Abstract

[배경 기술]
퍼옥시다제(E.C.1.11.1.7)는 과산화수소를 갖는 기질(전자 또는 수소 도너)의 산화를 촉매하는 효소이다. 그러한 효소는 미생물, 식물 및 동물 출처로 부터 알려져 있고, 에컨대 Coprinus cinereus 의 퍼옥시다제(가령 EP 특허출원 179,486참조)가 알려져 있다. 그것은 전형적으로 헤모단백질이다. 즉 그것은 보결분자단으로서 햄을 포함한다.

[퍼옥시다제 활성]
본 명세서에서 퍼옥시다제의 효소활성은 "퍼옥시다제 단위" (PODU)로 표현된다. 과산화수소의 존재하에서 퍼옥시다제(E.C.1.11.1.7)는 2,2'-아지노-비스(3-에틸벤조티아졸린-6-술포네이트)(ABTS)의 탈수소화를 촉매한다. 생성되는 녹색을 띤 청색을 418nm에서 광도계로 측정한다. 1PODU는 표준조건(즉, pH 7.0; 기질로서 과산화수소; 0.1M 인산염 완충액; 배양온도 30℃; 배양시간 3분에서 속도론적으로 측정함)하에서 분당 과산화수소 1μmol의 전환을 촉매하는 효소의 양이다.
[프로테아제 활성]

[서브틸리신]

[프로테아제의 안정화 방법]

그것은 PCR 반응에서 프라이머중 하나로서 화학적으로 합성된 DNA 올리고뉴클레오티드를 이용함으로써 원하는 돌연변이를 포함하는 PCR 단편의 3-단계 생성을 포함한다. PCR 합성된 단편으로부터 돌연변이를 갖는 DNA 단편은 제한 효소로의 절단에 의해 분리되고 발현 플라스미드 속에 재삽입될 수 있다. 세번째 돌연변이 유발 방법은 DNA 코딩 영역에서 제한 위치를 이용한다. 돌연변이 유발 표적 측면위치한 부위에서 DNA 를 제한효소로 분해시키고, 원하는 돌연변이를 포함하는 새로운 단편을 합성하고, 이 새로운 단편을 제한 부위 사이에서 클로닝함으로써, 돌연변이 코딩영역을 제조할 수 있다.
일반적으로 새로운 또는 변화된 특성을 가는 폴리펩티드의 개발을 다루는 단백질 공학이라고 부르는 분야에서의 탐구에 모든 방법이 적용될 수 있다.
형질전환 및 발현은 당업계에서 알려진 방법, 가령 명세서가 여기서 참고문헌으로 포함된 유럽특허출원 305,216에서 기술된 방법에 의하여 수행될 수 있다.
본 발명의 안정화된 효소를 생성할 수 있는 미생물은 동화할 수 있는 탄소 및 질소와 다른 필수 영양소를 포함하는 영양 배지에서 종래의 발효 방법에 의해 배양할 수 있으며, 배지는 당업계의 알려진 원리에 따라 구성된다. 또한 안정화된 효소의 정제 및 회수는 당업계에서 알려진 방법에 따라 수행할 수 있다.
[프로테아제 유전자의 클로닝]

[프로테아제 유전자에서 부위 특이적 돌연변이의 생성]

그 다음엔 유전자의 단일가닥 영역이 돌연변이된 올리고뉴클레오티드와의 혼성화에 이용될 수 있고, 남아 있는 틈은 DNA 폴리머라제 I 의 클레노우 단편에 의해 채워지고, 삽입체는 T4 DNA 리가제로 결합되고, 1 사이클의 복제후에는 원하는 돌연변이를 갖는 이중가닥 플라스미드가 생성된다. 모리나가 방법은 새로운 제한 위치를 제조하는 추가 조작을 제거하고, 따라서 다수의 위치에서 돌연변이 생성을 촉진한다. 미국특허 4,760,025는 카세트의 약간의 변형에 의한 다수의 돌연변이를 갖는 올리고뉴클레오티드의 도입을 개시한다. 그러나, 모리나가 방법에 의해서 한번에 훨씬 더 다양한 돌연변이가 도입될 수 있는데, 그것은 각종 길이의 무수한 올리고뉴클레오티드가 도입될 수 있기 때문이다.
[프로테아제 변이체의 발현]
본 발명에 따르면 상술한 방법, 또는 당업계에서 알려진 어떤 다른 방법에 의해 생성된 돌연변이된 프로테아제 유전자는 발현벡터를 이용하여 효소형태로 발현시킬 수 있다.

