Verfahren zur Herstellung von neuen Estern der Nicotinsäure Die vorliegende Erfindung bezieht sich auf die Herstellung von neuen Estern der Nicotinsäurc, die zufolge ihrer besonderen pharmakologischen Eigen schaften wertvolle Arzneimittel darstellen.
Es wurde gefunden, dass man zu diesen, in pharmakologischer Hinsicht sehr wertvollen Estern der Nicotinsäure gelangt, wenn man Nicotinsäure mit Methylmorphin oder N-Allylnormorph;in ver- estert.
Der so erhältliche Nicotinsäureester des Methyl- morphins zeigt bei ausgezeichneter Verträglichkeit eine signifikante Verbesserung der Hustenreflex dämpfenden Wirkung gegenüber dem Ausgangs stoff Methylmorphin; ausserdem besteht die bei letz terem beobachtete lähmende Wirkung auf die Peri staltik des Darmes nicht.
Der weiters erhältliche N-Allylnormorphin-bis- (nicotin-säureester) zeichnet sich gegenüber dem N-Allylnormorphin dadurch aus, dass er bereits in Mengen von Bruchteilen eines mg beim Menschen die atemzentrumlähmende Wirkung von Opiaten ver ringert. Auch bei der experimentellen Alkaloidver- giftung, z. B. mit Morphin, gelingt es, mit diesem Körper das Atemzentrum weitgehend zu schützen.
Zur Herstellung dieser neuen Ester werden zweck mässig funktionelle Derivate der Nicotinsäure, wie z. B. Nicotinsäurea.nhydrid oder Nicotinsäurehalogenide in Form ihrer halogenwasserstoffsauren Salze, mit Methylmorphin oder N-Allylnormorphin kondensiert. Gegebenenfalls kann auch Nicotinsäure unmittelbar mit den Hydroxylverbindungen unter Zusatz von beispielsweise Phosphoroxychlorid umgesetzt wer den.
Im Falle der Anwendung von Nicotinsäure- halogenid-Hydrohalogeniden, insbesondere Nicotin- säurechlorid-Hydrochlorid bzw. Nicotinsäure, wird die Kondensation mit den Hydroxylverbindungen vorzugsweise in Gegenwart von tertiären Basen durchgeführt. Als solche tertiäre Basen kommen bei spielsweise in Betracht Pyridin, Chinolin, Dimethyl- anilin oder dergleichen. Erforderlichenfalls kann die Umsetzung auch unter Zusatz inerter Lösungsmit tel, wie z.
B. Alkylbenzole, Chlorbenzole, durchge führt werden. Durch die Veresterung der erfindungs- gemäss als Ausgangsstoffe zur Anwendung gelangen den Hydroxylverbindungen werden die pharmako logischen Eigenschaften der verwendeten Kompo nenten in der oben angegebenen Weise modifiziert.
Das erfindungsgemässe Verfahren soll im nach steÜenden an Hand von Ausführungsbeispielen näher erläutert werden.
<I>Beispiel 1</I> 10 Gew.-T-.ile Nicotinsäureanhyrid werden in einem Bade bei einer Badtemperatur von 125 bis l40 C, vorzugsweise 135 C, zum Schmelzen ge bracht und .in die Schmelze 2,4 Gew.-Teile Methyl- morphin-hydrochlorid (wasserfrei) unter Rühren in Anteilen eingetragen.
Die Schmelze verfestigt sich schon nach wenigen Minuten; nach einer Einwirkungszeit von insge samt 30-35 Minuten wird das Erhitzen unterbro chen, das Reaktionsgut zerkleinert, mit etwa 60 Gew.-Teilen Wasser versetzt, Natriumbicarbonat bis zum Aufhören der Kohlendioxydentwicklung einge tragen und anschliessend Soda zugefügt, bis ein pH-Wert von etwa 9 erreicht ist. Das Reaktions produkt scheidet sich hierbei als zähe, klebrige Masse aus, welche mit Wasser durchgeknetet und so von den anorganischen .Salzen befreit wird.
