Gegenstand der Erfindung ist ein Verfahren zur Herstellung neuer substituierter Arylalkylamine der allgemeinen Formel

Für die bei der Definition der Reste R B, und Rs erwähnten Alkylreste kommen insbesondere die Bedeutung der Methyl-, Äthyl-, Propyl- oder Isopropylgruppe und für die bei der Definition der Reste R6 und R7 erwähnten Alkoxyreste insbesondere die der Methoxy-, Äthoxy-, Propoxy- oder Isopropoxygruppe in Betracht.
Die Verbindungen der obigen allgemeinen Formel (I) und deren Säureadditionssalze besitzen wertvolle pharmakologische Eigenschaften, insbesondere eine herzfrequenzsenkende Wirkung.
Das erfindungsgemässe Verfahren zu ihrer Herstellung ist dadurch gekennzeichnet, dass man ein Benzylhalogenid der allgemeinen Formel

in der R4 bis R7 und n wie eingangs definiert sind, umsetzt und das so erhaltene 1-Imino-phthalimidin hydrolysiert und gewünschtenfalls eine erhaltene Verbindung der allgemeinen Formel (1), in der R4 ein Wasser- stoffatomdarstellt, niederalkyliertund/oder eine erhaltene Verbindung der allgemeinen Formel (1) in ein physiologisch verträgliches Säureadditionssalz mit einer anorganischen oder organischen Säure überführt.
Die Umsetzung wird gegebenenfalls in einem Lösungsmittel, z. B. in Aceton, Äthanol, Dimethylformamid, Dimethylsulfoxyd oder Methylenchlorid, und zweckmässigerweise bei erhöhten Temperaturen, z. B. bei Temperaturen zwischen 50 und 150 C, durchgeführt. Vorteilhaft ist die Gegenwart eines säurebindenden

Mittels, wie z. B. eines Alkoholats, eines Alkalihydroxyds, eines Alkalicarbonats oder einer tertiären organischen Base wie Triäthylamin oder Pyridin, und/oder eines Reaktionsbeschleunigers wie beispielsweise Kaliumjodid.
Die anschliessende Hydrolyse wird zweckmässigerweise in Gegenwart einer Base wie Kaliumcarbonat oder in Gegenwart einer Säure wie Salzsäure in einem wässerigen Medium wie Äthanol/Wasser oder Dioxan/Was- ser und bei Temperaturen zwischen 500C und der Siedetemperatur des verwendeten Lösungsmittels durchgeführt.
Erhält man eine Verbindung der allgemeinen Formel (1), in der R4, ein Wasserstoffatom darstellt, so kann diese mittels Alkylierung, z. B. durch Umsetzung mit einem entsprechenden Alkylhalogenid oder Dialkylsulfat, niederalkyliert oder durch Umsetzung mit Formaldehyd/Ameisensäure methyliert werden.
Die erhaltenen Verbindungen der allgemeinen Formel (1) können mit anorganischen oder organischen Säuren in ihre physiologisch verträglichen Salze übergeführt werden. Als Säuren haben sich beispielsweise Salzsäure, Phosphorsäure, Bromwasserstoffsäure, Schwefelsäure, Milchsäure, Weinsäure oder Maleinsäure als geeignet erwiesen.
Die alsAusgangsstoffe verwendeten Verbindungen der allgemeinen Formeln (II) und (III) lassen sich nach an sich bekannten Verfahren herstellen. So erhält man beispielsweise ein Benzylhalogenid der allgemeinen Formel (H) vorzugsweise durch Halogenierung eines entsprechenden Toluols, z. B. mit N-Brom-succinimid in Tetrachlorkohlenstoff, gegebenenfalls unter Zugabe eines Radikalstarters wie Dibenzoylperoxyd und/oder unter UV-Bestrahlung.
Wie bereits eingangs erwähnt, weisen die neuen Verbindungen der allgemeinen Formel (I) und deren Säureadditionssalze wertvolle pharmakologische Eigenschaften auf, neben einer milden blutdrucksenkenden Wirkung insbesondere eine selektive herzfrequenzsenkende Wirkung.
Beispielsweise wurden folgende Verbindungen auf ihre biologischen Eigenschaften untersucht :

1. Wirkung auf die Herzfrequenz an narkotisierten Meerschweinchen : An Meerschweinchen in Urethan-Narkose wurde die Herzfrequenz per Elektrokardiogramm registriert.
Die zu untersuchenden Substanzen wurden in steigenden Dosen zwischen 0,5 und 20 mg/kg 1. v. gege- ben.

