PATENTANSPRUCH
Verfahren zur Herstellung des neuen Antibiotikums S 53210/A-III, dadurch gekennzeichnet, dass man einen Stamm von Microellobosporia brunea nov. Spez. auf oder in einem Nährmedium züchtet und das Antibiotikum aus der Fermentationsbrühe durch extraktive und/oder adsorptive Methoden isoliert und hierauf chromatographisch oder durch Gegenstromverteilung reinigt.
Die vorliegende Erfindung betrifft Verfahren zur Herstellung des neuen Antibiotikums S 53210/A-III.
Erfindungsgemäss gelangt man zu dem neuen Antibiotikum S 53210/A-III, indem man einen Stamm von Microellobosporia brunea nov. Spez. auf oder in einem Nährmedium züchtet und das Antibiotikum aus der Fermentationsbrühe durch extraktive und/oder adsorptive Methoden isoliert und hierauf chromatographisch oder durch Gegenstromverteilung reinigt.
Der neue erfindungsgemäss verwendete Stamm S 53210/A wurde aus Erde isoliert, die aus einem antiken, etwa 2000 Jahre alten, 1976 bei Jama Coaque (Equador) gefundenen Tonkopf herausgekratzt worden war. Eine Probe davon wurde beim United States Department of Agriculture, Peoria, Ill., USA deponiert und steht der Öffentlichkeit unter der Kulturnummer NRRL 11,122 zur Verfügung.
Da der Stamm S 53210/A zuerst nur Substratmycel mit seitlichen Einzel- und Doppelsporen bildete, wurde seine Zugehörigkeit zur Gattung Micromonospora vermutet. Auf einigen Kulturmedien bildet der Stamm jedoch nach zweiwöchiger Inkubation Luftmycel und keulenförmige, 2 bis 7 ,um lange Sporangien, die ihrerseits ein bis sieben, kugelige bis ellipsoidische, 1,0-2,7,um grosse, unbewegliche Sporen enthalten. Es scheint, dass die Sporen durch Septierung innerhalb kurzer, keulenförmiger Seitenzweige zu entstehen beginnen; die einzelnen Sporen vergrössern sich unterschiedlich stark, reifen als kurze Kette im Sporangium und werden durch apikales Aufreissen der Sporangienhülle befreit. Lange, streptomycetenartige Sporenketten wurden keine beobachtet.
Aufgrund der oben genannten Beschreibung stimmt der Stamm S 53210/A gut überein mit der von Cross T., Lechevalier M. & Lechevalier H., 1963, A new genus of Actinomycetales: Microellobosporia gen. nov . Journ. Gen. Microbiology 31,421 (1963), errichteten Gattung Microellobosporia. Diese Gattung umfasst bisher nur vier Arten (vgl. Bergey's Manual of Determinative Bacteriology, 8th Edition (1974), Seiten 843-845), wobei M. cinerea und M. violacea violette Farbtöne im vegetativen Mycel aufweisen und die Sporangien von M. grisea mit kristallinen Stachelchen überzogen sind. Der Stamm S 53210/A weist diese auffallenden Merkmale nicht auf.
Die vierte bisher beschriebene Art, M. flavea, hat aufgrund der Beschreibung weisses Luftmycel und bildet strohgelbe Kolonien ohne lösliche Pigmente, so dass der Stamm S 53210/A mit mit weissem Luftmycel und beige bis braunen Kolonien ohne lösliche Pigmente jener letzten Art am nächsten kommt. Da aber zwischen M. flavea und dem neuen Stamm S 53210/A bezüglich der Kohlenstoffverwertung erhebliche Unterschiede bestehen, wurde der Stamm S53210/Aals neue Art der Gattung Microellobosporia aufgefasst und aufgrund des braunen, vegetativen Miycels Microellobosporia brunea, nov. Sp. genannt.

