Description
すなわち、本発明は、以下に示す被験物質を検出するための組成物、被験物質を検出するための凍結乾燥物、被験物質を検出する方法、及び被験物質を検出するためのキットを提供するものである。
〔1〕金ナノプレートを含む、被験物質を検出するための組成物であって、
前記金ナノプレートの平均アスペクト比(金ナノプレートの最大長さを厚さで割った値)が、1より大きく10以下であり、
前記組成物中の第四級アンモニウムカチオンの濃度が、0~1mMであり、
前記第四級アンモニウムカチオンが、N+(R)4(各Rは、それぞれ独立して、炭素数1~20の直鎖又は分枝鎖アルキル基から選択される)で表されることを特徴とする、組成物。
〔2〕前記第四級アンモニウムカチオンが、デシルトリメチルアンモニウムカチオン、ヘキサデシルトリメチルアンモニウムカチオン、及び、ヘプタデシルトリメチルアンモニウムカチオンから成る群から選択される、前記〔1〕に記載の組成物。
〔3〕分子量500以上の水溶性高分子を含まない、前記〔1〕又は〔2〕に記載の組成物。

〔21〕前記工程(1)が、金ナノプレート種粒子を予備成長させる工程を含まない、前記〔20〕に記載の方法。
また、金ナノプレートは、厚みが薄いプレートの形状であるから、その平均最大長さと同じ粒子径の球状金ナノ粒子よりも体積及び質量が小さい。そのため、金ナノプレートは、分散媒中で安定に分散しやすく、分散剤が必要な場合であっても、球状金ナノ粒子の分散に必要な量よりも少ない量の分散剤で分散することができる。そして、抗体などの被験物質に対する特異的結合物質は、分散剤としても機能し得るが、高価であることが多いので、その使用量が少なければ製造コストを削減できる。その他の分散剤は、金ナノプレートと被験物質に対する特異的結合物質との結合を妨げることがあるので、金ナノプレートの懸濁液中の分散剤の量が少なければ、当該金ナノプレート上に被験物質に対する特異的結合物質を効率よく担持でき、そのことによっても、当該被験物質に対する特異的結合物質の使用量を節約することができる。
本発明の被験物質を検出するための組成物は、平均アスペクト比が1より大きく10以下である金ナノプレートを含み、かつ該組成物中のN+(R)4(各Rは、それぞれ独立して、炭素数1~20の直鎖又は分枝鎖アルキル基から選択される)で表される第四級アンモニウムカチオンの濃度が0~1mMであることを特徴としている。
また、金ナノプレートの表面に担持させる抗体などの被験物質に対する特異的結合物質の表面電荷が正電荷である場合、金ナノプレートのゼータ電位が好ましくはマイナスである。
金ナノプレートは吸収波長を制御可能であり、生体内で使用可能な電磁波(光)を照射することで治療や診断などへの利用も期待されている。例えば、吸収した光エネルギーを熱に変換する光熱変換効果を利用したがんの温熱療法(ハイパーサーミア)や、発生した熱により温められた周囲の媒質が体積膨張した際に生じる音響波、すなわち光音響効果(フォトアコースティック効果)を検出する光音響波イメージングへの利用も期待される。
金ナノプレートを含む組成物中の金含有率は、例えばICP発光分析装置や紫外可視分光光度計で測定してもよい。
ICP発光分析装置で測定する場合、具体的には、以下の手順に示すように、懸濁液を遠心分離後、上澄み液を除去し、得られた沈殿物を、除去した上澄み液と同量の超純水で再度懸濁する。そして、その懸濁液に王水を添加後、煮沸して、得られた溶液を、ICP発光分析装置を用いて分析する。
1)金ナノプレート懸濁液を遠心分離(8,000×g)後、上澄み液を除去し、得られた沈殿物を、除去した上澄み液と同量の超純水で再度懸濁する。
2)上記工程1で得られた懸濁液に王水を添加後、5分間煮沸して、金及び銀を王水中へ溶解させる。
3)上記工程2で得られた溶液を、ICP発光分析装置を用いて測定する。金の濃度は、任意濃度の標準サンプルを上述と同様に測定することで作成した検量線より算出する。
紫外可視分光光度計で測定する場合、具体的には、以下の手順に示すように測定して、金含有率を算出する。
1)金ナノプレート合成液のICP測定、または金添加量より金含有率を算出
精製前の金ナノプレート合成液(例えば、実施例の懸濁液AやKなど)中の金含有率をICP発光分析装置で測定する。または、金ナノプレート合成時に添加する金の総量に基づき、合成の収率が100%であると仮定して、金含有率Aを算出する。
2)金ナノプレート合成液の光学特性を紫外可視分光光度計で測定
金ナノプレート合成液を超純水で4倍希釈して紫外可視分光光度計で測定する。
3)消光度1あたりの金含有率の算出
得られた消光スペクトルの540nmの消光度B(プレート状ではない多面体又は球状金ナノ粒子の消光度)、及び、550~740nmの最大吸収波長の消光度Cの合計値が、金ナノプレート合成液中の金の総量に由来する消光であるとする。そうすると、消光度1あたりの金含有率は、以下の式によって求められる。
消光度1あたりの金含有率(質量%/消光度)=A÷(B+C)
4)金ナノプレート精製液の光学特性を紫外可視分光光度計で測定
金ナノプレートの精製液(例えば、実施例の懸濁液BやO)を超純水で4倍希釈して紫外可視分光光度計で測定する。
5)金含有率の算出
得られた消光スペクトルの540nmの消光度D(プレート状ではない多面体又は球状金ナノ粒子の消光度)、及び、550~740nmの最大吸収波長の消光度Eの合計値に、上記消光度1あたりの金含有率を乗算して、金含有率をそれぞれ算出する。
金含有率(質量%)=(D+E)×(A÷(B+C))
純度指数=B÷C
前記純度指数の値が低いほど金ナノプレートの純度が高いと言える。本発明の被験物質を検出するための組成物においては、被験物質の検出が可能である限り、金ナノプレートの純度指数は限定されるものではないが、例えば、前記純度指数は、0.05~2.00であってもよく、好ましくは0.10~1.0である。金ナノプレートの純度指数は、金含有率、金ナノプレートの平均最大長さ、及び金ナノプレートの最大吸収波長が等しい組成物同士を比較する場合に有用であり得る。
また、MSの場合、既知濃度の第四級アンモニウムカチオンを含む溶液を測定し、第四級アンモニウムカチオン由来のシグナル強度より検量線を作成する。次に、本発明の組成物を測定し、得られた検出強度より第四級アンモニウムカチオンの濃度を算出することができる。例えば、飛行時間型質量分析計を使用する場合、一定強度のイオン化レーザーにより既知濃度の第四級アンモニウムカチオンを含む試料を測定後、本発明の組成物を測定し、シグナル強度を比較することで第四級アンモニウムカチオンの濃度を算出することができる。若しくは、本発明の組成物に既知濃度内部標準物質を添加し、得られたスペクトル中の第四級アンモニウムカチオン由来のシグナル強度と内部標準物質由来のシグナル強度を比較することで第四級アンモニウムカチオンの濃度を算出することができる。
