Abstract
Description
本出願は、参照によりここに援用されるところの日本出願特願2015-041214号優先権を請求する。
このLWEは、瞬目運動による眼瞼結膜縁と眼表面との摩擦の上昇が原因となり眼瞼結膜縁表層の上皮の脱落と変性が起こると考えられており(非特許文献6)、コンタクトレンズ装用者に対するLWEの治療については、コンタクトレンズ装用を中止し、人工涙液点眼によって治療することが好ましいと考えられている。また、コンタクトレンズ非装用者に対するLWEの治療については、ヒアルロン酸点眼液や眼軟膏による眼表面の摩擦軽減が有効であると考えられている。
例えば、特許文献1では、ホホバワックスやマッコウクジラ油、オレンジラフィー油などを配合することで潤滑性を向上させる方法を示しているが、大部分が水である点眼剤に適用したとき、満足できる組成物を得ることは困難であった。
特許文献2では、「2-(メタ)アクリロイルオキシエチルホスホリルコリンと、アルキル(メタ)アクリル酸エステルとを含む単量体組成物を重合した共重合体において、アルキル(メタ)アクリル酸エステルのアルキル基の炭素数と、前記2-(メタ)アクリロイルオキシエチルホスホリルコリン及びアルキル(メタ)アクリル酸エステルによる構成単位の合計量に対する該アルキル(メタ)アクリル酸エステルによる構成単位のモル分率とを、特定なものとし、且つ分子量を制御した共重合体を含むコンタクトレンズ用装着液」を開示している。しかし、本発明の点眼剤に含まれる共重合体は、特許文献2記載の共重合体とは明らかに構造が異なる。
特許文献3では、ソフトコンタクトレンズの濡れ性を向上させ、コンタクトレンズ表面に水分をまとわせることで使用感が改善するものであるが、特許文献3の方法での潤滑性向上効果は不十分であった。
特許文献4では、大幅に潤滑性を向上可能な組成物が開示されているが、コンタクトレンズケア製剤に関するものであるが、点眼剤の例示や眼瞼結膜縁上皮障害については言及されていない。
すなわち、眼瞼結膜縁上皮障害を治療もしくは予防するために角膜表面やソフトコンタクトレンズ表面に対して十分な潤滑性を付与する点眼剤は、いまだ満足のいくものが得られていない状況である。
そこで、本発明の課題は、角膜表面やソフトコンタクトレンズ表面に対して十分な潤滑性を付与し、眼瞼結膜縁上皮障害の治療用又は予防用の点眼剤を提供することにある。
1.式(1a)~式(1c)で表される構成単位を有し、各構成単位の比率na:nb:ncが100:10~400:2~50であり、重量平均分子量が5,000~2,000,000である共重合体(P)を0.01~2.0W/V%含有する点眼剤。
2.前記(1a)が2-(メタクリロイルオキシ)エチル-2’-(トリメチルオンモニオ)エチルホスフェートから得られる構成単位であり、前記(1b)がN,N-ジメチル(メタ)アクリルアミドから得られる構成単位であり、及び前記(1c)がステアリル(メタ)アクリレートから得られる構成単位である前項1に記載の点眼剤。
3.前記(1a)が2-(メタクリロイルオキシ)エチル-2’-(トリメチルオンモニオ)エチルホスフェートから得られる構成単位である、前記(1b)がN,N-ジメチル(メタ)アクリルアミドから得られる構成単位であり、及び前記(1c)がラウリル(メタ)アクリレートから得られる構成単位である前項1に記載の点眼剤。
4.ソフトコンタクトレンズ装用者用である前項1~3のいずれか1に記載の点眼剤。
5.眼瞼結膜縁上皮障害の治療用又は予防用である前項1~4のいずれか1に記載の点眼剤。
6.式(1a)~式(1c)で表される構成単位を有し、各構成単位の比率na:nb:ncが100:10~400:2~50であり、重量平均分子量が5,000~2,000,000である共重合体(P)を用いて点眼剤を製造するための用途。
