Abstract
화장료 조성물에 관한 것으로, 천연물을 베타인 및 구연산의 공융 용매를 이용하여 추출함으로써 유기 용매 추출물과 비교하여 세포 독성이 낮을 뿐만 아니라, 추출물의 변색을 방지하여 안전성 및 안정성이 개선되었는 바, 피부 상태 개선용 화장품 및 피부 외용제로 유용하게 사용될 수 있다.
Description
화장료 조성물에 관한 것이다.
일반적으로 액상 추출에는 유기 용매가 사용된다. 유기 용매는 값이 싸고 추출 효율이 좋다는 장점이 있으나, 사용자의 건강에 해롭거나 또는 환경 오염의 문제를 야기할 수 있다는 문제점이 있다. 이에, 지속 가능한 발전에 대한 관심이 높아지며 유기 용매를 대체할 수 있는 추출 용매와 관련된 연구가 활발히 진행되고 있다.
그 중에서도 공융 용매(Eutectic solvent)가 대안으로 제시되고 있다. 공융 용매란 두 종류의 성분이 혼합되어 각 성분보다 더 낮은 녹는점을 나타내는 공융 혼합물이다. 공융 용매의 개발 단계 초기에는 이온성 액체(ionic liquids)를 활용하여 유기 용매의 대안으로 사용하였으나, 낮은 생분해성, 독성, 및 제조 공정 상 배출되는 오염 물질의 양 등에 의하여 환경 친화적인 용매로서 부적법하다는 보고가 있다. 이후, 종래 이온성 액체의 단점을 보완한 깊은 공융 용매(Deep eutectic solvent), 천연 공융 용매(Natural eutectic solvent) 등에 대한 연구가 진행되고 있다. 그러나, 깊은 공융 용매, 천연 공융 용매는 염화콜린(Choline chloride)과 같이 화장품에 사용될 수 없는 성분이 다수 포함되어 있을 뿐만 아니라, 높은 점도를 갖고, 공융 용매 제조 공정 중 용매가 변색되는 등의 문제점으로 인하여 화장품 및 피부외용제로 적용하기에 한계가 있다.
따라서, 상기와 같은 문제점을 해결하기 위하여, 친환경 천연 공융 용매를 개발할 필요가 있다.
일 양상은 친환경 천연 공융 용매로 추출한 천연물 추출물을 유효성분으로 포함하는 피부 상태 개선용 조성물을 제공하는 것이다.
다른 양상은 친환경 천연 공융 용매를 제조하는 방법을 제공하는 것이다.
또 다른 양상은 피부 상태를 개선하기 위한 화장료 조성물의 제조를 위한, 천연 공융 용매로 추출한 천연물 추출물의 용도로서, 상기 천연 공융 용매는 아미노산 또는 이의 유도체, 및 유기산을 포함하는 것인 용도를 제공하는 것이다.
또 다른 양상은 피부 상태를 개선하기 위한 피부 외용제 조성물의 제조를 위한, 천연 공융 용매로 추출한 천연물 추출물의 용도로서, 상기 천연 공융 용매는 아미노산 또는 이의 유도체, 및 유기산을 포함하는 것인 용도를 제공하는 것이다.
또 다른 양상은 피부 염증 질환의 예방 또는 치료를 위한 약학적 조성물의 제조를 위한, 천연 공융 용매로 추출한 천연물 추출물의 용도로서, 상기 천연 공융 용매는 아미노산 또는 이의 유도체, 및 유기산을 포함하는 것인 용도를 제공하는 것이다.
또 다른 양상은 탈모 예방 또는 치료, 또는 발모 또는 육모 촉진을 위한 약학적 조성물의 제조를 위한, 천연 공융 용매로 추출한 천연물 추출물의 용도로서, 상기 천연 공융 용매는 아미노산 또는 이의 유도체, 및 유기산을 포함하는 것인 용도를 제공하는 것이다.
또 다른 양상은 유효한 양의 상기한 조성물을 그를 필요로 하는 개체에 처리 또는 투여하는 단계를 포함하는 개체의 상태를 예방, 개선, 또는 치료하는 방법을 제공한다.
일 양상은 친환경 천연 공융 용매로 추출한 천연물 추출물을 유효성분으로 포함하는 피부 상태 개선용 조성물을 제공한다.
상기 천연 공융 용매는 아미노산 또는 이의 유도체, 및 유기산으로 구성된 군에서 선택되는 어느 하나 이상인 것일 수 있다.
상기 아미노산 또는 이의 유도체는 예를 들어, 베타인, 알라닌, 시스테인, 아스파르트산, 글루타민, 페닐알라닌, 글리신, 히스티딘, 이소로이신, 라이신, 로이신, 메티오닌, 아스파라긴, 프롤린, 글루타민, 아르기닌, 세린, 트레오닌, 발린, 트립토판, 티로신 등일 수 있다.
또한, 상기 유기산은 예를 들어, 구연산, 락트산, 말산, 말레산, 피루브산, 푸마르산, 석신산, 아세트산, 아코니트산, 타트르산, 아스코르브산, 말론산, 옥살산, 글루쿠론산, 뉴라민산, 시알산, 시킴산, 피트산, 갈락투론산, 이두론산, 히알루론산, 하이드록시시트르산, 락톤 유도체 등일 수 있다.
일 구체예에 있어서, 상기 천연 공융 용매는 베타인 및 구연산의 혼합물일 수 있다. 상기 베타인 및 구연산은 1:0.001 내지 10의 중량비로 혼합될 수 있다. 상기 베타인 및 구연산은 예를 들어, 1:0.001 내지 10, 1:001 내지 8, 1: 0.05 내지 7, 1:0.05 내지 5, 1:0.01 내지 10, 1:0.01 내지 6, 1:0.1 내지 4, 1:0.1 내지 3, 1:1 내지 10, 1:1 내지 4, 또는 1:1 내지 2의 중량비로 혼합될 수 있다. 이때, 상기 베타인 및 구연산의 혼합 중량비가 상기 범위 미만이거나 또는 상기 범위를 초과하는 경우, 베타인 및 구연산이 침전되거나, 공융 용매가 현탁해지거나 또는 변색되는 문제점이 있다.

