Abstract
Description
アンチジーンは、標的遺伝子の二重鎖DNA部位に対応するオリゴヌクレオチドである。該DNA部位とオリゴヌクレオチドで三重鎖を形成させることにより、DNAからmRNAへの転写を抑制する。
アプタマーは、特定の分子と特異的に結合するDNA、RNAおよび/またはそれらの構造類似体である。標的タンパク質と結合することにより該タンパク質の機能を阻害する。
デコイ核酸は、特定の転写調節因子の結合部位と同じ配列を含む短いDNAである。該転写調節因子と遺伝子の結合を阻害し、該転写調節因子によって活性化される遺伝子群の発現を抑制する。

以下に本明細書中で使用する各用語を説明する。なお、本明細書中、各用語は単独で使用されている場合も、または他の用語と一緒になって使用されている場合も、特に記載の無い限り、同一の意義を有する。
つまり、本発明における「核酸塩基部分」とは、核酸(DNA、RNA)の塩基部分を構成する置換若しくは非置換の複素環式基または置換若しくは非置換の炭素環式基である。

2環以上の非芳香族複素環式基は、単環または2環以上の非芳香族複素環式基に、上記「芳香族炭素環式基」、「非芳香族炭素環式基」、および/または「芳香族複素環式基」におけるそれぞれの環が縮合したものも包含する。
さらに、「非芳香族複素環式基」は、以下のように架橋している基、またはスピロ環を形成する基も包含する。

上記方法で得られた化合物(k-1)をアセトニトリル、1,2-ジクロロエタン、トルエン等の溶媒中、またはそれらの混合溶媒中、トリメチルシリルトリフルオロメタンスルホン酸や四塩化スズ等の酸存在下で、シリル化された核酸塩基部分と25℃~140℃で、1時間~24時間反応させることにより化合物(n)を得ることができる。
工程2 化合物(o)の合成
化合物(n)をメタノール、エタノール、テトラヒドロフラン等の溶媒中、またはそれらの混合溶媒中、パラジウム-炭素粉末や水酸化パラジウム-炭素粉末を加え、水素気流下、0℃~40℃で、1時間~24時間反応させることにより化合物(o)を得ることができる。必要に応じてR5に置換基を導入してもよい。
工程3 化合物(p)の合成
化合物(o)をピリジン中、4,4´‐ジメトキシトリチルクロリド等を加え、0℃~80℃で、1時間~24時間反応させることにより化合物(p)を得ることができる。
工程4 化合物(I‐b)の合成
化合物(p)をアセトニトリル、ジクロロメタン、テトラヒドロフラン等の溶媒中、またはそれらの混合溶媒中、ジイソプロピルエチルアミン、トリエチルアミン等の塩基の存在下で、2-シアノエチル-N,N-ジイソプロピルクロロホスホロアミダイト等あるいはテトラゾール、ジシアノイミダゾール等の酸存在下で2-シアノエチル-N,N,N´,N´-テトライソプロピルホスフォロアミダイト等を加え、0℃~60℃で、1時間~24時間反応させることにより化合物(I‐b)を得ることができる。

本発明のオリゴヌクレオチド中の式(II)で示されるヌクレオシド構造以外の糖部分としては、天然のリボース若しくはデオキシリボース、公知の修飾を有するリボース若しくはデオキシリボース等が挙げられる。公知の修飾としては、例えば、2´-O-CH2-CH2-O-CH3(2´MOE)、4´-CH2-O-2´(LNA、Locked nucleic acid)、AmNA(アミドBNA)(Bridged nucleic acid、WO2011/052436参照)等による修飾が挙げられる。

工程2 化合物3の合成
室温下、2mol/L塩酸水溶液(0.36ml)とアセトニトリル(1.8ml)の混合溶液にN-クロロスクシンイミド(533mg,3.99mmol)を加えたのち氷冷した。反応液に化合物2(457mg,0.997mmol)のアセトニトリル溶液(1.2ml)を加えて氷冷し、30分間撹拌した。反応液へ水を加えて酢酸エチルで抽出した。有機層を飽和食塩水で洗浄後、硫酸ナトリウムで乾燥した。溶媒を減圧留去して、得られた粗製物をシリカゲルカラムクロマトグラフィー(n-ヘキサン:酢酸エチル=90:10→70:30)で精製し、化合物3(372mg,77%)を白色固体物質として得た。

