La présente invention a pour objet un procédé de préparation de dérivés de l'acide prostanoique de formule générale:

dans laquelle n représente un nombre entier égal à 2, 3 ou 4 et m, un nombre entier égal à 3, 4 ou 5.
On sait que la prostaglandine A2 (PGA2) ou acide l5tt-hydroxy 9-oxo 5-cis 10,13-trans prostatriène olque ou encore médulline a été isolée de la médullaire rénale. Son extraction est effectuée à partir de médullaire congelée de rein de lapin.
On sait également que la prostaglandine A2 posséde de trés intéressantes propriétés thérapeutiques. On obtient des effets hypotenseurs avec des doses très faibles de ce composé. Par ailleurs l'efficacité de ce produit semble supérieure à celle des vasodilatateurs habituels.
La présente invention a principalement pour objet l'obtention de dérivés de l'acide prostanoîque purs par une voie synthétique qui ne nécessite ni de matiére premiére d'origine animale dont l'approvisionnement est coûteux, ni de procédé d'extraction, créant une purification du composé difficile et des rendements aléatoires.
Des synthéses de prostaglandine A2 ont certes déjà été décrites.
C'est ainsi que J. E. Pike et coll. décrivent la préparation de la
PGA2 par déshydratation de la prostaglandine E2 par l'acide acétique [Nobel Symposium 2(1966), 161]. Ce procédé de préparation présente divers inconvénients; d'une part, les rendements sont trés faibles du fait que le produit de départ n'est accessible que par une extraction présentant les désavantages énumérés ci-dessus ou que par un procédé de synthése totale [voir entre autres, à ce sujet, W. P. Schneider, Chem.
Com. 6D 304 < 1969)] nécessitant un nombre de stades réactionnels important; et d'autre part, les procédés de synthése utilisés n'étant pas toujours stéréospécifiques, ils peuvent pour certaines, conduire à un mélange d'isoméres que l'on doit séparer, soit à l'un des stades du procédé soit en fin de synthése, par des processus exigeant une mise en oeuvre difficilement réalisable sur le plan industriel.
Le procédé selon l'invention permet précisément d'éviter les inconvénients signalés ci-dessus.
Il est caractérisé en ce que l'on fait agir le brome en milieu
méthanolique sur l'acide prostatriéne olque de formule générale:

dont on soumet le groupement a-tétrahydropyranyloxy à une hydrolyse en milieu acide pour obtenir l'acide prostanolque de formule I désiré.
On décrit ci-dessus des modes opératoires préférés du procédé.
La bromo-méthoxylation sélective de l'acide prostatriène orque de formule générale V s'effectue, de préférence, dans des conditions particulièrement douces. On opère à des températures comprises entre -70 et - 30"C. Afin de neutraliser l'acidité du milieu, on ajoute avantageusement au milieu réactionnel, un sel alcalin d'acide faible tel que l'acétate de sodium ou le phosphate monoacide de sodium.
L'agent de déshydrohalogénation que l'on utilise pour effectuer la débromhydratation du bromocétal de formule générale VII', est de préférence un diaza bicyclo alcéne et plus particulièrement le diaza l,5-bicyclo (4-3-0) non Se ne, la réaction s'effectuant notamment dans un solvant aprotique dipolaire tel que le diméthylsulfoxyde, le diméthylformamide, le diméthylacétamide et l'hexaméthylphosphotriamide.
Pour l'hydrolyse du semi-acétal pyranyloxy de l'acide prosta triéne olque de formule générale VIII', on utilise de préférence comme agent acide, I'acide oxalique, la réaction s'effectuant dans un solvant organique tel que l'éthanol; il est également possible d'utiliser comme agent acide d'autres acides tels que l'acide acétique, I'acide chlorhydrique dilué ou l'acide sulfurique dilué.