7.式(1a)~式(1c)で表される構成単位を有し、各構成単位の比率na:nb:ncが100:10~400:2~50であり、重量平均分子量が5,000~2,000,000である共重合体(P)を0.01~2.0W/V%含有する点眼剤を、ヒトを含む哺乳類に投与する点眼方法。
8.前記ヒトを含む哺乳類は、ソフトコンタクトレンズ装用者である前項7に記載の点眼方法。
9.前記ヒトを含む哺乳類は、眼瞼結膜縁上皮障害の治療又は予防が必要な患者である前項7又は8に記載の点眼方法。
本発明の点眼剤に用いる共重合体(P)は、下記の(1)~(3)の3つの構成単位を有し、各構成単位の比率(モル比率)na:nb:ncが100:10~400:2~50である。
<(1)PC構成単位>
本発明の点眼剤に用いる共重合体(P)は、下記式(1a)で表される構成単位(以下「PC構成単位」と略記)を有する。
共重合体(P)中のPC構成単位は、共重合体(P)へ親水性及び含水ゲル形成能を付与し潤滑性を高めるために導入される。
共重合体(P)中のPC構成単位は、共重合体(P)の重合時に使用される下記式(2)で表されるホスホリルコリン類似基含有単量体(以下、PC単量体とする)から得られる。
PC単量体としては、入手性の観点から例えば、2-((メタ)アクリロイルオキシ)エチル-2’-(トリメチルアンモニオ)エチルホスフェートが好ましく、さらに下記式(3)で表される2-(メタクリロイルオキシ)エチル-2’-(トリメチルオンモニオ)エチルホスフェート(以下、MPCとする)が好ましい。
本発明の点眼剤に用いる共重合体(P)は、下記式(1b)で表される構成単位(以下「アミド構成単位」と略記)を有する。
共重合体(P)中のアミド構成単位は、共重合体(P)を高分子量化し、ソフトコンタクトレンズへの密着性を高めるために導入される。
共重合体(P)中のアミド構成単位の割合については、PC構成単位のモル数naを100としたときのモル数nbについて、nb/na=10~400/100であり、好ましくは30~250/100である。nbが大きすぎる場合には点眼剤を製造する際に必要となる無菌ろ過が困難となるおそれがあり、小さすぎる場合には潤滑性向上効果が見込めない。
上記式(1b’)で表される(メタ)アクリルアミドまたは(メタ)アクリルアミド誘導体として例えば、N,N-ジメチル(メタ)アクリルアミド、N,N-ジエチル(メタ)アクリルアミドまたはN-アクリロイルモルホリンが挙げられる。
本発明の点眼剤に用いる共重合体(P)は、下記式(1c)で表される構成単位(以下「疎水性構成単位」と略記)を有する。
共重合体(P)中の疎水性構成単位は、ソフトコンタクトレンズへの吸着性を高め、疎水性相互作用による物理架橋ゲル形成能を高め、潤滑性を向上させるために導入される。
共重合体(P)中の疎水性構成単位は、共重合体(P)の重合時に使用される下記式(1c’)で表される疎水性単量体から得られる。
式(1c’)で表される疎水性単量体として例えば、ラウリル(メタ)アクリレート、ステアリル(メタ)アクリレート、ベヘニル(メタ)アクリレート等の直鎖アルキル(メタ)アクリレートが挙げられる。
共重合体(P)は、本発明の効果を損なわない範囲において、式(1a)~式(1c)で表される構成単位以外の構成単位を導入することもできる。共重合体(P)の製造に用いる単量体組成物に、他の重合性単量体を配合する場合、その配合割合は、本発明の効果に影響を与えない範囲で適宜選択できるが、共重合体(P)を構成する上記式(1a)で示すnaを100とした場合、モル比で50以下が好ましい。
環状アルキル(メタ)アクリレートとしては、例えば、シクロヘキシル(メタ)アクリレート等が挙げられる。