또한, 다른 양상은 베타인 및 구연산을 혼합하여 혼합물을 제조하는 단계; 상기 혼합물에 시트르산삼나트륨(sodium citrate), 수산화나트륨(sodium hydroxide), 수산화칼륨(potassium hydroxide), 중탄산나트륨(sodium bicarbonate), 아르기닌(arginine), 트로메타민(tromethamine), 트리에탄올아민(triethanol amine), 아미노메틸프로판올(aminomethyl propanol), 디포타슘포스페이트(dipotassium phosphate), 포타슘포스페이트(potassium phosphate), 아니스산나트륨(sodium anisate), 젖산나트륨(sodium lactate), 탄산나트륨(sodium carbonate), 및 요소(urea)로 이루어진 군에서 선택되는 어느 하나를 추가하여 pH를 조절하는 단계를 포함하는, 천연 공융 용매의 제조방법을 제공한다.
또한, 다른 양상은 천연물을 아미노산, 유기산 또는 이들의 혼합물로 추출하여 추출물을 제조하는 단계를 포함하는 천연 공융 용매로 추출한 천연물 추출물의 제조방법을 제공한다. 상기 천연물, 천연 공융 용매 등의 구체적인 내용은 전술한 바와 같다.
상기 방법에 따라 제조된 천연물 추출물은 활성산소 및 자유라디컬 소거율이 우수하고, 멜라닌 합성을 억제하여, SPT, AQP3, RNase7의 발현을 증가시킴으로써, 피부 장벽 강화, 피부 보습, 탄력, 피부 노화 개선 등과 같은 피부 상태 개선에 유용하게 사용될 수 있다. 또한, 염증성사이토카인의 발현을 감소시킴으로써 피부 염증 질환의 예방, 개선 또는 치료에 유용하게 사용될 수 있다.
일 양상에 따른 화장료 조성물은 천연물을 베타인 및 구연산의 공융 용매를 이용하여 추출함으로써 유기 용매 추출물과 비교하여 세포 독성이 낮을 뿐만 아니라, 추출물의 변색을 방지하여 안전성 및 안정성이 개선되었는 바, 피부 상태 개선용 화장품 및 피부 외용제로 유용하게 사용될 수 있다.
이하, 본 발명의 이해를 돕기 위하여 바람직한 실시예를 제시한다. 그러나 하기의 실시예는 본 발명을 보다 쉽게 이해하기 위하여 제공되는 것일 뿐, 하기 실시예에 의해 본 발명의 내용이 한정되는 것은 아니다.
[제조예]
제조예 1. 아미노산 및 유기산을 포함하는 천연 공융 용매의 제조
아미노산으로서 베타인(betaine) 및 유기산으로서 구연산(citric acid)를 포함하는 천연 공융 용매를 제조하였다. 구체적으로, 베타인과 구연산을 동일한 함량으로 혼합한 후, 60100 ℃에서 150 rpm으로 2시간 동안 교반하여 pH 2인 천연 공융 용매를 제조하였다. .
제조예 2~4. pH가 조절된 천연 공융 용매의 제조
pH가 조절된 천연 공융 용매를 제조하였다. 구체적으로, 상기 제조예 1의 천연 공융 용매에 1M 시트르산 나트륨(sodium citrate) 수용액을 각각 1, 5 및 10 ㎏씩 첨가하여 pH 3(제조예 2), pH 5(제조예 3) 및 pH 7(제조예 4)의 천연 공융 용매를 제조하였다.
[실시예]
실시예 1. 무화과 열매 공융 용매 추출물의 제조
무화과 열매(Ficus carica fruit) 천연 공융 용매 추출물을 제조하였다. 구체적으로, 건조 및 분쇄한 무화과 열매 1㎏을 제조예 1의 천연 공융 용매로 6시간 동안 추출하였다. 이후, 와트만(Whatman) #5 여과지로 여과하여 무화과 열매 공융 용매 추출물을 제조하였다.
실시예 2. 권백 공융 용매 추출물의 제조
권백(Selaginella tamariscine)을 사용하였다는 점을 제외하고는 상기 실시예 1과 동일한 방법으로 권백 공융 용매 추출물을 제조하였다.
실시예 3. 발아 흑미 공융 용매 추출물의 제조
발아 흑미(Oryza sativa)를 사용하였다는 점을 제외하고는 상기 실시예 1과 동일한 방법으로 발아 흑미 공융 용매 추출물으 제조하였다.
실시예 4. 혈갈 공융 용매 추출물의 제조
혈갈(Daemonorops draco)을 사용하였다는 점을 제외하고는 상기 실시예 1과 동일한 방법으로 혈갈 공융 용매 추출물을 제조하였다.
실시예 5. 무화과 열매, 권백, 발아 흑미 및 혈갈 혼합물의 공융 용매 추출물의 제조
무화과 열매, 권백, 발아 흑미, 및 혈갈을 동일한 함량으로 사용하였다는 점을 제외하고는 상기 실시예 1과 동일한 방법으로 무화과 열매, 권백, 발아 흑미 및 혈간 혼합물의 공융 용매 추출물을 제조하였다.
실시예 6~8. pH 조절형 무화과 열매 천연 공융 용매 추출물의 제조
제조예 2~4의 천연 공융 용매를 사용하였다는 점을 제외하고는 상기 실시예 1과 동일한 방법으로 pH 조절형 무화과 열매 천연 공융 용매 추출물을 제조하였다.
실시예 9~11. pH 조절형 권백 천연 공융 용매 추출물의 제조
제조예 2~4의 천연 공융 용매를 사용하였다는 점을 제외하고는 상기 실시예 1과 동일한 방법으로 pH 조절형 권백 천연 공융 용매 추출물을 제조하였다.
실시예 12~14. pH 조절형 발아 흑미 공융 용매 추출물의 제조
제조예 2~4의 천연 공융 용매를 사용하였다는 점을 제외하고는 상기 실시예 1과 동일한 방법으로 pH 조절형 발아 흑미 공융 용매 추출물을 사용하였다.
제조예 15~17. pH 조절형 혈갈 천연 공융 용매 추출물의 제조
제조예 2~4의 천연 공융 용매를 사용하였다는 점을 제외하고는 상기 실시예 1과 동일한 방법으로 pH 조절형 혈갈 천연 공융 용매 추출물을 제조하였다.
제조예 18~20. pH 조절형 무화과 열매, 권백, 발아 흑미 및 혈갈 혼합물의 공융 용매 추출물의 제조
제조예 2~4의 천연 공융 용매를 사용하였다는 점을 제외하고는 상기 실시예 1과 동일한 방법으로 무화과 열매, 권백, 발아 흑미 및 혈간 혼합물의 공융 용매 추출물을 제조하였다.
[비교예]
비교예 1. 무화과 열매 에탄올 추출물의 제조
70%(v/v) 에탄올을 사용하였다는 점을 제외하고는 상기 실시예 1과 동일한 방법으로 무화과 열매 에탄올 추출물을 제조하였다.
비교예 2. 권백 에탄올 추출물의 제조
70%(v/v) 에탄올을 사용하였다는 점을 제외하고는 상기 실시예 1과 동일한 방법으로 권백 에탄올 추출물을 제조하였다.
비교예 3. 발아 흑미 에탄올 추출물의 제조
70%(v/v) 에탄올을 사용하였다는 점을 제외하고는 상기 실시예 1과 동일한 방법으로 발아 흑미 에탄올 추출물을 제조하였다.
비교예 4. 혈갈 에탄올 추출물의 제조
70%(v/v) 에탄올을 사용하였다는 점을 제외하고는 상기 실시예 1과 동일한 방법으로 혈갈 에탄올 추출물을 제조하였다.
[실험예]
실험예 1. 활성 성분 함량 확인
일 구체예에 따른 천연 공융 용매 추출물에 포함된 페놀 및 플라보노이드의 함량을 확인하였다. 구체적으로, 증류수 10 ㎖에 상기 실시예 1~20 및 비교예 1~4를 각각 1 ㎖씩 첨가한 후, 2 ㎖의 Folin-Ciocalteu phenol reagent(Sigma)를 첨가하여 혼합하였다. 이후, 실온에서 5분 동안 반응시킨 뒤, 20% 소듐 카보네이트(Sodium Carbonate) 2 ㎖를 첨가하여 상온에서 1시간 동안 추가로 반응시켰다. 680 ㎚에서 흡광도를 측정하여 총 페놀 함량을 측정하였다. 지표물질로 갈릭산(gallic acid)를 사용하였다.
다음으로, 상기 실시예 1~20 및 비교예 1~4 각각 1.5 ㎖과 동량의 메탄올에 용해된 2% AlCl3H2O를 혼합한 뒤, 상온에서 10분 동안 반응시켰다. 이후, 367 ㎚에서 흡광도를 측정하여 총 플라보노이드 함량을 측정하였다. 지표 물질로 카테킨(catechin)을 사용하였다.

