かかる構成であることにより、動物に適用した場合に、高い動物寄生虫駆除活性を示すことができる。
「ハロゲン原子」とはフッ素原子、塩素原子、臭素原子またはヨウ素原子を示す。
「Ca-Cb(a、bは1以上の整数を表す)」との表記、例えば、「C1-C3」とは炭素原子数が1~3個であることを意味し、「C2-C6」とは炭素原子数が2~6個であることを意味する。
「C1-C3ハロアルキル基」とは例えば、トリフルオロメチル、ペンタフルオロエチル、ヘプタフルオロ-n-プロピル、ヘプタフルオロ-i-プロピル、2,2-ジフルオロエチル、2,2-ジクロロエチル、2,2,2-トリフルオロエチル、2-フルオロエチル、2-クロロエチル、2-ブロモエチル、2-ヨードエチル、2,2,2-トリクロロエチル、2,2,2-トリブロモエチル、1,3-ジフルオロ-2-プロピル、1,3-ジクロロ-2-プロピル、1-クロロ-3-フルオロ-2-プロピル、1,1,1-トリフルオロ-2-プロピル、2,3,3,3-トリフルオロ-n-プロピル、1,1,1,3,3,3-ヘキサフルオロ-2-プロピル、1,1,1,3,3,3-ヘキサフルオロ-2-クロロ-2-プロピル、1,1,1,3,3,3-ヘキサフルオロ-2-ブロモ-2-プロピル、1,1,2,3,3,3-ヘキサフルオロ-2-クロロ-n-プロピル、1,1,2,3,3,3-ヘキサフルオロ-2-ブロモ-n-プロピル、1,1,2,3,3,3-ヘキサフルオロ-1-ブロモ-2-プロピル、2,2,3,3,3-ペンタフルオロ-n-プロピル、3-フルオロ-n-プロピル、3-クロロ-n-プロピル、3-ブロモ-n-プロピルなどの同一または異なっていてもよい1以上のハロゲン原子によって置換された直鎖状または分岐鎖状の炭素原子数1~3個のアルキル基を示す。
以下の製造方法に示される一般式においては、X1、X2、X3、X4、R1、R2、Q1、Y1、Y2、Y3、Y4、およびY5は前記一般式(1)におけるX1、X2、X3、X4、R1、R2、Q1、Y1、Y2、Y3、Y4、およびY5とそれぞれ同じものを示す。LGはハロゲン原子、ヒドロキシル基等の脱離能を有する官能基を示し、Halは塩素原子または臭素原子を示し、XaおよびXbは塩素原子、臭素原子、またはヨウ素原子を示す。R3はC1-C6アルキル基、C3-C9シクロアルキル基、C2-C6アルケニル基、または、C2-C6アルキニル基を示す。
<製造方法1>
一般式(11)で表される脱離基を有するニトロ芳香族カルボン酸誘導体と、一般式(12)で表される芳香族アミン誘導体とを適当な溶媒中もしくは無溶媒で反応させることにより、一般式(13)で表されるニトロ芳香族カルボン酸アミド誘導体を製造することができる。本工程では適当な塩基を用いることもできる。
これらの塩基は、一般式(11)で表される化合物に対して0.01~5倍モル当量の範囲で適宜選択して使用すればよい。
一般式(11)で表される化合物の中で、芳香族カルボン酸ハライド誘導体は、芳香族カルボン酸からハロゲン化剤を使用する常法により容易に製造することができる。ハロゲン化剤としては、例えば、塩化チオニル、臭化チオニル、オキシ塩化リン、オキザリルクロリド、三塩化リンなどのハロゲン化剤を示すことができる。
一方、ハロゲン化剤を使用せずにニトロ芳香族カルボン酸誘導体と一般式(12)で表される化合物から一般式(13)で表される化合物を製造することが可能である。その方法としては、例えば、Chem.Ber.788ページ(1970年)に記載の方法を挙げることができる。具体的には、1-ヒドロキシベンゾトリアゾールなどの添加剤を適宜使用し、N,N’-ジシクロヘキシルカルボジイミドなどの縮合剤を用いる方法を示すことができる。この場合に使用される他の縮合剤としては、1-エチル-3-(3-ジメチルアミノプロピル)カルボジイミド、1,1’-カルボニルビス-1H-イミダゾールなどを示すことができる。
