Abstract
본 발명은 변형되고, 소포체(ER)내에 위치하는 "이염기성 프로세싱 엔도프로테아제"를 함호화하는 DNA분자및 형질전환된 숙주내에서 이종 폴리펩타이드의 정확한 프로세싱을 위한 상기의 소포체내에 위치하는 "이염기성 프로세싱 엔도프로테아제"의 용도에 관한 것이다.

Claims (14)
- 소포체(ER) 내에 "이염기성 프로세싱 엔도프로테아제'활성을 갖는 숙제 세포를 사용함을 특징으로 하여, 숙주 세포내에서 프로-서열이 절단된 이종 단백질을 제조하는 방법.
- 제1항에 있어서, ER내에 위치하는 KEX2 또는 YAP3 프로테아제를 갖는 효모 숙제 제포를 사용하는 방법.
- 제1항에 있어서, ER내에 위치하는 KEX2 프로테아제를 갖는 효모 숙주 제포를 사용하는 방법.
- 제1항에 있어서, 효모-인자 프로 서열이 절단된 이종의 생물학적으로 활성인 단백질을 제조하는 방법.
- 제1항에 있어서, 인슐린-관련된 단백질을 제조하는 방법.
- ER내에 위치하는 "이염기성 프로세싱 엔도프로테아제"에 대한 발현 카세트를 암호화하는 재조합 DNA 분자.
- 제6항에 있어서, "이염기성 프로세싱 엔도프로테아제"및 ER-보유 시그날로 이루어진 ER내에 위치하는 "이염기성 프로세싱 엔도프로테아제"를 암호화하는 재조합 DNA분자.
- 제6항에 있어서, 가용성의 "이염기성 프로세싱 엔도프로테아제" 및 ER-보유 시그날로 이루어진 ER내에 위치하는 '이염기성 프로세싱 엔도프로테아제"를 암호화하는 재조합 DNA 분자.
- 제7항에 있어서, KEX2pHDEL, KEX2pSHDEL및 YAP3HDEL로 이루어진 그룹중에서 선택된 단백질을 암호화하는 재조합 DNA 분자.
- 제6항에 따른 재조합 DNA 분자를 포함하는 하이브리드 벡터.
- 제10항에 따른 하이브리드 벡터로 형질전환된 숙주 세포.
- 제11항에 있어서, 안정하게 형질전환된 숙주 세포.
- 제6항에 따른 재조합 DNA 분자를 제조하는 방법.
