Die neuen erfindungsgemässen Präparate sind antiandrogen wirksam; sie können demnach als Arzneimittel Verwendung finden, insbesondere zur Behandlung von Krankheiten, die mit einer erhöhten androgenen Aktivität in Verbindung stehen, wie z. B. Akne, Seborrhoe, Hirsutismus und Prostataadenom. Bevorzugte Präparate enthalten einen Wirkstoff der Formel I, worin der Rest

darstellt. Besonders bevorzugte antiandrogen wirksame Präparate enthalten eine Verbindung der Formel I, worin R eine Gruppe (b) darstellt, insbesondere eine solche mit X = Sauerstoff, d.h. ein Hydantoinderivat der Formel I.
Bevorzugte Hydantoinderivate sind: 3 -(3 -Trifluormethyl-4-fluor-phenyl)-5, 5-dimethyl-hydan- toin;
3-(3-Trifluormethyl4-chlor-phenyl)-5,5-dimethyl-hydantoin; 3-(3-Chlor-4-fluor-phenyl)-5,5-dimethyl-hydantoin;
3-(3,4-Dichlor-phenyl)-5,5-dimethyl-hydantoin;
3-(3,5-Dichlor-4-fluorphenyl)-5,5-dimethylhydantoin.
Bevorzugt ist ebenfalls 1 -(3-Trifluormethyl.4-chlor-phenyl)-5-imino-4,4-dime- thyl-2-imidazolidinon.
Die erfindungsgemässen Präparate können durch Verwendung von in der Pharmazie üblichen Herstellungsmethoden erhalten werden. Zu diesem Zweck wird der Wirkstoff der Formel I mit einem für die enterale oder parenterale Applikation geeigneten pharmazeutischen organischen oder anorganischen Trägermaterial, wie Gelatine, Milchzucker, Stärke, Gummi arabicum, Magnesiumstearat, Talcum, pflanzliche Öle, Polyalkylenglykole, Vaseline oder dergleichen vermischt. Die Präparate können in fester Form, z. B.
als Tabletten, Dragees, oder in flüssiger Form, z. B. als Lösungen, Suspensionen oder Emulsion, vorliegen. Sie können Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffer enthalten. Auch andere therapeutisch wirksame Stoffe können beigemischt werden.
Zweckmässige pharmazeutische Dosierungsformen enthalten 10 bis 500 mg, vorzugsweise etwa 100 mg einer Verbindung der Formel I.
Die Dosierung wird entsprechend den individuellen Erfordernissen gewählt. Zum Beispiel können diese Verbindungen in Dosierungen von etwa 0,1 mg/kg bis etwa 10 mg/kg pro Tag p. o., vorzugsweise 1 mg/kg pro Tag p. 0. verabreicht werden. Zweckmässigerweise wird diese Dosis in Anpassung an den Zustand des Patienten etwa 3 bis 8 Monate täglich verabreicht.
Die antiandrogene Wirkung der erfindungsgemässen Endprodukte kann durch folgenden Versuch nachgewiesen werden:
Einer Gruppe von 5 männlichen, sterilisierten Ratten werden täglich während 7 Tagen jeweils 10 mg des zu untersuchenden Präparates pro kg zusamrnen mit 0,5 mg Testosteronpropionat pro kg s.c. verabreicht. 3 Kontrollgruppen zu jeweils 5 Ratten erhaltenjeweils keine Behandlung bzw. nur das Präparat bzw. nur Testosteronpropionat.
Die Gewichtsabnahme der ventralen Prostata und der Samenblase gibt ein Mass für die antiandrogene Wirkung.
Die nachstehende Tabelle I gibt über einige Versuchsergebnisse Aufschluss.

Diese Additionsreaktion kann ohne Lösungsmittelzusatz, d. h. mittels einer Schmelze, oder auch durch Erwärmung in einem inerten, wasserfreien Lösungsmittel, z. B.
Tetrahydrofuran, Äther, Dioxan, Benzol oder Toluol, durchgeführt werden. Die Temperatur für die Umsetzung liegt vorzugsweise im Bereiche von etwa 0 bis 120 "C. Beim Arbeiten mit einer Schmelze ist darauf zu achten, dass höhere Temperaturen bzw. Iängere Reaktionszeiten zur Cyclisation unter Bildung des entsprechenden Hydantoins führt (vgl. nachstehend). Dieses Amid wird unter sauren Bedingungen und einem niederen Alkanol der Formel R3OH zum gewünschten Endprodukt alkoholysiert, z.B. unter Zuhilfenahme von methanolischer oder äthanolischer Salzsäure bei Raumtemperatur.
