Abstract
본 발명은 성장호르몬에 대해 증진된 친화성 및 저하된 호르몬 활성을 나타내는 변이형 인간 성장호르몬, 그를 코드하는 염기배열, 그의 제조 방법 및 그의 용도를 제공한다. 성장호르몬 수용체에 대한 증진된 친화성을 갖는 본 발명의 단백질은, 저화된 성장호르몬 활성을 유지하면서, 수용체에 대한 성장호르몬의 결합을 저해하며, 따라서 거인증 및 말단비대증 치료제로서 적용할 수 있다.

제1도는 실시예 1에서 수득된, 변이형 성장호르몬 (77번째 위치, R→C)아미노산 배열 및 상기 단백질을 코드하는 염기배열이다.
Claims (14)
- 제1도에 주어진 아미노산 배열을 나타내는 변이형 인간 성장호르몬 단백질.
- 청구항 1에서 청구된 아미노산 배열을 코드하는, 제1도에서 주어진 염기서열을 나타내는 데옥시리보뉴클레오티드.
- 제1항에 있어서, 상기 변이형 인간 성장호르몬이 성장호르몬 수용체 단백질에 대해 증진된 친화성을 나타내고 또한 감소된 성장호르몬 활성을 유지하는 것을 특징으로 하는 변이형 인간 성장호르몬 단백질.
- 성장호르몬 수용체에 대해 증진된 친화력을 나타내고 감소된 성장호르몬 활성이 유지되는 그들의 특징적인 성질을 잃지 않을 정도로 그의 아미노산 잔기 부위에 부분적인 치환, 삽입 또는 소실을 실시한, 청구항 1에서 청구한 아미노산 배열을 나타내는 변이형 인간 성장호르몬 단백질.
- 제2도에서 주어진 아미노산 배열, 및 그를 코드하는 데옥시리보뉴클레오티드를 나타내는 단백질.
- 제3도에 주어진 염기서열의 데옥시리보뉴클레오티드가 코드하는 아미노산 배열을 나타내는 단백질.
- 청구항 1에서 청구된 단백질의 수용체 단백질 결합 부위를 구성하는 펩티드 및 상기 펩티드의 아미노산 배열을 코드하는 데옥시뉴클레오티드.
- 청구항 4에서 청구된 단백질의 아미노산 배열을 코드하는 염기서열을 나타내는 데옥시리보뉴클레오티드.
- 청구항 2, 5, 6, 7 또는 8에서 청구된 데옥시리보뉴클레오티드를 프로모터의 하류에 각각 배열시킨 발현 플라스미드.
- 청구항 9에서 청구된 플라스미드로 형질전환된 생물체 세포에 의한 발현을 통해 산출된 생성물.
- 청구항 1, 4, 5 또는 6에서 청구된 변이형 인간 성장호르몬 단백질과 상호반응을 나타내는 항체.
- 제11항에 있어서, 단성항체인 것을 특징으로 하는 항체.
- 청구항 1, 4, 5, 6 또는 7에서 청구된 단백질 또는 펩티드를 활성성분으로 함유하는 거인증 및 말단비대증 치료제.
- 청구항 2, 5, 6, 7 또는 8에서 청구된 데옥시리보뉴클레오티드를 이용하는 유전자요법용 약제.※ 참고사항 : 최초출원 내용에 의하여 공개하는 것임.
