<1> トレオニン含量の増強された乾燥たばこ材料の生産方法であって、
内在性のトレオニンアルドラーゼの発現または活性が低下するよう改変されたタバコ植物またはその一部から、該乾燥たばこ材料を生産することを含み、
該トレオニンアルドラーゼは、以下の(a)および(b)の少なくとも何れかである、生産方法:
(a)配列番号1に記載の塩基配列を有するポリヌクレオチドに対して90%以上の配列同一性を有するポリヌクレオチドを含む遺伝子によってコードされるポリペプチドからなるトレオニンアルドラーゼ
(b)配列番号3に記載の塩基配列を有するポリヌクレオチドに対して90%以上の配列同一性を有するポリヌクレオチドを含む遺伝子によってコードされるポリペプチドからなるトレオニンアルドラーゼ。
<2> トレオニン含量の増強された乾燥たばこ材料の生産方法であって、

(b)配列番号4に記載のアミノ酸配列を有するポリペプチドに対して90%以上の配列同一性を有するポリペプチドからなるトレオニンアルドラーゼ。
本明細書において、「ポリヌクレオチド」は、「核酸」または「核酸分子」とも換言でき、ヌクレオチドの重合体を意図している。また、「塩基配列」は、「核酸配列」または「ヌクレオチド配列」とも換言でき、特に言及のない限り、デオキシリボヌクレオチドの配列またはリボヌクレオチドの配列を意図している。
本発明に係る生産方法について説明すれば以下の通りである。
(b)配列番号3に記載の塩基配列を有するポリヌクレオチドに対して90%以上の配列同一性を有するポリヌクレオチドを含む遺伝子によってコードされるポリペプチドからなるトレオニンアルドラーゼ。
(b)配列番号4に記載のアミノ酸配列を有するポリペプチドに対して90%以上の配列同一性を有するポリペプチドからなるトレオニンアルドラーゼ。
まず、本発明の生産方法に用いられるタバコ植物またはその一部について説明する。本明細書において、「タバコ植物またはその一部」とは、タバコ植物の、植物体全体、植物器官(葉、根、茎、わき芽、花、および種子など)、植物組織片(表皮、師部、柔組織、木部および維管束等)ならびに植物細胞(培養細胞、カルス、およびプロトプラスト等)を包含するものを意味する。好ましくは植物体全体、植物器官(葉、根、茎、わき芽、花、および種子など)ならびに植物組織片(表皮、師部、柔組織、木部および維管束等)から選択されるものであり、より好ましくは、植物体全体、および植物器官(葉、根、茎、わき芽、花、および種子など)等から選択されるものであり、さらに好ましくは、植物体全体および葉から選択されるものであり、最も好ましくは、葉である。なお、本発明の生産方法に用いられる葉は、植物体の茎の上部に位置する葉(上葉)であってもよく、茎の真ん中より上に位置する葉(本葉)であってもよい。または、茎に着いたままの葉であってもよい。
本発明の生産方法に係る遺伝子は、トレオニンからグリシンへの異化反応を触媒するトレオニンアルドラーゼをコードするものである。また、該遺伝子の一例は、トレオニンからグリシンへの異化反応を触媒するトレオニンアルドラーゼをコードしており、当該トレオニンアルドラーゼが活性を有していると、アスパラギン酸由来のアミノ酸生合成経路において、トレオニンからグリシンへの異化反応を触媒するものである。以下、本発明の生産方法に係る遺伝子は、「トレオニンアルドラーゼ遺伝子」としても記載する。
(b)配列番号2に記載のアミノ酸配列に対して90%以上の配列同一性を有し、トレオニンからグリシンへの異化反応を触媒する酵素活性を有するタンパク質をコードするポリヌクレオチド
(c)配列番号2に記載のアミノ酸配列において1~35個のアミノ酸が置換、欠失、挿入、および/または付加されたアミノ酸配列を有し、トレオニンからグリシンへの異化反応を触媒する酵素活性を有するタンパク質をコードするポリヌクレオチド
(d)上記(a)に記載のポリヌクレオチドと相補的な配列からなるポリヌクレオチドと、ストリンジェントな条件下でハイブリダイズし、トレオニンからグリシンへの異化反応を触媒する酵素活性を有するタンパク質をコードするポリヌクレオチド
(e)配列番号3に記載の塩基配列を有するポリヌクレオチドに対して90%以上の配列同一性を有するポリヌクレオチド
