含有生物活性亲脂化合物的初级组合物
本发明涉及含有生物活性亲脂化合物的初级组合物以及含有所述初级组合物的口服组合物及其制备方法。
我们已经知道在市场上购得的含有生物活性亲脂化合物即番茄红素的组合物，番茄红素是一种天然产物，已知它具有多种功能，特别是具有抗氧剂的功能。番茄红素存在于各种天然产物中，特别是存在于番茄、甜瓜、番石榴和柚子当中。一般可以从市场上买到而且含有番茄红素的组合物是一种含油树脂。这种含油树脂的问题是，它所含的番茄红素的生物可利用性不够。本发明的目的就是提供一种含有番茄红素，而且比目前市场上购买的产品具有更好生物可利用性的产物。
专利EP-278284涉及到含有合成类胡萝卜素的粉末状组合物。这种组合物的问题是，它不能在食品领域中使用，另外它预计用于着色的目的。
本发明涉及的初级组合物含有生物活性的亲脂组合物和乳清蛋白。所谓“生物活性化合物”，指的是对人类或动物的新陈代谢具有有益效果的化合物。本发明的目的是给消费者准备一种由天然产物得到的组合物。正如在下面所指出的，按照本发明还要寻求对此生物活性亲脂化合物的保护。
此生物活性亲脂化合物由植物、微生物、酵母或动物源的产物得到、提取、富集或者提纯。所谓“得到”，指的是此生物活性产物直接从市场上购买；所谓“提取”，指的是提取生物活性的主要成分；所谓“富集”，指的是尽可能分离掉不具有生物活性的化合物；而所谓“提纯”，指的是单独回收生物活性化合物。
当一种生物活性化合物来源于植物时，此植物选自西红柿(整个的西红柿、西红柿提取物、西红柿果肉、西红柿酱、带籽不带籽的西红柿粉)、大豆、绿茶、青咖啡豆、香料(生姜或者其它香料)、葡萄籽、可可和谷物.微生物是各种类型的产生生物活性素脂化合物的细菌，比如，作为微生物可以设想是有益生物(probiotique)的微生物，特别是乳酸菌。酵母可以是各种产生生物活性亲脂化合物的酵母。比如酵母菌。动物源的产物选自肝脏提取物和奶组分。所谓“奶组分”，指的是奶的各个部分。

类胡萝卜素特别存在于西红柿、胡萝卜、黄桃、杏和柑橘中。番茄红素是在本发明中特别优选的一种类胡萝卜素。多元酚特别存在于绿茶、咖啡、可可、红葡萄酒中。亲脂性维生素特别存在于许多蔬菜中。类黄酮和并黄酮特别存在于大豆、茶叶、洋葱头和葡萄酒中。类姜黄素特别存在于姜中。神经酰胺是特别存在于酵母衍生物和动物源衍生物中的糖酯。前花青素是特别存在于葡萄籽中的类黄酮。类萜存在于香料中。甾醇、植物甾醇和甾醇酯特别存在于植物油、谷物、胡桃和蔬菜中。生育三烯酚特别存在于水稻、大麦、小麦、棕榈油、黑麦和燕麦中。角鲨烯特别存在于鱼肝、橄榄油、麦芽油、稻糠油中。最后，视黄素特别存在于肝、蛋黄和奶类产品中。

本发明组合物含有直至50％的生物活性亲脂化合物和直至90％的乳清蛋白。
在本发明初级组合物的一个优选实施方案中，该组合物含有西红柿油树脂、大豆提取物和乳清蛋白。在本叙述中的所谓“西红柿油树脂”，指的是所述植物的脂类提取物，包括类胡萝卜素，象番茄红素、三甘油酯、磷脂、生育酚和其它更低级的化合物。所谓“大豆提取物”，指的是含有大量异黄酮的大豆提取物。也可以设想其它含有类胡萝卜素的植物，特别是甜瓜、番石榴、柚子、杏、蔷薇果、胡萝卜、桃和柑橘。
本发明还涉及在食品、食品增补剂、化妆制剂或在药物制剂中的含有10～50％如上所述初级组合物的口服组合物。
