本発明のシクロブチルプリン誘導体の製造方法において、X1は、前記製造例1のように、最初から導入してもよいが、例えば、前記製造例1のような方法が困難である場合等には、目的のX1とは別の置換基をカップリング反応後にX1に変換してもよい。例えば、X1がアルキルチオ基の場合、前記一般式(1)で表されるシクロブチルプリン誘導体の製造方法は、特に限定されないが、例えば、以下の工程を含む製造方法等が挙げられる。下記工程反応式2に、本製造例のスキームを示す。なお、下記工程反応式2において、Haloは、ハロゲノ基であり、R3は、アルキル基である。
本発明の促進剤は、前述のように、前記一般式(1′)で表されるシクロブチルプリン誘導体、その互変異性体もしくは立体異性体、またはそれらの塩、溶媒和物もしくは水和物を含み、血管新生促進機能、管腔形成促進機能および神経細胞成長促進機能からなる群から選択される少なくとも一つの機能を有する促進剤である。

本発明の医薬品は、前述のように、本発明のシクロブチルプリン誘導体、その互変異性体および立体異性体、およびその塩、溶媒和物および水和物、ならびに本発明の促進剤からなる群から選択される少なくとも一つを含み、血管新生促進用、管腔形成促進用および神経細胞成長促進用からなる群から選択される少なくとも一つの用途を有する。なお、本発明において、医薬品とは、医薬品、医薬部外品を含む。本発明の医薬品としては、特に制限されないが、例えば、外傷治療薬、火傷治療薬、瘢痕治療薬、褥瘡治療薬等の創傷治癒薬、アルツハイマー治療薬、アルツハイマー予防薬、梗塞性疾患治療薬、梗塞性疾患予防薬、育毛剤等が挙げられる。
本例では、前記化学式(6)に示す6-アミノ-2-クロロ-9-[トランス-トランス-2,3-ビス(ヒドロキシメチル)シクロブチル]プリン(C11H14ClN5O2、以下、「2-Cl-C.OXT-A」という。)が、血管内皮細胞の細胞増殖、管腔形成および細胞遊走性に与える影響を調べた。
前記2-Cl-C.OXT.Aは、以下の工程1~3により合成した。なお、本例において、1H NMRおよび13C NMRスペクトルは、UltrashieldTM 400 Plus FT NMR System(BRUKER社製)を用いて測定した。ケミカルシフトは、δで表し、カップリング定数(J)は、Hzで表した。融点は、微量融点測定装置 MP-500D(ヤナコ機器開発研究所製)により測定した。元素分析は、2400 Series II CHNS/O(Perkin Elmer社製)を用いて測定した。高分解能質量分析は、APEX IV mass spectrometer(BRUKER社製)を用い、エレクトロスプレイイオン化質量分析(ESI-MS)法により測定した。
本例において、シス-トランス-2,3-ビス(ベンゾイルオキシメチル)-1-シクロブタノールは、Basacchiらの方法(G.S.Basacchi,A.Braitman,C.W.Cianci,J.M.Clark,A.K.Field,M.E.Hagen,D.R.Hockstein,M.F.Malley,T.Mitt,W.A.Slusarchyk,J.E.Sundeen,B.J.Terry,A.V.Tuomari,E.R.Weaver,M.G.Young and R.Zahler,J.Med.Chem.,1991,34,1415)に基づき、以下のようにして合成した。まず、トランス-2,3-ビス(ベンゾイルオキシメチル)-1-シクロブタノン 14.00g(41.40mmol)を、乾燥THF350mLに溶解し、窒素気流下で-78℃に冷却した。冷却後、撹拌しながら、1mmol/LのLS-セレクトリドTHF溶液42mLを前記溶液に滴下し、2時間後、さらに酢酸6mLを滴下した。この溶液を、室温に戻るまで静置後、20mLに濃縮した。前記濃縮液に、トルエン250mLおよび水100mLを加え、分液ロート中で震盪した。得られた有機層を水100mLで洗浄後、硫酸マグネシウムを用いて乾燥した。乾燥後、ろ過により固形物を除き、ろ液を20mLに濃縮した。この濃縮液(残渣液)を、シリカゲルカラムを用いたクロマトグラフィー法により分離し、あめ状のシス-トランス-2,3-ビス(ベンゾイルオキシメチル)-1-シクロブタノール(10.15g、収率72%)を得た。