縮合剤を用いる方法、混合酸無水物法共に、前記文献記載の溶媒、反応温度、反応時間に限定されることは無い。溶媒としては適宜反応の進行を著しく阻害しない不活性溶媒を使用すればよい。また反応温度、反応時間についても、反応の進行に応じて、適宜選択すればよい。
一般式(13)で表されるニトロ基を有する芳香族カルボン酸アミド誘導体は、還元反応により、一般式(14)で表されるアミノ基を有する芳香族カルボン酸アミド誘導体に導くことができる。還元反応としては水素添加反応を用いる方法と金属化合物(例えば、塩化第一スズ(無水物)、鉄粉、亜鉛粉など)を用いる方法を例示することできる。
(方法A)
一般式(14)で表される化合物を溶媒中、アルデヒド類またはケトン類と反応させ、触媒を添加し、水素雰囲気下で反応させることにより、一般式(15)で表される化合物を製造することができる。
溶媒としては、本反応の進行を著しく阻害しないものであればよく、例えば、ヘキサン、シクロヘキサン、メチルシクロヘキサンなどの脂肪族炭化水素類、ベンゼン、キシレン、トルエンなどの芳香族炭化水素類、ジクロロメタン、クロロホルム、四塩化炭素、1,2-ジクロロエタンなどのハロゲン化炭化水素類、ジエチルエーテル、ジオキサン、テトラヒドロフラン、1,2-ジメトキシエタンなどのエーテル類、ジメチルホルムアミド、ジメチルアセトアミドなどのアミド類、アセトニトリル、プロピオニトリルなどのニトリル類、アセトン、メチルイソブチルケトン、シクロヘキサノン、メチルエチルケトンなどのケトン類、酢酸エチル、酢酸ブチルなどのエステル類、メタノール、エタノールなどのアルコール類、1,3-ジメチル-2-イミダゾリジノン、スルホラン、ジメチルスルホキシドなどの不活性溶剤、水などの溶媒を示すことができ、これらの溶媒は単独もしくは2種以上混合して使用することができる。
触媒としてはパラジウム-カーボン、水酸化パラジウム-カーボンなどのパラジウム触媒類、ラネーニッケルなどのニッケル触媒類、コバルト触媒類、プラチナ触媒類、ルテニウム触媒類、ロジウム触媒類などを示すことができる。
ケトン類としては、例えば、アセトン、パーフルオロアセトン、メチルエチルケトンなどのケトン類を示すことができる。
反応圧力は1気圧から100気圧の範囲でそれぞれ適宜選択すればよい。反応温度は-20℃から使用する溶媒の還流温度の範囲で適宜選択すればよい。また反応時間は、数分から96時間の範囲で適宜選択すればよい。
一般式(14)で表される化合物を溶媒中で、アルデヒド類またはケトン類と反応させて、還元剤を処理することにより、一般式(15)で表される化合物を製造することができる。
溶媒としては、本反応の進行を著しく阻害しないものであればよく、例えば、ヘキサン、シクロヘキサン、メチルシクロヘキサンなどの脂肪族炭化水素類、ベンゼン、キシレン、トルエンなどの芳香族炭化水素類、ジクロロメタン、クロロホルム、四塩化炭素、1,2-ジクロロエタンなどのハロゲン化炭化水素類、ジエチルエーテル、ジオキサン、テトラヒドロフラン、1,2-ジメトキシエタンなどのエーテル類、ジメチルホルムアミド、ジメチルアセトアミドなどのアミド類、アセトニトリル、プロピオニトリルなどのニトリル類、アセトン、メチルイソブチルケトン、シクロヘキサノン、メチルエチルケトンなどのケトン類、酢酸エチル、酢酸ブチルなどのエステル類、メタノール、エタノールなどのアルコール類、1,3-ジメチル-2-イミダゾリジノン、スルホラン、ジメチルスルホキシドなどの不活性溶剤、水などの溶媒を示すことができ、これらの溶媒は単独もしくは2種以上混合して使用することができる。
