Groeihormoon vrijstellend peptide.
De uitvinding betreft een nieuw peptide en de zouten ervan, die een groeihormoon (GH) vrijstellend effect bezitten alsmede een hormoonpreparaat met deze stof daarin als effectief bestanddeel.
GH is een van de hormonen van de hypofyse, die de groei 5 bevordert, waardoor direct of indirect de totale proteïnesynthese bij lichaamscellen van dieren wordt bevorderd en de afgifte van vrije vetzu- -ren ter verhoging van de energiebeschikbaarheid wordt verhoogd. Hoewel GH wel is gebruikt voor een therapie bij kleine mensen, kan van GH veel « meer worden verwacht bijvoorbeeld een versnelde wondgenezing door een 10 activering van lichaamscellen.
Anders dan insuline is GH echter zeer soortspecifiek en een GH geëxtraheerd uit hypofysen van runderen en varkens kan niet worden gebruikt voor mensen. Bovendien is menselijk GH een enkelvoudig polypeptide met 191 aminozuren en een betrekkelijk hoog molecuulgewicht van 15 ca. 22.000 dat niet langs organische weg kan worden gesynthetiseerd.
Daarom is GH geëxtraheerd uit hypofysen van dezelfde soort dieren, maar dit levert natuurlijk zelfs al te weinig op voor een therapie van kleine mensen.
GH en het thyroïde stimulerend hormoon (TSH), het lutel-20 niserend hormoon (LH) en dergelijke staan onder controle van de hypothalamus. Gevonden werd nu dat afscheiding van elk hypofysehormoon specifiek wordt gestimuleerd door het overeenkomstige vrijstellende hormoon (RH) dat wordt afgescheiden door deze hypothalamus.
Bijvoorbeeld zijn thyroitroop vrijstellend hormoon (TRH) 25 dat de afgifte van THS stimuleert en lutelniserend hormoon vrijstellend hormoon (hieronder LH-RH) dat de afgifte van LH stimuleert, geïsoleerd en geïdentificeerd. Wat betreft groeihormoon stimulerend hormoon (GH-RH) dat de afgifte van GH bevordert, daarvan is gesuggereerd, dat dit een peptide was uit tien aminozuren en geïsoleerd uit varkenshypothalamus-30 weefsel door Shary in 1971, maar bij een later onderzoek is gebleken, dat dit geïsoleerde peptide iets anders was dan GH-RH. Sindsdien is er geen GH-RH in enige diersoort gevonden.
Anderzijds werd gevonden, dat GH in geringe mate wordt vrijgesteld volgens een niet specifieke weg bij toepassing van stoffen als 8300413 * ί 2 arginine, glucagon en TRH. Bovendien, hoewel gezegd wordt dat prostaglan-dine E, theofylline, cyclisch adenylzuur en dergelijke een GH vrijstellend effect bezitten, is ook dit effect kennelijk niet specifiek. Voorts, hoewel wordt aangenomen, dat bepaalde zenuwoverdrachts-peptiden een licht 5 GE afgevend effect bezitten, is ook dit effect niet specifiek.
Anderzijds is bevestigd, dat RH in de hypothalamus geen soortspecifiteit bezit bij studies van TRH of LH-RH.
Er werd nu gevonden, dat een nieuw peptide afkomstig van de hypothalamus kan worden geïsoleerd uit het extract van zalm-hypo-10 fysen,en het aldus verkregen peptide een specifiek GH- vrijstellend effect heeft op andere dieren.
De uitvinding betreft derhalve een peptide of een zout daarvan met de structuur formule .. .
Η-Asp-Thr-Met-Arg-Cys -Met-Val-Gly-Arg-15 Val-Tyr-Arg-Pro-Cys-Trp-Glx-Val-CH ^ waarin Glx Glu of Gin is, en dat de volgende fysische eigenschappen bezit: ' (1) ultraviolet absorptiespectrum: max 280 nm, 20 (2) de Ehrlich-reactie, Sakaguchi-reactie en Pauly's-reactie zijn positief^ (3) een basische reactie, - (4) oplosbaar in water, methanol en azijnzuur, zwak oplosbaar in ethyl-acetaat, butylacetaat, ethylether, hexaan, petroleumether, benzeen en chloroform, 25 (5) het uiterlijk bezit van een wit poeder.
Volgens de uitvinding kan tevens een hormoonpreparaat worden verkregen dat dit peptide of een zout daarvan als actief bestanddeel bevat.
Een nadere toelichting volgt aan de hand van de tekening. 30 Daarin geeft figuur 1 een kolomchromatogram weer verkregen met gelfiltra-tiekolomchromatografie volgens voorbeeld I; figuur 2 een ionen-uitwisselingschromatogram verkregen door de ionen-uitwisselingschromatografie volgens voorbeeld I; figuur 3 een elueringskromme verkregen bij een omgekeer-35 de fase hoge-druk-chromatografie (HPLC) volgens voorbeeld I; figuur 4 een ultraviolet absorptiespectrum van het pep- 8300413 » . - - 3 tide volgens de uitvinding; figuur 5 en figuur 6 een eluëringskromme verkregen met HPLC volgens de analysevoorbeelden 7 en 8; figuur 7 een eluëringskromme verkregen met behulp van de 5 HPLC volgens voorbeeld II? figuur 8 een grafiek die de verandering weergeeft met de tijd wat betreft de GH vrijstellende activiteit van GH-RH in vivo bereikt bij farmacologisch, proef voorbeeld II; figuur 9 een grafiek die de verandering weergeeft met 10 de tijd wat betreft de GH afgevende activiteit van [Gin] -GH-RH in vivo, verkregen bij de farmacologisch proef voorbeeld m.

Voorbeelden van zouten van het onderhavige peptide zijn bijvoorbeeld zuuradditiezouten met zoutzuur, zwavelzuur, azijnzuur, melkzuur, citroenzuur, oxaalzuur, fumaarzuur, malelnezuur en dergelijke.
^ Het onderhavige peptide kan worden verkregen door een extractie van zalm-hypofysen. Een zalm-hypothalamus kan niet alleen en volledig worden geïsoleerd, omdat een zalm-hypothalamus anders dan bij zoogdieren in de hypofyse binnen penetreert. De extractie moet daarom worden uitgevo'erd bij een geïsoleerd mengsel van hypofyse en hypothalamus.
35 In de onderhavige beschrijving wordt met "zalm-hypofyse" een hypofyse bedoeld met hypothalamus daarbij.

