Oppfinnelsens område
Den foreliggende oppfinnelse angår mellomprodukter for syntesen av LHRH-antagonister, nærmere bestemt en fremgangsmåte for fremstillingen av disse mellomproduktene.
Bakgrunn for oppfinnelsen
Det luteiniserende hormonfrigjørende hormonet, LHRH, kontrollerer sekresjonen av follikkelstimulerende hormon (FSH) og luteiniserende hormon (LH). LHRH-antagonister er forbindelser som er i stand til å blokkere sekresjonen av FSH og LH. De er vanligvis nona- og dekapeptider (men kan være kortere eller lengre) omfattende del av eller hele strukturen til LHRH hvori én eller flere aminosyrer har blitt erstattet med andre naturlige aminosyrer og/eller aminosyrer som ikke finnes i naturen.
Syntetiske LHRH-antagonister kan anvendes som prevensjon og i behandlingen av godartet hyperplasi i prostatakjertelen, hormonavhengige tumorer i brystene og eggstokkene, dysmenorrea, endometriose og andre tilstander. Disse syntetiske LHRH-antagonistene har den generelle formelen
hvori X er fra 5-6 naturlige og/eller syntetiske aminosyrerester. Nærmere bestemt har de den foran nevnte generelle formel hvori X er AAl-AA2-Leu-AA3-Pro-D-Ala, spesielt hvori AA1 er en naturlig eller syntetisk aminosyre og AA2 er en naturlig eller syntetiske aminosyre eller null, AA3 er en naturlig eller syntetisk aminosyre.
Mens det er et antall kjente syntetiske fremgangsmåter for fremstilling av LHRH-analoger er det et behov for forbedring siden det totale utbyttet av LHRH-analoger oppnådd fra kjente fremgangsmåter ikke er høyt og at produktet i tillegg kan trenge omfattende rensing. Videre er de kjente fremgangsmåtene for syntese av LHRH-analoger ganske kostbare.
En syntesestrategi beskrevet i US patent nr. 5 710 246 for å danne dekapeptid- eller nonapeptid-LHRH-antagonister omfatter koplingen av et tripeptidmellomprodukt som representerer aminosyrerestene 1-3 (telling starter ved aminoterminalen til peptidet) med henholdsvis et heptapeptid eller et heksapeptid som representerer henholdsvis aminosyrerestene 4-10 og 4-9. Tripeptidmellomproduktet beskrevet i US 5 710 246 A er en ester, Boc-D-2Nal-D-4ClPhe-D-3Pal-0-Me eller den korresponderende benzyl- eller allylester.
Oppfinnelsens formål
Det er følgelig et formål ved den foreliggende oppfinnelse å tilveiebringe en fremgangsmåte for fremstilling av et tripeptidmellomprodukt for 3+7- og 3+6-syntesen av LHRH-analoger..
Ytterligere formål ved oppfinnelsen vil være tydelig fra det følgende sammendraget av oppfinnelsen, beskrivelsen av de foretrukne utførelsesformer og de tilhørende patentkrav.

Sammendrag av oppfinnelsen
Ifølge oppfinnelsen tilveiebringes en fremgangsmåte for fremstilling av et tripeptid som representerer aminosyrer 1-3 til en LHRH-antagonist, hvori den terminale aminogruppen er Boe- eller Ac-beskyttet og den terminale karboksylgruppen (dvs. den terminale gruppen av aminosyre nr. 3) ikke er beskyttet.
Tripeptidet (I)
er et anvendelig mellomprodukt i en fremgangsmåte for syntesen av en LHRH-antagonist med den generelle formel (II) hvori X er fra 5-7 naturlige og/eller syntetiske aminosyrerester, nærmere bestemt AAl-AA2-Leu-AA3-Pro-D-Ala, spesielt hvori AA1 er en naturlig eller syntetisk aminosyre og AA2 er en naturlig eller syntetisk aminosyre eller null, AA3 er en naturlig eller syntetisk aminosyre. Tripeptidet (I) er spesielt anvendelig i syntesen av et peptid med den generelle formel (Ila) hvori AA1 og AA2 har den ovenfor angitte betydning, spesielt en LHRH-antagonist med formelen (III) eller, enda mer foretrukket, formlene (Illa)
Tripeptidet
Boc-D-2Nal-D-4ClPhe-D-3Pal-OH (IX)
som har samme nytte som mellomprodukt i en fremgangmåte for fremstilling av en LHRH-antagonist som beskrevet over for tripeptidet (I).
