Description
Den her beskrevne oppfinnelse vedrører monoklonale antistoffer som reagerer spesifikt mot sporozoitter av parasitten Eimeria tenella. Det beskrives hybridomakulturer som produserer antistoffer mot E. tenella. Slike antistoffer fås ved hjelp av hybridomateknologi. Sporozoitt-antigener identifiseres og karakteriseres.
Nærmere bestemt vedrører oppfinnelsen monoklonale antistoffer for bruk in vitro fremstilt ved hjelp av hybridomas dannet ved sammensmelting av celler fra en mus-uiyelomalinje og miltceller fra en mus som tidligere er blitt immunisert med sporozoitter av Eimeria tenella. Antistoffene er karakterisert ved at de (a) reagerer spesifikt med antigener fra Eimeria spp. sporozoitter, (b) reagerer spesifikt med antigener fra Eimeria tenella som har en molekylvekt på omtrent 13 til 150 kd, og (c) de er fremstilt fra hybridomas dannet ved sammen-smelting av p3 x 63.Ag8.653 myelomaceller og miltceller fra BALB/c-mus som tidligere er blitt immunisert med E. tenella sporozoitter, hvilke hybridomas betegnes klon nr. s5E5 og er deponert som ATCC nr. HB8403; klon nr. slC4 og er deponert som ATCC nr. HB83 33; klon nr. s2G8 og er deponert som ATCC nr. HB8405; klon nr. s5B9 og er deponert som ATCC nr. HB8402; klon nr. slA og er deponert som ATCC nr. HB8404; klon nr. s3D3 og er deponert som ATCC nr. HB8331; eller klon nr. slE4 og er deponert som ATCC nr. HB8332.
Coccidiose er en sykdom hos dyr forårsaket av mange forskjellige protozo-parasitter. Fugle-coccidiose er en ødeleggende sykdom hos fjærfe forårsaket av mange forskjellige arter av slekten Eimeria. Denne sykdommen har en komplisert livssyklus bestående av både aseksuelle og seksuelle stadier. Kyllinger infiseres i utgangspunktet med sykdommen etter inntak av frittlevende oocyster som vanligvis er forbundet med avføringsmateriale. Oocyster utvikler seg til invaderende aseksuelle sporozoitter i kyllingens fordøyelseskanal. Sporozoittene infiserer epitelceller og utvikler seg til flerkjernede former kjent som schizonter. Hver schizont modnes og frigjør eventuelt flere invaderende, aseksuelle former kjent som merozoitter. Disse merozoittene forlater den infiserte cellen og invaderer på nytt andre epitelceller. De flerfoldige invaderende aseksuelle stadiene som omfatter sporozoitter og merozoitter, svarer for mye av patologien ved coccidiose. Coccidioses seksuelle syklus startes opp når merozoitter differensieres til gametocytter. Befruktning skjer, og befruktningsprodukter kjent som oocyster frigjøres i avføringen. Parasittens livssyklus er følgelig komplisert. Hos kyllinger fullføres livssyklusen til Eimeria tenella, en representativ art, i løpet av ca. 7 til 9 dager.