芳香族基含有(メタ)アクリレートとしては、例えば、ベンジル(メタ)アクリレート、フェノキシエチル(メタ)アクリレート等が挙げられる。
スチレン系単量体としては、例えば、スチレン、メチルスチレン、クロロメチルスチレン等が挙げられる。
ビニルエーテル単量体としては、例えば、メチルビニルエーテル、ブチルビニルエーテル等が挙げられる。
ビニルエステル単量体としては、例えば、酢酸ビニル、プロピオン酸ビニル等が挙げられる。
酸基含有単量体としては、例えば、(メタ)アクリル酸、スチレンスルホン酸、(メタ)アクリロイルオキシホスホン酸等が挙げられる。
アミノ基含有単量体としては、例えば、アミノエチルメタクリレート、ジメチルアミノエチル(メタ)アクリレート、N,N-ジメチルアミノプロピル(メタ)アクリルアミド等が挙げられる。
カチオン性基含有単量体としては、例えば、2-ヒドロキシ-3-(メタ)アクリロイルオキシプロピルトリメチルアンモニウムクロライド等が挙げられる。
窒素含有基含有単量体としては、例えば、N-ビニルピロリドン等が挙げられる。
本発明に用いる共重合体(P)のPC構成単位、アミド構成単位及び疎水性構成単位の好ましい組み合わせは、以下の通りであるが、特に限定されない。
PC構成単位が2-(メタクリロイルオキシ)エチル-2’-(トリメチルオンモニオ)エチルホスフェートから得られる構成単位であり、アミド構成単位がN,N-ジメチル(メタ)アクリルアミドから得られる構成単位であり、及び疎水性構成単位がステアリル(メタ)アクリレートから得られる構成単位である共重合体(P)
PC構成単位が2-(メタクリロイルオキシ)エチル-2’-(トリメチルオンモニオ)エチルホスフェートから得られる構成単位であり、アミド構成単位がN,N-ジメチル(メタ)アクリルアミドから得られる構成単位であり、及び疎水性構成単位がラウリル(メタ)アクリレートから得られる構成単位である共重合体(P)
さらに、これらの共重合体(P)のPC構成単位、アミド構成単位及び疎水性構成単位の構成単位の比率(モル比率na:nb:nc)は、100:10~400:2~50である。より詳しくは、nb/na=10~400/100であり、好ましくは30~250/100であり、nc/na=2~50/100であり、好ましくは5~25/100である。
本発明に用いる共重合体(P)は、重量平均分子量5,000~2,000,000であり、好ましくは100,000~1,500,000の重合体である。重量平均分子量が5,000未満の場合は、重合体のコンタクトレンズ表面への吸着力が十分でないため潤滑性向上が見込めないおそれがあり、2,000,000を超える場合は、点眼剤を製造する際に必要となる無菌ろ過が困難となるおそれがある。
共重合体(P)は、上記単量体の配合物をラジカル重合することによって得ることができる。共重合体(P)の製造は、例えば、上記単量体組成物を、ラジカル重合開始剤の存在下、窒素、二酸化炭素、アルゴン、ヘリウム等の不活性ガス置換または雰囲気下においてラジカル重合によりすることができる。重合方法は、塊状重合、懸濁重合、乳化重合、溶液重合等の公知の方法により行うことができる。重合方法は、精製等の観点から溶液重合が好ましい。共重合体(P)の精製は、再沈殿法、透析法、限外ろ過法等の公知の精製方法により行うことが出来る。
ラジカル重合開始剤としては、アゾ系ラジカル重合開始剤、有機過酸化物、過硫酸化物等を挙げることができる。
有機過酸化物としては、例えば、t-ブチルペルオキシネオデカノエート、過酸化ベンゾイル、ジイソプロピルペルオキシジカーボネート、t-ブチルペルオキシ-2-エチルヘキサノエート、t-ブチルペルオキシピバレート、t-ブチルペルオキシジイソブチレート、過酸化ラウロイル、t-ブチルペルオキシネオデカネート、コハク酸ペルオキシド(=サクシニルペルオキシド)等が挙げられる。
