Abstract
Description
(a):化合物(1)の無水度:上記スキームで化合物(1)からシュウ酸ジアミド誘導体である化合物(5)を製造する工程は、無水条件で実施することが、生成物である化合物(5)を高収率で得るために重要であり、そのため化合物(1)が、付着水として水分を含むものや、水和物結晶を使用した場合には、化合物(5)の収率が大きく低下するため、化合物(1)の無水度を維持することが課題である。
(b):cis-ジアミノ誘導体である化合物(1)の純度:化合物(2)から、アジド体である化合物(3)の製造では、目的のcis-体である化合物(3)の他に、類縁物質として異性体であるtrans-体の化合物(3-trans)が生成する(国際公開第2001/74774号パンフレット参照)。したがって、粗アジド体を還元して得られるアミノ体(1a)にもアジド化工程で生成したtrans異性体を同じ割合で含むことから、trans異性体の化合物(1a-trans)の除去が課題である。
(c):反応収率、操作性など、工業的スケールで実施可能な製造法の提供が課題である。
(d):国際公開2007/032498号パンフレットにおける化合物(1)の製造法は、無水シュウ酸溶液を添加するときに滴下近傍で非晶質の析出が観察されることから、非晶質を経由して結晶化が徐々に進行することが明らかになり、結晶化に長時間を要し、また非晶質が生成したときの撹拌が困難になる。
(e):無水シュウ酸溶液の滴下時間によって多形結晶が析出することが明らかになり、製造工程の安定性がなく、また多形結晶があることから単一結晶形での結晶化の完結に長時間を要する。
(f):(e)において、結晶化が完結しない場合は非晶質を含むため、ろ過操作が不可能になる程度に操作性が低下する。
(g):長時間撹拌することにより、撹拌溶液中に存在する類縁物質であるtrans-異性体の化合物(1-trans)の結晶が、過飽和状態により析出が開始することがわかり、化合物(1-trans)の混在によるcis-ジアミノ誘導体である化合物(1)の純度が低下する。
(h):trans異性体である化合物(1-trans)の混入は、撹拌時間や温度のコントロール、また結晶化溶媒の変更や溶媒量の増量である程度は防止が可能であるが、溶媒量の増加は生産装置の大型化を伴って好ましくなく、また撹拌時間のさらなる長時間化による化合物(1-trans)の再混入現象が観察され、堅牢な工業的製造方法として問題点が潜在していることが明らかになった。

で表される化合物の1水和物結晶を、加温下、1重量%未満の水分含量の有機溶媒中で攪拌する工程、を含む、式(1)で表される化合物の無水物結晶の製造方法。

で表される化合物を、含水有機溶媒中で無水シュウ酸と処理して、下記式(1b)

(5):さらに、撹拌工程において、有機溶媒全体に対して1/2~4/7容量の有機溶媒を40~75℃の範囲で減圧留去し、その後、留去した量に相当する量の有機溶媒を再添加することを含む、(1)~(4)のいずれか1に記載の製造方法。
(6):有機溶媒の減圧留去及び再添加において、有機溶媒中の水分含量を0.2重量%未満にすることを特徴とする、(5)に記載の製造方法。

ここで、上記の製造方法では、アジド化の工程でトランス異性体である化合物(3-trans)が副生するため、いずれかの工程で精製操作を行わない限り、化合物(3)、化合物(1a)及びそのシュウ酸塩である化合物(1)には、下記の類縁物質であるtrans-異性体の化合物(3-trans)、化合物(1a-trans)、及び化合物(1-trans)が副生物として混在することが知られている(国際公開第2001/74774号パンフレット参照)。FXa阻害薬である式(X)で表される化合物、その薬理上許容される塩又はそれらの水和物の製造において、製造中間体の類縁物質の分離除去は最終物である医薬品化合物の品質向上に重要であり、また分離除去における効率性は原薬製造の収率の向上に非常に大切である。