氷冷下、40%メチルアミン水溶液(0.85mL)に化合物3(167mg,0.347mmol)のテトラヒドロフラン溶液(1.7mL)を滴下し30分間撹拌した。溶媒を減圧留去した後、残渣へ水を加えて酢酸エチルで抽出した。有機層を水、飽和食塩水で洗浄後、硫酸ナトリウムで乾燥した。溶媒を減圧留去して、化合物4の粗製物(156mg)を得た。

工程4および工程5 化合物6の合成
室温下、化合物4の粗製物(51mg)を0.1%(v/v)濃硫酸-酢酸溶液(1.5mL)に溶解し、無水酢酸(121μL,1.28mmol)を加えて2時間撹拌した。反応液を飽和重曹水と酢酸エチルの混合液に注いだ後、有機層を分離した。水層を酢酸エチルで抽出し、有機層を水、飽和食塩水で洗浄した。硫酸ナトリウムで乾燥した後、溶媒を減圧留去し、化合物5の粗製物(65mg)を得た。室温下、化合物5の粗製物(64mg)のアセトニトリル溶液(1.0mL)にシリル化チミン(46mg,0.170mmol)、トリメチルシリルトリフルオロメタンスルホン酸(20μL,0.113mmol)を加え、90℃下2時間撹拌した。反応液へ飽和重曹水を加えて酢酸エチルで抽出した。有機層を水、飽和食塩水で洗浄後、硫酸ナトリウムで乾燥して溶媒を減圧留去した。得られた粗製物をシリカゲルカラムクロマトグラフィー(n-ヘキサン:酢酸エチル=60:40→30:70)で精製し、化合物6(50mg,74%(工程3~工程5の通算収率))を白色固体物質として得た。

氷冷下、化合物6(47mg,0.075mmol)のテトラヒドロフラン溶液(0.47mL)に、40%メチルアミン水溶液(0.24mL)を加えて、氷冷下35分間撹拌した。テトラヒドロフランを減圧留去した後、残渣へ水を加えて酢酸エチルで抽出した。有機層を水、飽和食塩水で洗浄後、硫酸ナトリウムで乾燥した。溶媒を減圧留去して、化合物7の粗製物(42mg)を得た。

工程7 化合物8の合成
室温下、化合物7の粗製物(42mg)のピリジン溶液(0.4mL)にメタンスルホニルクロライド(14μL,0.180mmol)を加えて2時間撹拌した。反応液へ水を加えて酢酸エチルで抽出し、有機層を10%クエン酸水溶液、飽和重曹水、水、飽和食塩水で洗浄した。硫酸ナトリウムで乾燥した後、溶媒を減圧留去し、化合物8の粗製物(46mg)を得た。

室温下、化合物8の粗製物(46mg)のテトラヒドロフラン-エタノール溶液(3/2、0.5mL)に2mol/L水酸化ナトリウム水溶液(0.15mL,0.300mmol)を加えて20時間撹拌した。氷冷下、反応液へ2mol/L塩酸水溶液を加えて中和した後、溶媒を減圧留去した。残渣へ水を加えて酢酸エチルで抽出し、有機層を水、飽和食塩水で洗浄後、硫酸ナトリウムで乾燥した。溶媒を減圧留去して得られた粗製物をシリカゲルカラムクロマトグラフィー(n-ヘキサン:酢酸エチル=50:50→20:80)で精製し、化合物9(28mg,68%(工程6~工程8の通算収率))を白色固体物質として得た。