这些可以通过口服途径食用的组合物能够增加生物活性亲脂化合物在人体中的生物可利用性、延缓皮肤的老化。作为用上述初级组合物补充的食品，可以举出酸奶、饮料、巧克力、冰激凌、谷物、巧克力粉、咖啡、烹调的食品，比如蛋黄酱、西红柿酱或色拉酱、婴儿营养品、肠饲营养物或动物饲料。在此情况下，粉末被溶于上述食品或饮料中，每天大约有0.001～50mg的生物活性亲脂化合物，比如番茄红素。优选每天剂量为大约5～20mg。
按照本发明还可以提供呈糖衣、药丸、胶囊、糖浆、凝胶、乳脂或片剂的食品增补剂，含有所述初级组合物为0.001～100％，此时可以直接溶解于水中食用或者用各种其它已知的手段食用。此增补剂还含有甜味剂、稳定剂、添加剂、香料和着色剂。
此口服组合物也可以是含有此初级组合物和本领域技术人员已知对皮肤有活性的化合物的化妆制剂。
此口服组合物也可以是含有此初级组合物和药物化合物如局部涂敷和经口服用的药物的药物制剂。
本发明还涉及含有上述初级组合物的化妆组合物。在此情况下，初级组合物的含量为10-10～10％。此化妆组合物优选含有10-8～5％的生物活性亲脂化合物。
可以通过局部途径使用的此组合物还含有可在化妆品使用的脂或油，比如在文献CTFA的《化妆成分手册》(Cosmetic IngredientsHandbook)，(华盛顿)出版中叙述的油脂。也可以加入其它的化妆活性成分。此组合物还含有结构剂和乳化剂。在此组合物中还可以加入赋形剂、着色剂、香精或遮光剂。

本发明还涉及上述初级组合物的制备方法，其中乳清蛋白与生物活性亲脂化合物相混合。
在本发明方法的第一个实施方案中，
-将乳清蛋白溶解于水；
-将生物活性亲脂化合物溶解于溶剂；
-将两种溶液混合；
-蒸发掉溶剂，以及

在两相混合以后，在比环境温度稍高的温度下，比如在大约30℃下，将混合物放置30～60分种，并在适当的真空下开始驱赶掉溶剂。所谓“适当的真空”，指的是200～300mbar的真空度。如果想得到粉末，可以借真空、或者借喷雾、或者借冷冻干燥来除去水分。所谓“真空”，指的是40～50mbar的真空度。要想得到凝胶，将乳液加热，或者按照本领域技术人员已知的其它技术进行操作，得到所述凝胶。
在本发明方法的第二种实施方式中，
-将生物活性亲脂化合物与溶剂混合；
-将得到的组合物与乳清蛋白粉末混合，以及
-蒸发溶剂，得到粉末状组合物。
使用的溶剂与上面所叙述的相同。
在第三种实施方式中，将或者呈油树脂形式、或者呈粉末形式或者呈各种其它干燥形式(比如将油树脂吸附在载体上)的生物活性亲脂化合物与乳清蛋白粉末(任选地含有大豆提取物)直接混合，得到本发明的初级组合物。
下面参考实施例进行叙述。
实施例1：制备粉末状的组合物
将13.3kg乳清蛋白分离物溶解于330L软化水中，在25～30℃下搅拌混合物6小时。另外，在438L丙酮中混合550g含有6％番茄红素的Lycored公司的油树脂并搅拌混合物。然后在30℃下混合这两种溶液60分钟。适度地加热最终混合物，在适当的压力下驱赶丙酮。最后，在40～50mbar的压力下部分驱赶出水分。得到200kg乳清蛋白分离物和油树脂的水溶液。
然后将此溶液进行喷雾干燥。
用此粉末进行试验，每天给试验者含有25mg油树脂中的番茄红素和其它类胡萝卜素以及12.5g乳清蛋白的粉末：给药的方式是将此粉末溶解于苹果汁中。相对于含有相同量番茄红素的番茄酱参照物，发现由本发明粉末的番茄红素的生物可利用性与由番茄酱得到的生物可利用性相等：此项研究进行了8周。应该注意到，番茄酱被本领域技术人员认为是具有最好番茄红素生物可利用性的产品。血浆中番茄红素的含量证实了这一点。
