Abstract

The present disclosure provides a feed additive containing Trollius chinensis Bunge and a feed, and belongs to the technical field, of feed, additives. The feed, additive containing Trollius chinensis Bunge includes a Trollius chinensis Bunge extract, compound amino acids, vitamin C, vitamin E and organic selenium. The Trollius chinensis Bunge extract containing components such as total flavonoids, phenolic acids and trolline has effects of clearing away heat and detoxifying, and antibacterial and anti— inflammatory effects, and can also improve resistance and growth performance of livestock and poultry; and the compound amino acids can improve the efficiency of protein synthesis in animals, relatively reduce fat content of animals, increase a proportion of lean meat, and improve meat quality, thereby effectively improving digestive functions of animals, reducing diseases and, enhancing disease resistance of animals.

TECHNICAL FIELD
The present disclosure belongs to the technical field of feed additives, and particularly relates to a feed additive containing
Trollius chinensis Bunge and a feed.
BACKGROUND ART
Trollius chinensis Bunge is an annual or perennial herb be- longing to genus Trollius in the family Ranunculaceae, with a plant height of 30-100 cm, soft climbing stems, round leaves like lotus leaves, and trumpet-shaped flowers with elongated calyx tubes, commonly yellow, orange and red.Trollius chinensis Bunge is often used to treat acute and chronic tonsillitis, acute otitis media, acute myringitis, acute conjunctivitis and acute lymphangi- tis because of its medicinal functions of clearing away heat and detoxifying. In addition, Trollius chinensis Bunge is spicy, and its tender shoots and buds can be used as food seasonings. Howev- er, there is no report on use of Trollius chinensis Bunge as a feed additive.
SUMMARY
In view of this, an objective of the present disclosure is to provide a feed additive containing Trollius chinensis Bunge and a feed which are capable of significantly improving resistance and growth performance of livestock and poultry.
To achieve the objective, the present disclosure provides the following technical solutions:
A feed additive containing Trollius chinensis Bunge, includ- ing the following raw materials by weight: 20-30 parts of Trollius chinensis Bunge extract, 10-20 parts of compound amino acids, 5-10 parts of vitamin C, 5-10 parts of vitamin E and 5-10 parts of or- ganic selenium.
Preferably, a method for preparing the Trollius chinensis
Bunge extract includes: mixing a Trollius chinensis Bunge powder with a fermentation medium, inoculating a Lactobacillus liquid for fermentation, extracting a resulting fermentation broth in an eth-
anol solution, filtering, concentrating and vacuum-drying a fil- trate to obtain the Trollius chinensis Bunge extract.
Preferably, the fermentation medium includes the following components by weight: 10-15 parts of glucose, 10-20 parts of pep- tone, 1-3 parts of potassium dihydrogen phosphate, 1-3 parts of magnesium sulfate and 40-50 parts of water.
Preferably, a mass ratio of the Trollius chinensis Bunge pow- der to the fermentation medium is (65-75): (22-41).
Preferably, the fermentation is carried out at 35-40 °C for 3-5 d.
Preferably, a volume percent fraction of the ethanol solution is 70-80%, and a volume ratio of the fermentation broth to the ethanol solution is 1:1-3.
Preferably, the ethanol extraction is carried out at 55-65 °C for 3-5 h.
Preferably, the compound amino acids include lysine, methio- nine, tryptophan and threonine; and a mass ratio of the lysine, the methionine, the tryptophan and the threonine is (10-18): (5- 10): (3-7): (2-5).
Preferably, the organic selenium is yeast selenium.
The present disclosure further provides a feed containing
Trollius chinensis Bunge, including the feed additive and a basic feed; and a weight ratio of the feed additive to the basic feed is (0.5-2):100.