Zur Herstellung einer Verbindung der Formel I, in der R die Gruppe (b) mit X = Sauerstoff bedeutet, kann man ein Amid der obigen Formel VI oder auch einen erhaltenen Ester der Formel I, worin R die Gruppe (a) bedeutet, cyclisieren. Als Ausgangsverbindungen kommen selbstverständlich auch in analoger Weise hergestellte homologe Verbindungen in Betracht, worin anstelle der beiden Methylgruppen Wasserstoff bzw. andere niedere Alkylgruppen vorliegen. Die Cyclisation erfolgt z. B. unter den Bedingungen einer sauren Hydrolyse, vorzugsweise durch Behandeln mit einer wässrigen Mineralsäure, wie wässriger Salzsäure. Die saure Hydrolyse von Amiden der Formel VI erfolgt vorteilhaft in einem inerten organischen Lösungsmittel, z. B.
Tetrahydrofuran oder Dioxan, und bei etwa Raumtemperatur, wobei auch bei höheren Temperaturen, z.B. bis zur Siedetemperatur des Reaktionsgemisches, gearbeitet werden kann. Die saure Hydrolyse von Estern der Formel I erfolgt bevorzugt mit einem mit Wasser mischbaren Lösungsmittel, wie Aceton, Methyläthylketon, Tetrahydrofuran, Dimethoxyäthan oder Dioxan. Ein bevorzugtes Cyclisierungsmittel besteht aus 6n wässriger Salzsäure und Aceton in einem Gewichtsverhältnis von etwa 1:1. Die Reaktionstemperatur liegt vorzugsweise bei der Siedetemperatur des Reaktionsgemisches, wobei auch niedere Reaktionstemperaturen, z. B.
bis Raumtemperatur, mit entsprechend längeren Reaktionszeiten angewendet werden können. Die Ester der Formel I können ebenfalls durch Erwärmen ohne Lösungsmittel, d.h.
mittels einer Schmelze, cyclisiert werden. Die Temperatur für diese Schmelzreaktion liegt vorzugsweise im Bereich von etwa 100 bis 200 "C, insbesondere im Bereich von etwa 120 bis 160 "C. Nach einer bevorzugten Ausführungsform wird diese Cyclisierung durch Einsatz der für die Herstellung der Ester der Formel I verwendbaren Verbindungen der obigen Formeln II und III durchgeführt, wobei die Reaktion durch Wahl genügender Reaktionszeit bzw. Reaktionstemperatur ohne Isolierung der intermediär gebildeten Ester der Formel I bis zum gewünschten Cyclisationsprodukt, d.h. Hydantoin der Formel I, läuft. Die Reaktion kann beispielsweise dünnschichtchromatographisch verfolgt werden.

in der R4 und R5 die obige Bedeutung haben, umsetzen. Die Reaktion erfolgt in derselben Weise wie die oben beschriebene Cyclisation unter Verwendung von Ausgangsverbindungen der Formeln II und III, d. h. mittels einer Schmelze.
Verbindungen der Formel I, worin R1 und/oder R2 Nitro darstellt, können durch Nitrierung der entsprechenden Ausgangsverbindung ohne Nitrogruppe(n) hergestellt werden. Die Nitrierung erfolgt in an sich bekannter Weise, z. B.
durch Behandeln mit Salpetersäure, mit Salpetersäure und Schwefelsäure, mit Salpetersäure und Eisessig oder Essigsäureanhydrid oder auch mit einem Alkalimetallnitrat und Schwefelsäure. Die Nitrierung wird vorzugsweise bei niedriger Temperatur durchgeführt, beispielsweise bei etwa 0 "C bis Raumtemperatur.
Die Verbindungen der Formel I sind zum Teil kristalline, feste Substanzen, die in niederen Alkanolen, wie Methanol oder Äthanol, in Dimethylsulfoxid, Dimethylformamid und Hexamethylenphosphorsäuretriamid und z.T. auch in chlorierten Kohlenwasserstoffen, wie Chloroform, Methylenchlorid und Tetrachlorkohlenstoff, relativ gut löslich und in Äther, Benzol und Wasser relativ schwer löslich sind.
In den nachfolgenden Beispielen sind alle Temperaturen in 0 DC angegeben.
Beispiel 1
Herstellung von Tabletten nachstehender Zusammensetzung: Wirkstoff der Formel I 250,0 mg Milchzucker 100,0 mg Maisstärke 85,0 mg Äthylcellulose 10,0 mg Talcum 4,5 mg Magnesiumstearat 0,5 mg
450,0 mg
Der Wirkstoff wird mit dem Milchzucker und der Maisstärke vermischt und mit einer Lösung der Äthylcellulose in 40 ml Methylenchlorid granuliert. Das Granulat wird bei 40"C getrocknet, mit dem Talcum und Magnesiumstearat vermischt und zu Tabletten gepresst.