過硫酸化物としては、例えば、過硫酸アンモニウム、過硫酸カリウム、過硫酸ナトリウム等が挙げられる。
これらのラジカル重合開始剤は、単独で用いることができ、また、2種以上を混合して用いることもできる。重合開始剤の使用量は、単量体組成物100質量部に対して通常0.001~10質量部、好ましくは0.01~5.0質量部である。
アルコール系溶媒としては、例えば、メタノール、エタノール、n-プロパノール、イソプロパノール等が挙げられる。
ケトン系溶媒としては、例えば、アセトン、メチルエチルケトン、ジエチルケトン等が挙げられる。
エステル系溶媒としては、例えば、酢酸エチル等が挙げられる。
直鎖または環状のエーテル系溶媒としては、例えば、エチルセルソルブ、テトラヒドロフラン等が挙げられる。
含窒素系溶媒としては、例えば、アセトニトリル、ニトロメタン、N-メチルピロリドン等が挙げられる。
好ましくは水またはアルコール又はこれらの混合溶媒が挙げられる。
加えて、本発明の点眼剤の共重合体(P)の濃度は、より好ましくは、0.1~2.0W/V%又は0.01~1.5W/V%であり、さらに好ましくは0.1~1.5W/V%である。
その他の成分としては、例えば、充血除去成分、消炎・収斂成分、ビタミン類、アミノ酸類、サルファ剤、糖類、粘稠化剤、清涼化剤、無機塩、有機酸の塩、酸、塩基、酸化防止剤、安定化剤、防腐剤、ムコ多糖、ムチン分泌促進剤を挙げることができる。
消炎・収斂成分としては、例えば、イプシロン-アミノカプロン酸、アラントイン、ベルベリンまたはその塩、アズレンスルホン酸ナトリウム、グリチルリチン酸またはその塩、乳酸亜鉛、硫酸亜鉛、塩化リゾチームが挙げられる。
アミノ酸類としては、例えば、アスパラギン酸またはその塩、アミノエチルスルホン酸が挙げられる。
サルファ剤としては、例えば、スルファメキサゾールまたはその塩、スルフイソキサゾール、スルフイソミジンナトリウムが挙げられる。
糖類としては、例えば、ブドウ糖、マンニトール、ソルビトール、キシリトール、トレハロース等が挙げられる。
粘稠化剤としては、例えば、ヒドロキシプロピルメチルセルロースが挙げられる。
無機塩としては、例えば、塩化ナトリウム、塩化カリウム、ホウ砂、炭酸水素ナトリウム、リン酸水素ナトリウム、無水リン酸二水素ナトリウムが挙げられる。
有機酸の塩としては、例えば、クエン酸ナトリウムが挙げられる。
酸としては、例えば、ホウ酸、リン酸、クエン酸、硫酸、酢酸、塩酸が挙げられる。
塩基としては、例えば、水酸化ナトリウム、水酸化カリウム、トリスヒドロキシメチルアミノメタン、モノエタノールアミンが挙げられる。
酸化防止剤としては、例えば、酢酸トコフェロール、ジブチルヒドロキシトルエンが挙げられる。
安定化剤としては、例えば、エデト酸ナトリウム、グリシンが挙げられる。
防腐剤としては、例えば、塩化ベンザルコニウム、クロルヘキシジングルコン酸塩、ソルビン酸カリウム、塩酸ポリヘキサニドが挙げられる。
ムコ多糖としては、例えば、ヒアルロン酸ナトリウム、コンドロイチン硫酸ナトリウムが挙げられる。
ムチン分泌促進剤としては、例えば、ジクアホソルナトリウム、レバミピドが挙げられる。
本発明の点眼剤の具体的な製品形態としては、次のようなものを例示することができる。一般点眼薬、抗菌性点眼薬、洗眼薬、コンタクトレンズ装着液、人工涙液などが挙げられる。
本発明の点眼剤は、共重合体(P)、及び所望により上記その他の成分を、室温~50℃程度の水中に添加、攪拌して溶解させることにより製造することが出来る。また共重合体(P)と上記その他の成分の添加順序としては、どの成分から添加してもよい。