Den foreliggende oppfinnelse tilveiebringer følgelig en fremgangsmåte for fremstilling av et tripeptid med formelen (I)
Ac-D-2Nal-D-4ClPhe-D-3Pal-OH (I)
omfattende de følgende etterfølgende trinn for fremstillingen av (I):
(a) omsetting av Boc-D-4ClPhe-OH med HONSu for å danne Boc-D-4ClPhe-OSu (VII); (b) omsetting av Boc-D-4ClPhe-OSu (VII) med H-D-3Pal-OH for å danne Boc-D-4ClPhe-D-3Pal-OH (VIII); (c) omsetting av Boc-D-4ClPhe-D-3Pal-OH (VIII) med Boc-D-2Nal-OSu fremstilt ved å omsette Boc-D-2Nal-OH med HONSu for å danne Boc-D-2Nal-D-4ClPhe-D-3Pal-OH (IX); (d) Omsetting av Boc-D-2Nal-D-4ClPhe-D-3Pal-OH (IX) med eddiksyre for å danne Ac-D-2Nal-4ClPhe-D-3Pal-OH (I).
Tripeptidet (I) fremstilt i følge oppfinnelsen er anvendelig ved fremstilling av en LHRH-antagonist omfatter trinnet å kople tripeptidet (I) med et heptapeptid (IV) med den generelle formel
P'-Ser(P2)-AAl-AA2-Leu-Lys(iPr,P4)-Pro-D-AlaNH2 (IV),
hvori P<4>er H eller en aminobeskyttende gruppe slik som Boe, hvori AA1 og AA2 har den forannevnte betydning, spesielt med et heptapeptid (V) med den generelle formel P,-Ser(P<2>)-NMeTyr(P<3>)-D-Lys(Nic)-Leu-Lys(iPr,P<4>)-Pro-D-AlaNH2(V), hvori P er valgt fra H eller aminobeskyttende gruppe og P og P er uavhengig valgt fra H og -OH-beskyttende gruppe, og P<4>har den ovenfor angitte betydning for fremstilling av LHRH-antagonisten Ac-D-2Nal-D-4ClPhe-D-3Pal-Ser-MeTyr-D-Lys(Nic)-Leu-Lys(iPr)-Pro-D-Ala-NH2 (III),
nærmere bestemt med et heptapeptid (Va) med den generelle formel P<]->Ser(P<2>)-NMeTyr(P<3>)-D-Asn-Leu-Lys (iPr,P<4>)-Pro-D-AlaNH2(Va), hvori P<1>er valgt fra H eller aminobeskyttende gruppe og P og P er uavhengig valgt fra H og -OH-beskyttende gruppe, og P<4>har den ovenfor angitte betydning for fremstilling av LHRH-antagonisten Ac-D-2Nal-D-4ClPhe-D-3Pal-Ser-MeTyr-D-Asn-Leu-Lys(iPr)-Pro-D-Ala-NH2(III).
Heptapeptidet (V) er beskrevet i US 5 710 246 A. Heptapeptidet med den generelle formel (IV), inkludert heptapeptidet (Va), kan syntetiseres gjennom rutinemessige modifikasjoner av syntesen til (V) eller ved å kople de korresponderende Boc-aminosyrene på en peptidsyntesemaskin (Beckman Modell 990), som beskrevet i WO 94/40757 hvor også LHRH-antagonisten (III) er beskrevet.
En spesiell fordel ved fremgangsmåten ifølge oppfinnelsen er at et billigere utgangsmateriale, H-D-Pal-OH-2HCl, kan anvendes i stedet for esteren H-Pal-OR-2HC1; beskyttelsesgruppen til utgangsmaterialet trenger ikke å fjernes. Derfor er syntesen ifølge oppfinnelsen ett trinn kortere og forhindrer at materialet tapes i det ytterligere trinnet. En annen fordel er at dannelsen av urenheter i saponifiseringstrinnet unngås. Dannelsen av slike urenheter er velkjent. De basiske betingelsene i esterhydrolysetrinnet forårsaker f.eks. delvis racemisering av D-Pal. De andre kjente alternativene for å fjerne estergruppen ved hjelp av katalytisk hydrogenering (i tilfelle av allyl- eller benzylestergrupper) risikerer å forårsake et tap av Cl fra 4ClPhe og produsere fenyl. Mens allylgruppene kan fjernes ved ytterligere andre reagenser er den fullstendige fjerningen vanskelig å kontrollere.

