Description
Foreliggende oppfinnelse angår en analogifremgangsmåte for fremstilling av terapeutisk brukbare [[[(3-pyridinyl)-metylen]amino]oksy]alkansyrer og estere derav.
Oppfinnelsen angår også mellomprodukter for anvendelse ved denne fremgangsmåte.
Et antall [[(3-pyridinyl)metylen]amino]oksyderivater er beskrevet i "J. Chem. Soc", Perkin Trans. I, (3), 984-987
(1981) og i "J. Med. Chem.", 7 (6), 824-826 (1964) som mellomprodukter eller som forbindelser med plantevekst-inhiberende og vitamin K-aktivitet.
I CH-PS 616.406 er det beskrevet et antall [[(3-pyridinyl)-metylen]amino]oksyderivater som er brukbare som lipidsenkende og plantebeskyttende midler.
I US-PS 4.518.602 er det beskrevet et antall vinylkarboksyl-syrederivater med tromboksan Å2-syntetaseinhiberende egenskaper.
I EP-publ. 0.175.304 er det beskrevet [[(metylen)amino]-oksy]propansyrer.
Forbindelsene som fremstilles ifølge oppfinnelsen skiller seg fra de angitte kjente forbindelser ved evnen til spesifikt å inhibere den enzymatiske syntese av tromboksan Å£ og ved arten av substituentene bundet til [(metylen)amino]oksy-gruppen.
Som nevnt ovenfor angår oppfinnelsen en analogifremgangsmåte for fremstilling av terapeutisk brukbare [[[(3-pyridinyl)-metylen]amino]oksy]alkansyrer og estere derav med formelen: farmasøytisk akseptable syreaddisjons- eller metallsubstitu-sjonssaltformer og stereokjemisk isomere former derav, der: R er hydrogen, Ci_ioalkyl, trifluormetyl, Ar eller (Ar)Ci_ løalkyl der Ar er fenyl, naftyl, pyridinyl eller tienyl hvorved fenyl eventuelt er substituert med opptil 3 substituenter uavhengig valgt blant C^.^alkyl, Ci.^alkoksy, mono-eller di(C^.^alkoksy)metyl, amino, Ci^alkylkarbonylamino, formyl, halogen, nitro og trifluormetyl;
R<1> er hydrogen eller C^.^alkyl; og
Alk er en C2_ioalkandiylrest» forutsatt at [[(3-pyridinyl)-metylen]amino]oksyresten og COOR<*> resten ikke er bundet til samme karbonatom.
I den foregående definisjon er uttrykket halogen ment å omfatte fluor, klor, brom og jod; "C^.^-alkyl" er ment å inkludere rette og forgrenede mettede hydrokarbonrester med fra 1-6 karbonatomer som for eksempel metyl, etyl, 1-metyl-etyl, 1,1-dimetyletyl, propyl, butyl, pentyl, heksyl og lignende; "Ci_io-alkyl" er ment å inkludere rette og forgrenede hydrokarbonrester med fra 1-10 karbonatomer; og
"C2_io-alkandiyl" er ment å inkludere toverdige rette eller forgrenede alkandiylrester med fra 2-10 karbonatomer.
N-oksydene av forbindelsene med formel I er ment å omfatte disse forbindelser med formel I der ett eller flere nitrogen-atomer, fortrinnsvis pyridinnitrogenatomer, er oksydert til den såkalte N-oksydform.
Foretrukne forbindelser innenfor oppfinnelsens ramme er de der [[(3-pyridinyl)metylen]amino]oksyrest og -COOR^-rest er adskilt med minst 3 og høyst 6 karbonatomer.
Spesielt foretrukne forbindelser er de der R er År eller Ar(C1_4-alkyl).
Mer spesielt foretrukne forbindelser er de spesielt foretrukne der R<1> er hydrogen og R er fenyl eller naftalenyl, hvori fenyl og naftalenyl hver eventuelt er substituert med opptil 2 substituenter som hver uavhengig er C^.^-alkyl, Ci_£,-alkyloksy, di(Ci_^,-alkyloksy )metyl, formyl, halogen eller trifluormetyl.
Helt spesielt foretrukne forbindelser innenfor oppfinnelsens ramme er de spesielt foretrukne forbindelser hvori pyridin-ringen og -0-Alk-COOR<1->substituentene på oksimdelen er i trans-konfigurasjon.
De mest foretrukne forbindelser er valgt blant (E)-5-[[[(3-pyridinyl) [3-( trif luormetyl) fenyl] metyl en] amino] oksy] pentan-syre og (E)-5-[[[(3-metylfenyl)(3-pyridinyl)metylen]amino]-oksy]pentansyre samt farmasøytisk akseptable salter og N-oksydformen derav.
