<Desc/Clms Page number 1> Die Erfindung betrifft ein Verfahren zur Gewinnung eines bisher unbekannten Pigmentantibioticums mit cancerostatischer, virostatischer und insbesondere mit neuartiger und überraschender Wirksamkeit gegen verschiedeneErreger von Mycoplasma-Infektionen, das als Violamycin bezeichnet wird. Gegen Tumorkrankheiten und durch Viren hervorgerufene Krankheiten bei Mensch und Nutztier gibt es nur wenige bzw. unzureichende Mittel. Es ist ein Nachteil, dass die gegen Mycoplasma-Erkrankungen eingesetzten Chemotherapeutica keine bakterizide Wirkung auf die Erreger zeigen. Zweck der Erfindung ist die Herstellung eines Heilmittels gegen Tumor-, Virus- und Mycoplasma-Krankheiten. Ihr liegt die Aufgabe zugrunde, ein geeignetes Herstellungsverfahren anzugeben. Es wurde gefunden, dass in den Fermentationslösungen des Stammes IMET JA 6844 ein für die Therapie von neoplastischen, viralen und insbesondere durch Mycoplasme hervorgerufenen Erkrankungen geeignetes neues Antibioticum, das als Violamycin bezeichnet wurde, in guter Ausbeute gebildet wird und unter den im folgenden beschriebenen Verfahrensbedingungen isoliert werden kann. Der Mikroorganismus zur Gewinnung des Violamycins ist in der Stammsammlung des Zentralinstituts für Mikrobiologie und experimentelle Therapie, Jena, der Deutschen Akademie der Wissenschaften zu Berlin unter der Stammsammlungs-Nummer IMET JA 6844 hinterlegt. Der Wildtyp des Bildners wurde 1958 aus einer Erdprobe von derSchwellenburg beiElxleben (Erfurt) isoliert. Der Stamm gehört zur Gattung Streptomyces und entspricht der Diagnose von Streptomyces violaceus (Rossi-Doria 1891, Kraszilnikov 1941, Waksman 1953), wie sie nach Untersuchung des Neo-Typ-Stammes ISP 5082 (= INMI-1, RIA 656, ATCC 15888) im Internationalen Streptomyces Project von Shirling und Gottlieb (Int. J. syst. Bact. 19 : 497 [1969]) gegeben wird. Der Stamm hat folgende Merkmale : Das Substratmycel ist auf den verschiedenen Medien und zu verschiedenen Zeiten rot, violett oder blau gefärbt. Das Pigment, das auch in die Agar-Medien diffundieren kann, hat Indikatoreigen- schaften : sauer-rot, alkalisch-blau. Das Luftmycel ist blassrosa, rosa oder graurosa gefärbt. Die Sporenket- ten sind in mehr oder weniger regelmässigen, meist offenen Spiralen an zum Teil langen Haupthyphen ange- ordnet. Die Sporenoberfläche ist stachelig. Auf Pepton bzw. Tyrosin-halogen Medien diffundiert ein braunes bis blauschwarzes Pigment in den Agar. Der Stamm wächst auf Basalmedium mit Zusatz von D-Glucose, L-Arabinose, D-Xylose, m-Inosit, D-Mannit, D-Fructose, Rhamnose, Saccharose und Raffinose. Das erfindungsgemässe Verfahren besteht darin, dass unter aeroben Kulturbedingungen in einem flüssigen Nährmedium mit entsprechenden Kohlenstoff- und Stickstoffquellen sowie Mineralsalzen der Streptomyces violaceus-Stamm IMET JA 6844 oder seine Mutanten und Varianten gezüchtet werden. Die Züchtung wird in geeigneten Nährmedien bei einer Temperatur von 25 bis 37 C während 2 bis 6 Tagen unter Belüftung und Rührung durchgeführt. Nach Abtrennen des Mycels aus der Kulturlösung wird das Antibioticum Violamycin mit organischen Lösungsmitteln aus dem Kulturfiltrat und Mycel extrahiert. Die Extrakte werden bis zur wässerigen Phase konzentriert, und aus den Rohkonzentraten wird das Antibioticum erneut in die organische Phase überführt und aus den