工程9 化合物10の合成
窒素気流下、化合物9(29mg,0.054mmol)のピリジン溶液(0.3mL)に、氷冷下、トリフルオロメタンスルホン酸無水物(27μL,0.161mmol)を加えて5時間撹拌した。さらに室温下、2時間撹拌した後、反応液へ水を加えて酢酸エチルで抽出した。有機層を水、飽和食塩水で洗浄後、硫酸ナトリウムで乾燥した。溶媒を減圧留去して、化合物10の粗製物(39mg)を得た。

窒素気流下、化合物10の粗製物(39mg)のアセトニトリル溶液(0.7mL)に室温下、炭酸カリウム(19mg,0.135mmol)を加えて、40℃下5時間半撹拌した。反応液へ水を加えて酢酸エチルで抽出し、有機層を飽和食塩水で洗浄した。硫酸ナトリウムで乾燥した後、溶媒を減圧留去し、得られた粗製物をシリカゲルカラムクロマトグラフィー(n-ヘキサン:酢酸エチル=70:30→40:60)で精製し、化合物11(24mg,86%(工程9~工程10の通算収率))を白色固体物質として得た。

工程11 化合物12の合成
化合物11(21mg,0.040mmol)のテトラヒドロフラン-メタノール溶液(1:1,0.6mL)に20%水酸化パラジウム-炭素粉末(5.3mg)を加え、水素気流下室温で20時間撹拌した。反応溶液をろ過した後、溶媒を留去して化合物12(17mg)を白色固体物質として得た。

窒素気流下、化合物12(31mg,0.090mmol)のピリジン溶液(0.6mL)に、室温下、4,4´‐ジメトキシトリチルクロリド(46mg,0.135mmol)を加えて15時間半撹拌した。室温下、4,4´‐ジメトキシトリチルクロリド(46mg,0.135mmol)を加えて7時間半撹拌した。さらに室温下、4,4´‐ジメトキシトリチルクロリド(46mg,0.135mmol)を加えて14時間半撹拌した後、反応液へ飽和重曹水を加えて酢酸エチルで抽出した。有機層を水、飽和食塩水で洗浄後、硫酸ナトリウムで乾燥した。溶媒を減圧留去して、得られた粗製物をシリカゲルカラムクロマトグラフィー(クロロホルム:メタノール=100:0→95:5)で精製し、化合物13(59mg)を白色固体物質として得た。

工程13 化合物I-1の合成
窒素気流下、化合物13(59mg,0.091mmol)の無水ジクロロメタン溶液(0.9mL)にジイソプロピルエチルアミン(63μL,0.363mmol)、2-シアノエチル-N,N-ジイソプロピルクロロホスホロアミダイト(61μL,0.272mmol)を加え、3時間撹拌した。反応液へ飽和重曹水を加えて酢酸エチルで抽出した。有機層を水、飽和食塩水で洗浄した後に、無水硫酸ナトリウムで乾燥させた。溶媒を留去して得られた粗製物をシリカゲルカラムクロマトグラフィー(n-ヘキサン:酢酸エチル=34:66)で精製し、化合物I-1(47mg,61%)を白色固体物質として得た。

窒素気流下、実施例1-1で合成した化合物13(191mg,0.294mmol)のN,N-ジメチルホルムアミド溶液(1.0mL)に、室温下、イミダゾール(160mg,2.35mmol)とクロロトリエチルシラン(199μL,1.18mmol)を加えて3時間撹拌した。反応液へ水を加えて酢酸エチルで抽出した。有機層を水、飽和食塩水で洗浄後、硫酸ナトリウムで乾燥した。溶媒を減圧留去して、得られた粗製物をシリカゲルカラムクロマトグラフィー(n-ヘキサン:酢酸エチル=75:25→50:50)で精製し、化合物14(214mg,95%)を白色固体物質として得た。

