Description
The present invention relates to a composition for predicting peritoneal fibrosis, a kit, and a method of providing information for predicting peritoneal fibrosis.
Epithelial-Mesenchymal Transition (EMT) is a form of cell formation in which one type of cell differentiates into another type of cell, and refers to the process by which epithelial cells are converted into a highly mobile representation of mesenchymal cells. . When EMT occurs, epithelial cells lose polarity and the cell shape changes from square to fibroblast-like, and epithelial cell markers decrease and mesenchymal cell markers increase. If this process progresses irreversibly, it can lead to heart, liver, kidney and vascular dysfunction as well as malignant transformation. In other words, EMT is involved in normal physiological processes such as embryo formation and organ development in the prenatal period and wound healing and tissue regeneration in adulthood, but pathologically it causes tissue fibrosis, cancer progression, metastasis, and drug resistance.
Meanwhile, peritoneal fibrosis (PF) is a major cause of peritoneal structural changes and ultrafiltration failure in patients undergoing continuous ambulatory peritoneal dialysis (CAPD). Encapsulating peritoneal sclerosis (ESP), an extreme form of peritoneal fibrosis, is a representative cause of difficulty in continuing peritoneal dialysis. To date, there is no clear prevention method, and due to the nature of fibro-sclerosis, return to normal function is impossible. For this reason, once diagnosed, it has a very fatal course.
To date, epithelial-mesenchymal transition (EMT) of peritoneal mesothelial cells (PMC) and peritoneal accumulation of extracellular matrix (ECM) proteins are the main characteristics of peritoneal fibrosis. 그 결과로부터 대조군(복막 기능 유지군)과 실험군(복막 기능 악화군)으로 분류하였다(도 1 참조).Peritoneal function tests (PET) were performed on 13 patients who started peritoneal dialysis at the time of initial dialysis and 3 months after dialysis. In each group, peritoneal mesothelial cells were isolated from the drainage fluid obtained after dialysis, and the cell shape was observed. From the results, they were classified into the control group (peritoneal function maintenance group) and the experimental group (peritoneal function deterioration group) (see Figure 1).
Afterwards, ribonucleic acid (RNA) was extracted from the peritoneal mesothelial cells of each group and subjected to ribonucleic acid sequencing (RNA-seq.).
2> RNA seq.<Example 2> RNA seq.
RNA seq. Genes whose expression levels changed in the group with worsening peritoneal function compared to the group with maintained peritoneal function were investigated through RNA seq.