還元剤としては、例えば、ソディウムボロハイドライド、ソディウムシアノボロハイドライド、ソディウムトリアセテートボロハイドライドなどのボロハイドライド類などを示すことができる。
ケトン類としては、例えば、アセトン、パーフルオロアセトン、メチルエチルケトンなどのケトン類を示すことができる。
反応温度は-20℃から使用する溶媒の還流温度の範囲で適宜選択すればよい。また反応時間は、数分から96時間の範囲で適宜選択すればよい。
一般式(14)で表される化合物を溶媒中、または無溶媒でアルデヒド類と反応させることにより、一般式(15)の化合物を製造することができる。
溶媒としては、本反応の進行を著しく阻害しないものであればよく、例えば、ヘキサン、シクロヘキサン、メチルシクロヘキサンなどの脂肪族炭化水素類、ベンゼン、キシレン、トルエンなどの芳香族炭化水素類、ジクロロメタン、クロロホルム、四塩化炭素、1,2-ジクロロエタンなどのハロゲン化炭化水素類、ジエチルエーテル、ジオキサン、テトラヒドロフラン、1,2-ジメトキシエタンなどのエーテル類、ジメチルホルムアミド、ジメチルアセトアミドなどのアミド類、アセトニトリル、プロピオニトリルなどのニトリル類、アセトン、メチルイソブチルケトン、シクロヘキサノン、メチルエチルケトンなどのケトン類、酢酸エチル、酢酸ブチルなどのエステル類、メタノール、エタノールなどのアルコール類、1,3-ジメチル-2-イミダゾリジノン、スルホラン、ジメチルスルホキシドなどの不活性溶剤、硫酸、塩酸などの無機酸類、蟻酸、酢酸などの有機酸類、水などの溶媒を示すことができ、これらの溶媒は単独もしくは2種以上混合して使用することができる。
アルデヒド類としては、例えば、ホルムアルデヒド、アセトアルデヒド、プロピオンアルデヒドなどを示すことができる。
反応温度は-20℃から使用する溶媒の還流温度、反応時間は、数分から96時間の範囲でそれぞれ適宜選択すればよい。
一般式(15)で表される芳香族アミン誘導体と一般式(16)で表されるカルボン酸誘導体または脱離基を有する炭酸エステル誘導体を適当な溶媒中で反応させることにより、一般式(1)で表される芳香族カルボン酸アミドを製造することができる。本工程では適当な塩基もしくは溶媒を用いることが可能であり、その塩基もしくは溶媒としては、1-(i)に例示したものを用いることができる。反応温度、反応時間などについても、1-(i)の例示に従うことができる。
一般式(16)の中で、カルボン酸クロリド誘導体はカルボン酸誘導体から、ハロゲン化剤を使用する常法により、容易に製造することができる。ハロゲン化剤としては、1-(i)の例示に従うことができる。
ハロゲン化剤を使用せずにカルボン酸誘導体(16)と一般式(15)で表される化合物から一般式(1)で表される化合物を製造する方法もあり、1-(i)の例示の方法に従うことで製造可能である。
<製造方法2>
一般式(17)で表される化合物と一般式(12)で表される化合物とを、1-(i)に記載の条件に従い反応させることにより、一般式(18)で表される化合物を製造することができる。
例えば、J.Org.Chem.280ページ(1958年)に記載の条件に従うことにより、アンモニア等のアミノ化剤を使用してアミノ化反応を行い、一般式(15)で表される化合物を製造することが可能である。反応溶媒などの条件は文献記載のものに限定されることは無く、適宜反応の進行を著しく阻害しない不活性溶媒を使用すればよい。また反応温度、反応時間についても、反応の進行に応じて、適宜選択すればよい。尚、アミノ化剤としては、アンモニアのほかに、メチルアミン、エチルアミンなどを示すことができる。