På grunn av de voldsomme økonomiske tapene som påføres fjærfenæringen av Eimeria-arter, er era vaksine mot parasitten svært ønskelig. På grunn av. kompleksiteten til parasittens livssyklus og variasjonen i mengde antigener til stede i hvert stadium, er det imidlertid tidligere blitt observert at de aktiverte eller drepte parasitter ikke har frembragt homogen immunitet. En løsning på dette problemet er å isolere og karakterisere bestemte antigener fra parasitten og administrere disse i en tilstrekkelig mengde til å tjene som et immuniserende middel. Slike antigener vil fortrinnsvis gis beskyttelse mot infeksjon fra alle viktige arter. Det er kjent at forskjellige arter av Eimeria, såvel som forskjellige stadier i livssyklusen til de samme artene, både har felles og spesifikke antigener (Cerna, Z., Folia Prasitologica (Prague) 17: 135-140 (1970; Davis et al., Immunol..34: 879-888 (1978); Rose, M.E., Immunol. 2: 112-122 (1959),;, Rose et al., Immunol. 5: 79-92 (1963); og Tanielian et al'. „ Acta Parasitol. Det er også kjent at utvikling* av immunitet mot Eimeria er arts-spesifikk ogj at. det hos noen arter av tamme fugler er betydelig stammespesifikk immunitet ('Jfe-ffers", T.K. ; In Long, P..L. et al. ;(eds.),. Aktuelle immunogener fra Eimeria-arter som er i stand til å stimulere beskyttende immunitet hos fjærfe- eller pattedyr-verter er ennå ikke blitt isolert eller identifisert. Slike Eimeria-immunogener vil sannsynligvis gi vellykket immunisering mot coccidiose. ;Utviklingen av lymfocytt-hybridoma-teknologi gir et ;redskap for å produsere forholdsvis store mengder spesifikke antistoffer mot forskjellige antigener hos Eimeria. Ved å ;smelte sammen bestemte antistoff-produserende celler (miltceller) med celler fra en myeloma-tumor er det mulig å produsere hybridoma-celler som utskiller monoklonale antistoffer rettet spesifikt mot det opprinnelige sensibiliserende antigen. ;(Kohler & Milstein, Nature (London) 256: 495-497 (1975)). ;Forskjellige patenter vedrørende hybridomakulturer og monoklonale antistoffer er kjent (US-patentskrift nr. 4.172.124; 4.196.265; 4.271.145; 4.361.549; 4.631.550; 4.364.932: 4.364.933; 4.364.934; 4.364.935; 4.364.936; 4.381.292; og 4.381.295). ;Et formål ved foreliggende oppfinnelse er å tilveiebringe nye og anvendelige monoklonale antistoffer mot sporozoitter fra slekten Eimeria. Et ytterligere formål er å isolere og identifisere spesifikke antigener fra E. tenella. Disse formålene trer frem i den følgende beskrivelse og er spesielt skissert i kravene. ;Det brukes et preparat av E. tenella sporozoitter til å immunisere mus for eventuelt å frembringe monoklonale antistoffer ved å følge fremgangsmåten til Kohler og Milstein slik den er beskrevet nedenunder. De monoklonale antistoffene brukes til å identifisere antigener hos parasitten. Antigenene som utløser monoklonale antistoffer som reagerer med sporozoitter, og gir nøytralisering av parasittvekst, anses som beskyttende antigener. ;Oppløselige antigener fås fra sporozoitter av E. tenella. Disse oppløselige antigenene separeres elektroforetisk etter molekylvekt, og de som reagerer spesifikt med monoklonale antistoffer ifølge foreliggende oppfinnelse, identifiseres. Ved å bruke passende standarder, karakteriseres så de reaktive antigenene på basis av molekylvekt. ;De følgende eksempler tjener ytterligere til å illustrere oppfinnelsen. ;EKSEMPEL 1 ;Konstruksjon av hvbridomalinier ;Sporozoitter av organismen Eimeria tenella fås vjed å frigjøre sporulerte occyster ved å bruke kjente fremgangsmåter ;(Doran et al., Proe. Helmintol. Soc. Wash. 34: 59-65 (1967)). Et således oppnådd sporozoitt-preparat av E. tenella brukes til å immunisere.^ 18 uker gamle BALB/c-mus av hunnkjønn ved hjelp av intraperitoneal injeksjon. Etter å ha fastslått at en immunisert mus produserer anti-sporozoitt-antistoffer ved å bruke en indirekte immunofluoresCensprøve-teknikk (IFA) som er kjent innenfor fagområdet, tas det ut miltceller fra mus og de smeltes sammen med myeloma-cellelinjen P3X63 Ag8.653 fra mus. Sammensmeltnings-fremgangsmåten utføres i nærvær av 30 til 35% polyetylenglykol (950* - 1050). Fremgangsmåten for frembringelse av hybridomas er tidligere blitt beskrevet (jfr. Kennett et al., Monoclonal Antibodies; — Plenum Press: 365-371, 1980). Hybridoma-sammensmeltingsprodukter dyrkes- i HAT-medium (Littlefield,
J.W. , Science, I4S>:: 7'09>-710 (1964)), som inneholder Iscove's modifiserte Dulbecco's' medium (IMDM); med 20% supplement av seruim fra kalvefoster. Dyrkningsmedier overvåkes med hensyn på anti-sporozoitt-antistoffproduksjon ved hjelp av indirekte immunofluorescensprøve (IFA) ved å bruke glutaraldehydfikserte sporozoitter av E. tenella som antigenkilde. Av alle de undersøkte kulturene ble 33 brønner påvist ved hjelp av IFA å være positive.