Compared with the prior art, the present disclosure has the following beneficial effects:
The present disclosure provides a feed additive containing
Trollius chinensis Bunge. The Trollius chinensis Bunge extract containing components such as total flavonoids, phenolic acids and trolline has effects of clearing away heat and detoxifying, and antibacterial and anti-inflammatory effects, and can also improve resistance and growth performance of livestock and poultry; and the compound amino acids can improve the efficiency of protein synthesis in animals, relatively reduce fat content of animals, increase a proportion of lean meat, and improve meat quality, thereby effectively improving digestive functions of animals, re- ducing diseases and enhancing disease resistance of animals. The
Trollius chinensis Bunge extract, the compound amino acids, the vitamin C, the vitamin E and the organic selenium of the present disclosure are combined to provide a synergistic effect, which can not only improve resistance of livestock and poultry, but also im- prove the growth performance.
DETAILED DESCRIPTION OF THE EMBODIMENTS
The present disclosure provides a feed additive containing
Trollius chinensis Bunge, including the following raw materials by weight: 20-30 parts of Trollius chinensis Bunge extract, 10-20 parts of compound amino acids, 5-10 parts of vitamin C, 5-10 parts of vitamin E and 5-10 parts of organic selenium.
In the present disclosure, the feed additive preferably in- cludes the following raw materials by weight: 25 parts of Trollius chinensis Bunge extract, 15 parts of compound amino acids, 8 parts of vitamin C, 8 parts of vitamin E and 8 parts of organic seleni- um.
In the present disclosure, a method for preparing the Trolli- us chinensis Bunge extract preferably includes: mixing a Trollius chinensis Bunge powder with a fermentation medium, inoculating a
Lactobacillus liquid for fermentation, extracting a resulting fer- mentation broth in an ethanol solution, filtering, concentrating and vacuum-drying a filtrate to obtain the Trollius chinensis
Bunge extract.
In the present disclosure, the fermentation medium preferably includes the following components by weight: 10-15 parts of glu- cose, 10-20 parts of peptone, 1-3 parts of potassium dihydrogen phosphate, 1-3 parts of magnesium sulfate and 40-50 parts of wa- ter, and more preferably includes 13 parts of glucose, 15 parts of peptone, 2 parts of potassium dihydrogen phosphate, 2 parts of magnesium sulfate and 45 parts of water. In the present disclo- sure, a mass ratio of the Trollius chinensis Bunge powder to the fermentation medium is preferably (65-75): (62-91), more preferably 70:77.
In the present disclosure, the Lactobacillus is preferably
Lactobacillus plantarum with collection number of CGMCC No.5494.
Preparation of a yeast liquid and the Lactobacillus liquid is not specifically limited in the present disclosure. As an implementa-
tion, a method for preparing the Lactobacillus liquid includes: subculturing a lyophilized Lactobacillus plantarum powder twice in an MRS broth until viable count reaches more than 5x10’ CFU/mL.
In the present disclosure, an inoculum dose of the Lactoba- cillus is preferably 2-4%, more preferably 3%; and the fermenta- tion is carried out preferably at 35-40 °C, more preferably 37 °C, preferably for 3-5 d, more preferably 4 d.
In the present disclosure, a volume percent fraction of the ethanol solution is preferably 70-80%, more preferably 75%, a vol- ume ratio of the fermentation broth to the ethanol solution is preferably 1:1-3, more preferably 1:2, and the ethanol extraction is carried out preferably at 55-65 °C, more preferably 60 °C, preferably for 3-5 h, more preferably 4 h.
In the present disclosure, the concentration by evaporation is carried out preferably at a relative density of 1.25-1.65, more preferably 1.5. The vacuum drying is carried out preferably at a vacuum degree of -0.15-0.05 MPa, more preferably -0.1 MPa, prefer- ably at 40-50 °C, more preferably 45 °C, preferably for 2-4 h, more preferably 3 h.
In the present disclosure, the compound amino acids prefera- bly include lysine, methionine, tryptophan and threonine; and a mass ratio of the lysine, the methionine, the tryptophan and the threonine is preferably (10-18):{5-10):{3-7): (2-5), more prefera- bly 14:8:5:3. In the present disclosure, the compound amino acids can improve the efficiency of protein synthesis in animals, rela- tively reduce fat content of animals, increase a proportion of lean meat, and improve meat quality, thereby effectively improving digestive functions of animals, reducing diseases and enhancing disease resistance of animals. Among them, the lysine can improve growth rate of pigs, reduce feed consumption, improve meat quality and increase lean meat percentage; the methionine can inhibit and kill bacteria and reduce heat stress; the tryptophan has effects of enhancing appetite and feed intake of pigs; and the threonine can balance amino acids and promote protein synthesis.