一般式(15)で表される化合物と一般式(16)で表される化合物とを、1-(i)に記載の条件に従い反応させることにより、一般式(1)で表されるアミド誘導体を製造することができる。
<製造方法3>
一般式(19)で表される化合物と一般式(12)で表される化合物とを、1-(i)に記載の条件に従い反応させることにより、一般式(20)で表される化合物を製造することができる。
一般式(20)で表されるニトロ芳香族カルボン酸アミド誘導体を適当な溶媒中、または無溶媒で、適当なフッ素化剤と反応させることにより、一般式(21)で表される化合物を製造することができる。
溶媒としては、本反応の進行を著しく阻害しないものであればよく、例えば、ヘキサン、シクロヘキサン、メチルシクロヘキサンなどの脂肪族炭化水素類、ベンゼン、トルエン、キシレンなどの芳香族炭化水素類、ジクロロメタン、クロロホルム、四塩化炭素、1,2-ジクロロエタンなどのハロゲン化炭化水素類、ジエチルエーテル、ジオキサン、テトラヒドロフラン、1,2-ジメトキシエタンなどの鎖状または環状エーテル類、酢酸エチル、酢酸ブチルなどのエステル類、アセトン、メチルイソブチルケトン、シクロヘキサノン、メチルエチルケトンなどのケトン類、アセトニトリル、プロピオニトリルなどのニトリル類、1,3-ジメチル-2-イミダゾリジノン、スルホラン、ジメチルスルホキシド、N,N-ジメチルホルムアミド、N-メチルピロリドン、N,N-ジメチルアセトアミドなどの非プロトン性極性溶媒を示すことができ、これらの溶媒は単独もしくは2種以上混合して使用することができる。
これらのフッ素化剤は一般式(20)で表されるニトロ芳香族カルボン酸アミド誘導体に対して1から10倍モル当量の範囲、または溶媒として適宜選択して使用すればよい。
反応温度は-80℃から使用する溶媒の還流温度の範囲で適宜選択すればよい。また反応時間は、数分から96時間の範囲で適宜選択すればよい。
<製造方法4>
一般式(22)で表されるアミノ基を有するカルボン酸類を出発原料として、1-(i)に記載の条件に従い、一般式(16)で表される化合物と反応させることにより、一般式(23)で表されるアシルアミノ基を有するカルボン酸類を製造することができる。
一般式(23)で表されるアミド化合物と、一般式(24)で表されるハロゲンなどの脱離基を有する化合物を溶媒中もしくは無溶媒で反応させることにより、一般式(25)で表されるアミド誘導体を製造することができる。本工程では適当な塩基もしくは溶媒を用いることが可能であり、その塩基もしくは溶媒としては、1-(i)に例示したものを用いることができる。反応温度、反応時間などについても、1-(i)の例示に従うことができる。
一般式(25)から一般的な手法を利用した加水分解または、Pd触媒を用いた方法により、一般式(26)で表される化合物を得ることができる。加水分解による方法としては、メタノール、エタノールまたはテトラヒドロフラン、ジオキサンの単独または混合溶媒中、等量から5倍モルの水性またはアルコール性の水酸化リチウム、水酸化ナトリウムまたは水酸化カリウムを使用し塩基加水分解することにより得ることができる。またトルエン、キシレンなどの非水溶性溶媒中でも、水性の水酸化ナトリウム、水酸化カリウムまたは水酸化リチウムなどの塩基と、テトラブチルアンモニウムブロマイド、ベンジルトリエチルアンモニウムクロライドまたはクラウンエーテルなどの相間移動触媒との組み合わせによって加水分解を行うこともできる。また、塩酸や硫酸等の無機酸類、酢酸やトリフルオロ酢酸等の有機酸、強酸性樹脂を使用した酸加水分解を行うこともできる。