Le dérivé de l'acide prostatriène olque de formule générale V, utilisé comme produit de départ, peut être obtenu comme suit:

Dans un mode opératoire préféré, on utilise le diazométhane comme agent de méthylation pour le traitement du composé de formule générale II.
La saponification du composé de formule générale III est conduite, de préférence, à une température inférieure à 50"C.
Comme agent basique, on peut utiliser un hydroxyde de métal alcalin, tel que la soude ou la potasse aqueuse.
Le traitement à chaud du composé de formule générale IV est effectué, de préférence, dans un solvant organique anhydre à une température comprise entre 50 et 1200 C. Comme solvant organique, on peut choisir notamment le xylène.
Les composés de formule générale II utilisés au départ peuvent être préparés par une méthode analogue à celle décrite dans la demande de brevet français publiée N0 2085654.

L'exemple suivant illustre le procédé selon l'invention, les préparations I, II, III et IV décrivant l'obtention du produit de départ.
Préparation I
Préparation du 7-bromo 5-hepténoate d'éthyle
Stade A: 6ia-tétrahydropyranyloxy) I-chloro 4-hexyne
On refroidit à - 35"C 150 cm3 d'ammoniac, ajoute 0,77 g de lithium en présence d'un peu de nitrate ferrique comme catalyseur et laisse en contact pendant une heure; on ajoute, sous agitation, 14 g d'éther pyranylique de l'alcool propargylique en solution dans 50 cm3 d'éther et continue l'agitation pendant deux heures; on ajoute alors une solution de 15,7 g de 1-chloro 3-bromopropane dans 25 cm3 d'éther et agite pendant trois heures à
-35"C; on neutralise par addition de 5,25 g de chlorure d'ammonium, évapore l'ammoniac, traite le résidu avec de l'éther saturé d'eau puis avec de l'eau, extrait à l'éther, lave les phases éthérées à l'eau, les sèche et évapore à sec;
on obtient 13 g dé 6ia-tétrahydropyranyloxy) 1-chloro 4-hexyne sous forme d'un produit liquide incolore, soluble dans les alcools, l'éther, le benzène et le chloroforme, bouillant à 100"C sous 0,6 mm de mercure, [n]n20= 1,485.
Le produit de départ, l'éther pyranylique de l'alcool propargylique est obtenu selon le procédé décrit par J. W. Conia, Bull.
Soc. Chim. 1955, 1449.
Stade B: 7ia-tétrahydropyranyloxy) 5-heptyne nitrile
On introduit 34,174 g de 6ia-tétrahydropyranyloxy) 1-chloro 4-hexyne dans 75 cm3 de diméthylformamide, ajoute une solution aqueuse de 13,6 g de cyanure de potassium et agite le mélange réactionnel pendant huit heures à 80 C; on refroidit, ajoute 100 cm3 d'eau et extrait à l'éther; on lave les phases éthérées à l'eau jusqu'à neutralité des eaux de lavage, sèche sur sulfate de magnésium et évapore à sec; on obtient 23 g de 7u-tétrahydro- pyranyloxy) 5-heptyne nitrile sous forme d'un liquide incolore, soluble dans les alcools, l'éther, le benzène et le chloroforme, insoluble dans l'eau, bouillant à 1 1 S C sous 0,1 mm de mercure, [n]20= 1,481.
Spectre I.R.
Présence de tétrahydropyranyloxy et de -C = N.
Stade C: acide 7-(a-tétrnhydropyranyloxy) 5-heptyne osque
On chauffe à 80"C pendant dix-huit heures, sous azote et sous agitation un mélange de 22,5 g de 7ia-tétrahydropyranyloxy) 5-heptyne nitrile, 9,5 g de potasse et 120 cm3 d'éthanol à 50%; puis on chasse l'méthanol sous vide, ajoute 50 cm3 d'eau, glace, ajoute 150 cm3 d'éther et amène le mélange à pH=4 par addition d'acide chlorhydrique 0,5 N glacé; on lave les phases éthérées à l'eau, sèche sur sulfate de magnésium, filtre et évapore sous pression réduite, on obtient 21,3 g d'acide 7-(cl-tétrahydropyranyloxy) 5-heptyne olque, sous forme d'un liquide incolore, soluble dans les alcools, l'éther, le benzène et le chloroforme, insoluble dans l'eau [n];20= = 1,485.
