Description
Foreliggende oppfinnelse angår nye kjemiske forbindelser med verdifull farmasøytisk aktivitet. Oppfin-nelsenangår i særdeleshét forbindelser som utviser hyper-tensiv og angiotensin omdannende enzym-inhiberende aktivitet og med generell formel
hvori R og Rg uavhengig er hydroxy, lavere alkoxy, lavere alkenoxy, di (lavere alkyl)amino-lavere alkoxy, hydroxy-lavere alkoxy, acylamino-lavere alkoxy, acyloxy-lavere alkoxy, aryloxy, aryloxyl-lavere alkoxy, amino, lavere alkylamino, di-lavere alkylamino, hydroxyamino eller aryl-
lavere alkylamino; R^, R2, R3, R 4. R5, Ry og Rg er uavhengig hydrogen, alkyl, alkenyl eller alkynyl inneholdende opp til 20 carbonatomer, aryl eller aryl-lavere alkyl med fra 7 til
12 carbonatomer, heterocyclisk gruppe eller heterocyclisk
gruppe-lavere alkyl med fra 6 til 12 carbonatomer, cycloalkyl eller cycloalkyl-alkyl inneholdende opp til 20 carbonatomer i cycloalkylgruppen; R2 og R^.kan sammen med carbon og nitrogenatomet til hvilke de er bundet, og R^ og R^kan sammen med nitrogenatomet og carbonatomet til hvilke de er bundetrdanne en N-heterocyclisk gruppe inneholdende fra 3 til 5 carbonatomer eller 2 til 4 carbonatomer og et svovelatom; Rg er cycloalkyl, polycycloalkyl, partielt mettet cycloalkyl og polycycloalkyl, cycloalkyl-lavere alkyl, aryl, aryl-lavere alkyl, aryl-lavere alkenyl, aryl-lavere alkynyl; og salter derav med syrer eller baser, spesielt farmasøytisk akseptable syre- og basesalter.
Alkylgruppene i seg selv eller når de er til stede som substituenter, er fortrinnsvis lavere alkylgrupper inneholdende fra 1 til 6 carbonatomer og kan være rettkjedet eller forgrenet. Disse grupper innbefatter methyl, ethyl, propyl, iso-propyl, butyl, isobutyl, amyl, hexyl og lignende.
Alkenyl-og alkynylgruppene kan hver for seg eller
når de er tilstede som substituenter fortrinnsvis inneholde fra 2 til 6 carbonatomer og kan være.rettkjedet eller forgrenet. Disse grupper innbefatter vinyl, propenyl, allyl, isopropenyl, ethynyl og lignende.
Alkyl, alkenyl og alkynylgruppene kan bære substituenter slik som hydroxy, lavere alkoxy, thio, lavere alkylmercapto, amino, lavere alkylamino, di(lavere alkyl) amino, halogen og nitro.
Aryl-lavere alkylgruppene og heterocycliskgruppe-lavere alkylgrupper inneholder benzyl, fenethyl, nafthylmethyl, indolylethyl, indanylmethyl, indanylethyl og lignende.
Cycloalkylgruppene kan være mono eller polycycliske og inneholde fra tre til 20 carbonatomer. Disse grupper innbefatter cyclopropyl, cyclopentyl, cyclohexyl, cycloheptyl, bornyl, nor-bornyl, indanyl og lignende. Disse grupper kan være delvis umettet og bære substituenter slik som halogen, hydroxy, lavere alkyl, lavere alkoxy, amino lavere alkylamino, di(lavere alkyl) amino, thiol, lavere alkylmercapto, nitro og trifluormethyl.
Arylgruppene inneholder fra 6 til 10 carbonatomer
og innbefatter slike grupper som fenyl og a- eller (3-nafthyl og,kondensert fenyl-cycloalkyl slik som indanyl.
Aryl og aralkylgruppene kan bære en eller flere substituenter slik som lavere alkyl, lavere alkenyl, lavere alkynyl, hydroxy, lavere alkoxy, amino, lavere alkylamino, di (lavere alkyl)-amino, thiol, lavere alkylmercapto, hydroxy-lavere alkyl, amino-lavere alkyl, thio-lavere alkyl, nitro, halogen, trifluormethyl, methylen-dioxy, ureido eller guanidin.
Acylgruppene er fortrinnsvis lavere alkanoyl inneholdende fra 1 til 6 carbonatomer og benzoyl.
Halogenatomet kan være fluor, klor, brom og jod.
Egnede syreaddisjonssalter kan dannes fra uorganiske syrer slik som saltsyre, svovelsyre og fosforsyre, og organiske syrer slik som eddiksyre, melkesyre, sitronsyre, malin-syre, maleinsyre, fumarsyre, ravsyre, benzoesyre, hydroxy- benzoesyre, aminobenzoesyre, nicotinsyre, toluensulfonsyre og lignende.
Egnede basiske salter kan innbefatte saltene av
alkali og jordalkali metaller slik som natrium, lithium, kalium, magnesium og calcium, såvel som jern og salter av ammoniakk og aminer, og kvartære ammoniumsalter.
Forbindelsene ifølge oppfinnelsen kan inneholde en eller flere asymmetriske carbonatomer og kan eksistere i racemisk og optisk aktive former. Alle disse former betraktes j å være innen oppfinnelsens ramme. v
Forbindelsene ifølge oppfinnelsen fremstilles ved
en amiddannende reaksjon av en forbindelse av formel
med et acetylerende derivat av en syre av formel
under dannelse av denønskede forbindelse.
Alternativt omsettes et dipeptid av formel
med en a-ketosyre eller ester av struktur under dannelse av det tilsvarende imin, hvorpå iminet reduseres under dannelse av en forbindelse av formel I hvori R 2 og R 3 hver er H. Som en ytterligere variant kan de nye forbindelser fremstilles ved omsetning av en forbindelse.av formel med en a-haloforbindelse av formel
ved avspaltning av hydrogenhalogenid.
Den ovenfor angitte kondensasjonsreaksjon under dannelse av iminer med etterfølgende hydrogenering kan hensiktsmessig utføres i en enkel reaksjonssone ved hensiktsmessig å blande a-ketosyren eller derivatet med den reaktive forbindelse av formel IV under hydrogeneringsbetingelser. For praktiske formål kan de ovenfor angitte reaktanter hydro-generes over en edel metallkatalysator slik som palladium, platina, rhodium, ruthenium og lignende og de to trinn forløper under slike reaksjonsbetingelser at det ønskede sluttprodukt dannes.
Som i enhver organisk reaksjon kan løsningsmidler anvendes slik som methanol, ethanol, propanol, aceton, tetra hydrofuran, dioxan, dimethylformamid, diethylacetamid og lignende. Reaksjonen utføres normalt ved eller nær romtemperatur, selv om temperaturer fra 0°C opp til reaksjonsblan-dingens tilbakeløpstemperatur kan anvendes.