그 결과, 표 1에 나타낸 바와 같이, pH가 조절된 실시예 6~20의 경우, pH가 조절되지 않은 실시예 1~5와 비교하여 총 폴리페놀 및 총 플라보노이드 함량이 유의하게 높은 것을 확인할 수 있었다. 구체적으로, pH5로 조절된 실시예 7, 10, 16 및 19에서 pH3 또는 pH7로 조절된 천연 공융 용매와 비교하여 총 폴리페놀 및 총 플라보노이드 함량이 높은 것을 확인할 수 있었다.
즉, 일 구체예에 따른 천연 공융 용매는 특정 pH에서 추출 대상의 활성 성분을 극대화할 수 있다.
실험예 2. 세포독성 확인
일 구체예에 따른 천연 공융 용매 추출물의 피부 세포에 대한 세포 독성을 확인하였다. 구체적으로, 96-웰 플레이트에 DMEM에 10% FBS가 첨가된 세포 배양 배지에 희석된 섬유아세포(fibroblast)를 1x105 cells/mL개씩 넣고 24시간 동안 부착시켰다. 이후, 실시예 1~20 및 비교예 1~4를 적절한 농도로 희석하여 각 웰에 각각 처리한 후 24시간 동안 배양하였다. 24시간 경과 후, 배지를 제거하고, 각 웰 당 MTT(3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide) 용액(2.5㎎/㎖)이 함유된 세포배양 배지 200㎕를 넣은 후 2시간 동안 37℃, CO2 배양기에서 배양하였다. 배지를 제거하고 DMSO(Dimethyl sulfoxide)를 100㎕씩 넣어주었다. 5분간 진탕하여 세포를 용해시킨 후, 마이크로플레이트 판독기에서 565 ㎚ 흡광도를 측정하였다. 하기 수학식 1에 의해 세포생존율(%)을 정하였으며, 세포의 생존에 영향을 미치지 않는 시료의 농도를 결정하여 그 결과를 하기 표 2에 나타내었다.

