Abstract
本発明の課題は、ミトコンドリアへの送達性の高いビタミンK誘導体からなるミトコンドリア機能障害改善剤を提供することである。 下記一般式(1)(式中、R1およびR2はそれぞれ水素原子、またはグリシン、N-アシルグリシン、N-アルキルグリシン、N,N-ジアルキルグリシン、N,N,N-トリアルキルグリシン、アシル、ジカルボン酸ヘミエステル及びその塩から選ばれる置換基を意味し、R1, R2の少なくとも一方はグリシン、N-アシルグリシン、N-アルキルグリシン、N,N-ジアルキルグリシン、N,N,N-トリアルキルグリシン、アシル、ジカルボン酸ヘミエステル及びその塩である。R3は下記一般式(2)もしくは下記一般式(3)で示される基を表す。nは1~7の整数を意味する。)で表される活性型ビタミンKのカルボン酸エステル類またはその塩の少なくとも一種類からなる、ミトコンドリア機能障害改善剤、筋萎縮性側索硬化症改善剤、神経変性改善剤、アルツハイマー病改善剤、及びパーキンソン病改善剤、及び 前記一般式(1)において、R1及びR2は、R4OOCCH2CH2CO-及びR4OOCCH2CH2CH2CO-からなる群より選択されるカルボン酸残基である。R3は上記一般式(2)または(3)の意味を有する。R4はC1~C3のアルキル基である。)で表される活性型ビタミンKのカルボン酸エステル類またはその塩の少なくとも一種類からなる、ミトコンドリア機能障害改善剤、筋萎縮性側索硬化症改善剤、神経変性改善剤、アルツハイマー病改善剤、及びパーキンソン病改善剤。

本出願は、2020年03月17日付け出願の日本国特許出願2020-046864号の優先権を主張しており、ここに折り込まれるものである。
本発明はミトコンドリア機能障害改善剤、特にビタミンKヒドロキノン誘導体を有効成分とするミトコンドリア機能障害改善剤に関する。
ミトコンドリア機能障害は神経変性疾患や2型糖尿病など、様々な疾病の原因と考えられている。
例えばアルツハイマー病(AD)は、最も一般的な神経変性疾患であり、アミロイド-βペプチド(Aβ)の強い凝集と過剰リン酸化タウ(p-tau)を病理学的特徴とする壊滅的な疾患である。AD病変を標的とした進行を抑制できる効果的かつ安全な治療法の開発が必要とされている。しかし、今日まで有効な治療法はない。ADの臨床試験では、Aβのクリアランスとp-tauの抑制に焦点が当てられている。
例えばアルツハイマー病(AD)は、最も一般的な神経変性疾患であり、アミロイド-βペプチド(Aβ)の強い凝集と過剰リン酸化タウ(p-tau)を病理学的特徴とする壊滅的な疾患である。AD病変を標的とした進行を抑制できる効果的かつ安全な治療法の開発が必要とされている。しかし、今日まで有効な治療法はない。ADの臨床試験では、Aβのクリアランスとp-tauの抑制に焦点が当てられている。
孤発性と家族性のヒトAD脳サンプル、AD誘導多能性幹細胞(iPSC)由来ニューロン、およびADトランスジェニック動物モデルにおいて、ADではニューロンのミトコンドリア品質管理の障害とミトコンドリア機能低下を呈しており、これらの機能不全ミトコンドリアの蓄積がADの病理学的要因Aβやp-tauの増加に寄与し、また、Aβやp-tauの増加は逆にミトコンドリアの機能不全を増悪する(非特許文献1~5)。ミトコンドリア機能障害によるエネルギー不足とAβ凝集はシナプス毒性と記憶喪失をもたらし(非特許文献3、6)、Aβとp-tauの増加はミトコンドリアの軸索輸送欠陥を誘発しシナプススターベーション、ATP枯渇、最終的に神経変性をもたらす(非特許文献7)。機能不全ミトコンドリア(ミトコンドリア機能やミトコンドリア品質管理)を薬学的あるいは遺伝子的方法で回復または強化することによりAD病変であるAβ毒性を低下させタウ過剰リン酸化を無効にできることが明らかにされている(非特許文献1)。
UBIAD1はビタミンK3 ヒドロキノンとゲラニルゲラニルピロリン酸からメナヒドロキノン-4 (MKH)を生成する酵素であり(化1)(非特許文献8、9)、AD患者では脳のUBIAD1発現量が低下しMKHレベルも低下している(非特許文献10)。UBIAD1は膜電位脱分極剤で脱分極されたミトコンドリアの膜電位を上昇させることが明らかにされ、ミトコンドリア機能障害を回復しエネルギー産生を亢進することが示唆されている(非特許文献11)。
また、パーキンソン病(PD)はドーパミン作動性ニューロンの神経変性疾患であり、PDにおいても、ミトコンドリア機能障害やミトコンドリア品質管理の破綻が神経細胞死をもたらしている(非特許文献12)。PDのミトコンドリア品質管理に関与するマイトファジーにはPINK1/Parkinのタンパク質が必須であり、ADのマイトファジーにおいてもPINK1/Parkinが含まれている。PDモデルでもUBIAD1がミトコンドリア品質管理を救済することが報告されている(非特許文献13)。筋萎縮性側索硬化症(ALS) 患者においても上位運動ニューロンのミトコンドリア機能障害が明らかにされている(非特許文献14、15)。