窒素気流下、化合物14(206mg,0.269mmol)のアセトニトリル溶液(2.0mL)に、室温下、トリエチルアミン(149μL,1.08mmol)、N,N-ジメチルアミノピリジン(6.6mg,0.054mmol)と2,4,6-トリイソプロピルベンゼンスルホニルクロライド(122mg,0.403mmol)を加えて20時間撹拌した。室温下、反応液へ28%アンモニア水(2.0mL)を加えて2時間撹拌した。溶媒を減圧留去し、得られた残渣へ水を加えて酢酸エチルで抽出した。有機層を水、飽和食塩水で洗浄後、硫酸ナトリウムで乾燥した。溶媒を減圧留去して、得られた粗製物をシリカゲルカラムクロマトグラフィー(クロロホルム:メタノール=100:0→95:5)で精製し、化合物15(180mg,88%)を白色固体物質として得た。

氷冷下、フェノキシアセチルクロライド(38μL,0.276mmol)のジクロロメタン溶液(2.1mL)に1-メチルイミダゾール(25μL,0.313mmol)を加えて10分間撹拌した。得られた懸濁液を化合物15のピリジン溶液(1.4mL)へ加えて1時間半撹拌した。反応液へ飽和重曹水を加えて酢酸エチルで抽出した。有機層を水、飽和食塩水で洗浄後、硫酸ナトリウムで乾燥した。溶媒を減圧留去して、化合物16の粗製物(171mg)を得た。室温下、化合物16の粗製物のテトラヒドロフラン溶液(2.1mL)に1Mフッ化テトラ-n-ブチルアンモニウムテトラヒドロフラン溶液を(221μL,0.221mmol)を加えて15分間撹拌した。反応液に酢酸エチルを加えた後、溶媒を留去した。得られた粗製物をシリカゲルカラムクロマトグラフィー(n-ヘキサン:酢酸エチル=60:40→15:85)で精製し、化合物17(77mg,53%)を白色固体物質として得た。

窒素気流下、化合物17(100mg,0.127mmol)の無水ジクロロメタン溶液(1.5mL)にジイソプロピルエチルアミン(89μL,0.509mmol)、2-シアノエチル-N,N-ジイソプロピルクロロホスホロアミダイト(85μL,0.382mmol)を加え、2時間半撹拌した。反応液へ飽和重曹水を加えて酢酸エチルで抽出した。有機層を水、飽和食塩水で洗浄した後に、無水硫酸ナトリウムで乾燥させた。溶媒を留去して得られた粗製物をシリカゲルカラムクロマトグラフィー(n-ヘキサン:酢酸エチル=60:40→35:65)で精製し、化合物I-2(69mg,55%)を白色固体物質として得た。

窒素気流下、化合物3(16.8g,34.8mmol)のテトラヒドロフラン溶液(120mL)に氷冷下、トリエチルアミン(12.1mL,87.0mmol)、2-ナフチルメチルアミン塩酸塩(J. Carbohydr. Chem. 30, 559-574 (2011))(7.41g,38.3mmol)を加え、1時間撹拌した。溶媒を減圧留去した後、残渣へ水を加えて酢酸エチルで抽出した。有機層を水、飽和食塩水で洗浄後、硫酸ナトリウムで乾燥した。得られた粗製物をシリカゲルカラムクロマトグラフィー(n-ヘキサン:酢酸エチル=90:10→60:40)で精製し、化合物18(16.6g,79%)を淡黄色泡状物質として得た。