Syntese av Ac-D-2Nal-4ClPhe-D-3Pal-OH (I).
EKSEMPEL 1. Boc- D- 4ClPhe- OSu. Boc-D-4ClPhe-OH (299,75 g; 1,0 eq.) og HONSu (184,1 g; 1,6 eq.) ble løst i 2-propanol (4,5 1). Blandingen ble avkjølt til 0°C og DIC (164,1 g; 1,3 eq.) ble tilsatt. Blandingen ble rørt i 16 timer under varming til romtemperatur. Produktet ble filtrert av, vasket med 2-propanol (1,5 1) og tørket. Utbytte: 85 %. HPLC-renhet: 98,8 %.
EKSEMPEL 2. Boc- D- 4ClPhe- D- 3Pal- OH. H-D-3Pal-OH, 2 HC1 (251,1 g; 1,05 eq.) og Boc-D-4ClPhe-OSu (396,8 g; 1,0 eq.) ble løst i DMSO (3,33 1) og NMM (318,8 g; 3,15 eq.) ble tilsatt. Blandingen ble rørt i 16 timer ved romtemperatur. Vann (17 1) ble tilsatt og pH ble justert til 4-4,5, hvilket forårsaket at produktet presipiterte. Blandingen ble filtrert og produktet ble vasket med vann (3x5 1) for å fjerne spor av DMSO, H-D-3Pal-OH og Boc-D-4ClPhe-OH. Produktet ble tørket. Utbytte: 80 %. HPLC-renhet: 97,8 %.
EKSEMPEL 3. Boc- D- 2Nal- OSu. Boc-D-2Nal-OH (315,4 g; 1,0 eq.) ble løst i 2-propanol (6,8 1) ved -10°C og IBC (157 g; 1,15 eq.) og NMM (116 g; 1,15 eq.) ble tilsatt. Etter røring i 5-10 min. ble en blanding av HONSu (230,1 g; 2,0 eq.) i 2-propanol (1,4 1) tilsatt. Ytterligere NMM (10,1 g; 0,1 eq.) ble tilsatt. Etter en halv time ble vann (0,82 1) tilsatt for å løse presipitert NMM-HC1. Produktet ble isolert ved hjelp av filtrering, vasket med 2-propanol (1 1) og tørket. Utbytte: 90 %. HPLC-renhet: 98,3 %.
EKSEMPEL 4. Boc- D- Nal- D- 4ClPhe- D- 3Pal- OH.
(a) Avbeskyttelse Boc-D-4ClPhe-D-3Pal-OH (447,93 g; 1,0 eq.) ble løst i en blanding av etylacetat (3,4 1), eddiksyre (675 ml) og MSA (454 ml; 7,0 eq.) ved 0°C og oppbevart ved denne temperaturen i 2 timer. c) Rensing og isolering. Reaksjonsblandingen ble ekstrahert to ganger ved pH 6 (2 x 4,5 1 vann) for å fjerne TEA, ved pH 9 (4,5 1 vann) for å fjerne MSA og endelig ved pH 7 (4,5 1 vann). Ekstraksjonene ble utført ved 40-45°C for å forhindre presipitering. Den organiske fasen ble tilsatt eddiksyre (4,5 1; 1 vol.) og blandingen ble konsentrert in vacuo og fordampet sammen med eddiksyre (4,5 1), hvilket ga et fast stoff.
EKSEMPEL 5. Ac- D- 2Nal- D- 4ClPhe- D- 3Pal- ONa.
a) Avbeskyttelse. Til det faste stoffet Boc-D-2Nal-D-4ClPhe-D-3Pal-OH ble vann (90 ml), eddiksyre (1,8 1) og MSA (454 ml; 7,0 eq.) tilsatt og blandingen ble rørt i
1-2 timer ved romtemperatur. Blandingen ble avkjølt til 0°C og nøytralisert med TEA (1071 ml; 7,7 eq.). Løsningen ble konsentrert in vacuo og fordampet sammen med toluen (2 x 2,5 1), hvilket ga en olje.
b) Acetylering. Oljen fra avbeskyttelsestrinnet ble løst i toluen (2,0 1) og acetylimidazol (132,14 g) ble tilsatt. Blandingen ble rørt ved romtemperatur i 1 time
og deretter ble vann (100 ml) tilsatt for å undertrykke gjenværende acetylimidazol.