한 구체예에 따르면, 돌연변이된 DNA 를 갖는 발현벡터에 의하여, B.subtilis 를 형질절환 시킨다. 발현이 B. subtilis 같은 분비미생물에서 일어나게 되면, 시그날 서열이 번역개시 시그날 뒤 그리고 관심있는 DNA 서열 앞에 있을 수 있다.
시그날 서열은 발현산물을 세포벽으로 운반하는 작용을 하고 거기서 분비시에 산물로 부터 그것이 절단된다.

[뉴클레오티드서열, 발현벡터 및 미생물]
본 발명은 또한 본 발명의 안정화된 프로테아제 변이체를 암호화하는 DNA 뉴클레오티드 서열에 관한 것이다. 안정화된 효소 변이체는 이 효소를 암호화 하는 DNA 뉴클레오티드 서열을 적당한 숙주 생물체에서 적당한 벡터속에 삽입시킬 때 발현되고 생성될 수 있다. 숙주 생물체는 모 유전자 출처인 생물체와 반드시 동일하지는 않다.

[세정제 조성물]

유체베드 기술에 의한 적용에 적합한 필름-형성 코팅물질의 실례는 영국특허 1483591에서 주어진다. 액체효소제제는 예컨데 확립된 방법에 따라 프로필렌글리콜 같은 폴리올, 당 또는 당 알코올, 락트산 또는 붕산을 첨가함으로써 안정화 될 수 있다.
다른 효소 안정화제도 당업계에서 알려져 있다. 보호된 효소는 EP 238,216에서 개시된 방법 따라 제제화 될 수 있다.
본 발명의 세정제 조성물은 어떤 편리한 형태, 예컨대 분말, 과립, 페이스트 또는 액체 일 수 있다. 액체 세정제는 수성일 수 있는데 전형적으로 70%까지의 물과 0-30%의 유기용매를 포함하고, 또는 비수성일 수 있다.

세정제는 하나 이상의 중합체를 포함할 수 있다. 실례는 카르복시메틸셀룰로스(CMC), 폴리(비닐피롤리돈)(PVP), 폴리에틸렌글리콜(PEG), 폴리(비닐알코올) (PVA), 폴리카르복실레이트로서 폴리아크릴레이트등, 말레/아크릴 산 공중합체 및 라우릴 메타크릴레이트/아크릴산 공중합체이다.

