Abstract

樹状細胞は、自然免疫系および獲得免疫系を高度に制御する免疫細胞である。樹状細胞の一種であるプラズマサイトイド樹状細胞(形質細胞様樹状細胞、plasmacytoid dendritic cell、以降「pDC」とも記す)は、Toll様受容体(TLR)7およびTLR9を発現しており、TLR-7やTLR-9でウイルス由来のRNAやDNAを認識し、ウイルスに対して増殖阻害活性を示す大量のI型インターフェロン(IFN)を産生する。また、pDCは抗原提示細胞としても働き、特定のT細胞を活性化する。
従来、免疫機能を増強させたりウイルス防御活性を高めたりすることへの関心は高く、IFNを産生させる成分が研究されてきた。例えば、特許文献1には乳酸菌がpDCを活性化してIFN産生を誘導し得ることが開示されている。
また、乳タンパク質の一つであるラクトフェリン(以下、「LF」とも記す)についても、IFN産生との関係について報告されている。
非特許文献1には、1日1gのLFの摂取によってpDCのIFN-α産生能が上昇する傾向があることが報告されている。非特許文献2には、リポソーム化LFを270mg/日摂取したことにより健常成人男性のIFN-α産生能が前後比較で有意に上昇することが報告されている。また、抗原提示細胞として働くpDCは、その活性化の際には共刺激分子であるCD86等を細胞表面に発現するところ、非特許文献3には、腸溶性LFを300mg/日摂取したことにより健常高齢者においてCD86を発現する細胞が有意に増加することが報告されている。
しかしながら、CD86は骨髄系樹状細胞、マクロファージ、単球など、抗原提示細胞一般に広く発現しており、末梢血単核球中に0.2~0.5%しか存在しないマイナーポピュレーションであるpDCに関しては、ラクトフェリンが、pDCを増加させたり、pDCの細胞表面マーカーであるCD86の発現を促進したりすることは知られていない(非特許文献4)。
非特許文献1には、1日1gのLFの摂取によってpDCのIFN-α産生能が上昇する傾向があることが報告されている。非特許文献2には、リポソーム化LFを270mg/日摂取したことにより健常成人男性のIFN-α産生能が前後比較で有意に上昇することが報告されている。また、抗原提示細胞として働くpDCは、その活性化の際には共刺激分子であるCD86等を細胞表面に発現するところ、非特許文献3には、腸溶性LFを300mg/日摂取したことにより健常高齢者においてCD86を発現する細胞が有意に増加することが報告されている。
しかしながら、CD86は骨髄系樹状細胞、マクロファージ、単球など、抗原提示細胞一般に広く発現しており、末梢血単核球中に0.2~0.5%しか存在しないマイナーポピュレーションであるpDCに関しては、ラクトフェリンが、pDCを増加させたり、pDCの細胞表面マーカーであるCD86の発現を促進したりすることは知られていない(非特許文献4)。

本発明は、pDCを活性化させることにより、免疫機能を維持、改善、又は強化することができる組成物を提供することを課題とする。
本発明者らは、上記の課題を解決すべく鋭意研究を行った結果、ヒト末梢血から回収した単核細胞をLF存在下で培養すると、pDCが増加し、またCD86陽性細胞も増加することを見出し、本発明を完成するに至った。
すなわち、本発明の第一の態様は、ラクトフェリン及び/又はラクトフェリン加水分解物を含有する、プラズマサイトイド樹状細胞(pDC)活性化用組成物である。
前記活性化は、単核細胞中のpDC数の割合、pDC中のCD86陽性細胞数の割合、pDCにおけるCD86発現量、及びpDCにおけるHLA-DR発現量からなる群から選択される一種又は二種以上の増加であってよい。
本発明の第二の態様は、ラクトフェリン及び/又はラクトフェリン加水分解物を含有する、pDC増加用組成物である。
本発明の第三の態様は、ラクトフェリン及び/又はラクトフェリン加水分解物を含有する、pDCにおけるCD86及び/又はHLA-DR発現促進用組成物である。
これら本発明の組成物は、免疫機能を維持、改善、又は強化するために好ましく用いられる。また、これら本発明の組成物は、飲食品又は医薬品の形態であってよい。