国際公開第2001/74774号パンフレットにおいて、メシルオキシ体である化合物(2)のアジド化によるアジド体の化合物(3)の製造では、目的のcis-体である化合物(3)の他に、類縁物質としてtrans-体の異性体である化合物(3-trans)が生成することが開示されている。また、アジド化工程の終了後には、化合物(3)の精製操作を行わないので、化合物(3)を還元して得られるアミノ体(1a)にも、trans-異性体である化合物(1a-trans)が類縁物質として含まれている。
(A-1):化合物(1a)に、有機溶媒、及び4容量%以上の含水有機溶媒となるように水を添加して、懸濁又はスラリーとする。
(A-2):撹拌しながら内温を50~70℃に加温する。
(A-3):シュウ酸を有機溶媒に溶解した溶液を調製し、上記(A-2)の溶液中に、内温50~70℃の範囲で、1~2時間をかけて滴下して懸濁液又はスラリーとする。
(A-4):滴下終了後、反応混合液を50~70℃の範囲で、2~5時間撹拌する。
(A-5):反応混合液を攪拌下に20~40℃に冷却する。
(A-6):析出する結晶をろ取し、有機溶媒で結晶を洗浄後、乾燥する。
(B-1):第1の工程である(A工程)で製造した式(1b)で表される化合物の1水和物結晶に、溶媒を添加し、溶媒中の水分含量を1%(1重量%)未満になるように、溶媒の無水度、反応系内に水分が入り込まないように、懸濁又はスラリーとする。
(B-2):内温を60℃~80℃に加温して、溶媒中の水分含量が1重量%未満であることを確認して撹拌する。
(B-3):内温を40℃以上、外温80℃以下に加温して、減圧下に溶媒を留去して、反応溶媒中の水分を共沸脱水して反応混合液の溶媒中の水分含量を0.2重量%未満にするために、使用溶媒の総容量の半量以上を留去する。
(B-4):留去した量と同量の溶媒を添加し、反応混合液の溶媒中の水分含量を0.2重量%未満にであることを確認後、混合液の内温を40~75℃で添加して、撹拌する。水分含量を0.2重量%未満とするためには、添加する溶媒の無水度、外部の湿度を反応系内に取り込まないようにする。
(B-5):混合液の水分含量を0.2重量%未満で、20~40℃に冷却する。冷却中に外部湿度を取り込まないように、反応を湿度(水分)の低い不活性ガスの環境下で実施してもよい。
(B-6):析出した結晶を集め、用いた溶媒で結晶を洗浄した後、乾燥する。
有機溶媒としては、単独又は混合溶媒でもちればよく、例えば、酢酸C1~C5アルキルエステル系溶媒、直鎖状又は分枝鎖状のC1~C8アルコール系溶媒、C1~C6ケトン系溶媒、トルエン系溶媒、及びC2~C5ニトリル系溶媒から選ばれる1種又は2種以上の混合溶媒を挙げることができ、これらの溶媒の例示としては、上述と同様である。これらの溶媒は、単独溶媒として、又は2種以上を混合有機溶媒として使用してもよく、本操作において低水分含量が必要であるため、市販の無水の有機溶媒を用いればよい。これらの有機溶媒中、アセトニトリル、トルエン、及びアセトニトリルとトルエンの混合溶媒が好ましく、アセトニトリルが特に好ましい。使用する溶媒量は、化合物(2)に対して5~30倍容量部(V/W)の範囲が好ましく;7~10倍容量部(V/W)の範囲がより好ましい。
ここで、溶媒中の水分含量を1%(1重量%)未満の確認方法としては、溶媒中の水分含量を測定する方法として公知の、例えば、カール・フィッシャー法等で実施でき、例えば、カール・フィッシャー水分計等の市販の測定装置を用いて実施すればよい。