Stade D: 7-(a-tétrakydropyranyloxy) 5-heptyne oate d'e'thyle
On agite pendant une heure, à température ambiante, un mélange de 20 g d'acide 7-(a-tétrahydropyranyloxy) 5-heptyne oique, 100 cm3 de méthanol et 5,4 g de méthylate de sodium; on évapore le méthanol sous vide et obtient 22 g de 7-tésra- hydropyranyloxy) 5-heptyne oate de sodium; on mélange 21 g du sel de sodium ainsi obtenu et 70 cm3 de diméthylformamide, ajoute 27,2 g de bromure d'éthyle et chauffe le mélange réactionnel à 80"C pendant deux heures, sous agitation; on verse dans l'eau, extrait à l'éther, lave les phases éthérées à l'eau; sèche sur sulfate de magnésium, évapore le solvant et fractionne le résidu;
on obtient 13 g de 7ia-tétrahydropyranyloxy) 5-heptyne oate d'éthyle, sous forme d'un liquide incolore, soluble dans les alcools, l'éther, le benzène et le chloroforme, insoluble dans l'eau, bouillant à 122"C sous 0,1 mm de mercure, [n]i20= 1,47.
Stade E: 7-hydroxy 5-heptyne oate d'éthylène
On mélange 10 g de 7-(a-tétrahydropyranyloxy) 5-heptyne oate d'éthyle, 20 cm3 d'éther, 5 cm3 d'acide sulfurique à 25%, et 20 cm3 de méthanol; après une heure d'agitation, on ajoute 30 cm3 d'eau, extrait à l'éther, lave les phases éthérées à l'eau, sèche sur carbonate de sodium et évapore l'éther sous vide: on obtient 4,3 g de 7-hydroxy 5-heptyne oate d'éthyle, sous forme d'un liquide incolore, soluble dans les alcools, l'éther, le benzène et le chloroforme, insoluble dans l'eau, bouillant à 91"C sous 0,5 mm de mercure, [n]20 = 1,465.
Le 7-hydroxy 5-heptyne oate d'méthyle peut encore être obtenu de la manière suivante:
On porte au reflux pendant trois heures, sous azote, un mélange de 20 g d'acide 7ia-tétrahydropyranyloxy) 5-heptyne orque, 12 g d'éthanol, 50 cm3 de benzène et 2 gouttes d'acide sulfurique concentré. On laisse ensuite le mélange revenir à la température ambiante puis ajoute 20 cm3 d'acide sulfurique à 12,5%, 30 cm3 d'éthanol et des traces de laurylsulfate de sodium. On agite pendant une nuit à 200C puis neutralise au carbonate de sodium, extrait à l'éther, sèche, évapore le solvant sous pression réduite et rectifie le résidu. On obtient ainsi 8,5 g de 7-hydroxy 5-heptyne oate d'éthyle identique à celui décrit ci-dessus.
Stade F: 7-hydroxy 5-heptène oate d'éthyle
On introduit 300 mg de palladium sur sulfate de baryum à 5% dans 20 cm3 d'acétate d'éthyle, purge et fait passer un courant d'hydrogène; après absorption de 6 cm3 d'hydrogène, on ajoute une solution de 4 g de 7-hydroxy 5-heptyne oate d'éthyle dans 7 cm3 d'acétate d'éthyle contenant 0,4 cm3 de quinoléine et rince avec 14 cm3 d'acétate d'éthyle; on purge et fait passer un courant d'hydrogène; après absorption de 570 cm3 d'hydrogène, on filtre, rince le filtre à l'acétate d'éthyle, lave les phases organiques à l'acide chlorhydrique 0,5 N, puis à l'eau, sèche sur sulfate de magnésium, traite au charbon actif, filtre et évapore à sec; on obtient 2,6 g de 7-hydroxy 5-heptène oate d'éthyle, sous forme d'un liquide incolore soluble dans les alcools, l'éther, le benzène et le chloroforme, insoluble dans l'eau [n]20 =1,453.