(f)配列番号4に記載のアミノ酸配列に対して90%以上の配列同一性を有し、トレオニンからグリシンへの異化反応を触媒する酵素活性を有するタンパク質をコードするポリヌクレオチド。
本発明の生産方法に係るポリペプチドは、上記(遺伝子)の欄に記載した遺伝子の翻訳産物であり、少なくともトレオニンからグリシンへの異化反応を触媒する活性を有する植物のトレオニンアルドラーゼである。また、「トレオニンからグリシンへの異化反応を触媒するトレオニンアルドラーゼ」は、一例としては、L-トレオニンアルドラーゼ(EC 4.1.2.5)である。L-トレオニンアルドラーゼは、L-トレオニンからグリシンとアセトアルデヒドとを生成する反応を触媒するものである。
(b)配列番号2に記載のアミノ酸配列において1~35個のアミノ酸が置換、欠失、挿入、および/または付加されたアミノ酸配列を有するポリペプチドからなるトレオニンアルドラーゼとしてのポリペプチド

(f)上記(d)に記載のポリペプチドをコードするポリヌクレオチドと相補的な配列からなるポリヌクレオチドと、ストリンジェントな条件下でハイブリダイズするポリヌクレオチドによってコードされる、トレオニンアルドラーゼとしてのポリペプチド。
本発明の生産方法において用いられるタバコ植物は、内在性のトレオニンアルドラーゼの発現または活性が低下するよう改変されたタバコ植物である。
また、改変されたタバコ植物におけるトレオニンアルドラーゼの発現または活性の低下は、恒常的に生じるものであっても、断続的に生じるものであってもよく、一過性のものであってもよい。例えば、タバコ植物が所定の温度以上の温度に曝露されたときにのみ、当該トレオニンアルドラーゼの発現または活性の低下が生じるものであってもよい。そのようなトレオニンアルドラーゼとして加熱条件下で失活する温度感受性変異を含むケースが挙げられる。
タバコ植物における内在性のトレオニンアルドラーゼの発現または活性の低下が、該変異対立遺伝子によるものである場合、該タバコ植物は、トレオニンアルドラーゼをコードする遺伝子座についてみれば、対立遺伝子同士で同一種類の変異を有するホモ接合体であるか、または対立遺伝子同士で異なる変異を有するヘテロ接合体であり得る。
一実施形態において、本発明の生産方法に用いるのに好適なタバコ植物であるか否かは、以下の(a)または(b)に示される内在性のトレオニンアルドラーゼの発現または活性の低下を指標として判定することができる。また、別の実施形態において、本発明の生産方法に用いるのに好適なタバコ植物はトレオニンアルドラーゼの発現または活性の低下と関連した(a)~(e)のいずれかに示される指標に基づく選抜を経て育成することができる。

一実施形態において、本発明の生産方法に、あるタバコ植物またはその一部を原料として適用するに当たり、候補のタバコ植物のトレオニンアルドラーゼの発現または活性の低下を確認することによって、当該タバコ植物またはその一部が本発明の生産方法に用いるのに好適なタバコ植物であるか否かを判定することができる。
トレオニンアルドラーゼの発現の低下は、タバコ植物またはその一部におけるトレオニンアルドラーゼタンパク質の存在量を検査することによって確認することができる。該タンパクの存在量の検査は、例えば、抗体を用いたウェスタン分析またはELISA分析等によって実施することができる。必要に応じて基準となる該タンパクの存在量と比較して、該タンパク質の存在量が低減されているか否か検査する。また、基準となるタンパク質の存在量とは、例えば、対照となる同種のタバコ植物(例えば、トレオニンアルドラーゼの発現または活性が、正常な状態から変更されていない、例えば低下するよう改変されていないタバコ植物)におけるトレオニンアルドラーゼタンパク質の存在量を指す。
トレオニンアルドラーゼの活性の低下は、例えば、以下の2-1~2-2の方法によって確認することができる。