Forbindelsene med formel I kan fremstilles ved
O-alkylering av et oksim med formelen
eller et metallsalt derav, med et mellomprodukt med formelen W-Alk-COOR<1> (III), der W betyr halogen, i et reaksjonsinert oppløsningsmiddel; og eventuelt omdanning av forbindelsene med formel I i hverandre ved å følge kjente funksjonell-gruppe-transformasjonsreaksjoner som (i) omdanning av forbindelsene der R<1> er Cj^alkyl til forbindelser med formel I der R<*> er hydrogen ved hydrolyse i et surt eller basisk vandig medium; (ii) omdanning av forbindelsene med formel I der R<1> er hydrogen til forbindelser med formel I der R<*> er C]_(,alkyl ved forestring med et C^.^alkanol i nærvær av en sur katalysator; (lii) redusering av forbindelsene med formel I der R er
nitrofenyl til forbindelser med formel I der R er aminofenyl ved reduksjon med natriumditionitt i en vandig alkohol; (iv) N-acylering av forbindelsene med formel I der R er aminofenyl til forbindelser med formel I der R er Ci_6alkylkarbonylaminofenyl ved omsetning med et
Ci_£,alkylsyreanhydrid; og, hvis ønskelig,
omdanning av forbindelsene med formel I til et syreaddisjons-eller metallsubstitusjonssalt ved behandling med en egnet syre eller base; og, hvis ønskelig,
fremstilling av stereokjemisk isomere former derav.
O-alkyleringsreaksjonene gjennomføres hensiktsmessig i et egnet reaksjonsinert oppløsningsmiddel eller en blanding av slike. Et egnet reaksjonsinert oppløsningsmiddel kan for eksempel være et aromatisk hydrokarbon som benzen, metylbenzen, dimetylbenzen og lignende; en laverealkanol som metanol, etanol, 1-butanol og lignende; et keton som 2-propanon, 4-metyl-2-pentanon og lignende; en eter som 1,4-dioksan, 1,1'-oksybisetan, tetrahydrofuran og lignende; et polart aprotisk oppløsningsmiddel, for eksempel N,N-dimetyl-formamid (DMF), N,N-dimetylacetamid (DMA), heksametyl-fosforsyretriamid (HMPT), dimetylsulfoksyd (DMSO), nitro-benzen, l-metyl-2-pyrrolidinon og lignende. Tilsetningen av en egnet base som for eksempel et alkalimetallkarbonat eller
-hydrogenkarbonat, natriumhydrid eller en organisk base som for eksempel N,N-dietyletanamin eller N-(1-metyletyl)-2-propanamin kan benyttes for å fange opp syre som settes fri
linder reaksjonen. Noe forhøyede temperaturer kan øke reaksjonshastigheten.
Det kan være hensiktsmessig på forhånd å omdanne oksimet med formel II til en metallsaltform derav, fortrinnsvis alkali-metallsaltformen, ved å omsette utgangsoksimet med for eksempel et alkalimetallhydrid eller et jordalkalimetall-hydrid, for eksempel natriumhydrid, kalsiumhydrid og lignende, eller et alkalimetallhydroksyd, for eksempel natriumhydroksyd, kaliumhydroksyd og lignende, og derefter å benytte metallsaltformen i reaksjonen med III.
I alle de foregående og de følgende preparater kan reaksjons-produktene isoleres fra reaksjonsblandingen og, hvis nødven-dig, renses ytterligere i henhold til generelt kjente metoder.
Fra formel I er det åpenbart at forbindelsene som fremstilles ifølge oppfinnelsen kan være til stede som E- eller Z-former, idet E- og Z-angivelsen er i overensstemmelse med de regler som er gitt i "International Union of Pure og Applied Chemistry: Tentative Rules for the Nomenclature of Organic Chemistry. Section E. Fundamental Stereochemistry" i "J. Org. Chem.", 35» 2849-2867 (1970).
Avhengig av arten av substituentene på oksimdelen kan det være fordelaktig å underkaste en gitt blanding av E- og Z-isomerer en isomeriseringsreaksjon før separering av de rene geometriske isomerer. Isomeriseringsreaksjonen er en like-vektsreaksjon som generelt gjennomføres i nærvær av en mineralsyre.
Rene stereokjemisk isomere former av forbindelsene med formel I og mellomprodukter med formel II kan oppnås ved anvendelse av kjente separeringsmetoder som selektiv krystallisering og kromatografiteknikker slik som motstrømsfordeling, kolonnekromatografi, høyytelses-væskekromatografi og lignende. Stereokjemisk isomere former av forbindelsene med formel I ligger naturligvis innenfor oppfinnelsens ramme.