(B-3)の操作は、溶媒を濃縮留去する。濃縮留去するときの温度としては、好ましくは内温40~75℃、外温80℃以下;より好ましくは内温45~60℃、外温80℃以下;である。濃縮留去としては、減圧又は常圧で実施できるが、減圧下で行うのが好ましい。濃縮留去する量は、総量の半量以下にするのが好ましく、具体的には、(B-1)の操作で添加した量の半量以上を濃縮留去するのが好ましい。この溶媒の濃縮留去操作は、系内の水分含量を有機溶媒との共沸留去によって混合液系内の水分量を減らし、無水物結晶から、再び1水和物結晶に再度水和物化するのを防止する。混合液の溶媒中の水分含量としては、0.2重量%未満が好ましく;0.15重量%以下がより好ましい。
(B-5)の操作は、(B-4)の操作で溶媒を添加後、撹拌した混合液を冷却する。冷却は反応装置の外部を、水などで冷却するか、又は自然放冷してもよく、冷却する温度としては、内温を20~40℃の範囲になるように冷却すればよい。上記(B-4)の操作及び(B-5)の冷却操作では、混合液の溶媒中の水分含量を、0.2重量%未満、より好ましくは0.15重量%以下に保持する。水分含量を0.2重量%未満に保持するためには上述のように、反応操作を不活性ガスの環境下で行う等の方策を採ればよい。このように、溶媒中の水分含量を、0.2重量%未満、より好ましくは0.15重量%以下に保持することにより、式(1)で表される化合物の無水物結晶が再水和化して、式(1b)で表される化合物の1水和物結晶へ転移するのを防止する。したがって、(B-5)の操作における撹拌時間は特に問題ではなく、製造スケジュールに合致した時間で撹拌すればよい。
(B-6)の操作は、析出結晶をろ過にて集め、結晶を洗浄後、乾燥する。洗浄に用いる溶媒は、上記で用いた溶媒でよく;アセトニトリルが好ましい。洗浄に使用する溶媒量は化合物(2)に対して1倍容量部(V/W)程度が好ましい。乾燥は、常圧又は減圧でよく、温度は40℃程度が好ましい。ここで、乾燥後、目的の式(1)で表される化合物の無水物結晶を、化合物(2)からの通算収率として、60%以上で得ることができる。

[アジド化工程]:化合物(2)を、溶媒中でアジ化試薬と処理して化合物(3)を製造する。
4級アンモニウム塩及びアジ化金属塩を水に添加して4級アンモニウム塩-アジ化金属塩からなるアジ化試薬複合体の水溶液を調製し、引き続いて該水溶液を、芳香族炭化水素系溶媒を用いて脱水処理を行って、水分含量が0.2重量%以下の4級アンモニウム塩-アジ化金属塩からなるアジ化試薬複合体の芳香族炭化水素系溶媒混合液とし、次いで化合物(2)と処理して化合物(3)を製造する。
4級アンモニウム塩としては、アルキルアミンの4級アンモニウム塩、及びピリジニウム塩が好ましく;具体的には塩化アンモニウム、1-ドデシルピリジニウム クロリド(別名:1-ラウリルピリジニウム クロリド)などが特に好ましい。アジ化金属塩としては、アジ化アルカリ金属塩が好ましく;アジ化ナトリウム及びアジ化リチウムがより好ましく;アジ化ナトリウムが特に好ましい。
4級アンモニウム塩の使用量としては、化合物(2)に対して0.5モル当量程度が好ましく、アジ化金属塩の使用量としては、化合物(2)に対して2.0モル当量程度が好ましいが、なんらこの範囲に限定されるものではない。
4級アンモニウム塩及びアジ化金属塩から、アジ化試薬複合体を調製するときの水の使用量としては、化合物(2)の1重量部に対して1~2容量部[1.0(v/w)]程度が好ましいが、なんらこの範囲に限定されるものではない。水は、次の共沸脱水操作で除去するため、少ない量が好ましい。