实施例2：制备糖衣
用乳化剂制备550g实施例1的油树脂在乙醇中的分散液。将此分散液与1100g乳清蛋白和1100g大豆提取物(含有40％的异黄酮)混合。除去溶剂得到粉末。
将如此得到的粉末与抗坏血酸以及其它添加剂，比如一种或几种甜味剂、增稠剂和制备糖衣可用的食品添加剂混合。然后将得到的混合物制成糖衣。
如此制备大约700mg糖衣，含有33mg番茄红素、70mg大豆提取物、70mg乳清蛋白、40mg抗坏血酸，其余的是甜味剂、增稠剂和食品添加剂，凑足700mg。
实施例3：化妆组合物
制备含有7％凡士林油、2％实施例1的粉末、3％单硬脂酸甘油酯、硬脂酸聚乙二醇酯、0.4％羧基乙烯基聚合物、0.7％硬脂醇、3％大豆蛋白、0.4％NaOH，防腐剂的洗面奶，用水补足到100％。
实施例4：化妆组合物
制备含有10％甘油、2％实施例1的粉末、1％椰子基两性二醋酸二钠，一种防腐剂的洗面凝胶，用水补足到100％。
实施例5：番茄红素稳定性研究
已经知道光线和氧能够引起番茄红素的降解。对单独使用番茄红素和按照本发明与乳清蛋白一起使用的番茄红素的稳定性进行水相分析。在第一天末，对于只有番茄红素的水相样品，只剩下40％的番茄红素，而对于与乳清蛋白一起使用的水相样品，实际上剩余90％。在第二天末，与乳清蛋白一起使用的水相样品，剩余60％，而单独使用番茄红素的水相样品，其番茄红素几乎完全降解。
因此，乳清蛋白对番茄红素有很好的保护效果。

Claims (37)

1.含有至少一种生物活性亲脂化合物和乳清蛋白的粉末状的初级组合物，该组合物通过包含以下步骤的方法获得：
-将生物活性亲脂化合物与溶剂混合；
-将得到的组合物与乳清蛋白粉末混合，以及
-蒸发溶剂，得到干粉末状组合物。
2.如权利要求1的初级组合物，其特征在于，此生物活性亲脂化合物获得、提取、富集或提纯自植物、微生物、酵母或动物源的产品。
3.如权利要求2中的初级组合物，其特征在于，此植物选自西红柿、大豆、绿茶、青咖啡豆、香料、葡萄籽、可可、姜和谷物。
4.如权利要求2中的初级组合物，其特征在于，此微生物是能够产生生物活性亲脂化合物的细菌。
5.如权利要求2中的初级组合物，其特征在于，此酵母是能够产生生物活性亲脂化合物的酵母。
6.如权利要求2中的初级组合物，其特征在于，此动物源的产品选自肝提取物和奶组分。
7.如权利要求1的初级组合物，其特征在于，此生物活性亲脂化合物选自单独的或混合的类胡萝卜素、多元酚、亲脂维生素、类黄酮、异黄酮、类姜黄素、神经酰胺、前花青素、类萜、甾醇、植物甾醇、甾醇酯、生育三烯酚、角鲨烯、类视黄素。
8.如权利要求1的初级组合物，其特征在于，此生物活性亲脂化合物是西红柿提取物、大豆提取物或西红柿提取物和大豆提取物的混合物。
9.如权利要求1的初级组合物，其特征在于，此组合物还含有维生素C和/或生育酚。
10.如权利要求1的初级组合物，其特征在于，此组合物还含有乳化剂、稳定剂和各种香料、着色剂。
11.如权利要求1的初级组合物，其特征在于，其生物活性亲脂化合物的含量多至50％，乳清蛋白含量多至90％。
12.如权利要求11的初级组合物，其特征在于，此组合物含有西红柿油树脂、大豆提取物和乳清蛋白。