つぎに、前記シス-トランス-2,3-ビス(ベンゾイルオキシメチル)-1-シクロブタノール(5.20g、15.28mmol)および2,6-ジクロロプリン(4.33g、22.9mmol)を、THF100mLに溶解し、氷冷下、トリフェニルフォスフィリン(7.87g、30mmol)を添加した。添加後、さらに、ジイソプロピルアゾジカルボキシレート(5.9mL、30mmol)を加え、一晩、50℃に保持した。この溶液を濃縮した後、残渣液を、シリカゲルカラムを用いたクロマトグラフィー法により分離し、下記化学式(10)に示す2,6-ジクロロ-9-[トランス-トランス-2,3-ビス(ベンゾイルオキシメチル)シクロブチル]プリンの白い結晶(4.44g、収率57%)を得た。
前記2,6-ジクロロ-9-[トランス-トランス-2,3-ビス(ベンゾイルオキシメチル)シクロブチル]プリン(3.73g、7.29mmol)を、MeOH50mLに懸濁し、アンモニアで飽和させた。この溶液を、200mLの鋼鉄製封菅内で、一晩、100℃に加熱した。加熱終了後、溶液を氷冷し、エバポレーターを用いて減圧下で溶媒を留去した。得られた残渣に水50mLを加え、CHCl330mLで2度洗浄した。洗浄後、水層を採取し、10mLに濃縮し、前記化学式(6)に示す6-アミノ-2-クロロ-9-[トランス-トランス-2,3-ビス(ヒドロキシメチル)シクロブチル]プリン(2-Cl-C.OXT-A)の白い結晶(1.49g、72%)を得た。以下に、この化合物の物性値を示す。
前記2-Cl-C.OXT-Aを、所定濃度(0.5、1、5、10、50、100mmol/L)になるように生理食塩水に溶解し、試料液を調製した。そして、前記所定濃度の試料液を、それぞれ10v/v%濃度になるように、2v/v%非働化ウシ胎児血清(FBS)を添加したHuMedia-EB2培地(クラボウ社製)に加え、細胞増殖用培地を調製した。なお、前記細胞増殖用培地中の2-Cl-C.OXT-Aの最終濃度は、50、100、500μmol/L、1、5および10mmol/Lであった。
管腔形成測定は、ヒト臍帯静脈内皮細胞(HUVEC)および正常ヒト皮膚線維芽細胞から構成された市販の血管新生キット(クラボウ社製)を用い、以下のようにして行った。まず、2-Cl-C.OXT-Aを、所定濃度(1、10、100、500μmol/L、1、5、10、50mmol/L)になるように生理食塩水に溶解し、試料液を調製した。そして、前記所定濃度の試料液を、それぞれ10v/v%濃度になるように、前記キット付属の培地に添加し、管腔形成用培地を調製した。なお、前記管腔形成用培地中の2-Cl-C.OXT-Aの最終濃度は、0.1、1、10、50、100、500μmol/L、1および5mmol/Lであった。
細胞遊走性測定は、CytoSelect 24-well Cell Migration Assay(CBA-100、Cell Biolabs社製)キットを用いて、以下のようにして行った。まず、2-Cl-C.OXT-Aを、所定濃度(0.1、0.5、1mmol/L)になるように生理食塩水に溶解し、試料液を調製した。そして、前記所定濃度の試料液を、それぞれ10v/v%濃度になるように、2v/v%非働化ウシ胎児血清(FBS)を添加したHuMedia-EB2培地(クラボウ社製)に加え、細胞遊走用培地を調製した。なお、前記細胞遊走用培地中の2-Cl-C.OXT-Aの最終濃度は、10、50、100μmol/Lであった。
本例では、下記化学式(11)に示す6-アミノ-9-[トランス-トランス-2,3-ビス(ヒドロキシメチル)シクロブチル]プリン(以下、「C.OXT-A」という。)による血管内皮細胞の管腔形成への影響を調べた。前記管腔形成の測定は、前記2-Cl-C.OXT-Aに代えて、C.OXT-Aを用い、前記管腔形成用培地中のC.OXT-Aの最終濃度を、1、5、10、50、および100μmol/Lとしたこと以外は、実施例1と同様にして行った。