製造における加熱、冷却及び攪拌は溶液全体を均一に加熱、冷却及び攪拌することができればよく、いずれも公知の器具、装置を用いることが出来る。
本発明の点眼方法は、式(1a)~式(1c)で表される構成単位を有し、各構成単位の比率na:nb:ncが100:10~400:2~50であり、重量平均分子量が5,000~2,000,000である共重合体(P)を0.01~2.0W/V%含有する点眼剤を、ヒトを含む哺乳類に投与する工程を含む。
点眼方法の対象は、特に限定されないが、ヒトを含む哺乳類であり、特に、ソフトコンタクトレンズ装用者を対象とし、また、好ましくは眼瞼結膜縁上皮障害の治療又は予防が必要な患者を対象とする。
得られた各重合体5mgをメタノール/クロロホルム混液(80:20)に溶かし、試料溶液とした。分析条件は以下を用いた。

重量平均分子量の算出法:分子量計算プログラム(SC-8020用GPCプログラム)

カラムオーブン:40℃付近の一定温度
重合体の重量平均分子量は、ポリエチレングリコールを標準サンプルとしてゲルパーミエーションクロマトグラフ(GPC)により測定した重量平均分子量の値である。
得られた重合体溶液を0.5質量%になるように水で希釈し、この液を0.45μmメンブランフィルターでろ過し、測定した。
各実施例及び比較例の点眼剤のpHは、「第16改正日本薬局方 一般試験法2.54 pH測定法」に従い測定した。
各実施例及び比較例の点眼剤の浸透圧は、「第16改正日本薬局方 一般試験法 2.57 浸透圧測定法(オスモル濃度測定法)」に従い行った。具体的には、氷点測定法によるオズモメーター(Fiske Model 210 マイクロサンプル・オズモメーター)を用いて測定した。
MPC(日油(株)製)31.8g、ステアリルメタクリレート(SMA、日油(株)製)3.6g及びN,N-ジメチルアクリルアミド(DMAA、興人フィルム&ケミカルズ(株)製)9.6gを、4つ口フラスコへ入れ、エタノール55.0gで溶解させ、30分間窒素ガスの吹き込みを行った。この後、重合開始剤としてt-ブチルペルオキシネオデカノエート(パーブチル(日本登録商標)ND(PB-ND)、日油(株)製)0.10gを加えて8時間重合反応を行った。重合反応後、重合液を3リットルのジエチルエーテル中にかき混ぜながら滴下し、析出した沈殿をろ過し、48時間室温で真空乾燥を行い、粉末を得た。収量は40.2gであった。GPCにより重合物の分子量分析を行い、その重量平均分子量は1,000,000であった。これを重合体1とした。以下に、IR、NMR、元素分析の分析結果を示す。
NMR分析結果:0.8-1.2ppm(CH3-C-)、1.4ppm(-CH2-)、3.3ppm(-N(CH3)3)、2.8-3.2ppm(-N-(CH3)2)、3.7-4.4ppm(-CH2CH2-)。
元素分析結果:
理論値:C;53.55%、H;8.44%、N;8.74%
実測値:C;53.40%、H;8.52%、N;8.80%
以上の結果から、得られた重合体1の化学構造は、MPC 50モル%、DMAA 45モル%、SMA 5モル%の割合で共重合された重合体であった。
下記表1に示す種類及び量の成分を使用した以外は、合成例1と同様の手順に従って重合体を製造した。その重量平均分子量は1,200,000、収量は42.4gであった。以下に、IR、NMR、元素分析の分析結果を示す。
NMR分析結果:0.8-1.2ppm(CH3-C-)、1.4ppm(-CH2-)、3.3ppm(-N(CH3)3)、2.8-3.2ppm(-N-(CH3)2)、3.7-4.4ppm(-CH2CH2-)。
元素分析結果:
理論値:C;56.37%、H;8.70%、N;10.90%
実測値:C;56.41%、H;8.69%、N;10.87%