Die Züchtung des Stammes S 53210/A erfolgt durch Anwendung an sich bekannter Methoden und besteht im wesentlichen darin, den genannten Stamm auf oder in einem geeigneten Medium und unter geeigneten Bedingungen zu züchten und anschliessend das im Laufe der Züchtung gebildete Antibiotikum S 53210/A-III zu isolieren.
Zu Beginn der Züchtung kann der pH-Wert des Fermenta
tionsmediums zwischen 6,5 und 7,8 liegen. Die zur Züchtung optimale Temperatur kann zwischen 25 und 35 "C liegen. Die Belüftung der Züchtung kann in weiten Grenzwerten schwanken. Die maximale Ausbeute an Antibiotikum S 53210/A-III wird nach 4- bis 7tägiger Züchtung erhalten. Für die Beimpfung der Fermentationsmedien kann eine sporulierende Kultur verwendet werden. Die Fermentationsmedien können hauptsächlich eine assimilierbare Kohlenstoffquelle, eine assimilierbare Stickstoffquelle und gegebenenfalls Mineralsalze und Wachstumsfaktoren enthalten, wobei alle diese Elemente in Form von gut definierten Produkten oder von komplexen Gemischen, wie man sie in biologischen Produkten verschiedenen Ursprungs antrifft, zugeführt werden können.
Weiterhin eignen sich organische und anorganische stickstoffhaltige Verbindungen, wie Pepton, Hefe- oder Fleischextrakte, Ammoniumnitrat, Ammoniumsulfat, Aminosäuren usw., als Stickstoffquelle. Spurenelemente, die für ein optimales Wachstum des Organismus, der zur Produktion des Antibiotikums S 53210/A-III verwendet wird, erforderlich sind, sollen ebenfalls in dem Kulturmedium enthalten sein. Solche Spurenelemente kommen gewöhnlich als Verunreinigungen in den anderen Bestandteilen des Mediums in Mengen vor, die für den Wachstumsbedarf des erfindungsgemäss verwendeten Stammes S 53210/A genügen.
Sobald bei der Züchtung eine maximale Menge des Antibiotikums S 53210/A-III produziert worden ist, was z. B. durch die Aktivität gegen Staphylococcus aureus festgestellt werden kann, wird der Mycelanteil in der Kulturbrühe homogenisiert und das Antibiotikum durch Extraktion mit einem mit Wasser nicht mischbaren Lösungsmittel wie z. B. Äthylacetat, Butylacetat, n-Butanol, gewonnen.
Aus dem erhaltenen Rohextrakt kann das Antibiotikum S 53210/A-III durch an sich bekannte chromatographische Methoden isoliert und rein dargestellt werden.
Das neue Antibiotikum S 53210/A-III zeigt folgende Eigenschaften: Gelbes amorphes Pulver Smp. > 300 (Zers.) [a0 = + 67,2" (c = 1,444 in Chloroform) Analyse: Gef. C52,0 H5,0 N12,0 020,0 S11,0'/6 UV-Spektrum in Acetonitril, vgl. Fig. 1 IR-Spektrum in KBr, vgl. Fig. 2 'H-NMR-Spektrum in DMSO, 90 MHz mit Tetramethylsilan als interner Standard, vgl. Fig. 3.
Bei der Aminosäureanalyse wurde neben unbekannten Aminosäuren eine Aminosäure gefunden, die die gleiche Retentionszeit wie Threonin aufweist.
Farbreaktion: Ninhydrin negativ Clt-Benzidin grün
Löslichkeit: S 53210/A-III ist leicht löslich in Chloroform, Dimethylformamid, Acetonitril, Dioxan, Dimethylsulfoxid, schwerlöslich in Methanol, Äthanol und unlöslich in Wasser und Hexan.
In der Tabelle 1 sind die RrWerte von S 53210/A-III im Dünnschichtchromatogramm angegeben (Laufstrecke 14 cm auf Kieselgel Merck 6(rPlatten, Schichtdicke 0,25 mm).