Forbindelsene med formel I kan omdannes til sine terapeutisk aktive ikke-toksiske syreaddisjonssalter ved behandling med egnede syrer, som for eksempel uorganiske syrer som hydrogen-halogensyrer som saltsyre, hydrobromsyre og lignende, og svovelsyre, salpetersyre, fosforsyre og lignende; eller organiske syrer som for eksempel eddik-, propan-, hydroksy-eddik-, 2-hydroksy-propan-, 2-oksopropan-, etandion-, propandion-, butandion-, (Z )-2-butendion-, (E)-2-butendion-, 2-hydroksybutandion-, 2,3-dihydroksybutandion-, 2-hydroksy-1,2,3-propantrikarboksyl-, metansulfon-, etansulfon-, benzensulfon-, 4-metylbenzensulfon-, cykloheksansulfamin-, benzo-, 3-fenyl-2-propen-, a-hydroksybenzeneddik-,
2-hydroksybenzo-, 4-amino-2-hydroksyenzo- og lignende syrer. Omvendt kan saltformen omdannes ved behandling med alikali til den frie baseform.
Forbindelsene med formel I inneholdende et eller flere sure protoner kan også omdannes til sine terapeutisk aktive ikke-toksiske metall- eller amin-substitusjonssaltformer ved behandling med egnede organiske eller uorganiske baser. Egnede uorganiske baser kan for eksempel være ammoniakk eller baser avledet fra alkali- eller jordalkalimetaller som alkalimetall- eller jordalkalimetalloksyder eller —hydrok-syder som litiumhydroksyd, natriumhydroksyd, kaliumhydroksyd, magnesiumhydroksyd, kalsiumhydroksyd, kalsiumoksyd og lignende; alkalimetall- eller jordalkalimetallhydrider som natriumhydrid eller kaliumhydrid og lignende; alkalimetall-hydrogenkarbonater eller -karbonater, for eksempel natrium-eller kaliumkarbonat, natriumhydrogenkarbonat, kalsium-karbonat og lignende. Egnede organiske baser kan f.eks være primære, sekundære eller tertiære alifatiske eller aromatiske aminer som for eksempel metylamin, etylamin, propylamin, isopropylamin, de fire butylamin-isomerer, dimetylamin, dietylamin, dietanolamin, dipropylamin, diisopropylamin, di-n-butylamin, pyrrolidin, piperidin, morfolin, N-metyl-morfolin, trimetylamin, tripropylamin, kinuklidin, pyridin, kinolin, isokinolin, dietanolamin og 1,4-diazabicyklo-[2,2,2]oktan; eller kvaternære ammoniumbaser som tetrametyl-ammonlumhydroksyd, trimetylbenzylajnmoniumhydroksyd, trletyl-benzylammoniumhydroksyd, tetraetylammonlumhydroksyd og trimetyletylammoniumhydroksyd. Omvendt kan saltformen omdannes ved behandling med syre til fri syreform.
Som nevnt ovenfor angår oppfinnelsen også et mellomprodukt for anvendelse ved fremgangsmåten for fremstilling av forbindelser med formel I, og dette mellomprodukt karakte-riseres ved formelen:
eller en stereokjemisk isomer form derav, der R er fenyl substituert med opptil 2 substituenter valgt blant Ci.fcalkyl, Cj^alkoksy, di(Ci_(,alkoksy )metyl, halogen og trifluormetyl, forutsatt at R er forskjellig fra 4-butyl-fenyl, 2-klorfenyl og 2,5-dimetylfenyl.
Mellomproduktene med formel II kan fremstilles fra de tilsvarende karbonylforbindelser med formel IV eller N-oksydet derav ved omsetning av det sistnevnte med hydroksylamin, IX, eller et syreaddisjonssalt derav:
Forbindelsene med formel I, deres N-oksydformer, syreaddisjons-, metall- eller aminsubstitusjonssalter eller stereokjemisk isomere former har brukbare farmakologiske egenskaper. De brukbare farmakologiske egenskaper kan for eksempel vises ved "platemalondialdehyd (MDA) produksjon"-prøven, beskrevet nedenfor, og som viser tromboksan-syntetase inhiberende egenskaper for forbindelsene som fremstilles ifølge oppfinnelsen.
I tillegg til det ovenfor angitte kan den selektive inhibi-toriske virkning på biosyntesen av tromboksan Ag og den økende virkning på fremstillingen av prostacyklin og prostaglandin Dg, Eg og F2a, for eksempel vises ved blodplateaggre-geringsforsøk ved bruk av forskjellige induktører slik som for eksempel kollagen, arachidonsyre og lignende; eller i forsøk som måler mellomproduktene i arachidonsyremetabolismen ved bruk av merket arachidonsyre eller radioimmunoanalyse.
Forbindelsene med formel I, deres N-oksyder, deres farmasøy-tisk aksepterbare syreaddisjons-, metall- eller amin-sustitu-sjonssalter og deres mulige stereokjemisk isomere former, viser en selektiv inhiberende virkning på biosyntesen av tromboksan Ag og også en økende virkning på fremstilling av prostacyklin og prostaglandin Dg, Eg og Fga, og virker like godt på blodplatene som på karveggen.
