Abstract
本发明公开了一种复合功能性雄蚕蛾粉及其应用,属于食品医药加工技术领域,所述复合功能性雄蚕蛾粉按照如下方法制备得到:取全只雄蚕蛾烘干,粉碎,收集雄蚕蛾膏;向收集的雄蚕蛾膏中加入生物肽或地龙蛋白、乳化剂、葡萄糖浆及水进行乳化均质,得雄蚕蛾悬浊液;将乳化均质后的悬浊液进行干燥,干燥后所得的固体进行粉碎,全部过筛,收集。本发明选用生物肽或地龙蛋白为包埋剂,制备的复合功能性雄蚕蛾粉能够有效保留雄蚕蛾成药效力,有效保护了雄蚕蛾的营养物质及活性成分不流失,同时可以有效控制雄蚕蛾粉的表面出油及出水率,可以应用于食品或医药领域。
Description
技术领域
本发明涉及一种复合功能性雄蚕蛾粉及其应用,属于食品医药加工技术领域。
背景技术
雄蚕蛾,又名“原蚕蛾”,系蚕蛾科昆虫家蚕蛾的去翅、足和鳞毛的雄性成虫,具有较高的药用价值。
明朝李时珍《本草纲目》记载:雄原蚕蛾益精气,强***,交接不倦,亦止精。壮阳事,止泄精、尿血、暖水脏,治暴风、金疮、冻疮、汤火疮、灭瘢痕。蚕蛾性淫,出茧即媾,至于枯槁而已,故强阴益精用之。中医认为雄蚕蛾味咸、性温,入肝、肾经,有补肝益肾、壮阳涩精的作用;内服可治阳痿、遗精、白浊、血淋等症,外敷可治刀斧及金创之伤。此外,雄蚕蛾体内还含有大量雄性激素、保幼激素、脑激素和蜕皮激素等生物活性物质,是集降糖,消除并发症、养生、保健于一体的“药食同源”佳品。
雄蚕蛾体内含的脂肪主要为不饱和脂肪酸,其中人体必需的亚麻酸占不饱和脂肪酸的35.9%;氨基酸含有18种,含人体必需氨基酸8种,如异亮氨酸、亮氨酸、赖氨酸、丝氨酸等;丰富的天然维生素,如硫胺素、核黄素、视黄醇、维生素A、维生素E、维生素D、水溶性维生素B等;激素类成分,如脑激素、蜕皮激素和保幼激素、***、孕酮、***、促卵泡释放激素、垂体泌乳等;丰富的矿物质及微量元素,如钾、钠、镁、铁、锌等。
雄蚕蛾作为蚕种培育工序产生的多余副产品并未得到合理的开发利用,通常仅是简单用作饲料或肥料,甚至丢弃,没能发挥应有的经济效益,造成资源浪费,仅有少量入药。目前国内关于雄蚕蛾粉制备的专利尚未出现。涉及雄蚕蛾的文献主要是关于雄蚕蛾的化学、营养成分,药理作用,功能作用以及开发利用的研究。如董立科等[中国食物与营养,2015,(03):72-73.]研究了雄蚕蛾功能作用及开发利用,结果表明,雄蚕蛾有补肝益肾、壮阳涩精、温经通络,强身健体等功效。此外,新鲜雄蚕蛾自身油脂含量、水分含量较高,粉碎后无法直接应用,而高温烘干会致使雄蚕蛾本身营养成分遭到一定程度破坏,同时对控制表面油含量并无明显效果,低温烘干无法达到原料水分含量要求。因此需要寻求一种在保证营养成分的同时,又能有效控制表面油及水分渗出的雄蚕蛾粉的制备方法。
发明内容
为克服现有技术的不足,本发明的目的在于提供一种复合功能性雄蚕蛾粉及其应用。
为实现上述目的,本发明采用如下技术方案:
一种复合功能性雄蚕蛾粉,由如下方法制备得到:
(1)取全只雄蚕蛾烘干,粉碎,收集雄蚕蛾膏;
(2)向收集的雄蚕蛾膏中加入生物肽或地龙蛋白、乳化剂、葡萄糖浆及水进行乳化均质,得雄蚕蛾悬浊液;
(3)将乳化均质后的悬浊液进行干燥,干燥后所得的固形物进行粉碎,全部过筛,收集,得到复合功能性雄蚕蛾粉。
优选的,所述生物肽为牛鞭肽、羊睾丸肽、牡蛎肽、海洋鱼低聚肽、羊骨髓肽、羊心肽、羊肝肽、羊脾肽、猪血肽、海参肽、牛脑肽、牛肝肽、牛脾肽中的至少一种。
优选的,所述乳化剂为单、双硬脂酸甘油酯。
优选的,所述葡萄糖浆购自上海好成食品发展有限公司,购买执行标准GB/T20885。
优选的,所述生物肽或地龙蛋白的加量为40-90wt%,所述乳化剂的加量为0.5-1.5wt%,所述雄蚕蛾粉的加量为8.5-59.5wt%,该三种物质混合形成固形物。
