Abstract
本发明公开一种雄蚕蛾油微乳的制备方法,包括以下步骤:雄蚕蛾油的制备、混合乳化剂的制备、微乳化。本发明的优点在于:本发明制备的雄蚕蛾微乳外观透明,营养价值高,可长期储存。将雄蚕蛾油制备成微乳液,可以减缓雄蚕蛾油中不饱和脂肪酸的氧化,保护雄蚕蛾油的营养价值,延长其保存时间,提高其生物利用率,拓展其在保健品和食品等领域的应用。

技术领域
本发明涉及一种微乳,尤其是一种雄蚕蛾油微乳。
背景技术
雄蚕蛾油是从家蚕雄蛾体内提取的一类具有生理活性的物质,富含亚油酸、亚油酸等不饱和脂肪酸,具有降低血清胆固醇,预防动脉硬化,提高人体免疫力等作用。此外,雄蚕蛾油内还含有多种类固醇类激素和甾类激素,具有补肝益肾、壮阳涩精等保健功效。因此,雄蚕蛾油在人体保健品与疾病治疗药物开发方面具有广阔的应用前景。
由于雄蚕蛾油不饱和度极高,其在自然环境下很容易氧化酸败。
防止油脂氧化的措施常用的有微胶囊技术和微乳技术。
目前还未出现将微乳技术用于蚕蛾油的保存的相关研究。
发明内容
本发明的目的在于提供一种雄蚕蛾油微乳,微乳简化了微胶囊的繁琐生产工艺,粒径也大大低于微胶囊化的产品,营养成分高度分散,利于吸收,大大提高了营养物质的生物利用性。
本发明的技术方案为:一种雄蚕蛾油微乳的制备方法,包括以下步骤:(1)雄蚕蛾油的制备:取干燥蚕蛹粉经超临界CO2萃取,得雄蚕蛾原油,在雄蚕蛾原油中加入维生素E和柠檬酸,得雄蚕蛾油。超临界CO2萃取的条件为:CO2的流量为25kg/hm2,萃取温度为45℃,压力为25MPa,萃取时间为3h。所述的维生素E的重量为雄蚕蛾原油重量的0.1~0.5%,柠檬酸的重量为雄蚕蛾原油重量的0.01~0.02%。(2)混合乳化剂的制备:以重量计,取10~20份复合乳化剂和5~10份助乳化剂,混合均匀得到混合乳化剂。所述复合乳化剂是在大豆磷脂、吐温、卖泽、苄泽、卵磷脂中任选两种等量混合而成,优选为大豆磷脂和吐温60等量混合。所述助乳化剂为乙醇、丙二醇、正丁醇、乙二醇中的一种,优选为乙醇。所述复合乳化剂和助乳化剂的重量比例优选为3:2。(3)微乳化:以重量计,将步骤(2)得到的混合乳化剂与5~10份雄蚕蛾油混合,在温度30~40℃下,以250~350转/分搅拌速度边搅拌边加入蒸馏水40~80份直至微乳形成。所述混合乳化剂和雄蚕蛾油的重量比例优选为3:1。
本发明的优点在于:本发明制备的雄蚕蛾微乳外观透明,营养价值高,可长期储存。将雄蚕蛾油制备成微乳液,可以减缓雄蚕蛾油中不饱和脂肪酸的氧化,保护雄蚕蛾油的营养价值,延长其保存时间,提高其生物利用率,拓展其在保健品和食品等领域的应用。
具体实施方式
原料:雄蚕蛾粉100g,维生素E1.0g,柠檬酸0.06g,大豆磷脂1.0g,吐温60 1.0g,乙醇1g,蒸馏水12.2g
制备方法:
(1)称取100g雄蚕蛾粉装于FY121-50-01型超临界装置的萃取罐中,设定萃取罐的温度为45℃,压力为25MPa, CO2的流量为25 kg/hm2,萃取3 h,获得雄蚕蛾原油43.1g。在雄蚕蛾原油中加入1.0g维生素E和0.06g柠檬酸,混匀,得到雄蚕蛾油,测得雄蚕蛾油中不饱和脂肪酸含量为73.17%。
(2)将1.0g大豆磷脂,1.0g吐温60和1.5g乙醇置于50mL离心管中混合均匀后,加入雄蚕蛾油1.0g,混合均匀,在30℃条件下边搅拌边逐滴加蒸馏水10.2g,即得雄蚕蛾油微乳。得到的微乳液外观澄清透明,是一种O/W型微乳,其包封率为80%,粒径范围为15~80nm,25℃保存3个月未发生相分离,稳定性良好。
Claims (8)
1.一种雄蚕蛾油微乳,其特征在于:该产品重量组分为:复合乳化剂10~20份、助乳化剂5~10份、雄蚕蛾油5~10份、水40~80份。
2. 根据权利要求1所述的雄蚕蛾油微乳,其特征在于:该产品的制备方法,其步骤:(1)雄蚕蛾油的制备:取干燥蚕蛹粉经超临界CO2萃取,得雄蚕蛾原油,在雄蚕蛾原油中加入维生素E和柠檬酸,得雄蚕蛾油;(2)混合乳化剂的制备:取10~20份复合乳化剂和5~10份助乳化剂,混合均匀得到混合乳化剂;(3)微乳化:将步骤(2)得到的混合乳化剂与5~10份雄蚕蛾油混合,在温度30~40℃下,以250~350转/分搅拌速度边搅拌边加入蒸馏水直至微乳形成。
3.根据权利1或2所述的雄蚕蛾油微乳,其特征在于:所述的复合乳化剂是在大豆磷脂、吐温、卖泽、苄泽、卵磷脂中任选两种等量混合而成,优选为大豆磷脂和吐温60等量混合。
4.根据权利1或2所述的雄蚕蛾油微乳,其特征在于:所述的助乳化剂为乙醇、丙二醇、正丁醇、乙二醇中的一种,优选为乙醇。
5.根据权利2所述的雄蚕蛾油微乳的制备方法,其特征在于:所述的超临界CO2萃取的条件为:CO2的流量为25kg/hm2,萃取温度为45℃,萃取时间为3h。
6.根据权利2所述的雄蚕蛾油微乳的制备方法,其特征在于:所述的维生素E的重量为雄蚕蛾原油重量的0.1~0.5%,柠檬酸的重量为雄蚕蛾原油重量的0.01~0.02%。
7.根据权利1或2所述的雄蚕蛾油微乳及其制备方法,其特征在于:所述复合乳化剂和助乳化剂的质量比例优选为3:2。
8.根据权利1或2所述的雄蚕蛾油微乳及其制备方法,其特征在于:所述混合乳化剂和雄蚕蛾油的重量比例优选为3:1。