For å sikre at hybridomakulturene er monoklonale ble det brukt en begrensende fortynningsfremgangsmåte. Etter eksponering av E. tenella sporozoitter mot forskjellige monoklonale antistoffer ifølge oppfinnelsen iakttas tre viktige IFA-reaktivitetsmønstre på behandlende sporozoitter: (1) reaksjon på hele overflaten til sporozoittene; (2) overflatereaksjon som flekker på sporozoittene; (3) indre reaksjon rundt kjerne-membranene til sporozoittene. Disse reaksjonsmønstrene bekreftes vedi hg,elp av ferritin-merking og "transmission election"-mikroskopi (Speer et al., J. of Protozoology under trykning). Som, bekreftet ved hjelp av IFA, frembringer hybridomakulturer som her beskrevet identiske antistoffer etter kloning. Kloner dyrkes enten in vitro eller i BALB/c-mus som peritoneale tumorer, og bukhulevæsken inneholder antistoffer i en konsen-trasjon opptil omtrent 10 mg/ml.
EKSEMPEL 2
Fremstilling av antigener forbundet med sporozoitter av E. tenella
Nylig frisatte sporozoitter av E. tenella brukes til anti-genfremstillingen. Bestanddelene i den ytre membranen hos sporozoittene ekstraheres ved å bruke detergenter (dvs. 0,5% "Nonidet P40", 0,5 til 2% "CHAPS" eller 0,5% til 1% "Triton X-100") i 5 mM natriumfosfatbuffer med en pH på 7,8. Bufferen inneholder de følgende proteaseinhibitorer: aprotinin (2 trypsin-enheter/ml), antipain (25 yg/ml), leupeptin (25 yg/ml), fenyl-metylsulfonylfluorid (4 mM) (Yoshida et al., J. Exp. Med., 154: 1225 - 1236 (1981)). Materialet som er gjort oppløselig ved hjelp av detergenter, sentrifugeres ved 100 000 x g i 10 min for å fjerne faststoffer. Den klare supernatanten inneholder opplø-selige antigener forbundet med sporozoitter av E. tenella.
EKSEMPEL 3
Antigenkarakterisering
Oppløselige antigener av E. tenella sporozoitter adskilles etter molekylvekt ved å bruke SDS-polyakrylamidgel-elektroforese (PAGE) (Laemmli, U.K., Nature 227: 680 - 685 (1970)). Proteinene som er adskilt ved hjelp av SDS-PAGE overføres elektroforetisk til nitrocellulosemembraner ved å bruke "Western blotting tech-nique" (Towbin et al., Proe, Nati. Acad. Sei. (USA) 76: 4350 - 4354 (1979)). Nitrocellulosefilteret omsettes så enten med for-tynnet bukhulevæske eller brukt hybridomakulturvæske som inneholder antistoffer. Bundede monoklonale antistoffer påvises så ved å bruke en radioimmunpåvisningsfremgangsmåte omfattende 125^ anti-mus IgG antistoff (New England Nuclear). Det andre anti-stoffet, som ikke er bundet, fjernes ved vasking og nitrocellu-losef Utrene eksponeres så med Kodak røntgenfilm "XAR-5".