Het onderhavige peptide kan worden verkregen door een extractie van vers gelyofiliseerde zalmhypofysen met HCl-aceton, uitzouten, van de extracten, isoleren van het gewenste peptide via gelfiltratie-chromatografie en ionen-uitwisselingschromatografie, waarna wordt ge-5 zuiverd met een HPLC. In dit geval komt het verkregen peptide overeen met de structuurformule I, waarbij Glx is gelijk . Glu. Dit peptide kan gemakkelijk worden omgezet in een .. " zuuradditiezout door lyofilisatie of kristallisatie en behandeling met een anorganisch zuur zoals zoutzuur.
10 Anderzijds is thans gevonden, dat dit peptide bok kan worden gesynthetiseerd met behulp van een peptidesynthesetechniek.
Bij de uitvinding kan het peptide worden gesynthetiseerd op basis van de boven weergegeven GH-RH-structuur, ontdekt bij 16 16 zalmhypofyse. Daarbij kunnen zowel Gin -GH-RH als Glu -GH-RH worden 15 gesynthetiseerd. Voor de toe te passen technieken bij een dergelijke synthese wordt verwezen naar Haruaki Yajima en Syunpei Sakakibara . (Tanpakushitsu no Kagaku (Protein Chemistry), SEIKAGAKU JIKKEN KOZA (Experimental Biochemistry) (I), _4, p. 208 - 495 (1977) van de Japanse Biochemical Soc. en gepubliceerd door Kabushiki kaisha Tokyo Kagaku Dohj1η)Λ 20 en Nobuo Izumiya et al (PEPTIDE GOHSEI (SYNTHESIS) (1975) gepubliceerd door Maruzen Kabushiki Kaisha).
Het onderhavige GH-RH heeft een specifieke GH vrijstellende activiteit maar geen soortspecifiteit. Daardoor kan een hormoon-preparaat met dit peptide als actief bestanddeel worden gebruikt voor 25 een therapie van menselijke dwergvormen en wondheling maar.ook voor het stimuleren van de groei van huisdieren.
Het onderhavige peptide heeft evenals andere fysiologisch actieve peptiden specifieke receptoren in het lichaam en veroorzaakt specifieke reacties door deze receptoren specifiek te beïnvloe-30 den. GH-RH kan GH vrijstellen in een hypofyse met specifieke receptoren maar heeft geen effect op de andere weefsels die deze specifieke receptoren missen. Bovendien wordt het onderhavige peptide dat uitsluitend uit L-aminozuren bestaat afgebroken door verschillende proteasen, zodat zijn effect niet blijvend is.
35 Anderzijds bezit een peptide met een dergelijk mole- cuulgewicht (ca. 2000) nog geen antigene eigenschappen, waardoor noch 8300413 . * w 5 een antilichaam-produktie op gang komt, noch een anafylactische schok wordt veroorzaakt,ook niet H.j herhaalde toediening. Het peptide is bovendien niet giftig.
Het onderhavige peptide heeft reeds een duidelijk effect 5 bij extreem lage doseringen van ca. ng/kg hetgeen minder is dan die van GH zelf op basis van de .hoeveelheid actief bestanddeel. Het effect wordt met de dosering evenredig verhoogd en deze dosering kan tot mg/kg bedragen. Een voorkeurs-doseringstraject ligt tussen 1 ng/kg en 10 mg/kg.
Het onderhavige peptide wordt bij voorkeur toegediend 10 via een injectie bijvoorbeeld een intraveneuze injectie, een intramuscu-laire injectie, een subcutane injectie of dergelijke, die gewoonlijk worden toegepast voor peptide-middelen. Bij orale toediening van het onderhavige peptide, wordt dit in de spijsverteringsorganen afgebroken en omgezet in een inactieve vorm. In dit geval kan het evenwel in sommige 15 preparaatvormen, zoals microcapsules in liposomen worden verwerkt, die niet in organen worden afgebroken. Voorts kan het onderhavige peptide ook worden toegediend via de mucosa van andere dan spijsverteringsorganen, zoals het rectum, sublinguaal, de neus en dergelijke. In dat geval kan het in een reeks preparaatvormen worden toegepast, zoals zetpillen, 20 sublinguale tabletten, en rhinologische spray's. Voorbeelden van excipiSn-tia zijn de gebruikelijke, waarbij het peptide volgens de uitvinding niet wordt afgebroken.
De uitvinding wordt hieronder meer specifiek beschreven aan de hand van de voorbeelden. De uitvinding is uiteraard niet tot 25 deze voorbeelden beperkt en diverse wijzigingen en modificaties zijn mogelijk zonder van de uitvinding af te wijken.
Voorbeeld 1
Ca. 100 g verse hypofyse verkregen uit vrouwelijke witte zalmen (Oncorhynchus keta) werd geëxtraheerd met 400 cm3 van een 30 35%'s zoutzuur-acetonmengsel in de verhouding 1 : 28, en vervolgens met 280 cm3 80%'s aceton. Beide extracten werden samengevoegd en toegevoegd aan 10 1 aceton onder roeren waarbij 3,0 g van een zuur-aceton-precipitaat werd verkregen. Dit precipitaat werd toegevoegd aan 90 cm3 water, gebracht op pH 3 en indien nodig, opnieuw geregeld met 0,1N na-35 tronloog of 0,1N zoutzuur. Vervolgens werd 10 cm3 van een verzadigde NaCl-oplossing toegevoegd om uit te zouten. Na centrifugeren van 8300413 t ï 6 het verkregen precipitaat werd de verkregen bovenstaande vloeistof ge-chromatografeerd over een kolom van 5/5 x 68 cm Sephadex G 25 (M). Geëlu-eerd werd met 0,1N azijnzuur (fracties van 10 cm3 per fractie). Het verkregen chromatogram is weergegeven in figuur 1. De fractie (III) weer-5 gegeven in figuur 1 met een volume van 320 cm3 werd ontzout met een UM-2 membraanfilter van Amicon Co.; daarna werd gelyofiliseerd tot 139 mg van een wit poeder. Dit poeder werd opnieuw gechromatografeerd over een ionenui twisselende kolom (1,6 x 63 cm) van CM-cellulose. Ditmaal werd geëlu-eerd met een gradiënt^verkregen door toepassing van 500 cm3 van een 0,01 10 M ammoniumacetaatbuffer, pH 4,6, onder druppelsgewijs toevoegen van 0,1 M ammoniumacetaatbuffer met pH 7,0. Het verkregen chromatogram is weergegeven in figuur 2. De fractie (8) weergegeven in figuur 2 met een volume van 110 cm3 werd gelyofiliseerd tot 1,5 mg onzuiver GH-RH.
Dit onzuivere GH-RH werd verder gechromatografeerd over 15 een kolom (0,4 x 2,5 cm) HPLC (fine packcyl C 18 van Nippon Bunko Kogyo Kabushiki Kaisha), geëquilibreerd met 0,01M ammoniumacetaat met pH 5,0.
Geëlueerd werd hierbij met een lineaire gradiënt van isopropanol (concentraties 15% tot 45%) in een inrichting met constant niveau en bij een stroomsnelheid van 1 cm3/minuut waarbij de kolom op 20 kamertemperatuur werd gehouden. Het kolomeffluënt werd opgevangen in fracties van 1 cm3. De verkregen eluëringskromme is weergegeven in figuur 3. De fractie (A) van figuur 3 met een volume van 13,3 cm3 werd gelyofiliseerd tot 0,51 mg onderhavig peptide met de hoge zuiverheid van 99% of meer.
25 De fysische eigenschappen van dit basische peptide lui den als volgt: uiterlijk: wit poeder; molecuulgewicht: 500 - 3000 (bepaald door gelfiltratie); kleurreacties: Ehrlich-reactie (+)·, Sakaguchi-reactie (+), Pauly's-30 reactie (+); oplosbaarheid: oplosbaar in water, methanol en azijnzuur; vrij slecht oplosbaar in ethylacetaat, butylacetaat, ethylether, petroleumether, hexaan, benzeen en chloroform; ultraviolet absorptiespectrum (in 0,1 N azijnzuur met een concentra-35 tie van 0,166 mM): zie figuur 4.
In 5 cm3 gedestilleerd water werd 500 ug van dit pepti- 83 0 0 4 1 3 # 7 de opgelost, zuur gemaakt met enige druppels 0,1 N zoutzuur en gelyo-filiseerd tot 500 ^ug hydrochloride van het onderhavige peptide.
De aminozuursamenstelling en de aminozuurvolgorde van het aldus verkregen peptide werden bepaald volgens de volgende analyti-, 5 sche voorbeelden 1-8.