In the present disclosure, the organic selenium is preferably yeast selenium, which has effects of promoting animal growth, en- hancing immune functions, improving carcass quality and improving growth performance.
In the present disclosure, the Trollius chinensis Bunge ex- tract containing components such as total flavonoids, phenolic ac- ids and trolline has effects of clearing away heat and detoxify- 5 ing, and antibacterial and anti-inflammatory effects, and can also improve resistance and growth performance of livestock and poul- try; and the compound amino acids can improve the efficiency of protein synthesis in animals, relatively reduce fat content of an- imals, increase a proportion of lean meat, and improve meat quali- ty, thereby effectively improving digestive functions of animals, reducing diseases and enhancing disease resistance of animals. The
Trollius chinensis Bunge extract, the compound amino acids, the vitamin C, the vitamin E and the organic selenium of the present disclosure are combined to provide a synergistic effect, which can not only improve resistance of livestock and poultry, but also im- prove the growth performance.
The method for preparing the feed additive is not specifical- ly limited in the present disclosure, and the feed additive can be prepared by a conventional method in the art. As an implementa- tion, the Trollius chinensis Bunge extract, the compound amino ac- ids, the vitamin C, the vitamin E and the organic selenium are mixed well, granulated and air-dried to obtain the feed additive.
The present disclosure further provides a feed containing
Trollius chinensis Bunge, including the feed additive and a basic feed; and a weight ratio of the feed additive to the basic feed is (0.5-2):100. Further preferably, a weight ratio of the feed addi- tive to the basic feed is 1:100.
The method for preparing the feed containing Trollius chinen- sis Bunge is not specifically limited in the present disclosure, and the feed can be prepared by a conventional method in the art.
As an implementation, the method for preparing the feed containing
Trollius chinensis Bunge includes: mixing the basic feed with the feed additive to obtain the feed.
Sources of the raw materials involved in the present disclo- sure are not specifically limited, and commercially available products known in the art can be used.
The technical solutions provided in the present disclosure will be described in detail with reference to embodiments below, but the embodiments cannot be construed as limiting the protection scope of the present disclosure.
Embodiment 1 130 parts of glucose, 150 parts of peptone, 20 parts of po- tassium dihydrogen phosphate, 20 parts of magnesium sulfate, 450 parts of water and 700 parts of Trollius chinensis Bunge powder are weighed; the glucose, the peptone, the potassium dihydrogen phosphate, the magnesium sulfate and the water are mixed and ster- ilized, and then mixed with the Trollius chinensis Bunge powder; 44.1 parts of Lactobacillus plantarum liquid with viable count more than 5x10’ CFU/mL is inoculated for fermentation at 37 °C for 4 d, then twice the volume of 75% ethanol is added to a fermenta- tion broth for extraction at 60 °C for 4 h before filtration, and a filtrate is concentrated to a relative density of 1.5, and dried at 45 °C under a vacuum degree of -0.1 MPa for 3 h to obtain a
Trollius chinensis Bunge extract.
Embodiment 2 100 parts of glucose, 100 parts of peptone, 10 parts of po- tassium dihydrogen phosphate, 10 parts of magnesium sulfate, 400 parts of water and 650 parts of Trollius chinensis Bunge powder are weighed; the glucose, the peptone, the potassium dihydrogen phosphate, the magnesium sulfate and the water are mixed and ster- ilized, and then mixed with the Trollius chinensis Bunge powder; 25.4 parts of Lactobacillus plantarum liquid with viable count more than 5x10’ CFU/mL is inoculated for fermentation at 35 °C for 5 d, then an equal volume of 80% ethanol is added to a fermenta- tion broth for extraction at 55 °C for 5 h before filtration, and a filtrate is concentrated to a relative density of 1.25, and dried at 50 °C under a vacuum degree of -0.15 MPa for 2 h to obtain a
Trollius chinensis Bunge extract.