前記活性化は、単核細胞中のpDC数の割合、pDC中のCD86陽性細胞数の割合、pDCにおけるCD86発現量、及びpDCにおけるHLA-DR発現量からなる群から選択される一種又は二種以上の増加であってよい。
本発明の第二の態様は、ラクトフェリン及び/又はラクトフェリン加水分解物を含有する、pDC増加用組成物である。
本発明の第三の態様は、ラクトフェリン及び/又はラクトフェリン加水分解物を含有する、pDCにおけるCD86及び/又はHLA-DR発現促進用組成物である。
これら本発明の組成物は、免疫機能を維持、改善、又は強化するために好ましく用いられる。また、これら本発明の組成物は、飲食品又は医薬品の形態であってよい。
本発明によれば、pDCを活性化させることにより、免疫機能を維持、改善、又は強化することができる。
次に、本発明を詳細に説明する。ただし、本発明は以下の実施形態に限定されず、本発明の範囲内で自由に変更することができるものである。
本発明の組成物は、ラクトフェリン及び/又はラクトフェリン加水分解物を有効成分として含有する。
ラクトフェリンは、哺乳動物、例えば、ヒツジ、ヤギ、ブタ、マウス、水牛、ラクダ、ヤク、ウマ、ロバ、ラマ、ウシ又はヒトの乳、涙、唾液、血液等に含まれる鉄結合性の糖タンパク質である。
本発明におけるラクトフェリンは、いずれの哺乳動物に由来するものであってもよく、特に限定されないが、含有量や入手容易性の点から、例えば、ウシ、ヒト等の乳由来のラクトフェリンが好ましい。前記乳としては、初乳、移行乳、常乳、末期乳のいずれでもよい。
また、本発明におけるラクトフェリンは、前記乳の処理物である脱脂乳、ホエイ等から常法(例えば、イオンクロマトグラフィー等)によって分離されたラクトフェリン、遺伝子操作によって微生物、動物細胞、トランスジェニック動物等から産生された組換えラクトフェリン、合成ラクトフェリン、又はそれらの混合物でもよい。また、ラクトフェリンは、非グリコシル化又はグリコシル化されたものでもよい。このようなラクトフェリンとして、工業的規模で製造されている市販のラクトフェリン(例えば、森永乳業社製等)を使用することができる。
本発明におけるラクトフェリンは、いずれの哺乳動物に由来するものであってもよく、特に限定されないが、含有量や入手容易性の点から、例えば、ウシ、ヒト等の乳由来のラクトフェリンが好ましい。前記乳としては、初乳、移行乳、常乳、末期乳のいずれでもよい。
また、本発明におけるラクトフェリンは、前記乳の処理物である脱脂乳、ホエイ等から常法(例えば、イオンクロマトグラフィー等)によって分離されたラクトフェリン、遺伝子操作によって微生物、動物細胞、トランスジェニック動物等から産生された組換えラクトフェリン、合成ラクトフェリン、又はそれらの混合物でもよい。また、ラクトフェリンは、非グリコシル化又はグリコシル化されたものでもよい。このようなラクトフェリンとして、工業的規模で製造されている市販のラクトフェリン(例えば、森永乳業社製等)を使用することができる。
本発明におけるラクトフェリン中の金属含有量は特に限定されず、ラクトフェリンを塩酸やクエン酸等により脱鉄したアポ型ラクトフェリン;該アポ型ラクトフェリンを、鉄、銅、亜鉛、マンガン等の金属でキレートさせて得られる飽和度100%以上の状態の金属飽和型ラクトフェリン;及び100%未満の各種飽和度で金属が結合している状態の金属部分飽和型ラクトフェリンからなる群から選ばれる、いずれか1種又は2種以上の混合物を用いることができる。
ここで、本技術に用いられるラクトフェリンの調製(乳等の原料からのラクトフェリンの分離、精製)方法の一例を以下に示すが、これに限定されるものではない。
ウシ由来の乳原料を陽イオン交換カラムに通液し、この通過液を回収し、適宜この通過液を繰り返しカラムに通液する。このカラムに脱イオン水を通液し、食塩水を通液し、この陽イオン交換カラムに吸着した塩基性タンパク質の溶出液を得る。この溶出液からタンパク質を回収し、適宜洗浄し、脱イオン水にて溶解し、この溶解液を限外ろ過膜にてろ過する。さらに、脱塩処理、凍結乾燥することで、粉末状のラクトフェリンが得られる。