I lys av evnen til selektivt å inhibere biosyntesen av tromboksan Ag, og den økende virkning på produksjon av prostacyklin og prostaglandin Dg, Eg og Fga, kan forbindelsene som fremstilles ifølge oppfinnelsen benyttes som inhibitorer på vasokonstriksjon og på blodplateaggregering, hvilke virkninger er kjent og frembringes av tromboksan Ag, prostacyklin og/eller prostaglandin Dg, Eg og Fga ("Science", 193. 1135-1137 (1976), "Pharmacolog. Rev.", 30, 293-331
(1979).
Fagmannen på behandling av disse kliniske tilstander vil lett bestemme den effektive mengde ut fra prøveresultatene som her gis. Det er generelt ansett at en effektiv mengde vil ligge innen området 0,005 - 20 mg/kg kroppsvekt, fortrinnsvis fra 0,01 - 10 mg/kg.
De følgende eksempler skal illustrere oppfinnelsen uten å begrense den, hvis ikke annet er sagt, er alle deler angitt på vektbasis.
EKSPERIMENTELL DEL
A. FREMSTILLING AV MELLOMPRODUKTER
Eksempel 1
Til en omrørt og til -78° C avkjølt blanding av 50 deler 1-brom-2-(dimetoksymetyl)benzen og 245 deler 1,1'-oksybisetan ble det dråpevis tilsatt 59 deler av en 2,5M 1-butyllitium-oppløsning i heksan. Efter ferdig tilsetning ble omrøringen fortsatt i 10 minutter ved romtemperatur. En oppløsning av 23,1 deler 3-pyridinkarboksaldehyd i 42 deler 1,1'-oksybisetan ble dråpevis tilsatt til reaksjonsblandingen. Efter ferdig tilsetning ble blandingen tillatt å nå romtemperatur. Blandingen ble tatt opp i isvann og produktet ekstrahert med 1,1'-oksybisetan. Ekstrakten ble tørket, filtrert og fordampet, og man oppnådde 56 deler (99, 9S6) a-[2-(dimetoksy-metyl)fenyl]-3-pyridinmetanol som en rest (mellomprodukt 1).
På samme måte ble det også fremstilt: a-[3-(dimetoksymetyl)fenyl]-3-pyridinmetanol som en rest (mellomprodukt 2).
Eksempel 2
Til en omrørt og til ca. -70 - 80°C avkjølt oppløsning av 71,4 deler av en 1,6M l-butyllitiumoppløsnlng i heksan i 105 deler 1,1'-oksybisetan ble det dråpevis tilsatt en oppløsning av 40,6 deler 3-brompyridin i 70 deler 1,1'-oksybisetan. Efter ferdig tilsetning ble omrøringen fortsatt i 15 minutter og en oppløsning av 30 deler 2-(trifluormetyl)benzonitril i 35 deler 1,1'-oksybisetan ble tilsatt dråpevis ved denne lave temperatur. Efter ferdig tilsetning ble blandingen omrørt i 3 timer ved denne lave temperatur. Blandingen ble oppvarmet til romtemperatur og helt i 84 deler ION saltsyreoppløsning og noe knust is. Det vandige sjikt ble separert og oppvarmet til kokepunktet. Efter avkjøling ble blandingen gjort alkalisk med en ammoniumhydroksydoppløsning og produktet ble ekstrahert med diklormetan. Ekstrakten ble tørket, filtrert og konsentrert. Resten ble renset ved filtrering over silikagel ved bruk av triklormetan som elueringsmiddel. De rene fraksjoner ble samlet og elueringsmidlet ble konsentrert og man oppnådde 27,8 deler (63*) (3-pyridinyl)[2-(trifluormetyl ) fenyl ] met anon som en oljeaktig rest (mellomprodukt 3).
På samme måte ble det også fremstilt: [3,5-bis(trifluormetyl)fenyl](3-pyridinyl)metanon som en rest (mellomprodukt 4).
Eksempel 3
Til en omrørt oppløsning av 12 deler magnesium og 70 deler 1,1'-oksybisetan ble det dråpevis tilsatt 88 deler 1-brom-oktan under avkjøling og under en nitrogenatmosfære. Efter ferdig tilsetning ble blandingen oppvarmet i en time under tilbakeløpstemperatur. 1,1'-oksybisetan ble fjernet og en blanding av 40 deler 3-pyridinkarbonitril og 180 deler benzen ble tilsatt. Det hele ble oppvarmet i 2 timer under tilbake-løp og efter avkjøling ble blandingen helt i 400 deler isvann. Den separert vandige fase ble behandlet med ammoniumhydroksyd og produktet ekstrahert tre ganger med 150 deler triklormetan. De kombinerte ekstrakter ble tørket, filtrert og fordampet og man oppnådde 70,7 deler (83 , 956) l-(3-pyridinyl)-l-nonanon som en rest (mellomprodukt 5).
