Description
Foreliggende oppfinnelse vedrører en ånalogifremgangsmåte ved
fremstilling av eh terapeutisk aktiv 3- (2-iæteksy-fenoksy)-3-metyl-benzo-2,4-dioxacykloheksanon med formel
Forbindelsen viser bemerkelsesverdige farmakologiske egen-
skaper. F.eks. har den høyere anti-hostevirkning enn kodein,
en antiinflammatorisk virkning som er praktisk talt lik acetylsalisylsyrens og antipyretisk virkning som bare er litt lavere enn den acetylsalisylsyren viste. Disse fordelaktige biologiske egenskaper samtidig med fullstendig mangel på
gastrolesiv virkning og en nesten neglisjerbar toksisitet (både akutt, subakutt og kronisk) gjør at denne substansen er nesten perfekt tolererbar i gastriske mengder og følgelig spesielt egnet for oral og rektal administrering.
Fremgangsmåten ifølge oppfinnelsen ved fremstilling av forbindelsen I som
i det følgende kalles MR 693 for korthets skyld, består av å omsette acetylsalisylsyreklorid, guaiakol og pyridin i molforholdet 1:1:1.
Reaksjonen utføres i inert organiske løsningsmidler så som
f.eks. metylenklorid.
Det følgende ikke-begrensende eksempel er gitt for å
illustrere oppfinnelsen bedre.
EKSEMPEL
a) Acetylsalisylsyreklorid ble fremstilt ved å
koke syren og tionylklorid i 3 timer, avdampe overskuddet
av tionylklorid og destillere den erholdte rest under vakuum
(150°C/30 mm Hg)
Utbytte: 50-70% av teoretisk.
Det erholdte klorid ble holdt ved 0-4°C i 18 timer.
b) En løsning av 709 g (3,57 mol) acetylsalisylsyreklorid erholdt som under a) i 685 ml metylenklorid ble satt
til 442 g (3,57 mol) guaiakol. Den resulterende blanding ble utvendig avkjølt med et vannbad, deretter ble 282 g (3,57 mol) pyridin tilsatt dråpevis, mens reaksjonstemperaturen ble holdt på 20-25°C. Etter at tilsetningen var ferdig ble rør-ingen forlenget med ca. 1 ytterligere time, og blandingen fikk deretter stå i 16 timer.
Blandingen ble vasket flere ganger med vann, den organiske fasen tørket over vannfritt natriumsulfat og tilslutt for-dampet under redusert trykk. Den erholdte gule rest krystal-liserte langsomt. Den ble etter 4 8 timer opptatt i en liten mengde etanol, den resulterende blanding ble filtrert og de erholdte krystaller ble vasket. Etter krystallisering fra etanol fikk man 291 g av forbindelsen med formel I, smelte-punkt 70-74°C.
Elementæranalysen stemmer med den antatte struktur, hvilket også er bekreftet ved krystallografiske undersøkelser: MR 693 (empirisk formel <C>16H14°5<;> m-v- = 286,29). Krystalliseres i det ortorombiske system, romgruppe
Pbca.
Celleparametrene er: a = 9,968 (3), b = 23,808(4), c = 11,668(2)8; V = 2769(2)A°3> De ble bestemt gjennom en minste

kvadraters analyse på sin & verdiene for 50 reflekser. Målingene ble utført med et automatisk diffraktometer CAD4
(Nonius) , ved bruk av Mo K^ stråling = 0,71069 8),
med grafittmonokromator. Densiteten beregnet for 8 molekyler i cellen er 1,373 g/cm; den eksperimentelle verdi (ved fIotasjon i ^H^I^ løsning) er en 1,370 g/cm 2.
På en tilnærmet sfærisk krystall (diameter ca. 0,28 mm) ved bruk av samme diffraktometer og ved anvendelse av omega-scan-ning teknikk ved varierbar hastighet ble 2377 intensiteter målt. Den periodiske kontroll av tre standard intensiteter viste ingen forsemring av krystallet. Dataene er korrigert gjennom Lorentz- og polarisasjonsfaktorer; ingen absorpsjons-korreksjon har vært nødvendig Jji (Mo K©\ ) = 1,1 cm .
Strukturen (se vedlagte tegning) er bestemt ved direktemetod-er. Den endelige bestemmelse av 247 parametre (skalafaktor, sekundær ekstinksjon koeffisient, koordinater og termiske anisotropifaktorer for 16C og 50, koordinater og isotropiske B faktorer for 14 H) er utført gjennom påfølgende cykler
av fullstendig minste kvadraters matrise, basert på 2055 reflekser med positiv intensitet. Den minimaliserte mengde var

Farmakokynetikk og metabolisme
Undersøkelser utført både in vitro og in vivo.
In vitro eksperimentene har vist at forbindelsen MR693 hydroliseres enzymatisk til acetylsalisylsyre og guaiakol. Organene som viser den høyeste esteraseaktivitet viste seg
å være lungene og nyrene. Da MR 6 93 ikke har noen ioniserbar
- intraperiotoneal administrering
MR 693 viser utmerket aktivitet også gitt ad denne vei.
I en dose på 100 mg/kg observeres en 100% reduksjon i sammen-ligning med kontrollene av antall hosterier, mens kodein ved 12,5 mg/kg , igjen gitt ad intraperiotoneal vei, forårsaket en reduksjon på 93%.