2. Wirkung auf die Herzfrequenz an Meerschweinchen-Vorhöfen :
Isolierte spontan schlagende Vorhöfe von Meerschweinchen beider Geschlechter mit einem Körperge- wicht von 300 bis 400 g wurden in einem Organbad in Tyrode-Lösung untersucht. Die Nährlösung wur- de mit Carbogen (95% 02 + 5% C02) versorgt und konstant bei 300C gehalten. DieKontraktionenwur- den isometrisch mittels eines Dehnungsmessstreifens auf einem Grass-Polygraphen registriert.
Die zu untersuchenden Substanzen wurden den Organbädern zugesetzt, so dass Endverdünnungen von 10-5 g/ml entstanden. 5 Vorhöfe pro Substanz.
In der folgenden Tabelle wird die prozentuale Verminderung der Herzfrequenz im Mittel von 5 Vor- höfen bei einer Substanzkonzentration von 10-5 g/ml angegeben.

Zur pharmazeutischen Anwendung lassen sich die Verbindungen der allgemeinen Formel (I) sowie deren physiologisch verträgliche Säureadditionssalze gegebenenfalls in Kombination mit andern Wirksubstanzen in die üblichen galenischen Zubereitungsformen wie Tabletten, Dragées, Pulver, Suspensionen, Lösungen oder Suppositorien einarbeiten. Schmelzpunkt : 170 bis 1720C.
Beispiel 2 : 5, 6-Dimethoxy-2N- {3- [2- (3, 4-dimethoxy)-phenyläthyl-methylamino]-propyl}-phthal- imidin-hydrochlorid
5 g (12,1 Mol) 5, 6-Dimethoxy-2N-{3-[2-(3,4-dimethoxy)-phenyläthylamino]-propyl}-phthalimidin-hydrochlorid werden in einer Mischung von 1, 38 g (30 mMol) Ameisensäure und 1, 5 g (20 mMol) Formalin eine Stunde lang auf 1000C erhitzt. Nach dem Abkühlen wird die Reaktionslösung durch Zugabe von 2n Natronlauge alkalisch gestellt, mit Chloroform extrahiert und die Chloroformphase mit Wasser gewaschen, getrocknet und im Vakuum eingeengt. Man chromatographiert den Rückstandan Kieselgel (Chloroform/Methanol =45/1), engt die Hauptfraktionen ein und fällt die Base aus ätherischer Salzsäure als Hydrochlorid.
Ausbeute : 3,2 g (57% der Theorie).
Schmelzpunkt : 170 bis 172 C.
Beispiel3 :5,6-Dimethoxy-2N-{3-[2-(3,4-dimethoxy)-phenyläthyl-n-propylamino]-propyl}-phthal- imidin-hydrochlorid
Eine Lösung von 2, 5 g (5,5 mMol) 5,6-Dimethoxy-2N-{3-[2-(3,4-dimethoxy)-phenyläthylamino]-propyl}- - phthalimidin in 100 ml Aceton wird nach Zugabe von 20 ml 1-Brompropan und 5 g Kaliumcarbonat 6 h unter Rückfluss erhitzt. Nach dem Abkühlen wird von der Festsubstanz abfiltriert und das Filtrat eingeengt. Man nimmt in Äther auf, filtriert erneut vom Unlöslichen ab und fällt nach dem Einengen das Hydrochlorid aus ätherischer Salzsäure.