d) Rensing. Blandingen fra acetyleringen ble varmet til 30-35°C og 1-butanol (4,5 1) ble tilsatt for å forhindre presipitering. Blandingen ble ekstrahert to ganger ved pH 5 (2 x 2,6 1 vann), og to ganger ved pH 11 (2 x 2,6 1 vann) ved anvendelse av NaOH for å justere pH til 11. Metanol (2,25 1) ble tilsatt til de siste ekstraksj onene for å forhindre presipitering. NaCl (130 g) ble tilsatt til den første og den siste ekstraksj onen for å minimalisere produkttap i de vandige fasene.
e) Isolering. Til den kraftige rørte organiske fasen fra ekstraksj onene ble heptan (15 1) tilsatt og den resulterende suspensjonen ble oppbevart ved romtemperatur under
røring i det minste i 1 time. Blandingen ble filtrert og produktet ble vasket to ganger med heptan (2 x 3,5 1) og tørket. Utbytte: 75 % (fra Boc-D-4ClPhe-D-3Pal-PH). HPLC-renhet: 92 %- Aminosyreanalyse: 2Nal: 1,1; 4ClPhe: 1,0; 3Pal: 0,9. MS: MW 586. Na: 4,6 %.
EKSEMPEL 6. Ac- D- 2Nal- D- 4ClPhe- D- 3Pal- OH- DCHA
a) Avbeskyttelse. Til det faste stoffet Boc-D-2Nal-D-4ClPhe-D-3Pal-OH ble vann (90 ml), eddiksyre (1,8 1) og MSA (454 ml; 7,0 eq.) tilsatt og blandingen ble rørt i
1-2 timer ved romtemperatur. Blandingen ble avkjølt til 0°C og nøytralisert med TEA (1071 ml; 7,7 eq.). Løsningen ble konsentrert in vacuo og fordampet to ganger sammen med toluen (2 x 2,5 1) hvilket ga en olje.

vann), én gang ved pH 9-9,5 (2,6 1 vann) og én gang ved pH 7 (2,6 1 vann). DCHA (disykloheksylamin) ble tilsatt og blandingen ble konsentrert in vacuo. Produktet ble suspendert i 1-butanol (4,5 1) ved 50°C og sakte tilsatt til kraftig rørt heptan (27
1). Blandingen ble rørt ved 0°C over natt, filtrert og produktet vasket to ganger med 1-butanol/heptan (1:3; 2 x 4,8 1) og to ganger med heptan (2 x 4,5 1). Utbytte: 65 %
(fra Boc-D-4ClPhe-D-3Pal-OH). HPLC-renhet: 94,2 %. Aminosyreanalyse: 2Nal: 1,1; 4ClPhe: 1,0; 3Pal: 0,9. MS: MW 586 (fritt peptid).
EKSEMPEL 7. Ac- D- 2Nal- D- 4ClPhe- D- 3Pal- OH.

Blandingen ble oppbevart under røring ved 0°C over natt. Blandingen ble filtrert, og produktet ble vasket to ganger med etylacetat/heptan (1:3; 2 x 3,6 1) og to ganger med heptan (2 x 3,6 1).
Utbytte: 70 % (fra Boc-D-4ClPhe-D-3Pal-OH). HPLC-renhet: 93,9 %. Aminosyreanalyse: Nal: 1,1; 4ClPhe: 1,0; 3Pal: 0,9 MS: MW 586 (fritt peptid).

1. Fremgangsmåte for fremstilling av et tripeptid, inkludert et salt derav, med formelen (I)
karakterisert vedat fremgangsmåten omfattende de følgende etterfølgende trinn for fremstillingen av (I): (a) omsetting av Boc-D-4ClPhe-OH med HONSu for å danne Boc-D-4ClPhe-OSu (VII); (b) omsetting av Boc-D-4ClPhe-OSu (VII) med H-D-3Pal-OH for å danne Boc-D-4ClPhe-D-3Pal-OH (VIII); (c) omsetting av Boc-D-4ClPhe-D-3Pal-OH (VIII) med Boc-D-2Nal-OSu fremstilt ved å omsette Boc-D-2Nal-OH med HONSu for å danne Boc-D-2Nal-D-4ClPhe-D-3Pal-OH (IX); (d) omsetting av Boc-D-2Nal-D-4ClPhe-D-3Pal-OH (IX) med eddiksyre for å danne Ac-D-2Nal-4ClPhe-D-3Pal-OH (I).