Extraktkonzentraten durch Ausfällen mit einer Fraktion niedrigsiedender Kohlenwasserstoffe gewonnen und durch geeignete chromatographische Verfahren gereinigt. Die Kultivierung des Stammes IMET JA 6844 sowie seiner Mutanten und Varianten erfolgt unter aeroben Bedingungen. An Erde lyophil getrocknetes Sporenmaterial des Stammes wird in geeignete Agarnährböden und nachfolgend in flüssige, vorher sterilisierte Nährmedien geimpft und das entstehende Mycel wird in an sich bekannter Weise bei einer Temperatur zwischen 25 und 370C (vorzugsweise 28 C) über einen Zeitraum von 2 bis 6 Tagen (vorzugsweise 4 Tagen) bei einer Acidität, die zu Beginn des Fermentationsprozesses zwischen PH 6, 2 7. 0 und am Ende zwischen PH 7, 5 und 8, 4 liegt, kultiviert. Das Nährmedium besteht aus Kohlenstoff- und Stickstoffquellen sowie aus anorganischen Salzen. Als Kohlenstoffquellen können Stärke, Glucose, Glycerin, Mannit, Dextrin, Saccharose, Sojaöl und Sojamehl verwendet werden. Als Stickstoffquellen können ausser den oben erwähnten stickstoffhaltigen Substraten auch Trockenhefe, Fleischpepton und Casein in Frage kommen. Gute Fermentationsergebnisse können bei Zusatz von Mineralsalzen erzielt werden. Letztere begünstigen den Verlauf der Fermentation je nach dem verwendeten Nährmedium. In komplexen Medien, die verschiedene Mehle enthalten, haben sich Zusätze von Kalziumkarbonat, Na-Phosphat bzw. K-Phosphat als vorteilhaft erwiesen. Die Fermentation des Bildners kann in Steilbrustflaschen und Rundkolben verschiedenen Inhalts, im Glasfermenter sowie in V2A-Tanks durchgeführt werden. Beide Violamycine unterscheidensich von den bisher bekannten Naturstoffen mit Wirksamkeit gegen Mycophasmen durch ihre mehrhundertfach stärkere bakterizide Wirkung. Eine derartige biologische Aktivität gegen Vertreter der Mycoplasmen, die bei Nutztieren Krankheitssymptome hervorrufen können, ist bisher für Anthracyclin-Antibiotica nicht beschrieben und überraschend. Das neue Antibioticum Violamycin und seine Derivate können als potentielle Chemotherapeutica gegen Mycoplasmen-Erkrankungen von Mensch und Nutztier, gegen Tumor- sowie Viruskrankheiten eingesetztwerden. Die folgenden Beispiele dienen dazu, die Erfindung zu erläutern, ohne sie hierauf zu beschränken. Jede Steilbrustflasche wird mit einer Sporen- und Mycelsuspension beimpft, welche mit steriler, isotonischer Kochsalzlösung von einer im Reagenzglas auf folgendem Anzucht-Nährboden gezüchteten, 10 bis 14 Tage alten Kultur des Violamycin-Bildners hergestellt wird : EMI3.3 <tb> <tb> Hafermehl <SEP> 10/0 <tb> Haferflocken, <SEP> gemahlen <SEP> 1% <tb> Agar-Agar <SEP> 2% <tb> in <SEP> Leitungswasser. <tb> Sterilisation : 35 min bei 120 C, natursauer. Nach 72stündiger Fermentation wird der höchste Gehalt an Violamycin erreicht (125 y/cm3). Mikrobiol. 8 (2), 139). Die Acidität liegt nach dem Sterilisieren bei PH 6, 2. Der höchste Gehalt an Violamycin wird nach 90 h erreicht (100 y /cm3). Beispiel 3 : Mit einer Kultur des Violamycin-Bildners von einem festen Nährboden, wie im Beispiel 1 beschrieben, beimpft man 80 cm3 des im Beispiel 1 angeführten flüssigen Vorzucht-Mediums, welches in einer EMI4.2 einer Schüttelfrequenz von 180 Umdr/min, ehe die so erhaltenen 400 cm3 Kulturbrühe zur Beimpfung von 20 l des im Beispiel 1 beschriebenen Produktionsmediums dienen. Letzteres ist in einem 32 1-Glasfermenter