Description
"Vorrichtung zur Elektrosprayionisierung einer flüssigen Probe""Apparatus for electrospray ionization of a liquid sample"
The invention relates to a device for electrospray ionization of a liquid sample having a capillary-shaped supply channel having an electrode for the liquid sample and a provided with at least one passage opening for the formed ion beam plate which is spaced to form a Ionenbildungsfrei- space spaced from the free end of the feed channel in which a high potential difference is applied between the electrode of the feed channel and the plate.
Im elektrischen Feld werden die Ionen bei Atmosphärendruck in die Gasphase transferiert, wobei sich dieser Prozess in vier Schritte unterteilt :Electrospray ionization is the dispersion of a liquid into many small charged droplets with the help of an electric field. In the electric field, the ions are transferred to the gas phase at atmospheric pressure, whereby this process is divided into four steps:
1. Die Bildung kleiner geladener Elektrolyt-Tröpfchen;1. The formation of small charged electrolyte droplets;
2. kontinuierlicher Lösungsmittelverlust dieser Tröpfchen durch Verdampfen, wobei die Ladungsdichte an der Tröpfchenoberfläche zunimmt;2. continuous solvent loss of these droplets by evaporation, whereby the charge density at the droplet surface increases;
Repeated spontaneous decay of the droplets in micro droplets (Coulomb explosions) and finally
Desolvation of the analyte molecules during transfer into an analysis device, for example a mass spectrometer.
For the detection of, for example, positively charged ions, the electrospray ionization (ESI) process begins with the continuous delivery of a dissolved analyte to the tip of a conductive capillary. The applied electric field between the capillary tip and the mass spectrometer inlet also penetrates the analyte solution. The positive ions are attracted to the liquid surface. Accordingly, the negative ions are pushed in the opposite direction until the electric field within the liquid is canceled by the redistribution of negative and positive ions. This suppresses other possible forms than soft ionization, such as ionization by removal of an electron from the analyte molecule (field ionization), which would require very high electric fields.
The positive ions accumulated on the liquid surface are further drawn toward the cathode. This creates a characteristic cone of liquid (Taylor cone), because the surface tension of the liquid counteracts the electric field. At a sufficiently high electric field (potential difference about 3 kV), the cone is stable and emits from its tip a continuous filamentous liquid flow of a few micrometers in diameter. This becomes unstable some distance from the anode and breaks up into tiny, strung droplets. The surface of the droplets is enriched with positive charges that no longer have any negative counterions, resulting in a net positive charge.
The charge in the droplets is due to the electrophoretic separation of the ions. The positive ions (as well as after polarity reversal of the field, the negative ions) observed in the spectrum are always the ions already present in the (electrolyte) solution. Additional ions are only observed at very high voltages when electrical (corona) discharges occur between the capillary tip and the mass spectrometer.
Bekannte Vorrichtungen zur Elektrosprayionisierung weisen
als Zuführkanal für die flüssige Probe eine elektrisch kontaktierte Kapillare auf, an welche ein Potential angelegt wird, d.h. die Kapillarspitze selbst bildet die Elektrode .Known devices for electrospray ionization have as an inlet channel for the liquid sample, an electrically contacted capillary to which a potential is applied, ie the capillary tip itself forms the electrode.
Alternatively, it is known to integrate the required capillary feed channel into a microchip. A special solution of this type is described for example in DE 199 47 496 C2.
Probe direkt in Verbindung gebracht. Both in devices with capillaries and in devices consisting of a microchip, the electrode is directly connected to the liquid or sample to be analyzed. As a result, the lifetime of these devices is severely limited, since the electrode inevitably corrodes so strongly after a certain period of use that it is no longer usable. In addition, in these known devices, the maximum applicable potential difference is limited, otherwise unwanted corona discharges arise.
Aus der Veröffentlichung "BANKS J. F. : Recent advances in capillary electrophoresis/electrospray/mass spectrometry. In: Electrophoresis.
spray. In: Anal. Chera. 1997, Vol. 69, S. 2955-2962." bekannt. From the publication "BANKS JF: Recent advances in capillary electrophoresis / electrospray / mass spectrometry." In: Electrophoresis, 1997, Vol. 18, pp. 2255-2266. " a generic device for Elektrosprayionisie- tion is known, in which in one embodiment, a dielectric separation layer is disposed between the feed channel and the electrode. The electrode is arranged on the outside of the dielectric separation layer and extends to the tip or to the outlet of the feed channel, so that there is a direct contact between the electrode and the liquid sample. A similar device is known from the publication "SZOSTEK B. et al .: Coupling Condensation Nucleation Light Scattering Detection with Capillary Electrophoresis Using Electro- spray. In: Anal. Chera. 1997, Vol. 69, pp. 2955-2962. "In this device as well, the electrode extends as far as the tip or the outlet of the feed channel, so that there is direct contact between the liquid sample and the electrode. These two known devices thus likewise have the above-described disadvantages of the prior art Further devices for electrospray ionization are known from WO 01/50499 A1 and DE 44 08 032 A1.