さらにHPLCの場合、初めに複数の既知濃度の第四級アンモニウムカチオンを含む溶液を測定し、得られたチャート中の第四級アンモニウムカチオン由来のピーク面積より検量線を作成する。次に、本発明の組成物を測定し、得られたチャート中の第四級アンモニウムカチオン由来のピーク面積より第四級アンモニウムカチオンの濃度を算出することができる。例えば、第四級アンモニウムカチオンの分離カラムにはShodex Asahipak GF-310 HQ(昭和電工株式会社製)、第四級アンモニウムカチオンの検出には電気伝導度検出器や示差屈折率検出器が使用できる。
本発明の組成物が水溶性高分子を含む場合には、その組成物中に溶解又は分散している水溶性高分子の濃度は、例えば100μM以下であってもよく、好ましくは50μM以下、さらに好ましくは10μM以下である。あるいは、前記水溶性高分子の濃度は、例えば1質量%以下であってもよく、好ましくは0.5質量%以下、さらに好ましくは0.1質量%以下である。
本発明は、特定の理論に拘束されるものではないが、本発明の組成物に水溶性高分子を配合すると、前記組成物中において金ナノプレートの分散安定性が向上する。また、水溶性高分子の濃度が100μM以下又は1質量%以下であると、その濃度が100μM又は1質量%を超える場合と比較して、例えば、前記被験物質に対する特異的結合物質が金ナノプレートに良好に担持されるか、又は、被験物質に対する特異的結合物質を担持した金ナノプレートが被験物質と安定した複合体を形成するため、結果としてその被験物質の検出感度を高めることができる。
NMRの場合、本発明の組成物を乾燥させ、得られた固形分を重溶媒で溶解(分散)させて測定する。重溶媒には既知濃度の内部標準物質(例えば、マレイン酸)を予め添加し、得られたスペクトル中の水溶性高分子由来のシグナルの積分値と内部標準物質由来のシグナルの積分値を比較することで水溶性高分子の濃度を算出することができる。
また、GPCの場合、初めに複数の既知濃度の水溶性高分子水溶液を分析し、得られたチャート中の水溶性高分子由来のピーク面積より検量線を作成する。次に、本発明の組成物を測定し、得られたチャート中の水溶性高分子由来のピーク面積より水溶性高分子の濃度を算出することができる。
さらに、TG-DTAの場合、本発明の組成物の乾燥固形分を測定した際の、重量変化から水溶性高分子量を算出することができる。
被験物質又は被験物質に結合する物質としての核酸は、例えば、DNA、RNA、オリゴヌクレオチド、ポリヌクレオチド、又は、それらの増幅物であってもよい。
前記複合体は、被験物質の検出の分野で通常用いられる手段又は凝集物若しくは沈殿物を検出するのに通常用いられる手段を、特に制限なく利用することによって検出することができる。例えば、前記複合体の形成を、消光度測定、吸光度測定、濁度測定、粒度分布測定、粒子径測定、ラマン散乱光測定、色調変化の観察、凝集又は沈殿形成の観察、イムノクロマトグラフィー法、電気泳動、及び、フローサイトメトリーから成る群から選択される手段によって検出してもよい。
消光度測定又は吸光度測定は、200~2500nmの範囲の波長領域で行ってもよく、430~2000nmの範囲の金ナノプレートの最大吸収波長において行ってもよい。消光度又は吸光度は、これらの測定のために通常用いられる紫外可視分光光度計(例えば、紫外可視近赤外分光光度計MPC3100UV-3100PC、株式会社島津製作所)などによって測定してもよい。
本発明の組成物においては、被験物質とその被験物質に対する特異的結合物質を担持した金ナノプレートとの複合体が形成することで複数の粒子が隣接して存在する場合、双極子-双極子相互作用により金ナノプレートの最大吸収波長よりも長波長側に、前記複合体の形成に依存して消光度又は吸光度が上昇する波長領域が存在している(Nano Lett., Vol. 4, No. 9, (2004), p.1627-1631)。この波長領域における消光度又は吸光度を濁度として測定することにより、前記複合体を検出してもよい。例えば、金ナノプレートの最大吸収波長が560nmである場合には、660~840nmの波長領域において濁度測定を行ってもよく、金ナノプレートの最大吸収波長が620nmである場合には、720~900nmの波長領域において濁度測定を行ってもよい。濁度は、その測定のために通常用いられる紫外可視分光光度計(例えば、紫外可視近赤外分光光度計MPC3100UV-3100PC、株式会社島津製作所)又は濁度計などによって測定してもよい。
1.被験物質を含まない展開液を使用したイムノクロマト試験時の検出ラインと検出ライン以外の部分(対照領域)との輝度差
2.被験物質を含む展開液を使用したイムノクロマト試験時のイムノクロマト担体上の検出ラインと検出ライン以外の部分(対照領域)との輝度差
また、近赤外域の光を吸収する金ナノプレートを使用したイムノクロマト試験においては、試験後に近赤外光を検出部に照射し、そのときの吸光を測定することで検出を確認することができる。
(1)金ナノプレート種粒子の懸濁液の作製
100mMのヘキサデシルトリメチルアンモニウムクロリド(CTAC)水溶液24gに、50mMの塩化金酸水溶液0.13g、10mMの水素化ホウ素ナトリウム水溶液1.5gを撹拌しながら添加した。得られた溶液をインキュベーター(30℃)中で120分間静置し、金ナノプレート種粒子懸濁液を作製した。作製した懸濁液(原液)の光学特性を図1に示す。光学特性の測定は、株式会社島津製作所製の紫外可視近赤外分光光度計MPC3100UV-3100PCを用い、光路長:1cm及び測定波長:190-1300nmの条件下で行われた。
金ナノプレート種粒子懸濁液の金含有率は、懸濁液の総質量に対して0.0049質量%であった。この懸濁液の光学特性を図1に示す。
(2-1)予備成長を含まない合成
(2-1-1)金ナノプレートの懸濁液(懸濁液A)の作製
50mMのCTAC水溶液36gに強力攪拌子(φ8×38mm)を入れ、500rpmで攪拌し、50mMの塩化金酸水溶液を0.5g、10mMのよう化ナトリウム水溶液を0.3g、そして100mMのL-アスコルビン酸水溶液を0.4g添加した。30秒間攪拌後、金ナノプレート種粒子懸濁液の10倍希釈溶液を0.3g添加し、5秒間攪拌した。得られた懸濁液をインキュベーター(30℃)内で12時間静置し、金ナノプレートの懸濁液(懸濁液A)を作製した。この懸濁液の作製に使用したCTAC水溶液からの希釈率により比例計算(ただし、各溶液の比重は1と仮定し、前記金ナノプレート種粒子懸濁液の10倍希釈溶液中のCTACの量は無視)すると、懸濁液Aの分散媒中のCTAC濃度は48.0mMと求められた(特に断らない限り、他の実施例でも同様の計算方法を採用した)。
懸濁液Aを超純水で4倍容に希釈したときの懸濁液の色調は青色であった。この4倍希釈懸濁液の光学特性を図2に示す。最大吸収を示す波長は648nm(消光度:0.86)であった。プレート状ではない多面体又は球状の金ナノ粒子の最大吸収波長である540nmの消光度は0.52であったので、金ナノプレートの純度指数は0.60と計算された。光学特性の測定は、株式会社島津製作所製の紫外可視近赤外分光光度計MPC3100UV-3100PCを用い、光路長:1cm及び測定波長:190-1300nmの条件下で行われた。得られた懸濁液A中の金ナノプレートを、超高分解能分析走査電子顕微鏡SU-70(株式会社日立製作所製)により、走査電子顕微鏡(SEM)観察を行った結果を図3に示す。SEM観察より、金ナノプレートの最大長さは50nm、厚さは24nmで、アスペクト比は2.1であることがわかった。