Stade G: 7-brnrno 5-heptène oate d'éthyle
On mélange sous agitation 10 g de 7-hydroxy 5-heptène oate d'éthyle et 120 cm3 d'éther de pétrole, refroidit à -10 C et ajoute une solution de 2,9 g de tribromure de phosphore dans 30 cm3 d'éther de pétrole; on agite pendant trente minutes à - 100C et pendant une heure à 0 C; on verse le mélange réactionnel dans l'eau, extrait à l'éther, lave les phases éthérées à l'eau jusqu'à neutralité des eaux de lavage, séche sur sulfate de magnésium et évapore sous pression réduite;
on recueille 11,55 g de produit huileux que l'on purifie par passage sur silice, et élution au mélange cyclohexane-acétate d'éthyle, sous forme d'un produit
liquide incolore, soluble dans les alcools, l'éther, le benzène et
le chloroforme; insoluble dans l'eau, [n]i20= 1,4825.
Préparation Il
Préparation du 3 3' < -tétrahydropyranyloxy trans octényl-l?
cyclopentanone 2-carboxylate d'éthyle
Stade A: 3-oxo 6-heptyne oate d'éthyle
1) Enolate magnésien du malonate acide d'éthyle
On dissout, sous azote, 7,3 g de malonate acide d'éthyle (obtenu
selon le procédé de Bram et Vilkas, Bull. Soc. Chim., 945, 1964)
dans 50 cm3 de tétrahydrofuran, ajoute 0,10 mole de bromure
d'isopropyl magnésium en solution tétrahydrofuranique, chauffe
légèrement, ramène à la température ambiante et conserve sous
azote.
2) N,N'-thionyl diimidazole
On dissout 15 g d'imidazole dans 100 cm3 de tétrahydrofuran,
ajoute une solution de 6 g de chlorure de thionyle dans 60 cm3
de tétrahydrofuran en agitant pendant quinze minutes, filtre, lave
le filtre au tétrahydrofuran et obtient une solution de N,N'-thionyl
diimidazole.
3) Propargyl acétyl imidazole
A la solution de N,N'-thionyl diimidazole obtenue ci-dessus,
on ajoute 5 g d'acide propargylacétique dissous dans 50 cm3 de
tétrahydrofuran et agite pendant quinze minutes à température
ambiante; on obtient une solution de propargyl acétyl imidazole
que l'on utilise telle quelle.
4) Condensation
On ajoute cette solution à l'énolate magnésien obtenu selon 1) et agite pendant une nuit, à température ambiante; on acidifie par addition d'acide chlorhydrique à 4%, extrait à l'éther, lave les phases éthérées avec une solution aqueuse de bicarbonate de sodium, sèche, traite au charbon actif, filtre et évapore à sec; on passe le résidu sur alumine, élue au chlorure de méthylène et obtient 6,03 g de 3-oxo 6-heptyne oate d'éthyle que l'on utilise tel quel dans le stade suivant. Le composé se présente sous forme de prismes jaune pâle, solubles dans les alcools, I'éther, le benzène et le chloroforme, insolubles dans l'eau, fondant à 25o C.