反応温度は、-20℃から使用する溶媒の還流温度の範囲で適宜選択すればよい。また反応時間は、数分から96時間の範囲で適宜選択すればよい。
Pdを用いる方法としては、例えば、Tetrahedron Letters 4371ページ(1987年)に記載されている方法を用いることができる。溶媒、反応温度などの条件は、文献記載のものに限定されることはない。
一般式(26)で表される化合物を塩化チオニル、オキザリルクロリド、ホスゲン、オキシ塩化リン、五塩化リン、三塩化リン、臭化チオニル、三臭化リン、ジエチルアミノ硫黄トリフルオリドなどと反応させるという公知の常法により、一般式(27)で表される化合物を製造することができる。
一般式(27)で表される化合物と一般式(12)で表される化合物とを、1-(i)に記載の条件に従い反応させることにより、一般式(1)で表される化合物を製造することができる。
一般式(26)で表される化合物を、1-(i)に記載の縮合剤を用いる条件、もしくは混合酸無水物法を用いる条件に従い、一般式(12)で表される化合物と反応させることにより、一般式(1)で表されるアミド誘導体を製造することができる。
<製造方法5>
一般式(28)で表される化合物を、3-(ii)に記載の条件に従い、フッ素化することにより一般式(29)で表される化合物を製造することができる。
一般式(29)で表される化合物を、1-(ii)に記載の条件に従い、還元することにより一般式(30)で表される化合物を製造することができる。

具体的には例えば、一般的に通常使用される製剤処方に従って適当な固体担体及び/又は液体担体等及び必要に応じて補助剤等と共に適当な割合に配合して溶解、懸濁、混合、含浸、吸着若しくは付着させ、使用目的に応じて適当な剤型に調製して使用すれば良い。
本発明の動物寄生虫駆除用組成物を経口投与する場合、カプセル剤、錠剤、丸剤、粉剤、顆粒剤、細粒剤、散剤、シロップ、腸溶剤、懸濁剤、ペースト或いは薬物配合飲料または飼料の形態で、投与することが可能である。
投与方法としては、対象動物の肩甲背部の皮膚等に滴下剤を滴下することにより外部寄生虫を駆除するスポットオン処理、対象動物の背中線に沿って液状の薬剤を処理し、処理された薬剤が体表面に拡散することにより、外部寄生性害虫を防除するポアオン処理等の局所処理法の他に、首輪等に薬剤を担持させ、薬剤を放出する処理法、直接体表面に液剤又は軟膏剤等を塗布する処理法、スプレー等でエアゾル剤等を処理する方法、注射剤を筋肉内、皮下等に注射する方法、座剤を用いた肛門投与、等が挙げられる。
本発明の動物寄生虫駆除用組成物中の有効成分の濃度は、一般に0.0001~100重量%、好ましくは0.001~99%、更に好ましくは0.005~80重量%程度である。寄生虫駆除剤は、一般に、使用時に適当な濃度に希釈して使用される高濃度な組成物として提供しうる。
本明細書に記載された全ての文献、特許出願、および技術規格は、個々の文献、特許出願、および技術規格が参照により取り込まれることが具体的かつ個々に記された場合と同程度に、本明細書に参照により取り込まれる。

1H-NMR(CDCl3,ppm)δ7.19-7.25(2H,m),7.35(1H,t,J=7.8Hz),7.87-7.97(4H,m),8.08(1H,s),8.25(1H,d,J=12.0Hz),8.37(1H,s),8.65(1H, t,J=8.0Hz).