懸濁液AのpHは、室温下(20℃)で測定した結果、2.4であった。pH測定は株式会社堀場製作所のTwin pHメータ B-212(ガラス電極法)を用いた。
懸濁液Aの金含有率は、懸濁液の総質量に対して0.0131質量%であった。
(2-2-1)金ナノプレートの懸濁液(懸濁液K)の作製
15mMのCTAC水溶液3.16gを攪拌し、20mMのよう化ナトリウム水溶液を0.004g、50mMの塩化金酸水溶液を0.013g、そして100mMのL-アスコルビン酸水溶液0.013gを添加し、予備成長液を作製した。次いで、51mMのCTAC水溶液40gを攪拌し、20mMのよう化ナトリウム水溶液を0.15g、50mMの塩化金酸水溶液を0.5g、そして100mMのL-アスコルビン酸水溶液0.4gを添加し、本成長液を作製した。予備成長液へ金ナノプレート種粒子懸濁液の10倍希釈溶液を0.031g添加し、予備成長金ナノプレート懸濁液を作製した。予備成長金ナノプレート懸濁液全量を本成長液へ添加した。得られた懸濁液をインキュベーター(30℃)内で12時間静置し、金ナノプレートの懸濁液(懸濁液K)を作製した。懸濁液Kの分散媒中のCTAC濃度は47.2mMと計算された。
懸濁液Kを超純水で4倍容に希釈したときの懸濁液の色調は青色であった。この4倍希釈懸濁液の光学特性を図17に示す。最大吸収を示す波長は644nm(消光度:0.94)であった。プレート状ではない多面体又は球状の金ナノ粒子の最大吸収波長である540nmの消光度は0.53であったので、金ナノプレートの純度指数は0.56と計算された。光学特性の測定は、株式会社島津製作所製の紫外可視近赤外分光光度計MPC3100UV-3100PCを用い、光路長:1cm及び測定波長:190-1300nmの条件下で行われた。得られた懸濁液K中の金ナノプレートを、超高分解能分析走査電子顕微鏡SU-70(株式会社日立製作所製)により、走査電子顕微鏡(SEM)観察を行った結果を図18に示す。また、懸濁液KのpHは、室温下(20℃)で測定した結果、2.5であった。pH測定は株式会社堀場製作所のTwin pHメータ B-212(ガラス電極法)を用いた。
懸濁液Kの金含有率は、懸濁液の総質量に対して0.0114質量%であった。
上記金ナノプレート種粒子懸濁液の10倍希釈溶液の添加量を0.031gから0.016gに変更した以外は、金ナノプレートの懸濁液(懸濁液K)と同様にして、金ナノプレートの懸濁液(懸濁液L)を作製した。懸濁液Lの分散媒中のCTAC濃度は47.1mMと計算された。
懸濁液Lを超純水で4倍容に希釈したときの懸濁液の色調は青色であった。この4倍希釈懸濁液の光学特性を図19に示す。最大吸収を示す波長は668nm(消光度:0.99)であった。プレート状ではない多面体又は球状の金ナノ粒子の最大吸収波長である540nmの消光度は0.48であったので、金ナノプレートの純度指数は0.48と計算された。光学特性の測定は、株式会社島津製作所製の紫外可視近赤外分光光度計MPC3100UV-3100PCを用い、光路長:1cm及び測定波長:190-1300nmの条件下で行われた。得られた懸濁液L中の金ナノプレートを、超高分解能分析走査電子顕微鏡SU-70(株式会社日立製作所製)により、走査電子顕微鏡(SEM)観察を行った結果を図20に示す。また、懸濁液LのpHは、室温下(20℃)で測定した結果、2.5であった。pH測定は株式会社堀場製作所のTwin pHメータ B-212(ガラス電極法)を用いた。
懸濁液Lの金含有率は、懸濁液の総質量に対して0.0114質量%であった。
上記金ナノプレート種粒子懸濁液の10倍希釈溶液の添加量を0.031gから0.062gに変更した以外は、金ナノプレートの懸濁液(懸濁液K)と同様にして、金ナノプレートの懸濁液(懸濁液M)を作製した。懸濁液Mの分散媒中のCTAC濃度は47.1mMと計算された。
懸濁液Mを超純水で4倍容に希釈したときの懸濁液の色調は青色であった。この4倍希釈懸濁液の光学特性を図21に示す。最大吸収を示す波長は628nm(消光度:0.84)であった。プレート状ではない多面体又は球状の金ナノ粒子の最大吸収波長である540nmの消光度は0.59であったので、金ナノプレートの純度指数は0.70と計算された。光学特性の測定は、株式会社島津製作所製の紫外可視近赤外分光光度計MPC3100UV-3100PCを用い、光路長:1cm及び測定波長:190-1300nmの条件下で行われた。得られた懸濁液M中の金ナノプレートを、超高分解能分析走査電子顕微鏡SU-70(株式会社日立製作所製)により、走査電子顕微鏡(SEM)観察を行った結果を図22に示す。また、懸濁液MのpHは、室温下(20℃)で測定した結果、2.4であった。pH測定は株式会社堀場製作所のTwin pHメータ B-212(ガラス電極法)を用いた。
懸濁液Mの金含有率は、懸濁液の総質量に対して0.0114質量%であった。
上記金ナノプレート種粒子懸濁液の10倍希釈溶液の添加量を0.031gから0.155gに変更した以外は、金ナノプレートの懸濁液(懸濁液K)と同様にして、金ナノプレートの懸濁液(懸濁液N)を作製した。懸濁液Nの分散媒中のCTAC濃度は47.0mMと計算された。
懸濁液Nを超純水で4倍容に希釈したときの懸濁液の色調は紫色であった。この4倍希釈懸濁液の光学特性を図23に示す。消光スペクトルの内、最大吸収を示す波長の長波長側に有るピークの位置(金ナノプレート由来のピーク)は602nm(消光度:0.58)であった。プレート状ではない多面体又は球状の金ナノ粒子の最大吸収波長である540nmの消光度は0.64であったので、金ナノプレートの純度指数は1.10と計算された。光学特性の測定は、株式会社島津製作所製の紫外可視近赤外分光光度計MPC3100UV-3100PCを用い、光路長:1cm及び測定波長:190-1300nmの条件下で行われた。得られた懸濁液N中の金ナノプレートを、超高分解能分析走査電子顕微鏡SU-70(株式会社日立製作所製)により、走査電子顕微鏡(SEM)観察を行った結果を図24に示す。また、懸濁液NのpHは、室温下(20℃)で測定した結果、2.4であった。pH測定は株式会社堀場製作所のTwin pHメータ B-212(ガラス電極法)を用いた。
懸濁液Nの金含有率は、懸濁液の総質量に対して0.0114質量%であった。
(3-1)懸濁液Aの精製(懸濁液Bの作製)
懸濁液A30gに、1.0MのCTAC水溶液を4.0g注入して攪拌した。30℃で、12時間静置し、上澄み液を除去した。沈殿物を50mMのCTAC水溶液20gで分散し、懸濁液Aの精製物(懸濁液B)を作製した。懸濁液Bの分散媒中のCTAC濃度は50mMとみなした(再分散時に使用した分散媒におけるCTAC濃度)。