以上の背景から、本発明者は、活性体メナヒドロキノン-4(MKH)あるいは活性体フィロヒドロキノン(PKH)をミトコンドリアに送達させることで、ミトコンドリア機能障害とミトコンドリア品質管理の回復強化が可能となり、その結果AD病変(Aβとp-tauの増加)を軽減または回復できると仮説を立てた。
そして、本発明者は仮説を実現できる低分子化合物を創薬標的とし、MKH、PKHのプロドラッグから候補化合物の探索を行うこと、ミトコンドリア機能障害改善剤の開発、ひいてはADの予防、治療薬の開発、PDの治療薬、ALSの治療薬の開発を企図した。
そして、本発明者は仮説を実現できる低分子化合物を創薬標的とし、MKH、PKHのプロドラッグから候補化合物の探索を行うこと、ミトコンドリア機能障害改善剤の開発、ひいてはADの予防、治療薬の開発、PDの治療薬、ALSの治療薬の開発を企図した。
現在、アルツハイマー病(AD)薬は、コリンエステラーゼ阻害薬とNMDA型受容体アンタゴニストが存在するが、AD症状の管理だけであり有効な治療法はない。AD病変を標的とし進行を変更できる効果的かつ安全な治療法の開発が必要とされており、ADの臨床試験では、Aβとp-tauの増加抑制に焦点が当てられている。
本発明は、前記従来技術に鑑みなされたものであり、その解決すべき課題はミトコンドリア機能障害及びミトコンドリア品質管理を効果的に行い得るビタミンK誘導体を提供することにある。

前述のとおり、本発明者等は特定の構造を有するビタミンKヒドロキノン誘導体が、ミトコンドリアへの送達性に優れることを見出し、本発明を完成するに至った。
すなわち、本発明にかかるミトコンドリア機能障害改善剤(ビタミンKヒドロキノン誘導体)は下記一般式(1)で表される。

また、本発明にかかるビタミンKヒドロキノンジカルボン酸ダブルエステルは、下記一般式(4)で示される。
(一般式(4)において、R1及びR2は、R4OOCCH2CH2CO-及びR4OOCCH2CH2CH2CO-からなる群より選択されるカルボン酸残基である。R3は上記一般式(2)または(3)の意味を有する。R4はC1~C3のアルキル基である。)
本発明は、前記一般式(1)及び一般式(4)で表される、ビタミンKヒドロキノンのカルボン酸エステルまたはその塩またはビタミンKヒドロキノンジカルボン酸ダブルエステルの少なくとも一種類を有効成分としたミトコンドリア機能障害改善剤は、活性型ビタミンKをミトコンドリアに効率的に送達させ、アルツハイマー病やパーキンソン病や筋萎縮性側索硬化症などの神経変性疾患、2型糖尿病などの予防、治療に有効である。
前記一般式(1)で表される化合物は、単独で製剤に含有させることもできるし、その塩として製剤に配合することもできる。前記一般式(4)で表される化合物は、単独で製剤に含有させることもできる。本発明において、窒素置換基を有するカルボン酸残基R1、R2としては次のものが例示される。
窒素原子に対し水素原子ないし、1または2のアルキル基、アシル基が結合したもの。
前記アルキル基としては、炭素数1~6の直鎖、もしくは分枝のアルキル基であり次のものが例示される。メチル基、エチル基、n-プロピル基、n-ブチル基、n-ペンチル基、n-ヘキシル基、イソプロピル基、イソブチル基、1-メチルプロピル基、tert-ブチル基、1-エチルプロピル基、イソアミル基。上記アルキル基としてはメチル基、エチル基が好ましい。また、アシル基を有する場合の炭化水素鎖も同様に定義可能である。