アジ化試薬複合体を調製する温度としては、室温でよく、20~40℃の範囲が好ましい。脱水処理としては、水を共沸させる有機溶媒を用いた共沸脱水処理を意味し、芳香族炭化水素系溶媒を用いた共沸脱水が好ましい。芳香族炭化水素系溶媒としては、ベンゼン、トルエン、キシレン、クロルベンゼン及びジクロルベンゼンが好ましく、これら溶媒を単独(1種類)で、又は2種以上を混合した混合溶媒として用いてもよい。芳香族炭化水素系溶媒としては、トルエンがより好ましい。共沸脱水処理によって、水分含量を0.2重量%未満にするのが好ましく;0.1重量%以下にするのがより好ましく;0.1重量%以下が特に好ましい。
(A工程)及び(B工程):上述の(A工程)及び(B工程)と同様に行う。
本願発明で製造した高純度の式(1)で表される無水物結晶を用いた、FXa阻害薬である化合物(X)又は化合物(X-a)の製造は、以下に示すとおりであり、高純度の式(1)で表される化合物の無水物結晶を用いたことにより、高純度の化合物(X)又は化合物(X-a)が製造できる。
化合物(4)は、下記に示すように製造することができ、製造の具体例としては、例えば参考例4に記載された方法を挙げることができる。すなわち、化合物(4)は、市販であるアニリン誘導体の化合物(4b)をC2-C4ニトリル系溶媒中で攪拌下に、化合物(4c)を添加して製造することができる。
本願発明の特徴である高純度の化合物(1)の化合物の無水物結晶を使用することにより、高純度のFXa阻害薬の化合物(X)、及び化合物(X)のモノp-トルエンスルホン酸塩・1水和物である化合物(X-a)を製造することができる。
[工程1] tert-ブチル{(1R,2R,5S)-5-[(ジメチルアミノ)カルボニル]-2-ヒドロキシシクロヘキシルカルボニル}カルバマート(11)の合成
(1S,3S,6R)-N,N-ジメチル-7-オクサビシクロ[4.1.0]ヘプタン-3-カルボキサミド(10)(1g)に28%アンモニア水溶液(5ml)を室温で加えた。混合液を、40℃で時間攪拌後、溶媒を減圧濃縮し、(1S,3R,4R)-3-アミノ-4-ヒドロキシ-N,N-ジメチルシクロヘキサンカルボキサミド(1.18g)を得た。

[工程2] (1R,2R,4S)-2-[(tert-ブトキシカルボニル)アミノ]-4-[(ジメチルアミノ)カルボニル]シクロヘキシルメタンスルホナート(2)の合成
tert-ブチル {(1R,2R,5S)-5-[(ジメチルアミノ)カルボニル]-2-ヒドロキシシクロヘキシルカルボニル}カルバマート(11)(214.59g)の4-メチル-2-ペンタノン(MIBK)溶液(1875ml)に、室温攪拌下で塩化メタンスルホニル(159.07g)を加えた。混合液に、室温でトリエチルアミン(170.62g)を加え、同温度で1時間攪拌した。反応液に水を加えた後、有機層を分取した。溶媒を減圧濃縮後、濃縮残渣にMIBK(750ml)を加え、室温で3時間攪拌した。析出した結晶をろ取、乾燥して標題化合物(2)(242.57g)を得た。

(1R,2R,4S)-2-[(tert-ブトキシカルボニル)アミノ]-4-[(ジメチルアミノ)カルボニル]シクロヘキシルメタンスルホナート(2)(20.0g)のN,N-ジメチルアセトアミド(DMAC)溶液(40ml)に、室温にて、アジ化ナトリウム(7.14g)及びドデシルピリジニウム クロリド(7.80g)を加えた。混合液を60℃で72時間攪拌後、室温に放冷した。反応液に水を加え酢酸エチル エステルで抽出した。抽出液を飽和重層水、水で洗浄後、溶媒を減圧濃縮した。濃縮残渣にn-ヘキサン-酢酸エチルエステル(5:1)の混合溶媒(300ml)を加え、室温で1時間攪拌した。析出した結晶をろ取した。得られた結晶にn-ヘキサン-酢酸エチルエステル(5:1)の混合溶媒(300ml)を加え、攪拌・結晶ろ過の操作を2回繰り返し、標題化合物(3)(4.6g,26.9%)を得た。