懸濁液Bを超純水で4倍容に稀釈したときの懸濁液の色調はシアン調であった。この4倍希釈懸濁液の光学特性を図4Aに示す。最大吸収を示す波長は644nm(消光度:0.61)であった。プレート状ではない多面体又は球状の金ナノ粒子の最大吸収波長である540nmの消光度は0.22であったので、金ナノプレートの純度指数は0.36と計算された。消光度比率より、懸濁液Bの金含有率の概算値は、懸濁液の総質量に対して0.0079質量%であった。最大吸収波長における消光度の高さを合せるために測定波長域すべての消光度に1.41を乗算した懸濁液Bの消光スペクトル(実線)と、上記懸濁液Aの消光スペクトル(破線)との比較を図4Bに示す。懸濁液Bの消光スペクトルでは、プレート状ではない多面体又は球状の金ナノ粒子の最大吸収波長である540nmの消光度が、39%減少していた。光学特性の測定は、株式会社島津製作所製の紫外可視近赤外分光光度計MPC3100UV-3100PCを用い、光路長:1cm及び測定波長:190-1300nmの条件下で行われた。得られた懸濁液B中の金ナノプレートを超高分解能分析走査電子顕微鏡SU-70(株式会社日立製作所製)により、走査電子顕微鏡(SEM)観察を行った結果を図5に示す。SEM観察より、金ナノプレートの最大長さは52nm、厚さは19nmで、アスペクト比は2.7であることがわかった。
懸濁液B中の金ナノプレートのゼータ電位は+41.4 ± 1.2であった。ゼータ電位測定は大塚電子株式会社のPhotal ELS-Zを用いた。
また、上記懸濁液Aの沈殿物を50mMのCTAC水溶液0.02gで分散し、懸濁液Aの精製物(懸濁液W)を作製した時、金含有率の概算値は、懸濁液の総質量に対して7.9質量%であった。

懸濁液O中の金ナノプレートのゼータ電位は+42.0 ± 1.5であった。ゼータ電位測定は大塚電子株式会社のPhotal ELS-Zを用いた。

懸濁液Pを超純水で4倍容に稀釈したときの懸濁液の色調は薄水色調であった。この4倍希釈懸濁液の光学特性を図27Aに示す。最大吸収を示す波長は668nm(消光度:0.58)であった。プレート状ではない多面体又は球状の金ナノ粒子の最大吸収波長である540nmの消光度は0.17であったので、金ナノプレートの純度指数は0.29と計算された。消光度比率より、懸濁液Pの金含有率の概算値は、懸濁液の総質量に対して0.0059質量%であった。最大吸収波長における消光度の高さを合せるために測定波長域すべての消光度に1.70を乗算した懸濁液Pの消光スペクトル(実線)と、上記懸濁液Lの消光スペクトル(破線)との比較を図27Bに示す。懸濁液Pの消光スペクトルでは、プレート状ではない多面体又は球状の金ナノ粒子の最大吸収波長である540nmの消光度が、40%減少していた。光学特性の測定は、株式会社島津製作所製の紫外可視近赤外分光光度計MPC3100UV-3100PCを用い、光路長:1cm及び測定波長:190-1300nmの条件下で行われた。得られた懸濁液中の金ナノプレートを超高分解能分析走査電子顕微鏡SU-70(株式会社日立製作所製)により、走査電子顕微鏡(SEM)観察を行った結果を図28Aに示す。鮮明な像を観察するため、懸濁液Pにチオール末端ポリエチレングリコール(Mw:20,000)を添加し、透過走査電子顕微鏡(STEM)観察を行った結果を図28Bに示す。SEM、STEM観察より、金ナノプレートの最大長さは91nm、厚さは41nmで、アスペクト比は2.2であることがわかった。
懸濁液P中の金ナノプレートのゼータ電位は+41.6 ± 2.0であった。ゼータ電位測定は大塚電子株式会社のPhotal ELS-Zを用いた。

懸濁液Rを超純水で4倍容に稀釈したときの懸濁液の色調は薄水色調であった。この4倍希釈懸濁液の光学特性を図31Aに示す。最大吸収を示す波長は602nm(消光度:0.28)であった。プレート状ではない多面体又は球状の金ナノ粒子の最大吸収波長である540nmの消光度は0.16であったので、金ナノプレートの純度指数は0.57と計算された。消光度比率より、懸濁液Rの金含有率の概算値は、懸濁液の総質量に対して0.0041質量%であった。最大吸収波長における消光度の高さを合せるために測定波長域すべての消光度に2.06を乗算した懸濁液Rの消光スペクトル(実線)と、上記懸濁液Nの消光スペクトル(破線)との比較を図31Bに示す。懸濁液Rの消光スペクトルでは、プレート状ではない多面体又は球状の金ナノ粒子の最大吸収波長である540nmの消光度が、48%減少していた。光学特性の測定は、株式会社島津製作所製の紫外可視近赤外分光光度計MPC3100UV-3100PCを用い、光路長:1cm及び測定波長:190-1300nmの条件下で行われた。得られた懸濁液中の金ナノプレートを超高分解能分析走査電子顕微鏡SU-70(株式会社日立製作所製)により、走査電子顕微鏡(SEM)観察を行った結果を図32Aに示す。鮮明な像を観察するため、懸濁液Rにチオール末端ポリエチレングリコール(Mw:20,000)を添加し、透過走査電子顕微鏡(STEM)観察を行った結果を図32Bに示す。SEM、STEM観察より、金ナノプレートの最大長さは37nm、厚さは19nmで、アスペクト比は1.9であることがわかった。
懸濁液R中の金ナノプレートのゼータ電位は+41.1± 1.8であった。ゼータ電位測定は大塚電子株式会社のPhotal ELS-Zを用いた。
上記(3)で作製した懸濁液Bを40mL遠心分離(8,000×g、30℃、10分間)して金ナノプレートを沈殿させ、上澄み液39.6mLを除去した。その後、遠沈管底部の金ナノプレートに20μMのポリ(p-スチレンスルホン酸ナトリウム)(Na-PSS)水溶液を39.6mL添加して再分散させ、PSS被覆金ナノプレート中間体1懸濁液(懸濁液C)を作製した。懸濁液Cの分散媒中のCTAC濃度は0.50mMと計算され、PSS濃度は20μMとみなした(再分散時に使用した分散媒におけるPSS濃度)。懸濁液C中の金ナノプレートのゼータ電位は-51.0 ± 0.7であった。ゼータ電位測定は大塚電子株式会社のPhotal ELS-Zを用いた。
懸濁液Cを遠心分離(8,000×g、30℃、10分間)してPSS被覆金ナノプレート中間体1を沈殿させ、上澄み液39.6mLを除去した。その後、遠沈管底部のPSS被覆金ナノプレート中間体1に20μMのPSS水溶液を39.6mL添加して再分散させ、PSS被覆金ナノプレート中間体2懸濁液(懸濁液D)を作製した。懸濁液Dの分散媒中のCTAC濃度は0.50×10 -2 mMと計算され、PSS濃度は20μMとみなした。懸濁液D中の金ナノプレートのゼータ電位は-51.0 ± 0.4であった。ゼータ電位測定は大塚電子株式会社のPhotal ELS-Zを用いた。懸濁液Dを遠心分離(8,000×g、30℃、10分間)してPSS被覆金ナノプレート中間体2を沈殿させ、上澄み液39.6mLを除去した。その後、PSS被覆金ナノプレート中間体2に20μMのPSS水溶液を39.6mL添加して再分散させ、PSS被覆金ナノプレート懸濁液(懸濁液E)を作製した。懸濁液Eの分散媒中のCTAC濃度は0.50×10 -4 mMと計算され、PSS濃度は20μMとみなした。懸濁液E中の金ナノプレートのゼータ電位は-54.5 ± 2.0であった。ゼータ電位測定は大塚電子株式会社のPhotal ELS-Zを用いた。PSS被覆金ナノプレートの懸濁液のpHは、室温下(20℃)で測定した結果、7.0であった。pH測定は株式会社堀場製作所のTwin pHメータ B-212(ガラス電極法)を用いた。