前記アルキル基としては、炭素数1~6の直鎖、もしくは分枝のアルキル基であり次のものが例示される。メチル基、エチル基、n-プロピル基、n-ブチル基、n-ペンチル基、n-ヘキシル基、イソプロピル基、イソブチル基、1-メチルプロピル基、tert-ブチル基、1-エチルプロピル基、イソアミル基。上記アルキル基としてはメチル基、エチル基が好ましい。また、アシル基を有する場合の炭化水素鎖も同様に定義可能である。
アミノ基とカルボニル基の間は、好ましくは炭素数1~7の直鎖、分枝または環状のアルキレン基で結合される。分枝状のアルキレン基としては、次のものが例示される。イソプロピル、イソブチル、tert-ブチル、1-エチルプロピルなどのアルキル基から誘導されたもの。
前記環状アルキレン基としては、次のものが例示される。
シクロペンタン環、シクロヘキサン環、あるいはメチルシクロヘキサン環などを構造中に含むもの。上記アルキレン基としては、メチレン基あるいはエチレン基が特に好ましい。
前記環状アルキレン基としては、次のものが例示される。
シクロペンタン環、シクロヘキサン環、あるいはメチルシクロヘキサン環などを構造中に含むもの。上記アルキレン基としては、メチレン基あるいはエチレン基が特に好ましい。
ハロゲン化水素酸塩としては、塩酸塩、臭化水素酸塩などが好ましい。本発明において、ハロゲン化水素酸塩は結晶化ないし固形化する場合が多く、製剤にあたっての取り扱いが容易になるという利点がある。その他の塩としては次のものが例示される。アルキルスルホン酸塩としてはメタンスルホン酸塩等、糖酸塩としてはグルコン酸塩、グルコヘプタン酸塩、ラクトビオン酸塩等。
本発明において、ジカルボン酸残基R1、R2はジカルボン酸及びそのアルカリ金属塩またはメグルミン塩の残基から選ばれる。ジカルボン酸残基のカルボニル基間は炭素数2~4の直鎖のアルキレン基で結合される。アルキレン基としては、エチレン基またはプロピレン基が特に好ましい。アルカリ金属塩としてナトリウム塩、カリウム塩が好ましい。
本発明において、ジカルボン酸ダブルエステルはジカルボン酸残基のカルボニル基間は炭素数2~4の直鎖のアルキレン基で結合される。アルキレン基としては、エチレン基またはプロピレン基が特に好ましい。ダブルエステルのアルコール残基は炭素1~3のアルキル基が好ましく特にエチル基が好ましい。
また、本発明において、一般式(1)及び一般式(4)で表される化合物の製造方法は種々考えられるが,代表的な方法を述べれば以下の通りである。
一般式(5)で表されるビタミンK類を還元剤で還元し、一般式(6)で表されるビタミンKヒドロキノンとし、この ビタミンKヒドロキノンと、窒素置換基を有するカルボン酸、若しくはその反応性酸誘導体またはこれらのハロゲン化水素酸塩、または酸無水物とを常法によりエステル化反応を行なうことにより、本発明の目的物質(1)を得ることができる。ここで用いられる還元剤はビタミンK類のナフトキノン骨格をナフトヒドロキノン骨格に還元するものであり、水素化ホウ素ナトリウム、ハイドロサルファイトナトリウム、トリ-n-ブチルホスフィン、塩化亜鉛、塩化第一スズ、亜鉛末などを挙げることができる。
ビタミンKヒドロキノンのエステル化反応は常法に従うが、1級、2級アミノ基あるいは側鎖に水酸基、チオール基を有するアミノ酸のエステル化を行なう際は、tert-ブトキシカルボニル基(以下 t-BOC 基と略記) 、ベンジルオキシカルボニル基(以下Z基と略記)、9-フルオレニルメトキシカルボニル基(以下FMOC基と略記)などの適切な保護基で保護して用い、N,N-ジアルキルアミノ酸はハロゲン化水素酸塩を用いて、ジシクロヘキシルカルボジイミド(以下DCC と略記) 、N,N-ジサクシニミドオキザレート(以下DSO と略記) などの活性エステル化試薬の存在下に反応を行なうことが好ましい結果を与える。この際溶媒としては無水ピリジンが好ましい。また、反応性酸誘導体を用いる方法では、酸ハロゲナイトとりわけ、酸クロリドを用いる方法が好ましい結果を与える。この際溶媒としては無水ベンゼン-無水ピリジン混合物が好ましい。ハロゲン化水素酸塩、アルキルスルホン酸塩、糖酸塩は常法により遊離のビタミンKヒドロキノン窒素含有カルボン酸エステルとハロゲン化水素酸、アルキルスルホン酸、酸性糖のラクトン体を反応させて製造する。また、 N-アシルアミノ酸エステルを製造した後、常法によりハロゲン化水素酸で脱保護基化することによってハロゲン化水素酸塩を製造することができる。
一般式(4)で表されるジカルボン酸ダブルエステルは、一般式(5)で表されるビタミンK類を酢酸酸性下、亜鉛末、酸無水物とを加熱下反応させビタミンKヒドロキノンカルボン酸ヘミエステルとし、さらに酸性触媒下、アルコールでエステル化させ目的物質(4)を得る。また、上記一般式(1)で得られたビタミンKヒドロキノンカルボン酸ヘミエステルを酸性触媒下、アルコールでエステル化させ目的物質(4)を得る。
本発明のビタミンKヒドロキノン誘導体は投与剤型の自由度が高く、経口投与、経皮投与、経鼻投与、注射投与などが採用できる。添加剤として、等張化剤、緩衝剤、pH調節剤、可溶化剤、増粘剤(分散剤)、粘膜付着剤、安定化剤(抗酸化剤)、保存剤(防腐剤)、吸収促進剤等を適宜配合することにより、周知の方法で製剤化することができる。また、pH調節剤、増粘剤、分散剤等を添加し、薬物を懸濁化させることによって、安定な点眼剤や点鼻液剤を得ることもできる。
等張化剤としては、医薬上、薬理学的に(製薬上)又は生理学的に許容されるものであれば特に制限されず、例えば、イオン性等張化剤としては、塩化ナトリウム、塩化カリウム、塩化カルシウム、塩化マグネシウム等が挙げられ、非イオン性等張化剤としてはグリセリン、プロピレングリコール、ソルビトール、マンニトール等が挙げられる。
緩衝剤としては、医薬上、薬理学的に(製薬上)又は生理学的に許容されるものであれば特に制限されず、例えば、リン酸、リン酸塩、クエン酸、酢酸若しくはε-アミノカプロン酸等を挙げることができる。
pH調節剤としては、医薬上、薬理学的に(製薬上)又は生理学的に許容されるものであれば特に制限されず、例えば、塩酸、リン酸、クエン酸、酢酸、水酸化ナトリウム、水酸化カリウム、ホウ酸、ホウ砂、炭酸ナトリウム、炭酸水素ナトリウム等が挙げられる。点眼剤のpHは眼科製剤に許容される範囲内にあればよいが、4.0~9.0であり、より好ましくは5.5~8.5となる範囲が挙げられる。
可溶化剤としては、医薬上、薬理学的に(製薬上)又は生理学的に許容されるものであれば特に制限されず、例えば、ポリオキシエチレンヒマシ油、ポリオキシエチレン硬化ヒマシ油、ポリオキシエチレンソルビタン脂肪酸エステル、ビタミンE TPGS、ポリオキシエチレン脂肪酸エステル、ポリオキシエチレンポリオキシプロピレングリコール、ショ糖脂肪酸エステル等が挙げられる。
増粘剤、分散剤及び粘膜付着剤としては、医薬上、薬理学的に(製薬上)又は生理学的に許容されるものであれば特に制限されず、例えば、ヒドロキシプロピルメチルセルロース若しくはヒドロキシプロピルセルロース等のセルロース系高分子;ポリビニルアルコール;又はポリビニルピロリドン等を挙げることができる。
安定化剤としては、医薬上、薬理学的に(製薬上)又は生理学的に許容されるものであれば特に制限されず、例えば、エデト酸、エデト酸一ナトリウム、エデト酸二ナトリウム、エデト酸四ナトリウム、クエン酸ナトリウム等が挙げられ、エデト酸ナトリウムは水和物であってもよい。
抗酸化剤としては、医薬上、薬理学的に(製薬上)又は生理学的に許容されるものであれば特に制限されず、例えば、アスコルビン酸、ビタミンE、ジブチルヒドロキシトルエン、ブチルヒドロキシアニソール、エリソルビン酸ナトリウム、没食子酸プロピル、亜硫酸ナトリウム等が挙げられる。
保存剤(防腐剤)としては、医薬上、薬理学的に(製薬上)又は生理学的に許容されるものであれば特に制限されず、例えば、ベンザルコニウム塩化物、ベンザルコニウム臭化物、ベンゼトニウム塩化物、ソルビン酸、ソルビン酸カリウム、パラオキシ安息香酸メチル、パラオキシ安息香酸プロピル、クロロブタノール等が挙げられ、これらの保存剤を組み合わせて使用することもできる。
以上説明したように本発明にかかるミトコンドリア機能障害改善剤は、ミトコンドリアへ活性型ビタミンK送達を可能とし、ミトコンドリア機能障害に起因する各種疾患の改善を行うことができる。
以下、本発明の好適な実施形態について詳細な説明を行う。
孤発性と家族性のヒトAD脳サンプル、AD誘導多能性幹細胞(iPSC)由来ニューロン、およびADトランスジェニック動物モデルにおいて、ADではニューロンのミトコンドリア品質管理の障害とミトコンドリア機能低下を呈しており、これらの機能不全ミトコンドリアの蓄積がAD病変要因であるAβやp-tauの増加に寄与していることが明らかにされた(非特許文献1~5)。機能不全ミトコンドリア(ミトコンドリア機能やミトコンドリア品質管理)を薬学的あるいは遺伝子的方法で回復または強化することによりAβ毒性を低下させタウ過剰リン酸化を無効にできることが明らかにされている(非特許文献1)。UBIAD1はメナヒドロキノン-4(MKH)を生合成する酵素であり(化1)、AD患者では脳のUBIAD1発現が低下しており、MKHレベルも低下していること、UBIAD1は分極したミトコンドリアの膜電位を上昇させること、即ちミトコンドリア機能障害の改善が示されている(非特許文献13)。