그 결과, 표 2에 나타낸 바와 같이, 비교예 1~4와 비교하여 실시예 1~20에서 100% 세포 생존 농도가 5배 내지 40배 이상 높은 것을 확인할 수 있었다. 특히, 천연 공융 용매의 pH 3 내지 pH 7의 경우, pH 2.5와 비교하여 세포 생존 농도가 유의하게 높은 것을 확인할 수 있었다. 구체적으로, pH가 조절된 실시예 6~20의 경우, pH가 증가함에 따라 세포 독성이 감소하는 경향을 나타내었으며, 특히 pH5로 조절한 천연 공융 용매 추출물에서 100% 세포 생존 농도가 가장 높은 것을 확인할 수 있었다.
따라서, 일 구체예에 따른 천연 공융 용매는 피부 자극의 유발 가능성이 작고, 유용 농도의 범위가 넓은 바, 안전하고 효율적인 용매로 사용할 수 있다.
실험예 3. 상온 안정성 확인
일 구체예에 따른 천연 공융 용매 추출물의 상온 안정성을 확인하였다. 구체적으로, 상기 실시예 1~20 및 비교예 1~4에 디프로필렌글리콜 60 중량%를 혼합하여 시료를 제조하였다. 이후, 상기 시료를 초가 용기에 각각 담은 뒤, 45℃로 일정하게 유지되는 항온조에서 4주 동안 보관하여 변색 정도를 측정하였다. 시료의 색상 변화는 크로마메타(Minolta CR300)를 이용하여 색의 변화(ㅿE)를 측정한 후, 안정성을 평가하여 그 결과를 하기 표 3에 나타내었다.

그 결과, 표 3에 나타낸 바와 같이, 비교예 1~4의 유기 용매 추출물과 비교하여 실시예 6~20의 경우, 상온에서의 색상 변화가 비교적 적은 것을 확인할 수 있었다. 구체적으로, pH5로 조절된 천연 공융 용매로 추출한 천연물 추출물(실시예 7, 10, 14, 16 및 19)에서 색상 변화가 적은 바, 가장 높은 안정성을 나타내는 것을 확인할 수 있었다.
상기 실험예 1 내지 3에 의할 때, 비교예 1~4는 실시예 1~20과 비교하여 추출 효율이 우수하였으나, 세포 독성 및 상온 안정성이 떨어지는 문제가 있다. 그러나, 일 구체예에 따른 천연 공융 용매는 유기 용매 추출에 의해 발생할 수 있는 세포 독성, 상온 안정성의 문제를 개선하였다는 점에서, 화장품 및 피부외용제의 제조를 위한 용매로 유용하게 사용될 수 있다.
실험예 4. 항산화 활성 확인(1)
상기 실험예 3의 결과에 따라, 이하의 실험예에서는 pH 5의 천연 공융 용매로 추출한 천연물 추출물의 활성 실험을 수행하였다.
NBT(Nitro Blue Tetrazolium) 법을 이용하여 일 구체예에 따른 천연 공융 용매 추출물의 항산화 활성을 확인하였다. 구체적으로, 바이엘 병에 0.05M의 Na2CO3 2.4㎖, 3mM의 잔틴 용액 0.1㎖, 3mM의 EDTA 용액 0.1㎖, BSA 용액 0.1㎖, 및 0.72mM의 NBT 용액 0.1㎖를 가하고 상기 실시예 7, 10, 13, 16 및 19와 비교예 1~4를 0.1 중량%로 각각 첨가한 후 25 ℃에서 10 분 동안 방치하였다. 이후, 잔틴옥시다제 용액 0.1 ㎖를 첨가하여 빠르게 교반한 뒤, 25 ℃에서 20 분 동안 배양하고, 6 mM의 CuCl2용액 0.1 ㎖을 가하여 반응을 정지킨 후, 560 ㎚에서의 흡광도(St)를 측정하였다. 음성대조군으로는 상기 실시예의 추출물 대신 증류수(Bt)를 사용하였다. 시료 용액의 블랭크(Blank)는 반응 효소인 잔틴옥시다제 용액 대신 증류수(Bo)를 사용하였으며, 측정 결과를 하기 수학식 2에 의하여 산출하였다. 양성대조군으로 BHT(Butylated hydroxytoluene)를 사용하였다.

실험예 5. 항산화 활성 확인(2)
DPPH법[2,2-Di(4-tert-octylphenyl)-1-picrylhydrazyl free radical]을 이용하여 일 구체예에 따른 천연 공융 용매 추출물의 항산화 활성을 확인하였다. 구체적으로, 상기 실시예 7, 10, 13, 16, 및 19와 비교예 1~4를 0.1% 농도로 희석한 뒤, 96-웰 플레이트에 넣어주었다. 이후, 100 ㎕ 메탄올 용액으로 제조된 DPPH를 첨가하여 용액의 총 부피가 200 ㎕가 되도록 한 뒤 37℃에서 30분간 방치한 후, 560 ㎚에서 흡광도를 측정하였다. 자유라디컬 소거활성(%)은 하기 수학식 3에 의하여 산출하였고, 그 결과를 하기 표 5에 나탸내었다. 양성대조군으로 BHT(Butylated hydroxytoluene)를 사용하였다.

