Gegenstand der vorliegenden Anmeldung sind somit die neuen 1H-Pyrazolo[3,4-b]pyridine der obigen Formel I und deren physiologisch verträglichen Salze mit anorganischen oder organischen Säuren sowie Verfahren zu ihrer Herstellung, und die sie enthaltenden pharmazeutischen Zubereitungen.
Für die bei der Definition des Restes R1 erwähnten Bedeutungen kommt somit die der Methylamino-, Äthylamino-, Propylamino-, Isopropylamino-, Butylamino-, Isoamylamino-, Hexylamino-, Benzylamino-, Dimethylamino-, Di äthylamino-, Dipropylamino-, Dibutylamino-, Diisobutyl amino , Dipentylamino-, N-Methyl-äthylamino-, N-Methyl -propylamino-, N-Methylhexylamino-, N-Äthyl-propylamino-, N-Äthyl-butylamino-, N-Äthyl-pentylamino-, N-Propyl-butylamino-, N Butyl-pentylamino-, Diallylamino-, N-Methyl -cyclohexylamino-, N-Äthyl-cyclohexylamino-, N-Propyl -cyclohexylamino-, N-Butyl-cyclohexylamino-, N-Pentyl -cyclohexylamino-, N-Methyl-benzylamino-, N-Äthyl-benzylamino-, N-Isopropyl-benzylamino-, N-Isobutyl-benzylamino-, Pyrrolidino-, Piperidino-, Methylpiperidino-, Dimethylpiperidino-, Hydroxypiperidino-, Hydroxymethylpiperidino , Phenylpiperidino-, Benzylpiperidino-,

Als sogenannte leaving -Gruppe, d.h. eine nukleophile, austauschbare Gruppe, kommt für X beispielsweise die Bedeutung des Chlor- der Bromatoms oder eines Aryloxyrestes wie die der Phenoxygruppe in Betracht.
Die Umsetzung wird zweckmässigerweise in einem Lösungsmittel wie Dimethylformamid, Dimethylsulfoxid, Wasser, Äthylenglykol, Propylenglykol, 2-Äthoxyäthanol oder in einem Überschuss eines verwendeten Hydrazins der Formel III, gegebenenfalls in Gegenwart einer Base wie Natriumcarbonat, Pyridin oder in einem Oberschuss des verwendeten Hydrazins, im allgemeinen bei erhöhten Temperaturen, z.B. bei Temperaturen zwischen 70 und 1800C, vorzugsweise jedoch bei Temperaturen zwischen 100 und 1500C durchgeführt. Die Umsetzung kann jedoch auch ohne Lösungsmittel durchgeführt werden.
Die erhaltenen Verbindungen der Formel I können gewünschtenfalls mit anorganischen oder organischen Säuren in ihre physiologisch verträglichen Säureadditionssalze übergeführt werden. Als Säuren haben sich beispielsweise Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Milchsäure, Zitronensäure, Weinsäure, Maleinsäure oder Fumarsäure als geeignet erwiesen.
Die als Ausgangsstoffe verwendeten Verbindungen der Formel II sind neu und lassen sich nach an sich bekannten Verfahren aus den entsprechenden 2,6-Dihalogen-pyridinen durch Umsetzung mit einem entsprechenden Amin der Formel H-R1, bei erhöhten Temperaturen, z.B. bei der Siedetemperatur des verwendeten Lösungsmittels, herstellen.
Wie bereits eingangs erwähnt, besitzen die neuen Verbindungen der Formel I wertvolle pharmakologische Eigenschaften, neben einer antiphlogistischen insbesondere antithrombotische Wirkungen.
Beispielsweise wurden die Substanzen A = 3-Amino-6-morpholino-4-phenyl- 1 H-pyrazolo [3,4-b] - pyridin, B = 3-Amino-4-phenyl-6-thiomorpholino- 1 H-pyrazolo- [3,4-b]pyridin, C = 3-Amino-4-phenyl-6-piperidino- 1 H-pyrazolo [3,4-b] pyridin, D = 3-Amino-6-(3-methylpiperidino)-4-phenyl- lH-pyrazolo [3,4-blpyridin, E = 3-Amino-6-hexamethylenimino-4-phenyl- 1H-pyrazolo f3,4-bjpyridin, F = 3-Amino-6-diäthylamino-4-phenyl- 1 H-pyrazolo [3,4-b]- pyridin, G = 3-Amino-4-(4'-methoxy-phenyl)-6-piperidino-1H -pyrazolo [3,4-b]pyridin und H = 3-Amino-4-(4'-fluorphenyl)-6-hexamethylenimino -lH-pyrazolo[3,4-b]pyridin im Vergleich zu I = <RTI