そこで本発明者は、AD患者の脳神経ミトコンドリアへのMKHの効率的送達によってミトコンドリア機能障害やミトコンドリア品質管理の回復・強化を可能とし、AD病変(Aβとp-tau)を軽減または回復できると仮説を立て、MKH、PKH送達を可能にしAD病変(Aβとp-tauの増加)を軽減または回復をもたらす低分子化合物の開発を創薬標的とした。
MKH はVitamin K(VK)の1種類であるvitamin K2(20) (menaquinone-4, MK-4)の2電子還元体であり、小胞体において、VKの活性体として、ビタミンK作用の一つであるビタミンK依存性タンパク質(VKDP)の翻訳後修飾において、Glu残基をGla残基にカルボキシル化する酵素(GGCX)の補因子として機能する。MKHは極めて酸化され易いため、臨床にはキノン型のMK-4が用いられているが、光安定性が低く、光毒性が高い、水難溶性、還元的活性化過程が必須であることがMK-4によるMKHの薬物送達上の課題であった。本発明者は、これまでに還元的活性化過程を必要としない、光安定性が高く、光毒性の無いMKH送達剤をMKHのプロドラッグ化によって可能にしている。

まず、本研究で仮説したMKH送達により、ミトコンドリア機能の回復強化をもたらしAβタンパク毒性の低下を可能にできる低分子化合物を創薬標的としてMKHプロドラッグを取り上げ、その機能を検証し、AD予防、治療薬としての可能性をin vitroで評価した。
また、パーキンソン病(PD)はドーパミン作動性ニューロンの神経変性疾患であり、PDにおいてもミトコンドリア機能障害やミトコンドリア品質管理の破綻が神経細胞死をもたらしている。さらに、PDモデルにおいてUBIAD1がミトコンドリア品質管理を救済することが報告されている。
そこで、低分子化合物の創薬標的としてMKHプロドラッグを用いてミトコンドリア機能障害の回復が可能か検証し、PDの治療薬としての可能性をin vitroで評価した。
また、パーキンソン病(PD)はドーパミン作動性ニューロンの神経変性疾患であり、PDにおいてもミトコンドリア機能障害やミトコンドリア品質管理の破綻が神経細胞死をもたらしている。さらに、PDモデルにおいてUBIAD1がミトコンドリア品質管理を救済することが報告されている。
そこで、低分子化合物の創薬標的としてMKHプロドラッグを用いてミトコンドリア機能障害の回復が可能か検証し、PDの治療薬としての可能性をin vitroで評価した。
1)アルツハイマー病(AD)の予防、治療薬としての可能性
単一ニューロン・アストロサイト共培養標本にAβペプチドの処置を施したAβペプチドによる神経細胞障害モデルにおいて、MKH誘導体の神経変性改善効果を評価した。MKH誘導体によるMKH送達では、誘導体のミトコンドリア親和性とMKHへの再変換性が影響することが予測されるので、親和性が高いと予想されるカチオン性誘導体(MKH-DMG)と、細胞内再変換性が高いアニオン性誘導体(MKH-SUC)及び臨床に用いられているMKHの酸化体(MK-4)に関して評価した(化7)。
単一ニューロン・アストロサイト共培養標本にAβペプチドの処置を施したAβペプチドによる神経細胞障害モデルにおいて、MKH誘導体の神経変性改善効果を評価した。MKH誘導体によるMKH送達では、誘導体のミトコンドリア親和性とMKHへの再変換性が影響することが予測されるので、親和性が高いと予想されるカチオン性誘導体(MKH-DMG)と、細胞内再変換性が高いアニオン性誘導体(MKH-SUC)及び臨床に用いられているMKHの酸化体(MK-4)に関して評価した(化7)。
β-アミロイドによる神経障害に対するMKH誘導体の改善効果
生後0-1日齢ICRマウスの大脳皮質より単離したアストロサイトを培養(2週間)後、スライドガラス上にドット状に領域を区分して培養した。さらに1週間培養後、生後0-1日齢ICRマウス海馬由来のニューロンを播種し、共培養した。ニューロン播種1日よりAβ25-35 (1 μM)並びに試験化合物(0.3 μM)を3日間暴露し、ニューロンの形態変化を観察した。免疫染色法を用いて神経の樹状突起(MAP2抗体)、軸索(Tau抗体)を染色し解析した。樹状突起ならびに軸索はSholl analysis法により、樹状突起及び軸索の分岐・伸長の変化について評価した。
ニューロンの染色結果を図1に示した。神経軸索の分岐・伸長と神経樹状突起の分岐・伸長はどちらもAβ25-35によって有意に抑制され神経障害が観察された。MKH誘導体(DMG(MKH-DMG)とSUC(MKH-SUC))は神経軸索と神経樹状突起どちらの神経障害も有意に改善し(図2,3)、神経障害の改善効果が明らかとなった。MK4(MK-4)はいずれの神経障害に対しても改善効果は観察されなかった。MKH誘導体は低濃度0.3μMでβ-アミロイド神経変性に対する改善効果を有することから、高い安全性が予想される。