Gastrolesiv aktivitet
MR 693 og acetylsalisylsyre som sammenligningsforbindelse ble gitt ad oral vei til rotter som hadde fastet i minst 18 timer.
Etter 6 timer fra begynnelsen av eksperimentet ble rottene avlivet og magene åpnet og undersøkt for å fastslå nærværet av lesjoner på de gastriske slimhinner.
De erholdte resultater uttrykt som gjennomsnitlig diameter (i mm) av lesjonene er angitt i den følgende tabell 2: De viser klart at forbindelsen som fremstilles ifølge foreliggende oppfinnelse i motsetning til acetylsalisylsyre tolereres fullstendig i gastriske mengder.

Antiinflammatorisk virkning
De antiinflammatoriske egenskapene til MR 693 ble under-søkt ved hjelp av karagenin indusert ødemforsøk hos rotter ved å gi forbindelsen som fremstilles ifølge oppfinnelsen ad oral vei og bruke acetylsalisylsyre som sammenligningssubstans i ekvi-molære doser.
De oppnådde resultater viser klart at MR 693 forårsaker en markert reduksjon av ødem og at denne reduksjon er praktisk talt av samme størrelse som den sammenligningssubstansen viser. Disse resultater er sammenfattet i den følgende tabell 3.

Antipyretisk aktivitet
Denne akivitet ble undersøkt ved hjelp av gjærindusert hyper-termiforsøk hos rotter, idet acetylsalisylsyre gitt i ekvimolare doser ble brukt som sammenligningssubstans.
De erholdte resultater som er sammenfattet i den følgende tabell 4 viser at MR 693 har en markert antipyretisk virkning, som bare er litt lavere enn den acetylsalisylsyren viser.

Toksisitet
Akutt toksisitet
Akutte tosisitetsforsøk ble utført i mus og rotter ved å gi forbindelsen som fremstilles ifølge oppfinnelsen ad oral og intraperitoneal vei. Som man ser fra resultatene som er angitt i den følg-ende tabell 5, kan MR 693 ansees å være en meget lite toksisk forbindelse.

Subakutt toksisitet
Subakutt toksisitetsstudier ble utført på rotter og hunder ved å gi MR 693 ad oral vei i 4 påfølgende uker.
Parametrene som er tatt i betraktning er de følgende:

- vektøkning
- hematologiske data
- hematokjemiske data
- glykosuri, proteinuri, albuminuri og hematuri
- autoptisk undersøkelse
- vekt av hovedorganer
- histologiske undersøkelser av hovedorganene; men ikke i noen tilfeller fant man endringer eller misdannelser som kunne tilskrives behandlingen med forbindelsen som fremstilles ifølge oppfinnelsen.
Kronisk toksisitet
Kroniste toksisitetsstudier ble utført på rotter som ad oral vei fikk MR 693 i 24 etterfølgende uker, og hunder som ad oral og rektal vei fikk forbindelsen som fremstilles ifølge oppfinnelsen i 24 uker.
Parametrene som er tatt i betraktning er som følger:

- biokjemisk undersøkelse av blodet
- glykosuri, proteinuri, hematuri, albuminuri
- autoptisk undersøkelse

Teratogenese
Teratogenesestudier ble utført på rotter og kaniner som
ad oral vei ble behandlet med MR 693 under hele svangerskaps-tiden. Undersøkelsen av:
- antall gravide hunner
- antall døde hunner
- opprinnelig kropsvekt/sluttvekt (av mødrene)
- totalt antall foetuses
- gjennomsnittstall av levende fuetuses
- totalt antall døde foetuses
- totalt antall re-absorpsjoner
- gjennomsnittsvekt av levende foetuses
- mulige misdannelser
viste at behandlingen med MR 6 93 ikke forårsaket noen forandring. Hverken somatiske eller skjelettmisdannelser ble observert, og heller ingen variasjon i antall av overlevne kull.
Farmasøytiske blandinger
Forbindelsen ifølge oppfinnelsen kan gi oralt eller rektalt, og kan anvendes i forskjellige farmakologiske blandinger så som f.eks: Tabletter og kapsler som inneholder 0,5 g av den aktive be-standdel
5, 0% og 3, 3% ( vekt) suspensjoner
suppositorier som inneholder 1,2 og 0,5 g av den aktive be-standdel .
Claims (1)
- Analogifremgangsmåte ved fremstilling av terapeutisk aktiv 3-(2-metoksy-fenoksy)-3-metyl-benzo-2,4-dioksa-cykloheksanon med formelkarakterisert ved at man omsetter acetylsalisylsyreklorid, guaiakol og pyridin i det molare forhold 1:1:1, i nærvær av et inert organisk løsningsmiddel.
