具体实施方式
以下实施例用于进一步说明本发明,但不限制本发明的范围。若未特别指明,所用的是本领域技术人员所熟知的常规技术方法。
实施例1 水稻雄性不育突变基因及其功能型育性基因的定位克隆和功能验证
用钴60-γ射线诱变水稻粳稻品种02428,获得1个单座位隐性孢子体雄性不育突变体,命名为msl12(图2)。将msl12与籼稻品种黄华占杂交获得F2定位群体。以表1所列的基于PCR的***/缺失多态性分子标记,利用约10500株F2个体将msl12定位在第6染色体的一个43 kb的区间(图3A)。该区间存在2个预测基因(基因号 Os06g006700和Os06g006800)。DNA测序分析表明,与原亲本的基因组序列相比,msl12的Os06g006800没有变异,而在Os06g006700缺失了一段包括1个外显子和2个内含子的1479 bp片段(图3B,SEQ ID No.1)。本发明将该野生型(功能型)基因命名为OsABCG15(SEQ ID No.2,SEQ ID No.3),它编码一个454氨基酸的ABC运转子(ATP-binding cassette transporter)蛋白(SEQ ID No.4)。其功能可能是从花药绒毡层运输脂肪酸到小孢子,合成花粉壁的主要成分花粉素。用特异引物5’-CATGTGTGGCCAACCAAAGA-3 和5’-GTG GTCTTGCCACTGCC AGA-3’对不同发育时期花药进行定量RT-PCR分析表明,OsABCG15主要在四分体期至单核小孢子早期表达(图3C)。

为了验证OsABCG15的功能,用特异引物5’-TTCTTGGTACCTG ATTAAAAGGAGAG-3’(下划线为Kpn I切点)和5’-GCCAAGCTTCGCTTATGCT TACAAGCTGA-3’ (下划线为Hind III切点)以PCR扩增OsABCG15 全长cDNA,与玉米泛素基因启动子Pubi连接后克隆到植物转化双元载体pCAMBIA1305.1改造的载体。把此载体以农杆菌转化方法转化由MSL12/msl12 杂合体产生的种子(有1/4比例是msl12/msl12纯合体)诱导的愈伤组织。从获得的T0转化体用分子标记625051(表1)筛选出2个msl12/msl12纯合体,它们的花药发育正常并产生可育花粉(图3D)。此结果证明OsABCG15是具有育性功能的目标基因,突变基因osabcg15(SEQ ID No.1)的功能缺失导致小孢子早期不能正常发育而退化,产生无花粉型败育。
实施例2 从水稻分离化学诱导型基因启动子
本发明用0.05%丙环唑和0.05%腈菌唑混合水溶液施加到水稻叶片,24h后取处理和对照的减数***期至小孢子期的花药,提取RNA。利用水稻全基因组表达芯片(OsAffx)杂交(由博奥生物有限公司完成)筛查,发现1个基因探针(OsAffx.13615.1.S1_at)代表的基因在非处理样品的表达水平很低,而在处理样品的表达水平大大高于非处理对照。进一步用定量RT-PCR验证,证实其本底表达较低,而丙环唑或腈菌唑可提高其表达水平,这2种诱导剂共用的表达水平更高(图4)。
实施例3 水稻雄性育性可控系的构建
以特异引物5’-CTTGAGGCCGGCCTAAATTATTATTTTTCCAATATTTA-3’ (下划线为FseI切点)和TCAGGTACCGATCGGTGATCCCTCCTCAA-3’ (下划线为Kpn I切点)从水稻基因组PCR扩增所述的启动子序列;用5’-GATCGGTACCTGATTAAAAGGAGAGA AG-3’ (下划线为Kpn I切点)和5’-GGAGGATCCAGACCTCGCGCA-3’ (下划线为BamH I切点) PCR扩增野生型 OsABCG15 近全长cDNA编码序列;以5’-CTCT GGATCCTCCTCGCCATGGC-3’ (下划线为BamH I切点)和5’-CCTACTCGAG CTCCCTGCAGGTGAGGA-3’(下划线为Xho I切点) PCR扩增OsABCG15 的小部分cDNA编码序列和3’终止子序列。将这3个片段用所示的限制性内切酶切出粘性末端,连接成重组基因表达盒结构(图5),其DNA序列如SEQ ID No.6所示。将此表达盒序列克隆到基于可转化人工染色体载体(Lin等 2003,PNAS, 100, 5962-5967)的改造载体。将含有此重组基因表达盒的载体以农杆菌介导方法(Hiei 等, 1994 Plant J. 6, 271-282) 转化导入所述的雄性不育突变系,获得转化体。将转化体与1个籼稻品系TB杂交,并以TB为轮回亲本回交3代,选育出偏籼型的雄性育性可控系HNLS-1。每次回交时以引物组5’-CCTGTTCCATCGAGGAGT-3’ /5’-CTCAACAATGGCCATCTCCA-3’以及5’-CTCGGCCTCCTCTGGTGGAAGT-3’/ 5’-CGGCTGCCGCAGTTGTAGGT-3’筛选含有转基因和内源突变基因osabcg15 的个体作为回交的母本。
实施例4 水稻雄性育性可控系的繁殖
在雄性育性可控系的花药减数***期至单核小孢子期,以0.05%丙环唑或0.05%腈菌唑或0.05%丙环唑+0.05%腈菌唑混合水溶液喷施到叶片,3天后重施一次。结果表明,单独使用丙环唑或腈菌唑的自交结实率约为60%,将2种诱导剂混用的自交结实率约为86%(图6)。
实施例5 用水稻雄性育性可控系进行杂交制种
将本发明育成的水稻雄性育性可控系与1个籼稻品系HNLR-1按12:2的比例种植,在抽穗杨花时辅以人工赶花粉以提高杂交率,获得F1杂交种子(结实率约55%)。F1杂种植株的花粉正常可育,自交结实率达到约90%(图7)。