本例では、下記化学式(12)に示す2-アミノ-9-[トランス-トランス-2,3-ビス(ヒドロキシメチル)シクロブチル]-3H-プリン-6-オン(以下、「C.OXT-G」という。)による血管内皮細胞の管腔形成への影響を調べた。前記管腔形成の測定は、前記2-Cl-C.OXT-Aに代えて、C.OXT-Gを用い、前記管腔形成用培地中のC.OXT-Gの最終濃度を、1、5、10、50、および100μmol/Lとしたこと以外は、実施例1と同様にして行った。
本例では、下記化学式(13)に示す2-クロロ-6-アミノ-9-[3、4-ジヒドロキシ-5-(ヒドロキシメチル)オキソラン-2-イル]プリン(以下、「2-Cl-ADN」という。)による血管内皮細胞の管腔形成への影響を調べた。前記管腔形成の測定は、前記2-Cl-C.OXT-Aに代えて、2-Cl-ADNを用い、前記管腔形成用培地中の2-Cl-ADNの最終濃度を、500nmol/L、1、5、および10μmol/Lとしたこと以外は、実施例1と同様にして行った。
本例では、下記化学式(14)に示す2-クロロ-6-アミノ-9-[4-ヒドロキシ-5-(ヒドロキシメチル)オキソラン-2-イル]プリン(以下、「2-Cl-DAD」という。)による血管内皮細胞の管腔形成への影響を調べた。前記管腔形成の測定は、前記2-Cl-C.OXT-Aに代えて、2-Cl-DADを用い、前記管腔形成用培地中の2-Cl-DADの最終濃度を、500nmol/L、1、5および10μmol/Lとしたこと以外は、実施例1と同様にして行った。
本例では、アデノシンによる血管内皮細胞の管腔形成への影響を調べた。前記管腔形成の測定は、前記2-Cl-C.OXT-Aに代えて、アデノシンを用い、前記管腔形成用培地中のアデノシンの最終濃度を、100μmol/L、1、2.5および5mmol/Lとしたこと以外は、実施例1と同様にして行った。
本例では、デオキシアデノシンによる血管内皮細胞の管腔形成への影響を調べた。前記管腔形成の測定は、前記2-Cl-C.OXT-Aに代えて、デオキシアデノシンを用い、前記管腔形成用培地中のデオキシアデノシンの最終濃度を、100、500μmol/L、1および2.5mmol/Lとしたこと以外は、実施例1と同様にして行った。
本例では、前記2-Cl-C.OXT-Aに代えて、ポジティブコントロールとしてVEGFを用い、前記細胞増殖用培地、前記管腔形成用培地および前記遊走性測定用培地中のVEGFの最終濃度を、10ng/mLとしたこと以外は、実施例1と同様にして、血管内皮細胞の細胞増殖、管腔形成および細胞遊走性への影響を調べた。
本例では、前記試料液に代えて、生理食塩水を、前記細胞増殖用培地、前記管腔形成用培地および前記遊走性測定用培地へ添加したこと以外は、実施例1と同様にして、血管内皮細胞の細胞増殖、管腔形成および細胞遊走性への影響を調べた。
本例では、前記試料液に代えて、牛胎児血清(FBS)を前記遊走性測定用培地に添加し、FBSの前記培地中の最終濃度を5v/v%としたこと以外は、実施例1と同様にして、血管内皮細胞の細胞遊走性への影響を調べた。
本例では、前記試料液に代えて、牛胎児血清(FBS)を前記遊走性測定用培地に添加し、FBSの前記培地中の最終濃度を10v/v%としたこと以外は、実施例1と同様にして、血管内皮細胞の細胞遊走性への影響を調べた。
図2に、細胞増殖測定における前記相対吸光度を比較したグラフを示す。同図のグラフにおいて、縦軸は、前記相対吸光度であり、各バーは、左から順に、比較例8(生理食塩水)、比較例7(VEGF)、実施例1(2-Cl-C.OXT-A)の結果である。なお、横軸下に記載した各濃度は、実施例1における細胞増殖用培地中の試料の最終濃度を示す。