실험예 6. 자외선에 의한 피부세포 방어 효과 확인
일 구쳬예에 따른 천연 공융 용매 추출물의 자외선에 의한 세포 독성 완화 효과를 확인하였다. 구체적으로, 섬유아세포(fibroblast)를 96-웰 플레이트에 1x105cells/㎖ 개씩 넣고 24시간 동안 부착시켰다. 24시간 후 상기 실시예 7, 10, 13, 16, 및 19와 비교예 1~4를 처리하여 동일한 배양조건에서 24시간 동안 추가로 배양하였다. 그 후 배지를 제거하고 각 웰을 PBS로 1회 세척한 뒤, 각 웰에 100 ㎕의 PBS를 넣었다. 이후, 자외선 B(UVB) 램프(Model: F15T8, UV B 15W, Sankyo Dennki사, Japan)를 이용하여 자외선 10 mJ/㎠를 조사한 후 PBS를 덜어내고 10% FBS가 첨가된 DMEM 200 ㎕을 첨가하였다. 여기에 각각의 시료를 세포독성을 나타내지 않는 농도로 처리한 후 24시간 동안 배양하였다. 24시간이 지나면 배지를 제거하고, 각 웰 당 세포배양 배지를 함유하는 MTT 용액(2.5㎎/㎖) 200 ㎕를 넣은 후 2시간 동안 37℃, CO2 배양기에서 배양하였다. 배지를 제거하고 DMSO(Dimethyl sulfoxide)을 100 ㎕씩 넣어준 뒤, 2분간 진탕하여 세포를 용해하고 마이크로플레이트 판독기에서 565 ㎚ 흡광도를 측정하였다. 세포 생존율(%) 및 자외선에 의한 세포독성 완화율을 각각 하기 수학식 4 및 수학식 5에 의하여 산출하였고, 그 결과를 하기 표 6에 나타내었다.

실험예 7. 염증유발관련 효소 활성 억제 효과 확인
일 구체예에 따른 천연 공융 용매 추출물의 염증 예방, 개선 또는 치료 효과를 확인하였다. 구체적으로, 상기 실시예 7, 10, 13, 16, 및 19와 비교예 1~4 100 ㎕에 히알루로니디아제 용액(type 쿞igma, 400 U/㎖) 50 ㎕를 각각의 웰에 넣고 37℃에서 20분 동안 반응시킨 후, 효소활성화용액(compound 48/80 CaCl22H2O, Sigma, 0.1㎎/㎖)을 100 ㎕를 첨가하고 37℃에서 20분 동안 추가로 반응시켰다. 이후, 히알루론산(hyaluronic acid) 용액(0.4㎎/㎖)을 250 ㎕ 넣고 37℃에서 40분 동안 반응시킨 뒤, 0.4N NaOH 100 ㎕를 넣어 반응을 종결시켰다. 포타슘보레이트 용액을 100 ㎕ 첨가하여 95℃에서 3분 동안 반응시키고 냉각시킨 다음 ρ-디메틸아미노벤즈알데히드 용액을 3 ㎎ 넣고 37℃에서 20분 동안 추가로 반응시킨 후 발색시켰다. 이후, 585㎚에서 흡광도를 측정하여 히알루로니디아제 활성 저해율을 측정하였다. 양성대조군으로 유용성 감초 추출물을 사용하였다. 히알루로니다아제의 활성 저해율(%)은 하기 수학식 6에 의하여 계산하였으며, 그 결과를 하기 표 7에 나타내었다.

실험예 8. 자외선에 의한 염증성 사이토카인 억제 효과 확인
일 구체예에 따른 천연 공융 용매 추출물의 자외선에 의한 염증 예방, 개선 또는 치료 효과를 확인하였다. 구체적으로, 10%의 FBS가 첨가된 배지에 35φ dish에 3X105 cells/ml로 희석한 각질형성세포주를 3 ml씩 분주하여 24시간 동안 배양하였다. 이후, UVB 15 mJ/cm2로 조사하고 상기 실시예 7, 10, 13, 16, 및 19와 비교예 1~4를 세포독성이 나타나지 않는 농도로 FBS 2% 함유된 배지에 희석한 후 24시간 동안 추가 배양하였다. 배양 후 RNeasy mini kit (Qiagen, Hilden)을 이용하여 총 RNA를 추출하고, 1㎍의 총 RNA를 oligo (dT)15 primer, dNTP (0.5uM), 1 unit RNase 억제제 및 4 unit Omniscript reverse transcriptase(Qiagen, Hilden, Germany)로 37℃에서 60 분, 93℃에서 5분 동안 가열 후, 반응을 중지시킴으로써 cDNA를 합성하였다. 상기 cDNA로부터 IL-1α, TNF-α, IL-1β, IL-6, COX-2, GAPDH를 증폭하기 위해 PCR을 실시하였다. PCR에 의하여 생성된 산물을 2% 아가로즈 겔에서 전기영동하여 유전자의 발현을 image analyzer로 확인하였으며, 각 밴드의 밀도는 Image J program (NIH Image software, Maryland, USA)을 이용하였다. 양성대조군으로 마데카소사이드를 사용하였다. 염증성사이토카인 및 염증매개물질의 발현 억제율을 하기 수학식 7에 의하여 계산하였으며, 그 결과를 하기 표 8에 나타내었다.

