B16 세포를 준비한 각 웰에 천연소재 혼합추출물 500ppm을 투여하고 배양하였다. 그리고 각 웰(well)에 MTT 용액(5㎎/㎖)을 성장배지의 10분의 1을 가해주고 다시 37℃에서 4시간 더 배양하여 MTT를 환원시킴으로써 생성된 포르마존(formazon)이 배지에 따라나가지 않도록 배지를 조심스럽게 제거하였다. 남아 있는 배지를 완전히 제거하기 위해 실온에서 30분간 방치한 후 DMSO를 이용하여 용해시킨 실시예 1을 570nm에서 흡광도를 측정하였다. 흡광도 측정시 공시료는 DMSO로 하였고, 세포의 생존율은 하기 수학식 1에 의해 계산되었다.