本例では、前記化学式(9)に示す6-アミノ-2-チオメトキシ-9-[トランス-トランス-2,3-ビス(ヒドロキシメチル)シクロブチル]プリン(C12H17N5O2S、以下、「2-SMe-C.OXT-A」という。)を合成した。なお、前記化学式(9)において、SMeは、メチルチオ基である。
本例では、実施例1と同様にして、前記工程1~3を行い、6-アミノ-2-クロロ-9-[トランス-トランス-2,3-ビス(ヒドロキシメチル)シクロブチル]プリン(2-Cl-C.OXT-A)を合成した。そして、前記2-Cl-C.OXT-Aを用いて、さらに下記工程4~6を実施し、前記2-SMe-C.OXT.Aを合成した。なお、本例では、実施例1と同様にして、各化合物を測定した。
前記6-アミノ-2-クロロ-9-[トランス-トランス-2,3-ビス(ヒドロキシメチル)シクロブチル]プリン(170.4mg、0.60mmol)溶液、トリチルクロライド(501.8mg、1.80mmol)、トリエチルアミン(0.4mL)および4-ジメチルアミノピリジン(11.6mg、0.06mmol)をDMF2.0mLに溶解させ、室温で一晩撹拌した。撹拌開始12時間後、得られた黄濁液を、氷水に加え、酢酸エチルで抽出した。この有機抽出物を、飽和塩化アンモニウム溶液および水で洗浄し、硫酸ナトリウムを用いて乾燥させた。溶媒留去後、残渣をシリカゲルカラムクロマトグラフィーにより精製し、下記化学式(15)に示す6-アミノ-2-クロロ-9-[トランス-トランス-2,3-ビス(トリフェニルメトキシメチル)シクロブチル]プリンの白い結晶(273.9mg、収率59%)を得た。なお、下記化学式(15)において、Trは、トリチル基である。
前記6-アミノ-2-クロロ-9-[トランス-トランス-2,3-ビス(トリフェニルメトキシメチル)シクロブチル]プリン(92.2mg、0.12mmol)およびチオメトキシドナトリウム(84.1mg、1.20mmol)をDMF3.1mLに溶解し、110℃で3.5時間加熱した。加熱終了後、減圧下で溶媒を留去し、得られた残渣を酢酸エチルで抽出した。得られた有機抽出物を、水および飽和塩化ナトリウム溶液で洗浄し、硫酸ナトリウムを用いて乾燥後、減圧下で溶媒を留去した。得られた残渣を、シリカゲルカラムクロマトグラフィー(エチルアセテート:ヘキサン=1:1)により精製し、下記化学式(16)に示す6-アミノ-2-チオメトキシ-9-[トランス-トランス-2,3-ビス(トリフェニルメトキシメチル)シクロブチル]プリンの黄色味を帯びた結晶(66.3mg、収率71%)を得た。なお、下記化学式(16)において、SMeは、メチルチオ基であり、Trは、トリチル基である。
前記6-アミノ-2-チオメトキシ-9-[トランス-トランス-2,3-ビス(トリフェニルメトキシメチル)シクロブチル]プリン(65.0mg、0.08mmol)を、ギ酸1.7mLおよびジエチルエーテル1.7mLの混合液に溶解し、室温で15分間撹拌した。撹拌終了後、この混合液を酢酸エチルに溶解し、抽出した。得られた有機抽出物を、飽和炭酸水素ナトリウム溶液および飽和塩化ナトリウム溶液で洗浄し、硫酸ナトリウムを用いて乾燥させた。有機溶媒留去後、残渣を薄層クロマトグラフィー(ジクロロメタン:エタノール=5:1)により精製し、前記化学式(9)に示す2-SMe-C.OXT-A(6-アミノ-2-チオメトキシ-9-[トランス-トランス-2,3-ビス(ヒドロキシメチル)シクロブチル]プリン)の黄色味を帯びた結晶(9.4mg、収率38%)を得た。以下に、この化合物の物性値を示す。
本例では、実施例1で用いた2-Cl-C.OXT-Aが、ERK、Akt、JNKおよびp38の酵素の活性化(リン酸化)に与える影響を調べた。なお、これらのタンパク質は、前記ERKは、内皮細胞における最も代表的な血管新生促進系タンパク質リン酸化酵素の一つである。前記Aktは、ERK経路と並ぶ、内皮細胞における最も代表的な血管新生促進系タンパク質リン酸化酵素の一つである。また、JNKおよびp38は、ERK経路以外の、代表的なMAPキナーゼタンパク質リン酸化酵素である。