Eksempel 4
En blanding av 19 deler a-[3-(dimetoksymetyl)fenyl]-3-pyridinmetano, 36 deler aktivert mangan(IV)oksyd og 280 deler diklormetan ble omrørt i helgen ved romtemperatur. Reaksjonsblandingen ble filtrert over silikagel og filtratet filtrert igjen over diatomejord og silikagel. Filtratet ble fordampet og resten destillert i vakuum. Den ønskede fraksjon ble samlet og man oppnådde 15,7 deler (82*) [3-(dimetoksymetyl)-fenyl](3-pyridinyl)metanon (mellomprodukt 6).
På samme måte ble det også fremstilt: [2-(dimetoksymetyl)fenyl](3-pyridinyl)metanon; kokepunkt 142-147°C (mellomprodukt 7).
Eksempel 5
Til en omrørt og til 0°C avkjølt oppløsning av 70 deler 2-fenyl-l-(3-pyridinyl)etanon i 225 deler N,N-dimetylformamid ble det dråpevis tilsatt 46,9 deler 2-metyl-2-propanol, kaliumsalt under nitrogen. Eksempel 6
En blanding av 27,6 deler [3-(trifluormetyl)fenyl]-(3-pyridinyl)etanon, 8,3 deler hydroksylaminhydroklorid, 6,4 deler natriumkarbonat og 160 deler etanol ble omrørt i 24 timer ved tilbakeløpstemperatur. Efter avkjøling ble utfelt produkt filtrert av, satt til side og vasket med vann. Etanolsjiktet ble konsentrert og den gjenværende rest ble også vasket med vann, kombinert med den første fraksjon som ble satt til side (se ovenfor) og krystallisert fra 120 deler 4-metyl-2-pentanon. Produktet ble filtrert av og tørket og man oppnådde 20 deler (68*) (E+Z)-(3-pyridinyl)[3-(trifluormetyl)fenyl]metanon,oksim; smeltepunkt 185,4°C (mellomprodukt 9).

Eksempel 7
En blanding av 70 deler l-(3-pyridinyl)-l-nonanon, 44,8 deler hydroksylaminhydroklorid, 88,2 deler kaliumkarbonat og 320 deler etanol ble omrørt i 2 timer ved tilbakeløpstemperatur. Efter fordamping til tørr tilstand ble resten helt i 1000 deler vann og produktet ekstrahert tre ganger med 105 deler 1,1'-oksybisetan. De kombinerte ekstrakter ble tørket, filtrert og fordampet og man oppnådde 68,5 deler (91,6*) 1-(3-pyridinyl)-l-nonanon,oksim som en rest (mellomprodukt 18).
På samme måte ble det også fremstilt:

Til en omrørt blanding av 4,3 deler (4-metoksyfenyl)-(3-pyridinyl)etanon, 1,53 deler hydroksylaminhydroklorid og 80 deler etanol ble det dråpevis tilsatt 2,1 deler N,N-dietyl-etanamin ved romtemperatur. Efter ferdig tilsetning ble omrøringen fortsatt over natten under tilbakeløpstemperatur. Reaksjonsblandingen ble fordampet. Resten ble tatt opp i vann under omrøring. Produktet ble filtrert av, vasket med vann og krystallisert to ganger: først fra acetonitril og så fra etylacetat. Produktet ble filtrert av, vasket med etylacetat og 2,2'-oksybispropan og tørket under vakuum ved 60"c, noe som ga 1,68 deler (36,8*) (E+Z )-(4-metoksyfenyl)(3-pyridinyl)metanon,oksim; smeltepunkt 176,0°C

Eksempel 9
Til en omrørt blanding av 54,2 deler (3-metylfenyl) (3-pyridinyl)metanon, 25,2 deler natriumhydrogenkarbonat og 480 deler etanol ble det tilsatt 20,8 deler hydroksylaminhydroklorid. Omrøringen ble fortsatt over natten under tilbake-løpstemperatur. Utfelt produkt ble filtrert av, filtratet ble satt til side, og omrørt i vann. Filtratet som ble satt til side (se ovenfor) ble fordampet. Resten ble omrørt i vann. De vandige oppløsninger ble kombinert. En blanding av 9 deler [3,5-bis(trifluormetyl)fenyl](3-pyridinyl)metanon, 2,09 deler hydroksylaminhydroklorid, 2,46 deler natriumacetat, 60 deler metanol og 50 deler vann ble omrørt i 8 timer under tilbakeløpstemperatur. Etanolsjiktet ble fordampet. Utfelt produkt ble filtrert av, vasket suksessivt med vann, 2-propanol og 2,2'-oksybispropan og tørket, noe som ga 7,6 deler (81*) [3,5-bis(trifluormetyl)-fenyl](3-pyridinyl)metanon,oksim; smeltepunkt 188°C (mellomprodukt 25).