ウシ由来の乳原料を陽イオン交換カラムに通液し、この通過液を回収し、適宜この通過液を繰り返しカラムに通液する。このカラムに脱イオン水を通液し、食塩水を通液し、この陽イオン交換カラムに吸着した塩基性タンパク質の溶出液を得る。この溶出液からタンパク質を回収し、適宜洗浄し、脱イオン水にて溶解し、この溶解液を限外ろ過膜にてろ過する。さらに、脱塩処理、凍結乾燥することで、粉末状のラクトフェリンが得られる。
より詳細には、まず、イオン交換体をカラムに充填し、塩酸を通液し、水洗してイオン交換体を平衡化する。続いて、4℃に冷却したpH6.9の脱脂乳をカラムに通液し、透過液を回収し、再度同様にカラムに通液する。次いで、脱イオン水をカラムに通液し、食塩水を通液し、イオン交換体に吸着した塩基性タンパク質の溶出液を得る。この溶出液から回収したタンパク質を洗浄し、脱イオン水を添加して溶解し、得られた溶液を限外ろ過膜モジュールを用いて脱塩し、凍結乾燥して、粉末状ウシラクトフェリンを得る。このようにして、純度が95質量%以上のウシラクトフェリンが得られる。
ラクトフェリンにおいては、種、属、個体等の違いによって、1又は複数の位置での1又は複数の塩基の置換、欠失、挿入、付加、又は逆位等の遺伝子変異が当然存在し、このような変異を有する遺伝子がコードするタンパク質のアミノ酸においても変異が生じている場合がある。本発明における本技術に用いることができるラクトフェリンには、本発明の効果を損なわない限りにおいて、このような変異を含むものも含有される。
また、本発明におけるラクトフェリンには、本発明の効果を損なわない限りにおいて、熱処理、酸処理、又はアルカリ処理を行ったラクトフェリン処理物も含まれてもよい。
また、本発明におけるラクトフェリンには、本発明の効果を損なわない限りにおいて、熱処理、酸処理、又はアルカリ処理を行ったラクトフェリン処理物も含まれてもよい。
本発明におけるラクトフェリン加水分解物は、前述のラクトフェリンを加水分解処理したものをいう。
加水分解処理としては、例えば特開2012-235768号公報に記載された方法が挙げられる。
具体的には、ラクトフェリン溶液を、酵素反応処理を行う前に塩酸、クエン酸、酢酸等の酸によりpHを2~4、好ましくは2.5~3.5、特に好ましくはpH3に調整する。
pHを調整したラクトフェリン溶液に、タンパク質分解酵素を所望の量で添加した後、酵素反応の温度を35~55℃、好ましくは40~50℃、より好ましくは42~48℃に保持して、6時間~24時間、好ましくは12~18時間、攪拌しながらラクトフェリンを加水分解させる。
次いで、例えば反応溶液を80℃に昇温して10分間維持し、酵素を加熱失活させる。さらに、好ましくは、水酸化ナトリウム溶液等のアルカリ溶液を添加して、pHを5~7、例えば6に調整する。
なお、pH調整後の反応溶液(ラクトフェリン加水分解物)は、溶液のままでもよいが、凍結乾燥等を行って粉末化することが好ましい。また、ラクトフェリン分解物は、クロマトグラフィー、又は限外濾過等により、分画したものを用いることもできる。
加水分解処理としては、例えば特開2012-235768号公報に記載された方法が挙げられる。
具体的には、ラクトフェリン溶液を、酵素反応処理を行う前に塩酸、クエン酸、酢酸等の酸によりpHを2~4、好ましくは2.5~3.5、特に好ましくはpH3に調整する。
pHを調整したラクトフェリン溶液に、タンパク質分解酵素を所望の量で添加した後、酵素反応の温度を35~55℃、好ましくは40~50℃、より好ましくは42~48℃に保持して、6時間~24時間、好ましくは12~18時間、攪拌しながらラクトフェリンを加水分解させる。
次いで、例えば反応溶液を80℃に昇温して10分間維持し、酵素を加熱失活させる。さらに、好ましくは、水酸化ナトリウム溶液等のアルカリ溶液を添加して、pHを5~7、例えば6に調整する。
なお、pH調整後の反応溶液(ラクトフェリン加水分解物)は、溶液のままでもよいが、凍結乾燥等を行って粉末化することが好ましい。また、ラクトフェリン分解物は、クロマトグラフィー、又は限外濾過等により、分画したものを用いることもできる。
本態様の組成物において、ラクトフェリン及び/又はラクトフェリン加水分解物の組成物全体に対する含有量は、好ましくは0.001質量%以上100質量%未満、より好ましくは0.005~95質量%、さらに好ましくは0.01~85質量%である。
