Abstract

Se proporcionan métodos para amplificación de ácido nucleico que incluyen poner en contacto un ácido nucleico de doble cadena con transposasas unidas a un transposón de ADN, en donde el transposón de ADN incluye un sitio de unión a transposasa y una secuencia promotora de polimerasa de ARN polimerasa, en donde el complejo de transposasas/transposón de ADN se une a las ubicaciones objetivo a lo largo del ácido nucleico de doble cadena y descompone el ácido nucleido de doble cadena en una pluralidad de fragmentos de doble cadena, cada fragmento de doble cadena tiene el transposón de ADN unido a cada extremo 5'del fragmento de doble cadena, extender los fragmentos de doble cadena a lo largo del transposón de ADN para hacer productos de extensión de doble cadena con secuencias promotoras de ARN polimerasa de doble cadena en cada extremo, poner en contacto los productos de extensión de doble cadena con una ARN polimerasa para hacer una pluralidad de transcripciones de ARN de cada producto de extensión de doble cadena, transcribir de forma inversa las transcripciones de ARN en un ADN de un copia de una sola cadena, formar cadenas complementarias en el ADN de copia de una sola cadena para formar una pluralidad de amplicones de ADN de doble cadena que corresponden a cada fragmento de doble cadena, en donde los amplicones de ADN de doble cadena se amplifican de manera lineal desde los fragmentos de doble cadena originales.
