L'invention a pour objet la définition et la mise en oeuvre de moyens d'identification positive d'échantillons analysés par électrophorèse. Plus précisément, l'invention concerne des moyens d'identification d'applicateurs d'échantillons sur un support d'électrophorèse, qui permettent d'associer au profil électrophorétique d'une ou plusieurs séries d'échantillons analysés, le ou les applicateurs d'échantillons utilisés et donc, d'identifier de façon fiable le résultat de l'analyse par rapport aux échantillons primaires.
Dans le cadre des analyses d'échanfiillons après séparation par électrophorèse, notamment à des fins cliniques, et en particulier lors d'analyses de routine pour la détermination de pathologies, il est nécessaire pour la sécurité des résultats produits d'être en mesure de suivre de façon fiable le devenir de chaque échantillon au cours de l'analyse. En particulier, il est nécessaire de s'assurer que le résultat obtenu à l'issue de la séparation par éiectrophorèse peut sans aucun doute ëtre attribué à l'échantillon auquel il correspond en théorie (échantillon primaire).
L'incertitude quant à l'attribution d'un résultat au bon échantillon primaire, peut résulter par exemple des opérations exécutées manuellement et dont la bonne exécution dépend de ce fait essentiellement de l'attention de l'opérateur.
Dans la pratique actuelle de ce type d'analyse, une erreur peut ne pas être d étectée.
L'invention propose ainsi des moyens d'identification qui permettent de repérer un échantillon ou une série d'échantillons et de l'identifier sur le support d'électrophorèse, à l'issue de la séparation électrophorétique. Ces moyens peuvent être mis en oeuvre dans le cadre de toute séparation électrophorétique, employant toute technique appropriée pour réaliser une électrophorèse de zone sur un support. Dans un milieu d'électrophorèse contenant une solution tampon, les échantillons contenant les protéines sont déposés au moyen d'un applicateur d'échantillons à la surface du gel, où elles s'ionisent et, sous l'effet d'un champ électrique, migrent à des vitesses différentes en fonction de leur charge respective. Après l'étape de migration qui permet la séparation des constituants protéiques des échantillons, ces protéines sont révélées, par exemple par coloration au moyen d'un colorant spécifique et quantifiées par exemple par une lecture densitométrique.
Comme la majorité des techniques modernes d'analyse, l'électrophorèse a été portée à un degré important d'automatisation afin d'une part, de faciliter sa mise en oeuvre et d'autre part de fa fiabifiser. L'invention permet d'identifier sans erreur un applicateur et en conséquence une série d'échantillons à analyser associés à cet applicateur, par l'examen de l'image électrophorétique de l'applicateur obtenue, après les opérations de dépôt, migration, coloration et séchage, des moyens d'identification de l'applicateur et simultanément, des échantillons analysés.
L'utilisation de la présente invention, en association avec les méthodes classiques d'identification (par exemple les codes à barres ou les codes bidimensionnels tels que les codes à matrice) des différentes composantes du système d'analyse notamment les tubes échantillons, l'applicateur et le gel, permet l'identification et la traçabilité des échantillons depuis le tube primaire contenant le prélèvement, jusqu'au résultat final.

Un procédé d'analyse par électrophorèse, des constituants d'échantillons, notamment d'échantillons biologiques prélevés chez des patients comprend les étapes suivantes - le dépôt des échantillons biologiques, au moyen de plusieurs applicateurs d'échantillons, sur un ou plusieurs supports d'électrophorèse, chaque applicateur comprenant des moyens pour l'identifiier (marqueur d'identification), identifiables par (examen du support d'électrophorèse à l'issue de l'électrophorèse ;

Le procédé ainsi défini pour l'analyse par électrophorèse des constituants d'échantillons à analyser, peut être intégré dans un système dans lequel les moyens d'identification de l'applicateur sont associés à un système de repérage complet de l'ensemble des composantes du système d'analyse dans le processus d'identifiication des échantillons à analyser par rapport au résultat obtenu à l'issue de l'électrophorèse.
Chaque marqueur d'identification, déposé sur le support d'électrophorèse, en même temps que les échantillons, et soumis à
l'électrophorèse, donne une image électrophorétique particulière qui devra être repérable, notamment visible, sur le support d'électrophorèse, après les difFérentes étapes de l'analyse : migration, coloration et séchage. Ce repérage peut se faire de manière manuelle par un opérateur ou de façon automatique, par exemple lors de l'analyse du support d'électrophorèse par un densitomètre.