本発明の組成物は、pDCを活性化することができる。
ここでpDCの活性化には、単核細胞中のpDC数の割合の増加が含まれる。これは、任意の単位の単核細胞群全体に占めるpDCの細胞数の割合が、本発明の組成物の適用前に比べて適用後に、又は本発明の組成物の適用の場合に非適用の場合に比べて、増加することをいう。なお、係る割合の増加は、pDC数の絶対的な増加によるものであってよい。言い換えると、本発明の組成物は、pDCを増加することができる。本発明の組成物によりpDC数の割合や絶対数が増加することは、pDC前駆細胞へ本発明の組成物が働きかけてpDCへの分化を促進することによると推測される。本発明の組成物の適用後のpDC細胞数は、適用前に比べて、少なくとも1.1倍であり、1.5倍以上であることが好ましく、2倍以上であることが好ましく、2.5倍以上であることが好ましく、3倍以上であることが好ましく、3.5倍以上であることが好ましく、4倍以上であることがより好ましい。上限値は、特に制限されないが、8倍以下、7倍以下、6倍以下、又は5倍以下であってよい。また、上記数値の矛盾しない組み合わせの範囲であってもよく、具体的には、1.1~8倍、1.5~8倍、2~8倍、2.5~8倍、3~8倍、3.5~8倍、又は4~8倍であって良い。
ここでpDCの活性化には、単核細胞中のpDC数の割合の増加が含まれる。これは、任意の単位の単核細胞群全体に占めるpDCの細胞数の割合が、本発明の組成物の適用前に比べて適用後に、又は本発明の組成物の適用の場合に非適用の場合に比べて、増加することをいう。なお、係る割合の増加は、pDC数の絶対的な増加によるものであってよい。言い換えると、本発明の組成物は、pDCを増加することができる。本発明の組成物によりpDC数の割合や絶対数が増加することは、pDC前駆細胞へ本発明の組成物が働きかけてpDCへの分化を促進することによると推測される。本発明の組成物の適用後のpDC細胞数は、適用前に比べて、少なくとも1.1倍であり、1.5倍以上であることが好ましく、2倍以上であることが好ましく、2.5倍以上であることが好ましく、3倍以上であることが好ましく、3.5倍以上であることが好ましく、4倍以上であることがより好ましい。上限値は、特に制限されないが、8倍以下、7倍以下、6倍以下、又は5倍以下であってよい。また、上記数値の矛盾しない組み合わせの範囲であってもよく、具体的には、1.1~8倍、1.5~8倍、2~8倍、2.5~8倍、3~8倍、3.5~8倍、又は4~8倍であって良い。
また、pDCの活性化には、pDCの活性化マーカーの発現が高いことが含まれる。
前述の通りpDCは抗原提示細胞としても働き、ヒトのpDCは、主要組織適合抗原クラスII(MHC-II)である白血球型抗原遺伝子複合体(HLA)-DR陽性細胞である。pDCはHLA-DRにより外来抗原を提示することで、CD4陽性T細胞やCD8陽性T細胞を活性化することができる。その際に、共刺激分子であるCD86、CD40、CD80、CD83などによる共刺激シグナルがT細胞の活性化に必要とされる。したがって、pDCのMHC-IIであるHLA-DRや共刺激分子であるCD86などの発現が高いことは、pDCの抗原提示細胞としての活性が高いことを示す指標となる(T. S. Mathan et al., Front Immunol., 2013;4:372.、Swiecki M. et al., Nat Rev Immunol., 2015;15:471-485.参照)。すなわち、HLA-DR、CD86、CD40、CD80、CD83等はpDCの活性化マーカーとして評価に用いることができる。