Stade B: 3-éthoxy 6-yne 2-heptène oate d'éthyle
On dissout 10 g de 3-oxo 6-heptyne oate d'éthyle dans 60 cm3 d'éthanol, ajoute 30 cm3 d'orthoformiate d'éthyle et 0,35 cm3 d'acide sulfurique et distille 25 cm3 de solvant, sous barbotage d'azote, en quarante-cinq minutes; on refroidit, ajoute au chlorure de méthylène, lave à la soude 2 N, extrait au chlorure de méthylène, sèche les phases organiques sur sulfate de sodium, traite au noir, filtre et évapore à sec sous vide; on chromatographie le résidu sur gel de silice, élue au chlorure de méthylène et obtient 7,05 g de 3-éthoxy 6-yne 2-heptène oate d'éthyle, sous forme d'un liquide jaune pâle, soluble dans les alcools, I'éther, le benzène et le chloroforme, insoluble dans l'eau.
Le 3-éthoxy 6-yne 2-heptène oate d'éthyle peut encore être
obtenu de la manière suivante:
On mélange, sous azote, 10,1 g de 3-céto 6-heptyne oate
d'éthyle 100 cm3 d"éthanol, 20 cm3 d'orthoformiate d'éthyle et 1 g
d'acide paratoluènesulfonique et porte au reflux pendant une heure
vingt minutes; on refroidit, ajoute du chlorure de méthylène, verse
le mélange réactionnel dans 100 cm3 de soude N glacée, lave
les phases organiques à la soude N glacée, puis avec une solution
aqueuse de chlorure de sodium, réextrait les solutions de lavage
au chlorure de méthylène, sèche les phases organiques réunies
sur sulfate de sodium, filtre et évapore à sec sous vide; on
chromatographie le résidu sur gel de silice, élue au chlorure de
méthylène et obtient 6,716 g de 3-éthoxy 6-yne 2-heptène oate
d'éthyle, identique au précédent.
Stade C: 3-éthoxy 8-hydroxy 9-chloro 6-yne 2-tétradécène
oate d'éthyle
On refroidit à - 25 C un mélange de 2 g de 3-éthoxy 6-yne 2-heptène oate d'éthyle et 15 cm3 de tétrahydrofuran et ajoute 9,25 cm3 d'une solution éthérée de butyllithium 1,085 N; on laisse en contact pendant une heure trente minutes à - 25" C, refroidit à - 30C et ajoute 3 g de -chloro heptanal (obtenu selon le procédé de Krattiger, Bull. Soc.
Chim. 1953, 222), on laisse en contact pendant trente minutes à - 20 C, puis pendant trente minutes à 0 C et laisse revenir à la température ambiante; on verse le mélange réactionnel dans une solution aqueuse de phosphate monosodique glacée, extrait à l'éther, lave les phases organiques avec une solution aqueuse de bicarbonate de sodium, puis à Feau et enfin avec une solution aqueuse de chlorure de sodium; on sèche sur sulfate de sodium, traite au charbon actif, filtre et évapore à sec; on chromatographie le résidu sur gel de silice, élue au chlorure de méthylène à 0,5% d'acétone; on purifie le résidu par une nouvelle chromatographie sur gel de silice et élution au chlorure de méthylène à 0,25% d'acétone puis à 0,50% d'acétone;
on obtient 1,85 g de 3-éthoxy 8-hydroxy 9-chloro 6-yne 2 tétradécéne oate d'éthyle, sous forme d'un produit amorphe jaune pâle, soluble dans les alcools et l'éther et insoluble dans l'eau.

On peut également obtenir le 3-oxo 8-hydroxy 9-chloro 6-tétradécyne oate d'éthyle à partir du 3-éthoxy 6-yne 2-heptène oate d'éthyle (obtenu au stade B) de la manière suivante:
1) Condensation
On refroidit à - 60 C un mélange de 6,5 g de 3-éthoxy 6-yne 2-heptène oate d'éthyle et 110 cm3 de tétrahydrofuran et ajoute 29 cm3 d'une solution de butyllithium 1,6 N dans l'hexane, en agitant pendant trois heures;
on aJoute ensuite une solution de 9,7 g de x-chloroheptanal dans 25 cm3 de tétrahydrofuran et agite pendant une heure à -60 C, puis laisse revenir la température à OC; on verse le mélange réactionnel dans une solution aqueuse saturée de phosphate monosodique glacée, extrait à l'éther, lave les phases éthérées avec une solution aqueuse saturée de bicarbonate de sodium puis à l'eau jusqu'à neutralité des eaux de lavage, sèche sur sulfate de magnésium et évapore à sec.
