Die vorliegende Erfindung betrifft Ultramikroemulsionen aus spontan dispergierbaren Konzentraten mit schwer löslichen Carotinen, ihre Zusammensetzung, sowie ihre Verwendung als Mittel zur Herstellung von Präparaten mit verbesserter Bioverfügbarkeit für die Behandlung von Tumoren, Psoriasis und Ekzemen. The present invention relates to ultramicroemulsions from spontaneously dispersible concentrates with poorly soluble carotenes, their composition and their use as agents for the preparation of preparations with improved bioavailability for the treatment of tumors, psoriasis and eczema.
Ausgewählte, in Wasser schwer lösliche Carotine haben überraschenderweise eine sehr gute Wirkung gegen Tumore, Psoriasis und Ekzeme, insbesondere dann, wenn sie in spontan dispergierbare Konzentrate eingearbeitet worden sind, welche mit Wasser, Glucoselösung oder physiologischer Kochsalzlösung verdünnt thermodynamisch stabile Ultramikroemulsionen ergeben mit Mizellen, die einen hydrodynamischen Radius im untersten Nanometerbereich aufweisen. (Messungen ausgeführt mit dynamischer, d.h. quasielastischer Lichtstreuung und stark verbesserter Laserdetektion haben Grössenordnun-gen aufgezeigt von einheitlich 1,5-3 nm.) Eingesetzt wurden normale 8-wöchige weibliche BALB/c nAncr (H-2d)-Mäuse; LIEFERANT: Charles River Laboratories, Calco (Italien). Während 4 Wochen wurden täglich zweimal 250 jxl einer wässrigen Mikroemulsion der angegebenen MARIGENOL®-KONZENTRATE uv. (Schwanzvene) injiziert, bzw. die gleiche Menge physiologischen Puffers für die Kontrollen. Färbung mit May Grünwald-Giemsa. Normal 8-week female BALB / c nAncr (H-2d) mice were used; SUPPLIER: Charles River Laboratories, Calco (Italy). 250 jxl of an aqueous microemulsion of the indicated MARIGENOL® CONCENTRATES etc. (tail vein) or the same amount of physiological buffer for the controls were injected twice daily for 4 weeks. Coloring with May Grünwald-Giemsa.

ERGEBNIS: Es traten keine signifikanten Unterschiede auf zwischen den Testgruppen und den Kontrollen. Die MARIGENOL®-KONZENTRATE weisen keinerlei Toxizität auf gegenüber der im Versuch eingesetzten Leucocyten-Population. Alle Versuchstiere waren und blieben während des ganzen Versuches gesund. RESULT: There were no significant differences between the test groups and the controls. The MARIGENOL® CONCENTRATES are not toxic to the leucocyte population used in the experiment. All test animals were and remained healthy throughout the test.

1) Identifikation von CAROTIN-ESTERN 1) Identification of CAROTINE ESTERS
Kapillarzonen-Elektrophorese mit einem Gerät von Beckman Instruments, bzw. von BIORAD. Bedingungen: DL-a-Tocopherol 50 mM; Puffer pH = 9,5 Na-tetra-Borat Run 15 kV, Messung bei 195 nm Der Wirksubstanz-Peak erscheint nach ca. 4 Minuten. Auflösung bis 0,5 ppm. Capillary zone electrophoresis with a device from Beckman Instruments or from BIORAD. Conditions: DL-a-tocopherol 50 mM; Buffer pH = 9.5 Na-tetra-Borat Run 15 kV, measurement at 195 nm The active substance peak appears after approx. 4 minutes. Resolution up to 0.5 ppm.

Der charakteristische Peak für die Carotin-Ester erscheint nach ca. 8 Minuten; die Verzögerung gegenüber der reinen W.S.-Messung ist eine Folge der Ummantelung der inneren Phase der Konzentrate mit Tensiden. Sie gibt einen wichtigen Hinweis auf das Diffusionsverhalten der Ultramikroemulsion. 3) Nachweis der Membran-Penetration an der Tumorzelle The characteristic peak for the carotene esters appears after about 8 minutes; the delay compared to the pure W.S. measurement is due to the coating of the inner phase of the concentrates with surfactants. It gives an important indication of the diffusion behavior of the ultramicroemulsion. 3) Detection of membrane penetration on the tumor cell
Am Lichtmikroskop (wie auch am Elektronenmikroskop) lässt sich aufzeigen, dass wenige Stunden nach der Inkubation (Beispiel 3T3 Py6-Zellen der Maus; dünn ausgesät, mittlere Verdünnung der Wirkstoff-Konzentrate) sich ein Kranz von Vakuolen um den Zellkern herum ausbildet. On the light microscope (as well as on the electron microscope) it can be shown that a ring of vacuoles forms around the cell nucleus a few hours after the incubation (example 3T3 Py6 cells of the mouse; thinly sown, medium dilution of the active ingredient concentrates).