Anschlie ssend wird die Masse in so viel n/2 Salzsäure auf genommen, bis auf dem Dampfbade vollständige Lösung eingetreten ist; der pH-Wert dieser Lösung soll 4 nicht unterschreiten. Nach Klärung der Lösung mit Aktivkohle wird das Filtrat zur Trockene verdampft, wobei der Niko- tinsäureester des Methylmorphins in Form seines Hydrochlorides als glänzende kristallinische Masse zurückbleibt. Ausbeute: 2,2 Gew.-Teile.
Nach dem Umkristallisieren aus 9511/o igem Alkohol wird die Substanz völlig rein erhalten; F = 263,0-263,5 C.
<I>Beispiel 2</I> 22 Gew.-Teile Nicotinsäurechlorid-hydrochlorid werden mit 49 Gew.-Teilen wasserfreien Pyridins verrührt und 34 Gew: Teile Methylmorphin-hydro- chlorid (wasserfrei) zugegeben.
Der Ansatz wird 15 Minuten im Bade bei 90 C Badtemperatur erwärmt, die nach dem Erkalten anfallende Kristallmasse in 250 Gew.-Teilen Wasser aufgenommen und mit Natriumbicarbonat und Soda auf pH = 9 eingestellt.
Die als Öl abgeschiedene Base und das freige setzte Pyridin werden mit Chloroform ausgezogen und die Chloroformlösung am Dampfbade einge dampft, wodurch das Chloroform und das Pyridin vertrieben werden. Die nun als Öl verbleibende Base wird mit n-Salzsäure auf pH = 4 eingestellt und am Dampfbade eingeengt. Nach dem Erkalten fällt das Hydrochlorid der Verbindung kristallin an, das aus Alkohol umkristallisiert und am Dampf bade getrocknet wird.
Man erhält so 30 Gew.-Teile Methylmorphin- nicotinsäurcester-hydrochlorid, das sind 70,011/o der Theorie; F = 262-263 C unter Zersetzung und Rot färbung.
<I>Beispiel 3</I> 1 Gew.-Teil N-Allyl-normorphin-hydrochlorid wird in 5 Gew.-Teilen geschmolzenes Nicotinsäure- anhydrid bei einer Badtemperatur von 125-140 C, vorzugsweise 130 C, unter Rühren eingetragen und die Schmelze insgesamt 1 Stunde bei dieser Tem peratur gehalten.
Nach dem Erkalten wird die Schmelze zerklei nert, mittels 500 Gew.-Teilen Wasser in der Wärme in Lösung gebracht, die schwach braun gefärbte Lösung mit Aktivkohle entfärbt und das Filtrat nach dem Erkalten mit Natriumbicarbonatlösung und an schliessend mit Sodalösung auf pH = 9 eingestellt. Die erhaltene Verbindung kristallisiert unmittelbar in glänzenden Flittern aus, die auf dem Filter ge sammelt, sodann mit Wasser gewaschen und schliess lich im Vakuum bei Normaltemperatur über festem Natriumhydroxyd getrocknet werden.
Ausbeute: 1,3 Gew.-Teile N - Allyl - normorphin - bis - (nicotinsäure- ester).
Zur Reinigung wird der Ester durch Erwärmen in 4 Gew.-Teilen n-Salzsäure und 26 Gew.-Teilen Wasser gelöst, die Lösung filtriert und das Filtrat im Vakuum über Natriumhydroxyd eingedampft, wobei glänzende Kristalle in einer Menge von 1,2 Gew.-Teilen erhalten werden.
Zum Umkristallisieren werden die Kristalle in 47 Gew.-Teilen Alkohol gelöst und die Lösung auf die Hälfte des Volumens im Vakuum über Na triumhydroxyd eingedampft, die auskristallisierte Substanz wird am Filter gesammelt, mit absolutem Alkohol gewaschen und getrocknet; F = 238,0 bis 239,0 C.
Die Marquissche Probe (Morphin und morphin- ähnlich wirkende Verbindungen, O. Schaumann, Springer Verlag, Berlin-Göttingen-Heidelberg 1957, Seite 25) fällt mit der neuen Verbindung negativ aus, während das als Ausgangsmaterial verwendete N-Allyl-normorphin eine starke rotviolette Färbung liefert.