På samme måte ble det også fremstilt: (E+Z)-(3-pyridinyl)[2-(trifluormetyl)fenyl]metanon,oksim som en rest (mellomprodukt 26).
Eksempel 11
En blanding av 40 deler 2-fenyl-l-(3-pyridinyl)etanon, 36,8 deler 1-bromheksan, 25 deler 2-metyl-2-propanol, kaliumsalt og 200 deler dimetylsulfoksyd ble omrørt i 2 timer ved romtemperatur under nitrogen. Produktet ble ekstrahert med etylacetat. Ekstraktet ble vasket med en fortynnet natrium-hydrogenkarbonatoppløsning, tørket, filtrert og fordampet. Resten ble tatt opp i 160 deler etanol og 32 deler hydroksylaminhydroklorid ble tilsatt. Efter omrøring i en time ved 50°C ble blandingen fordampet og produktet ekstrahert med etylacetat. Ekstraktet ble tørket, filtrert og fordampet. Resten ble renset ved kolonnekromatografi over silikagel ved bruk av triklormetan som elueringsmiddel. De rene fraksjoner ble samlet og elueringsmidlet fordampet, noe som ga 7,8 deler (13*) (E+Z)-2-fenyl-l-(3-pyridinyl)-l-oktanon,oksim som en rest (mellomprodukt 27).
B. FREMSTILLING AV SLUTTFORBINDELSER
Eksempel 12
Til en omrørt blanding av 5 deler (E)-3-pyridinkarboksaldehyd,oksim og 90 deler N,N-dimetylformamid ble det porsjonsvis tilsatt 4,02 deler 50 *-ig natriumhydrid-dispersjon ved romtemperatur. Efter ferdig tilsetning ble omrøringen fortsatt ved 40"C inntil hydrogengassutviklingen ga seg. Efter avkjøling ble en oppløsning av 8,3 deler 6-bromheksansyre I 13,5 deler N,N-dimetylformamid tilsatt dråpevis ved 15°C. Efter ferdig tilsetning ble reaksjonsblandingen omrørt I 30 minutter. Den ble helt i vann og surgjort med saltsyre til pH - 5. Det utfelte produkt ble filtrert av og krystallisert fra en blanding av vann og 2-propanol, hvorved man oppnådde 6,37 deler (65,9*) (E)-6-[C[(3-pyridinyl)metylen]amino]oksy]heksansyre; smeltepunkt 117,1°C (forbindelse 1).
På tilsvarende måte ble det også fremstilt: (E)-5-[[[(3-pyridinyl)metylen]amino]oksy]pentansyre, smeltepunkt 126,3°C (forbindelse 2); og (E)-3-[[[(3-pyridinyl)metylen]amino]oksy]propansyre, smeltepunkt 155,5<*>C (forbindelse 3).
Eksempel 13
Til en omrørt blanding av 8 deler (E+Z)-fenyl(3-pyridinyl)-metanon,oksim og 108 deler N,N-dimetylformamid ble det porsjonsvis tilsatt 4,0 deler av en 50 *-ig natriumhydrid-dispersjon ved romtemperatur. Efter ferdig tilsetning ble omrøringen fortsatt inntil hydrogengassutviklingen ga seg. En oppløsning av 8,3 deler 5-brompentansyre i 18 deler N,N-dimetylformamid ble tilsatt dråpevis ved romtemperatur. Efter ferdig tilsetning ble reaksjonsblandingen omrørt og oppvarmet over natten ved 80°C. Oppløsningsmidlet ble fordampet. Resten ble tatt opp i en natriumhydrogenkarbonatoppløsning i vann og ekstrahert tre ganger med diklormetan. pH-verdien for den separerte vandige/NaHC03~fase ble bragt til 5 ved hjelp av saltsyreoppløsning. Produktet ble ekstrahert med diklormetan. Ekstrakten ble tørket, filtrert og fordampet. Resten ble renset ved kolonnekromatografi over silikagel ved bruk av triklormetanrmetanol i volumforholdet 98:2. De rene fraksjoner ble samlet og elueringsmidlet fordampet. Resten ble tatt opp i 2-propanol og det sistnevnte fordampet. Resten ble omrørt i en blanding av etylacetat og 2,2'-oksybispropan. Produktet ble filtrert av (filtratet satt til side), vasket med 2,2'-oksybispropan og tørket i vakuum ved 70"C, noe som ga en første fraksjon på 4,3 deler (35,8*) (E+Z)-5-[[[fenyl-(3-pyridinyl)metylen]amino]oksy]pentansyre; smeltepunkt 97,4°C. Filtratet som ble satt til side, ble fordampet. Resten ble tørket i vakuum ved 60°C og man oppnådde en andre fraksjon på 0,8 deler (6,6*) (E+Z)-5-[[[fenyl(3-pyridinyl)-metylen]amino]oksy]pentansyre; smeltepunkt 92,4°C (forbindelse 4).