ニューロンの染色結果を図1に示した。神経軸索の分岐・伸長と神経樹状突起の分岐・伸長はどちらもAβ25-35によって有意に抑制され神経障害が観察された。MKH誘導体(DMG(MKH-DMG)とSUC(MKH-SUC))は神経軸索と神経樹状突起どちらの神経障害も有意に改善し(図2,3)、神経障害の改善効果が明らかとなった。MK4(MK-4)はいずれの神経障害に対しても改善効果は観察されなかった。MKH誘導体は低濃度0.3μMでβ-アミロイド神経変性に対する改善効果を有することから、高い安全性が予想される。
2)MKH誘導体によるミトコンドリア機能障害の改善効果
神経系細胞において、complex I阻害剤ロテノン(10mM)、complex II阻害剤(3-nitro propionic acid (3-NP)、complex III阻害剤(antimycin A)、脱分極剤(カルボニルシアニド-m-クロロフェニルヒドラゾン, CCCP)によるミトコンドリア機能障害に対する、MKH誘導体とPKH誘導体の効果を評価した。細胞死(cell titer blueで生存測定)、ATP産生阻害(cell titer gloでATP測定)、膜電位低下(JC-1染色 脱分極(Green), 過分極 (Red))で評価した。
MKH誘導体投与によってcomplex I阻害剤ロテノン処理による細胞死は抑制され(図4)、ATP産生阻害と(図5)、ミトコンドリア膜電位低下を回復した(図6)。PKH誘導体投与においてロテノン処理による細胞死は抑制され(図7)、ATP産生阻害と(図8)、ミトコンドリア膜電位低下を回復した(図9)。
神経系細胞において、complex I阻害剤ロテノン(10mM)、complex II阻害剤(3-nitro propionic acid (3-NP)、complex III阻害剤(antimycin A)、脱分極剤(カルボニルシアニド-m-クロロフェニルヒドラゾン, CCCP)によるミトコンドリア機能障害に対する、MKH誘導体とPKH誘導体の効果を評価した。細胞死(cell titer blueで生存測定)、ATP産生阻害(cell titer gloでATP測定)、膜電位低下(JC-1染色 脱分極(Green), 過分極 (Red))で評価した。
MKH誘導体投与によってcomplex I阻害剤ロテノン処理による細胞死は抑制され(図4)、ATP産生阻害と(図5)、ミトコンドリア膜電位低下を回復した(図6)。PKH誘導体投与においてロテノン処理による細胞死は抑制され(図7)、ATP産生阻害と(図8)、ミトコンドリア膜電位低下を回復した(図9)。
また、各誘導体は、膜電位低下を回復(図10)した。さらに、complex III阻害剤(antimycin A)による生存率低下、ATP産生低下、膜電位低下を回復(図11)した。加えて、脱共役剤(CCCP)による生存率低下を回復させた(図12)。また、各誘導体は、ミトコンドリア生合成を活性化させるPGC-1α(peroxisome proliferator-activated receptor-c coactivator-1α)の発現を増加させた。
以上の結果、MKH誘導体とPKH誘導体はミトコンドリア機能障害(膜電位低下ミトコンドリア)を回復できることが明らかになった。

3)PD治療薬としての可能性
孤発性PDはComplex I活性が低下しており、ロテノンcomplex I阻害モデルは運動緩慢、硬直、振戦などPDの特徴的症状に加えてドーパミン作動性ニューロンにおいて形態学的及び機能的損傷をもたらす。また、Complex I阻害によって損傷したミトコンドリア(膜電位低下ミトコンドリア)は、ミトコンドリア品質管理を誘導することから遺伝性PDの評価としても用いられている。したがって、得られた結果からMKH誘導体とPKH誘導体はロテノンPDモデルにおいて有効であることが明らかになりPD治療薬としての可能性が明らかになった。

・MKH-DMGとMKH-SUCは、ミトコンドリア機能(呼吸鎖)障害を改善できることが明らかになった。
これらの結果は、AD患者の脳神経ミトコンドリアへのMKHの効率的送達によって、ミトコンドリア機能障害やミトコンドリア品質管理の回復・強化を可能とし、AD病変(Aβとp-tauの増加)を軽減・回復を可能にする仮説を支持するものであり、MKH誘導体は仮説に基づいた創薬標的の可能性が高いことが示された。
また、MKH誘導体のロテノンPDモデルにおける効果はミトコンドリア品質管理の回復・強化によるPD治療薬としての可能性を支持するものである。
また、ミトコンドリア機能回復による筋萎縮性側索硬化症治療薬としての可能性を支持するものである。