I den ovenfor angitte reaksjonsrekke er R - Rg som tidligere beskrevet og Hal er halogen. Fortrinnsvis er R og Rg hydrogen eller lavere alkyl, R2, R5, R-y og Rg er hydrogen,
R^og R^er lavere alkyl, R3er hydrogen og R^er cycloalkyl, aryl eller aralkyl.
De her angitte amiddannende betingelser innbefatter bruk av kjente derivater av de beskrevne syrer, slik som syrehalogenider, anhydrider, blandede anhydrider, lavere alkylestere, carbodiimider, carbonyldiimidazoler og lignende. Reaksjonene utføres i organiske løsningsmidler slik som acetonitril, tetrahydrofuran, dioxan, eddiksyre, methylenklorid, ethylenklorid og lignende løsningsmidler. Den amid-dannende reaksjon vil forløpe ved romtemperatur eller ved forhøyet temperatur. Anvendelse av forhøyet temperatur er hensiktsmessig idet dette muliggjør kortere reaksjonstider. Temperaturer varierende fra 0°C opp til reaksjonssystemets tilbakeløpstemperatur kan anvendes. Ytterligere hensiktsmessig kan den amiddannende reaksjon utføres i nærvær av en base slik som tertiære organiske aminer, for eksempel trimethyl-amin, pyridin, picolin og lignende, i særdeleshet hvor hydrogenhalogenid dannes ved den amiddannende reaksjon, for eksempel acylhalogenid og aminoforbindelse. I de reaksjoner hvor hydrogenhalogenidet dannes kan selvsagt en hvilken som helst av de vanlig anvendte hydrogenhalogenidakseptorer også
anvendes.
Ved kondensasjonen av et alfa halosyrederivat av
formel VIII kan lignende reaksjonsbetingelser, løsningsmidler og hydrogenhalogenidakseptorer anvendes som ved amiddannelsen.
Forskjellige substituenter på de nye forbindelser,
for eksempel som definert for Rg kan være til stede i utgangs-forbindelsene eller adderes etter dannelse av amidproduktene etter kjente substitusjonsmetoder eller omdanneIsesreaksjoner. Således kan nitrogruppen adderes til sluttproduktet ved nitrering
av den aromatiske ring, og nitrogruppen kan omdannes til andre grupper slik som amino ved reduksjon, og halo ved diazo-tering av aminogruppen og fortrengning av diazogruppen. Andre reaksjoner kan utføres på det dannede amidprodukt. Amino-grupper kan alkyleres under dannelse av mono- og dialkylamino-grupper, mercapto og hydroxygrupper kan alkyleres under dannelse av tilsvarende ethere. Således kan substitusjons-reaksjoner og andre reaksjoner anvendes for å tilveiebringe et utall substituenter på molekylet i sluttproduktene. Hvor reaktive grupper er til stede må disse selvsagt beskyttes med egnede blokkerende grupper under hvilke som helst av de ovenfor angitte reaksjoner, i særdeleshet kondensasjonsreak-sjoner under dannelse av amidkjedene.
Syre og basesalter av de nye forbindelser kan dannes under anvendelse av standardprosedyre. Ofte dannes disse in situ under fremstilling av de nye amidoaminosyrer.
De nye forbindelser eksisterer selvsagt i stereo-isomere former og de erholdte produkter kan være blandinger av isomerene, som kan oppløses. Alternativt kan den foretrukne stereoisomer fremstilles ved valg av spesifikke isomerer som utgangsforbindelser. De foretrukne former hvor hvert asymmetrisk senter (kiralt senter) er av S-konfigurasjon fremstilles derfor fortrinnsvis ved den stereospesifikke rute i steden for å foreta oppløsning av blandinger av isomerer. Forbindelsene i hvilke S-konfigurasjonen eksisterer ved alle asymmetriske sentra er de mest aktive, de hvori R-konfigurasjonen eksisterer har mindre aktivitet, og de hvori både R- og S-konfigurasjon eksisterer har mellomliggende aktivitet.
Oppfinnelsen illustreres ytterligere i de etterfølgende eksempler.
Eksempel 1
A. Tert- butyl N-( exo- norbornyl) glycinat
50 g (0,451 mol) exo-aminonorboran ble løst i 800 ml ethanol og deretter ble 55 g (0,545 mol) triethylamin tilsatt. Til denne løsning ble dråpevis tilsatt 87 g (0,446 mol)
tertiær butylbromacetat, som var oppløst i 50 ml ethanol.
Den resulterende blanding ble omrørt over natten ved romtemperatur. Ethanolen ble fordampet og vann ble tilsatt til residuuet. Produktet ble ekstrahert to ganger i kloroform.
Det kombinerte kloroformekstrakt ble vasket to ganger med
vann, ble tørket over magnesiumsulfat, filtrert og fordampet under dannelse av produktet som en lys gul olje (78 g, 78%).
B. Tert-butyl N-carbobenxyloxy (L-alanyl)-N-(exo-norbornyl) glycinat
22,4 g (0,1 mol) tert-butyl N-(exo-norbornyl) glycinat og 22,3 g (0,1 mol) N-carbobenzyloxy-L-alanin ble oppløst i 3 00 ml methylenklorid og løsningen ble avkjølt på et isbad.
Til denne løsning ble tilsatt 20,6 g (0,1 mol) dicyclohexylcarbodiimid. Reaksjonsblandingen ble omrørt i 15 minutter med ytre kjøling og deretter i en og en halv time ved romtemperatur. Utfelt dicyclohexylurea ble filtrert og vasket med ether. Filtratet ble fordampet under dannelse først av en farveløs olje som ble krystallisert fra n-hexan/ether under dannelse av et farveløst fast materiale (33,4 g, 77,7%;
sm.p. 93-95°C; [a] £HC13 = +3^7<0>^
Fra filtratet ble det erholdt en andre masse som
en lys gul olje som krystalliserte langsomt (8 g).
C. N- carbobenzyloxy ( L- alanyl)- N-( exo- norbornyl) glycin
20 g (0,0465 mol) tert-butyl N-carbobenzyloxy (L-alanyl)-N-(exo-norbornyl) glycinat ble tilsatt til 125 ml kald trifluoreddiksyre. Reaksjonsblandingen ble omrørt i 15 minutter med ytre kjøling (isbad) og deretter i to og en halv time ved romtemperatur. Trifluoreddiksyre ble destillert ved 30°C under vakuum. Residuuet ble løst i kloroform, vasket to ganger med vann, ble tørket over magnesiumsulfat, filtrert og fordampet under dannelse av10,9g (60%) av det ønskede produkt som et lyst gult skum; sm.p. 52°, £<HCl>3"= -1,62°.
