Abstract

La presente se refiere a moléculas superagonistas de unión a antígeno con diana tumoral capaces de unión multivalente a CD28, a métodos para su producción, a composiciones que contienen dichos anticuerpos y a métodos de utilización de los mismos. Reivindicación 1: Una molécula superagonista de unión al antígeno CD28, caracterizada porque es capaz de unión bivalente a CD28 y que comprende: (a) dos o más dominios de unión a antígeno capaces de unión específica a CD28, (b) por lo menos un dominio de unión a antígeno capaz de unión específica a un antígeno asociado a tumor, y (c) un dominio Fc compuesto de una primera y una segunda subunidades capaces de asociación estable, que comprende una o más sustituciones de aminoácidos que reducen la afinidad de unión de la molécula de unión a antígeno a un receptor de Fc y/o la función efectora. Reivindicación 23: La molécula superagonista de unión al antígeno CD28 según cualquiera de las reivindicaciones 1 a 17, o la composición farmacéutica según la reivindicación 21 caracterizada porque se usa en el tratamiento del cáncer.