Beispiel 2
Herstellung von Kapseln nachstehender Zusammensetzung: Wirkstoff der Formel I 250,0 mg Milchzucker 155,0 mg
Maisstärke 30,0 mg Talcum 15,0 mg
450,0 mg
Der Wirkstoff wird mit dem Milchzucker und der Maisstärke homogen vermischt, durch eine Siebmaschine passiert und nach Untermischen des Talcums in Gelatinekapseln Nr. 4 abgefüllt.
Herstellung der Wirkstoffe
Beispiel 3
23,5 g (83 mMol) 1-(4-Chlorphenyl)-3-[1-(dimethylcar- bamoyl)-1-methyläthyl]-harnstoff werden in 1000 ml absolutem Methanol gelöst und mit 7,5 ml (83 mMol) methanolischer Salzsäure zwei Tage bei Zimmertemperatur gerührt.
Das Gemisch wird eingedampft und der Rückstand nach Zugabe von 50 ml Wasser mehrmals mit Chloroform extrahiert. Die Chloroformlösung wird über Natriumsulfat getrocknet und eingedampft. Der Rückstand wird aus Chloroform-Hexan kristallisiert. Man erhält N-[(4-Chlorphenyl)- carbamoyl]-2-methylalanin-methylester, der bei 148 bis 1500 schmilzt.
Analyse für Cl2HlscIN2O3 (270,72).
Berechnet: C 53,24 H 5,58 N 10,35 Cl 13,10% Gefunden: C 53,40 H 5,69 N 10,30 Q 13,19%
Der als Ausgangsverbindung eingesetzte 1-(4-Chlor- phenyl)-3-[ 1 -(dimethylcarbamoyl)- 1 -methyläthyWharnstoff kann wie folgt hergestellt werden:
43,7 g (0,255 Mol) p-Chlorbenzhydroxamsäure werden in 1300 ml absolutem Tetrahydrofuran gelöst und in einer Argonatmosphäre mit 33 ml (0,255 Mol) 3-Dimethylamino2,2-dimethyl-2H-azirin versetzt. Nach viertägigem Rühren bei Zimmertemperatur wird das ausgefallene Produkt abfiltriert und aus Tetrahydrofuran umkristallisiert. Man erhält
1 -(4-Chlorphenyl)-3-[I -(dimethylcarbamoyl)-l-methyläthy- harnstoff vom Schmelzpunkt 216 bis 2180.
Analyse für C33H18ClN3O2: Berechnet: C 55,03 H 6,39 N 14,81 CI 12,49%
Gefunden: C 55,07 H 6.53 N 14,74 Cl 12,67%
In der gleichen Weise erhält man 1-(4-Nitrophenyl)-3-[1- (dimethylcarbamoyl)- 1 -methyläthylj-harnstoff, der bei 215Z (Zers.) schmilzt.
Beispiel 4
15,1 g (119 mMol) p-Chloranilin werden in 300 ml absolutem Tetrahydrofuran gelöst und I5 Minuten mit einer Lösung von 17,0 g (119 mMol) 2-Isocyanato-2-methylpro pionsäuremethylester in 150 ml absolutem Tetrahydrofuran versetzt. Nach dreitägigem Kochen unter Rückflussbedingungen wird die Lösung zur Trockne eingedampft. Das verbleibende Produkt wird zweimal aus Methylenchlorid kristallisiert. Nach 6stündigem Trocknen unter stark vermindertem Druck bei 40c erhält man N-[(4-Chlorphenyl)-carb amoyl]-2-methylalanin-methylester, der bei 147 bis 1480 schmilzt.
In der gleichen Weise können hergestellt werden: N-[(3 -Chlorphenyl) -carbamoyl]-2-methylalanin-methyl- ester (2 Tage unter Rückflussbedingungen); Smp. 135 bis 136".
N-[(3 ,4-Dichlorphenyl)-carbamoyl]-2-methylalanin- methylester (2 Tage unter Rückflussbedingungen); Smp. 153 bis 1540.
N-[(4-Bromphenyl)-carbamoyl]-2-methylalanin-methyl- ester (2 Tage unter Rückflussbedingungen); Smp. 143 bis 1450.
N-[(4-Fluorphenyl)-carbamoyl]-2-methyl-alanin-methyl- ester (1 Tag unter Rückflussbedingungen); Smp. 132 .
Beispiel 5
9,75 g (50 mMol} 5-Amino-2-chlorbenzotrifluorid und 7,15 g (50 mMol) 2-Isocyanato-2-methylpropionsäure-methylester werden bei 80" geschmolzen und 30 Minuten bei dieser Temperatur gehalten. Das dabei auskristallisierte Produkt wird aus Isopropanol/Methylenchlorid umkristallisiert und 20 Stunden unter stark vermindertem Druck bei 50 getrocknet. Man erhält N-[(3-Trifluormethyl-4-chlorphenyl)- carbamoyl]-2-methylalanin-methylester, der bei 141 bis 142" schmilzt.