tert-ブチル (1R,2S,5S)-2-アミノ-5-(ジメチルアミノカルボニル)シクロヘキシルカルバマート 1・シュウ酸塩(1)(100.1g)のアセトニトリル懸濁液(550ml)に、60℃にてトリエチルアミン(169ml)を加えた。そのままの温度にて、2-[(5-クロロピリジン-2-イル)アミノ]-2-オキソアセタート エチルエステル 1・塩酸塩(4)(84.2g)を加え、6時間攪拌後、室温にて16時間攪拌した。反応液に水を加え、10℃にて1時間30分攪拌後、結晶をろ取して標題化合物(5)(106.6g)を得た。

(参考例8)
国際公開第2007/032498号パンフレットに記載に従って、化合物(1a)と無水シュウ酸から、式(1)で表される化合物を製造し、化合物(1)に混在するtrans-異性体である化合物(1-trans)の割合を測定した。なお、基質は異なるが、国際公開第2001/74774号パンフレットの参考例157に記載のアジド化によるcis-異性体とtrans-異性体の生成比とほぼ同じような比率で、化合物(1a)中には、化合物(1a)のtrans-異性体である化合物(1a-trans)を約10%程度含む。
国際公開第2007/032498号パンフレットに記載の、2回の結晶化工程を経由する化合物(1)の製造における、第1回目の結晶化における類縁物質である化合物(1-trans)の、経時的な混入の割合を検討したところ、Run1では1~3%程度であったが、Run2及びRun3では、それぞれ4.2%以上及び3.7~6.7%(撹拌時間に比例して増加)となり、類縁物質(1-trans)の混入割合が、撹拌時間が増すにつれて急激に増加し、国際公開第2007/032498号パンフレットに記載の含有率に比べて約1.5~2.5倍に増加する結果であった。
(参考例9)
国際公開第2007/032498号パンフレットの記載にしたがって、化合物(1a)と無水シュウ酸から、化合物(1)を製造し、撹拌時間による化合物(1)に混在するtrans-異性体である化合物(1-trans)の割合を測定した。結果は表2に示した。
上記の表1に示した類縁物質である化合物(1-trans)の混入割合の低減を目的に、(第2回目の結晶化)における結晶化の溶媒量を増量して、その効果を検討した。表2に検討結果を示したが、溶媒量を増量しても、撹拌時間が長くなると類縁物質(1-trans)の混入の割合が急激に増加することが明らかになり、結晶化による製造方法の堅牢性に懸念を残す結果であった。
(参考例10)
化合物(1a)と無水シュウ酸から、従来法(国際公開第2007/032498号パンフレット)の第1回目の結晶化における析出結晶の結晶形の経時変化を粉末X線で測定した。
従来法(国際公開第2007/032498号パンフレット)の第1回目の結晶化において、結晶析出直後はアモルファスであることが粉末X線結晶回析図から明らかになった。また、第1回目の結晶化における結晶析出直後に、シュウ酸溶液滴下箇所における局所的な過飽和により非晶化が目視で観察された。さらに、シュウ酸溶液の滴下速度(滴下時間)によって結晶形が異なり、結晶化工程の安定性に問題があることが明らかになり、工業的製造方法としては適さないことが判明した。
上記の参考例3に記載の方法で製造した化合物(1)の無水物結晶は、25℃で撹拌を行うと母液の水分含有率により、1水和物に結晶転移することがわかった。一方、上記の参考例3の製造方法とは異なり、アセトニトリル溶液中の水分含有率を0.65%以下に保ち、撹拌温度を60~80℃で1時間以上処理する方法では、先のForm 1型の無水物結晶とは異なり、1水和物に結晶転移しにくいForm 2型の無水物結晶が得られた。Form 2型の無水物結晶はろ過性などの操作性にも優れていることがわかった。化合物(1)の無水物結晶のForm 1型及びForm 2型の粉末X線回析図を図1、2に、熱分析による回析図を、それぞれ図5、6に示した。