また、ミトコンドリア機能回復による筋萎縮性側索硬化症治療薬としての可能性を支持するものである。
したがって、本発明者はMKH誘導体をミトコンドリア機能障害や品質管理の回復・強化を作用機構とするADの予防・治療薬ならびにPD治療薬の創薬標的として提案する。

以下に実施例を挙げて本発明を更に具体的に説明するが、本発明がこれらに限定されるものではない。

製造方法A
アミノ酸0.1 molを蒸留水-ジオキサン(1:1、v/v) 100mlに溶解し、トリエチルアミン30 mlを加え、ジ-tert-ブチルジカルボネートを徐々に加え30分間室温で撹拌する。減圧下ジオキサンを留去し、炭酸水素ナトリウム水溶液(0.5M)50 mlを加え酢酸エチル100 mlで洗う。酢酸エチル層を50 mlの炭酸水素ナトリウム液で洗い、水層を合わせて氷冷下でクエン酸水溶液(0.5M)を加えて酸性(pH3)とし、塩化ナトリウムを飽和させた後、酢酸エチルで抽出する(100 ml×3回)、抽出液を無水硫酸ナトリウムで脱水後減圧下溶媒を留去し、油状残渣をイソプロピルエーテルを加えるか、または冷却にて結晶化させて、N-t-BOC-アミノ酸を得る。ビタミンK6.75 mmolをイソプロピルエーテル40 mlに溶解し、水素化ホウ素ナトリウム47 mmolをメタノール15 mlに溶解して加え、溶液の黄色が無色になるまで室温で撹拌する。反応液にイソプロピルエーテル60 mlと蒸留水100 mlを加え、イソプロピルエーテル層を分離し、更に水層にイソプロピルエーテル100 mlを加えて可溶画分を抽出し、イソプロピルエーテル層を合わせて無水硫酸ナトリウムで脱水後減圧下濃縮する。残渣に n-ヘキサンを加えて白色沈殿を析出させてビタミンKヒドロキノンを得る。

ビタミンKヒドロキノン、 N-t-BOC-アミノ酸13.55 mmol 、DCC 13.55 mmolを無水ピリジン50 mlに加え室温で20時間撹拌する。溶媒を減圧下留去し、残渣に酢酸エチルを加えて可溶画分を抽出する(100ml×2回)、抽出液を減圧下濃縮し、残渣をシリカゲルカラムクロマトグラフィー(溶離溶媒; n-ヘキサン-イソプロピルエーテル)で分離精製し、ビタミンKヒドロキノン-1,4-ビス-N-t-BOC-アミノ酸を得る。ビタミンKヒドロキノン-1,4-ビス-N-t-BOC-アミノ酸を少量のアセトンに溶解し、塩酸-ジオキサン(2.5~4.0N) をエステル量の約20倍モル量の塩酸量に相当する量加え1時間撹拌後、減圧下溶媒を留去する。残渣をアセトン-メタノール系で再結晶してビタミンKヒドロキノン-1,4-ビス-アミノ酸エステルの塩酸塩を得る。
製造方法B
ビタミンK 6.75 mmolをイソプロピルエーテル40 mlに溶解し、水素化ホウ素ナトリウム47 mmolをメタノール15 mlに溶解して加え、溶液の黄色が無色になるまで室温で撹拌する。反応液にイソプロピルエーテル60mlと蒸留水100 mlを加え、イソプロピルエーテル層を分離し、更に水層にイソプロピルエーテル100 mlを加えて可溶画分を抽出、イソプロピルエーテル層を合わせて無水硫酸ナトリウムで脱水後減圧下濃縮する。残渣に n-ヘキサンを加えて白色沈殿を析出させてビタミンKヒドロキノンを得る。ビタミンKヒドロキノン、塩酸N,N-ジアルキルアミノ酸13.55 mmolまたは塩酸N,N,N-トリアルキルアミノ酸13.55 mmol、DCC 13.55 mmol を無水ピリジン50mlに加え室温で20時間撹拌する。溶媒を減圧下留去し、残渣を、蒸留水に懸濁させ炭酸水素ナトリウムを加えて溶液のpHを7~8にした後に酢酸エチルで抽出する(100 ml×3回)、抽出液を無水硫酸ナトリウムで脱水後減圧下溶媒を留去し、残渣をシリカゲルカラムクロマトグラフィー(溶離溶媒;イソプロピルエーテル-酢酸エチル)で分離精製し、ビタミンKヒドロキノン-1,4-ビス-N,N-ジアルキルアミノ酸エステルまたはビタミンKヒドロキノン-1,4-ビス-N,N,N-トリアルキルアミノ酸エステルを得る。