De aminozuuranalyse werd uitgevoerd met een LKB 4400 aminozuur-autoanalyzer (LKB Biochrom Co., Ltd.) na een hydrolyse van 10 het peptide volgens voorbeeld I in 6N HC1 gedurende 18 uren bij 110eC.

De gevonden waarde voor Cys werd bepaald door een oxy-25 datie met permierezuur. analytisch voorbeeld 2 [N-terminaal aminozuur]
Het N-terminale aminozuur van het peptide volgens voorbeeld I werd bepaald volgens de Dansyl-methode (Biochemical J., 89, 30 P379 (1963)) en dit bleek Asx te zijn.

Het C-terminale aminozuur van het peptide volgens voorbeeld I werd bepaald volgens een hydrazinolyse-methode van Akabori et al, 35 Bulletin Chemical Soc. Japan 25, 214 (1952) en bleek Val te zijn.
üit de analytische voorbeelden 1-3 blijkt, dat het 8300413 δ onderhavige peptide een heptadecapeptide is bestaande uit elf verschillende aminozuren met een berekend molecuulgewicht van 2211 in de vorm van een trihydrochloride en met N-terminaal Asx en C-terminaal Val.
• Analytisch voorbeeld 4 5 [Aminozuurvolgorde (1)]
Onder toepassing van 66 ug (ca. 30 nmol) van het ge-J carboxymethyleerde peptide volgens voorbeeld I werd de aminozuurvolg-orde bepaald van het N-terminale residu volgens de Edman-Dansyl-methode (Biochimica. Biophysica. Acta., 21_, p. 58 (1956). Het resultaat luidde

Analytisch voorbeeld 5 [Aminozuurvolgorde (2)] 15 . ' Onder toepassing van 88 ^ug (ca. 40 nmol) van het ge- carboxymethyleerde peptide volgens voorbeeld {I, werd de aminozuurvolgorde bepaald van het N-terminale residu volgens’ de fluorescelne-iso-cyanaatmethode (Muramoto et al, Agricultural and Biological Chemistry, 44, 1559 - 1563 (1978)). Het resultaat luidde als volgt: 20 H-Asp-Thr^Met-Arg-Cys -Met-Val-Gly-Arg-

Deze fluoresceïne-isocyanaatmethode van Muramoto is een bijzonder betrouwbare methode voor het bepalen van een aminozuurvolgorde met een kleine hoeveelheid monster.
25 Analytisch voorbeeld 6 [Carboxypeptidase Y afbraak]
Aan 0,2M N-ethylmorfolineacetaat-buffer met pH 8,0 met daarin carboxypeptidase Y (Oriental Yeast Company Limited) werd 10 nmol van het peptide volgens voorbeeld I toegevoegd, waarna het mengsel bij 30 37°C werd geïncubeerd. Carboxypeptidase Y : substraat = 1 : 60 (gewichts verhouding) . De aminozuur samenstelling van de behandelde vloeistof werd na 1 en na 5 uren bepaald. Deze luiden als volgt:

Uit. deze resultaten gecombineerd met analytisch voor beeld 3 bleek de C-terminale volgorde -Glu-Val-OH te zijn.

Uit analytische voorbeelden 1-6 kan de aminozuur- volgorde van het gehele peptide worden afgeleid en deze luidt als volgt: H-Asp-Thr-Met-Arg-Cys -Met-Val-Gly-Arg-Val-Tyr-Arg-Pro-Cys-Trp-Glu-Val-QH 5 Voorts werd het onderhavige peptide afgebroken met chymotrypsine of trypsine en daarna de aminozuursamenstelling, het N-tenninale aminozuur en de aminozuurvolgorde van het afbraakprodukt onderzocht volgens de volgende analytische voorbeelden 7 en 8 om de volledige aminozuurvolgorde te bevestigen.
10 analytisch voorbeeld 7 [ Chymo trypsine-afbraak ] aan Q,2M ammoniumacetaatbuffer met pH 8,0 met daarin chymotrypsine werd 110 ^ug van het peptide volgens voorbeeld I toegevoegd en het reactiemengsel 1 uur bij 37 °C gelncubeerd. Chymotrypsine : sub-15 straat is 1 : 250 gew.dln. Het verkregen afbraakprodukt werd vervolgens via een HPLC geanalyseerd die op dezelfde wijze werd uitgevoerd als bij voorbeeld met dit verschil dat met 5 - 50% isopropanol werd geëlueerd. De resulterende eluëringskromme is weergegeven in figuur 5. De fracties (C-2), (c-4) en (C-13) weergegeven in figuur 5 werden geanalyseerd en wel 20 op dezelfde wijze, als in de analytische voorbeelden 1 en 2, waarna de aminozuursamenstelling en het N-terminale aminozuur daarvan werden bepaald. De resultaten luidden als volgt:

De fracties (C-2), (C-4) en (C-13) werden -vervolgens geanalyseerd ' - op dezelfde wijze als in analytisch voorbeeld 4 ter bepaling van de aminozuurvolgorde. In dit geval werden de fracties 15 C-2 en C-4 als zodanig geanalyseerd en de fractie C-13 na een carboxy- methylering. De identificatie van Trp werd uitgevoerd volgens de Edman-Dansyl-methode, waarbij Trp werd bepaald als DNS-Trp via een hydrolyse met thioglycolzuur. Cys werd bepaald als DNS-Cm-Cys en wel op dezelfde wijze als boven weergegeven. Aan 0,2M ammoniumacetaatbufferoplossing met pH 8,0 30 met daarin trypsine werd 88 ^,ug (ca. 30 nmol) van het peptide volgens voorbeeld I toegevoegd en het reactiemengsel vervolgens 1 uur bij 37°C geincubeerd. De trypsine : substraatverhouding bedroeg 1 : 50 gew.dln. Het verkregen afbraakmateriaal werd vervolgens met HPLC geanalyseerd, die op dezelfde wijze werd uitgevoerd als bij voorbeeld i met dit ver-35 schil dat werd geëlueerd met 5 tot 50% isopropanol. Aan 300 cm3 van een dioxanoplossing met daarin 16,4 g Boc-Gln-ONp en H-Val-Obzl, verkregen uit 17,5 g p-tolueensulfonaat en • 6,45 cm3 triëthylamine (hieronder aangeduid als Et^N) werd 6,45 cm3 Et^N toegevoegd. Het verkregen, mengsel werd 24 uren bij kamertemperatuur be-5 waard en daarna geconcentreerd. Het residu werd opgelost in ethylacetaat (hieronder AcOEt), gewassen met 5% citroenzuur, 5% Na^CO^ en NaCl-water, en daarna gedroogd met Na^SO^. Na afdestilleren van het ethylacetaat werd het residu begiftigd met petroleumether ter kristallisatie. Her-kristallisatie uit ethylacetaat/ether leverde 16,45 g (79% van de theo-10 retische hoeveelheid).

Aan 7,94 cm3 anisool werd 8,00 g Boc-Gln-Val-OBzl toegevoegd. Dit reactiemengsel werd behandeld met 30 cm3 trifluorazijnzuur (TFA) en wel gedurende 60 minuten, terwijl met ijs werd gekoeld; hierna werd het TFA bij kamertemperatuur af gedestilleerd. Het residu werd toe-20 gevoegd aan n-hexaan, door decanteren gewassen en daarna gedroogd met KOH. Hierna werd het verkregen olievormige materiaal opgelost in 80 cm3 dimethylformamide (DMF) en geneutraliseerd met 2,57 cm3 Et^N, waarna 8,60 g Boc-Trp-ONp werd toegevoegd. Dit reactiemengsel werd 24 uur bij kamertemperatuur bewaard, waarbij de reactie optrad. Na afdestilleren 25 van bet oplosmiddel beneden 30°C werd het residu opgelost in ethylacetaat, gewassen met 5%'s Na^O^, 5%'s citroenzuur en NaCl-water en daar-: na met Na2SO^ gedroogd. Na afdestilleren van het ethylacetaat werd het residu aan ether toegevoegd ter kristallisatie. Aan 9,54 g Boc-Trp-Gln-Val-OBzl werd 16,56 cm3 2%'s 8300413 * · 15 ethaandithiol-anisool toegevoegd. Dit reactiemengsel werd onder stikstof-gas Z''*·, behandeld met 50 'cm3 WA en wel gedurende 120 minuten, terwijl met ijs werd gekoeld. Na af destilleren van het TFA onder 30 °C werd het residu aan ether toegevoegd, waarbij een poeder ontstond dat boven KOE 5 werd gedroogd- Het verkregen witte poeder werd opgelost in 50 cm3 DMF en daarna 2,36 cm3 ET^N alsmede 7,09 g Boc-Cys(MBzl)-ONp toegevoegd.
Dit reactiemengsel werd 18 uren bij kamertemperatuur weggezet om de reactie te doen verlopen en daarna met azijnzuur geneutraliseerd. Na afdestilleren van de oplosmiddelen werd het residu opgelost in ethylacetaat, met 10 5% I3a.^30^t 5% citroenzuur en NaCl-water gewassen en met Na2S0^ gedroogd.
Na afdestilleren van het ethylacetaat werd het verkregen gelachtige ma- « teriaal met ether aangewreven. Een precipitatie met methanol/ether werd .

Dit mengsel . liet men onder de boven weergegeven voorwaarden 24 uur staan om de reactie te doen verlopen. Na affiltreren van neergeslagen dicycloureum werd het filtraat gewassen met 5% citroenzuur, 5% soda-oplossing en NaCl-water, waarna werd gedroogd met natriumsulfaat. Na afdestilleren van het ethylacetaat 30 werd n-hexaan ter kristallisatie aan het residu toegevoegd. Herkristal-liseren uit ethylacetaat/petroleumether leverde 7,80 g (73% van de theoretische hoeveelheid) aan gewenste verbinding.

Nadat de verkregen olie was opgelost in 30 cm3 DMF, 20 werd hieraan 2,1 cm3 Et^N toegevoegd, terwijl met ijs werd gekoeld. Aan het verkregen reactiemengsel in 35 cm3 tetrahydrofuran werd Z(0Me)-Arg-(Mts)—OH^verkregen uit 10,23 g van het zout van cyclohexylamine en 17 cm3 IN zoutzuur, alsmede het gemengde, zuur anhydride^ verkregen uit 2,31 cm3 : Et^N en 2,36 cm3 isobutylchloorformiaat onder watervrije voorwaarden, 25 toegevoegd. Dit reactiemengsel werd 2 uren bij -15 °C en daarna 4 uren bij 0°C bewaard. Na afdestilleren van het oplosmiddel werd het residu opgelost in ethylacetaat, met 5% citroenzuur, 5% soda-oplossing en NaCl-water gewassen en met natriumsulfaat gedroogd. Na afdestilleren van het ethylacetaat werd isopropylether aan het residu ter kristallisatie toe-30 gevoegd. Vervolgens werd het verkregen kristallijne produkt opgelost in een kleine hoeveelheid chloroform en over een kolom (8x5 cm) silicagel gechromatografeerd, waarbij werd geëlueerd met chloroform : methanol = 100 : 1, waarna een onzuivere verbinding met een van 0,74 werd verkregen. Na af destilleren van het oplosmiddel werd de verbinding met pe-35 troleumether gewassen waarna 9,80 g (81% van de theoretische hoeveelheid) aan gewenste verbinding werd verkregen.

