Abstract
Description
The invention relates to colloidal systems and solutions. Micellar complexes obtained by the proposed method can be used in the formulation of cosmetic prophylactic agents with a fungicidal and antiseptic effect, in the preparation of compositions with a fungicidal and antiseptic effect for use in medicine, veterinary medicine and construction.
At present, micellar complexes are of considerable scientific interest. Due to its properties, these structures are used in many fields: food, pharmaceutical, oil, and cosmetics. The structure of micelles allows the active component to be placed inside the shell, stabilizing it and protecting it from environmental factors, and, in addition, due to its size and structure similar to the structure of biological liposome objects, it facilitates penetration through cell membranes, which leads to a more intense effect. Obtaining micelle stable complexes of copper cations will improve the known antiseptic and fungicidal properties of copper-based compositions.
A number of works are known in which the preparation of fungicidal and antiseptic preparations based on copper is considered, as well as the placement of active cosmetic components in the micellar membrane.
A known method of combating bacterial-fungal infections (RF Patent for the invention No. 2094988, priority date: 04/02/1993). Copper-containing compositions are generally not stable. In this method, a protected object is coated with a copper-containing composition consisting of an aqueous solution of a copper complex and 0.2-80 wt. % partially neutralized water-soluble carboxyl-containing polymer. The resulting preparation is referred to as composite compounds, stabilization of copper hydroxide is carried out due to interaction with polycarboxylic acids. The use of this method is limited to agriculture.
Also known composition with bactericidal properties (RF Patent for the invention No. 21868100, priority date: 07/20/2000). Coatings with bactericidal properties considered in the patent are mainly used for the manufacture of paints and varnishes, film formers, impregnations, dry mixes that can be used in construction. In this case, nanostructured metal particles, including copper, which are introduced into the composition of the composite material for surface treatment, are used as a metal-containing bactericidal component. The disadvantages of this invention include the high cost of metal nanoparticles and their high consumption when introduced into the composition.
When considering works related to the development of drugs and prophylactic drugs that can be used in medicine, a list of antiseptic compositions using copper and silver metal particles in the form of colloidal or nanoparticles in which these metals are null-valent is highlighted.
For example, a patent of the Russian Federation for invention No. 2429820 “Antiseptic ointment for external use (2 variants)” is known (priority date: September 24, 2009). Bentonite pots with intercalated silver or copper cations, which are obtained by modifying the inorganic salts of these metals, are dispersed into a petroleum jelly or a mixture of petrolatum and lanolin. This composition has limitations on the method of application and is used only in the form of ointments.
In the technical solution according to the RF patent for the invention No. 2331407 “Gel formulation (options)” (priority date: 09/09/2006), a gel in the form of a liposomal form of water-soluble substances with an antiseptic agent based on electrolytic silver is proposed to increase the activity of the drug. Additional energy costs during the electrolytic process and the use in the form of a gel limit its use.
The most interesting to use are aqueous solutions containing copper in various forms, since they expand the scope. The use of aqueous solutions involves additional stabilization of metal particles, no matter in what form they are.
So, in the RF Patent for the invention No. 2574400 “Imidazolmalate of copper (II), showing antibacterial activity, and a method for its preparation” (priority date: 03/20/2015) receive coordination compounds of metals, namely copper, showing antibacterial activity. The invention can be used in medicine and veterinary medicine for chemotherapy purposes. The disadvantage of this invention is that the synthesis is carried out in several stages and seems to be time-consuming. In addition, according to the description, the obtained compound has good antiseptic properties, but there is no information about the stability and preservation of activity after some time.
Known methods for producing organic salts of copper with organic acids. In the patent of the Russian Federation for invention No. 2256648 “Method for the preparation of copper (II) salts with dicarboxylic acids” (priority date: 02/27/2004), tartrates, succinates and some other copper salts are obtained from solutions of inorganic copper salts by reaction with dicarboxylic acids. In the work “Compounds of leucine with cations of copper (II) in aqueous solutions” (Bondareva LP, Rusina EV, Ovsyannikova DV Compounds of leucine with cations of copper (II) in aqueous solutions // Bulletin of Voronezh State University engineering technologies. 2017. No. 4 (74), pp. 175-180) receive coordination compounds of copper with amino acids, in particular with leucine. The obtained salts are readily soluble in water, exhibit antibacterial properties, but with prolonged storage, the probability of hydrolysis is high.