Als Nachweisreagens kann Jod verwendet werden. Beim Ansprühen mit einer 0,2%igen Ce(SO4)2-Lösung in 500/oiger Schwefelsäure und Erwärmen auf 1300 gibt S 53210/A-III eine graue bis braune Anfärbung.
Das Antibiotikum S 53210/A-III ist antimikrobiell wirksam; insbesondere hemmt es vorwiegend das Wachstum von grampositiven Bakterien. Es zeigt eine sehr breite Aktivität gegen grampositive Bakterien, aber keine Wirkung gegen Hefen und Pilze.
Die Wirkung erstreckt sich auch auf die pathogenen Vertreter der grampositiven Bakterien, wie Staphylococcen, Streptokokken, Corynebakterien, Mycobakterien. Auch gegen Mycoplasmen und Neisserien ist das neue Antibiotikum wirksam.
Im Reihenverdünnungstest wurden folgende Mindesthemmkonzentrationen erhalten (mcg/ml): Organismus MIC 5. aureus 0,01 S. aureus res. Penicillin 0,01 S. aureus res. Tetracyclin 0,1 S. aureus res. Das neue Antibiotikum S 53210/A-III kann für humanmedizinische Zwecke als Arzneimittel allein, und zwar in reiner Form oder in den entsprechenden Arzneiformen für topicale, enterale oder parenterale Verabreichung verwendet werden.
In den nachfolgenden Beispielen, welche die Ausführung des Verfahrens erläutern, den Umfang der Erfindung aber in keiner Weise einschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden.
Beispiel 1: Züchtung des Stammes S 53210/A in einer Schüttelkultur a) Agar-Ausgangskultur
Die als Ausgangsmaterial verwendete Agarkultur des Stammes S 53210/A erhält man, indem man ein Kulturmedium (I) folgender Zusammensetzung Liter Cerelose (Glucose) 4,0 Malz-Extrakt 10,0 Hefe-Extrakt 4,0 Agar (Bacto) 20,0 destilliertes Wasser 1 Liter mit einer auf an sich bekannten Weise hergestellten Sporen suspension des ursprünglich isolierten Stammes S 53210/A beimpft. Das Medium wird vor der Sterilisation mit NaOH auf pH 8,3 bis 8,6 eingestellt und beträgt nach der Sterilisation (20 Minuten bei 1200)6,9 bis 7,3.
b) Sporensuspension
Eine gut sporulierende Agar-Ausgangskultur des Stammes S 53210/A wird mit 5 ml steriler,0,9%iger Kochsalzlösung versetzt, was eine dichte Sporen- und Mycelsuspension ergibt.
c) Vorkulturen
Von dieser Sporensuspension wird 1 ml zur Beimpfung eines 200.ml-Erlenmeyer-Kolbens verwendet, der 50 ml des folgenden Vorkulturmediums (II) enthält: Liter Dextrin 10,0 Glucose 10,0 Pepton 5,0 Hefeextrakt 5,0 CaC03 1,0 destilliertes Wasser 1 Liter
Der pH-Wert wird mit NaOH auf 7,2 eingestellt. Das Vorkulturmedium wird in einem Autoklav bei 1200 während 20 Minuten sterilisiert, worauf der pH-Wert 7,0 bis 7,2 beträgt.
Die so angesetzte Vorkultur wird während drei Tagen bei 27 auf einer Rundschüttelmaschine (220 U/Min.) aerob inkubiert und wird zur Beimpfung einer zweiten Vorkultur verwendet, indem 5 ml der ersten Vorkultur it inem 500-ml-Erlenmeyer Kolben mit 100 ml des folgenden Mediums (III) geimpft werden: Liter Fleischextrakt 3,0 Trypton 5,0 Glucose 1,0 Stärke löslich 24,0 Hefeextrakt 5,0 CaC03 2,0 destilliertes Wasser 1 Liter
Der pH-Wert wird mit NaOH auf 7,2 eingestellt und das Medium in einem Autoklav bei 1200 während 20 Minuten sterilisiert, worauf der pH-Wert 7,0 bis 7,2 beträgt.