Die Thrombozytenaggregation wurde im plättchenreichen Plasma gesunder Versuchspersonen gemessen. Hierbei wurde der Verlauf der Veränderung der optischen Dichte nach Zugabe von handelsüblichem Collagen der Firma Hormonchemie, München, welches 1 mg Collagenfibrillen pro ml enthält, photometrisch gemessen und registriert. Aus dem Neigungswinkel der Dichtekurve wurde auf die Aggregationsgeschwindigkeit (Vmax) geschlossen. Der Punkt der Kurve bei dem die grösste Lichtdurchlässigkeit vorlag, diente zur Berechnung der optical density (O.D.).
Die Collagen-Dosen wurden möglichst gering gewählt, aber doch so, dass sich eine irreversible Aggregation ergab.
Zur maximalen Aggregationsauslösung werden ca. 0,01 ml der Collagenlösung zu 1 ml plättchenreichem Plasma gegeben.
Die Substanzen wurden vor Aggregationsauslösung 10 Minuten lang bei 37"C mit dem Plasma inkubiert. Die Substanzen wurden mit etwas Salzsäure oder Dimethylformamid gelöst und mit aqua dest. auf die gewünschte Endkonzentration verdünnt.
Es wurde die Es50, das heisst die Dosis bestimmt, die eine 50%ige Hemmung der Aggregation gegenüber Kontrolle bewirkte:

Die erfindungsgemäss hergestellten neuen Verbindungen der Formel I und deren physiologisch verträgliche Salze mit anorganischen und organischen Säuren eignen sich somit insbesondere zur Prophylaxe arterieller Thromboembolien und arterieller Verschlusskrankheiten und lassen sich zur pharmazeutischen Anwendung in die üblichen pharmazeutischen Zubereitungsformen wie Tabletten, Dragees, Kapseln, Suppositorien, Ampullen und Lösungen,
gegebenenfalls in Kombination mit anderen Wirksubstanzen, einarbeiten. Die Einzeldosis beträgt hierbei am Erwachsenen zweckmässiger weise 50-200 mg, vorzugsweise jedoch 120-170 mg, 3- bis 4mal täglich.
Die nachfolgenden Beispiele sollen die Erfindung näher erläutern: Vorbemerkung:
Bei den Schmelzpunktangaben handelt es sich um unkorrigierte Schmelzpunkte.
Beispiele zur Herstellung der Ausgangsverbindungen:
Beispiel A 2,6-Dihydroxy-4- phenyl-nicotinonitril
84 g (1 Mol) Cyanacetamid und 192 g (1 Mol) Benzoylessigsäureäthylester werden in 200 ml absolutem Äthanol unter Erwärmen gelöst. In die erhaltene Lösung lässt man dann unter weiterem Erwärmen eine Lösung von 56 g (1 Mo)l Kaliumhydroxid in 200 ml absolutem Äthanol im Verlaufe von etwa 2 Stunden langsam zutropfen. Das erhaltene Gemisch wird schliesslich etwa 20 Stunden lang unter Rückfluss erhitzt, wobei sich das Kaliumsalz des 2,6-Dihydroxy -4-phenylnicotinonitrils allmählich abscheidet. Es wird nach dem Erkalten abgesaugt und in 2-3 1 heissem Wasser gelöst Durch Ansäuern mittels konzentrierter Salzsäure wird dann das 2,6-Dihydroxy-4-phenylnicotinonitril als elfenbeinfarbener kristalliner Niederschlag ausgefällt.
Nach dem Erkalten wird abgesaugt, mit Wasser und wenig Aceton gewaschen und getrocknet.
Ausbeute: 89 g (42% der Theorie); F.: ca. 280"C (Zers.)
Beispiel B 2,6-Dichlor-4-phenyl-nicotinonitril
42,4 g (0,2 Mol) 2,6-Dihydroxy-4-phenyl-nicotinonitril werden mit 300 ml Phosphoroxychlorid in einem Glas-Druckgefäss unter Schütteln 6 Stunden lang auf etwa 1800C erhitzt. Nach dem Erkalten wird das Reaktionsgemisch portionsweise unter Rühren in einem Gemisch von Eis und Wasser (ca. 3 1) aufgenommen. Der abgeschiedene Niederschlag wird abgesaugt, mit Wasser gewaschen und getrocknet. Ausbeute: 47,4 g (95% der Theorie). Das erhaltene Reaktionsprodukt wird in Chloroform gelöst (Abfiltrieren eines geringen Rückstands) und über eine kurze Kieselgelsäule mittels Chloroform als Laufmittel gereinigt.
Nach Vereinigung und Eindampfen der Chloroformfraktionen wird das 2,6-Dichlor-4-phenyl-nicotinonitril in Form fast farbloser Kristalle erhalten..
Ausbeute: 37,5 g (75% der Theorie); F.: 167-169"C.
In analoger Weise wurde auch hergestellt: 2,6-Dibrom-4- phenyl-nicotinonitril durch Umsetzen von 2,6-Dihydroxy-4-phenyl-nicotinonitril mit Phosphoroxybromid bzw. mit Phosphortribromid und Triäthylamin, jedoch jeweils ohne Druck.
F.: 186-1880C (Essigester).
Beispiel C 2-Chlor-6-morpholino4-phenyl-nicotinonitril
Eine Suspension von 24,9 g (0,1 Mol) 2,6-Dichlor-4 -phenyl-nicotinonitril in 400 ml Äthanol wird langsam mit 17,4 g (0,2 Mol) Morpholin versetzt und anschliessend etwa 2 Stunden lang unter Rückfluss erhitzt. Schon während des Kochens, vor allem jedoch beim Erkalten des Reaktionsgemisches (evtl. Kühlen mit Eiswasser) scheidet sich das Reaktionsprodukt als farbloser kristalliner Niederschlag ab. Es wird abgesaugt, sehr gründlich mit Wasser gewaschen und getrocknet.
Ausbeute: 25,8 g (86% der Theorie); F.: 198-200"C.

Beispiel 1
3-A mino-6-morpholino-4-phenyl-lH-pyrazolo[3,4-bjpyridia
15,0 g (0,05 Mol) 2-Chlor-6-morpholino-4-phenyl-nicotinonitril (F.: 198-2000C) werden mit 20 ml Hydrazinhydrat (80%ig) in 100 ml Äthylenglykol 3 Stunden lang auf 1300C erhitzt. Die erhaltene Lösung wird in etwa 11 Wasser eingegossen, wobei sich das Reaktionsprodukt als fast farbloser Niederschlag abscheidet. Es wird abgesaugt, mit Wasser gewaschen und sofort einmal aus etwa 700 ml 0,ln-Salzsäure mittels konzentriertem Ammoniak umgefällt. Nach Absaugen, Waschen und Trocknen beträgt die Ausbeute 13,7 g (93 % der Theorie). Das so erhaltene 3 Amino-6-morpho lino-4-phenyl- 1 H-pyrazolo [3,4-b] pyridin ist praktisch rein, es schmilzt bei 178-1800C.

Das Hydrochlorid des 3-Amino-6-morpholino-4-phenyl -1H-pyrazolo[3,4-bpyfldins wird durch Lösen in etwa 2n Salzsäure unter Erwärmen und anschliessendes Auskristallisieren erhalten.