前述の通りpDCは抗原提示細胞としても働き、ヒトのpDCは、主要組織適合抗原クラスII(MHC-II)である白血球型抗原遺伝子複合体(HLA)-DR陽性細胞である。pDCはHLA-DRにより外来抗原を提示することで、CD4陽性T細胞やCD8陽性T細胞を活性化することができる。その際に、共刺激分子であるCD86、CD40、CD80、CD83などによる共刺激シグナルがT細胞の活性化に必要とされる。したがって、pDCのMHC-IIであるHLA-DRや共刺激分子であるCD86などの発現が高いことは、pDCの抗原提示細胞としての活性が高いことを示す指標となる(T. S. Mathan et al., Front Immunol., 2013;4:372.、Swiecki M. et al., Nat Rev Immunol., 2015;15:471-485.参照)。すなわち、HLA-DR、CD86、CD40、CD80、CD83等はpDCの活性化マーカーとして評価に用いることができる。
pDCの活性化マーカーの発現が高いことには、pDC中のCD86陽性細胞数の割合の増加が含まれる。これは、任意の単位のpDC群全体に占めるCD86陽性細胞数の割合が、本発明の組成物の適用前に比べて適用後に、又は本発明の組成物の適用の場合に非適用の場合に比べて、増加することをいう。なお、係る割合の増加は、CD86陽性細胞数の絶対的な増加によるものであってよい。CD86陽性細胞はCD86を発現している細胞を指し、pDCにおいてCD86の発現はpDCの活性化を表す。なお、pDCは元々HLA-DR陽性細胞であるため、一部のpDCのみが発現するCD86とは異なり、HLA-DR陽性細胞数の割合はpDC活性化の指標とはならない(Dopheide J. F. et al., Thromb Haemost. 2012;108:1198-207.)。
また、pDCの活性化マーカーの発現が高いことには、pDCにおけるCD86及び/又はHLA-DR発現量の増加が含まれる。これは、単位pDCにおいて(言い換えるとpDC1細胞当たりで)発現するCD86及び/又はHLA-DR量が、本発明の組成物の適用前に比べて適用後に、又は本発明の組成物の適用の場合に非適用の場合に比べて、増加することをいう。言い換えると、本発明の組成物は、pDCにおけるCD86及び/又はHLA-DR発現を促進することができる。
ここで、CD86及び/又はHLA-DRの発現は、周知の方法で定性的又は定量的に確認することができ、例えば蛍光標識化抗体を用いた免疫学的手法等により行うことができる。
また、pDCの活性化マーカーの発現が高いことには、pDCにおけるCD86及び/又はHLA-DR発現量の増加が含まれる。これは、単位pDCにおいて(言い換えるとpDC1細胞当たりで)発現するCD86及び/又はHLA-DR量が、本発明の組成物の適用前に比べて適用後に、又は本発明の組成物の適用の場合に非適用の場合に比べて、増加することをいう。言い換えると、本発明の組成物は、pDCにおけるCD86及び/又はHLA-DR発現を促進することができる。
ここで、CD86及び/又はHLA-DRの発現は、周知の方法で定性的又は定量的に確認することができ、例えば蛍光標識化抗体を用いた免疫学的手法等により行うことができる。
本発明の組成物はpDCを活性化させることから、種々の免疫細胞の働きを活性化したり、IFN産生を促進したりすることが見込まれるため、免疫機能を維持、改善、又は強化することが期待できる。免疫機能の維持とは正常又は良好な免疫機能が発揮されている状態を保ち機能低下することを防ぐことを含み、免疫機能の改善とは低下した免疫機能を正常な状態にすることを含む。
本発明の組成物を投与する(摂取させる)対象は、動物であれば特に限定されないが、通常は哺乳動物であり、ヒトが好ましい。また、対象は特に限定されないが、健康な人が好ましい。ここで健康な人とは、疾患や疾病に罹患していない人をいう。
本発明の別の側面は、pDC活性化用組成物の製造における、ラクトフェリン及び/又はラクトフェリン加水分解物の使用である。

本発明の組成物を経口摂取(投与)するときの含有量としては、前述した組成物全体における含有量としてもよいし、適宜希釈等してもよい。例えば、経口摂取(投与)時のラクトフェリン及び/又はラクトフェリン加水分解物の組成物全体に対する含有量は、好ましくは0.001~100質量%、より好ましくは0.005~95質量%、さらに好ましくは0.01~85質量%である。
これらは、通常、経口組成物として流通するときの含有量の範囲であってよい。

本発明の組成物の摂取(投与)量は、摂取(投与)対象の年齢(月齢)、性別、状態、その他の条件等により適宜選択される。