化合物18(16.6g,27.4mmol)の酢酸溶液(80mL)に室温下、濃硫酸(80μL,1.44mmol)、無水酢酸(31.1mL,329mmol)を加えて4時間撹拌した。反応液を飽和重曹水に注いだ後、酢酸エチルで抽出した。有機層を水、飽和食塩水で洗浄した後、硫酸マグネシウムで乾燥した。溶媒を減圧留去し、化合物19および20の混合物(18.3g)を得た。窒素気流下、化合物19および20の混合物(18.3g)のアセトニトリル溶液(150mL)に室温下、チミン(5.18g,41.1mmol)、N,O-ビス(トリメチルシリル)アセトアミド(23.5mL,96.0mmol)を加え、40℃下20分間撹拌した。室温下、トリメチルシリルトリフルオロメタンスルホン酸(4.95mL,27.4mmol)を加え、5時間加熱還流した。反応液へ飽和重曹水を加えて酢酸エチルで抽出した。有機層を水、飽和食塩水で洗浄後、硫酸マグネシウムで乾燥して溶媒を減圧留去した。得られた粗製物をシリカゲルカラムクロマトグラフィー(n-ヘキサン:酢酸エチル=70:30→45:55)で精製し、化合物21および22の混合物(18.6g)を白色固体物質として得た。
化合物21および22の混合物(18.6g)のテトラヒドロフラン溶液(90mL)に氷冷下、40%メチルアミン水溶液(45mL)を加えて、40分間撹拌した。テトラヒドロフランを減圧留去した後、残渣へ水を加えて酢酸エチル/メタノール(5/1)で抽出した。有機層を水、飽和食塩水で洗浄後、硫酸マグネシウムで乾燥した。溶媒を減圧留去して、化合物23の粗製物(16.7g)を得た。

化合物24の粗製物(18.7g)のテトラヒドロフラン-エタノール溶液(3/2、150mL)に室温下、2mol/L水酸化ナトリウム水溶液(61.5mL,123mmol)を加えて22時間撹拌した。氷冷下、反応液へ濃塩酸(10.2mL,123mmol)を加えて中和した後、飽和重曹水を加えてテトラヒドロフラン、エタノールを減圧留去した。残渣へ水を加えて酢酸エチルで抽出し、有機層を水、飽和食塩水で洗浄後、硫酸マグネシウムで乾燥した。溶媒を減圧留去して得られた粗製物をシリカゲルカラムクロマトグラフィー(n-ヘキサン:酢酸エチル=60:40→30:70)で精製し、化合物25(11.9g,64%(工程2~工程6の通算収率))を白色固体物質として得た。

窒素気流下、化合物25(11.9g,17.7mmol)のピリジン溶液(60mL)に、氷冷下、トリフルオロメタンスルホン酸無水物(8.95mL,53.0mmol)を加えて15時間撹拌した。反応液へ水を加えて酢酸エチルで抽出した。有機層を水、飽和食塩水で洗浄後、硫酸マグネシウムで乾燥した。溶媒を減圧留去して、化合物26の粗製物(22.6g)を得た。窒素気流下、化合物26の粗製物(22.6g)のアセトニトリル溶液(220mL)に室温下、炭酸カリウム(6.10g,44.2mmol)を加えて、40℃下4時間半撹拌した。反応液へ水を加えて、アセトニトリルを減圧留去した。残渣を酢酸エチルで抽出し、有機層を水、飽和食塩水で洗浄した。硫酸マグネシウムで乾燥した後、溶媒を減圧留去し、得られた粗製物をシリカゲルカラムクロマトグラフィー(n-ヘキサン:酢酸エチル=65:35→50:50)で精製し、化合物27(8.73g,76%)を白色固体物質として得た。

窒素気流下、化合物27(4.00g,6.12mmol)のジクロロメタン溶液(40mL)に-78℃下、三塩化ホウ素(1mol/Lジクロロメタン溶液、61.2mL,61.2mmol)を滴下し、徐々に室温まで昇温して40時間撹拌した。-78℃下、反応液にメタノール(40mL)を滴下し、室温下、20分攪拌した。溶媒を減圧留去し、得られた粗製物をシリカゲルカラムクロマトグラフィー(クロロホルム:メタノール=96:4→85:15)で精製し、化合物28(1.59g,78%)を白色固体物質として得た。

窒素気流下、化合物28(1.76g,5.28mmol)のピリジン溶液(18mL)に、室温下、4,4´‐ジメトキシトリチルクロリド(3.04g,8.98mmol)を加えて15時間撹拌した。溶媒を減圧留去した後、残渣へ飽和重曹水を加えて酢酸エチルで抽出した。有機層を水、飽和食塩水で洗浄後、硫酸ナトリウムで乾燥した。溶媒を減圧留去して、得られた粗製物をシリカゲルカラムクロマトグラフィー(クロロホルム:メタノール=100:0→94:6)で精製し、化合物29(3.30g,98%)を白色固体物質として得た。