あるタバコ植物が有するトレオニンアルドラーゼタンパク質そのものの酵素活性の低下は該植物から単離したトレオニンアルドラーゼ遺伝子のcDNAを用いた相補性試験により明らかにすることができる。相補試験において、グリシン要求性酵母株であるYM13(gly1 shm1 shm2)がホストとして好適に利用することができる(Monschauet al. 1997, FEMS microbiology letters 150: 55-60., Joshi et al. 2006. The Plant Cell 18: 3564-3575.)。単離したトレオニンアルドラーゼ遺伝子の翻訳産物が酵素活性を有する場合は機能相補するため、この遺伝子を導入された宿主はグリシンを含まない培地で生育することができる。試験の容易さの観点からは、下記のポリペプチドの活性の測定よりも、酵母を用いた相補試験を行うことが一般的である。
対象となるタバコ植物またはその一部から、トレオニンアルドラーゼ遺伝子がコードするポリペプチドを単離する。または対象となる植物におけるトレオニンアルドラーゼ遺伝子のクローンを発現ベクターに導入して、宿主細胞に導入することにより、一過的にポリペプチドを発現させる。これを培養、抽出および精製したものを用いて、トレオニンを基質としたグリシンへの異化反応を触媒する酵素活性の有無を検査し、当該酵素活性が無いか、当該酵素活性が、対照となる同種の植物(例えば、野生型のトレオニンアルドラーゼ遺伝子のホモ接合体)と比較して低下している場合に、トレオニンアルドラーゼの活性が低下しているとみなす。
また、上記の<2-1>および<2-2>に加え、本発明の生産方法に用いるのに好適なタバコ植物であるか否かの判定は、そのタバコ植物またはその一部を加熱処理し、加熱処理後のタバコ植物またはその一部が含有する遊離トレオニンの量を測定し、測定結果を確認することによって簡単に行うことができる。例えば、加熱処理した後のタバコ植物またはその一部が含有する遊離トレオニンの量が、対照の植物またはその一部を加熱処理した場合と比べて、有意に多いか否かを確認することによって好適なタバコ植物であるか否かが判定できる。
一実施形態において、本発明の生産方法に用いられるのに好適なタバコ植物は、上述した(a)~(e)からなる群より選択される指標に基づいて選抜を行うことを包含する選抜方法を経て育成されたタバコ植物であり得る。
トレオニンアルドラーゼ遺伝子における、該トレオニンアルドラーゼの発現の低下または活性の低下を生じさせる変異を検出する。該変異の一例は挿入、欠失、フレームシフト変異、ナンセンス変異、ノンストップ変異、およびスプライス部位変異からなる群より選択される突然変異である。該変異の検出には様々な方法が利用可能であるが、例えば、PCR-RFLP法またはリアルタイムPCRを利用した方法(Cycling Probe Method, TaqMan(登録商標) SNP Genotyping Assays)などが好適に利用される。トレオニンアルドラーゼの遺伝子座の遺伝子型は、交配または選抜などによる一般的な育種プログラムにおける育種過程を通して、常にモニターすることが可能である。したがって、トレオニンアルドラーゼの発現または活性の低下したタバコの品種は、トレオニンアルドラーゼの変異対立遺伝子を利用した育種プログラムにおいて、変異対立遺伝子の変異そのものを指標とすることによって、容易に選抜し、育成することが可能である。
ある局面においては、トレオニンアルドラーゼの発現の低下は、トレオニンアルドラーゼ遺伝子の発現を抑制するために導入された外因性のポリヌクレオチドによるものが含まれる。そのような場合はトレオニンアルドラーゼ遺伝子の発現を抑制するために導入された外因性のポリヌクレオチドの有無を指標として利用することによって、該形質を有するタバコ植物を選抜および育成することができる。あるいは、該外因性ポリヌクレオチドと共に植物に導入されたコンストラクトの構成要素(プロモータ配列など)も選抜の指標として利用してもよい。
トレオニンアルドラーゼの発現または活性の低下と関連するトレオニンアルドラーゼの変異対立遺伝子の有無を、当該変異対立遺伝子に連鎖するDNAマーカーを利用して、検査することも可能である。上記の連鎖DNAマーカーは、一例において、トレオニンアルドラーゼ遺伝子が座乗する位置から約10cM以内に位置しており、好ましくは約5cM以内に位置しており、より好ましくは約2cM以内に位置しており、さらに好ましくは約1cM以内に位置している。