优选的,所述葡萄糖浆的加量占固形物、葡萄糖浆、水的总量的1-3wt%,所述水的加量占固形物、葡萄糖浆、水的总量的57-79wt%。
优选的,步骤(1)中,烘干温度为50-60℃,烘干时间为6-8小时,粉碎条件为3500r/min,粉碎时间20-60s。
优选的,步骤(2)中,乳化条件为2500-2800r/min,乳化时间为15-30min,乳化细度为200-300目,均质条件为16000-21000r/min,均质时间为10-15min。
优选的,步骤(3)中,干燥条件为于45-65℃条件下进行低温加热干燥或冷冻干燥,粉碎粒度为18目。
将如前所述的复合功能性雄蚕蛾粉用于制药或食品领域中。
相对于现有技术,本发明的有益效果在于:
全只鲜雄蚕蛾粉碎有利于有效保留雄蚕蛾成药效力,而通过功能性肽,乳化剂,葡萄糖浆对粉状雄蚕蛾进行包埋,有效保护了雄蚕蛾的营养物质及活性成分不流失,同时可以有效控制雄蚕蛾粉的表面出油及出水率。

具体实施方式
现结合实施例对本发明进一步详细的说明。应当理解,本实施例仅用于说明本发明,而不用于限定本发明的保护范围。
实施例1
(1)取全只鲜雄蚕蛾55℃烘干8小时,放入粉碎机,设定转速35000r/min,粉碎40s,收集雄蚕蛾膏。
(2)取膏状雄蚕蛾59.5g放入烧杯中,加入羊睾丸肽、牛鞭肽、牡蛎肽共40g(其中羊睾丸肽、牛鞭肽及牡蛎肽的重量的添加比例为1:1:1),单、双硬脂酸甘油酯0.5g均匀混合后,加入纯净水147.5g,葡萄糖浆2.5g,搅拌均匀后,倒入胶体磨中,转速2800r/min,乳化时间30min,乳化细度200目。所得混合液过均质机,设定转速15000r/min,均质10min,得雄蚕蛾悬浊液;
其中葡萄糖浆购自上海好成食品发展有限公司,购买执行标准GB/T 20885。
(3)均质后的雄蚕蛾悬浊液经50℃低温干燥,得到干燥块状雄蚕蛾,放入粉碎机粉碎,过18目筛,得到复合功能性雄蚕蛾粉Ι。
实施例2
(1)取全只鲜雄蚕蛾60℃烘干7小时,放入粉碎机,设定转速35000r/min,粉碎30s,收集雄蚕蛾膏。
(2)取膏状雄蚕蛾8.5g放入烧杯中,加入羊睾丸肽、牛鞭肽、牡蛎肽共90g(其中羊睾丸肽、牛鞭肽及牡蛎肽的重量的添加比例为1:1:1),单、双硬脂酸甘油酯1.5g均匀混合后,加入纯净水385g,葡萄糖浆15g,搅拌均匀后,倒入胶体磨中,设定转速2800r/min,乳化时间35min,乳化细度250目,所得混合液过均质机,设定转速17000r/min,均质8min,得雄蚕蛾悬浊液;
其中,葡萄糖浆购自上海好成食品发展有限公司,购买执行标准GB/T 20885。
(3)均质后的雄蚕蛾混合液经55℃低温干燥,得到干燥块状雄蚕蛾,放入粉碎机粉碎,过18目筛,得到复合功能性雄蚕蛾粉Ⅱ。
实施例3
(1)取全只鲜雄蚕蛾低温65℃烘干6小时,放入粉碎机,35000r/min,粉碎20s,所得膏状雄蚕蛾全部收集。
(2)取膏状雄蚕蛾40g放入烧杯中,加入羊睾丸肽、牛鞭肽、牡蛎肽共59.25g(其中羊睾丸肽、牛鞭肽及牡蛎肽的重量的添加比例为1:1:1),单、双硬脂酸甘油酯0.75g,加入纯净水226.7g,葡萄糖浆6.6g,搅拌均匀后,倒入胶体磨中,设定转速2800r/min,乳化时间40min,乳化细度300目,所得混合液过均质机,设定转速20000r/min,均质6min,得雄蚕蛾悬浊液;
其中,葡萄糖浆购自上海好成食品发展有限公司,购买执行标准GB/T 20885。
(3)均质后雄蚕蛾混合液经60℃低温干燥,得到干燥块状雄蚕蛾,放入粉碎机粉碎,过18目筛,得到复合功能性雄蚕蛾粉Ⅲ。
实施例4感官评价试验
将实施例1-3中制备的复合功能性雄蚕蛾粉Ⅰ、Ⅱ、Ⅲ分别进行压片试验。施加量分别为片剂重量的70%、80%、90%,同时以未包埋的雄蚕蛾粉为对照样。将制得的压片片剂进行感官质量评价,结果如下表所示:
实施例5出油及出水实验
1、表面油测定实验
(1)试验试剂
石油醚,待测样品,该待测样品为根据实施例3的制备方法得到的雄蚕蛾粉。对照组1为采用酪蛋白酸钠为包埋剂其余与实施例3相同的方法制备得到的雄蚕蛾粉,对照组2为采用大豆分离蛋白为包埋剂其余与实施例3相同的方法制备得到的雄蚕蛾粉,对照组3为采用乳清蛋白和大豆分离蛋白的混合物为包埋剂其余与实施例3相同的方法制备得到的雄蚕蛾粉。
(2)萃取方式
采用数次浸泡式萃取法(即将萃取溶剂分若干次萃取样品的方法),具体做法为:将准确称量的待测样品放在已知重量的滤纸上(滤纸已折叠好放在φ50mm的玻璃漏斗上),然后用一定体积的溶剂(溶剂量以能浸没样品为妥),浸泡洗涤玻璃漏斗中滤纸上的样品,待溶剂全部从漏斗中流出后,紧接着加入与第一次等量的溶剂浸泡洗涤样品,重复操作数次后,滤液收集在一起,滤液脱溶,准确称量,萃取出的油量W(g)除以样品重量G(g),乘以100%即为样品表面油含量。
(3)试验条件
30-60℃的石油醚,数次浸泡式萃取,料溶比=1:20,萃取温度25℃,称取待测样品量2.0g。
(4)实验结果
测试试验结果如表1所示,根据表1绘制出如图1所示的萃取油量曲线图。

(5)萃取油量结果分析:由图1可知,随着包埋剂含量的增加,雄蚕蛾粉表面萃取油量明显降低,说明此方法对雄蚕蛾粉表面油方面起到良好控制作用。通过测量两种雄蚕蛾粉表面萃取的油量比较可知,当包埋剂含量为30%时,多种肽包埋雄蚕蛾粉表面油含量为0.0375g,酪蛋白酸钠包埋雄蚕蛾表面油含量为0.0405g;大豆分离蛋白包埋雄蚕蛾表面含油量为0.0412g,复合包埋剂包埋雄蚕蛾表面含油量为0.0435g,当包埋剂含量为95%时,多肽包埋雄蚕蛾粉表面油含量为0.019g,酪蛋白酸钠包埋雄蚕蛾表面油含量为0.0236g,大豆分离蛋白表面油含量为0.0412g,复合包埋剂包埋雄蚕蛾表面油含量为0.0435g,可见采用本发明的多种生物肽类作为包埋剂对控制雄蚕蛾粉表面出油量起到显著的作用。其中,包埋剂含量达95%时,虽然雄蚕蛾粉表面油量很少,但雄蚕蛾含量较少,因此,选择90%为包埋剂添加上限;当包埋剂含量为30%时,表面油含量较高,因此,选择40%为包埋剂添加下限。
2、表面水测定实验
(1)实验原理
食品中的水分在达到40kPa~53kPa压力后加热至60℃±5℃时,可以采用减压烘干方法去除试样中的水分,再通过烘干前后的称量数值计算出水分的含量。
(2)仪器和设备
扁形铝制或玻璃制称量瓶,真空干燥箱,干燥器,天平,精确度为0.1mg。
(3)试样准备
粉末或结晶试样直接取样,按照实施例3的制备方法制得,较大硬块经研钵粉碎,混匀备用。
(4)测定:取已恒重的称量瓶称取2-10g(精确至0.0001g)试样,放入真空干燥箱内,将真空干燥箱连接真空泵,抽出真空干燥箱内空气,所需压力一般为40kPa~53kPa,并同时加热至所需温度60℃±5℃。关闭真空泵上的活塞,停止抽气,使真空干燥箱内保持一定的温度和压力,经4h后,打开活塞,使空气经干燥装置缓缓通入真空干燥箱内,待压力恢复正常后再打开。取出称量瓶,放入干燥器中0.5h后称量,并重复以上操作至前后两次质量差不超过2mg,即为恒重。
(5)分析结果的表述
试样中的水分含量,按式(1)进行计算:

水分含量测定结果分析:实验结果如表1所示,根据表1绘制曲线图,如图2所示,由图2可知,随着包埋剂含量的增加,雄蚕蛾粉水分渗出含量明显降低,说明此方法对控制雄蚕蛾粉内部水分渗出方面起到作用。通过测量雄蚕蛾粉水分含量比较可知,当包埋剂含量为30%时,多种肽包埋雄蚕蛾粉水分含量为5.78%,酪蛋白酸钠包埋雄蚕蛾水分含量为0.677g;大豆分离蛋白包埋雄蚕蛾水分含量为0.665g,复合包埋剂包埋雄蚕蛾水分含量为0.683g;当包埋剂含量为95%时,多肽包埋雄蚕蛾粉水分含量为0.421g,酪蛋白酸钠包埋雄蚕蛾水分含量为0.495g,大豆分离蛋白水分含量为0.531g,复合包埋剂包埋雄蚕蛾水分含量为0.542g。