化合物(1a)を、6%含水アセトニトリル中で、無水シュウ酸と処理して、標題の2種の式(1b)で表される化合物の1水和物結晶の多形結晶が得られた(製造方法は下記の実施例3に記載した)。Form 1型及びForm 2型の1水和物結晶の粉末X線回析を図3、4に、熱分析による回析図を、それぞれ図7、8に示した。Form 1型の1水和物結晶は、室温で約2時間放置するとForm 2型の1水和物結晶に転移することがわかり、Form 2型の1水和物結晶が準安定晶であることがわかった。さらに、Form 1型の1水和物結晶を、化合物(1)の化合物の無水物結晶に結晶転移を行った場合、Form 1型の1水和物結晶が、1水和物結晶の準安定晶であるForm 2型への結晶転移を経由して無水物結晶に転移することから、化合物(1)の無水物結晶を製造する場合、出発物質として用いる1水和物結晶としては、Form 2型の1水和物結晶が好ましいことがわかった。
[A工程]
化合物(1a)(45.61g,0.16mol)にアセトニトリル(595ml)及び水(43ml)を加え、攪拌下に内温が50~70℃になるように加熱した。あらかじめ無水シュウ酸(18.87g,0.21mol)とアセトニトリル(255ml)から調製した溶液を、上記の溶液に、内温を50~70℃に保ちながら、1時間をかけて滴下した。滴下終了後、反応混合液を50~70℃で5時間撹拌した後、内温が20~40℃になるように冷却した。析出結晶を集め、アセトニトリルで洗浄後、乾燥して式(1b)で表される化合物の1水和物結晶(59.14g,94.1%)を得た。得られた式(1b)で表される1水和物結晶の粉末X線回析図、及び熱分析の結果は、図4、8に示した、実施例2のForm 2型の1水和物結晶のものと同じであった。
[B工程]
式(1b)で表される化合物の1水和物結晶(13.25g,33.68mmol)に、アセトニトリル(126ml)を加え、反応系内の水分含量が約0.7%であることを確認後、内温70~75℃で、5時間撹拌した。反応液を内温40~70℃(外温は80℃以下)に保って、アセトニトリル(66ml)を減圧留去した。次に、濃縮留去した量と同量の市販の無水アセトニトリル(66ml)を反応混合液に添加した後、反応混合液の水分含量が約0.15%であることを確認後、反応混合液を50~70℃で1時間撹拌した。反応混合液を内温20~40℃に冷却後、析出結晶をろ取し、アセトニトリルで洗浄、及び乾燥して表題物である式(1)で表される化合物の無水物結晶(12.46g,98.6%)を得た。得られた式(1)で表される化合物の無水物結晶の粉末X線回析図、及び熱分析の結果は、図2、6に示した実施例1のForm 2型の無水物結晶のものと同じであった。
[図9]に示したように、式(1b)で表される化合物の1水和物の結晶、新たに調製したtrans-異性体である式(1b-trans)で表される化合物の1水和物結晶、及び式(1-trans)で表される化合物の無水物結晶の、trans-異性体3種の結晶の水溶性を比較した。
13%トルエン/アセトニトリル溶液に水を添加して含水率を変化させ、式(1b)で表される化合物の1水和物の結晶、及びtrans-異性体である式(1b-trans)で表される化合物の1水和物の水溶性を比較した。