化合物番号7-1574或いは7-1733の化合物を10部、ソルポール355S(東邦化学製、界面活性剤)6部、ソルベッソ150(エクソン製)84部、以上を均一に攪拌混合し、乳剤として、動物寄生虫駆除用組成物を得た。
化合物番号7-1574或いは7-1733の化合物を1部、サラシミツロウ50部、白色ワセリン49部を十分混合し、軟膏剤として、動物寄生虫駆除用組成物を得た。
化合物番号7-1574或いは7-1733の化合物を2部、植物油(オリーブ湯)10部、結晶セルロース3部、ホワイトカーボン20部、カオリン65部を十分混合した後、打錠し、錠剤として、動物寄生虫駆除用組成物を得た。
化合物番号7-1574或いは7-1733の化合物を10部、食品添加物用プロピレングリコール10部、植物油(コーンオイル)80部を混合し、注射剤として、動物寄生虫駆除用組成物を得た。
化合物番号7-1574或いは7-1733の化合物を5部、界面活性剤20部、イオン交換水75部を十分混合し、液剤として、動物寄生虫駆除用組成物を得た。
ネコノミ(Ctenocephalides felis)殺虫試験
所定濃度に調製した有効成分のアセトン溶液0.5mlを、底面が平滑なガラス管(内径2.6cm、底面積5.3cm2、高さ12cm)に滴下し、室温下で溶媒を蒸散させ、底面に薬剤のドライフィルムを形成させた。1ガラス管当たりノミ成虫約10頭を入れ、試験化合物にそれぞれ接触暴露させた。暴露後24時間のノミの状態を、生存、死亡(瀕死を含む)に分けて調査し、死虫率を算出した(3連制)。結果を第4表に示した。
フタトゲチマダニ(Haemaphyxalis longicornis)殺虫試験
所定濃度に調製した有効成分のアセトン溶液1mlを、9cm径シャーレ内面の濾紙に滴下し、室温下でアセトンを蒸散させた。濾紙上に1シャーレ当たりマダニ幼虫約100頭を放ち、ポリエチレンシートで覆って輪ゴムで密封し、温度25℃、相対湿度100%の暗所で保存して、試験物質を暴露させた。暴露後24時間のマダニの状態を、生存、死亡(瀕死を含む)に分けて調査した。
結果、本発明にかかるアミド誘導体は、フタトゲチマダニに対しても有効な殺虫効果が認められた。

なお、比較化合物として、国際公開第2009/080203号パンフレットに開示された下記化合物(D)、化合物(E)およびフィプロニルを用いた。
クリイロコイタマダニ(Rhipicephalus sanguineus)殺虫試験
所定薬量になるよう調製した有効成分のアセトン溶液0.5mlを、10cm径ガラスシャーレ内面に滴下し、室温下でアセトンを蒸散させた。1シャーレ当たりマダニ成虫約10頭を放ち、同様に有効成分を処理したガラスシャーレで蓋をした。18時間試験物質を暴露させた後、有効成分を処理していないガラスシャーレ内にマダニ成虫を移動させた。暴露開始48時間後のマダニの状態を、生存、死亡(瀕死を含む)に分けて調査し、死虫率を算出した(1連制)。結果を第6表に示した。
なお、比較化合物として、国際公開第2009/080203号パンフレットに開示された前記化合物(B)、化合物(C)、化合物(D)、下記化合物(F)およびフィプロニルを用いた。
Claims (7)
- 前記一般式(1)において、Q1はフェニル基または1つのフッ素原子で置換されているフェニル基であり、R1は水素原子またはメチル基であり、Y1およびY5はそれぞれ独立に、臭素原子、ヨウ素原子またはトリフルオロメチル基である請求項1に記載の動物寄生虫駆除用組成物。
- 前記一般式(1)で表されるアミド誘導体が、2-フルオロ-3-(N-メチルベンズアミド)-N-(2-ブロモ-6-トリフルオロメチル-4-(ヘプタフルオロプロパン-2-イル)フェニル)ベンズアミド、2-フルオロ-3-(4-フルオロ-N-メチルベンズアミド)-N-(2-ヨード-6-トリフルオロメチル-4-(ヘプタフルオロプロパン-2-イル)フェニル)ベンズアミド、2-フルオロ-3-(3-フルオロ-N-メチルベンズアミド)-N-(2-ヨード-6-トリフルオロメチル-4-(ヘプタフルオロプロパン-2-イル)フェニル)ベンズアミド、2-フルオロ-3-(4-フルオロベンズアミド)-N-(2-ヨード-6-トリフルオロメチル-4-(ヘプタフルオロプロパン-2-イル)フェニル)ベンズアミド、または、2-フルオロ-3-(N-メチルベンズアミド)-N-(2,6-ジブロモ-4-(ヘプタフルオロプロパン-2-イル)フェニル)ベンズアミドである請求項2に記載の動物寄生虫駆除用組成物。
- 請求項1~請求項3の何れか1項に記載の動物寄生虫駆除用組成物を動物に投与することを含む、動物寄生虫の駆除方法。
- 前記動物寄生虫が外部寄生虫である、請求項4に記載の駆除方法。
- 前記外部寄生虫がノミ目害虫である、請求項5に記載の駆除方法。
- 前記外部寄生虫がダニ目害虫である、請求項5に記載の駆除方法。