以上の結果から、得られた重合体1の化学構造は、MPC 30モル%、DMAA 67モル%、SMA 3モル%の割合で共重合された重合体であった。
下記表1に示す種類及び量の成分を使用した以外は、合成例1と同様の手順に従って重合体を製造した。その重量平均分子量は700,000、収量は36.1gであった。以下に、IR、NMR、元素分析の分析結果を示す。
IR分析結果:2964cm-1(-CH)、1733cm-1(O-C=O)、1651cm-1(N-C=O)、1458cm-1(-CH)、1253cm-1(P=O)、1168cm-1(C-O-C)、997cm-1(P-O-C)。
NMR分析結果:0.8-1.2ppm(CH3-C-)、1.4ppm(-CH2-)、3.3ppm(-N(CH3)3)、2.8-3.2ppm(-N-(CH3)2)、3.7-4.4ppm(-CH2CH2-)。
元素分析結果:
理論値:C;50.55%、H;8.15%、N;6.72%
実測値:C;50.46%、H;8.14%、N;6.72%
以上の結果から、得られた重合体1の化学構造は、MPC 70モル%、DMAA 24モル%、SMA 6モル%の割合で共重合された重合体であった。
下記表1に示す種類及び量の成分を使用した以外は、合成例1と同様の手順に従って重合体を製造した。その重量平均分子量は1,000,000、収量は39.9gであった。以下に、IR、NMR、元素分析の分析結果を示す。
NMR分析結果:0.8-1.2ppm(CH3-C-)、1.4ppm(-CH2-)、3.3ppm(-N(CH3)3)、2.8-3.2ppm(-N-(CH3)2)、3.7-4.4ppm(-CH2CH2-)。
元素分析結果:
理論値:C;54.17%、H;8.55%、N;8.03%
実測値:C;54.12%、H;8.54%、N;8.04%
以上の結果から、得られた重合体1の化学構造は、MPC 50モル%、DMAA 40モル%、ラウリルメタクリレート(LMA) 10モル%の割合で共重合された重合体であった。
精製水約80gを45℃に加温し、これにホウ酸0.4g、水酸化ナトリウム0.0072g、塩化ナトリウム0.55g、塩化カリウム0.1g、Cosmocil CQ(日本登録商標(trade mark))0.0004g(20%水溶液であるため、塩酸ポリヘキサニドとしては0.00008gを含む)及び重合体1(0.1g)を順次加え、攪拌した。この溶液を45℃のまま、1時間攪拌混合した。この後、これに全量100mLとなるように精製水を加えた。この後、ろ過滅菌を行い、無菌の点眼剤とした。この点眼剤の浸透圧は281mOsm/kg、pHは7.4、外観は無色澄明であった。その詳細を下記表2に示す。
表2に示す種類及び量の成分を使用した以外は、実施例1と同様の手順に従って製造し、無菌の点眼剤とした。各実施例の外観、pH及び浸透圧を下記表2に示す。
下記表3に示す種類及び量の成分を使用した以外は、実施例と同様の手順に従って製造し、無菌の点眼剤とした。各比較例の外観、pH及び浸透圧を下記表3に示す。
文献「D.R.Korb et al.,2005,Eye&Contact Lens,31,2-8.」によれば、眼瞼結膜上皮障害の治療については、角膜表面の摩擦軽減が有効となる例もあることが考えられるが、角膜表面における摩擦軽減効果の検証は困難であることから、実使用を想定したソフトコンタクトレンズを用いた潤滑性向上効果を以下の手順で検討した。
なお、SCLの潤滑性向上効果の検討には、1Day ACUVUE(日本登録商標(trade mark))(ジョンソンエンドジョンソン(株)製、FDA分類:グループIV)及びメダリストプラス(日本登録商標(trade mark))(ボシュロムジャパン(株)製、FDA分類:グループI)を用いた。
1)実施例1の点眼剤を生理食塩液で10倍希釈した。