窒素気流下、化合物29(21mg,0.033mmol)のアセトニトリル溶液(0.4mL)に室温下、2-シアノエチル-N,N,N’,N’-テトライソプロピルホスホロジアミダイト(21μL,0.065mmol)、5-エチルチオ-1H-テトラゾール(6.4mg,0.049mmol)を加え、7時間撹拌した。反応液へ飽和重曹水を加えて酢酸エチルで抽出した。有機層を水、飽和食塩水で洗浄した後に、無水硫酸ナトリウムで乾燥させた。溶媒を留去して得られた粗製物をシリカゲルカラムクロマトグラフィー(n-ヘキサン:酢酸エチル=50:50→25:75)で精製し、化合物III-4(18mg,65%)を白色固体物質として得た。

実施例1で得られた化合物I-1から調製されるオリゴヌクレオチド(1)~(5)(表4)およびIII-4から調製されるオリゴヌクレオチド(6)及び(7)(表5)を、nS-8(ジーンデザイン社製)により0.2μmolスケールで合成した。化合物I-1およびIII-4(アミダイトユニット)は、アセトニトリルに溶解して用いた。なお、表4において、Xaは化合物I-1のヌクレオシド構造(下記式II-1)を意味する。表5においてXbは化合物III-4のヌクレオシド構造(下記式II-2)を意味する。アミダイトユニット(化合物I-1、III-4)と5´-末端の水酸基とのカップリング時間は32秒(標準)から16分に延長した。5´-末端がDMTr基により保護され、かつ固相に支持されたオリゴヌクレオチドを28%アンモニア水:40%メチルアミン水溶液(1:1)で処理し、溶媒を留去した。得られた粗製物をSep-Pak C18 Plus Short Cartridge(Waters社製)により粗精製した後、逆相HPLC(オリゴヌクレオチド(1)~(5)に関してはGilson PLC2020、分取カラムとしてWatersXBridgeTM Shield RP18 Column 5.0 μm (10mm x 50mm)、オリゴヌクレオチド(6)及び(7)に関してはYMC Hydrosphere C18 Column 5.0 μm (10mm x 150mm))により精製した。
合成したオリゴヌクレオチドの純度は、逆相HPLC(オリゴヌクレオチド(1)~(5)に関してはWatersXBridgeTM C18 Column 5.0μm(4.6mm x 50mm)、オリゴヌクレオチド(6)及び(7)に関してはYMC Hydrosphere C18 Column 5.0μm (4.6mm x 100mm))により確認した(オリゴヌクレオチド(1)~(5)に関してはWatersXBridgeTM C18 Column条件:0.1Mトリエチルアンモニウム酢酸バッファー(pH7.0)中、1mL/分で30分の6→10%(v/v)アセトニトリルのグラジエント、オリゴヌクレオチド(6)及び(7)に関してはYMC Hydrosphere C18 Column条件:0.01Mトリエチルアンモニウム酢酸バッファー(pH7.0)中、1mL/分で30分の6→10%(v/v)アセトニトリルのグラジエント)。また、分子量は、MALDI-TOF-MASS測定(オリゴヌクレオチド(1)~(5))またはESI-TOF-MASS測定(オリゴヌクレオチド(6)及び(7))により決定した。結果を表4および表5に示す。
実施例2で合成したオリゴヌクレオチド(1)~(4)および(6)(アンチセンス鎖)とセンス鎖(3´-CGC AAA AAA CGA-5´)とをアニーリング処理した後、Tm値を測定することにより、オリゴヌクレオチド(1)~(4)および(6)のハイブリッド形成能を調べた。オリゴヌクレオチドのヌクレオシド部分が非修飾であるヌクレオチド(0)をコントロールとして利用した。