懸濁液Eを超純水で4倍容に希釈した懸濁液の色調はシアン調であった。この4倍希釈懸濁液の光学特性を図6に示す。最大吸収を示す波長は638nm(消光度:0.64)であった。プレート状ではない多面体又は球状の金ナノ粒子の最大吸収波長である540nmの消光度は0.25であったので、金ナノプレートの純度指数は0.39と計算された。消光度比率より、懸濁液Eの金含有率の概算値は、懸濁液の総質量に対して0.0085質量%であった。光学特性の測定は、株式会社島津製作所製の紫外可視近赤外分光光度計MPC3100UV-3100PCを用い、光路長:1cm及び測定波長:190-1300nmの条件下で行われた。
懸濁液E中の金ナノプレートを超高分解能分析走査電子顕微鏡SU-70(株式会社日立製作所製)により、走査電子顕微鏡(SEM)観察を行った結果を図7に示す。SEM観察より、金ナノプレートの最大長さは53nm、厚さは19nmで、アスペクト比は2.8であることがわかった。
(5-1)クエン酸被覆金ナノプレート懸濁液(懸濁液F~H)の作製(PSS被覆金ナノプレート水懸濁液E中のCTAC濃度の低減及びPSS濃度の低減)
上記(4)で作製したPSS被覆金ナノプレート水懸濁液(懸濁液E)の40mLを遠心分離(8,000×g、30℃、10分間)してPSS被覆金ナノプレートを沈殿させ、上澄み液39.6mLを除去した。その後、遠沈管底部のPSS被覆金ナノプレートに4.5mMのクエン酸三ナトリウム水溶液を39.6mL添加して再分散させ、クエン酸被覆金ナノプレート中間体1懸濁液(懸濁液F)を作製した。懸濁液Fの分散媒中のCTAC濃度は0.50×10 -6 mMと計算され、PSS濃度は0.20μMと計算された。懸濁液F中の金ナノプレートのゼータ電位は-47.5 ± 1.0であった。ゼータ電位測定は大塚電子株式会社のPhotal ELS-Zを用いた。懸濁液Fを遠心分離(8,000×g、30℃、10分間)してクエン酸被覆金ナノプレート中間体1を沈殿させ、上澄み液39.6mLを除去した。その後、遠沈管底部のクエン酸被覆金ナノプレート中間体1に4.5mMのクエン酸三ナトリウム水溶液39.6mLを添加して再分散させ、クエン酸被覆金ナノプレート中間体2懸濁液(懸濁液G)を作製した。懸濁液Gの分散媒中のCTAC濃度は0.50×10 -8 mMと計算され、PSS濃度は0.20×10 -2 μMと計算された。懸濁液G中の金ナノプレートのゼータ電位は-44.2 ± 1.7であった。ゼータ電位測定は大塚電子株式会社のPhotal ELS-Zを用いた。懸濁液Gを遠心分離(8,000×g、30℃、10分間)してクエン酸被覆金ナノプレート中間体2を沈殿させ、上澄み液39.6mLを除去した。その後、クエン酸被覆金ナノプレート中間体2に4.5mMのクエン酸水溶液を39.6mL添加して再分散させ、クエン酸被覆金ナノプレート懸濁液(懸濁液H)を作製した。懸濁液Hの分散媒中のCTAC濃度は0.50×10 -10 mMと計算され、PSS濃度は0.20×10 -4 μMと計算された。懸濁液H中の金ナノプレートのゼータ電位は-42.2 ± 1.4であった。ゼータ電位測定は大塚電子株式会社のPhotal ELS-Zを用いた。懸濁液HのpHを室温下(20℃)で測定した結果、pH8.4であった。pH測定は株式会社堀場製作所のTwin pHメータ B-212(ガラス電極法)を用いた。
懸濁液Hを超純水で4倍容に希釈した懸濁液の色調はシアン調であった。この4倍希釈懸濁液の光学特性を図8に示す。最大吸収を示す波長は638nm(消光度:0.33)であった。プレート状ではない多面体又は球状の金ナノ粒子の最大吸収波長である540nmの消光度は0.14であったので、金ナノプレートの純度指数は0.42と計算された。消光度比率より、懸濁液Hの金含有率の概算値は、懸濁液の総質量に対して0.0045質量%であった。光学特性の測定は、株式会社島津製作所製の紫外可視近赤外分光光度計MPC3100UV-3100PCを用い、光路長:1cm及び測定波長:190-1300nmの条件下で行われた。
懸濁液H中の金ナノプレートを超高分解能分析走査電子顕微鏡SU-70(株式会社日立製作所製)により、走査電子顕微鏡(SEM)観察を行った結果を図9に示す。SEM観察より、金ナノプレートの最大長さは55nm、厚さは19nmで、アスペクト比は2.9であることがわかった。
金ナノプレート懸濁液Bを懸濁液Hとする上記工程を、懸濁液O~Rについても同様に実施し、クエン酸被覆金ナノプレート懸濁液(懸濁液S~V)を作製した。
懸濁液S
懸濁液Sの分散媒中のCTAC濃度は0.50×10 -10 mMと計算され、PSS濃度は0.20×10 -4 μMと計算された。懸濁液S中の金ナノプレートのゼータ電位は-41.5 ± 0.5であった。懸濁液SのpHを室温下(20℃)で測定した結果、pH7.9であった。懸濁液Sを超純水で4倍容に希釈した懸濁液の色調はシアン調であった。最大吸収を示す波長は634nm(消光度:0.32)であった。プレート状ではない多面体又は球状の金ナノ粒子の最大吸収波長である540nmの消光度は0.12であったので、金ナノプレートの純度指数は0.38と計算された。消光度比率より、懸濁液Sの金含有率の概算値は、懸濁液の総質量に対して0.0034質量%であった。
懸濁液T
懸濁液Tの分散媒中のCTAC濃度は0.50×10 -10 mMと計算され、PSS濃度は0.20×10 -4 μMと計算された。懸濁液T中の金ナノプレートのゼータ電位は-42.2 ± 1.2であった。懸濁液TのpHを室温下(20℃)で測定した結果、pH8.2であった。懸濁液Tを超純水で4倍容に希釈した懸濁液の色調は薄水色調であった。最大吸収を示す波長は658nm(消光度:0.23)であった。プレート状ではない多面体又は球状の金ナノ粒子の最大吸収波長である540nmの消光度は0.07であったので、金ナノプレートの純度指数は0.30と計算された。消光度比率より、懸濁液Tの金含有率の概算値は、懸濁液の総質量に対して0.0023質量%であった。
懸濁液U
懸濁液Uの分散媒中のCTAC濃度は0.50×10 -10 mMと計算され、PSS濃度は0.20×10 -4 μMと計算された。懸濁液U中の金ナノプレートのゼータ電位は-41.7 ± 2.7であった。懸濁液UのpHを室温下(20℃)で測定した結果、pH8.1であった。懸濁液Uを超純水で4倍容に希釈した懸濁液の色調は青色調であった。最大吸収を示す波長は618nm(消光度:0.28)であった。プレート状ではない多面体又は球状の金ナノ粒子の最大吸収波長である540nmの消光度は0.13であったので、金ナノプレートの純度指数は0.46と計算された。消光度比率より、懸濁液Uの金含有率の概算値は、懸濁液の総質量に対して0.0033質量%であった。
懸濁液V