Eksempel 2
A. Tert-butyl N-carbobenzyloxy(L-fenylalanyl)-N-(exo-norbornyl-glycinat
18,8 g (0,0836 mol) tert-butyl N-(exo-norbornyl) glycinat og 25 g (0,0836 mol) N-carbobenzyloxy-L-fenylalanin ble løst i 600 ml methylenklorid. Til den resulterende løsning ble tilsatt 19,0 g (0,0922 mol) dicyclohexylcarbodiimid. Reaksjonsblandingen ble omrørt ved romtemperatur i 4 timer. Utfelt dicyclohexylurea ble filtrert og vasket med en liten mengde ether. Filtratet ble konsentrert og ether ble tilsatt til residuuet for å utfelle mer dicyclohexylurea. Dicyclohexylurea ble filtrert fra og filtratet ble fordampet under dannelse av produktet som en lys gul olje (49,7 g) .
B. N- carbobenzyloxy( L- fenylalanyl)- N-( exo- norbornyl)- glycin Til 9,5 g (0,0188 mol) tert-butyl N-carbobenzyloxy (L-fenylalanyl)-N-(exo-norbornyl) glycinat ble tilsatt 75 ml trifluoreddiksyre. Den resulterende løsning ble omrørt i 15 minutter med ytre kjøling (isbad) og deretter i en og en halv time ved romtemperatur. Trifluoreddiksyre ble fordampet ved 30°C under vakuum. Residuuet ble løst i kloroform og vasket to ganger med vann. Det organiske lag ble tørket over magnesiumsulfat, filtrert og fordampet under dannelse av en lys gul gummi.'Produktet krystalliserte fra n-hexan/ether under dannelse av et farveløst fast materiale (6,0 g, 71%) smeltepunkt 63,5°C.
C. L- fenylalanyl- N-( exo- norbornyl) glycin
2,5 g (5,56 mmol) N-carbobenzyloxy (L-fenylalanyl)-N-(exo-norbornyl) glycin.ble tilsatt til en mettet løsning av HBr i 20 ml eddiksyre, som var blitt avkjølt på et isbad.
Den resulterende løsning ble lagret i 15 minutter med ytre kjøling og deretter i en time ved romtemperatur. Mesteparten av eddiksyren ble fordampet og ether ble tilsatt til residuuet under dannelse av hydrobromidet som et gult fast materiale som ble filtrert og vasket med ether. Saltet ble oppløst på nytt i kloroform og omrørt med kull. Kullet ble filtrert fra og filtratet ble fordampet. Produktet ble krystallisert fra ether under dannelse av lyse gule krystaller (1,7 g, 75%).
Eksempel 3
A. Tert- butyl N-( cyclopentyl)- glycinat
50,0 g (587 mmol) cyclopentylamin ble løst i 500 ml vannfri ether og 69,3 g (685 mmol) triethylamin ble tilsatt. Den resulterende løsning ble avkjølt på et isbad hvorpå 95,4 g (489 mmol) tertiær butylbromacetat i 150 ml ether ble langsomt tilsatt i løpet av tre timer. Reaksjonsblandingen ble omrørt over natten ved romtemperatur. Reaksjonsblandingen ble surgjort til pH 1 med vandig saltsyre og lagene ble fraskilt.
Det vandige lag ble vasket med ethylacetat og ble deretter gjort basisk til pH 10 med vandig ammoniumhydroxyd. Produktet ble ekstrahert flere ganger med methylen klorid, og det kombinerte organiske ekstrakt ble vasket to ganger med vann,
en gang med saltvann, ble tørket over magnesiumsulfat, filtrert og fordampet under dannelse av det ønskede produkt som en svakt gul olje (56,9 g, 58,4%).
B. Tert-butyl N-carbobenzyloxy (L-alanyl)-N-(cyclopentyl)-glycinat
16,3 g (81,8 mmol) tert-butyl N-(cyclopentyl) glycin ble løst i 500 ml methylenklorid. Reaksjonsblandingen ble avkjølt på et isbad hvoretter 16,9 g (81,8 mmol) dicyclohexylcarbodiimid løst i en liten mengde methylenklorid ble tilsatt.
18,3 g (81,8 mmol) N-carbobenzyloxy-L-alanin i 50 ml methylenklorid ble dråpevis tilsatt i løpet av 30 minutter med ytre kjøling. Reaksjonsblandingen ble omrørt over natten ved romtemperatur. Utfelt dicyclohexylurea ble filtrert og vasket med en liten mengde methylenklorid. Filtratet ble

Ved skraping, og avkjøling ble det dannet farveløse krystaller av det ønskede produkt (30 g, 91%); sm.p. 73-74°, [a] ^1(EtOH) = -23.044°.
C. N- carbobenzyloxy( L- alanyl)- N-( cyclopentyl)- glycin
19,9 g (49,2 mmol) tert-butyl N-carbobenzyloxy (L-alanyl)-N-(cyclopentyl)-glycinat ble løst i 50 g (492 mmol) anisol, hvorpå 111 g (974 mmol) trifluoreddiksyre ble tilsatt. Reaksjonsblandingen ble omrørt i tre timer ved romtemperatur. Trifluoreddiksyren ble fordampet ved 3 0°C under vakuum. Residuuet ble løst i ether og vasket fire ganger med vann.
Det sure produkt ble ekstrahert flere ganger i vandig kalium-carbonat og lagene ble fraskilt. De kombinerte vandige ekstrakter ble vasket to ganger med ether og ble deretter surgjort forsiktig til pH 1 ved dråpevis tilsetning av konsentrert saltsyre. Det utfelte produkt ble ekstrahert flere ganger i ether. Etheren ble vasket med vann, tørket over magnesiumsulfat, filtrert og fordampet. Residuuet ble oppløst på nytt i ether og ble omrørt med kull i 3 0 minutter. Kullet ble filtrert fra og filtratet fordampet under dannelse
av 16,0 g urent produkt. Det urene produkt ble renset ved HPLC under anvendelse av eddiksyre/ethylacetat/n-hexan (2:49:49) som elueringsmiddel. Det rene produkt ble erholdt som et farveløst fast materiale (9,9 g,52,6%); sm.p. 48-54°; [a] ^ (EtOH) = -20,644°.
D. L- alanyl- N-( cyclopentyl)- glycin
8,7 g (25,0 mmol) N-carbobenzyloxy-(L-alanyl)-N-(cyclopentyl)glycin ble løst i 200 ml methanol og 1 g 10%-ig Pd/C ble tilsatt. Den resulterende blanding ble hydrogenert ved et begynnelsestrykk på 3,5 kg/cm 2 over natten. Reaksjonsblandingen ble filtrert gjennom celite, filterkaken ble vasket med methanol og filtratet ble fordampet til tørrhet ander dannelse av et hvitt fast materiale. Dette materiale ble delt mellom vann og ether. Det vandige lag ble frysetørket
under dannelse av detønskede produkt som et farveløst fast materiale (2,7 g, 50,5%); sm.p. 153-156°; W ^<1>(H20 = +11,677°.
