При решении поставленной задачи получен положительный эффект за счет замены губчатой кости на компактную. Компактная кость структурно более монолитна и обладает большим запасом прочности. При вертебрологических операциях, когда необходимо резецировать тело позвонка или осуществить расклинивающий корпородез, данный трансплантат может с успехом использоваться как альтернатива металлическим "кейджам". Предлагаемый костный трансплантат с течением времени полностью биодеградирует и замещается полноценной костной тканью, а так как процесс биодеградации протекает с интенсивностью 4% в год, сохраняется надежная фиксация. К преимуществам использования предлагаемого трансплантата следует отнести и тот факт, что всегда можно осуществить контрольное МРТ-исследование, что невозможно при использовании металлоимплантатов. Поставленная задача решается за счет того, что аллогенные или ксеногенные фрагменты длинных трубчатых костей депротеинизируют в растворе химопсина в течение 96 ч, а при обработке 10% раствором перекиси водорода их помещают в переменное магнитное поле при 45oC. Способ осуществляется следующим образом. Аллогенные или ксеногенные фрагменты диафизов длинных трубчатых костей освобождают от мягких тканей и костного мозга. Трубку распиливают на 2-3 части не менее 6 см длиной или распиливают на фрагменты размером 10•0,2•0,5 см. Подвергают механической очистке и промывают холодной проточной водой в течение 4 ч, затем осуществляют депротеинизацию, помещая костные фрагменты в 0,01% раствор химопсина, и выдерживают в термостате при 37,5oС в течение 96 ч. Затем материал промывают холодной проточной водой в течение 40 мин. Далее материал заливают 10% раствором перекиси водорода и помещают в переменное магнитное поле с подогревом до 37,5oС на 48 ч (за этот период осуществляют 4-5-кратную смену раствора перекиси водорода). Критерием для смены раствора является обильное выпадение осадка и помутнение раствора (плавающие частицы тормозят дальнейшее очищение кортикальных фрагментов от продуктов деструкции белков). При величине фрагментов 5-6 см процесс депротеинизации проводят дважды (обработка 0,01% раствором химопсина и 10% раствором перекиси водорода). В случае депротеинизации крупных фрагментов (более 6 см) - трижды. Затем фрагменты компактной кости промывают проточной водой 4 ч. Контроль первичный осуществляется в луче света, костная ткань равномерно белая, полупрозрачная без очагов затемнений. Морфологический контроль осуществляется на срединном распиле фрагмента под увеличением х3 диоптрии. Отсутствие элементов костного мозга свидетельствует о полной депротеинизации костной ткани. Далее материал помещают в жидкий эфир на 6 ч, с трехкратной сменой раствора (каждые 2 ч), при комнатной температуре. Затем высушивают в стерильном боксе под вентилятором 15 мин, после чего заливают 10% раствором хлористого лития на 16 ч при 45oС, промывают стерильной дистиллированной водой 5 мин.The method is as follows. In the case of deproteinization of large fragments (more than 6 cm) - three times. Then the fragments of the compact bone are washed with running water for 4 hours. The primary control is carried out in a ray of light, the bone tissue is uniformly white, translucent without foci of dimming. Morphological control is carried out at the midline fragment cut under an increase of x3 diopter. The absence of bone marrow elements indicates complete deproteinization of bone tissue. Next, the material is placed in liquid ether for 6 hours, with a three-time change of solution (every 2 hours), at room temperature. Then it is dried in a sterile box under a fan for 15 minutes, after which it is poured with a 10% lithium chloride solution for 16 hours at 45 ° C, washed with sterile distilled water for 5 minutes.