Het hierbij neergeslagen gelvormige materiaal werd met ethylether aangewreven. Na affiltreren, herkristalliseren uit warme methanol/ethylether en etha- , nol werd 4,1 g (83% van de theoretische hoeveelheid) aan gewenste ver- i 10 binding verkregen.

(B-v) Synthese van Z (OMe) -Arg (Mts) -Val-Tyr-Arg (Mts) -Pro-OH (9-13 stuk .)
In 80 cm3 methanol werd 4,7 g Z (OMe) -Arg (Mts) -Pro-OBzl opgelost en hieraan enige druppels azijnzuur toegevoegd. Dit reactie-15 mengsel werd gereduceerd met een katalytisch effectieve hoeveelheid 10%'s palladium-op-kool. Het oplosmiddel werd na affiltreren van de katalysator afgedestilleerd. Indien de verwijdering van het Z(OMe) niet volledig was, werd nog 2,9 cm3 anisool aan het residu toegevoegd en het reactiemengsel 30 minuten onder koelen met ijs met 12 cm3 TFA behandeld- Na af-20 destilleren van het TFA bij kamertemperatuur werd het residu met n-hexaan onder decanteren gewassen en gedroogd met Κ0Ξ. In 20 cm3 DMF werd dit residu vervolgens opgelost, waarna 0,93 cm* Et^N werd toegevoegd onder koelen met ijs. Het verkregen mengsel werd bij -10°C gemengd met een DMF-oplossing van Z(OMe)-Arg(Mts)-Val-Tyr-N,, bereid uit 5,26 g van het 25 hydrazide-derivaat daarvan, 0,87 cm3 isoamylnitraat en 0,92 cm3 Et^N, en hieraan 0,93 cm3 Et^N toegevoegd. Het verkregen reactiemengsel werd 24 uur bij -10°C bewaard om de reactie te doen verlopen en daarna met 2 tot 3 druppels azijnzuur geneutraliseerd. Na afdestilleren van het oplosmiddel werd het residu opgelost in 5%'s NaHCO^ / met ethylether ge-30 wassen, zuur gemaakt met citroenzuur en geëxtraheerd met ethylacetaat.
De verkregen ethylacetaatfase werd gewassen met 5%'s citroenzuur, 5%'s soda-oplossing en NaCl-water en daarna met Na^SO^ gedroogd. Na afdestilleren van het ethylacetaat werd petroleumether aan het residu ter kristallisatie toegevoegd. Herkristalliseren uit methanol/ether leverde 35 5,40 g (67% van de theoretische hoeveelheid) van de gewenste verbinding.

De verkregen oplossing werd aan n-hexaan toegevoegd en door decanteren gewassen. Het verkregen olievormige materiaal werd met KOH gedroogd en opgelost in 30 cm3 DMF. Aan de verkregen oplossing werd 1,4 cm3 Et^N toegevoegd en wel onder koelen met ijs, gevolgd 10 door het . gemengde anhydride van z(OMe)-Arg(Mts)-OH, verkregen uit 6,20 g van het zout van cyclohexylamine daarvan en 10 cm3 IN zoutzuur, 1,54 cm3 Et^N en 1,58 cm3 isobutylchloorformiaat in 50 cm3 THF onder watervrije voorwaarden. Het verkregen reactiemengsel werd 3 uren bij -10°C bewaard om de reactie te doen verlopen. Na afdestilleren van het 15 oplosmiddel werd het residu opgelost in ethylacetaat, met 5%'s citroenzuur, 5%'s sodaoplossing en NaCl-water gewassen en daarna met natrium-sulfaat gedroogd. Na afdestilleren van het oplosmiddel, kristalliseren uit ether en opnieuw neerslaan met methanol/ether (2 maal herhaald) werd 6,2 g (70% van de theoretische hoeveelheid) aan gewenste verbinding ver-20 kregen.

Het verkregen mengsel werd een nacht bij kamertemperatuur bewaard, waar-. na een gelachtig materiaal was ontstaan. Het verkregen materiaal werd met ether verwreven en deze ether afgefiltreerd. Opnieuw neerslaan uit DMF-ethanol leverde 3,0 g van de gewenste verbinding (68%).

(C-vii) Synthese synthese van Z(OMe)-Arg(Mts)-Cys(MBzl)-Met-Val-Gly-OH 35 (4-8 .stuk ), M.G.: 1,031,254
In 50 cm3 methanol werd 1,3 g Boc-Val-Gly-OBzl opge- 8300413 21 lost en hieraan diverse druppels azijnzuur toegevoegd. Het verkregen mengsel werd gereduceerd met een katalytisch effectieve hoeveelheid 10%'s palladium-op-koolstof en wel gedurende 3 uren. Na affiltreren werd het oplosmiddel af gedestilleerd. Het residu werd aangewreven met n-5 hexaan. Aan het verkregen witte poeder met een gewicht van ca. ir06 g werd 2,09 cm3 anisool toegevoegd en het geheel behandeld met 10 cm3 TFA gedurende 45 minuten onder koelen met ijs. Na afdestilleren van het TFA bij kamertemperatuur werd het residu boven KQH gedroogd, daarna opgelost in een mengsel van 2 cm3 d imethylsulfoxyde (DMSO) en 10 cm? DMF en 10 hieraan 1,1 cm3 Et^N onder koelen met ijs toegevoegd. Het verkregen mengsel werd gemengd met een DMF-oplossing van Z(OMe) -Arg(Mts)-Val-Tyr-N^, verkregen uit 3,8 g 2(OMe)-Arg(Mts)-Val-Tyr-^H^ en 0,62 cm3 isoamyl-nitraat alsmede 1,32 cm3 Et^N, en hieraan 0,6 cm? Et^N toegevoegd, waarna men de reactie 36 uren bij -10°C liep verlopen. Na neutralisatie van 15 het mengsel met enige druppels azijnzuur en af destilleren van het oplosmiddel, werd citroenzuur aan het residu toegevoegd, waarna men het enige tijd liet staan. Het verkregen neerslag werd aangewreven met 5%'s citroenzuur en water en daarna gedroogd. Neerslaan uit DMF/ethylacetaat werd enige malen uitgevoerd en daarbij 2,8 g (69%) van de gewenste ver-20 binding verkregen.