É. N-(1-carbethoxyethyl) -L-alanyl -N'-cyclopentyl glycin maleat
En blanding av 4,39 g (0,01 mol) CBZ-L-alanyl-N-cyclopentyl glycin benzylester, 3,5g (0,03 mol) ethylpyruvat,
300 mg 10%-ig palladium på carbon i 50 ml ethanol ble hydrogenert inntil hydrogenopptaket var stanset.
Produktet ble filtrert, 1,16 g (0,01 mol) maleinsyre ble tilsatt og blandingen ble fordampet til tørrhet. Den urene olje ble vasket med ethylacetat og krystallisert fra ethanol-ether.
Eksempel 4
A. Ethyl N-carbobenzylloxy-L-isoleucyl-N-(exo-norbornyl) glycinat
7,8 g (39,5 mmol) ethyl N-(exo-norbornyl) glycinat
og 10 g (39,5 mmol) N-carbobenzyloxy-L-isoleucin ble løst i 250 ml methylenklorid. Til den resulterende løsning ble tilsatt 8,3 g (40,3 mmol) dicyclohexylcarbodiimid. Reaksjonsblandingen ble omrørt i to og en halv time ved romtemperatur. Utfelt dicyclohexylurea ble filtrert og vasket med ether. Filtratet ble konsentrert og ether ble tilsatt til residuuet
for å utfelle en andre masse dicyclohexylurea som ble filtrert. Filtratet ble fordampet under dannelse av 16,8 g (96%) av det ønskede produkt som en lys gul viskøs olje.
Eksempel 5
A. Ethyl N-( 5- indanyl)- glycinat
Til en løsning av 15,0 g (0,1128 mol) 5- aminoindan
og 11,92 g (0,1184 mol) triethylamin i 200 ml ethanol ble dråpevis tilsatt 18,83 g (0,1128 mol) ethylbromacetat. Etter at tilsetningen var fullført ble blandingen omrørt ved romtemperatur i 24 timer og ble konsentrert i vakuum. Residuuet ble løst i ether, ble vasket med H20 (4 x 300 ml), saltvann
og ble tørket (MgSO^). Konsentrering av de flyktige bestanddeler i vakuum gav et krystallinsk materiale som ble omkrystallisert fra petroleumether; kokepunkt 30-60°C, smeltepunkt 4 9°C.
B. Ethyl N- carbobenzoxy- L- alanyl- N-( 5- indanyl)- glycinat
Til en løsning av 10,6 g (48,4 mmol) ethyl N-(5-indanyl)-glycinat og 10,5 g (50,8 mmol) N, N'-dicyclohexylcarbodiimid i 200 ml CH2C12ved 0°C, ble dråpevis tilsatt en løsning av 11,3 g (50,8 mmol) N-carbobenzyloxy-L-alanin i 100 ml CH2C12. Etter at tilsetningen var fullført ble løsningen omrørt i ti minutter, ble oppvarmet til romtemperatur og omrørt over natten. Løsningen ble filtrert og konsentrert i vakuum. Residuuet ble løst i ether, ble filtrert, vasket med mettet NaHC03, 10%-ig vandig HC1 (5x), saltvann og ble tørket over (MgSO^). Konsentrering av de flyktige bestanddeler i vakuum gav produktet.
C. N- carbobenzoxy- L- alanyl- N-( 5- indanyl)- glycin
Til en løsning av 14,0 g (33,0 mmol) ethyl N-carbobenzoxy-L-alanyl-N- ( 5-indanyl) -glycinat i 150 ml ethanol ble tilsatt 25 ml av en 2N vandig natriumhydroxydløsning (49,5 mmol) og blandingen ble omrørt ved romtemperatur i 20 timer. De flyktige bestanddeler ble fjernet under redusert trykk, og residuuet ble løst i ether og mettet natriumbicarbonat. Det organiske lag ble fraskilt og ekstrahert med mettet natriumbicarbonat. De kombinerte basiske ekstrakter ble avkjølt på et isbad og ble surgjort med konsentrert HC1. Den vandige løsning ble ekstrahert grundig med CH2C12. De kombinerte organiske ekstrakter ble tørket (MgSO^) og konsentrert i vakuum.
D. L- alanyl- N-( 5- indanyl)- glycin
En løsning av 4,5 g N-carbobenzoxy-L-alanyl-N-(5-indanyl)-glycin og 1,0 g 10%-ig palladium på carbon i 250 ml ethanol ble hydrogenert ved 1,75 kg/cm 2 i to timer og ble deretter filtrert. Konsentrering av de flyktige bestanddeler i vakuum gav et residuum som ble løst i vann og filtrert over celite. Lyophilisering av filtratet gav det krystallinske materiale med smeltepunkt 119-120°C.
E. N-( 1- carboxyethyl)- L- alanyl- N-( 5- indanyl)- glycinpyruvat
Til en løsning av 3,8 g (9,59 mmol) N-carbobenzoxy-L-alanyl-N- (5-indanyl)-glycin og 4,2 g (47,9 mmol) pyruvsyre
i 200 ml ethanol ble tilsatt 9,5 g 10%-ig palladium på carbon. Blandingen ble hydrogenert .ved 3,5 kg/cm 2 i fire timer, ble filtrert og konsentrert i vakuum. Residuuet ble løst i f^O
og ethylacetat. Det vandige lag ble vasket to ganger med ethylacetat, en gang med ether og ble lyophilisert under dannelse av det krystallinske produkt med smeltepunkt 140°C (spaltning).
Eksempel 6
A. N'-( carbobenzyloxy)- alanyl- N-( p- tolyl)- glycin t- butylester
20 g (0,0954 mol) p-tolyl-glycin t-butylester ble tatt opp i en enliters trehals rundkolbe utstyrt med en dråpetrakt, kondensator (tørkerør), termometer og rører. Esteren ble løst i 250 ml methylenklorid, og til dette ble tilsatt en løsning av 27 g (0,1311 mol) N,N-dicyclohexylcarbociimid i 100 ml methylenklorid. Temperaturen på reaksjonsblandingen ble brakt til 0°C (isbad). 33,5 g (0,15 mol) carbobenzyloxy-L-alanin i methylenklorid ble dråpevis tilsatt i løpet av
en og en halv time. Etter endt tilsetning fikk reaksjonsblandingen oppvarming til romtemperatur og ble omrørt over natten. Reaksjonsblandingen ble igjen kjølt ned til 0°C
for fullstendig å utfelle DCC urea, ble filtrert hvorpå filtratet ble konsentrert på en rotasjonsfordamper under dannelse av en uren lys tykk væske. Det urene produkt ble oppløst i 400 ml vannfri diethylether og ble ekstrahert med 3x1N HC1 og 3 ganger natriumbikarbonat (mettet). Etherløsningen ble tørket over vannfri magnesiumsulfat og etheren ble fordampet under dannelse av en meget tykk gul væske.