本発明に係るトレオニンアルドラーゼの発現または活性が低下するよう改変されたタバコ植物は、以下に記載する方法で作出されたタバコ植物を含む。
トレオニンアルドラーゼの発現または活性の低下に繋がるトレオニンアルドラーゼをコードする遺伝子の変異対立遺伝子は、挿入、欠失、フレームシフト変異、ナンセンス変異、ノンストップ変異、およびスプライス部位変異からなる群より選択される突然変異を含むものである。
上記変異対立遺伝子は、変異原処理、ゲノム編集およびノックアウト法から選択されるいずれかの方法によって生じる人工突然変異を含むものであるか、天然に存在する自然突然変異を含むものである。変異原処理の具体例としては、EMS(ethyl methane sulfonate:エチルメタンスルホン酸)処理、電磁波、紫外線、X線、γ線、粒子線、中性子線、α線およびβ線などの各種放射線の照射、または重イオンビーム等の照射等の方法が挙げられる。
次に、外因性のポリヌクレオチドを導入することによってトレオニンアルドラーゼ遺伝子の発現を抑制したタバコ植物の作出方法について説明する。
本発明に係るタバコ植物に適用される組換え発現ベクターおよび本発明に係る外因性のポリヌクレオチドが発現可能に導入された形質転換体について以下に説明する。

また、本発明は、上記の生産方法により生産された、乾燥たばこ材料も提供する。上記のように、本明細書において、「乾燥たばこ材料」とは、採取されたタバコ植物またはその一部を乾燥させたものを包含するものである。
本発明に係る乾燥たばこ材料から調製された抽出物は本発明の範疇である。当該抽出物は、乾燥たばこ材料から1つ以上の成分を抽出する抽出工程を含む調製方法によって調製されたものであり得る。
本発明は、本発明の生産方法により生産された乾燥たばこ材料および/または当該乾燥たばこ材料から調製した抽出物を原料として用いて製造されたたばこ製品も提供する。たばこ製品は、上述の乾燥たばこ材料、更に加工処理の加えられた乾燥たばこ材料、または抽出物を用いて製造されるものである。上述の乾燥たばこ材料、さらに加工された乾燥たばこ材料または抽出物は、たばこ製品中のたばこ材料の全体として使用されてもよいし、たばこ製品中のたばこ材料の一部として使用されてもよい。あるいは、本発明の方法により生産された乾燥たばこ材料から粉末、液体などの形態にさらに加工された材料はたばこ製品中の他のたばこ材料に添加されて使用されてもよい。本発明により得られる乾燥たばこ材料は、たばこ製品中のたばこ材料の一部として使用される場合、任意の割合で使用することができる。
本発明は以下の<1>~<18>の何れかを包含する。
<1> トレオニン含量の増強された乾燥たばこ材料の生産方法であって、
内在性のトレオニンアルドラーゼの発現または活性が低下するよう改変されたタバコ植物またはその一部から、該乾燥たばこ材料を生産することを含み、
該トレオニンアルドラーゼは、以下の(a)および(b)の少なくとも何れかである、生産方法:
(a)配列番号1に記載の塩基配列を有するポリヌクレオチドに対して90%以上の配列同一性を有するポリヌクレオチドを含む遺伝子によってコードされるポリペプチドからなるトレオニンアルドラーゼ
(b)配列番号3に記載の塩基配列を有するポリヌクレオチドに対して90%以上の配列同一性を有するポリヌクレオチドを含む遺伝子によってコードされるポリペプチドからなるトレオニンアルドラーゼ。
<2> トレオニン含量の増強された乾燥たばこ材料の生産方法であって、
内在性のトレオニンアルドラーゼの発現または活性が低下するよう改変されたタバコ植物またはその一部から、該乾燥たばこ材料を生産することを含み、
該トレオニンアルドラーゼは、以下の(a)および(b)の少なくとも何れかである、生産方法:
(a)配列番号2に記載のアミノ酸配列を有するポリペプチドに対して90%以上の配列同一性を有するポリペプチドからなるトレオニンアルドラーゼ、
(b)配列番号4に記載のアミノ酸配列を有するポリペプチドに対して90%以上の配列同一性を有するポリペプチドからなるトレオニンアルドラーゼ。
<3> 上記改変されたタバコ植物における内在性のトレオニンアルドラーゼの発現または活性の低下は、該トレオニンアルドラーゼをコードする遺伝子の変異対立遺伝子によるものであるか、または該トレオニンアルドラーゼの発現を抑制するために導入された外因性のポリヌクレオチドによるものである、<1>または<2>に記載の生産方法。
