본 발명은, 신규한 항-PD-1 항체 또는 이의 기능적인 단편을 제공한다. 특히, 본 발명은 추가적으로, 종양을 치료하기 위한 약제의 제조에서 항체의 용도에 관한 것이다.

PD-1 유전자는, 프로그램된 세포 사멸 단백질 1(programmed cell death protein 1)로서 명명되고, T 세포 하이브리도마가 아포토시스를 받은 경우에, 상향-조절된다. PD-1 (CD279)는 활성화된 T 세포, B 세포, 및 활성화된 골수성 세포로 표시되고(Ishida Y, Agata Y, Shibahara K, Honjo T. 지금까지, PD-L1-PD-1 조절 축을 특이적으로 차단하고, 항-종양 및 항-바이러스 면역 반응의 활성화를 제공하도록, 암 환자에 대한 효과적인 면역 반응을 유도할 수 있는 만족스러운 방법이 여전히 없다. 따라서, 암 또는 만성 감염을 가지는 환자의 면역 저해를 극복하기 위해, PD-L1-PD-1 조절 축을 특이적으로 차단하기 위한 치료학적 방법의 설계 및 발전에 대한 절실한 필요성이 있다.

Claims (13)
- PD-1에 특이적으로 결합할 수 있는, 중쇄 및 경쇄를 포함하는 항체로서,
(ⅰ) 상기 중쇄는, SEQ ID NO: 7, 8 및 11 또는 9, 10 및 11에 의해 각각 나타낸 아미노 서열을 가지는 H-CDR1, H-CDR2 및 H-CDR3을 포함하고; 및
(ⅱ) 상기 경쇄는, SEQ ID NO:12, 13 및 14에 의해 각각 나타낸 아미노 서열을 가지는 L-CDR1, L-CDR2 및 L-CDR3을 포함하는 것인, 항체.
- 제1항에 있어서,
(ⅰ) 상기 중쇄는 SEQ ID NO: 1, 2, 4 또는 5에 의해 나타낸 아미노 서열을 가지는 중쇄 가변 영역을 포함하고; 및
(ⅱ) 상기 경쇄는 SEQ ID NO: 3 또는 6에 의해 나타낸 아미노 서열을 가지는 경쇄 가변 영역을 포함하는 것인, 항체.
- 제1항에 있어서,
(ⅰ) 상기 중쇄는 SEQ ID NO: 4 또는 5에 의해 나타낸 아미노 서열을 가지는 중쇄 가변 영역을 포함하고; 및
(ⅱ) 상기 경쇄는 SEQ ID NO: 6에 의해 나타낸 아미노 서열을 가지는 경쇄 가변 영역을 포함하는 것인, 항체.
- 제1항에 있어서,
상기 항체는 10F8, 15H6, BA08-1 및 BA08-2로부터 선택된 것인, 항체.
- 제1항 내지 제4항 중 어느 한 항에 따른 항체를 인코딩하는, 분리된 폴리뉴클레오티드.
- 제1항 내지 제4항 중 어느 한 항에 따른 항체의 경쇄를 인코딩하는 폴리뉴클레오티드, 및 제1항 내지 제4항 중 어느 한 항에 따른 항체의 중쇄를 인코딩하는 폴리뉴클레오티드를 포함하는, 분리된 폴리뉴클레오티드의 조합물(combination).
- 제5항에 따른 폴리뉴클레오티드 또는 제6항에 따른 폴리뉴클레오티드의 조합물을 포함하는 발현 벡터로서,
상기 폴리뉴클레오티드는, 숙주 세포 또는 세포-프리 발현 시스템(cell-free expression system)에서 상기 폴리뉴클레오티에 의해 인코딩되는 폴리펩티드의 발현을 가능하게 하는 조절 서열(regulatory sequence)에 효과적으로 연결하는 것인, 발현 벡터.
- 제1항 내지 제4항 중 어느 한 항에 따른 항체, 및 약제학적으로 용인가능한 담체를 포함하는, 약제학적 조성물.
- 제1항 내지 제4항 중 어느 한 항에 따른 항체, 제5항에 따른 폴리뉴클레오티드, 제6항에 따른 폴리뉴클레오티드의 조합물, 제7항에 따른 발현 벡터 및/또는 제8항에 따른 약제학적 조성물을 대상에 투여하는 것을 포함하는, 이를 필요로 하는 대상에서 암 또는 감염성 질환을 치료하거나 예방하기 위한 방법.
- 제9항에 있어서,
상기 암은 폐암 또는 흑색종이고, 상기 감염성 질병은 HIV 감염 또는 간염 B 바이러스 감염인 것인, 방법.
- 제1항 내지 제4항 중 어느 한 항에 따른 항체, 제5항에 따른 폴리뉴클레오티드, 제6항에 따른 폴리뉴클레오티드의 조합물, 제7항에 따른 발현 벡터 및/또는 제8항에 따른 약제학적 조성물을 대상에 투여하는 것을 포함하는, 이를 필요로 하는 대상에서 T 세포 면역 반응을 증진시키기 위한 방법.
- 제11항에 있어서,
T 세포 면역 반응을 증진시키는 것은, T 세포의 사이토카인 생산을 증진시키는 것을 포함하고, 바람직하게는, 상기 사이토카인은 IL-2 및/또는 IFN-γ를 포함하는 것인, 방법.
- 제11항에 있어서,
상기 방법은, 상기 대상에 항-CD3 항체를 투여하는 것을 더 포함하는 것인, 방법.