3-Amino-6-morpholino-4-phenyl-IH-pyrazol b]pyridilz
3,4 g (0,01 Mol) 2-Brom-6-morpholino-4-phenyl-nicotinonitril (F.: 198-200 C) werden mit 5 ml Hydrazinhydrat (80Zig) in 20 ml Äthylenglykol 3 Stunden lang auf etwa 130 C erhitzt. Nach Aufarbeitung und Reinigung analog Beispiel 1 schmilzt das Reaktionsprodukt bei 180-182"C.
Ausbeute: 2,3 g (78% der Theorie).
Beispiel 3
3-Amino-6-morpholino-4-phenyl-1H-pyrazolo[3,4-b]pyridin
3,6 g (0,01 Mol) 6-Morpholino-2-phenoxy-4-phenyl -nicotinonitril (F.: 150-1530C; erhalten aus 2-Chlor-6-morpholino-4-phenyl-nicotinonitril und Natriumphenolat in Phenol durch 3stündiges Erhitzen auf 1400C) wurden mit 5 ml Hydrazinhydrat (80%mg) in 25 ml Äthylenglykol 5 Stunden lang auf etwa 1500C erhitzt (Rückfluss). Nach einmaligem Umfällen aus 0,1n- Salzsäure mittels konz. Ammoniak beträgt die Ausbeute 2,8 g (91% der Theorie). Die so erhaltene Substanz schmilzt bei 200-202 C.

3-Amino-l -methyl-6-morpholino-4-phenyl-l H- pyrazolo- [3,4-b]pyridin
3,0 g (0,01 Mol) 2-Chlor-6-morpholino-4-phenyl-nicotinonitril werden mit 1,4 g (0,03 Mol) Methylhydrazin und 4 ml Äthylenglykol 3 Stunden lang auf 1700C (Badtemperatur) unter Rückfluss erhitzt. Die erhaltene Lösung wird in 50 ml Wasser aufgenommen, wobei das Reaktionsprodukt zunächst als schmieriger Niederschlag ausfällt. Es wird mit Wasser digeriert und nach dem Erstarren abgesaugt, gewaschen und getrocknet.
Ausbeute: 2,8 g (91% der Theorie).
Nach einmaligem Umkristallisieren aus Methanol schmilzt das 3-Amino- 1 -methyl-6-morpholino-4-phenyl- IH- pyrazolo [3,4-b]pyridin bei 149-152"C.

Die mit Milchzucker und Maisstärke gemischte Wirksubstanz wird mit einer 20%igen äthanolischen Lösung des Polyvinylpyrrolidons gleichmässig befeuchtet, durch ein Sieb der Maschenweite 2,0 mm granuliert, bei 45"C getrocknet und nochmals durch ein Sieb der Maschenweite 1,5 mm geschlagen.
Das so erhaltene Granulat wird mit Magnesiumstearat gemischt und zu Drageekernen verpresst.

Die so hergestellten Drageekeme werden nach bekannten Verfahren mit einer Hülle überzogen, die im wesentlichen aus Zucker und Talkum besteht. Die fertigen Dragees werden mit Hilfe von Bienenwachs poliert.

Die Lösung wird mit vorbehandeltem Wasser auf das gegebene Volumen aufgefüllt und steril filtriert. Alle Arbeitsgänge müssen in diffusem Licht erfolgen.
Abfüllung: In braune 5 ml-Ampullen unter Stickstoff begasung.
Sterilisation: 20 Minuten bei 121"C.

Die Sorbinsäure wird in Alkohol gelöst und die gleiche Menge Wasser zugesetzt. Darin wird die Wirksubstanz gelöst (Lösung 1). Der Zucker wird im restlichen Wasser gelöst (Lösung 2).
Lösung 2, Polyäthylenglykol 600 und die Kakaoessenz werden der Lösung 1 unter Rühren zugesetzt. Man filtriert durch ein geeignetes Filter.
1 ml Tropflösung 9 50 mg Wirksubstanz
Herstellung, Abfüllung und Lagerung der Lösung müssen unter Stickstoffbegasung und unter Lichtschutz erfolgen.

Solution 2, polyethylene glycol 600 and the cocoa essence are added to solution 1 with stirring. Filter through a suitable filter.