d) Hauptkultur
Die zweite Vorkultur wird während 3 Tagen bei 27 auf einer Rundschüttelmaschine (220 U/Min.) aerob inkubiert und wird dann direkt zur Beimpfung einer Hauptkultur verwendet, indem 5 ml der zweiten Vorkultur in einem 500-ml-Erlenmeyer Kolben mit 100 ml des folgenden Mediums (IV) geimpft werden: Liter Malzextrakt 30,0 Blutpepton 7,5 destilliertes Wasser 1 Liter
Der pH-Wert wird mit NaOH auf 7,0 eingestellt und das Medium in einem Autoklav bei 1200 während 20 Minuten sterilisiert, worauf der pH-Wert 6,8 bis 7,0 beträgt. Die so angesetzte Hauptkultur wird während 5 Tagen bei 27 auf einer Rundschüttelmaschine (220 U/Min.) inkubiert.
Beispiel 2: Züchtung des Stammes S 53210/A in einem Fermenter
Die Ausgangskultur für die Züchtung eines 500-Liter-Fermentationsansatzes ist eine Standkultur (Erlenmeyerkolben zu 500 ml mit 100 ml Agar) des ursprünglich isolierten Stammes S 53210/A. Diese Kultur mit dem Agarmedium, das pro Liter 20 g Agar, 20 g Malzextrakt, 4 g Hefeextrakt und entmineralisiertes Wasser enthält, wird 14 Tage bei 33 bebrütet. Mit 200 ml Sporen- und Mycelsuspension aus 2 Standkulturen wird 1 Glasfermenter mit 10 Liter Nährlösung der folgenden Zusammensetzung angeimpft: 20 g Glukose, 5 g Pepton, 5 g Hefeextrakt, 1 g CaCO3 und entmineralisiertes Wasser ad 1 Liter.
Diese Vorkultur wird 4 Tage bei 30 mit einer Luftrate von 0,7 Liter V/V/Min. und einer Rührerdrehzahl von 250 UPM inkubiert. Die Zwischenkultur im 75-Liter-Stahlfermenter (50 Liter Gärvolumen) wird mit 7 Liter aus der Vorkultur angeimpft, das Medium hat die gleiche Zusammensetzung wie die Vorkultur: 20 g Glucose, 5 g Pepton, 5 g Hefeextrakt, 1 g CaC03 und entmineralisiertes Wasser ad 1 Liter. Die Inkubation erfolgt 3 Tage bei 30 mit einer Luftrate von 0,7 Liter VW/Min. bei 0,5 bar Druck und einer Rührerdrehzahl von 200 UPM.
Die Hauptstufen-Fermentation im 750-Liter-Gärtank (500 Liter Gärvolumen) erfolgt bei 30 mit einer Nährlösung folgender Zusammensetzung: pro Liter 30 g Malzextrakt, 7,5 g Blutpepton, 1 mg FeSO4- 7 HzO, 1 mg MnCk-4 HzO, 1 mg ZnS0 .7 H20 und entmineralisiertes Wasser. Die Inkubation dauert 4 Tage bei einer Rührerdrehzahl von 130 UPM und einer Luftrate von 0,7 Liter VlV/Min. und bei 0,5 bar Druck. Der pH-Endwert liegt bei 7,8.
Beispiel 3: Isolierung von S 53210/A-III
500 Liter Kulturbrühe werden mit 2N Schwefelsäure auf pH 7 gestellt, mit 500 Liter Essigester versetzt und 90 Minuten mit einem Dispax-Reaktor homogenisiert. Die Abtrennung der organischen Phase erfolgt mit einem Westfalia-Trennseparator. Der Extrakt wird nach Waschen mit 100 Liter Wasser im Vakuum bei 20 bis 40 Konzentrattemperatur zur Trockene eingeengt (146 g Rohextrakt). Die Wiederholung der Extraktion ergab nochmals 51 g Rohextrakt.