従来法(国際公開第2007/032498号パンフレット)の第1回目の結晶化における類縁物質であるtrans-異性体の式(1-trans)で表される化合物の無水物結晶の割合と、本願発明の(A法)によって製造した式(1b)で表される化合物の1水和物結晶中のtrans-異性体(1-trans)の含有割合を比較した。
[アジド化工程]
アジ化ナトリウム(32.82g)及びドデシルピジニウム クロリド(35.83g)に水(184ml)を加え、60℃で1時間撹拌した。反応溶液にトルエン(460ml)を加え、同温度で減圧下に水をディーン・スターク装置を用いて共沸脱水した。トルエン懸濁液の水分が0.1%程度であるのを確認後、留去したトルエン量を添加した。(1R,2R,4S)-2-[(tert-ブトキシカルボニル)アミノ]-4-[(ジメチルアミノ)カルボニル]シクロヘキシルメタンスルホナート(2)を加え、内温60~65℃で48時間撹拌した。反応液を40℃に冷却後、5%炭酸水素ナトリウム水溶液(460ml)を添加し、40℃に加温したトルエン(184ml)で3回抽出した。抽出液を合わせ、40℃に加温した水(138ml)で2回洗浄後、抽出液を約半分の容量まで濃縮した。得られた化合物(3)のトルエン溶液は、これ以上の精製を行うことなく次の工程で使用した。
[還元工程]
上記の[アジド化工程]で得られた化合物(3)のトルエン溶液に、メタノール(460ml)、市販の7.5%パラジウム-炭素(川研ファインケミカル株式会社製:12.88g)、及びギ酸アンモニウム(17.48g)を加え、内温30~50℃の範囲で1時間撹拌した。不溶物である金属触媒をろ過にて除去し、これをメタノール(184ml)で洗浄した。ろ液を減圧濃縮し、濃縮残渣にトルエン(230ml)を加え、減圧濃縮を行って、粗生成物である化合物(1a)を得た。得られた粗生成物の化合物(1a)はこれ以上の精製を行うことなく次の工程で使用した。
[A工程]
上記の[還元工程]で得られた化合物(1a)にアセトニトリル(644ml)及び水(46ml)を加え、攪拌下に内温が50~70℃になるように加熱した。ここに、あらかじめ無水シュウ酸(18.18g)とアセトニトリル(276ml)から調製した溶液を、内温を50~70℃に保ち、1時間をかけて滴下した。滴下終了後、反応混合液を50~70℃で5時間撹拌した後、内温が20~40℃になるように冷却した。析出結晶を集め、アセトニトリル(92ml)で洗浄後、乾燥して式(1b)で表される化合物の1水和物結晶を得た。
[B工程]
上記の[A工程]で得られた式(1b)で表される化合物の1水和物結晶に、アセトニトリル(920ml)を加え、反応系内の水分含量が約0.7%であることを確認後、内温が70~75℃で、5時間撹拌した。反応液を内温40~70℃(外温は80℃以下)に保って、アセトニトリル(526ml)を減圧留去した。あらたに留去した量と同容量のアセトニトリル(526ml)を添加し、反応混合液の水分含量が約0.15%であることを確認後、反応混合液を50~70℃で1時間撹拌した。反応混合液を内温20~40℃に冷却後、析出結晶をろ取し、乾燥して表題物[52.12g、化合物(2)から55%]を得た。
本発明の実施例7で製造した tert-ブチル {(1R,2S,5S)-2-アミノ-5-[(ジメチルアミノ)カルボニル]シクロヘキシル}カルバマート オキサレート (1)の無水物結晶を用い、参考例7に記載された方法を用い、式(X-a)で示した化合物の純度測定をHPLC法で行った。
Claims (36)
- 加温が、50~80℃である、請求項1又は2に記載の製造方法。
- 加温が、70~75℃である、請求項1又は2に記載の製造方法。
- さらに、撹拌工程において、有機溶媒全体に対して1/2~4/7容量の有機溶媒を40~75℃の範囲で減圧留去し、その後、留去した量に相当する量の有機溶媒を再添加することを含む、請求項1~4のいずれか1項に記載の製造方法。