<4> 上記変異対立遺伝子は、挿入、欠失、フレームシフト変異、ナンセンス変異、ノンストップ変異、およびスプライス部位変異からなる群より選択される突然変異を含むものである、<3>に記載の生産方法。
<5> 上記突然変異は、突然変異誘発、ゲノム編集およびノックアウト法からなる群から選択される方法によって導入された人工突然変異を含むものであるか、天然に存在する自然突然変異を含むものである、<4>に記載の生産方法。
<6> 上記人工突然変異は、突然変異誘発またはゲノム編集によって導入されたものであり、該突然変異誘発またはゲノム編集は、化学物質または放射線への暴露、CRISPR system、Transcription Activator-Like Effector Nucleases、Zinc Finger Nucleases、Oligonucleotide-directed mutagenesis、T-DNA挿入、およびトランスポゾン挿入からなる群より選択される方法によって行われるものである<5>に記載の生産方法。
<7> 上記改変されたタバコ植物は、上記トレオニンアルドラーゼをコードする遺伝子として、対立遺伝子同士で同一種類の変異を有するホモ接合体であるか、または対立遺伝子同士で異なる変異を有するヘテロ接合体である、<3>~<6>の何れか一項に記載の生産方法。
<8> 上記外因性のポリヌクレオチドは、アンチセンスRNA分子、RNAi分子および共抑制因子分子からなる群より選択される分子を含む、<3>に記載の生産方法。
<9> 上記改変されたタバコ植物は、内在性のトレオニンアルドラーゼの発現または活性が低下するよう改変されたタバコ植物の自殖によって育成された、または該改変されたタバコ植物同士の交配によって育成された、タバコ品種の植物である、<1>~<8>の何れか一項に記載の生産方法。
<10> 上記改変されたタバコ植物は、以下の(a)~(e)からなる群より選択される指標に基づき選抜および育成されたタバコ植物である、<9>に記載の生産方法:
(a)上記トレオニンアルドラーゼの発現の低下
(b)上記トレオニンアルドラーゼの活性の低下
(c)加熱処理した葉におけるトレオニン含量の増強
(d)上記トレオニンアルドラーゼの変異対立遺伝子の有無
(e)上記(d)の変異対立遺伝子に連鎖するDNAマーカー
(f)上記遺伝子の発現を抑制するために導入された外因性のポリヌクレオチドの有無。<11> 上記タバコ植物の一部は、葉、根、茎、わき芽、花、種子、植物培養細胞、カルスおよびプロトプラストからなる群より選択される、<1>~<10>の何れか一項に記載の生産方法。
<12> 上記乾燥たばこ材料におけるトレオニン含量の増強は、上記改変されたタバコ植物またはその一部の乾燥によって生じたものである、<1>~<11>の何れか一項に記載の生産方法。
<13> 上記乾燥が、熱風乾燥、空気乾燥、火力乾燥および日干し乾燥からなる群より選択される乾燥によって行われる、<12>に記載の生産方法。
<14> 上記改変されたタバコ植物は、ニコチアナ タバカムおよびニコチアナ ルスティカからなる群より選択されるタバコ植物である、<1>~<13>の何れか一項に記載の生産方法。
<15> 上記乾燥たばこ材料は、葉材料であって、当該乾燥たばこ材料におけるトレオニン含量は、トレオニンアルドラーゼの発現または活性が低下するよう改変されていない対照のタバコ植物またはその一部から作製した乾燥たばこ材料のトレオニン含量と比べて20%以上高い、<1>~<14>の何れか一項に記載の生産方法。
<16> <1>~<15>の何れか一項に記載の生産方法によって生産される、乾燥たばこ材料。
<17> <16>に記載の乾燥たばこ材料から調製した、抽出物。
<18> <16>に記載の乾燥たばこ材料および/または<17>に記載の抽出物を使用して製造された、たばこ製品。
トレオニンアルドラーゼ(Threonine aldolase(THA))は、生物におけるトレオニンの代謝に関わる酵素であり、Arabidopsis thalianaではTHA1(At1g08630)、THA2(At3g04520)という2つの遺伝子によってコードされている(非特許文献10)ことが知られている。アスパラギン酸に由来するアミノ酸の代謝経路を図1に示す。