2) Hydrolyse
On dissout le résidu dans 100 cm3 d'èthanol et 56,5 cm3 d'acide chlorhydrique 2 N, en chauffant à 50-60 C pendant une heure trente minutes, sous agitation et sous azote; on refroidit et verse la solution dans le mélange eau-glace; on extrait à l'éther, lave les phases éthérées à l'eau jusqu'à neutralité des eaux de lavage, sèche sur sulfate de magnésium et évapore à sec; on obtient 16 g de 3-oxo 8-hydroxy 9-chloro 6-tétradécyne oate d'éthyle brut.
3) Purification
On élimine les produits volatils contenus dans les 16 g de composé brut par distillation sous pression réduite; à la fin de la distillation, on refroidit, reprend le résidu à l'éther, traite au noir, filtre et évapore à sec.
On dissout le résidu dans 5 cm3 d'éther isopropylique, ajoute 4 volumes d'éther de pétrole sous agitation, refroidit la solution à -60' C, ajoute de l'éther de pétrole jusqu'à formation d'un dépôt et décante.
On mélange le dépôt avec 22 cm3 d'éthanol et 113 cm3 d'une solution saturée d'acétate cuivrique, agite le mélange pendant une nuit, décante, lave le complexe cuivrique formé avec de l'eau et essore; on met le complexe en suspension dans l'éther de pétrole, agite pendant une heure dans un bain glace-eau, filtre, lave le filtre avec de l'éther de pétrole glacé puis avec un mélange glacé d'éther de pétrole et d'éther (2-1); on dissout le précipité dans du chlorure de méthylène, acidifie par addition d'acide acétique dilué, ajoute au chlorure de méthylène, lave les phases chlorométhyléniques à l'eau, puis avec une solution aqueuse de bicarbonate de sodium et enfin à l'eau jusqu'à neutralité des eaux de lavage; on sèche sur sulfate de magnésium et évapore à sec; on obtient le 3-oxo 8-hydroxy 9-chloro 6-tétradécyne oate d'éthyle, identique au précédent.
Stade E: 3-oxo 8-hydroxy 9-chloro cis 6-tétradécène oate
d'éthyle
On introduit 240 mg de palladium sur sulfate de baryum à 5% dans 15 cm3 d'acétate d'éthyle, purge et fait passer un courant d'hydrogène pendant trente minutes; après absorption de 6 cm3 d'hydrogène, on ajoute une solution de 3,03 g de 3-oxo 8-hydroxy 9-chloro 6-tétradécyne oate d'éthyle dans 5 cm3 d'acétate d'éthyle contenant 0,3 cm3 de quinoléine et rince avec 15 cm3 d'acétate d'éthyle; on purge, fait passer un courant d'hydrogène pendant deux heures; après absorption de 223 cm3 d'hydrogène, on filtre, rince le filtre à l'acétate d'éthyle, lave les phases organiques à l'acide chlorhydrique 0,5 N puis à l'eau, sèche sur sulfate de sodium, traite au charbon actif filtre et évapore à sec sous vide;
on obtient 3 g de 3-oxo 8 hydroxy 9-chloro cis 6-tétradécène oate d'éthyle, sous forme d'un produit amorphe jaune pâle, soluble dans les alcools et l'éther et insoluble dans l'eau.

It is known that prostaglandin A2 (PGA2) or 15tt-hydroxy 9-oxo 5-cis 10,13-trans prostatriene olque acid or even medulline has been isolated from the renal medulla. It is extracted from frozen rabbit kidney medulla.