Also known are works in which surfactants were used as the micellar shell, and active components, for example, vitamins, were encapsulated inside the structure.
Closest to the proposed invention in terms of essential features is the RF Patent for invention No. 2303036 “Solubilizate from ascorbic acid and an emulsifier, a mixture of solubilizates, a method for the manufacture of solubilizate and its use (options)” (priority date: May 29, 2003), including obtaining stable solubilizates due to the addition of polysorbate in high concentration. In the resulting solution, the presence of micelles with a size of 100 nm was recorded. This composition has increased biological activity and is used as biologically active additives and antioxidants. The disadvantages of the invention include the lack of information about the possibility of inclusion in the micelle of metal particles in one form or another.
The objective of the present invention is to develop a method for producing a stable micellar complex based on copper (II) cations stabilized by a two-layer shell of nonionic surfactants, which will significantly increase the sedimentation stability and light fastness of the obtained atiseptic and fungicidal compositions.
This problem is solved by selecting a surfactant capable of complexing with a copper (II) cation and an auxiliary surfactant (co-surfactant) that stabilizes the complex. The ratio of the components for effective micelle formation is determined through the critical micelle concentration (CMC) of the surfactant mixture in the presence of a copper salt. The stability of the complex (K mouth ) was determined spectrophotometrically, and the change in the particle size distribution in the obtained colloidal system by dynamic light scattering in time.
When solving this problem, a technical result is created, which consists in obtaining micellar complexes of copper (II) using a non-ionic surfactant complexing agent and stabilizing co-surfactant with high stability, sedimentation stability and biological (antiseptic and fungicidal) activity.
K=99,45%.The method was carried out as follows. Decyl glucoside was chosen as a surfactant complexing agent, and Tween-80 co-surfactant was chosen as a stabilizer. Both substances are nonionic and are widely used in cosmetic formulations. A fixed volume of double-distilled water or water for laboratory analysis (GOST R 52501-2005) is added to the calculated mass of decyl glucoside at room temperature and constant stirring on a magnetic stirrer (manual stirring is allowed) until a clear solution is formed. Then take the calculated sample of copper acetate (PSA) and place in the resulting surfactant solution, while the solution takes on an intense blue color. Next, the calculated sample of Tween-80 and bidistilled water are successively added to the resulting solution to the required mass of the solution. In this case, the solution becomes almost colorless. After that, the container with the solution is closed and placed in a dark place for at least one day. Samples are calculated on the basis of experimentally determined CMC values of a mixture of surfactants taken in conjunction with copper acetate. CMC was determined by viscometric and refractometric methods. Stable micellar complexes are formed with the following mass fractions of components: 2% decyl glucoside, 4% tween-80, 0.1% copper acetate. The calculated degree of micellation during spectrophotometric determination of residual copper was K = 99.45%.
The stability of the systems was controlled by measuring the size of micelles in the resulting colloidal system by dynamic light scattering.
Table 1 - Particle size distribution and change over time
Table 1 shows that the obtained colloidal system contains particles of three types. Particles with a size of 300-400 nm are micellar complexes containing copper (II). When analyzing a surfactant mixture without the addition of a copper (II) salt, such particles are absent. Within 6 months, no significant changes in the particle size distribution have occurred. Additionally, the temperature was determined by conductometry at which the complex was destroyed, and which amounted to 47 ° C.
12038-84.Biological activity was checked using a microbiological test system, as well as in accordance with the procedure specified in GOST 12038-84.

Thus, the use of the proposed method for producing stable micellar complexes of copper (II) allows to obtain colloidal systems with thermal stability up to 45 ° C, stability for at least a year when stored in an unlit place and at least six months when stored in the light, and increased biological activity compared with unmicellated forms.