2)試験ソフトコンタクトレンズをブリスターパックから1枚取り出し、15mL遠心チューブへ入れた。
3)これに生理食塩液10mLを加え、終夜振とうした。
4)この後、生理食塩液を取り除き、1)で準備した溶液10mLを加え、終夜振とうした。
5)振とう後、ソフトコンタクトレンズを取り出し、人差し指に乗せて潤滑性評価を行った。
メダリストプラス(日本登録商標(trade mark))については、実施例4では評価点数5点となり、実施例1~3及び実施例5及び実施例6について評価点数9点となった。比較例1~比較例5では1Day ACUVUE(日本登録商標(trade mark))と同様に基準となる4点を下回る結果となり、潤滑性を低下させる結果であった。
1Day ACUVUE(日本登録商標(trade mark))とメダリストプラス(日本登録商標(trade mark))とでは、1Day ACUVUE(日本登録商標(trade mark))の方がより潤滑性が向上する結果であった。
以上により、本発明の点眼剤は、優れた潤滑性能を示す。
眼瞼結膜上皮障害の治療効果について検討した。検討に際して、文献「T.Nagano et al.,Atarashii Ganka(Journal of the Eye),13,267-270,1996」、文献「A.Shiraishi et al.,Nippon Ganka Gakkai Zasshi(J. Jpn. Ophthalmol Soc.),113,596-600,2009.」及び文献「D.R.Korb et al.,CLAO J,28,211-216, 2002.」を参考に以下の手順で検討した。
1)ウサギを強制開瞼させた後、ソフトコンタクトレンズ(1Day ACUVUE(ジョンソンエンドジョンソン(株)製)(日本登録商標(trade mark)))を装着させた。
2)ソフトコンタクトレンズを16時間装着させ、眼瞼結膜縁上皮障害を引き起こさせた。
3)この後、ソフトコンタクトレンズを取り外し、メチレンブルーを用いて上眼瞼結膜及び下眼瞼結膜の上皮組織を染色し、眼瞼結膜縁上皮障害の程度を観察した。上皮障害の程度は染色によってスコア化した。
0点:異常所見なし
1点:軽度
2点:中度
3点:重度
試験には、上下の眼瞼の総点が5点以上のウサギを用いた。
4)5点以上のウサギに、実施例1の点眼剤を1回1滴、1日3回点眼し、これを3日間行った。
5)3日間点眼後に再び、メチレンブルーを用いて染色し、眼瞼結膜縁上皮障害の程度を観察した。
なお、スコア化は上眼瞼結膜及び下眼瞼結膜ともに行い、合計点数により評価を行った。実施例2~実施例6及び比較例1~比較例5の点眼剤についても本手順に従って評価を行った。
以上により、本発明の点眼剤及び点眼方法は、優れた眼瞼結膜縁上皮障害治療・予防効果を示す。
Claims (5)
- 前記(1a)が2-(メタクリロイルオキシ)エチル-2’-(トリメチルオンモニオ)エチルホスフェートから得られる構成単位であり、前記(1b)がN,N-ジメチル(メタ)アクリルアミドから得られる構成単位であり、及び前記(1c)がステアリル(メタ)アクリレートから得られる構成単位である請求項1に記載の点眼剤。
- 前記(1a)が2-(メタクリロイルオキシ)エチル-2’-(トリメチルオンモニオ)エチルホスフェートから得られる構成単位である、前記(1b)がN,N-ジメチル(メタ)アクリルアミドから得られる構成単位であり、及び前記(1c)がラウリル(メタ)アクリレートから得られる構成単位である請求項1に記載の点眼剤。
- ソフトコンタクトレンズ装用者用である請求項1~3のいずれか1に記載の点眼剤。
- 眼瞼結膜縁上皮障害の治療用又は予防用である請求項1~4のいずれか1に記載の点眼剤。
