Abstract
Se proporciona un método para conjugar luciferasa con una entidad química, especialmente con un agente de enlace específico como, por ejemplo, un anticuerpo, antígeno o bien ácido nucleico, y más especificamente un anticuerpo, que comprende (a) la mezcla de la luciferasa con uno varios de los siguientes: D-luciferina, iones magnesio y adenosintrifosfato y (b) realizar una reaccion de acoplamiento covalente entre la luciferasa y el reactivo de enlace usando un reactivo de acoplamiento donde la cantidad de D-luciferina, iones magnesio y/o adenosinatrifosfato es suficiente para proteger la actividad de la luciferasa contra la inhibicion por el reactivo de acoplamiento covalente. De preferencia el paso (a) se lleva a cabo mediante la mezcla de la luciferasa con sus sustratos en la solucion y de preferencia tanto el magnesio como el adenosintrifosfato se encuentran presentes como adenosintrifosfato de magnesio (Mg++ATP), opcionalmente junto con D-luciferina. En un segundo aspecto de la presente invencion, se proporciona una entidad química marcada que comprende una entidad química conjugada con luciferasa activa de conformidad con lo proporcionado por el método de la presente invencion. De preferencia, la entidad química es un agente de enlace especifico para su uso en un ensayo de enlace especifico, de preferencia un anticuerpo, antígeno o bien ácido nucleico. Cuando el agente de enlace es un ácido nucleico, es de preferencia un oligonucleotido, pero puede ser un polinucleotido o bien un nucleosido, y puede emplearse como sonda de hidratacion o bien como iniciador de extension de cadena, por ejemplo, un iniciador de reaccion en cadena de polimerasa. De manera más provechosa, la entidad es un anticuerpo puesto que intentos previos en inactividad. Se proporcionan adicionalmente conjuntos de implementos de prueba.