本発明の組成物の摂取(投与)量は、ラクトフェリン及び/又はラクトフェリン加水分解物の摂取量として、例えば、好ましくは0.01~1000mg/kg/日、より好ましくは0.1~500mg/kg/日、さらに好ましくは1~300mg/kg/日の範囲となる量を目安とするのがよい。
なお、摂取(投与)の量や期間にかかわらず、本発明の組成物は1日1回又は複数回に分けて投与することができる。
本発明の組成物の摂取(投与)量は、ラクトフェリン及び/又はラクトフェリン加水分解物の摂取量として、例えば、好ましくは0.01~1000mg/kg/日、より好ましくは0.1~500mg/kg/日、さらに好ましくは1~300mg/kg/日の範囲となる量を目安とするのがよい。

なお、飲食品としてサプリメントの形態とする場合は、腸溶性コーティング等により腸溶処理されてもよい、散剤、顆粒剤、錠剤、カプセル剤等の固形製剤;溶液剤、シロップ剤、懸濁剤、乳剤等の液剤;等に製剤化することができる。かかる製剤化に際しては、後述する医薬品の製剤化に係る成分、担体、及び方法の説明に準ずることができる。
また、飲食品の一態様として飼料とすることもできる。飼料としては、ペットフード、家畜飼料、養魚飼料等が挙げられる。
飼料の形態としては特に制限されず、ラクトフェリン及び/又はラクトフェリン加水分解物の他に例えば、トウモロコシ、小麦、大麦、ライ麦、マイロ等の穀類;大豆油粕、ナタネ油粕、ヤシ油粕、アマニ油粕等の植物性油粕類;フスマ、麦糠、米糠、脱脂米糠等の糠類;コーングルテンミール、コーンジャムミール等の製造粕類;魚粉、脱脂粉乳、ホエイ、イエローグリース、タロー等の動物性飼料類;トルラ酵母、ビール酵母等の酵母類;第三リン酸カルシウム、炭酸カルシウム等の鉱物質飼料;油脂類;単体アミノ酸;糖類等を含有するものであってよい。

本発明の組成物が飲食品(飼料を含む)の態様である場合、免疫機能の維持等の用途が表示された飲食品として提供・販売されることが可能である。また、本明細書に係るラクトフェリン及び/又はラクトフェリン加水分解物は、これら飲食品等の製造のために使用可能である。
かかる「表示」行為には、需要者に対して前記用途を知らしめるための全ての行為が含まれ、前記用途を想起・類推させうるような表現であれば、表示の目的、表示の内容、表示する対象物・媒体等の如何に拘わらず、全て本発明における「表示」行為に該当する。
また、「表示」は、需要者が上記用途を直接的に認識できるような表現により行われることが好ましい。具体的には、飲食品に係る商品又は商品の包装に前記用途を記載したものを譲渡し、引き渡し、譲渡若しくは引き渡しのために展示し、輸入する行為、商品に関する広告、価格表若しくは取引書類に上記用途を記載して展示し、若しくは頒布し、又はこれらを内容とする情報に上記用途を記載して電磁気的(インターネット等)方法により提供する行為等が挙げられる。
また、「表示」は、需要者が上記用途を直接的に認識できるような表現により行われることが好ましい。具体的には、飲食品に係る商品又は商品の包装に前記用途を記載したものを譲渡し、引き渡し、譲渡若しくは引き渡しのために展示し、輸入する行為、商品に関する広告、価格表若しくは取引書類に上記用途を記載して展示し、若しくは頒布し、又はこれらを内容とする情報に上記用途を記載して電磁気的(インターネット等)方法により提供する行為等が挙げられる。
一方、表示内容としては、行政等によって認可された表示(例えば、行政が定める各種制度に基づいて認可を受け、そのような認可に基づいた態様で行う表示等)であることが好ましい。また、そのような表示内容を、包装、容器、カタログ、パンフレット、POP等の販売現場における宣伝材、その他の書類等へ付することが好ましい。