13.包含至少一种生物活性亲脂化合物和乳清蛋白的液体状或凝胶状的初级组合物，该组合物通过包含以下步骤的方法获得：
-将乳清蛋白溶解于水；
-将生物活性亲脂化合物溶解于溶剂；
-将两种溶液混合；和
-蒸发掉溶剂，以形成分散液状组合物。
14.如权利要求13的初级组合物，其特征在于，此生物活性亲脂化合物获得、提取、富集或提纯自植物、微生物、酵母或动物源的产品。
15.如权利要求14的初级组合物，其特征在于，此植物选自西红柿、大豆、绿茶、青咖啡豆、香料、葡萄籽、可可、姜和谷物。
16.如权利要求14的初级组合物，其特征在于，此微生物是能够产生生物活性亲脂化合物的细菌。
17.如权利要求14的初级组合物，其特征在于，此酵母是能够产生生物活性亲脂化合物的酵母。
18.如权利要求14的初级组合物，其特征在于，此动物源的产品选自肝提取物和奶组分。
19.如权利要求13的初级组合物，其特征在于，此生物活性亲脂化合物选自单独的或混合的类胡萝卜素、多元酚、亲脂维生素、类黄酮、异黄酮、类姜黄素、神经酰胺、前花青素、类萜、甾醇、植物甾醇、甾醇酯、生育三烯酚、角鲨烯、类视黄素。
20.如权利要求13的初级组合物，其特征在于，此生物活性亲脂化合物是西红柿提取物、大豆提取物或西红柿提取物和大豆提取物的混合物。
21.如权利要求13的初级组合物，其特征在于，此组合物还含有维生素C和/或生育酚。
22.如权利要求13的初级组合物，其特征在于，此组合物还含有乳化剂、稳定剂和各种香料、着色剂。
23.如权利要求13的初级组合物，其特征在于，其生物活性亲脂化合物的含量多至50％，乳清蛋白含量多至90％。
24.如权利要求23的初级组合物，其特征在于，此组合物含有西红柿油树脂、大豆提取物和乳清蛋白。
25.在食品中、在食品增补剂中、在化妆制剂中或在药物制剂中含有如权利要求1～24中之一项的初级组合物的口服组合物。
26.如权利要求25的口服组合物，其特征在于，此食品选自酸奶、饮料、巧克力、冰激凌、谷物、巧克力粉、咖啡、动物饲料。
27.如权利要求25的口服组合物，其特征在于，此食品增补剂还包括甜味剂、稳定剂、香料和着色剂，并且呈糖衣剂、丸剂、胶囊、糖浆、凝胶或乳脂的形式。
28.如权利要求25的口服组合物，其特征在于，此化妆制剂还含有对皮肤有活性的化合物。
29.如权利要求25～28中之一项的口服组合物，其特征在于，此初级组合物的含量为0.001～100％。
30.如权利要求29的口服组合物，其特征在于，此初级组合物的含量为10～50％。
31.含有如权利要求1～24中之一项的初级组合物的化妆组合物。
32.如权利要求31的化妆组合物，其特征在于，此初级组合物的含量为10-10％～10％。
33.如权利要求25～30中之一项的口服组合物在制备用于防止皮肤组织老化方面的产品中的应用。
34.如权利要求31-32中之一项的化妆组合物在制备用于防止皮肤组织老化方面的产品中的应用。
35.如权利要求13的初级组合物的制备方法，其中
-将乳清蛋白溶解于水；
-将生物活性亲脂化合物溶解于溶剂；
-将这两种溶液混合；
-蒸发掉溶剂，以及
-得到分散液。
36.如权利要求1的初级组合物的制备方法，其中
-将生物活性亲脂化合物与溶剂混合；
-将得到的组合物与乳清蛋白粉末混合，以及
-蒸发掉溶剂，得到粉末状组合物。
37.如权利要求35或36中之一项的方法，其特征在于，此溶剂选自丙酮、乙醇和异丙醇。