本例では、実施例1の2-Cl-C.OXT-Aおよび比較例2のC.OXT-Gが、ERK活性化(リン酸化)に与える影響を調べた。本例では、2-Cl-C.OXT-Aを所定濃度(0、10および100μmol/L)で添加した培地と、100μmol/L C.OXT-Gを添加した培地とを用い、添加後の培養時間を 20分とし、1次抗体として、抗phospho ERKポリクローナル抗体(カタログ番号9101、Cell Signaling Technology社製)および抗総ERKモノクローナル抗体(カタログ番号610408、BD Transduction Laboratory社製)のみを用い、ERKの2次抗体として、ヒツジHRP標識抗マウスIgG抗体(Pierce社製)を用い、pERKの2次抗体として、ヒツジHRP標識抗ラビットIgG抗体(Pierce社製)を用いた以外は、実施例3と同様にして、各酵素量を測定し、リン酸化型酵素相対値(倍)を算出した。
本例では、実施例1で用いた2-Cl-C.OXT-Aが、MEKのリン酸化に与える影響を調べた。なお、前記MEKは、実施例3において測定したERKの上流側に位置する、血管新生のシグナル伝達経路のタンパク質リン酸化酵素であり、血管新生促進に作用する。本例では、2-Cl-C.OXT-A添加後の培養時間を、所定時間(0、5、10、15および20分)とし、1次抗体として、抗phospho MEKポリクローナル抗体(カタログ番号9121、CellSignaling Technology社製)および抗総MEKポリクローナル抗体(カタログ番号9122、Cell Signaling Technology社製)を用い、2次抗体として、ヒツジHRP標識抗ラビットIgG抗体(Pierce社製)を用いた以外は、実施例3と同様にして、各酵素量を測定し、リン酸化型酵素相対値(倍)を算出した。
本例では、MEK阻害剤であるPD98059が、実施例1で用いた2-Cl-C.OXT-AによるERK活性化(リン酸化)に与える影響を調べた。本例では、2v/v%非働化ウシ胎児血清(FBS)を添加したHuMedia-EB2培地(クラボウ社製)に、下記表1に示す濃度で2-Cl-C.OXT-AおよびPD98059を添加した培地No.1~4を用いた以外は、実施例4と同様にして、各酵素量を測定し、リン酸化型酵素相対値(倍)を算出した。

本例では、VEGFR阻害剤であるSU5416が、実施例1で用いた2-Cl-C.OXT-Aによる管腔形成に与える影響を調べた。

本例では、理化学研究所バイオリソースセンターより入手した、神経細胞モデル細胞であるラット副腎髄質由来株化細胞PC12を用いて、実施例1の2-Cl-C.OXT-Aが、PC12細胞の分化に与える影響を調べた。
Fisher Scientific社製)を用いて、前記試料溶液のタンパク質濃度(D)を測定した。前記吸光度増加量(C)およびタンパク質濃度(D)を、下記式(IV)に代入して、AChE活性を算出した。なお、1Uは、1分あたりに1μmolの反応生成物を産生する酵素量(μmol/分)である。そして、AChEの測定値を下記式(V)に代入し、AChE活性相対値(倍)を算出した。

図15(A)~(D)に、PC12細胞の顕微鏡写真を示す。図15(A)は、PBS、図15(B)は、NGF(神経成長因子、ポジティブコントロール)、図15(C)は、2-Cl-C.OXT-A 50μmol/L、図15(D)は、2-Cl-C.OXT-A 100μmol/Lを添加して培養した細胞の写真である。図15(A)に示すバーの長さは、100μmである。図15において、線状の軸策が伸長した細胞は、PC12細胞が神経細胞様に分化した細胞である。図15(A)に示すように、コントロールでは、線状の軸策が伸長した細胞はほぼ観察されなかった。これに対して、図15(C)および(D)に示すように、図15(B)のNGFと同様に、2-Cl-C.OXT-A添加により、線状の軸策が伸長した細胞が観察された。