세정제는 또한 음이온성 계면활성제 및/또는 표백시스템으로 이루어질수도 있다.
본 발명의 프로테아제 변이체는 세정제에 종래에 사용된 농도들로 포함된다.
본 발명의 세정제 조성물에서, 프로테아제 변이체는 세척액 리더당 0.001-100mg 의 프로테아제 변이체에 해당하는 양으로 첨가될 수도 있다.
다음 실시예들은 본 발명을 더 예시하며 특허청구된 본 발명의 범위를 어떤식으로든 제한하려는 것은 아니다.
[실시예 1]
[퍼옥시다제 시스템]
Corprinus cinereus 퍼옥시다제(CiP, EP 특허출원 179,486에 따라 얻음), 과산화수소 (H2O2) 및 퍼옥시다제 강화제로서 p-히드록시벤젠술포네이트(pHBS)로 이루어지는 염료 전이억제(Dye Transfer Inhibition: DTI)를 위해 사용되는 퍼옥시다제(POD) 시스템을 인산나트륨 완충액에서 실험하였다: [pHBS]: 50μM, [H2O2]: 200μM, [CiP]: 2PODU/㎖, 20mM 인산나트륨, pH 8.5.
서브틸리신 309 (Savinase,Novo Nordisk A/S 덴마크제)의 활성을 퍼옥시다제 시스템과 35℃에서 20분간 배양후 조사하였다.
서브틸리신 309의 잔류활성을 상이한 바와 같이 기질 N-숙시닐-Ala-Ala-Pro-Phe-p-니트로아닐리드를 사용하여 측정하였다. 프로테아제의 안정성을 각각 40nM 과 400mM의 농도를 사용하여 분석적으로 평가하였다.
두 농도들에 대해 서브틸리신 309의 잔류활성은 미처리된 서브틸리신 309에 관하여 15%로 측정하였다.
따라서 서브틸리신은 POD 시스템의 존재하여 분명히 불활성화되었다.
[실시예 2]
서브틸리신 309 를 실시예 1에 기술된 것과 같이 [14c]-pHBS 의 존재하에 POD 시스템과 배양하였다. POD 처리후 서브틸리신 309를 정제하고 변성조건들에서 아미노산분석과 겔여과 크로마토그라피에 의해 더 조사를 하였다.
[POD 처리된 서브틸리신 309의 정제]
POD 시스템과 400nM 농도로 서브틸리신 309를 함유하는 인산나트륨 완충액 2리터를 35℃에서 20분간 배양하였다. 불활성화후 YM10 멤브레인(Amicon)을 사용하여 Amicon 셀에서 3㎖로 농축하였다. 겔여과 컬럼; 0.1M 아세트산암모늄으로 용리하는 Superdex 75(16/60)에서 더 정제를 수행하였다. POD 처리된 서브틸리신 309 의 대부분이 미처리된 서브틸리신 309보다 약간 더 높은 분자량을 용리되었다.
POD 처리된 서브틸리신 309 를 농축하고 Amicon 셀에서 Milli Q 물 10부피로 세척하였다.
[아미노산 분석]
Applied Biosystems 420A 아미노산 분석시스템에서 제조업자의 지시에 따라 아미노산분석을 수행하였다.
아미노산 분석으로 부터의 결과를 이하 표 1 에 제공한다. 표에서 알수 있는 바와 같이 POD 불활성화에 의해 상당히 영향을 받는 단지 검출가능한 아미노산은 티로신(Tyr)이다.
[14C]-pHBS 의 존재하에 서브틸리신 309의 POD 불화성화는 효소의 방사성 표지를 가져온다. 이 변형의 공유성질은 2M 요소에서 변성조건하에 겔여과를 통해 나타났다.

[실시예 3]
퍼옥시다제 시스템에 대한 서브틸리신 309의 안정화를 한개이상의 티로신이 다른 아미노산 잔기에 의해 치환된 몇개의 변이체를 사용하여 조사하였다.
퍼옥시다제 시스템을 퍼옥시다제 강화제로서 7-히드록시쿠마린(7HCm)을 사용하여 Britton-Robinson 완충액에서 실험하였다:
[7Hcm]: 10μM, [H2O2]: 200μM, [CiP]: 2PODU/㎖, 20mM Britton-Robinson 완충액 (20mM 아세트산나트륨, 20mM 붕산나트륨 및 20mM 인산나트륨), pH 8.5.
다른 변이체들을 80 내지 120nM 의 다양한 농도들에서 퍼옥시다제 시스템과 배양하였다. 35℃에서 10분간 배양후 변이체의 잔류활성을 상기한 바와 같이 기질 N-숙시닐-Ala-Ala-Pro-Phe-p-니트로아닐리드를 사용하여 분석하였다.
잔류활성을 미처리된 변이체에 관하여 측정하였다.
결과를 이하 표 2에 제공한다. 표로부터 알 수 있는 바와 같이, 167, 171, 및 192 위치들에서 치환을 함유하는 두 변이체가 강화제로서 7HCm 을 사용하여 퍼옥시다제 시스템에 대해 상당히 안정화되어 있다.