It is also known that prostaglandin A2 has very interesting therapeutic properties. Hypotensive effects are obtained with very low doses of this compound. In addition, the effectiveness of this product appears to be greater than that of the usual vasodilators.
The main object of the present invention is to obtain pure prostanoic acid derivatives by a synthetic route which requires neither raw material of animal origin, the supply of which is expensive, nor an extraction process, creating a purification. difficult compound and random returns.
Syntheses of prostaglandin A2 have certainly already been described.
Thus, J. E. Pike et al. describe the preparation of the

To a solution of 3.3 g of 3 3'a-tetrahydropyranyloxy trans 1'-octenyl) cyclopentanone 2-carboxylate of ethyl in 15 cm3 of benzene is added 1.165 g of potassium terbutoxide, under nitrogen and stirred for one hour, at room temperature; a solution of 2.76 g of ethyl 7-bromo 5-heptenoate in 5 cm3 of benzene is then added and the reaction mixture is left for twenty-four hours at room temperature;
the mixture is poured into an ice-cold aqueous monosodium phosphate solution, extracted with methylene chloride and the organic phases washed with water until the washing water is neutral; the washing water is re-extracted with methylene chloride, the combined organic phases are dried over magnesium sulfate and evaporated to dryness: the residue is chromatographed on silica gel, eluted with a cyclohexane-ethyl acetate mixture (80-20) and obtained 3.9 g of ethyl 15 α - (α - tetrahydropyranyloxy) 9-oxo 8-carbethoxy Scis 13-trans prostadiene oate, as a colorless liquid, soluble in organic solvents and insoluble in water.

420 mg of 15zx-tetrahydropyranyloxy 10-carbethoxy 9-oxo Scis 13-trans prostadiene ethyl oate are dissolved in 10 cm3 of ether, added at 0 C 40 cm3 of a chloromethylene solution of diazomethane titrating 1.5 g per 100 cm3 and stir;
the reaction mixture is allowed to return to room temperature and stirring is continued for eight hours, the excess diazomethane is evaporated off under reduced pressure while maintaining the temperature below 40 C; extracted with ether, dried over magnesium sulfate and evaporated to dryness; the residue is chromatographed on silica gel and eluted with a cyclohexane-ethyl acetate triethylamine mixture (80-20-0.3); after evaporation of the eluent, 295 mg of 15z (z-tetrahydropyranyloxy) 10-carbethoxy 9-methoxy 5cis, 9,13-trans prostatriene oate is obtained in the form of a clear yellow liquid, soluble in most organic solvents and insoluble in water.

A mixture of 2.6 g of 15cl- (cl-tetrahydropyranyloxy) 9-methoxy 5cis, 9,13-trans prostatriene oate of methyl 8.7 cm3 of normal sodium hydroxide, 20 cm3 of alcohol is heated at 40 ° C. for five hours. methyl and 5 cm3 of dioxane; the methanol is then evaporated off, the reaction mixture is extracted with diethyl ether, then the aqueous phase frozen beforehand with a solution of normal hydrochloric acid and extracted with ethyl acetate is acidified.
The organic phase is dried over magnesium sulfate to obtain, by evaporation of the solvent, 2.45 g of 15 α - (α -tetrahydro-pyranyloxy) 9-methoxy 5cis, 9,13-trans orc prostatriene acid.

9,9-dimethoxy 5cis.13 trans oic prostadiene
Cooled to - 70 C, 2.45 g of 15 α - (α -tetrahydro-pyranyloxy) 9-methoxy 5cis, 9,13-trans prostatriene oic acid, 200 cm3 of methyl alcohol and 2.13 g of sodium acetate, then 18.5 cm3 of a methanolic bromine solution titrating 5.5 g per 100 cm3 are added dropwise over one hour; the reaction mixture is allowed to return to room temperature and stirred for one hour;