Eksempel 14
Til en omrørt blanding av 4,08 deler (E)-l-(3-pyridinyl)-etanon,oksim og 45 deler N,N-dimetylformamid ble det porsjonsvis tilsatt 1,44 deler 50 *-ig natriumhydriddisper-sjon ved romtemperatur. Blandingen ble omrørt inntil total reaksjon og oppvarmet en stund i et vannbad ved 40-50°C. Efter avkjøling ble 6,9 deler etyl-5-brompentanoat dråpevis tilsatt ved romtemperatur, eksoterm reaksjon. Blandingen ble satt hen over natten. Efter fordamping ble vann tilsatt og produktet ekstrahert med diklormetan. Ekstraktet ble tørket, filtrert og konsentrert. Konsentratet ble renset ved kolonnekromatografi over silikagel ved bruk av triklormetan tmetanol i volumforholdet 95:5 som elueringsmiddel. En blanding av 7,6 deler etyl-(E+Z)-6-[[[2-fenyl-l-(3-pyridinyl)propyliden]amino]oksy]heksanoat og 100 deler 3N natriumhydroksydoppløsning ble omrørt over natten ved romtemperatur. Reaksjonsblandingen ble nøytralisert med 3N saltsyreoppløsning til pH 5,5. Reaksjonsblandingen ble konsentrert og konsentratet oppløst i 2-propanon. På tilsvarende måte ble det også fremstilt: nat r ium - (E)-5-[[[f enyl(3-pyr idinyl)metylen]amino]oksy]-pentanoat; smeltepunkt 153,4°C (forbindelse 122); na t r ium- (E )-5 - [ [ [ (1-naftalenyl)(3-pyridinyl )me tylen] amino] - oksy]pentanoat; smeltepunkt 172,0"C (forbindelse 123); og på tilsvarende måte ble det også fremstilt: natrium-5 - [ [ [ ( 4-metoksy-1-naftalenyl)(3-pyridinyl )metylen]-amino]oksy]pentanoat (forbindelse 124).
Eksempel 26
Til en omrørt oppløsning av 1 del (E)-5-[[[(1-naftalenyl)(3-pyridinyl )metylen]amino]oksy]pentansyre i 75 deler triklormetan ble det porsjonsvis tilsatt 0,625 deler 3-klorbenzen-karboperoksosyre ved 0-5°C. Efter ferdig tilsetning ble reaksjonsblandingen tillatt å innta romtemperatur. Efter fordamping ble resten krystallisert to ganger fra etylacetat. Produktet ble filtrert av, vasket med etylacetat og 2,2'-oksybispropan og tørket i vakuum ved 50°C, noe som ga 0,64 deler (61,1*) (E)-5-[[[(l-naftalenyl)(3-pyridinyl)metylen]-amino]oksy]pentansyre, N-oksyd; smeltepunkt 165,6°C (forbindelse 125).
Eksempel 27
En blanding av 6,5 deler sølv(I)oksyd, 50 deler vann, 63 deler etanol og 6,5 deler 5-[[[(3-formylfenyl)(3-pyridinyl)-metylen]amino]oksy]pentansyre omrøres i en time ved noe forhøyet temperatur hvorved man oppnår 3-[[(4-karboksy-butoksy)imino](3-pyridinyl)metyl]benzosyre (forbindelse 126).
C. FARMAKOLOGISKE EKSEMPLER
Eksempel 28
Platemalondialdeh<y>d ( MDA) produksjon
MDA-produksjon av plater i plasma, som indikator på tromboksan Ag-biosyntesen, ble evaluert spektrofluometrisk idet 0,4 ml citrert platerikt plasma (250 x IO<3> plater/pl) i nærvær av oppløsningsmiddel (dimetylsulfoksyd 0,2*) eller en forbindelsesoppløsning ble preinkubert i 10 minutter ved 37°C og så prøvet med 50 pl CaClg 1 niM sluttkonsentrasjon og 50 pl trombin 20 N.I.H. enheter/ml sluttkonsentrasjon. Efter 30 minutter ved 37°C ble reaksjonen avsluttet ved tilsetning av 1 ml 10* vekt/volum trikloreddiksyre. Efter isolering av supernatanten ved sentrifugering ved 320 g pr. 20 minutter, ble tiobarbitursyrereaksjonen på MDA gjennomført ved bruk av syrehydrolysert 1,1,3,3-tetrametoksypropan som standard. Fluorescensen av prøvene ved 553 nm efter eksitering ved 510 nm ble målt. Resultatene ble beregnet som nM MDA/10<8> plater pr. 30 minutter og ble uttrykt som prosentandel Inhibering produsert av forbindelsene i forhold til dannelsen av MDA efter trombinstimulering i oppløsningsmidlet. For in vitro-forsøk ble det benyttet humanplater 1 plasma. Prosentandel inhlbering ble beregnet mot oppløsnlngsmlddelverdien. IC50-verdler ble bestemt som de doser 1 pM som ga 50* inhlbering.