懸濁液Vの分散媒中のCTAC濃度は0.50×10 -10 mMと計算され、PSS濃度は0.20×10 -4 μMと計算された。懸濁液V中の金ナノプレートのゼータ電位は-40.8 ± 1.5であった。懸濁液VのpHを室温下(20℃)で測定した結果、pH8.2であった。懸濁液Vを超純水で4倍容に希釈した懸濁液の色調は青色調であった。最大吸収を示す波長は592nm(消光度:0.11)であった。プレート状ではない多面体又は球状の金ナノ粒子の最大吸収波長である540nmの消光度は0.07であったので、金ナノプレートの純度指数は0.64と計算された。消光度比率より、懸濁液Uの金含有率の概算値は、懸濁液の総質量に対して0.0017質量%であった。
懸濁液S~Vのゼータ電位測定は大塚電子株式会社のPhotal ELS-Zを用いた。懸濁液S~VのpH測定は株式会社堀場製作所のTwin pHメータ B-212(ガラス電極法)を用いた。懸濁液S~Vの光学特性の測定は、株式会社島津製作所製の紫外可視近赤外分光光度計MPC3100UV-3100PCを用い、光路長:1cm及び測定波長:190-1300nmの条件下で行われた。また、懸濁液S~Vの懸濁液の光学特性を図33、最大吸収波長の消光度を1.0とした際の光学特性を図34に示す。
(1)銀ナノプレート種粒子の作製
2.5mMのクエン酸三ナトリウム水溶液20mLに、0.5g/Lの分子量70,000ポリスチレンスルホン酸水溶液1mLと、10mMの水素化ホウ素ナトリウム水溶液1.2mLとを添加し、次いで、20mL/分で攪拌しながら、0.5mMの硝酸銀水溶液50mLを添加した。得られた溶液をインキュベーター(30℃)中で60分間静置し、銀ナノプレート種粒子の懸濁液を作製した。
作製した懸濁液(原液)の光学特性を図10に示す。光学特性の測定は、株式会社島津製作所製の紫外可視近赤外分光光度計MPC3100UV-3100PCを用い、光路長:1cm及び測定波長:190-1300nmの条件下で行われた。銀ナノプレート種粒子の最大吸収波長は、球状銀ナノ粒子のLSPR由来の最大吸収波長に相当する396nm(消光度3.3)であった。なお、本発明の消光度とは懸濁液を分光光度計で測定した際の吸光度の値である。また、超高分解能分析走査電子顕微鏡SU-70(株式会社日立製作所製)により、走査電子顕微鏡(SEM)観察を行った結果を図11に示す。SEM観察より、粒子径は主に3nm以上、10nm未満の粒子(アスペクト比は約1.0)であることがわかった。
超純水200mLに、10mMのアスコルビン酸水溶液4.5mLを添加し、上記(1)で作製した銀ナノプレート種粒子の懸濁液2mLをさらに添加した。得られた溶液に、0.5mMの硝酸銀水溶液120mLを30mL/分で攪拌しながら添加した。硝酸銀水溶液の添加が終了した4分後に攪拌を停止し、25mMのクエン酸三ナトリウム水溶液20mLを添加し、得られた溶液を大気雰囲気下のインキュベーター(30℃)中で100時間静置し、銀ナノプレートの懸濁液を作製した。
作製した懸濁液を超純水で4倍容に希釈した懸濁液の光学特性を図12に示す。最大吸収を示す波長は618nm(消光度1.20)であった。光学特性の測定は、株式会社島津製作所製の紫外可視近赤外分光光度計MPC3100UV-3100PCを用い、光路長:1cm及び測定波長:190-1300nmの条件下で行われた。懸濁液中の銀ナノプレートをSEMにより観察したところ、銀ナノプレートの平均粒子径は50nmであり、平均厚さは10nmで、アスペクト比は5.0であることがわかった。SEM観察写真の解析には株式会社日立製作所製の超高分解能分析走査電子顕微鏡SU-70を用いた。
上記(2)で作製した銀ナノプレートの懸濁液120mLに、0.125mMのポリビニルピロリドン(PVP)(分子量:40,000)の水溶液9.1mLを添加し、0.5Mのアスコルビン酸水溶液1.6mLを添加した後、0.14mMの塩化金酸水溶液9.6mLを0.5mL/分で攪拌しながら添加した。得られた溶液をインキュベーター(30℃)中に24時間静置し、銀ナノプレートの表面が金で被覆された金被覆銀ナノプレートの懸濁液を作製した。
金被覆銀ナノプレートの懸濁液の主要分散媒は水であった。この懸濁液に含まれる金被覆銀ナノプレートは、主面の最大長(粒子径)の平均が50nmの三角形状を含む多角形状、円形状であるプレートの混合物であり、厚さの平均は10nmであった。また、当該金被覆銀ナノプレートにおける金の厚さの平均は0.30nmであった。
金被覆銀ナノプレートの懸濁液のpHは、室温下(20℃)で4.0であった。pH測定は株式会社堀場製作所のTwin pHメータ B-212(ガラス電極法)を用いた。
金被覆銀ナノプレートの懸濁液の銀含有率は、懸濁液の総質量に対して0.0016質量%であった。
上記(3)で得られた懸濁液の1.95mLに、200mMのリン酸緩衝生理食塩水(二価イオンを含まない;以下、「PBS(-)緩衝液」ということもある)0.05mLおよび190mMの炭酸ナトリウム水溶液0.025mLを攪拌しながら添加し、pH調整された金被覆銀ナノプレートの懸濁液(懸濁液I)を作製した。
懸濁液Iを超純水で3.4倍容に希釈した懸濁液の光学特性を図13に示す。最大吸収を示す波長は628nm(消光度1.07)であった。光学特性の測定は、株式会社島津製作所製の紫外可視近赤外分光光度計MPC3100UV-3100PCを用い、光路長:1cm及び測定波長:190-1300nmの条件下で行われた。
懸濁液Iの主要分散媒は水であった。
懸濁液Iに含まれる金被覆銀ナノプレートは、三角形状を含む多角形状であるプレートと円形状であるプレートとの混合物であり、主面の最大長の平均は50nmで、厚さの平均は10nmであった。また、懸濁液Iに含まれる金被覆銀ナノプレートにおける金の厚さの平均は0.30nmであった。超高分解能分析走査電子顕微鏡SU-70(株式会社日立製作所製)により、走査電子顕微鏡(SEM)観察を行った結果を図14に示す。
懸濁液Iの3倍希釈溶液の色調はシアンであった。
懸濁液IのpHは、室温下(20℃)で7.3であった。
懸濁液Iの銀含有率は、懸濁液の総質量に対して0.0015質量%であった。
(1)PVP安定化球状金ナノ粒子懸濁液の作製
市販の球状金ナノ粒子(粒子径:40nm、濃度:0.0068質量%)12mLに0.125mMのポリビニルピロリドン(PVP)(分子量:40,000)の水溶液0.91mLを攪拌しながら添加した。得られた溶液をインキュベーター(30℃)中に24時間静置し、PVP安定化球状金ナノ粒子懸濁液を作製した。
(2)PVP安定化球状金ナノ粒子懸濁液のpH調整
(1)で得られた懸濁液Jの1.95mLに200mMのPBS緩衝液0.05mLおよび190mMの炭酸ナトリウム水溶液0.025mLを攪拌しながら添加し、pH調整されたPVP安定化球状金ナノ粒子懸濁液(懸濁液J)を作製した。
調製した懸濁液Jを超純水で3.8倍容に希釈した水懸濁液の光学特性を図15に示す。最大吸収を示す波長は522nm(消光度0.38)であった。光学特性の測定は、株式会社島津製作所製の紫外可視近赤外分光光度計MPC3100UV-3100PCを用い、光路長:1cm及び測定波長:190-1300nmの条件下で行われた。