The object of the invention is to develop a generic device so that increases their life and also the creation of higher potential differences without corona discharges is possible.
This object is achieved with a device of the type described in the present invention, that between the electrode and the feed channel a direct contact between the liquid sample and electrode preventing separation layer is disposed of a dielectric material, wherein the electrode at a distance from the free end of the feed channel is arranged.
Electrospray ionization is thus made possible by contactless application of a potential, since the electrode of the feed channel has no direct contact with the sample liquid by dielectrically coupling the electric field. The electric field is transmitted through a dielectric shift of the charges through the channel walls, without affecting the functional mechanism, which is due to the fact that the liquid sample is not at rest, but flows through the feed channel. Since there is no direct contact of the electrodes with the sample liquid, corrosion of the electrodes is completed avoided, so that significantly increases the life of the device. Furthermore, it has been found that with such a device much higher voltages or potential differences can be applied without igniting a corona discharge. Thus, potential differences in the order of 6 to 8 kV can be realized. It has surprisingly been found that, unlike previously assumed, the electrode does not have to be arranged in the end region of the feed channel, this can also be arranged clearly spaced from the free end, without affecting the functionality. As a result, the structure of the device is much more flexible.
In a particularly preferred embodiment, it is provided that the channel wall of the feed channel itself consists of a dielectric material and is surrounded on the outside by the electrode. Thus, the channel wall of the feed channel made of glass or plastic.
According to a first embodiment, it is provided that the supply channel is formed by a microfluidic microchip, the electrode / n is / are then attached to the outside of the microchip.
Alternatively, the feed channel can also be formed by a tube, which preferably consists of glass, which is externally coated ring-shaped with an electrode (preferably made of gold).
Diese zeigt in:The invention is explained in more detail below by way of example with reference to the drawing. This shows in:
Fig. 2 eine erste Ausführungsform des wesentlichen Teils der Vorrichtung in Draufsicht,1 is a schematic representation of an apparatus for electrospray ionization, 2 shows a first embodiment of the essential part of the device in plan view,
Fig. 3 eine Seitenansicht auf Figur 2 undFig. 3 is a side view of Figure 2 and
Fig. 4 shows the essential part of a device according to a second embodiment.
Der kapillarförmige Zuführkanal 1 ist im Bereich seines vorderen freien Endes Ia, aber beabstandet von diesem, von einer Elektrode 3 umgeben.FIG. 1 generally shows an apparatus for electrospray ionization, which first comprises a capillary feed channel 1, generally designated 1, in which a liquid sample, i. a dissolved analyte is supplied continuously, which is indicated by the arrow 2. The capillary feed channel 1 is surrounded by an electrode 3 in the region of its front free end 1a, but spaced from it.
Spaced to the free end Ia of the feed channel 1 is a plate 4 with a passage opening 5 is arranged to form a free space for ion formation, which allows the passage of the indicated with an arrow 6 ion beam in a not shown mass spectrometer or a similar analyzer. Between the electrode 3 and the second electrode forming plate 4, a potential difference ΔP of about 3 kV is applied.
Wesentlich ist nun die Ausbildung des Zuführkanals 1 mit der Elektrode 3.The formation of the feed channel 1 with the electrode 3 is essential.
A first preferred embodiment is shown in FIGS. 2 and 3. The feed channel 1 is integrated in this embodiment in a microfluidic microchip 7, which on both sides of the feed channel 1 two Has layers 8 of dielectric material, for example plastic film. It can also be provided only a layer 8. The front free end of the microchip 7 is formed nozzle-shaped, this area is denoted by 7a.
The electrode 3 is arranged on the feed channel 1 such that there can be no contact between the electrode 3 and the liquid sample 2 flowing through the feed channel 1. For this purpose, in the embodiment according to FIGS. 2 and 3, a bipartite electrode 3 is arranged on the upper and lower sides on the outside of the two layers 8 of dielectric material.
Thus, when the liquid sample flows through the passageway 1, the electric field is transmitted through a dielectric shift of the charges through the channel walls (layers 8) of the microchip 7. Thus, an electrospray ion beam 6 is generated without the electrodes 3 coming into contact with the liquid.
Unlike in FIGS. 2 and 3, the electrode 3 can also be arranged at a considerable distance from the front free end 1a of the feed channel 1.
An alternative embodiment is shown in FIG. The feed channel 1 is surrounded or formed in this embodiment by a tube 9, for example made of glass. This tube 9, which thus consists of a dielectric material, is spaced from the free end 1a of the feed channel 1 enclosed by an annular electrode 3, which consists for example of gold and is preferably formed as a coating on the outside of the tube 9. Of course, the invention is not limited to the illustrated embodiments. Further embodiments are possible without departing from the basic idea. Thus, the feed channel 1 may of course also have a different shape, it is essential that the at least one electrode 3 is separated from the feed channel 1 by a dielectric separation layer so that the electrode 3 can not get into contact with the liquid sample.