また、「表示」には、健康食品、機能性食品、経腸栄養食品、特別用途食品、保健機能食品、特定保健用食品、栄養機能食品、機能性表示食品、医薬用部外品等としての表示も挙げられる。この中でも特に、消費者庁によって認可される表示、例えば、特定保健用食品、栄養機能食品、若しくは機能性表示食品に係る制度、又はこれらに類似する制度にて認可される表示等が挙げられる。具体的には、特定保健用食品としての表示、条件付き特定保健用食品としての表示、身体の構造や機能に影響を与える旨の表示、疾病リスク減少表示、科学的根拠に基づいた機能性の表示等を挙げることができ、より具体的には、健康増進法に規定する特別用途表示の許可等に関する内閣府令(平成二十一年八月三十一日内閣府令第五十七号)に定められた特定保健用食品としての表示(特に保健の用途の表示)及びこれに類する表示が典型的な例である。
かかる表示としては、例えば、「pDC(プラズマサイトイド樹状細胞)に働きかけ」、「免疫細胞pDCに働きかける」、「免疫細胞pDCを活性化する」「免疫細胞pDCを増やす」、「健康な人の免疫機能の維持に役立つ」、「免疫機能を高める」、「免疫機能をサポート」、「健康な人の免疫機能の維持をサポート」、「pDC (プラズマサイトイド樹状細胞)に働きかけ、健康な人の免疫機能の維持に役立つ」等と表示することが挙げられる。
かかる表示としては、例えば、「pDC(プラズマサイトイド樹状細胞)に働きかけ」、「免疫細胞pDCに働きかける」、「免疫細胞pDCを活性化する」「免疫細胞pDCを増やす」、「健康な人の免疫機能の維持に役立つ」、「免疫機能を高める」、「免疫機能をサポート」、「健康な人の免疫機能の維持をサポート」、「pDC (プラズマサイトイド樹状細胞)に働きかけ、健康な人の免疫機能の維持に役立つ」等と表示することが挙げられる。
本発明の組成物は、医薬品の態様とすることもできる。
医薬品の投与経路は、経口又は非経口のいずれでもよいが経口が好ましい。また、非経口摂取(投与)としては、経皮、静注、直腸投与、吸入等が挙げられる。
医薬品の形態としては、投与方法に応じて、適宜所望の剤形に製剤化することができる。例えば、経口投与の場合、散剤、顆粒剤、錠剤、カプセル剤等の固形製剤;溶液剤、シロップ剤、懸濁剤、乳剤等の液剤等に製剤化することができる。また、腸溶性コーティング等により、腸溶剤とすることもできる。非経口投与の場合、座剤、軟膏剤、注射剤等に製剤化することができる。
製剤化に際しては、ラクトフェリン及び/又はラクトフェリン加水分解物の他に、通常製剤化に用いられている賦形剤、pH調整剤、着色剤、矯味剤等の成分を用いることができる。また、他の薬効成分や、公知の又は将来的に見出されるpDC活性化作用を有する成分等を併用することも可能である。
加えて、製剤化は剤形に応じて適宜公知の方法により実施できる。製剤化に際しては、適宜、通常製剤化に用いる担体を配合して製剤化してもよい。かかる担体としては、賦形剤、結合剤、崩壊剤、滑沢剤、安定剤、矯味矯臭剤等が挙げられる。

結合剤としては、例えば、上記賦形剤の他、ゼラチン;ポリビニルピロリドン;マクロゴール等が挙げられる。
崩壊剤としては、例えば、上記賦形剤の他、クロスカルメロースナトリウム、カルボキシメチルスターチナトリウム、架橋ポリビニルピロリドン等の化学修飾されたデンプン又はセルロース誘導体等が挙げられる。
滑沢剤としては、例えば、タルク;ステアリン酸;ステアリン酸カルシウム、ステアリン酸マグネシウム等のステアリン酸金属塩;コロイドシリカ;ピーガム、ゲイロウ等のワックス類;硼酸;グリコール;フマル酸、アジピン酸等のカルボン酸類;安息香酸ナトリウム等のカルボン酸ナトリウム塩;硫酸ナトリウム等の硫酸塩類;ロイシン;ラウリル硫酸ナトリウム、ラウリル硫酸マグネシウム等のラウリル硫酸塩;無水珪酸、珪酸水和物等の珪酸類;デンプン誘導体等が挙げられる。
安定剤としては、例えば、メチルパラベン、プロピルパラベン等のパラオキシ安息香酸エステル類;クロロブタノール、ベンジルアルコール、フェニルエチルアルコール等のアルコール類;塩化ベンザルコニウム;無水酢酸;ソルビン酸等が挙げられる。
矯味矯臭剤としては、例えば、甘味料、酸味料、香料等が挙げられる。
なお、経口投与用の液剤の場合に使用する担体としては、水等の溶剤等が挙げられる。

本発明の医薬品を摂取するタイミングは、例えば食前、食後、食間、就寝前など特に限定されない。
以下に実施例を用いて本発明をさらに具体的に説明するが、本発明はこれら実施例に限定されるものではない。
[実施例1]