懸濁液の主要分散媒は水であった。
懸濁液Jに含まれる球状金ナノ粒子は、平均粒子径が40nmであった。
懸濁液Jの色調は赤色であった。
懸濁液JのpHは、室温下(20℃)で7.0であった。
懸濁液Jの金含有率は、懸濁液の総質量に対して質量0.0061質量%であった。
実施例及び比較例の各懸濁液を下記手順のイムノクロマト試験を用いて、試験結果を評価した。
(1)イムノクロマト試験に使用する展開液の作製(金ナノプレートの各種懸濁液への特異的結合物質の担持;検出試薬の標識)
1mMのPBS(-)緩衝液中における濃度50μg/mLのB型肝炎ウイルス抗原(HBs抗原)に対する抗体(品名:Goat anti HBsAg、製造元:Arista Biologicals,Inc.)(イムノクロマト法における検出抗体)の溶液0.2mLと、金ナノプレートの各種懸濁液(懸濁液A~H、S~V)1.8mLとを混合し、得られた混合物を室温下で60分間振とうした。次いで、1mMのPBS(-)緩衝液中における濃度0.4mMの末端がチオール基で修飾されたポリエチレングリコールであるSUNBRIGHT ME-020SH(分子量:2,000、NOF CORPORATION製)の溶液0.044mLを添加し、得られた懸濁液を室温下で30分間振とうした。次いで、1mMのPBS(-)緩衝液中における濃度0.5mMのウシ血清アルブミン(品名:Bovine Serum Albumin、略称:BSA、分子量:66kDa、製造元:Sigma-Aldrich)の溶液を0.100mL添加し、得られた懸濁液を室温下で30分間振とうした。次いで、この懸濁液を遠心分離(8,000×g、30℃、10分間)して抗体-金ナノプレート複合体を沈殿させ、上澄み液1.90mLを除去した。その後、抗体-金ナノプレート複合体に、1mMのPBS(-)緩衝液(4.9μMのBSA含有)を0.40mL添加して再分散させ、株式会社島津製作所製の紫外可視近赤外分光光度計MPC3100UV-3100PCを使用して消光度(Extinction)が1.0になるように調整し、展開液を作製した。
1mMのPBS(-)緩衝液中における濃度50μg/mLのB型肝炎ウイルス抗原(HBs抗原)に対する抗体(品名:Goat anti HBsAg、製造元:Arista Biologicals,Inc.)(イムノクロマト法における検出抗体)の溶液0.2mLと、金被覆銀ナノプレート懸濁液(懸濁液I)1.8mLとを混合し、得られた混合物を室温下で60分間振とうした。次いで、1mMのPBS(-)緩衝液中における濃度0.4mMの末端がチオール基で修飾されたポリエチレングリコールであるSUNBRIGHT ME-020SH(分子量:2,000、NOF CORPORATION製)の溶液0.044mLを添加し、得られた懸濁液を室温下で30分間振とうした。次いで、1mMのPBS(-)緩衝液中における濃度0.5mMのウシ血清アルブミン(品名:Bovine Serum Albumin、略称:BSA、分子量:66kDa、製造元:Sigma-Aldrich)の溶液0.100mLを添加し、得られた懸濁液を室温下で30分間振とうした。次いで、この懸濁液を遠心分離(26,000×g、30℃、10分間)して抗体-金被覆銀ナノプレート複合体を沈殿させ、上澄み液1.90mLを除去した。その後、抗体-金被覆銀ナノプレート複合体に、1mMのPBS(-)緩衝液(4.9μMのBSA含有)を1.90mL添加して再分散させ、株式会社島津製作所製の紫外可視近赤外分光光度計MPC3100UV-3100PCを使用して消光度(Extinction)が1.0になるように調整し、展開液I(色調:シアン)を作製した。
1mMのPBS(-)緩衝液中における濃度50μg/mLのB型肝炎ウイルス抗原(HBs抗原)に対する抗体(品名:Goat anti HBsAg、製造元:Arista Biologicals,Inc.)(イムノクロマト法における検出抗体)の溶液0.2mLと、PVP安定化球状金ナノ粒子懸濁液(懸濁液J)1.8mLとを混合し、得られた混合物を室温下で60分間振とうした。次いで、1mMのPBS(-)緩衝液中における濃度0.8mMの末端がチオール基で修飾されたポリエチレングリコールであるSUNBRIGHT ME-020SH(分子量:2,000、NOF CORPORATION製)の溶液0.044mLを添加し、得られた懸濁液を室温下で30分間振とうした。次いで、1mMのPBS(-)緩衝液中における濃度0.5mMのウシ血清アルブミン(品名:Bovine Serum Albumin、略称:BSA、分子量:66kDa、製造元:Sigma-Aldrich)の溶液0.100mLを添加し、得られた懸濁液を室温下で30分間振とうした。次いで、この懸濁液を遠心分離(4200×g、30℃、10分間)して抗体-球状金ナノ粒子複合体を沈殿させ、上澄み液1.90mLを除去した。その後、抗体-球状金ナノ粒子複合体に、1mMのPBS(-)緩衝液(4.9μMのBSA含有)を1.90mL添加して再分散させ、株式会社島津製作所製の紫外可視近赤外分光光度計MPC3100UV-3100PCを使用して消光度(Extinction)が1.0になるように調整し、展開液J(色調:赤色)を作製した。
図16に示す手順により、B型肝炎ウイルス抗原(HBs抗原)を被験物質としたイムノクロマト試験を、抗HBs抗体(捕獲抗体)を直線状(図16の検出ライン)に固定化したイムノクロマト試験紙を用いて行った。
イムノクロマト試験紙はイムノクロマト試験紙の受託作製会社(有限会社バイオデバイステクノロジー)より購入したものを使用した。捕獲抗体を直線状に固定する際、捕獲抗体を5mM PBS(-)緩衝液で濃度1mg/mLに調整したものを使用した。
第1展開液(図16(a)で用いる展開液)は、1mMのPBS(-)緩衝液中におけるHBs抗原(品名:HBsAg Protein(Subtype adr)、製造元:Fitzgerald Industries International Inc.)の溶液であった。第1展開液として、HBs抗原の濃度が6.0μM、0.6μM、0.06μM、0.006μM、0M(ブランク)の溶液を用意した。
第2展開液は1mMのPBS(-)緩衝液であった。
第3展開液(図16(c)で用いる展開液)は、上述の展開液A~Jである。
イムノクロマト試験紙に、第1展開液15μLを展開させた(図16(a))。第1展開液の展開により、HBs抗原が試験紙の検出ライン上に固定化された捕獲抗体によって捕獲される(図16(b))。
次いで、第2展開液30μLを展開させて、イムノクロマト試験紙上の余剰抗原の洗浄を行った。
最後に、第3展開液60μLを展開させた(図16(c))。第3展開液の展開により、試験紙の検出ライン上で捕獲されたHBs抗原へ検出抗体(抗体-PSS被覆金ナノプレート複合体など)が結合する(図16(d))。
同一の手順をHBs濃度の異なる第1展開液ごとに繰り返した。
第3展開液展開後の検出ラインにおける金属ナノ粒子の着色を目視で確認することにより、HBs抗原の有無を判定した。結果を次の表2に示す。