L'applicateur ou les applicateurs ainsi chargés et le support d'électrophorèse sont introduits dans l'instrument d'électrophorèse après lecture de leur code respectif. A ce niveau, chaque support d'électrophorèse est associé à un ou plusieurs applicateurs. Après les étapes de migration, coloration et séchage, le support est introduit dans le densitomètre après lecture de son code. Le densitomètre, ou l'opérateur, repère sur chaque rangée le marqueur, ce qui permet au système d'identifier sans erreur les échantillons de cette rangée et donc de leur attribuer un résultat.
Ce système permet donc d'assurer la traçabilité totale des échantillons depuis le tube primaire jusqu'au résultat final.
Selon le premier mode de réalisation préféré de l'invention, le procédé
d'analyse par électrophorèse comprend, pour la détection des constituants séparés et/ou des marqueurs d'identification d'applicateurs, une étape de révélation des constituants séparés à l'issue de l'électrophorèse.
Selon un mode particulier de l'invention, on met en oeuvre différents applicateurs d'échantillons pour déposer plusieurs rangées d'échantillons différents sur un même support d'électrophorèse.
Selon un autre mode de réalisation de l'invention ci-dessus décrite, on met en oeuvre différents applicateurs d'échantillons pour déposer plusieurs rangées d'échantillons différents sur différents supports d'électrophorèse.
Dans ce cas, on peut réaliser en parallèle, plusieurs électrophorèses sur différents instruments, en parallèle.
Dans un procédé d'analyse défini selon les modalités précédentes y compris les combinaisons des modes de réalisation particuliers proposés ci-dessus, le marqueur d'identification de chaque applicateur est constitué par une composition spécifique pour chaque applicateur, déposée sur une piste déterminée sur le support d'électrophorèse, simultanément aux échantillons.
Alternativement, le marqueur d'identification de chaque applicateur est constitué par une composition commune à tous les applicateurs, ladite composition étant déposée sur le support d'électrophorèse au niveau d'une piste difFérente pour chaque applicateur et donc pour le support d'électrophorèse, simultanément aux échantillons.
Dans le cadre de la réalisation du procédé de l'invention, le marqueur d'identification d'applicateurs d'échantillons est constitué par toute composition telle que définie précédemment.
Les différents marqueurs précédemment décrits peuvent étre mis en oeuvre dans un procédé comprenant les étapes suivantes - le dépôt des échantillons biologiques, au moyen de plusieurs applicateurs d'échantillons, sur un ou plusieurs supports d'électrophorèse, chaque applicateur comprenant des moyens pour l'identifier (marqueur d'identification), identifiables par l'examen du support d'électrophorèse à l'issue de l'électrophorèse ;
- l'application d'un champ électrique pour permettre la migration des constituants des échantillons ;
- le séchage du ou des supports d'électrophorèse, - la coloration du ou des supports d'éiéctrophorèse, - la décoloration du ou des supports d'électrophorèse, - le séchage du ou des supports d'électrophorèse.
Dans le cadre de ce procédé, le marqueur d'identification peut être identifié avant l'étape de coloration du support d'électrophorèse, ou s'il ne s'agit pas d'un marqueur coloré, par coloration lors de l'étape de détection des constituants des échantillons qui comprend la coloration du support d'électrophorèse.
Dans un autre mode de réalisation de l'invention, les espèces chimiques colorées sont des anticorps, de préférence monoclonaux de façon à obtenir une bande distincte, et dans l'hypothèse de l'utilisation d'un mélange d'anticorps monoclonaux, chaque anticorps a une mobilité telle qu'il conduit à l'obtention d'une bande distincte de celles obtenues avec les autres anticorps du mélange.
Ces anticorps peuvent être détectés par toute méthode connue en soi, applicable dans le cadre d'une analyse comportant unie séparation électrophorétique.
De même, on peut ajouter au procédé ainsi défini, une étape de détection par analyse quantitative des différents constituants séparés.
La séparation électrophorétique est réalisée sur tout type de support d'électrophorèse et en particulier sur des supports tels que des gels, notamment des gels d'agarose, des gels de polyacrylamide, ou sur des membranes d'acétate de cellulose. De façon générale, on aura recours à tout support poreux permettant la diffusion des échantillons et leur migration dans le support pour permettre la séparation des constituants contenus dans ces échantillons.
Selon un autre mode de réalisation de l'invention, le procédé est un procédé d'immunofixation dans lequel les constituants séparés des échantillons sont révélés de toute façon en soi connue, au moyen d'anticorps, ledit procédé
incorporant, dans l'étape de dépôt des échantillons à analyser, le dépôt d'un marqueur d'identification de l'applicateur d'échantillons.
L'invention concerne aussi un procédé d'identification positive d'échantillons, pour permettre d'associer l'image électrophorétique d'un échantillon analysé, à un échantillon prélevé, comprenant une étape de marquage de l'applicateur d'échantillons au moyen d'un marqueur d'identification, identifiable par son image électrophorétique et l'utilisation de l'applicateur d'échantillons ainsi marqué pour le dépôt des échantillons à
analyser sur un support d'électrophorèse.
Pour la mise en oeuvre de ce procédé, tous les marqueurs décrits précédemment, utilisables dans le cadre du procédé d'analyse par électrophorèse de l'invention, peuvent être mis en aeuvre.
L'invention a également pour objet un kit (trousse) pour la réalisation de l'analyse des constituants d'échantillons par électrophorèse, comprenant ~ un support d'électrophorèse comprenant un matériau poreux approprié pour recevoir les échantillons à analyser et le marqueur d'identification de l'applicateur d'échantillon et permettre leur migration électrophorétique, ~ une ou plusieurs compositions constituées) par des espèces chimiques, utilisables lors d'une analyse électrophorétique pour l'identification d'applicateur(s) d'échantillons, cette (ces) compositions) étant identifiables) par son(leur) image électrophorétique.
Le marqueur d'applicateur d'échantillons utilisé dans ce kit répond aux définitions données précédemment, prises isolément ou en combinaison.
En outre, le kit peut comprendre tout réactif habituellement utilisé pour des analyses par électrophorèse, en particulier tout ou partie des réactifs suivants - un ou plusieurs réactifs pour la révélation des constituants séparés des échantillons ;
- un ou plusieurs réactifs pour la révélation des marqueurs d'identification des applicateurs.
D'autres propriétés et avantages particuliers de l'invention apparaissent encore dans les exemples qui suivent et les figures qui les accompagnent.