Eventuelt identifiseres bestemte komplekser mellom antigen og monoklonalt antistoff ved hjelp av en ELISA-teknikk under anvendelse av pepperrot-peroksydase-forbundet IgG-antistoff fra kanin mot mus-immunoglobulin (Burnette et al., Anal. Biochem.
112: 195-203 (1981)). Prøvesettet "Bio-Rad Immuno Blot" anvendes.
De tilsynelatende molekylvektene til de reaktive sporozoitt-antigenene bestemmes ved å sammenligne de elektroforetiske Rf-verdiene til antigenene med Rf-verdiene til forbindelser med kjent molekylvekt kjørt som standarder sammen med antigenene i det samme systemet. Molekylvektdata funnet eksperimentelt for forskjellige antigener er vist i tabell I.
Hybridomcellelinjene nr. slC4, nr. s3D3, nr. slE4, nr. s5B9, nr. s5E5, nr. slA og nr. s2G8, isolert som beskrevet ovenfor, er blitt deponert ved American Type Culture Collection (ATCC), Rockville, Maryland og er der blitt tilført den permanente samlingen. Nr. slC4 er blitt tildelt nummeret HB8333; nr. s3D3 har nummeret HB8331, nr. slE4 er blitt betegnet med nr. HB8332, nr. s5B9 er blitt betegnet nr. HB8402, nr. s5E5 er blitt betegnet nr. HB 8403, nr. slA er blitt betegnet nr. HB8404 og nr. s2G8 er blitt betegnet nr. HB8405. Tilgang til antistoffene er tilgjengelig under behandlingen av foreliggende søknad for de som Comissioner of Patents and Trademarks bestemmer er berettiget til det under 37 CF.R. 1.14 og 35 U.S.C. 122, og alle restriksjoner på almenhetens tilgjengelighet til HB8331, HB8332, HB8333, HB8402, HB8403, HB8404 og HB8405 vil bli ugjenkallelig fjernet etter meddelelse av et patent på foreliggende søknad.
Nr. slC4, HB8333, nr. s3D3, HB8331 og nr. slE4 , HB8332 ble deponert ved ATCC 11. august 1983. De nye monoklonale antistoffene nr. s5B9, HB8402, nr. s5E5, HB8403, nr. slA, HB8404 og nr. s2G8, HB8405 ble deponert ved ATCC 1. november 1983.
Claims (1)
- Monoklonale antistoffer for bruk in vitro fremstilt ved hjelp av hybridomas dannet ved sammensmelting av celler fra en mus-myelomalinje og miltceller fra en mus som tidligere er blitt immunisert med sporozoitter av Eimeria tenella, karakterisert ved at antistoffene (a) reagerer spesifikt med antigener fra Eimeria spp.sporozoitter,(b) reagerer spesifikt med antigener fra Eimeria tenella somhar en molekylvekt på omtrent 13 til 150 kd, og(c) de er fremstilt fra hybridomas dannet ved sammen-smelting av p3 x 63.Ag8.653 myelomaceller og miltceller fra BALB/c-mus som tidligere er blitt immunisert med E. tenellasporozoitter,hvilke hybridomas betegnes klon nr. s5E5 og er deponert som ATCC nr. HB8403; klon nr. slC4 og er deponert som ATCC nr.HB8333; klon nr. s2G8 og er deponert som ATCC nr. HB8405; klon nr. s5B9 og er deponert som ATCC nr. HB8402; klon nr. slA og er deponert som ATCC nr. HB8404; klon nr. s3D3 og er deponert som ATCC nr. HB8331; eller klon nr. slE4 og er deponert som ATCC nr. HB8332.