ent- EMI4.3 auf PH 5, 0 eingestellt und nach Separieren des Mycels werden 18 l Kulturfiltrat erhalten. Der Butanolextrakt des Kulturfiltrates wird im Vakuum auf 1 : 10 des Ausgangsvolumens eingeengt und mit dem Butanolextrakt des Mycelextraktkonzentrates vereinigt. Zuvor wird das Mycel mit Methanol zweimal im Verhältnis 1 : 6 ausgerührt. Die vereinigten Extrakte werden unter vermindertem Druck bis auf 1/20 des Methanolvolumens eingeengt und die dabei entstehende wässerige Lösung wird mit Butanol erschöpfend extrahiert. Die vereinten Butanolextrakte werden zusammen mit dem Butanolextrakt des Kulturfiltrates auf 1/25 des Kulturlösungsvolumens ge- bracht. Aus dem Endkonzentrat werden unter Zufügen des 3fachen Volumens an Petroläther 300 g biologisch aktives Rohprodukt (A 6844) pro m3 Kulturlösung erhalten. B e i s p i e l 4: Das Verfahren unterscheidetsich von demjenigen des Beispiels 3 dadurch, dass der ViolamycinBildner nach Vorzucht in einem 2 1-Glaskolben anstatt in einem 32 1-Glasfermenter in einem 400 1-V2A-Tank überimpft wird, der das im Beispiel 1 beschriebene Produktionsmedium enthält. DieRührgeschwindigkeitbeträgt 280 Umdr/min und die Luftzufuhr wird auf 400 l/min eingestellt. Aus 300 1 Kulturlösung werden nach dem in Beispiel 3 beschriebenen Verfahren 300 g Rohprodukt A 6844/m 3 gewonnen. Das Rohprodukt A 6844 wird für die Gewinnung der biologisch aktiven Komplexe Violamycin A und B benutzt. Beispiel 5 : 200 g Rohprodukt A 6844 werden in 5 l Chloroform suspendiert und nach 1stündigem Rühren wird die Lösung vom unlöslichen Rückstand abgetrennt. Die Chloroformlösung wird 5-bis 6mal mit der Hälfte des Volumens Wasser gewaschen und dadurch von wasserlöslichen Pigmenten vollständig befreit. Nach dem Trocknen wird die Chloroformlösung unter vermindertem Druck auf 1/50 desAusgangsvolumens konzentriert und mit der 10fachen Menge an Petroläther verrührt. Dabei fällt die biologisch aktive Violamycin-Fraktion A aus. Die Fraktion A wird abfiltriert, mit reinem Petroläther gewaschen und unter vermindertem Druck im Exsikkator über konz. Schwefelsäure getrocknet. Die Ausbeute an Fraktion A beträgt 16 g. Die Violamycin-Fraktion A wird säulenchromatographisch in das Antibioticum Violamycin A und den biologisch inaktivenChromophorkomplex Violamycin A getrennt. Aus 10 g Violamycin-Fraktion A werden 5 g Violamycin A erhalten. Die Violamycin-Fraktion B ist aus dem Waschwasser des Chloroformextraktes nach Einstellen der Acidität auf pH 8, 5 durch Butanolextraktion zu gewinnen. Nach Konzentrieren der Butanolextrakte und nach deren Verrühren mit Petroläther wird der entstehende Niederschlag abfiltriert und getrocknet. Claims (1)
- PATENTANSPRUCH : Verfahren zur Gewinnung des neuen Antibioticums Violamycin sowie seines-Aglycons durch Züchtung eines Streptomyceten unter aeroben Kulturbedingungen in einem flüssigen Nährmedium mit entsprechenden KohlenStoff- und Stickstoffquellen sowie Mineralsalzen, d a d u r c h g e k e n n z e i c h n e t, dass Streptomyces violaceus IMET JA 6844 oder seine Mutanten und Varianten eingesetzt werden, die Züchtung in geeigneten Nährmedien bei Temperaturen von 25 bis 370C während 2 bis 6 Tagen unter Belüftung und Rührung durchgeführtwird, das gebildete Antibioticum durch Extraktion aus dem Kulturfiltrat und dem Mycel gewonnen, gereinigt und in seine Komponenten getrennt wird.