Aan 480 mg Z(OMe)-Arg(Mts)-Cys-(MBzl)-Met-Val-Gly-OH 35 werd 0,6 cm3 anisool toegevoegd. Het verkregen mengsel werd 45 minuten met 3 cm3 TEA onder koelen met ijs behandeld, waarna n-hexaan werd toe- 8300 4 1 3 22 gevoegd; gewassen werd door decanteren en daarna werd ether toegevoegd, waarbij een wit poeder werd verkregen. Dit poeder werd boven ΚΟΞ gedroogd, vervolgens opgelost in 5 cm3 DMF en daarna onder koelen 3 met ijs hieraan 0,055 cm .Et^N teegèvoegdjEet verkregen mengsel werd ge-5 mengd met 15 cm3 van een DMF-oplossing van Z (OMe)-Thr-Met-N^, verkregen uit 254 mg Z(OMe)-Thr-Met-^H^, 0,094 cm3 isoamylnitraat en 0,18 cm3 Et^N,waarna 0,055 cm3 Et^N werd toegevoegd, en men het reactiemengsel 24 uren bij -10°C liet staan. Na neutraliseren met enkele druppels azijnzuur en afdestilleren van het oplosmiddel werd 5% citroenzuur aan 10 het residu toegevoegd. Het verkregen neerslag werd fijngewreven en gewassen met 5% citroenzuur en water en daarna gedroogd. Neerslaan uit DMF-ethylacetaat, vervolgens uit DMF-methanol leverde 430 mg (90% van de theoretische hoeveelheid) aan de gewenste verbinding met =0,41.

Aan 348 mg Z(OMe)-Thr-Met-Arg(Mts)-Cys(Mbzl)-Met-Val-20 Gly-OH werd 0,3 cm3 anisool toegevoegd, waarna onder koelen met ijs 45 minuten met 1,5 cm3 TFA werd behandeld. Het reactiemengsel werd begiftigd met n-hexaan, daarmee door decanteren gewassen en vervolgens met ether aangewreven, waarbij een wit poeder ontstond. Dit poeder werd boven KOH gedroogd, opgelost in 3 cm3 DMSO en 3 cm3 DMF en daarna begiftigd met · 25 0,077 cm3 Et^N alsmede Boc-Asp (OBzl) onder koelen met ijs. Het reac tiemengsel werd 24 uur bij kamertemperatuur bewaard, waarna geneutraliseerd werd met enkele druppels azijnzuur. Na afdestilleren van het oplosmiddel werd het residu begiftigd met 5% citroenzuur, waarbij een neerslag ontstond. Dit werd met 5%*s citroenzuur en water gewassen en daarna 30 gedroogd. Het verkregen materiaal werd met ethanol begiftigd en 24 uur geroerd, waarbij, het merendeel Van het succinimide-derivaat overging in de ethanol, en 300 mg (78% van de theoretische hoeveelheid) van de gewenste verbinding werd verkregen.

Aan 0,85 g Boc-Cys(MBzl)-Trp-Gln-Val-OBzl werd 1,08 5 cm3 2%*s ethaandithiol/anisool toegevoegd en het geheel 60 minuten in een stroom stikstofgas bij -5°C behandeld met 5,00 cm3 TFA. Het reactie-mengsel werd begiftigd met n-hexaan en door decanteren gewassen. Het verkregen residu werd met ether begiftigd waarbij een wit poeder ontstond, dat met 5%'s soda-oplossing werd aangewreven waardoor het vrije 10 amine ontstond, dat met H20 en ether werd gewassen. Het verkregen materiaal werd boven KOH gedroogd en daarna opgelost in 10 cm3 DMF. Aan deze oplossing werd 1,22 g Z(QMe)-Arg(Mts)-Val-Tyr-Arg(Mts)-Pro-OH en 0,153 g N-hydroxy-benzotriazool (hierna HQBt) toegevoegd en daarna begiftigd met __ 0,25 g DCC terwijl werd gekoeld met een vriezend mengsel, waarna men de 15 reactie 24 uren bij kamertemperatuur liet verlopen. Na affiltreren van het dicycloureum werd het oplosmiddel af gedestilleerd, waarbij de temperatuur beneden 30°C werd gehouden. Nadat het residu aan ethylacetaat en 5%*s NaHCO^ was toegevoegd, werd het verkregen neerslag af gefiltreerd, onder roeren gewassen in 5%'s citroenzuur en en daarna gedroogd.
20 Opnieuw neerslaan uit DMF-ethylacetaat en DMF-ethylacetaat plus ethanol leverde 1,1 g van de gewenste verbinding (56% van de theoretische opbrengst) .

Trp werd geanalyseerd met 4M methaansulfonaat. Cys werd geanalyseerd na een oxydatie met permierezuur, (D-ii) Synthese van Boc-Asp (OBzl)-Thr-Met-Arg (Mts)-Cys (MBzl)-Met-Val-Gly-Arg(Mts)-Val-Tyr-Arg(Mts)-Pro-Cys(MBzl)-Trp-Gln-Val-OBzl 5 (1-17 .peptide, 16 beschermd [Gin ]-GH-RH), M..G.: 3*167^846
Aan 230 mg van de volgens (D-i) verkregen verbinding werd 0,254 cm3 2%'s ethaandithiol-anisool toegevoegd en het geheel met 1,00 cm3 TFA in een stroom stikstofgas bij 5°C'behandeld. Het verkregen 10 residu werd begiftigd met n-hexaan, door decanteren gewassen en met ether behandeld, waarbij een wit poeder werd verkregen.
Dit poeder werd verder gewassen met 5% 's soda, water en ether, met KOH gedroogd en daarna opgelost in 5 cm3 DMF. Aan de verkregen oplossing werd 166 mg Boc-Asp (OBzl)-Thr-Met-Arg (Mts) -Cys (MBzl) -Met-'15 Val-Gly-OH en 21,7 mg HOBt toegevoegd. Na roeren van het reactiemengsel werd 26,8 mg DCC toegevoegd, terwijl met een vriezend mengsel werd gekoeld; het reactiemengsel liet men 48 uur uitreageren. Hierna werd het reactiemengsel begiftigd met 11 mg HOBt en 14 mg DCC en nogmaals het reactiemengsel 48 uren omgezet. Na affiltreren van dicycloureum werd het 20 oplosmiddel afgedestilleerd bij een temperatuur beneden 30°C. Het residu werd begiftigd met water, waarbij een neerslag ontstond, aangewreven en gewassen met 5% citroenzuur en water en daarna gedroogd. Na neerslaan uit DMF/methanol (2 maal) werd 275 mg onzuivere verbinding verkregen.
Dit materiaal werd opgelost in een kleine hoeveelheid DMF en daarna ge-25 chromatografeerd over een kolom van 3 x 75 cm (volume 530 cm3) Sephadex : LH-20. Geëlueerd werd met DMF en de hoofdfractie werd opgevangen. Na afdestilleren van DMF beneden 30 °C werd het residu aangewreven met en daarbij 220 mg (59% van de theoretische hoeveelheid) aan gewenste verbinding in de vorm van een wit poeder verkregen.