B. N' -( carbobenzyloxy)- alanyl- N-( p- tolyl)- glycin
Til en løsning av 9,5 g N'-(carbobenzyloxy)-alanyl-N-(p-tolyl)-glycin t-butylester i 16 ml methylenklorid ble dråpevis tilsatt 10 ml TFA ved 0°C. Ved en fullførelse av TFA tilsetningen ble temperaturen på reaksjonsblandingen hevet til romtemperatur, og blandingen ble omrørt i 54 timer. Etter endt omsetning ble methylenklorid fordampet i vakuum og residuuet ble løst i 100 ml carbontetraklorid. Denne løsning ble vasket tre ganger med destillert vann (50 ml's porsjoner) og carbontetraklorid, og ble fordampet. Det resulterende residuum ble løst i 150 ml diethylether og ble ekstrahert tre ganger med natriumbicarbonat (mettet). De kombinerte natriumbicarbonatlag ble surgjort med 10%-ig saltsyre til pH 4-5 og ble ekstrahert med diethylether. Ether ble fordampet i vakuum under dannelse av den rene syre. TLC (methanol-ethylacetat, 2:3) gav "en flekk. NMR spekteret var i overensstemmelse med den angitte struktur.
C. Alanyl- N-( p- tolyl)- glycinhydrobromid
1,7 g (0,0046 mol) N1 -(carbobenzyloxy)-alanyl-N-(p-tolyl)-glycin ble løst i 5 ml iseddik og løsningen ble avkjølt til 0°C (isbad) Til denne løsning ble tilsatt en mettet løsning av hydrogenbromid i iseddik (4,6 ml). Reaksjonsblandingen fikk oppvarmes til romtemperatur og
ble omrørt i ytterligere en halv time. Reaksjonsblandingen ble fortynnet med 150 ml ether under dannelse av et halvfast hydrobromidsalt. Saltet ble løst i 100 ml destillert vann og ble vasket tre ganger med 50 ml porsjoner av ether. Det vannløselige salt gav en enkel flekk ved TLC (methanol) og også n-butanol-eddiksyre-vann. Det faste salt ble erholdt etter fordampning av vannet på en frysetørker. Hydrobromidet var hygroskopisk, med smeltepunkt 83-87°C.
D. N '-(1-carboethoxyethyl) -alanyl-N- (p-tolyl) -glycin t- butylester
I en tørr hydrogeneringsflaske ble 25 g (0,059 0 mol) av N'-(carbobenzyloxy)-alanyl-N-(p-tolyl)glycin t-butylester løst i 60 ml absolutt ethanol. Til denne løsning ble tilsatt 30 ml iseddik, 13 g (0,1126 mol) ethylpyruvat og 4,5 g 10%-ig palladium/carbon. Hydrogenering over natten av denne blanding ble foretatt under et trykk på 0,77 kg/cm 2. Reaksjonsblandingen ble opparbeidet ved tilsetning av 5 g celite og palladium/carbonet ble filtrert. Ethanol og eddiksyre ble fjernet i vakuum. Det urene produkt ble undersøkt ved TLC (ethylacetat) og ble funnet å inneholde to diastereomerer av produktet. Diastereomerene kunne separeres ved HPLC (hexan-ethylacetat). En blanding av diastereomerer av de separerte isomerer blekarakterisert vedNMR og massespektroskopi.
E. N<1->(1-carboethoxyethyl)-alanyl-N-(p-tolyl)-glycin og dets calciumsalt
Til en løsning av 2,6 g. (0,0066 mol) N'-(1-carbo-ethoxyethyl)-alanyl-N-(p-tolyl)-glycin t-butylester i 26 ml methylenklorid ble dråpevis tilsatt 14,4 ml (0,19 mol) trifluoreddiksyre ved 0°C (isbad). Temperaturen fikk anta romtemperatur og reaksjonsblandingen ble omrørt i 72 timer. Etter endt reaksjon ble methylenklorid og trifluoreddiksyre fordampet i vakuum. Det således erholdte urene produkt ble renset ved kromatografi på IR-120 harpiks. Harpiksen ble vasket med vann og surgjort med 6NHC1. Forbindelsen ble løst i 2 riil destillert vann og fylt på kolonnen. Kolonnen ble vasket med vann og eluert med 5% ammoniakk under dannelse av 0,65gav den ønskede forbindelse. 0,57 g av den fri syre ble løst i 25 ml vann og ble omsatt med 47,6 mg (0,0009 mol) calciumoxyd. Et hvert uomsatt calciumoxyd ble fjernet ved filtrering, og den resulterende løsning ble frysetørket under dannelse av 0,61 g av calciumsaltet med smeltepunkt 165-170°C.
Eksempel 7
A. Ethyl N- carbobenzyloxy- L- alanyl- N- benzyl- glycinat
Til en løsning av 47,5 g (0,268 mol) ethyl N-benzyl-glycinat og 60,0 g (0268 mol) N-carbobenzyloxy-L-alanin i 600 ml methylenklorid ved 0°C, ble dråpevis tilsatt en løsning av 55,2 g (0,268 mol) N, N'-dicyclohexylcarbodiimid i 600 ml mehylenklorid i løpet av en time. Etter at tilsetningen var fullført ble løsningen omrørt i ti minutter,
ble oppvarmet til romtemperatur og omrørt over natten. Løsningen ble filtrert, vasket med fortynnet vandig HC1,
H20, mettet NaHCO-j, saltvann, ble tørket (MgSO^) . Konsentrering av løsningen i vakuum gav det oljeaktige produkt..
B. N- carbobenzyloxy- L- alanyl- N- benzyl- glycin
Til 11,2 g (28,1 mmol) ethyl N-carbobenzyloxy-L-alanyl-N-benzylglycinat i 60 ml ethanol ble tilsatt 2,2 g
(55,0 mmol) natriumhydroxyd. Blandingen ble omrørt ved romtemperatur i 18 timer. Løsningen ble konsentrert i vakuum og det gjenværende residuum ble løst i vann. Den vandige løsning ble vasket to ganger med ether, ble avkjølt på et isbad og surgjort med konsentrert HC1. Den surgjorte løsning ble ekstrahert grundig med ether. De kombinerte organiske ekstrakter ble tørket (MgSO^) og konsentrert i vakuum under dannelse av carboxylsyren.
C. L- alanyl- N- benzyl- glycin
En løsning av 6,0 g N-carbobenzyloxy-L-alanyl-N-benzylglycin og 2,0 g 10%-ig palladium på carbon i 200 ml ethanol ble hydrogenert ved 3,5 kg/cm 2 i fire timer og ble filtrert. Konsentrering av de flyktige bestanddeler i vakuum gav et residuum som ble oppløst i H20 og filtrert. Den vandige løsning ble surgjort med 25%-ig vandig HBr inntil pH lik 1 ble erholdt. Løsningen ble lyophilisert. Det krystallinske produkt ble omkrystallisert fra ether/acetonitril (smeltepunkt 84-85°C).