タバコにおけるTHA遺伝子の発現を確認するために、品種つくば1号の播種後10日の芽生え、播種後4週の苗の葉身(ラミナ)、播種後5週の苗の根、加熱処理を施したラミナおよび種子からRNAを抽出してRT-PCRに供試した。葉身の加熱処理は、播種後8週の個体から切り取った葉を温度37℃および湿度85%の条件下に、0、8、24または48時間暴露して行った。種子のRNAは10、25、50または100粒から調製した。全RNAはRNeasy Plant Mini Kit(QIAGEN)を用いて抽出した。cDNA合成はHigh Capacity RNA-to-cDNA Master Mix (Thermo Fisher Scientific Inc.)を用いて行った。手法は抽出Kitに添付のマニュアルに従った。
(NtTHA1_s遺伝子およびNtTHA1_t遺伝子の変異体の選抜)
NtTHA1_s遺伝子およびNtTHA1_t遺伝子に変異を有する系統はニコチアナタバカム(つくば1号)のEMS変異体集団(M2)の中から選抜した。選抜は SingleStrand Conformation Polymorphism(SSCP)解析によって行った。SSCP解析に供試するPCR断片は、以下の表2に示すNtTHA1遺伝子に特異的なプライマーセットおよびMultiplex PCR Kit(QIGEN)を用いて増幅した。その後、増幅産物の分析はPOP(商標)Conformational Analysis Polymer(Thermo Fisher Scientific Inc.)を充填したキャピラリー電気泳動装置(3130xl PRISM(登録商標)DNA analyzer, Thermo Fisher Scientific Inc.)を用いて実施した。得られたデータをGeneMapper(登録商標)Version4.0(Thermo Fisher Scientific Inc.)で解析し、多型性のPCR産物が増幅された系統を目的変異体の候補として選抜した。
液体培地で一晩培養した後、QiaPrep Spin Miniprep Kit(QIAGEN)を用いてプラスミドを抽出した。その後、得られたプラスミドおよびBigDye(登録商標)Terminator v3.1 Cycle Sequencing Kit(Thermo Fisher Scientific Inc.)を用いてPCR産物の塩基配列を解読した。得られたデータをATGC (Ver.7) Sequence Assembly Software(GENETYX CORPORATION)で解析し、NtTHA1遺伝子に導入された変異を特定した。各手順における全ての手法はKitに添付のマニュアルに従った。
NtTHA1遺伝子の機能欠失の影響を明らかにするために、tha1-s1またはtha1-s2のホモ接合体であり、且つ、tha1-tのホモ接合体である二重変異体植物を育成した。
二重変異体におけるNtTHA1遺伝子の発現は以下のように調査した。まず播種後6週の苗からRNAを抽出し、PrimeScript(商標)RT reagent kit with gDNA Eraser(TAKARA BIO INC.)を用いてcDNAを合成した。得られたcDNAを鋳型とし、以下の表4に示すプライマーおよびプローブセットとTaqMan(登録商標)Fast Advanced Master Mix(Thermo Fisher Scientific Inc.)とを用いて、Real-Time PCRを実施した。解析にはStepOnePlus Real-Time PCR System(Thermo Fisher Scientific Inc.)を用いた。PCRの手順についてはキットおよび装置のマニュアルに従った。
(生育状態および形態の観察)
上記のように選抜した二重変異体(tha1-s2/tha1-t)および対照(control)となる植物を、一般的なたばこ耕作法に従い、圃場にて栽培した。図6の(b)は二重変異体であり、図6の(a)は対照植物である。