실험예 9. 티로시나제 활성 억제 효과 확인
일 구체예에 따른 천연 공융 용매 추출물의 미백 개선 활성을 확인하였다. 구체적으로, L-티로신(Sigma)을 0.05M 인산나트룸 완충용액(pH 6.8)에 용해하여 0.1㎎/㎖ 용액으로 제조한 후, 제조된 용액 0.5 ㎖를 시험관에 넣었다. 여기에 0.05M 인산나트륨 완충 용액(pH 6.8)에 적당한 농도로 조절하여 용해한 상기 실시예 7, 10, 13, 16, 및 19와 비교예 1~4를 각각 0.5 ㎖씩 추가하고 37℃ 항온기에서 10분간 방치하였다. 이후, 200 unit/㎖ 티로시나제 0.5 ㎖를 첨가하고 37℃에서 10분간 반응시킨 뒤, 얼음 위에 놓아 급냉시켜 반응을 중지시켰다. 분광광도계를 파장 475 ㎚에서의 흡광도를 측정하였다. 음성대조군으로 완충용액 0.5 ㎖를 사용하였고, 양성대조군으로 0.05M 인산나트륨 완충 용액(pH 6.8)에 적당한 농도로 조절하여 용해한 알부틴 용액을 사용하였다. 티로시나제에 대한 저해율(%)을 하기 수학식 8에 의하여 계산하였으며, 그 결과를 하기 표 9에 나타내었다.

실험예 10. 멜라닌 합성 억제 효과 확인
일 구체예에 따른 천연 공융 용매 추출물의 미백 개선 활성을 확인하였다. 구체적으로, 멜라노마 세포인 B16F10(기탁번호: ATCC 6323)를 6웰 플레이트에 각 웰당 2 x 106 cells/㎖ 농도로 분주하고 세포를 부착시킨 후 독성을 유발하지 않는 농도로 상기 실시예 7, 10, 13, 16, 및 19와 비교예 1~4를 처리하여 72시간 동안 배양하였다. 이후, 세포를 트립신(trypsin)-EDTA로 떼어낸 후 세포 수를 측정한 다음 원심분리하여 세포를 회수하였다. 세포 내 멜라닌의 정량은 로탄[Lotan: Cancer Res., 40, 3345- 3350(1980)]의 방법을 약간 변형하여 실시하였다. 셀 펠릿을 PBS(phosphate buffered saline)로 1회 세척한 후 균질화 완충액(50mM 소듐 포스페이트, pH 6.8, 1% Triton X-100, 2mM PMSF(phenyl methane sulphonyl fluoride)) 1 ㎖를 첨가하여 5분간 와류하여 세포를 파쇄하였다. 원심분리(3,000 rpm, 10분)하여 얻은 세포 여액에 1N NaOH 10% DMSO(Dimethyl sulfoxide)를 첨가하여 추출된 멜라닌을 용해하였다. 이후, 마이크로 플레이트 판독기로 405 ㎚에서 멜라닌의 흡광도를 측정한 뒤, 멜라닌을 정량하여 시료의 멜라닌 생성 저해율(%)을 측정하였다. 멜라닌 생성 저해율을 하기 수학식 9에 의하여 계산하였으며, 그 결과를 하기 표 10에 나타내었다. 양성대조군으로 알부틴을 사용하였다.

실험예 11. 피부 장벽 강화 효과 확인
일 구체예에 따른 천연 공융 용매 추출물의 피부 장벽 강화 효과를 확인하였다. 구체적으로, 인간 각질형성 세포주(human keratinocyte)인 HaCaT 세포를 10% 우혈청 (fetal bovine serum)을 포함한 DMEM 배지(Dulbecco'smodified Eagle's Medium, Gibco 11965-092)에서 배양하였고, 배양은 모두 37℃ 5% CO2 배양기에서 수행하였다. 상기 배양된 세포주에 실시예 7, 10, 13, 16 및 19 및 비교예 1~4를 처리하고 24시간 동안 추가로 배양하였다. 이후, 각 시료의 세포에서 RNeasy mini kit(Qiagen, Germany)를 이용하여 총 RNA를 분리한 뒤 nanodrop으로 260 ㎚에서 RNA를 정량한 후, 각각 2㎍의 RNA를 사용하여 증폭기에서 cDNA를 합성하였다(C1000 Thermal Cycler, Bio-Rad, 미국). 합성된 cDNA를 주형으로 타겟 유전자인 피부 장벽 강화 및 보습과 관련된 인자인 SPT(serine palmitoyl transferase), AQP3과 항균 펩타이드인 RNase7에 대한 프라이머 및 cDNA를 첨가하여 실시간(real-time) PCR 기계 (Mic qPCR, biomolecular system, 오스트레일리아)에서 실시간 중합효소 연쇄반응을 수행하였다. 유전자의 발현량은 GAPDH 유전자에 대한 보정을 통해 최종적으로 분석하였고, 그 결과를 하기 표 11에 나타내었다.

실험예 12. 피부 주름 및 보습 효과 확인
일 구체예에 따른 천연 공융 용매 추출물의 피부 주름 및 보습 개선 활성을 확인하였다. 구체적으로, DMEM/F-12(3:1)배지에 10% 우태아혈청(FBS)을 첨가하여 인간 진피섬유아세포(Modern Tissue Technology, Korea)를 1Х104 cell/cm2 농도로 배양하였다. 70~80% 정도 자라면 1:3의 비율로 분주하여 계대 배양하였고, 3~4차 계대배양된 세포를 실험에 이용하였다. 상기 섬유아세포를 48 웰 플레이트(well plate)에 90% 이상 배양한 후 세포독성이 나타나지 않는 농도로 상기 실시예 7, 10, 13, 16, 및 19와 비교예 1~4를 가하고, 24시간 후에 배지 중에 유리된 프로콜라겐의 양을 프로콜라겐타입-1C-펩타이드 EIA 키트(procollagen type-1 C-peptide EIA kit)(MK101, Takara, Japan)를 사용하여 측정하였다. 양성대조군으로 아스코르브산을 사용하였다.