<I>Beispiel 4</I> 4,3 Gew.-Teile Nicotinsäurechlorid-hydrochlorid werden in 19 Gew.-Teilen wasserfreies Pyridin ein getragen und hierauf 1 Gew.-Teil N-Allyl-normor- phin zugefügt. Der Ansatz wird im Bade durch 30 Minuten auf 90 C erwärmt und die Reaktions masse nach dem Erkalten in 100 Gew.-Teilen Was ser aufgenommen.
Die Lösung wird sodann vorerst mit Natriumbicarbonat und anschliessend mit Na- triumcarbonat versetzt und auf pH = 9 eingestellt, wobei sich das Reaktionsprodukt gemeinsam mit dem Pyridin ölig ausscheidet.
Das Reaktionsprodukt wird mit Chloroform extra hiert, das Chloroform sowohl als auch das mit extrahierte Pyridin auf dem Dampfbade abgetrie ben und der Rückstand mit n12 Salzsäure auf pH = 4 eingestellt, wobei das Hydrochlorid des N-Allyl-normorphin-bis-(nicotinsäurcesters) auskri stallisiert. Beim Eindampfen im Vakuum über festem Natriumhydroxyd zur Trockene bleibt der kristalline Rückstand in einer Menge von 1,1 Gew.-Teilen zurück.
Durch Auflösen des Rückstandes in 47 Gew.- Teilen Alkohol in der Wärme und Einengen im Vakuum auf die Hälfte des Volumens kristallisiert der erhaltene N-Allyl-normorphin-bis-(nicotinsäure- ester) aus; F = 238,0-239,0 C.
Die Marquissche Probe ist negativ; das so ge wonnene Produkt ist identisch mit dem nach Bei spiel 3 hergestellten Ester.

The nicotinic acid ester of methylmorphine obtainable in this way shows, with excellent tolerance, a significant improvement in the cough reflex suppressing effect compared to the starting material methylmorphine; in addition, the paralyzing effect observed in the latter on the peristalsis of the intestine does not exist.
The N-allylnormorphine bis (nicotinic acid ester), which is also available, is distinguished from the N-allylnormorphine in that it reduces the respiratory center paralyzing effect of opiates in humans in fractions of a mg. Even with experimental alkaloid poisoning, e.g. B. with morphine, it is possible to largely protect the respiratory center with this body.
To prepare these new esters, functional derivatives of nicotinic acid such. B. Nicotinsäurea.nhydrid or nicotinsäurehalogenide in the form of their hydrohalic acid salts, condensed with methylmorphine or N-allylnormorphine. If necessary, nicotinic acid can also be reacted directly with the hydroxyl compounds with the addition of, for example, phosphorus oxychloride.

After recrystallization from 9511% alcohol, the substance is obtained completely pure; F = 263.0-263.5 C.
<I> Example 2 </I> 22 parts by weight of nicotinic acid chloride hydrochloride are stirred with 49 parts by weight of anhydrous pyridine and 34 parts by weight of methylmorphine hydrochloride (anhydrous) are added.
The batch is heated in the bath for 15 minutes at a bath temperature of 90 ° C., and the crystal mass obtained after cooling is taken up in 250 parts by weight of water and adjusted to pH = 9 with sodium bicarbonate and soda.

<I> Example 3 </I> 1 part by weight of N-allyl normorphine hydrochloride is introduced into 5 parts by weight of molten nicotinic anhydride at a bath temperature of 125-140 ° C., preferably 130 ° C., with stirring and the Melt held at this temperature for a total of 1 hour.

<I> Example 4 </I> 4.3 parts by weight of nicotinic acid chloride hydrochloride are introduced into 19 parts by weight of anhydrous pyridine, and 1 part by weight of N-allyl-normorphine is then added. The reaction product is extracted with chloroform, both the chloroform and the extracted pyridine are removed on the steam bath and the residue is adjusted to pH 4 with n12 hydrochloric acid, the hydrochloride of the N-allyl-normorphine-bis- (nicotinic acid ester) auskri installed. When evaporating to dryness in vacuo over solid sodium hydroxide, the crystalline residue remains in an amount of 1.1 parts by weight.
By dissolving the residue in 47 parts by weight of alcohol while warming and concentrating it in vacuo to half the volume, the N-allyl-normorphine-bis (nicotinic acid ester) obtained crystallizes out; F = 238.0-239.0 C.
The Marquis trial is negative; the product thus obtained is identical to the ester prepared according to Example 3.