Embodiment 3 150 parts of glucose, 200 parts of peptone, 30 parts of po- tassium dihydrogen phosphate, 30 parts of magnesium sulfate, 500 parts of water and 750 parts of Trollius chinensis Bunge powder are weighed; the glucose, the peptone, the potassium dihydrogen phosphate, the magnesium sulfate and the water are mixed and ster- ilized, and then mixed with the Trollius chinensis Bunge powder; 66.4 parts of Lactobacillus plantarum liquid with viable count more than 5x10’ CFU/mL is inoculated for fermentation at 40°C for 3 d, then 3 times the volume of 70% ethanol is added to a fermenta- tion broth for extraction at 65°C for 2 h before filtration, and a filtrate is concentrated to a relative density of 1.65, and dried at 40°C under a vacuum degree of -0.05 MPa for 4 h to obtain a
Trollius chinensis Bunge extract.
Embodiment 4 250 g of the Trollius chinensis Bunge extract prepared in Em- bodiment 1, 70 g of lysine, 40 g of methionine, 25 g of trypto- phan, 15 g of threonine, 80 g of vitamin C, 80 g of vitamin E and 80 g of yeast selenium are weighed, mixed well, granulated and air-dried to obtain a feed additive.
Embodiment 5 200g of the Trollius chinensis Bunge extract prepared in Em- bodiment 2, 50g of lysine, 25g of methionine, 15g of tryptophan, 10g of threonine, 50 g of vitamin C, 50 g of vitamin E and 50 g of yeast selenium are weighed, mixed well, granulated and air-dried to obtain a feed additive.
Embodiment 6 300g of the Trollius chinensis Bunge extract prepared in Em- bodiment 3, 90g of lysine, 50g of methionine, 35g of tryptophan, 25g of threonine, 100g of vitamin C, 100g of vitamin E and 100g of yeast selenium are weighed, mixed well, granulated and air-dried to obtain a feed additive.
Comparative Example 1
The specific implementation is the same as that of Embodiment 4, except that the Trollius chinensis Bunge extract prepared in
Embodiment 1 is not added.
Comparative Example 2
The specific implementation is the same as that of Embodiment 4, except that the compound amino acids are not added.
Comparative Example 3
The specific implementation is the same as that of Embodiment 4, except that the vitamin C and the vitamin E are not added.
Comparative Example 4
The specific implementation is the same as that of Embodiment 4, except that the yeast selenium is not added.
Experiment Example 1
A total of 240 DLY pigs with similar hereditary basis and in- itial weight of (20.0+0.7}) kg were randomly divided into 8 treat- ments, with 3 replicates for each treatment and 10 pigs for each replicate, with half male and half female. Pigs in the 8 treat- ments were raised in pigpens with cement floor and steel pipe par- titions without automatic feeding system.
A feed includes feed additives and a basic feed, the basic feed includes the following raw materials by percentage: 40% of corn, 12% of wheat bran, 5% of peanut bran, 20% of soybean meal, 5% of grass meal, 12.5% of rice, 3% of pine needle powder and 2.5% of water.
Pigs in the 8 treatments were fed freely with the feed con- taining the feed additives in Embodiments 4-6 and Comparative Ex- amples 1-4, respectively, and pigs in one treatment were fed with a basic feed without the feed additives as a control. After the pigs were raised to about 60 kg, growth performance of the pigs in each treatment was detected and calculated at the same time. Refer to Table 1 for details.

According to data in Table 1, compared with Comparative Exam- ples 1-4 and the control, Embodiment 4 in the present disclosure can significantly improve daily feed intake and daily gain, and reduce feed conversion ratio. It can be seen that the Trollius chinensis Bunge extract, the compound amino acids, the vitamin C, the vitamin E and the organic selenium in the feed additive of this application provide a synergistic effect, and can signifi- cantly improve growth performance of growing pigs.

It should be noted that those of ordinary skill in the art can make a plurality of improvements and modifications to the present disclosure without departing from the principle of the present disclosure, and these improvements and modifications should fall within the protection scope of the present disclosure.
