Abstract
En la presente invencion se ha desarrollado un sistema de expresion adecuado para la produccion y secrecion de hormona de crecimiento humano (HGH) en su forma natural y biologicamente activa, en el cual se emplea como organismo hospedero una levadura metilotrofica, tal como Pichia pastoris. En donde la presente invencion comprende la transformacion de una levadura metilotrofica con al menos una copia de una secuencia funcional del ADNc que codifica para HGH, la cual está funcionalmente asociada con una segunda secuencia de ADN que codifica para la secuencia pre-pro del factor alfa de S. cerevisiae (incluyendo el sitio de procesamiento proteolítico: Iys-arg), y en donde ambas secuencias de ADN se encuentran bajo la regulacion de un promotor de un gen de una levadura metilotrofica, el cual es inducible con metanol. Levaduras metiolotroficas conteniendo en us genoma al menos una copia de esta secuencia de ADN producen y secretan de forma eficiente HGH madura, correctamente procesada y biologicamente activa hacia el medio de cultivo.