Trp - 1 gem. teruggewonnen: 78% (E) Verwijdering schermgroepen en zuivering 10 aan 120-mg Boc-(l-17 aminozuren)-OB werd 0,33 cm^' m-cresol en 0,11.cm^.ethaandiol toegevoegd, waarna nog 3,02 cm van een TFA oplossing van 1M trifluormethaansulfonaat-thioanisool werd toegevoegd onder roeren met ijs. Het reactiemengsel werd 3 uren bij 0°C weggezet-ter verwijdering van alle schermgroepen. Vervolgens werd het oplosmiddel met 15 een vacuumpomp af gedestilleerd, het residu aan ether toegevoegd en daarbij een poeder verkregen. Dit poeder werd afgefiltreerd, het residu onmiddellijk opgelost in een kleine hoeveelheid water die ontgast was en vervangen door stikstofgas, waarna het onoplosbare materiaal werd afgefiltreerd. Het verkregen filtraat werd verdund met 400 cm? 0,01M amrao-20 niumacetaat met pH 7,0 tot een concentratie van 0,1 ^.umol/1 en 3 dagen in een donkere kamer bij kamertemperatuur bewaard om het aan de lucht te laten oxyderen. : ..... Nadat, door de proef van Elman was bevestigd, dat de absorptie bij 412 nm van 0,27 was teruggelopen tot 0,12 en daarop constant bleef, werd het verdunde filtraat op pH 5,0 gebracht met 5%'s 16 25 azijnzuur, en gelyofiliseerd tot 40 mg onzuiver [Gin 3-GH-RH.
Dit .onzuivere [Gin ]-GH-RH werd gesuspendeerd in een kleine hoeveelheid IN azijnzuur en gecentrifugeerd. De verkregen bovenstaande vloeistof werd gechromatografeerd over een kolom van 3 x 95 cm, volume 672 cm3, Sephadex G-25, waarbij werd geëlueerd met IN azijnzuur 30 en het kolomeffluent werd opgevangen in fracties van 4 cm3. De verkregen gel-gefiltreerde verbinding werd gelyofiliseerd, opgelost in 0,1N azijnzuur en daarna gecentrifugeerd ter verwijdering van onoplosbaar materiaal. De verkregen bovenstaande vloeistof werd gechromatografeerd over een kolom HPLC (^u Bondapak TM/C18). Geëlueerd werd met een line-35 aire gradiënt verkregen uit 10% en 50% acetonitrile in 0,1% TFA gedurende 30 minuten in een inrichting met constant niveau bij een stroom- - 83 00 4 1 3 ...............................
26 snelheid van 2 cm3 /minuut (4 cm3 /minuut) voor fractioneren).
De verkregen elutiekromme is weergegeven in figuur 7.
In figuur 7 heeft het gezuiverde peptide een enkele piek in het HPLC en de retentietijd is nagenoeg gelijk aan die van GH-RH 5 verkregen uit zalmen.
De fysische eigenschappen van het peptide luiden als : volgt: '

Analytisch voorbeeld 9 .
[Carboxypeptidase P-afbraak]
Bij de aminozuur-analyseruitgevoerd na een hydrolyse 30 met 6N HC1, kon niet worden aangetoond of Glu danwel Gin aanwezig is.
Om deze reden werd een C-terminale aminozuuranalyse met carboxypeptidase uitgevoerd.
In 150 mm3 0,1N pyridine-acetaat met pH 5,5 werd ca.
* ’ 16 4,4 ^,ug (ca. 2 nmol) [Gin ]-GH-RH volgens voorbeeldll of .hét GH-RH ver-35 kregen uit zalm volgens voorbeeld I opgelost en hieraan ca. 2 mm3 1 mg/ cm3 H^O aan carboxypeptidase P (Protein Research Foundation) toegevoegd.

Het reactiemengsel werd 3 uren bij 37°C geincubeerd.
Na verwijdering van het oplosmiddel werd het residu opgelost in 300 mm3 0,02N HC1 en geanalyseerd met de Hitachi 835-50 aminozuur auto-analyzer (Hitachi Ltd.)· De resultaten zijn weergegeven 5 in tabel O. Carboxypeptidase P is een exopeptidase die de C-terminale peptidebinding aantast, waarbij slechts één aminozuur wordt vrijgesteld.

Uit tabel O blijkt, dat Gin op de tweede plaats zit na 16 het C-terminale aminozuur in [Gin ]-GH-RH, terwijl Glu dezelfde positie inneemt in een GH-RH verkregen uit zalm.
25 - Dezelfde resultaten werden verkregen bij toepassing van carboxypeptidase A (Sigma Chemical Company) dat gemakkelijk Gin vrijstelt, maar nauwelijks Glu vrijstelt. De reactie werd uitgevoerd onder dezelfde voorwaarden als bij carboxypeptidase P, met dit verschil, dat een pyridine-acetaat met pH 7,8 werd toegepast in plaats van een pyridine-30 acetaat met pH 5,5. De resultaten zijn weergegeven in tabel E.

Re.cept-voorbeeld (Injectie-oplossing)
In 5 cm3 gedestilleerd en gesteriliseerd water werd 30 0,1 mg peptide volgens de uitvinding en 0,50 mg glycine opgelost, waarna de verkregen oplossing werd verdeeld in 10 flesjes en voor opslag werd gelyofiliseerd. Aan het gelyofiliseerde mengsel werd een passende hoeveelheid van een steriele fysiologische zoutoplossing toegevoegd om vlak voor toepassing een injectievloeistof te verkrijgen.

Farmacologisch, voorbeeld T·»
Kleine stukjes van de voorste hypofyse verkregen van mannelijke Wistar ratten werden op het boveneinde van een kolom aangebracht bestaande uit een injectiecilinder met 1 millimeter diameter, ge-5 vuld met 0,3 - 0,5 cm3 Sephadex G 25 geëquilibreerd met Krebs-Ringer-bufferoplossing met daarin 0,1% kalverserum waarbij een stroomsnelheid werd toegepast van 0,5 cm3 /minuut. Over deze kolom werd 1 cm3 van een . controle-pplossing (een Klebs-Ringer-buf feroplossing) gechromatografeerd; . 1 cm3 van een 2 ^ug/cm? oplossing van TRH en 1 cm3 van een 2 ^ug/cm3 10 oplossing van GH-RH verkregen volgens voorbeeld 1 in deze volgorde.
Het kolcm-effluent werd opgevangen in 1 cm3 fracties en elke fractie geanalyseerd met behulp van een radio-immunoproef met een anti-ratten GH antilichaam (NIH standaard) om de hoeveelheid vrijgesteld ratten-GH bij de proef te bepalen. De resultaten zijn weergegeven in tabel A.