次に、二重変異体および対照植物のそれぞれから葉を採取し、新鮮な葉および熱風乾燥した葉における遊離トレオニン含量を分析した。熱風乾燥における標準的な乾燥スケジュール(Tabacco Curing schedule)を図7に示す。図7の横軸は、経過時間(Time)(hr)を示し、左側の縦軸は温度(Tempreture)(℃)を示し、右側の縦軸は相対湿度(Relative humidity、RH)(%)を示している。また、図8に、二重変異体および対照植物の新鮮な葉(flesh leaves)の遊離トレオニン含量を示す。図9に、二重変異体および対照植物の熱風乾燥した葉(cured leaves)の遊離トレオニン含量を示す。
熱風乾燥の温度が二重変異体(tha1-s2/tha1-t)および対照植物(THA1-s/THA1-t)の葉の遊離トレオニン含量に及ぼす影響を調査した。乾燥開始から0h、8h、16h、24h、30h、36h、48h、60hおよび72h後に乾燥中のタバコ葉3枚をランダムに選び、それらから凡そ10cm四方の葉片を採取した。3つの葉片を1つのサンプルとしてまとめて遊離トレオニン含量を測定した。なお、熱風乾燥チャンバーは一定温度(25℃、34℃、38℃または42℃)を維持し、湿度85%で運転した。図10に、NtTHA1遺伝子の二重変異体および対照のタバコ植物から採取した葉を各温度条件で熱風乾燥した場合のトレオニン含量を示す。乾燥開始から24時間以降の二重変異体の葉ではどの温度条件でも野生型と比べて遊離トレオニン含量が高かった。
各種遺伝子型の個体におけるNtTHA1遺伝子の発現をさらに詳しく調べるために、NtTHA1s遺伝子またはNtTHA1t遺伝子を特異的に検出するプライマーおよびプローブを作成した(表5)。表中の‘position’カラムは配列番号1または3における位置を示す。
二重変異体(tha1-s1/tha1-t)と空気乾燥タイプのタバコ品種(TN90)を交配し、BC1F2の種子を得た。BC1F2植物の幼苗からDNAを抽出しNtTHA1遺伝子の遺伝子型を調査した。二重変異体(tha1-s1/tha1-t)と対照植物(THA1-s/THA1-t)とを選抜し、それぞれ200個体以上を一般的なたばこ耕作法に従って圃場にて栽培した。
CRISPR-Cas9 を用いたゲノム編集はLiら(Li et al., Nat Biotechnol. 2013 Aug;31(8):688-912013)の方法を参照して実施した。ターゲット配列(5’-GGAGGCATTGCCACTCTCGGAGG-3’:配列番号42)はNtTHA1s遺伝子(配列番号1)およびNtTHA1t遺伝子(配列番号3)の共通部分から選択された。この配列は配列番号1または配列番号3の283番目から305番目の塩基配列であり、3’末端のAGG(下線部)はPAM配列である。plant codon‐optimized SpCas9 (pcoCas9)とsynthetic-guide RNA (sgRNA)をタバコで発現させるための一体型の植物発現ベクターは、pRI201-AN(タカラバイオ社)のCaMV 35Sプロモータの下流のNdeI-SalI間にpcoCas9遺伝子を挿入し、KpnI-BamHI間にArabidopsis U6 polymerase III promoterと連結したsgRNA発現カセットを挿入して作成した。

- トレオニン含量の増強された乾燥たばこ材料の生産方法であって、
内在性のトレオニンアルドラーゼの発現または活性が低下するよう改変されたタバコ植物またはその一部から、該乾燥たばこ材料を生産することを含み、
該トレオニンアルドラーゼは、以下の(a)および(b)の少なくとも何れかである、生産方法:
(a)配列番号1に記載の塩基配列を有するポリヌクレオチドに対して90%以上の配列同一性を有するポリヌクレオチドを含む遺伝子によってコードされるポリペプチドからなるトレオニンアルドラーゼ
(b)配列番号3に記載の塩基配列を有するポリヌクレオチドに対して90%以上の配列同一性を有するポリヌクレオチドを含む遺伝子によってコードされるポリペプチドからなるトレオニンアルドラーゼ。 - トレオニン含量の増強された乾燥たばこ材料の生産方法であって、