Analyse für C13H14ClF3N2O3 (338,71): Berechnet: C 46,10 H 4,17 N 8,27% Gefunden: C 46,17 H 4,14 N 8,26%
In der gleichen Weise erhält man: N-[(4-Chlorphenyl)-carbamoyli-2-methylalanin-methyl- ester (Schmelze 10 Minuten bei 1400); Smp.148 bis I50".
N-[(4-Trifluormethylphenyl)-carbamoyW2-methyl- alaninmethylester (Schmelze 60 Minuten bei 1200); Smp 1560 (Sinterung) bis 2200 (Zers.).
N-[(44odphenyl > carbamoyW2-methylalanin-methyl- ester (Schmelze 30 Minuten bei 90 ); Smp. 148 bis 1490.
N-[(3-Nitro-4-chlorphenyl)-carbamoylj-2-methylalanin- methylester (Schmelze 30 Minuten bei 100 ); Smp. 142 bis 143".
N-[(3-Trifluormethylphenyl)-carbamoyl]-2-methylala- ninmethylester (Schmelze 120 Minuten bei 1300).
Beispiel 6
14,5 g (100 mMol) 3-Chlor-4-fluoranilin und 17,2 g (120 mMol) 2-Isocyanato-2-methylpropionsäure-methylester werden bei 1400 geschmolzen und 6 Stunden bei dieser Temperatur gehalten. Das erhaltene Produkt wird aus Tsopropa- nol/Methanol/Methylenchlorid kristallisiert. Nach 1 6stündi- gem Trocknen unter stark vermindertem Druck bei 55" erhält man 3-(3-Chlor-4-fluorphenyl)-5,5-dimethylhydantoin, das bei 165 bis 166 schmilzt Analyse für C31 H1 0ClFN2O2 (256,66): Berechnet: C 51,48 H 3,93 N 10,91 Cl 13,81% Gefunden:

Beispiel 7
In der in Beispiel 6 angegebenen Weise erhält man aus 3,5-Dichlor-4-fluor-anilin und 2-Isocyanato-2-methylprn- pionsäure-methylester und durch Kristallisieren aus Ather/ Petroläther 3-(3,5-Dichlor-4-fluorphenyl)-5,5-dimethyl-hy- dantoin, das bei 175 bis 176 schmilzt.
Analyse für C11H9Cl2FN2O2 (291,11): Berechnet: C 45,39 H 3,12 N 9,62 Cl 24,36% Gefunden: C 45,55 H 3,19 N 9,57 Cl 24,58%
Beispiel 8
Eine Lösung von 8,00 g (23,6 mMol) N-[(3-Trifluorme- thyl-4-chlorphenyl)-carbamoyll-2-methylalanin-methylester in 20 ml 6n wässriger Salzsäure und 20 ml Aceton werden 2 Stunden auf dem Dampfbad erwärmt und anschliessend eingeengt. Das ausgefallene Produkt wird zuerst aus Methylenchlorid und anschliessend aus Methylenchlorid/Petroläther umkristallisiert. Nach 20stündigem Trocknen unter stark vermindertem Druck bei 50 erhält man 3-(3-Trifluorme thyl-4-chlorphenyl)-5,5-dimethylhydantoin vom Schmelzpunkt 152 bis 153".
Analyse für C12H10C1F3N2O2 (306,67): Berechnet: C 47,00 H 3,29 N 9,I3% Gefunden: C 46,89 H 3,23 N 8,93%
In der gleichen Weise erhält man: 3-(4-Bromphenyl)-5 ,5-dimethylhydantoin; Smp. 172 bis 173".

Beispiel 9
14,5 g (65 mMol) 3-Trifluonmethyl-4-chlorphenyl-iso- cyanat und 5,5 g (65 mMol) a-Amino-isobutyronitril werden 20 Minuten bei 100" gehalten. Das erhaltene dickflüssige Öl wird zur Reinigung an 500 g Kieselgel (Korngrösse etwa 0,06 bis 0,2 mm) mit Methylenchlorid/Methanol chromatographiert. Das erhaltene Produkt wird aus Methylenchlorid/ Petroläther kristallisiert. Nach 1 Sstündigem Trocknen unter stark vermindertem Druck bei 50 erhält man 1 -(3-Trifluor- methyl-4-chlorphenyl)-5-imino-4,4-dimethyl-2-imidazolidinon, das bei 148 bis 149 schmilzt.