図16に、AChE(アセチルコリンエステラーゼ)活性の測定結果を示す。AChE活性はPC12細胞の分化のマーカーである。図16において、縦軸は、AChE活性相対値であり、各バーは、左から順に、PBS、NGF(ポジティブコントロール)、2-Cl-C.OXT-A 50μmol/Lおよび2-Cl-C.OXT-A 100μmol/Lの結果である。図16に示すように、2-Cl-C.OXT-Aにより、NGFと同様に、AChE酵素活性が増加した。このように、2-Cl-C.OXT-Aによる神経細胞様への分化促進活性が示され、神経細胞成長促進活性が確認された。
本例では、ウサギ角膜法による、実施例1の2-Cl-C.OXT-Aが、血管新生に与える影響を調べた。
下記化学式(17)に示す9-[トランス-トランス-2,3-ビス(アセチルオキシメチル)シクロブチル]グアニンについて、実施例1と同様にして、血管内皮細胞の細胞増殖、管腔形成および細胞遊走性への影響を調べた。その結果、下記化学式(17)の化合物は、血管内皮細胞の細胞増殖促進活性、管腔形成促進活性および細胞遊走性促進活性を示した。
本例では、下記化学式(18)に示す8-ブロモ-9-[トランス-トランス-2,3-ビス(アセチルオキシメチル)シクロブチル]グアニンを合成した。
下記化学式(19)で表される8-ブロモ-9-[トランス-トランス-2,3-ビス(ヒドロキシメチル)シクロブチル]グアニンを合成した。
下記化学式(20)で表される2-アミノ-6,8-ジクロル-9-[トランス-トランス-2,3-ビス(アセチルオキシメチル)シクロブチル]プリンを合成した。
下記化学式(21)で表される2-アミノ-6,8-ジメトキシ-9-[トランス-トランス-2,3-ビス(ヒドロキシメチル)シクロブチル]プリンを合成した。
Claims (14)
- 下記一般式(1)で表されるシクロブチルプリン誘導体、その互変異性体もしくは立体異性体、またはそれらの塩、溶媒和物もしくは水和物。
X1は、ハロゲノ基、アルキル基、アルキルチオ基、チオ基(チオール基)、アミノ基、ヒドロキシ基、アルコキシ基、アルキニル基またはシアノ基であり、
X2は、ハロゲノ基、アミノ基、ヒドロキシ基、アルコキシ基、チオ基(チオール基)またはアルキルチオ基であり、
X3は、水素原子、ハロゲノ基またはアルコキシ基であり、
R1およびR2は、同一であるかまたは異なり、それぞれ、水素原子、ハロゲノ基、カルボキシル基、アルキル基、アシル基、カルバモイル基、アシルオキシ基、ヒドロキシアルキル基、アシルオキシアルキル基、アルコキシアルキル基、ハロアルキル基またはホスホノオキシアルキル基であり、
X1がアミノ基である場合は、
X2およびX3がともにハロゲノ基であるか、
X2およびX3がともにアルコキシ基であるか、または、
X2がヒドロキシ基であり、X3がハロゲノ基であり、かつ、R1およびR2がともにアシルオキシアルキル基であり、
前記X1、X2、X3、R1およびR2において、前記アルキル基、前記アルキルチオ基、前記チオ基(チオール基)、前記ヒドロキシ基、前記アルコキシ基、前記アルキニル基、前記アミノ基、前記カルボキシル基、前記アシル基、前記カルバモイル基、前記アシルオキシ基、前記ヒドロキシアルキル基、前記アシルオキシアルキル基、前記アルコキシアルキル基および前記ホスホノオキシアルキル基は、それぞれの一つ以上の水素原子が任意の置換基に置換されていてもよい。 - 前記一般式(1)において、前記X1が、クロロ基である請求の範囲1記載のシクロブチルプリン誘導体、その互変異性体もしくは立体異性体、またはそれらの塩、溶媒和物もしくは水和物。
- 前記一般式(1)において、前記X2が、アミノ基である請求の範囲1記載のシクロブチルプリン誘導体、その互変異性体もしくは立体異性体、またはそれらの塩、溶媒和物もしくは水和物。
- 前記一般式(1)において、前記R1およびR2が、ヒドロキシメチル基である請求の範囲1記載のシクロブチルプリン誘導体、その互変異性体もしくは立体異性体、またはそれらの塩、溶媒和物もしくは水和物。