For in vivo-forsøk ble Wistar-hannrotter fastet over natten. 2 timer før blodprøving ble de behandlet med prøveforbin-delsesoppløsningen/-suspensjonen eller oppløsningsmiddel. En 50* inhlbering i forhold til MDA-verdiene som ble oppnådd med oppløsningsmidlet ble ansett som kriterium for inhiberende aktivitet. EDsQ-verdiene ble bestemt ved probit-analyse. ICsQ-verdiene og EDsg-verdiene for et antall forbindelser med formel I er angitt i tabellen nedenfor.
Claims (8)
1.
Analogifremgangsmåte for fremstilling av terapeutisk brukbare [[[(3-pyridinyl)metylen]amino]oksy]alkansyrer og estere derav med formelen:
farmasøytisk akseptable syreaddisjons- eller metall substitusjonssaltformer og stereokjemisk isomere former derav, der:
R er hydrogen, C^igalkyl, trifluormetyl, Ar eller (Ar)C^_ løalkyl der Ar er fenyl, naftyl, pyridinyl eller tienyl hvorved fenyl eventuelt er substituert med opptil 3 substituenter uavhengig valgt blant C^.^alkyl, C^_£,alkoksy, mono-eller di(C^.^alkoksy)metyl, amino, C^.^alkylkarbonylamino, formyl, halogen, nitro og trifluormetyl;
r<1> er hydrogen eller C^.^alkyl; og
Alk er en C2_ioalkandlylres"t» forutsatt at [[(3-pyridinyl)-metylen]amino]oksyresten og COOR<1> resten ikke er bundet til samme karbonatom,
karakterisert ved
O-alkylering av et oksim med formelen
eller et metallsalt derav, med et mellomprodukt med formelen W-Alk-COOR<1> (III), der W betyr halogen, i et reaksjonsinert oppløsningsmiddel; og eventuelt omdanning av forbindelsene med formel (I) 1 hverandre ved å følge kjente funksjonell-gruppe-transformasjonsreaksjoner som (i) omdanning av forbindelsene der R<1> er C-^fcalkyl til forbindelser med formel (I) der R<1> er hydrogen ved hydrolyse i et surt eller basisk vandig medium; (ii) omdanning av forbindelsene med formel (I) der R<1> er hydrogen til forbindelser med formel (I) der R<1> er C]_6alkyl ved forestring med et C^.^alkanol i nærvær av en sur katalysator; (ili) redusering av forbindelsene med formel (I) der R er
nitrofenyl til forbindelser med formel (I) der R er aminofenyl ved reduksjon med natriumditionitt i en vandig alkohol; (iv) N-acylering av forbindelsene med formel (I) der R er aminofenyl til forbindelser med formel (I) der R er C]_6alkylkarbonylaminofenyl ved omsetning med et C]_6alkylsyreanhydrid; og, hvis ønskelig,
omdanning av forbindelsene med formel (I) til et syreaddisjons-eller metallsubstitusjonssalt ved behandling med en egnet syre eller base; og, hvis ønskelig,
fremstilling av stereokjemisk isomere former derav.

Analogifremgangsmåte ifølge krav 1 for fremstilling av terapeutisk brukbare (E)-5-[[[(3-pyridinyl)[3-(trifluormetyl)fenyl]-metylen]amino]oksy]pentansyre eller farmasøytisk akseptable syreaddisjons- eller metallsubstitusjonssalter derav .karakterisert ved at man går ut fra de tilsvarende utgangsforbindelser.

Analogifremgangsmåte ifølge krav 1 for fremstilling av terapeutiske brukbare (E)-5[[[(3-metylfenyl)-(3-pyridinyl )-metylen]amino]oksy]pentansyre eller et farmasøytisk akseptabelt syreaddisjonssalt eller et metallsubstitusjonssalt derav, karakterisert ved at man går ut fra de tilsvarende utgangsforbindelser.

Mellomprodukt for anvendelse ved fremgangsmåten Ifølge krav 1,karakterisert ved formelen
eller en stereokjemisk isomer form derav, der R er fenyl substituert med opptil 2 substituenter valgt blant <C>^.^alkyl, Ci^alkoksy, di(C^.^alkoksy )metyl, halogen og trifluormetyl, forutsatt at R er forskjellig fra 4-butyl-fenyl, 2-klorfenyl og 2,5-dimetylfenyl.