ビタミンK 6.75 mmolをイソプロピルエーテル40mlに溶解し、ハイドロサルファイトナトリウム50 mmolを蒸留水50mlに溶解して加え、イソプロピルエーテルが褐色を呈し、さらに無色になるまで室温で撹拌する。イソプロピルエーテル層を分離し、更に水層にイソプロピルエーテル100 mlを加えて可溶画分を抽出、イソプロピルエーテル層を合わせて無水硫酸ナトリウムで脱水後減圧下濃縮する。残渣にn-ヘキサンを加えて白色沈殿を析出させてビタミンKヒドロキノンを得る。ビタミンKヒドロキノンに塩酸N,N-ジアルキルアミノ酸6.75 mmol、DCC 6.75 mmolを加え無水ピリジン50ml中で20時間撹拌する。溶媒を減圧下留去し、残渣を、蒸留水に懸濁させ炭酸水素ナトリウムを加えて溶液のpHを7~8にした後酢酸エチルで抽出する(100 ml×3回)、抽出液を無水硫酸ナトリウムで脱水後減圧下溶媒を留去し、残渣をシリカゲルカラムクロマトグラフィー(溶離溶媒;イソプロピルエーテル-酢酸エチル、3:2)で分離精製し、ビタミンKヒドロキノン-1-N,N-ジアルキルアミノ酸エステルおよびビタミンKヒドロキノン-4-N,N-ジアルキルアミノ酸エステルを得る。
ビタミンK 6.75 mmolをイソプロピルエーテル40mlに溶解し、ハイドロサルファイトナトリウム50 mmolを蒸留水50mlに溶解して加え、イソプロピルエーテルが褐色を呈し、さらに無色になるまで室温で撹拌する。イソプロピルエーテル層を分離し、更に水層にイソプロピルエーテル100 mlを加えて可溶画分を抽出、イソプロピルエーテル層を合わせて無水硫酸ナトリウムで脱水後減圧下濃縮する。残渣にn-ヘキサンを加えて白色沈殿を析出させてビタミンKヒドロキノンを得る。ビタミンKヒドロキノンに塩酸N,N-ジアルキルアミノ酸6.75 mmol、DCC 6.75 mmolを加え無水ピリジン50ml中で20時間撹拌する。溶媒を減圧下留去し、残渣を、蒸留水に懸濁させ炭酸水素ナトリウムを加えて溶液のpHを7~8にした後酢酸エチルで抽出する(100 ml×3回)、抽出液を無水硫酸ナトリウムで脱水後減圧下溶媒を留去し、残渣をシリカゲルカラムクロマトグラフィー(溶離溶媒;イソプロピルエーテル-酢酸エチル、3:2)で分離精製し、ビタミンKヒドロキノン-1-N,N-ジアルキルアミノ酸エステルおよびビタミンKヒドロキノン-4-N,N-ジアルキルアミノ酸エステルを得る。

製造方法E
ビタミンKヒドロキノン-1,4-ビス-N,N-ジアルキルアミノ酸エステル又はビタミンKヒドロキノン-1,4-ビス-ピリジンカルボン酸2mmolをアセトン20mlに溶解し、塩酸-ジオキサン(2.5~4.0 N)を塩酸量がエステルの10倍モル量に相当する量加え、溶媒を減圧下留去し、残渣をアセトン-メタノールで再結晶してビタミンKヒドロキノン-1,4-ビス-N,N-ジアルキルアミノ酸又はビタミンKヒドロキノン-1,4-ビス-ピリジンカルボン酸の塩酸塩を得る。

ビタミンKヒドロキノン-1,4-ビス-N,N-ジアルキルアミノ酸又はビタミンKヒドロキノン-1,4-ビス-ピリジンカルボン酸2mmolをジクロロメタン20mlに溶解し、アルキルスルホン酸2mmolを加え撹拌する。析出する結晶を濾取してビタミンKヒドロキノン-1,4-ビス-N,N-ジアルキルアミノ酸エステル又はビタミンKヒドロキノン-1,4-ビス-ピリジンカルボン酸エステルのアルキルスルホン酸塩を得る。
ビタミンKヒドロキノン-1,4-ビス-N,N-ジアルキルアミノ酸又はビタミンKヒドロキノン-1,4-ビス-ピリジンカルボン酸2mmolをジクロロメタン20mlに溶解し、アルキルスルホン酸2mmolを加え撹拌する。析出する結晶を濾取してビタミンKヒドロキノン-1,4-ビス-N,N-ジアルキルアミノ酸エステル又はビタミンKヒドロキノン-1,4-ビス-ピリジンカルボン酸エステルのアルキルスルホン酸塩を得る。