塩化ナトリウム100mM、リン酸ナトリウム緩衝液(pH7.2)10mM、オリゴヌクレオチド(アンチセンス鎖)4.0μM、およびセンス鎖4.0μMを含むサンプル溶液(150μM)をヒートブロック(95℃)で5分間、加熱した後、12時間かけて室温まで冷却した。分光高度計(SHIMADZU UV-1800)のセル室内に結露防止のために窒素気流を通し、サンプル溶液を15℃まで徐々に冷却し、さらに15分間15℃に保った後、測定を開始した。温度は90℃まで毎分0.5℃ずつ上昇させ、0.5℃間隔で260nmにおける紫外部吸収を測定した。なお、温度上昇による濃度変化を防止するため、セルは蓋付きのものを用いた。結果を表6に示す。
表6から明らかなように、本発明のオリゴヌクレオチドは、RNA相補鎖に対して、天然オリゴヌクレオチドと比較してTm値を上昇させた。一方で、DNA相補鎖に対しては、天然オリゴヌクレオチドと比較してTm値を低下させた。また、本発明のヌクレオシド構造(II-1)の割合が高いほどTm値の上昇が見られた。従って、本発明のヌクレオチドから調製されるオリゴヌクレオチドは、一本鎖RNAに対する親和性が高いので、mRNAに作用しやすい。また、一本鎖DNAに対する親和性が低いのでDNA複製への影響は小さく、毒性の懸念が低いと考えられるため、核酸医薬品を合成するための材料として有用であると考えられる。
実施例2で合成したオリゴヌクレオチド(5)(Sulfonamide-NMe)及びオリゴヌクレオチド(7)(Sulfonamide-NH)について、オリゴヌクレオチドを3´側から分解するエキソヌクレアーゼに対する耐性を検討した。オリゴヌクレオチド(5)のXa部分が非修飾であるヌクレオチド(nature)、オリゴヌクレオチド(5)のXa部分が下記2´,4´‐BNA/LNAであるヌクレオチド(LNA)、オリゴヌクレオチド(5)のXa部分が下記2´,4´‐BNA-amideであるヌクレオチド(AmNA(Amide-BNA))および、オリゴヌクレオチド(5)のXa部分が下記2´,4´‐BNANC-Meであるヌクレオチド(BNANC-Me)をコントロールとして利用した。
移動相 A液: 0.1Mトリエチルアンモニウム酢酸バッファー(pH7.0)
B液: 0.1Mトリエチルアンモニウム酢酸バッファー:アセトニトリル =1:1(v/v)(pH7.0)
グラジエント
オリゴヌクレオチド(5)及び(7):B液濃度 16%-20%(16min)
その他のオリゴヌクレオチド:B液濃度 14%-22%(16min)
カラム WatersXBridgeTM Shield RP18 Column 2.5μm(3.0mm x 50mm)
流速 0.8mL/min
カラム温度 50℃
検出 UV(254nm)
この結果、ヌクレオチド(nature)およびヌクレオチド(LNA)は10分後にはすべて分解している。ヌクレオチド(AmNA(Amide-BNA))は40分後の残存率は50%以下であり、ヌクレオチド(BNANC-Me)は40分後の残存率は70%以下であるが、オリゴヌクレオチド(5)(Sulfonamide-NMe)は40分後においても残存率が80%以上である。また、オリゴヌクレオチド(7)(Sulfonamide-NH)は40分後においても残存率が95%以上である。よって、本発明のスルホンアミド構造を有するオリゴヌクレオチドは、非修飾のオリゴヌクレオチドや公知の人工ヌクレオチドから調製されるオリゴヌクレオチド(LNA、AmNA(Amide-BNA)、BNANC-Me)を大きく上回る酵素耐性を有することが分かった。従って、本発明のオリゴヌクレオチドは体内での持続性が非常によいと考えられることから、核酸医薬品を合成するための材料として有用であると考えられる。
Claims (8)
- 式(I):