由此可见多种肽包埋效果最好,其中,包埋剂含量达95%时,虽然雄蚕蛾粉水分渗出很少,但雄蚕蛾含量较少,因此,选择90%为包埋剂添加上限;当包埋剂含量为30%时,水分渗出较高,因此,选择40%为包埋剂添加下限。
由实验1及实验2可知,采用新型包埋剂对雄蚕蛾粉进行包埋,制得雄蚕蛾粉表面油含量及雄蚕蛾内部水分渗出现象得到了较好的控制,制得复合功能雄蚕蛾粉可作为食品原料被广泛应用于生产食品中。
实施例6复合雄蚕蛾粉功能性实验
本发明经两组临床验证,其中一组为治疗组,使用本发明按照实施例3制得的雄蚕蛾粉加工成片剂,每天食用1次,服用2周,另一组对照组服用补肾壮阳片,每天食用1次,服用2周,每组选择130例,其中男60例,女70例,最大年龄70,最小年龄30岁,临床表现为腰膝酸软,身体乏力,精神不佳,肾虚,免疫力低下易感冒,体质虚弱,表2为服用2周后对照数据:

由表2可以看出,根据本发明的复合功能雄蚕蛾粉制备的片剂在临床上的应用比普通补肾壮阳类药品有明显的疗效,尤其是在恢复体力方面尤为突出,同时在治疗肾虚、腰膝酸软,提高免疫力方面也具有显著疗效,改善体质方面也具有一定作用。
本发明的实施例内容揭露如上,然而本实施例并非用以限定本发明实施的范围,依据本发明的权利要求书及说明书内容所作的简单的等效变化与修饰,仍属于本发明技术方案的范围内。
Claims (10)
1.一种复合功能性雄蚕蛾粉,其特征在于,由如下方法制备得到:
(1)取全只雄蚕蛾烘干,粉碎,收集雄蚕蛾膏;
(2)向收集的雄蚕蛾膏中加入生物肽或地龙蛋白、乳化剂、葡萄糖浆及水进行乳化均质,得雄蚕蛾悬浊液;
(3)将乳化均质后的悬浊液进行干燥,干燥后所得的固体进行粉碎,全部过筛,收集,得到复合功能性雄蚕蛾粉。
2.如权利要求1所述的复合功能性雄蚕蛾粉,其特征在于,所述生物肽为牛鞭肽、羊睾丸肽、牡蛎肽、海洋鱼低聚肽、羊骨髓肽、羊心肽、羊肝肽、羊脾肽、猪血肽、海参肽、牛脑肽、牛肝肽或牛脾肽中的至少一种。
3.如权利要求1所述的复合功能性雄蚕蛾粉,其特征在于,所述乳化剂为单、双硬脂酸甘油酯。
4.如权利要求1所述的复合功能性雄蚕蛾粉,其特征在于,所述葡萄糖浆购自上海好成食品发展有限公司,购买执行标准GB/T 20885。
5.如权利要求1所述的复合功能性雄蚕蛾粉,其特征在于,所述生物肽或地龙蛋白的加量为40-90wt%,所述乳化剂的加量0.5-1.5wt%,所述雄蚕蛾粉的加量为8.5-59.5wt%,该三种物质混合形成固形物。
6.如权利要求5所述的复合功能性雄蚕蛾粉,其特征在于,所述葡萄糖浆的加量占固形物、葡萄糖浆及水的总量的1-3wt%,所述水的加量占固形物、葡萄糖浆、水的总量的57-79%wt%。
7.如权利要求1所述的复合功能性雄蚕蛾粉,其特征在于,步骤(1)中,烘干温度为50-60℃,烘干时间为6-8小时,粉碎条件为3500r/min,粉碎时间20-60s。
8.如权利要求1所述的复合功能性雄蚕蛾粉,其特征在于,步骤(2)中,乳化条件为2500-2800r/min,乳化时间为15-30min,乳化细度为200-300目,均质条件为16000-21000r/min,均质时间为10-15min。
9.如权利要求1所述的复合功能性雄蚕蛾粉,其特征在于,步骤(3)中,干燥条件为于45-65℃条件下进行低温加热干燥或冷冻干燥,粉碎粒度为18目。
10.将权利要求1-9任一项所述的复合功能性雄蚕蛾粉用于制药或食品领域中。