ビタミンK 4.55 mmolをイソプロピルエーテル40mlに溶解し、水素化ホウ素ナトリウム31.5 mmolをメタノール15 mlに溶解して加え、溶液の黄色が無色になるまで室温で撹拌する。反応液にイソプロピルエーテル60mlと精製水100mlを加え、イソプロピルエーテル層を分離し、更に水層にイソプロピルエーテル100mlを加えて可溶画分を抽出、イソプロピルエーテル層を合わせて、無水硫酸ナトリウムで脱水後、減圧下溶媒を留去する。残渣にジメチルアミノピリジン8.97 mmol、ジカルボン酸無水物18.0 mmolを加え、イソプロピルエーテル-ジオキサン(6:4、v/v)100mlに溶解して、室温で3時間撹拌後、50~60℃に加熱しながら2時間反応させ、さらに室温で放冷しながら10時間反応させる。反応液に精製水100mlを加え、イソプロピルエーテル層を分離し、無水硫酸ナトリウムで脱水後、減圧下溶媒を留去する。残渣をイソプロピルエーテルに懸濁し、遠心して得た沈殿物に酢酸エチル100mlと精製水100mlを加え酢酸エチル可溶画分を抽出し、無水硫酸ナトリウムで脱水後、減圧下溶媒を留去する。残渣をイソプロピルエーテルに懸濁し不溶物を酢酸エチルで再結晶して、ビタミンKヒドロキノン-1,4-ビス-ジカルボン酸ヘミエステルを得る。
ビタミンK 4.55 mmolをイソプロピルエーテル40mlに溶解し、水素化ホウ素ナトリウム31.5 mmolをメタノール15 mlに溶解して加え、溶液の黄色が無色になるまで室温で撹拌する。反応液にイソプロピルエーテル60mlと精製水100mlを加え、イソプロピルエーテル層を分離し、更に水層にイソプロピルエーテル100mlを加えて可溶画分を抽出、イソプロピルエーテル層を合わせて、無水硫酸ナトリウムで脱水後、減圧下溶媒を留去する。残渣にジメチルアミノピリジン8.97 mmol、ジカルボン酸無水物18.0 mmolを加え、イソプロピルエーテル-ジオキサン(6:4、v/v)100mlに溶解して、室温で3時間撹拌後、50~60℃に加熱しながら2時間反応させ、さらに室温で放冷しながら10時間反応させる。反応液に精製水100mlを加え、イソプロピルエーテル層を分離し、無水硫酸ナトリウムで脱水後、減圧下溶媒を留去する。残渣をイソプロピルエーテルに懸濁し、遠心して得た沈殿物に酢酸エチル100mlと精製水100mlを加え酢酸エチル可溶画分を抽出し、無水硫酸ナトリウムで脱水後、減圧下溶媒を留去する。残渣をイソプロピルエーテルに懸濁し不溶物を酢酸エチルで再結晶して、ビタミンKヒドロキノン-1,4-ビス-ジカルボン酸ヘミエステルを得る。
製造方法H
ビタミンK 6.75 mmol、亜鉛18.4 mmol、無水ジカルボン酸33.0 mmol、無水酢酸ナトリウム 13.8 mmol、酢酸161.5 mmolを100 mlのナスフラスコに入れ、ジムロートを取り付けよく撹拌しながら85℃で3時間加熱。室温冷却し生じた白い固形に酢酸エチル200 mlと精製水100mlを加え酢酸エチル可溶画分を抽出し、無水硫酸ナトリウムで脱水後、減圧下溶媒を留去する。残渣を酢酸エチルで再結晶して、ビタミンKヒドロキノン-1,4-ビス-ジカルボン酸ヘミエステルを得る。
ビタミンK 6.75 mmol、亜鉛18.4 mmol、無水ジカルボン酸33.0 mmol、無水酢酸ナトリウム 13.8 mmol、酢酸161.5 mmolを100 mlのナスフラスコに入れ、ジムロートを取り付けよく撹拌しながら85℃で3時間加熱。室温冷却し生じた白い固形に酢酸エチル200 mlと精製水100mlを加え酢酸エチル可溶画分を抽出し、無水硫酸ナトリウムで脱水後、減圧下溶媒を留去する。残渣を酢酸エチルで再結晶して、ビタミンKヒドロキノン-1,4-ビス-ジカルボン酸ヘミエステルを得る。
製造方法I
ビタミンKヒドロキノン-1,4-ビス-ジカルボン酸ヘミエステル2mmolを2倍molの0.1N水酸化ナトリウム水溶液または2倍molのメグルミン水溶液を加え溶解させ凍結乾燥させる。メタノール-アセトニトリルで再結晶しビタミンKヒドロキノン-1,4-ビス-ジカルボン酸ヘミエステル-ビス-ナトリウム塩またはビタミンKヒドロキノン-1,4-ビス-ジカルボン酸ヘミエステル-ビス-メグルミン塩を得る。
ビタミンKヒドロキノン-1,4-ビス-ジカルボン酸ヘミエステル2mmolを2倍molの0.1N水酸化ナトリウム水溶液または2倍molのメグルミン水溶液を加え溶解させ凍結乾燥させる。メタノール-アセトニトリルで再結晶しビタミンKヒドロキノン-1,4-ビス-ジカルボン酸ヘミエステル-ビス-ナトリウム塩またはビタミンKヒドロキノン-1,4-ビス-ジカルボン酸ヘミエステル-ビス-メグルミン塩を得る。
製造方法J
ビタミンKヒドロキノン-1,4-ビス-ジカルボン酸ヘミエステルを1級または2級アルコールに溶解し塩酸酸性下、撹拌する。減圧下溶媒を留去しビタミンKヒドロキノン-1,4-ビス-ジカルボン酸アルコールエステルを得る。
ビタミンKヒドロキノン-1,4-ビス-ジカルボン酸ヘミエステルを1級または2級アルコールに溶解し塩酸酸性下、撹拌する。減圧下溶媒を留去しビタミンKヒドロキノン-1,4-ビス-ジカルボン酸アルコールエステルを得る。
Claims (10)
- 下記一般式(1)
(式中、R1およびR2はそれぞれ水素原子、またはグリシン、N-アシルグリシン、N-アルキルグリシン、N,N-ジアルキルグリシン、N,N,N-トリアルキルグリシン、アシル、ジカルボン酸ヘミエステル及びその塩から選ばれる置換基を意味し、R1, R2の少なくとも一方はグリシン、N-アシルグリシン、N-アルキルグリシン、N,N-ジアルキルグリシン、N,N,N-トリアルキルグリシン、アシル、ジカルボン酸ヘミエステル及びその塩である。R3は下記一般式(2)
もしくは下記一般式(3)
で示される基を表す。n は1~7の整数を意味する。)で表される活性型ビタミンKのカルボン酸エステル類またはその塩の少なくとも一種類からなる、ミトコンドリア機能障害改善剤。 - 前記一般式(1)で表される化合物またはその塩を含有する神経変性改善剤。
- 前記一般式(1)で表される化合物またはその塩を含有する筋萎縮性側索硬化症改善剤。
- 前記一般式(1)で表される化合物またはその塩を含有するアルツハイマー病改善剤。
- 前記一般式(1)で表される化合物またはその塩を含有するパーキンソン病改善剤
- 前記一般式(4)で表される化合物またはその塩を含有する神経変性改善剤。
- 前記一般式(4)で表される化合物またはその塩を含有する筋萎縮性側索硬化症改善剤。
- 前記一般式(4)で表される化合物またはその塩を含有するアルツハイマー病改善剤。
- 前記一般式(4)で表される化合物またはその塩を含有するパーキンソン病改善剤。
