Treg增加有关的疾病或医学病况。
治疗方法
本发明提供治疗有癌症风险(或易患癌症)的受试者或其它细胞增殖相关疾病或病症的预防性和治疗性方法两者。这类疾病或病症包括但不限于例如与GITR的异常表达有关的那些疾病或病症。例如,所述方法用于治疗、预防或减轻癌症症状。或者,所述方法用于治疗、预防或减轻其中GITR在T细胞应答中起负调节作用的癌症的症状。或者,所述方法用于治疗、预防或减轻例如乳腺癌、肺癌、卵巢癌、***癌、结肠癌、***、脑癌、皮肤癌、肝癌、胰腺癌或胃癌等实体瘤的症状。此外,本发明的方法用于治疗血癌例如白血病和淋巴瘤。或者,所述方法用于治疗、预防或减轻已转移的癌症的症状。
因此,一方面,本发明提供通过给予受试者本发明的单克隆抗体、scFv抗体或本发明的双特异性抗体预防、治疗或减轻受试者癌症症状或细胞增殖性疾病或病症的方法。例如,huGITR抗体可以治疗有效量给予。
有癌症或细胞增殖相关疾病或病症风险的受试者包括有癌症家族史的患者或暴露于已知或疑似致癌物的受试者。预防药的给药可发生在癌症显现之前,使得预防疾病或延迟其进展。
另一方面,通过使细胞与本发明的GITR抗体接触,抑制肿瘤细胞生长、降低Treg活性、提高Teff活性或增加NK细胞细胞毒性。细胞是表达GITR的任何细胞。例如细胞是T细胞或NK细胞。
本发明还包括提高或增强对抗原的免疫应答的方法。通过给予受试者本发明的单克隆抗体或scFv抗体,提高或增强免疫应答。例如通过提高抗原特异性T效应子功能,加强免疫应答。抗原是病毒(例如HIV)、细菌、寄生物或肿瘤抗原。免疫应答是天然免疫应答。所谓天然免疫应答意指是感染的结果的免疫应答。感染是慢性感染。提高或增强对抗原的免疫应答可通过本领域已知的多种方法测量。例如,免疫应答可通过测量下列的任一个来测量:T细胞活性、T细胞增殖、T细胞活化、效应细胞因子的产生和T细胞翻译概况。
或者,免疫应答是由于接种诱导的应答。因此,另一方面,本发明提供通过给予受试者本发明的单克隆抗体或scFv抗体和疫苗来提高疫苗效力的方法。抗体和疫苗序贯或同时给予。疫苗是肿瘤疫苗、细菌疫苗或病毒疫苗。
组合方法
本发明提供通过给予与GITR蛋白的相同表位或GITR蛋白的2个不同表位结合的2种抗体治疗患者的癌症的方法。或者,通过给予与GITR结合的第一抗体和与GITR以外的蛋白质结合的第二抗体来治疗癌症。例如,GITR以外的其它蛋白质可包括但不限于PD-1、PD-L1、CAIX、CCR4和IL-10。例如,GITR以外的其它蛋白质是肿瘤相关抗原。
在一些实施方案中,本发明提供huGITR抗体单独给予或与识别GITR以外的另一种蛋白质的其它抗体、与能够引起或提高免疫应答的细胞一起给予。例如,这些细胞可以是外周血单核细胞(PBMC)或PBMC中存在的任何细胞类型,例如细胞毒性T细胞、巨噬细胞和天然杀伤(NK)细胞。

b. 包含SEQ ID NO: 6的氨基酸序列的可变重链区和包含SEQ ID NO: 8的氨基酸序列的可变轻链区;
c. 包含SEQ ID NO: 10的氨基酸序列的可变重链区和包含SEQ ID NO: 12的氨基酸序列的可变轻链区;
d. 包含SEQ ID NO: 14的氨基酸序列的可变重链区和包含SEQ ID NO: 16的氨基酸序列的可变轻链区;
e. 包含SEQ ID NO: 18的氨基酸序列的可变重链区和包含SEQ ID NO: 20的氨基酸序列的可变轻链区;
f. 包含SEQ ID NO: 22的氨基酸序列的可变重链区和包含SEQ ID NO: 24的氨基酸序列的可变轻链区;

其中所述抗体或抗体结合片段结合人-糖皮质激素诱导的肿瘤坏死因子受体(GITR)。
3.权利要求1的抗体,其中所述抗体是单价的或二价的。
4.权利要求1的抗体,其中所述抗体是单链抗体。