[실시예 4]
상기한 몇개의 서브틸리신은 위치 104에서 티로신 잔기를 함유한다.
이 위치에서 티로신의 중요성을 서브틸리신 309변이체 V104Y 를 사용하여 조사하였다.
퍼옥시다제 시스템을 퍼옥시다제 강화제로서 7-히드록시쿠마린(7HCm)을 사용하여 Britton-Robinson 완충액에서 실험하였다:
[7Hcm]: 5μM, [H2O2]: 200μM, [CiP]: 2 PODU/㎖, 20mM Britton-Robinson 완충액 (20mM 아세트산 나트륨, 20mM 붕산나트륨 및 20mM 인산나트륨), pH 8.5.
프로테아제를 80nM의 농도로 퍼옥시다제 시스템과 배양하였다. 35℃에서 1,5 및 10분간 배양후, 프로테아제의 잔류활성을 상기한 바와 같이 N-숙시닐-Ala-Ala-Pro-Phe-p-니트로아닐리드 기질을 사용하여 분석하였다.
잔류활성은 미처리된 변이체에 관하여 측정하였다.
결과를 제1도에 제공한다. 위치 104에서 추가의 티로신의 도입후 서브틸리신 309변이체는 POD 시스템에 대해 훨씬 더 민감하게 되었다.
Claims (11)
- 퍼옥시다제 또는 퍼옥시다제 활성을 나타내는 화합물, 과산화수소원 및 퍼옥시다제 증강제를 포함하는 퍼옥시다제 시스템에 의해 야기되는 불활성화에 대한 서브틸리신 프로테아제 변이체의 안정화 방법으로서, 6, 57, 91, 104, 143, 167, 171, 192, 206, 209, 214, 256, 262, 263 (BPN' 넘버링) 위치의 하나 이상의 티로신 잔기가 결실되거나 페닐알라닌 잔기, 류신 잔기, 이소류신 잔기, 발린 잔기, 글루타민 잔기, 아스파라긴 잔기, 세린 잔기, 트레오닌 잔기, 글루탐산 잔기, 또는 히스티딘 잔기로 치환된 것을 특징으로 하는 서브틸리신 프로테아제 변이체의 안정화 방법.
- 제1항에 있어서, 프로테아제 변이체는 Bacillus lentus 프로테아제 변이체, 바람직하게는 서브틸리신 309변이체 또는 서브틸리신 147 변이체인 것을 특징으로 하는 방법.
- 제1항에 있어서, 프로테아제 변이체는 Bacillus amyloliquefaciens 프로테아제 변이체, 바람직하게는 서브티리신 BPN' 변이체인 것을 특징으로 하는 방법.
- 제1항에 있어서, 프로테아제 변이체는 Bacillus licheniformis 프로테아제 변이체, 바람직하게는 서브틸리신 Carlsberg 변이체인 것을 특징으로 하는 방법.
- 퍼옥시다제 또는 퍼옥시다제 활성을 나타내는 화합물, 과산화수소원 및 퍼옥시다제 증강제를 포함하는 퍼옥시다제 시스템에 의해 야기되는 불활성화에 대한 서브틸리신 프로테아제 변이체의 안정화 방법으로서, 6, 57, 91, 104, 143, 167, 171, 192, 206, 209, 214, 256, 262, 263 (BPN' 넘버링) 위치의 하나 이상의 티로신 잔기가 결실되거나 페닐알라닌 잔기, 류신 잔기, 이소류신 잔기, 발린 잔기, 글루타민 잔기, 아스파라긴 잔기, 세린 잔기, 트레오닌 잔기, 글루탐산 잔기, 또는 히스티딘 잔기로 치환되며: 다음 돌연변이들Y91F, Y167E, F, Y171F, V, Y192E, F, Y209F, L, Y214F, T, Y263F, (BPN' 넘버링)중 적어도 한 가지를 갖는 프로테아제 변이체와 적어도 하나의 아미노산이 다음과 같이, Y21X, Y104X, Y171X, Y214X, Y217X 치환되는 Bacillus amylololiquefaciens 서브틸리신 변이체는 제외하는 것을 특징으로 하는 서브틸리신 프로테아제 변이체.

- 제9항에 있어서, 세정제에 종래에 포함되는 리파제, 아밀라제, 셀룰라제, 옥시다제 및 퍼옥시다제에서 선택된 하나 이상의 다른 효소를 더욱 포함하는 것을 특징으로 하는 세정제 조성물.