Zoals blijkt uit tabel A was bij toepassing van het peptide volgens de uitvinding de hoeveelheid vrijgesteld GH vijf naai zo groot ' als bij de controle en gelijk aan die bij toepassing van TRH 25 met een GH-RH-achtig niet-specifiek effect.
De wijze van vrijstellen evenwel is specifiek, d.w.z. de vrijstellingswijze volgens de uitvinding is scherp, terwijl die bij TRH breed is.
Farmacologisch voorbeeld II.
30 [GH-vrijstellende activiteit van GH-RH in vivo]
Mannelijke Wistar-ratten van 10 weken elk werden toege past. Deze ratten werden geanesthetiseerd met pentabarbital (i.p.) toegediend in een dosering van 50 mg/kg en gefixeerd. Een .buisvormige (Dow coring 0,020 inwendige diameter bij 0,037 uitwendige diameter) cannu-35 la van 3,5 cm werd in de hartzijde van de rechter jugulaire ader van de ........8300413 30 rat geplaatst. In een rat met chronische ader-cannula-implantatie werd de andere zijde van de cannula met 3 cm uit de rugzijde Van de nek getrokken en gefixeerd op de buitenlaag van de huid.
Via de cannula werd 0,5 cm3 20 eenheden/cm3 heparine 5 geïnjecteerd en daarna de cannula gesloten met een roestvrijstalen stop. Hierna werd de rat geïnjecteerd met GH-RH en bloed gewonnen onder niet-anesthetische conditie op de volgende dag. Chloorpromazine-hydrochloride werd i.p. toegediend telkens 4 uur voor de injectie en wel in een dosering van 2 mg/kg. Vier ratten kregen eën injectie via de cannula van 10 440 ^.ug/kg GH-RH verkregen uit zalm volgens voorbeeld ï. Anderzijds kre gen drie ratten een fysiologische zoutoplossing als controle.
Van elke rat werd 0,5 cm3 bloed af genomen 15 minuten vóór de injectie en daarna 15, 30, 60, 120 en 180 minuten nd de injectie.
Het afgenomen bloed werd met 2000 g gedurende 10 minuten . 15 gecentrifugeerd. Voor het meten van de GH-vrij stellende activiteit werd . de bovenstaande vloeistof (plasma) onderzocht met een radio-immuno-proef (RIA) volgens een dubbele antilichaamsmethode (zie Birge et al Endocrinology SI, p.195 (1967)). De RIA van ratten-GH werd uitgevoerd onder toepassing van een NIH doos. De resultaten zijn weergegeven in ta-20 bel B en fig. 8, waarbij de symbolen A - G in tabel B corresponderen met die van figuur 8.

Uit figuur 8 blijkt een duidelijke verhoging aan GH-concentratie in het plasma, die werd waargenomen na 60 minuten na de injectie bij de groep die GH-RH had gekregen vergeleken met de controlegroep, zelfs wanneer het verschil tussen individuële ratten werd beke-5 ken.
Bij het vergelijken met de GH-vrijstellende activiteit van GH-vrijstellende factor geïsoleerd uit humane pancreas-kanker (hieronder aangeduid als hpGRF), vertoonde hpGRF een toename aan GH na. 5 mi-. nuten na de injectie (Guillemin et alf Science, 218, 585 (1982)), ter-10 Wijl het GH-RH verkregen uit zalm een verhoging van de GH een uur na de injectie aangaf. Uit deze resultaten blijkt, dat het GH-vrijstellend effect van het GH-RH verkregen uit zalm niet direct op de hypofyse werkt, maar dit effect op een andere wijze tot stand doet komen. · . .

De procedure van het voorgaande farmacologische voorbeeld werd herhaald met dit verschil, dat zes ratten werden gebruikt als de groep die [Gin16]-GH-RH kreeg en~zes TOr'dé~y^p~aië~^s^ól^isëh¥" -zoutoplossing 'kreeg~£öegediehd','' terwi‘j r"di“filoé<föpva£glwefd~uiÏ^revoerd_ 20 15 minuten vóór de injectie en'15','"'30',' 60, 120, 18Ó, 24Ó en 300 minuten ‘ na de injectie.

34 ‘ Uit figuur 9 blijkt een duidelijke toename aan GH· concen tratie in het plasma 30 tot 60 minuten na de injectie bij de groep die 16 [Gin ]-GH-RH kreeg toegediend vergeleken met de groep die als controle fungeerde, waarbij zelfs het verschil tussen de individuële ratten door- 16 5 slaggevend was. Hoew'el het GH-vrijstellend effect van [Gin ]-GH-RH iets eerder komt dan dat van een GH-RH verkregen uit zalm, is het duidelijk, 16 dat dit [Gin]-GH-RH een GH-vrijstellend effect bezit.
Uit de resultaten van de farmacologische voorbeelden „I -ΙΊΓ blijkt duidelijk, dat de onderhavige peptiden een GH-vrij stellend ef-10 fect hebben op vreemde dieren.

Claims (6)
1- Een peptide of zout ervan met de structuurformule: H-Asp-Thr-Met-Arg-C^s-Met-Va1-Gly-Arg ___Val-Tyr-Arg-Pro-Cys-Trp-Glx-Val-OH ^ waarin Glx: Glu of Gin voorstelt en met de volgende fysische eigenschappen 5 (1) ultraviolet absorptiespectrum: maximum 2S0 nm, (2) Ehrlich-reactie, Sakaguchi-reactie en Pauly's-reactie: positief, C3) basische reactie, (4) oplosbaar in water, methanol en azijnzuur, slecht oplosbaar in ethylacetaat, butylacetaat, ethylether, hexaan, petroleumether, 10 benzeen en chloroform, {5) met het uiterlijk van een wit poeder.
2. Peptide volgens conclusie 1, waarin Glx: Glu is.
3. Peptide volgens conclusie 1, met het kenmerk, dat Glx: Gin is.
4. Groeihormoon-vrij stellend hormoonpreparaat met daarin een peptide of een zout ervan met de structuurformule: H-Asp-Thr-Met-Arg-Cys -Met-Val-Gly-Arg- I , (1) Val-Tyr-Arg-Pro-Cys-Trp-Glx-Val-OH waarin Glx: Glu of Gin voorstelt en de in conclusie 1 weergegeven fysi- 20 sche eigenschappen.
5. Groeihormoon-vrijstellend hormoonpreparaat volgens conclusie 4, met het kenmerk, dat Glx gelijk is aan Glu.
6. Groeihormoon-vrij stellend hormoonpreparaat volgens conclusie 4, met het kenmerk, dat Glx gelijk is aan Gin. 25 8300 4 1 3 ........................