D. N-( 1 - carboxyethyl)- L- alanyl- N- benzyl- glycinpyruvat
Til en løsning av 5,8 g (15,6 mmol) N-carbobenzyloxy-L-alanyl-N-benzyl-glycin og 3,5 g (39,1 mmol) pyruvsyre i
200 ml absolutt ethanol ble tilsatt 0,5 g 10%-ig palladium på carbon. Blandingen ble hydrogenert ved 3,5 kg/cm 2 i fire timer.
Løsningen ble filtrert og konsentrert i vakuum ved romtemperatur. Residuuet ble løst i J^O og ethylacetat. Det vandige lag ble vasket to ganger med ethylacetat og en gang med ether, og ble lyophilisert under dannelse av det krystallinske produkt med smeltepunkt 105-108°C.
Eksempel 8
A. Ethyl N-( 2- indanyl) glycinat
50 g (0,295 mol) 2-aminoindan hydroklorid ble løst
i en liter acetonitril og 65 g (0,644 mol) triethylamin ble tilsatt. Til den resulterende løsning ble dråpevis tilsatt 50 g (0,297 mol) ethylbromacetat i 100 ml acetonitril. Den resulterende blanding ble omrørt over natten ved romtemperatur. Acetonitril ble fordampet og residuuet ble løst i kloroform. Kloroformen ble vasket to ganger med vann, ble tørket over magnesiumsulfat, ble filtrert og fordampet under dannelse av et produkt som en lys brunfarvet olje (39,4 g; 62%) som ble anvendt i neste reaksjon uten ytterligere rensning. Produktet blekarakterisertsom dets hydrokloridsalt.
B. Ethyl N- carbobenzyloxy ( L- alanyl)- N-( 2- indanyl) glycinat
39,4 g (0,180 mol) ethyl N-(2-indanyl) glycinat og 40 g (0,179 mol) N-carbobenzyloxy-L-alanin ble løst i 600 ml methylenklorid, og den resulterende løsning ble avkjølt på isbad. 40 g (0,190 mol) dicyclohexylcarbodiimid ble porsjons-vis tilsatt, og den resulterende blanding ble omrørt i 30 minutter under ytre avkjøling og deretter over natten ved romtemperatur. Utfelt dicyclohexylurea ble filtrert og vasket med en liten mengde ether. Filtratet ble fordampet under dannelse av det urene produkt som en mørk olje som ble kromatografert på silicagel under anvendelse av ether som elueringsmiddel, under dannelse av det rene produkt som en lys gul gummi (70,2 g, 92,4%).
C. N- carbobenzyloxy-( L- alanyl)- N-( 2- indanyl) glycin
40 g (0,0991 mol) ethyl N-carbobenzyloxy-(L-alanyl)-N-(2-indanyl) glycinat ble tilsatt til en løsning av 11,1 g (0,198 mol) kaliumhydroxyd i 500 ml ethanol.
Den resulterende blanding ble omrørt i tre timer
ved romtemperatur. Ethanol ble fordampet og residuuet ble løst i vann. Den vandige løsning ble vasket tre ganger med ether. Den vandige løsning ble surgjort til pH 2 med konsentrert saltsyre og det utfelte produkt ble ekstrahert flere ganger i kloroform. De kombinerte kloroformekstrakter ble vasket med vann, ble tørket over magnesiumsulfat, filtrert og fordampet under dannelse av produktet som en lys brun gummi (25 g, 63,8%). Produktet blekarakterisertsom dets dicyclohexylaminsalt med smeltepunkt 180°C.
Eksempel 9
A. N<1->(carbobenzyloxy)-isoleucyl-N-(p-tolyl)-glycin t-butylester;
Reaksjonen ble utført på lignende måte som beskrevet
i eksempel 6 A. 8,67 g (0,0396 mol) p-tolyl-glycin t-butylester ble løst i 45 ml methylenklorid, ble blandet med en løsning av 9,15 g (0,0396 mol) N, N-dicyclohexylcarbodiimid i 30 ml methylenklorid og ble omsatt med en løsning av 10 g (0,0377 mol) carbobenzyloxyisoleucin i 60 ml methylenklorid.
B. N'-( carbobenzyloxy)- isoleucyl- N-( p- tolyl) glycin
8 g N'-CBZ-isoleucyl-N-(p-tolyl)-glycin t-butylester ble løst i 13,5 ml methylenklorid og ble omsatt med 8 ml TFA ved 0°C. Reaksjonsblandingen ble omrørt ved romtemperatur i 72 timer og ble opparbeidet som beskrevet i eksempel 7 B.
Den rene isolerte syre var en enkel flekk på TLC (methanol-ethylacetat, 2:3) og hadde det forventede NMR spekter.
C. Isoleucyl- N-( p- tolyl) glycin
1 g (0,00024 mol) N'-CBZ-isoleucyl-N-(p-tolyl)-
glycin ble løst i 3,5 ml iseddik og omsatt med 2,5 ml hydrogen-
bromid/eddiksyre. Reaksjonsprosedyren er som beskrevet i eksempel 6 C. Det erholdte produkt etter frysetørring blekarakterisert vedTLC (methanol), NMR og massespektroskopi.
Eksempel 10
N-( 1- carboxyethyl)- L- valyl- N- benzyl- glycinpyruvat
Til en løsning av 17,2 g (44,6 mmol) N-carbobenzoxy-L-valyl-N-benzylglycin og 19,6 g (0,223 mol) pyruvsyre i 400 ml absolutt ethanol ble tilsatt 1,5 g 10%-ig palladium på carbon. Blandingen ble hydrogenert ved 3,5 kg/cm 2 i fire timer. Filtrering og konsentrering i vakuum gav et oljeaktig
produkt som ble renset som beskrevet i eksempel 7 D.
Eksempel 11
N-( 1- carboxyethyl)- L- alanyl- N-( 4- methylbenzyl)- glycinpyruvat
Til en løsning av 5,8 g (15,1 mmol) N-carbobenzoxy-L-alanyl-N- (4-methylbenzyl)-glycin og 3,3 g (37,8 mmol) pyruvsyre i 200 ml absolutt ethanol ble tilsatt 0,5 g 10%-ig palladium på carbon. Blandingen ble hydrogenert ved 2,1 kg/cm<2>i fire timer og ble filtrert. Konsentrering av de flyktige bestanddeler i vakuum gav et oljeaktig produkt som ble renset som beskrevet i eksempel 7 D.
Eksempel 12
A. Tert- butyl N-( 3- fenylethyl) glycinathydroklorid
800 ml acetonitril ble tilsatt til 50 g (0,413 mol) f}-fenetylamin, etterfulgt av tilsetning av 100 ml vann og 75 ml konsentrert ammoniumhydroxyd. Til den resulterende omrørte løsning ble 82 g (0,421 mol) tertiærbutylbromacetat i 150 ml acetonitril dråpevis tilsatt ved romtemperatur.