- 前記一般式(1)において、前記X1が、クロロ基またはチオメトキシ基であり、前記X2が、アミノ基であり、前記R1およびR2が、ヒドロキシメチル基である、請求の範囲1記載のシクロブチルプリン誘導体、その互変異性体もしくは立体異性体、またはそれらの塩、溶媒和物もしくは水和物。
- 6-アミノ-2-クロロ-9-[トランス-トランス-2,3-ビス(ヒドロキシメチル)シクロブチル]プリンもしくは6-アミノ-2-チオメトキシ-9-[トランス-トランス-2,3-ビス(ヒドロキシメチル)シクロブチル]プリン、その互変異性体もしくは立体異性体、またはそれらの塩、溶媒和物もしくは水和物である請求の範囲1記載のシクロブチルプリン誘導体、その互変異性体もしくは立体異性体、またはそれらの塩、溶媒和物もしくは水和物。
- 下記一般式(1′)で表されるシクロブチルプリン誘導体、その互変異性体もしくは立体異性体、またはそれらの塩、溶媒和物もしくは水和物を含み、血管新生促進機能、管腔形成促進機能および神経細胞成長促進機能からなる群から選択される少なくとも一つの機能を有する促進剤。
X1′は、ハロゲノ基、アルキル基、アルキルチオ基、チオ基(チオール基)、アミノ基、ヒドロキシ基、アルコキシ基、アルキニル基またはシアノ基であり、
X2′は、ハロゲノ基、アミノ基、ヒドロキシ基、アルコキシ基、チオ基(チオール基)またはアルキルチオ基であり、
X3′は、水素原子、ハロゲノ基、アルキル基、アルキルチオ基、アミノ基、ヒドロキシ基、アルコキシ基、ヒドロキシフェニル基またはカルバモイル基であり、
R1′およびR2′は、同一であるかまたは異なり、それぞれ、水素原子、ハロゲノ基、カルボキシル基、アルキル基、アシル基、カルバモイル基、アシルオキシ基、ヒドロキシアルキル基、アシルオキシアルキル基、アルコキシアルキル基、ハロアルキル基またはホスホノオキシアルキル基であり、
X1′がアミノ基かつX3′が水素原子である場合は、R1′およびR2′は、ヒドロキシアルキル基以外の原子または置換基であり、
前記X1′、X2′、X3′、R1′およびR2′において、前記アルキル基、前記アルキルチオ基、前記チオ基(チオール基)、前記ヒドロキシ基、前記アルコキシ基、前記アルキニル基、前記アミノ基、前記ヒドロキシフェニル基、前記カルバモイル基、前記カルボキシル基、前記アシル基、前記アシルオキシ基、前記ヒドロキシアルキル基、前記アシルオキシアルキル基、前記アルコキシアルキル基および前記ホスホノオキシアルキル基は、それぞれの一つ以上の水素原子が任意の置換基に置換されていてもよい。 - 前記一般式(1′)において、前記X1′が、クロロ基である、請求の範囲7記載の促進剤。
- 前記一般式(1′)において、前記X2′が、アミノ基である、請求の範囲7記載の促進剤。
- 前記一般式(1′)において、前記R1′およびR2′が、ヒドロキシメチル基である、請求の範囲7記載の促進剤。
- 前記一般式(1′)において、前記X1′が、クロロ基またはチオメトキシ基であり、前記X2′が、アミノ基であり、前記X3′が、水素原子であり、前記R1′およびR2′が、ヒドロキシメチル基である、請求の範囲7記載の促進剤。
- 請求の範囲6記載のシクロブチルプリン誘導体、その互変異性体もしくは立体異性体、またはそれらの塩、溶媒和物もしくは水和物を含み、血管新生促進機能、管腔形成促進機能および神経細胞成長促進機能からなる群から選択される少なくとも一つの機能を有する促進剤。
- 請求の範囲1記載のシクロブチルプリン誘導体、その互変異性体および立体異性体、およびそれらの塩、溶媒和物および水和物、ならびに請求の範囲7記載の促進剤からなる群から選択される少なくとも一つを含み、血管新生促進用、管腔形成促進用および神経細胞成長促進用からなる群から選択される少なくとも一つの用途を有する医薬品。
- 創傷治癒薬、アルツハイマー治療薬、アルツハイマー予防薬、梗塞性疾患治療薬、梗塞性疾患予防薬および育毛剤からなる群から選択される少なくとも一つである請求の範囲13記載の医薬品。