Die 197 g vereinigten Extrakte werden unter Rühren zu 2 Liter Hexan gegeben. Die überstehende Lösung wird abdekantiert und der ölige Rückstand im Vakuum bei 50 vom Lösungsmittel befreit, wobei 80,2 g Fällungsprodukt mit Aktivität gegen Staphylococcus aureus anfallen. Dieses Material wird in Chloroform-Methanol (1:1) gelöst und die Lösung auf eine mit Chloroform-Methanol (1:1) bereitete Säule von 1 kg Sephadex LH2o gebracht. Die Elution mit Chloroform-Methanol (1:1) liefert 14,5 g Fraktionen, die S 53210/A-III enthalten. Diese werden in Methylenchlorid + 3% Methanol gelöst und die Lösung auf eine mit Methylenchlorid + 3% Methanol bereitete Säule von 250 g Kieselgel Merck (Korngrösse 0,063-0,2 mm) aufgegeben.
Die Elution erfolgt zuerst mit Methylenchlorid unter Zusatz von 3 bis 5% Methanol, dann mit Methylenchlorid + 10% Methanol. Man erhält 4,8 g angereicherte Fraktionen von S 53210/ A-III. Die Fraktionen werden in 25 ml Methylenchlorid gelöst und nach Zugabe von 50 ml Methanol bei Raumtemperatur eingeengt, wobei S 53210/A-III ausfällt. Die Substanz wird abfiltriert und mit Methanol und Äther gewaschen. Man erhält S 53210/A-III als amorphes gelbes Pulver. Smp. > 300 nach 15 Stunden Trocknen im Hochvakuum bei Raumtemperatur.
Die Reinheitsprüfung der gegen Staphylococcus aureus aktiven Chromatographiefraktionen erfolgt dünnschichtchromatographisch auf Kieselgel mit Methylenchlorid-Methanol Wasser (88:11:1) und Methylenchlorid-Methanol-Wasser (92:7,5:0,5) als Fliessmittel.

Since the S 53210 / A strain initially only formed substrate mycelium with lateral single and double spores, it was assumed that it belongs to the Micromonospora genus. On some culture media, however, after two weeks of incubation, the strain forms aerial mycelium and club-shaped, 2 to 7, around long sporangia, which in turn contain one to seven, spherical to ellipsoid, 1.0-2.7, around large, immobile spores. It appears that the spores begin to form through short, club-shaped side branches; the individual spores enlarge to different extents, mature as a short chain in the sporangium and are released by apical tearing of the sporangial covering. No long, streptomycete-like spore chains were observed.
Based on the above description, the strain S 53210 / A agrees well with that of Cross T., Lechevalier M. & Lechevalier H., 1963, A new genus of Actinomycetales: Microellobosporia gen. Nov. Journ. Gene. Microbiology 31, 421 (1963), established the genus Microellobosporia. So far, this genus only includes four species (see Bergey's Manual of Determinative Bacteriology, 8th Edition (1974), pages 843-845), whereby M. cinerea and M. violacea have violet shades in vegetative mycelium and the sporangia of M. grisea with crystalline spikes are covered. The strain S 53210 / A does not have these striking features.
The fourth species described so far, M. flavea, has white aerial mycelium due to the description and forms straw-yellow colonies without soluble pigments, so that strain S 53210 / A with white aerial mycelium and beige to brown colonies without soluble pigments comes closest to the last species . The fractions are dissolved in 25 ml of methylene chloride and, after addition of 50 ml of methanol, concentrated at room temperature, S 53210 / A-III precipitating. The substance is filtered off and washed with methanol and ether. S 53210 / A-III is obtained as an amorphous yellow powder. Mp> 300 after 15 hours of drying in a high vacuum at room temperature.