Den resulterende blanding ble omrørt over natten ved romtemperatur. Acetonitrilet ble fordampet på en rotasjonsfordamper hvoretter vann ble tilsatt til residuuet, og produktet ble ekstrahert flere ganger i methylenklorid. De kombinerte methylenkloridekstrakter ble vasket to ganger med vann, ble tørket over magnesiumsulfat, ble filtrert og fordampet under dannelse av det urene produkt som en lys brun olje. Det urene produkt ble renset ytterligere ved silicagelkromatografi (kloroform). De ønskede fraksjoner ble konsentrert og hydro-kloridet ble fremstilt med vannfri hydrogenklorid i ether under dannelse av tertiærbutyl(N- 3-fenylethyl) glycinathydroklorid som et farveløst fast materiale (62 g, 55%); Sm.p. 143°; massespektrum (CI): 143,8 (30,84%) ;- 133f8 (40,93%); 104,8 (38,33%; 87,8 (100%).
B. Tert-butyl N- [1-(S)-ethoxycarbonyl-3-fenylpropylJ -(S)-alanyl- N-( 3 - fenylethyl) glycinat
Til urent N- |l-(S)-ethoxycarbonyl-3-fenylpropyl]-(S)-alanyl-N-carboxyanhydrid som var blitt fremstilt fra 5 g av den tilsvarende (SS)-syre i methylenklorid ble tilsatt 5,1 g (21,3 mmol) tertiærbutyl N-( 3-fenylethyl) glycinat. C. N-[1-(S)-ethoxycarbonyl-3-fenylpropyl] -(S)-alanyl-N-
( 3 ~ f enethyl) glycin hydroklorid
Til 2 g (4,03 mmol) tertiærbutyl N-[i -(S)-ethoxycar-bonyl-3-fenylpropylj-(S)-alanyl-N-(3 -fenethyl) glycinat ble tilsatt p-dioxan som var blitt mettet med 75 ml vannfri hydrogenklorid. Den resulterende løsning ble omrørt i tre timer ved 4 5°C, hvorpå løsningsmiddelet ble fordampet under dannelse av N-[.1 - (S)-ethoxycarbonyl-3-f enylpropyl] - (S) - alanyl-N-(3 -fenethyl)glycinhydroklorid som et farveløst pulver (1,62 g, 84,2%); Sm.p. 72°; [a] D CHC1J,= '- + 10,95°; massespektrum (CI): 423 (m+1 - H20, 100%.

Eksempel 13
A. t- butyl N- carbobenzoxy- L- alanyl- N-( 5- indanyl) glycinat
Til en løsning av 24,5 g (99,1 mmol) t-butyl N-(5-indanyl)glycinat og 20,4 g (99,1 mmol) dicyclohexylcarbodiimid i 130 ml methylenklorid ble dråpevis tilsatt en løsning av 22,1 g (99,1 mmol) N-carbobenzoxy-L-alanin i 150 ml methylenklorid.. Etter at tilsetningen var fullført ble løsningen omrørt i 18 timer, ble filtrert og konsentrert i vakuum. Residuuet ble fortynnet med ether, ble filtrert, vasket med vandig syre, vandig NaOH og ble tørket (MgSO^). Fjerning av de flyktige bestanddeler i vakuum og kromatografi på silicagel under anvendelse av 10% ethylacetat i hexan som elueringsmiddel gav det oljeaktig produkt.
B. t- butyl L- alanyl- N-( 5- indanyl) glycinat
Til en løsning av 7,3 5 g (16,3 mmol) t-butyl N-carbobenzoxy-L-alanyl-N- (5-indanyl) -glycinat og 3,09 g (48,9 mmol) ammoniumformiat med 110 ml methanol ble tilsatt 2,70 g 10%-ig palladium på carbon. Blandingen ble omrørt i ti minutter, ble filtrert og konsentrert i vakuum. Residuuet ble delt mellom ether og vann. Det vandige lag ble mettet med natriumklorid og ble ekstrahert grundig med methylenklorid. De kombinerte organiske ekstrakter ble tørket . (MgS04) og konsentrert i vakuum under dannelse av det krystallinske produkt.
C. t-butyl N-(1 -(S)-ethoxycarbonyl-3-fenylpropyl)-L-alanyl-N- ( 5- indanyl) glycinat
En løsning av 4,80 g (15,09 mmol) t-butyl L-alanyl-N-(5-indanyl)glycinat, 15,50 g (75,47 mmol) ethyl 4-fenyl-2-oxobutyrat og 30 g pulverformet 3A molekylsiler i 100 ml absolutt ethanol ble omrørt i 60 minutter, ved hvilket tids-punkt 1,05 g (16,6 mmol) natriumcyanborhydrid i 15 ml absolutt ethanol ble dråpevis tilsatt i løpet av 90 minutter. Etter at tilsetningen var fullført ble løsningen omrørt i
18 timer, ble filtrert og konsentrert i vakuum. Kromatografi av residuuet på silicagel under anvendelse av 20% ethylacetat i hexan som elueringsmiddel gav denønskede diastereo-mer som en olje.
D. N-(1-(S)-ethoxycarbonyl-3-fenylpropyl)-L-alanyl-N-(5-indanyl)- glycin maleatsalt
Til 2,30 g t-butyl N-(1 -(S)-ethoxycarbonyl-3-fenyl-propyl-L-alanyl-N-(5-indanyl)glycinat i 8 ml eddiksyre ved 0°C og under nitrogen ble tilsatt 5 ml 3,5 N HBr i eddiksyre. Løsningen ble holdt kald i 10 minutter, ble oppvarmet til romtemperatur og omrørt i 20 minutter. Løsningen ble konsentrert i vakuum ved 35°C. Residuuet ble løst i 1:1 ethanol-vann, ble avkjølt med et isbad og nøytralisert med vandig NaOH. Løsningen ble overført til 50 g av en Dowex 50x2-100 kolonne og ble først eluert med 1:1 ethanol-vann og 3% pyridin i vann. Produktrike fraksjoner ble lyophilisert. Residuuet (1,340 g) ble løst i 5 ml acetonitril og 0,3439 g maleinsyre i 10 ml acetonitril ble tilsatt. Løsningen ble konsentrert i vakuum. Residuuet ble krystallisert fra ethylacetat med hexan. Triturering av det faste materiale med ether og hexan gav det krystallinske salt, smeltepunkt 64°C (mykner).