協力者(n=3)から採取したヒト末梢血から単核細胞(PBMC)を回収した。ラクトフェリン(森永乳業株式会社製MLF-2)(100~200μg/mL)を添加又は非添加(対照群)した、5%ヒト血清、Glutamax、1% Penicillin Streptomycinを加えたRPMI1640培地を用いて、1.0×106cells/mLで、37℃、5%CO2条件下で培養した。培養開始24時間後に細胞を回収し、フローサイトメーターを用いて、PBMCの各免疫細胞の細胞表面マーカーCD86の発現を評価した。なお、CD86の発現(陽性)は、pDCの活性化を表す。
結果を表1に示す。いずれの試料においても、ラクトフェリン存在下で培養した場合に、計測した全細胞数におけるpDC数の割合も、pDC中のCD86陽性細胞数の割合も増加したことが認められた。また、pDC1細胞当たりのCD86発現量も増加したことが認められた。

結果を表2に示す。ラクトフェリン存在下で培養した場合に、pDC1細胞当たりのCD86とHLA-DR発現量も増加したことが認められた。

[製造例1]
本発明の組成物の一実施態様として、ラクトフェリンを含有する発酵乳の製造法を下記に示す。
まず、乳原料、ラクトフェリン(ミライ社製)、及びその他の成分等を、必要に応じて水を添加して混合溶解して調乳液を調製する。ラクトフェリンは、調乳液100g中の含有量が100mgになるように添加する。
次に、当該調乳液を、常法により、均質化処理、および加熱殺菌処理をする。加熱殺菌された殺菌調乳液に乳酸菌スターターを添加(接種)し、所定の発酵温度に保持して発酵させ、発酵物を得る。発酵によりカードが形成される。乳酸菌スターターとしては、例えば、ラクトバチルス・ブルガリクス(Lactobacillus bulgaricus)、ラクトコッカス・ラクチス(Lactococcus lactis)、ストレプトコッカス・サーモフィラス(Streptococcus thermophilus)等のヨーグルト製造に通常用いられている乳酸菌を用いることができる。
pHが目標の値に達したら、形成されたカードを撹拌により破砕し、10℃以下に冷却して、ラクトフェリン含有発酵乳を得る。
pDCはヒトの腸管にも存在することから、上記ラクトフェリン含有発酵乳を摂取することで、プラズマサイトイド樹状細胞(pDC)活性化、pDC増加、及び/又はpDCにおけるCD86やHLA-DRの発現促進効果が期待される。
本発明の組成物の一実施態様として、ラクトフェリンを含有する発酵乳の製造法を下記に示す。
まず、乳原料、ラクトフェリン(ミライ社製)、及びその他の成分等を、必要に応じて水を添加して混合溶解して調乳液を調製する。ラクトフェリンは、調乳液100g中の含有量が100mgになるように添加する。

以下の組成の原料を混合し、常法により打錠して1g当たりラクトフェリンを300mg含有する錠剤を得る。

当該ラクトフェリン含有錠菓を摂取することで、プラズマサイトイド樹状細胞(pDC)活性化、pDC増加、及び/又はpDCにおけるCD86やHLA-DR発現促進効果が期待される。
本発明の組成物の一実施態様として、ラクトフェリンを含有する錠菓の製造法を下記に示す。
以下の組成の原料を混合し、常法により打錠して1g当たりラクトフェリンを300mg含有する錠剤を得る。

Claims (7)
- ラクトフェリン及び/又はラクトフェリン加水分解物を含有する、プラズマサイトイド樹状細胞(pDC)活性化用組成物。
- 前記活性化が、単核細胞中のpDC数の割合、pDC中のCD86陽性細胞数の割合、pDCにおけるCD86発現量、及びpDCにおけるHLA-DR発現量からなる群から選択される一種又は二種以上の増加である、請求項1に記載の組成物。
- ラクトフェリン及び/又はラクトフェリン加水分解物を含有する、pDC増加用組成物。
- ラクトフェリン及び/又はラクトフェリン加水分解物を含有する、pDCにおけるCD86発現及び/又はHLA-DR発現の促進用組成物。
- 免疫機能を維持、改善、又は強化するために用いられる、請求項1~4のいずれか一項に記載の組成物。
- 飲食品である、請求項1~5のいずれか一項に記載の組成物。
- 医薬品である、請求項1~5のいずれか一項に記載の組成物。
