Abstract

La presente invención provee un novedoso método de producir una pluralidad de polinucleótidos modificados que tienen diferentes combinaciones de diversas mutaciones en múltiples sitios mediante un ensamblaje combinacional en sitios múltiples, a la medida, comprendiendo añadir al menos dos o al menos tres cebadores a un polinucleótido de plantilla de doble cadena en una sola mezcla de reacción, donde los cebadores no traslapan, y donde cada uno de los cebadores comprende al menos una mutación diferente de los demás cebadores, donde al menos un cebador es un cebador hacia adelante que puede templarse en una cadena menos de la plantilla y al menos un cebador es un cebador en reversa que puede templarse en una cadena mas de la plantilla, y someter la mezcla de reacción a una reacción de extensión de polimerasa para rendir una pluralidad de polinucleótidos modificados, extendidos a partir de los al menos tres cebadores. El método puede ser llevado a cabo sin emplear un paso de ligación antes de transformar los polinucleótidos modificados, extendidos en una célula. La pluralidad de polinucleótidos modificados, extendidos puede ser tratada con una enzima para destruir el polinucleótido de plantillas antes de transformarlo en la célula.