Y1-Y2はS(=O)-NR6、S(=O)2-NR6、NR6-S(=O)またはNR6-S(=O)2であり、
R6は水素原子、置換若しくは非置換のアルキル、置換若しくは非置換のアルケニルまたは置換若しくは非置換のアルキニルであり、
Bxは核酸塩基部分であり、
Z1およびZ2はそれぞれ独立して、水素原子、水酸基保護基または反応性リン基であり、
R1およびR2はそれぞれ独立して、水素原子、ハロゲン、シアノ、置換若しくは非置換のアルキル、置換若しくは非置換のアルケニルまたは置換若しくは非置換のアルキニルであり、
R3はそれぞれ独立して、水素原子、ハロゲン、シアノ、置換若しくは非置換のアルキル、置換若しくは非置換のアルケニルまたは置換若しくは非置換のアルキニルであり、
R4はそれぞれ独立して、水素原子、ハロゲン、シアノ、置換若しくは非置換のアルキル、置換若しくは非置換のアルケニルまたは置換若しくは非置換のアルキニルであり、
R5は水素原子、ハロゲン、シアノ、置換若しくは非置換のアルキル、置換若しくは非置換のアルケニルまたは置換若しくは非置換のアルキニルであり、
nは0~3の整数である)
で示される化合物、またはその塩。 - Y1-Y2がS(=O)2-NR6であり、R6が水素原子またはアルキルである、請求項1記載の化合物、またはその塩。
- Bxが置換若しくは非置換のプリン-9-イルまたは置換若しくは非置換の2-オキソ-ピリミジン-1-イルである、請求項1または2記載の化合物、またはその塩。
- Z1が水素原子または水酸基保護基である、請求項1~3いずれかに記載の化合物、またはその塩。
- 該水酸基保護基がアセチル、t-ブチル、t-ブトキシメチル、メトキシメチル、テトラヒドロピラニル、1-エトキシエチル、1-(2-クロロエトキシ)エチル、2-トリメチルシリルエチル、p-クロロフェニル、2,4-ジニトロフェニル、ベンジル、ベンゾイル、p-フェニルベンゾイル、2,6-ジクロロベンジル、レブリノイル、ジフェニルメチル、p-ニトロベンジル、トリメチルシリル、トリエチルシリル、t-ブチルジメチルシリル、t-ブチルジフェニルシリル、トリフェニルシリル、トリイソプロピルシリル、ギ酸ベンゾイル、クロロアセチル、トリクロロアセチル、トリフルオロアセチル、ピバロイル、イソブチリル、9-フルオレニルメチルオキシカルボニル、メタンスルホニル、p-トルエンスルホニル、トリフルオロメタンスルホニル、トリチル、モノメトキシトリチル、ジメトキシトリチル、トリメトキシトリチル、9-フェニルキサンチン-9-イルまたは9-(p-メトキシフェニル)キサンチン-9-イルである、請求項4記載の化合物、またはその塩。
- Z2が水素原子または反応性リン基である、請求項1~5いずれかに記載の化合物、またはその塩。
- 該反応性リン基がジイソプロピルシアノエトキシホスホロアミダイトまたはH-ホスホネートである、請求項6記載の化合物、またはその塩。
- 式(II):
Y1-Y2はS(=O)-NR6、S(=O)2-NR6、NR6-S(=O)またはNR6-S(=O)2であり、
R6は水素原子、置換若しくは非置換のアルキル、置換若しくは非置換のアルケニルまたは置換若しくは非置換のアルキニルであり、
Bxは核酸塩基部分であり、
R1およびR2はそれぞれ独立して、水素原子、ハロゲン、シアノ、置換若しくは非置換のアルキル、置換若しくは非置換のアルケニルまたは置換若しくは非置換のアルキニルであり、
R3はそれぞれ独立して、水素原子、ハロゲン、シアノ、置換若しくは非置換のアルキル、置換若しくは非置換のアルケニルまたは置換若しくは非置換のアルキニルであり、
R4はそれぞれ独立して、水素原子、ハロゲン、シアノ、置換若しくは非置換のアルキル、置換若しくは非置換のアルケニルまたは置換若しくは非置換のアルキニルであり、