일 구체예에 따른 천연 공융 용매 추출물의 여드름 및 피지, 또는 탈모의 예방 또는 개선 효과를 확인하였다. 구체적으로, 마이크로플레이트 홀(microplate hole)마다 3H(트리튬)으로 방사선표지(radio labelling)된 테스토스테론 0.1μCi 및 상기 실시예 7, 10, 13, 16, 및 19와 비교예 1~4를 첨가한 후 섬유아세포를 10,000개의 세포가 들어가도록 접종하여 배양하였다. 24시간 배양한 후, 상등액을 얻고 에틸아세테이트(ethyl acetate)-사이크로헥산(cyclohexane)(1:1) 추출용매 1 ㎖로 스테로이드(steroids)를 얻었다. 상기 스테로이드를 박막크로마토그래피판에 올려놓고 클로로포름/메탄올 혼합액(98/2 (v/v))로 전개시켰다. 테스토스테론과 디하이드로테스토스테론(Dihydrotestosterone, DHT) 에 해당하는 점의 방사 활성을 덴시토미터를 이용하여 측정하여 전환율을 계산하고, 그 결과를 음성대조군과 비교하여 하기 수학식 10에 의하여 5알파-환원효소 저해율을 평가하였다.

실험예 14. 피부 탄력 개선 효과 확인
일 구체예에 따른 천연 공융 용매 추출물을 포함하는 화장료 조성물의 피부 탄력 개선 효과를 확인하였다. 먼저, 하기 표 14의 성분 및 함량을 포함하는 크림 제형의 화장료 조성물을 제조하였다. 구체적으로, 하기 표 14의 B상을 가열하여 70℃에서 보관한 뒤, A상을 가하여 예비 유화하였다. 이후, 호모믹서로 균일하게 균일하여 유화한뒤, 서서히 냉각하여 화장료 조성물을 제조하였다. 본 명세서에서 다른 언급이 없는 한 함량은 중량%이다.
20~35세의 여성 20명에게 제형예 1~6 및 비교 제형예 1~4를 각각 1일 2회씩 2개월 동안 사용하게 하였다. 이후, 피부탄력측정기 (cutometer SEM 575, C+K Electronic Co., Germany)를 이용하여 상기 제형예 1, 5 및 비교 제형예 1을 사용하기 전과 후의 피부 탄력을 측정하였다. 측정값은 Cutometer SEM 575의 R7 값으로 기재하였으며, 상기 R7값은 피부의 점탄성(viscoelasticity)을 나타낸다.

일 구체예에 따른 천연 공융 용매 추출물을 포함하는 화장료 조성물의 피부 주름 개선 효과를 확인하였다. 구체적으로, 상기 실험예 14의 제형예 1, 5, 및 비교 제형예 1을 동일한 대상자에게 동일한 방법으로 사용하게 하였다. 이후, 실리콘 재질의 모사판(replica)을 제작한 뒤, 화상분석기인 비지오미터(visiometer: SV60, C+K Electronic Co., 등록특허 10-1905517-13-Germany)를 이용하여 상기 제형예 1, 5 및 비교 제형예 1을 사용한 후의 피부 주름 상태를 측정하였다. 측정 값은 2개월 사용 후의 각각의 파라미터(parameter) 값에서 사용 전 파라미터 값을 뺀 뒤 평균으로 나타내었으며, 측정 값이 음수가 나올수록 주름 개선 효과가 높음을 의미한다.

일 구체예에 따른 천연 공융 용매 추출물을 포함하는 화장료 조성물의 피부 미백 개선 효과를 확인하였다. 구체적으로, 상기 실험예 14의 제형예 1, 5, 및 비교 제형예 1을 동일한 대상자에게 동일한 방법으로 사용하게 하였다. 이후, 도포 부위 피부를 화상분석기 및 크로마메타(Minolta CR300)를 이용하여 피부색의 밝기 변화(L)를 측정하였다. 또한, 복수의 숙련자에 의하여 객관적 육안 관찰과 피험자에 의한 주관적 육안 관찰을 실시하여 미백 효능 정도를 하기의 등급에 따라 평가하였다.
-3: 매우 악화, -2: 악화, -1: 약간 악화, 0: 변화 없음, 1: 약간 개선, 2: 개선, 3: 매우 개선