Ved å følge de prosedyrer som er beskrevet i de foregående eksempler ble følgende ytterligere forbindelser fremstilt: N-[l -(S) -ethoxycarbonyl-2-fenylethyl] alanyl-N-(1-indanyl) glycin

Forbindelsene fremstilt.ifølge oppfinnelsen har
vist seg å ha kraftig aktivitet (i størrelsesorden I5Qpå
0,1 til 10,0 mikromol) ved inhibering av det angiotensin-omdannende enzym (ACEI aktivitet) når disse ble testet ved den metode som er beskrevet i Science 196, 441-4 (1977).. Forbindelsene fremstilt ifølge oppfinnelsen har også utvist
en 1,-q på 1 til 10 mg/kg P.O. ved inhibering av infusert angiotensin I i rotter. Som sådanne vil disse kunne være anvendbare ved behandling av hypertensjon.
Forbindelsene kan administreres oralt eller parente-ralt ved behandling av hypertensjon. Egnede doseringsformer innbefatter"tabletter, kapsler, eliksirer og injiserbare løsninger.
Claims (14)
1. Fremgangsmåte for fremstilling av terapeutisk aktive forbindelser av formel
hvori R og Rg uavhengig er hydroxy, lavere alkoxy, lavere alkenoxy, di(lavere alkyl)amino-lavere alkoxy, hydroxy-lavere alkoxy, acylamino-lavere alkoxy, acyloxy-lavere alkoxy, aryloxy, aryloxyl-lavere alkoxy, amino, lavere alkylamino, dilaverealkylamino, hydroxy-amino, eller aryl-lavere alkylamino, R^ , R2 , R3 , R^ , R^ , Ry og Rg er uavhengig alkyl med fra 1 til 20 carbonatomer, alkenyl med fra 2 til 20 carbonatomer, alkynyl med fra 2 til 20 carbonatomer, aryl-lavere alkyl med fra 6 til 12 carbonatomer eller heterocyclisk gruppe-lavere alkyl med fra 7 til 12 carbonatomer, Rg er cycloalkyl, polycycloalkyl, partielt mettet cycloalkyl og polycycloalkyl, cyckoalkyl-lavere alkyl med fra 3 til 20 carbonatomer, aryl med fra 6 til 10 carbonatomer, aryl-lavere alkyl, aryl-lavere alkenyl eller aryl-lavere alkynyl, og R2 og R^ kan sammen med carbon og nitrogenatomet til hvilke de er bundet og R^ og R,, kan sammen med nitrogen og carbonatomet til hvilket de er bundet danne en N-heterocyclisk gruppe inneholdende fra 3 til 5 carbonatomer eller 2 til 4 carbonatomer og et svovelatom; og hvori angitte alkyl, alkenyl og alkynylgrupper kan være substituert med hydroxy, lavere alkoxy, thio, lavere alkylmercapto, amino, lavere alkylamino, di(lavere alkyl) amino, halogen og nitrogrupper; hvilke cycloalkylgrupper innbefatter mono og polycycliske, fullstendig mettede og partielt umettede cycloalkylgrupper såvel som slike grupper substituert med halogen, hydroxy, lavere alkyl, lavere alkoxy, amino lavere alkylamino, di(lavere alkyl)amino, thiol, lavere alkylmercapto, nitro eller trifluormethyl; og arylgruppene kan være substituert med hydroxy, lavere alkoxy, amino, lavere alkylamino, di(lavere alkyl)amino, thiol, lavere alkylmercapto, hydroxy-lavere alkyl, amino-lavere alkyl, thio-lavere alkyl, nitro, halogen, trifluormethyl, methylendioxy, ureid eller guanidin; og salter derav med syrer eller baser, spesielt farmasøytisk akseptable syre og basesalter, karakterisert ved ata) en aminoforbindelse av formel II
omsettes under amid-dannende betingelser med et acylerende derivat av en syre av formel III
eller
b) det tilsvarende imin reduseres, eller
c) en forbindelse av formel VI omsettes med en haloforbindelse av formel VII
for å eliminere hydrogenhalogenid, og
at den erholdte forbindelse eventuelt underkastes substi-tusjons eller omdannelsesreaksjoner for å innføre de forskjellige substituenter på den erholdte forbindelse; og eventuelt at en erholdt forbindelse omdannes til salter derav, spesielt farmasøytisk akseptable salter med syrer og baser.
2. Fremgangsmåte ifølge krav 1, karakterisert ved at R og Rg er uavhengig hydroxy eller lavere alkoxy, R^ og R2 er hydrogen eller lavere alkyl, aryl-lavere alkyl med fra 7 til 12 carbonatomer, eller heterocyclisk gruppe-lavere alkyl med fra 6 til 12 carbonatomer, R3 , R4 ,
R^ , R^ og Rg er hydrogen eller lavere alkyl, Rg er cycloalkyl, polycycloalkyl, partielt mettet cycloalkyl og polycycloalkyl, cycloalkyl-lavere alkyl med fra 3 til 20 carbonatomer, aryl eller aryl-lavere alkyl, og hvor arylgruppen inneholder fra 6 til 10 carbonatomer, hvori arylgruppene kan være substituert med lavere alkyl, lavere alkenyl, lavere alkynyl, hydroxy, lavere alkoxy, amino, lavere alkylamino, di(lavere alkyl)amino, thiol, lavere alkylmercapto, hydroxy-lavere alkyl, amino-lavere alkyl, thio-lavere alkyl, nitro, halogen, trifluormethyl, methylendioxy, ureid og guanidin og salter derav, spesielt farmasøytisk akseptable syre og basesalter.
3. Fremgangsmåte ifølge krav 1 eller 2, karakterisert ved at R^ er lavere alkyl 'eller aryl-lavere alkyl, R^ er lavere alkyl og R2 , R^ , R^ , Ry og Rg er hydrogen.
4. Fremgangsmåte ifølge hvilket som helst av kravene 1-3, karakterisert ved at R og Rg er hydroxy.
5. Fremgangsmåte ifølge hvilket som helst av kravene 1-3, karakterisert ved at R er ethoxy og Rg er hydroxy.
6. Fremgangsmåte ifølge hvilket som helst av kravene 1-3, karakterisert ved at R^ og R4 er methyl.
7. Fremgangsmåte ifølge hvilket som helst av kravene 1-3, karakterisert ved at R^ er fenethyl og R 4 er methyl.
8. Fremgangsmåte ifølge hvilket som helst av kravene 1-3, karakterisert ved at Rg er benzyl.
9. Fremgangsmåte ifølge hvilket som helst av kravene 1-3, karakterisert ved at Rg er p-tolyl.
10. Fremgangsmåte ifølge hvilket som helst av kravene 1-3, karakterisert ved at Rg er indanyl.
11. Fremgangsmåte ifølge hvilket som helst av kravene 1-3, karakterisert ved at Rg er 2-indanyl.
12. Fremgangsmåte ifølge hvilket som helst av kravene 1-3, karakterisert ved at Rg er 5-indanyl.
13. Fremgangsmåte ifølge hvilket som helst av kravene 1-3, karakterisert ved at Rg er cycloalkyl med fra 3 til 10 carbonatomer.