- 有機溶媒の減圧留去及び再添加において、有機溶媒中の水分含量を0.2重量%未満にすることを特徴とする、請求項5に記載の製造方法。
- 含水有機溶媒が、4%以上の含水有機溶媒である、請求項2~7のいずれか1項に記載の製造方法。
- 含水有機溶媒が、4~10%の含水有機溶媒である、請求項2~7のいずれか1項に記載の製造方法。
- 無水シュウ酸との処理が、無水シュウ酸の有機溶媒溶液を滴下するものである、請求項2~9のいずれか1項に記載の製造方法。
- 滴下を50~80℃で行う、請求項10に記載の製造方法。
- 滴下終了後、さらに50~80℃で2~5時間撹拌を行う、請求項11に記載の製造方法。
- 有機溶媒が、酢酸C1~C5アルキルエステル系溶媒、直鎖状又は分枝鎖状のC1~C8アルコール系溶媒、C1~C6ケトン系溶媒、トルエン系溶媒、及びC2~C5ニトリル系溶媒からなる群より選ばれる1種又は2種以上の溶媒である、請求項1~12のいずれか1項に記載の製造方法。
- 有機溶媒が、アセトニトリル、トルエン、又はアセトニトリルとトルエンの混合溶媒である、請求項1~12のいずれか1項に記載の製造方法。
- 有機溶媒が、アセトニトリルである、請求項1~12のいずれか1項に記載の製造方法。
- 高純度の式(1b)で表される化合物。
- 1水和物結晶である、請求項16に記載の化合物。
- 式(1b)で表される化合物の純度が97.0%以上である、請求項7に記載の製造方法。
- 式(1b)で表される化合物の純度が99.0%以上である、請求項7に記載の製造方法。
- 高純度の式(1)で表される化合物。
- 無水物結晶である、請求項20に記載の化合物。
- 式(1)で表される化合物の純度が97.0%以上である、請求項1又は2に記載の製造方法。
- 式(1)で表される化合物の純度が99.0%以上である、請求項1又は2に記載の製造方法。
- 粉末X線回折スペクトルが図2に示すパターンを有することを特徴とする、請求項24に記載の式(1)で表される化合物のForm-2型の無水物結晶。
- 粉末X線回折スペクトルが図4に示すパターンを有することを特徴とする、請求項26に記載の式(1b)で表される化合物のForm-2型の1水和物結晶。
- 粉末X線回折スペクトルが図3に示すパターンを有することを特徴とする、請求項28に記載の式(1b)で表される化合物のForm-1型の1水和物結晶。
- 請求項1に記載の方法で製造した、式(1)で表される化合物の無水物結晶を使用することを特徴とする、下記式(X-a)
で表される化合物を製造する方法であって、
式(1)で表される無水物結晶を、塩基の存在下に、下記式(4)
で表される化合物と処理して、下記式(5)
(式中、Bocはtert-ブトキシカルボニル基を示す。)
で表される化合物を得る工程;
式(5)の化合物のBoc基を脱保護した後、塩基の存在下に、下記式(7)
で表される化合物と処理して、下記式(X)
で表されるフリー体の化合物を得る工程;及び
式(X)で表される化合物を、溶媒中、p-トルエンスルホン酸又はその水和物と処理して、式(X-a)で表される化合物を得る工程、
を含む、式(X-a)で表される化合物の製造方法。 - 高純度の式(X-a)で表される化合物。
- 純度が99.50重量%以上である、請求項32に記載の化合物。
- 純度が99.75重量%以上である、請求項32に記載の化合物。
- 式(X-a)で表される化合物の純度が99.50重量%以上である、請求項30又は31に記載の製造方法。
- 式(X-a)で表される化合物の純度が99.75重量%以上である、請求項30又は31に記載の製造方法。