그 결과, 표 17에 나타낸 바와 같이, 제형예 1 및 5는 비교 제형예 1과 비교하여 피부색의 밝기 변화가 현저하게 증가한 것을 확인할 수 있었다. 구체적으로, 천연 공융 용매의 산도를 pH 5로 조절한 제형예 5에서 피부색의 밝기 변화가 유의하게 증가한 것을 확인할 수 있었다.
따라서, 일 구체예에 따른 천연 공융 용매 천연물 추출물은 피부 미백 개선에 유용하게 사용될 수 있다.
전술한 본 발명의 설명은 예시를 위한 것이며, 본 발명이 속하는 기술분야의 통상의 지식을 가진 자는 본 발명의 기술적 사상이나 필수적인 특징을 변경하지 않고서 다른 구체적인 형태로 쉽게 변형이 가능하다는 것을 이해할 수 있을 것이다. 그러므로 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적이 아닌 것으로 이해해야만 한다.
Claims (17)
- 천연 공융 용매로 추출한 천연물 추출물을 유효성분으로 포함하는 피부 상태 개선용 화장료 조성물로서, 상기 천연 공융 용매는 아미노산 또는 이의 유도체, 및 유기산을 포함하는 것인 조성물.
- 청구항 1에 있어서, 상기 천연물은 무화과, 권백, 흑미, 혈갈, 검은콩, 능소화, 설련, 마가목, 화분, 물개구리밥, 향나무, 봉미초, 금낭화, 겨우살이, 돌나물, 정금, 상황버섯, 토마토, 어성초, 진피, 모란, 밤꽃, 칡, 호두로 이루어진 군에서 선택된 것인, 조성물.
- 청구항 1에 있어서, 상기 천연 공융 용매는 pH 2.5 내지 pH 8인 것인, 조성물.
- 청구항 1에 있어서, 상기 아미노산 또는 이의 유도체는 베타인, 알라닌, 시스테인, 아스파르트산, 글루타민, 페닐알라닌, 글리신, 히스티딘, 이소로이신, 라이신, 로이신, 메티오닌, 아스파라긴, 프롤린, 글루타민, 아르기닌, 세린, 트레오닌, 발린, 트립토판, 및 티로신로 이루어진 군에서 선택되는 것인, 조성물.
- 청구항 1에 있어서, 상기 유기산은 구연산, 락트산, 말산, 말레산, 피루브산, 푸마르산, 석신산, 아세트산, 아코니트산, 타트르산, 아스코르브산, 말론산, 옥살산, 글루쿠론산, 뉴라민산, 시알산, 시킴산, 피트산, 갈락투론산, 이두론산, 히알루론산, 하이드록시시트르산, 및 락톤 유도체로 이루어진 군에서 선택되는 것인, 조성물.
- 청구항 1에 있어서, 상기 아미노산 또는 이의 유도체 및 유기산은 1:0.001 내지 10의 중량비로 혼합된 것인, 조성물.
- 청구항 1에 있어서, 상기 피부 상태는 피부 노화, 보습, 주름, 탄력, 피부 장벽 강화, 여드름 또는 피지 분비인 것인, 조성물.
- 청구항 7에 있어서, 상기 피부 노화는 자외선 또는 활성산소에 의한 것인, 조성물.
- 천연 공융 용매로 추출한 천연물 추출물을 유효성분으로 포함하는 피부 상태 개선용 피부 외용제 조성물로서, 상기 천연 공융 용매는 아미노산 또는 이의 유도체, 및 유기산인 것인 조성물.
- 천연 공융 용매로 추출한 천연물 추출물을 유효성분으로 포함하는 피부 염증 질환의 예방 또는 치료용 약학적 조성물로서, 상기 천연 공융 용매는 아미노산 또는 이의 유도체, 및 유기산인 것인 조성물.
- 청구항 8에 있어서, 상기 염증 질환은 피부 상처, 피부염, 아토피 피부염, 소양증, 습진성 피부질환, 건성 습진, 홍반, 두드러기, 건선, 약발진, 및 여드름으로 이루어진 군에서 선택된 것인, 조성물.
- 천연 공융 용매로 추출한 천연물 추출물을 유효성분으로 포함하는 탈모 예방 또는 치료, 또는 발모 또는 육모 촉진용 약학적 조성물로서, 상기 천연 공융 용매는 아미노산 또는 이의 유도체, 및 유기산인 것인 조성물.
- 아미노산 또는 이의 유도체, 및 유기산을 혼합하여 혼합물을 제조하는 단계;상기 혼합물에 시트르산삼나트륨(sodium citrate), 수산화나트륨(sodium hydroxide), 수산화칼륨(potassium hydroxide), 중탄산나트륨(sodium bicarbonate), 아르기닌(arginine), 트로메타민(tromethamine), 트리에탄올아민(triethanol amine), 아미노메틸프로판올(aminomethyl propanol), 디포타슘포스페이트(dipotassium phosphate), 포타슘포스페이트(potassium phosphate), 아니스산나트륨(sodium anisate), 젖산나트륨(sodium lactate), 탄산나트륨(sodium carbonate), 및 요소(urea)로 이루어진 군에서 선택되는 pH 조절제를 추가하는 단계를 포함하는, 천연 공융 용매의 제조방법.
- 피부 상태를 개선하기 위한 화장료 조성물의 제조를 위한, 천연 공융 용매로 추출한 천연물 추출물의 용도로서, 상기 천연 공융 용매는 아미노산 또는 이의 유도체, 및 유기산을 포함하는 것인 용도.
- 피부 상태를 개선하기 위한 피부 외용제 조성물의 제조를 위한, 천연 공융 용매로 추출한 천연물 추출물의 용도로서, 상기 천연 공융 용매는 아미노산 또는 이의 유도체, 및 유기산을 포함하는 것인 용도.
- 피부 염증 질환의 예방 또는 치료를 위한 약학적 조성물의 제조를 위한, 천연 공융 용매로 추출한 천연물 추출물의 용도로서, 상기 천연 공융 용매는 아미노산 또는 이의 유도체, 및 유기산을 포함하는 것인 용도.
- 탈모 예방 또는 치료, 또는 발모 또는 육모 촉진을 위한 약학적 조성물의 제조를 위한, 천연 공융 용매로 추출한 천연물 추출물의 용도로서, 상기 천연 공융 용매는 아미노산 또는 이의 유도체, 및 유기산을 포함하는 것인 용도.