+:検出ラインにおける着色を確認できた。
-:検出ラインにおける着色を確認できなかった。
第3展開液展開後のイムノクロマト試験紙をスキャニング(装置名:Cano Scan LiDE500F、製造元:キヤノン株式会社)し、検出ライン(捕獲抗体の固定化部分)の最低輝度と、検出ライン以外の部分(対照領域)の最低輝度とを画像解析ソフト(Image-J:アメリカ国立衛生研究所でWayne Rasbandが開発したオープン・ソースで公有の画像処理ソフトウェア(http://imagej.nih.gov/ij/))を用いて数値化した。最低輝度は、検出ラインと対照領域における異なる5箇所の輝度をそれぞれ1回ずつ測定し、得られた数値の中央値を採用した。輝度差(検出ラインの最低輝度-対照領域の最低輝度)を計算し、結果を次の表3に示す。
第1展開液中のHBs抗原濃度が6.0μMであり、第3の展開液展開後のイムノクロマト試験紙を温度20℃、湿度50%の室内で乾燥させた。乾燥開始時(0時間)、乾燥開始後24時間、48時間、168時間、720時間、720時間、2160時間経過時にイムノクロマト試験紙をスキャニング(装置名:Cano Scan LiDE500F、製造元:キヤノン株式会社)し、得られた画像をビットマップ画像編集ソフトウェアのAdobe Photoshopで取り込み、検出ライン、及び対象領域(検出ライン又は検出ライン以外の部分)のCIELAB D50を測定した。検出ラインと対象領域の色差(ΔE)を、次の式1より算出した。

(補色空間の一種であるLab色空間において、L値は明度を示し、a値及びb値は色味の強弱を示す。a値がプラスのときは赤味を示し、マイナスのとき緑味を示す。b値がプラスのときは黄味を示し、マイナスのときは青紫味を示す。式1中、L1は検出ラインのL値、L2は対照領域のL値、a1は検出ラインのa値、a2は対照領域のa値、b1は検出ラインのb値、そして、b2は対照領域のb値をそれぞれ意味する。)
本実施例のイムノクロマト試験と同様の結果は、ヒト絨毛性ゴナドトロピン(hCG)などの他の被験物質を対象としたイムノクロマト試験においても得ることができたので、本発明の利点は、特定の実施態様に限定されるべきものではないと理解される。
Claims (21)
- 金ナノプレートを含む、被験物質を検出するための組成物であって、
前記金ナノプレートの平均アスペクト比(金ナノプレートの最大長さを厚さで割った値)が、1より大きく10以下であり、
前記組成物中の第四級アンモニウムカチオンの濃度が、0~1mMであり、
前記第四級アンモニウムカチオンが、N+(R)4(各Rは、それぞれ独立して、炭素数1~20の直鎖又は分枝鎖アルキル基から選択される)で表されることを特徴とする、組成物。 - 前記第四級アンモニウムカチオンが、デシルトリメチルアンモニウムカチオン、ヘキサデシルトリメチルアンモニウムカチオン、及び、ヘプタデシルトリメチルアンモニウムカチオンから成る群から選択される、請求項1に記載の組成物。
- 分子量500以上の水溶性高分子を含まない、請求項1又は2に記載の組成物。
- 分子量500以上の水溶性高分子をさらに含み、前記組成物中の前記分子量500以上の水溶性高分子の濃度が、0より大きく1質量%以下である、請求項1又は2に記載の組成物。
- 分子量500以上の水溶性高分子をさらに含み、前記組成物中の前記分子量500以上の水溶性高分子の濃度が、0より大きく100μM以下である、請求項1又は2に記載の組成物。
- 前記金ナノプレートと前記水溶性高分子とが複合体を形成している、請求項4又は5に記載の組成物。
- 前記水溶性高分子が、ポリスチレンスルホン酸ナトリウム、ポリビニルピロリドン、及び、チオール末端ポリエチレングリコールから成る群から選択される、請求項4~6のいずれか一項に記載の組成物。
- クエン酸ナトリウムを含まない、請求項1~7のいずれか一項に記載の組成物。
- クエン酸ナトリウムをさらに含み、前記組成物中の前記クエン酸ナトリウムの濃度が、0より大きく50mM以下である、請求項1~7のいずれか一項に記載の組成物。
- 前記金ナノプレートの平均最大長さが、100nmより小さい、請求項1~9のいずれか一項に記載の組成物。
- 前記金ナノプレートが、表面プラズモン共鳴による光吸収ピークを550~740nmの波長領域に有する、請求項1~10のいずれか一項に記載の組成物。
- 赤色、マゼンタ、紫色、紺色、青色、シアン、又は、薄水色の色調を呈する、請求項1~11のいずれか一項に記載の組成物。
- 前記金ナノプレートが、前記被験物質に対する特異的結合物質を担持している、請求項1~12のいずれか一項に記載の組成物。
- 前記被験物質と前記特異的結合物質の組み合わせが、抗原とそれに結合する抗体、抗体とそれに結合する抗原、糖鎖又は複合糖質とそれに結合するレクチン、レクチンとそれに結合する糖鎖又は複合糖質、ホルモン又はサイトカインとそれに結合する受容体、受容体とそれに結合するホルモン又はサイトカイン、タンパク質とそれに結合する核酸アプタマー若しくはペプチドアプタマー、酵素とそれに結合する基質、基質とそれに結合する酵素、ビオチンとアビジン又はストレプトアビジン、アビジン又はストレプトアビジンとビオチン、IgGとプロテインA又はプロテインG、プロテインA又はプロテインGとIgG、及び、第1の核酸とそれに結合する第2の核酸から成る群から選択される、請求項13に記載の組成物。
- 被験物質を検出するための、請求項1~14のいずれか一項に記載の組成物の凍結乾燥物。
- 請求項13又は14に記載の組成物又は請求項15に記載の凍結乾燥物を使用して、前記被験物質を検出する方法であって、
該組成物、又は、該凍結乾燥物の再懸濁液を、該被験物質と混合して、該被験物質と前記特異的結合物質を担持した金ナノプレートとの複合体を形成する工程、及び、
該複合体を検出する工程を含むことを特徴とする、方法。 - 前記複合体の形成を、消光度測定、吸光度測定、濁度測定、粒度分布測定、粒子径測定、ラマン散乱光測定、色調変化の観察、凝集又は沈殿形成の観察、イムノクロマトグラフィー、電気泳動、及び、フローサイトメトリーから成る群から選択される手段によって検出する、請求項16に記載の方法。
- 請求項1~12のいずれか一項に記載の組成物中の金ナノプレートに、前記被験物質に対する特異的結合物質を担持させて、請求項13又は14に記載の組成物を調製する工程、及び/又は、請求項15に記載の凍結乾燥物を再懸濁する工程をさらに含む、請求項16又は17に記載の方法。
- 請求項1~14のいずれか一項に記載の組成物又は請求項15に記載の凍結乾燥物を含む、請求項16~18のいずれか一項に記載の方法に使用するためのキット。
- 請求項1~14のいずれか一項に記載の組成物又は請求項15に記載の凍結乾燥物を製造する方法であって、
(1)金ナノプレート種粒子の懸濁液を調製する工程、及び、
(2)金ナノプレートの懸濁液を調製する工程
を含むことを特徴とする、方法。 - 前記工程(1)が、金ナノプレート種粒子を予備成長させる工程を含まない、請求項20に記載の方法。
