Abstract
영양가를 높이기 위해 식품 첨가용으로 특히 유용한 촉생 조성물이 개시된다. 그 조성물은 Bifidobacterium 같은 하나 이상의 촉생 미생물과 미생물을 대장 또는 위장관의 다른 영역으로 수송하기 위한 담체로 이루어진다. 담체는 변형 또는 비변형 내성 전분, 특히 대장 또는 위장관의 다른 영역에서 미생물용 생장 또는 유지 배지로서 작용하는 고아밀로스 전분이다.
Description
인간의 건강 및 행복이 위장관, 특히 대장에 서식하는 미생물에 의해 긍정적 또는 부정적으로 영향받을 수 있는지는 많은 과학자들의 쟁점이다. 이들 미생물은 독소, 대사 부산물, 단쇄 지방산 등의 생산을 통해 숙주의 생리상태에 영향을 준다. 장내 미생물상(microflora)의 구성 및 양은 질병, 생활스타일, 여행 및 다른 요인으로 인한 상태 또는 스트레스에 의해 영향받을 수 있다. 만약 개체의 건강 및 행복에 긍정적으로 영향을 주는 미생물이 대장에서 번식하도록 조장될 수 있다면, 이것은 숙주의 생리적 행복을 개선할 것이다.
유익한 미생물 또는 촉생제의 도입은 일반적으로 음료, 요구르트, 캡슐 및 미생물이 대장에 생존상태로 도달가능한 다른 형태를 통해 미생물을 섭취함으로써 성취된다.
대장에서의 내성(resistant) 전분의 박테리아 발효는 상승된 레벨의 단쇄 지방산, 특히 프로피오네이트 및 부티레이트 같은 유리한 유형을 생성한다는 것이 Englyst H.N. et al (1987)에 의해 ″Polysaccharides breakdown by mixed populations of human faecal bacterial″, EMS Microbiology Ecol 95: 163-71에서 증명되었다.
본 발명자는 촉생 미생물을 대장으로 전달하는 것뿐만 아니라 대장에 도착시 미생물의 생장을 촉진하는 기능을 할 수 있는 배지(medium)를 공급하는 것도 바람직할 것이라는 것을 깨달았다.
놀랍게도 변형 또는 비변형된 내성 전분이 촉생 미생물을 대장으로 수송하는 수단으로서 그리고 대장의 표적영역에 전달된 미생물을 위한 생장배지로서 둘다 기능할 수 있다는 것이 밝혀졌다. 본 명세서에서 사용된 ″내성 전분″은 RS1, RS2, RS3 및 RS4로 정의된 형태들을 포함한다.
본 발명은 영양 조성물, 구체적으로 위장관, 특히 대장내로 촉생(probiotic) 미생물을 전달 및 유지시키기 위한 조성물에 관한 것이다. 본 명세서에 사용된 촉생제 또는 촉생 미생물은 장내 미생물 균형을 개선함으로써 숙주동물을 이롭게 하는 생미생물성 사료보충물이다. 이 용어의 정의는 R. Fuller(AFRC Institute of Food Research, Reading Laboratory, UK)에 의해 Journal of Applied Bacteriology, 1989, 66, pp. 365-378. ″Probiotics in Man and Animals - A Review″에 제공된다.

(발명의 개시)
따라서 본 발명은 하나 이상의 촉생 미생물과 하나 이상의 촉생 미생물을 대장 또는 위장관의 다른 영역에 수송하는 기능을 하는 담체로 이루어진 촉생 조성물에 있으며, 여기서 담체는 변형 또는 비변형된 내성 전분 또는 그것의 혼합물로 이루어지고 대장 또는 위장관의 다른 영역에서 미생물을 위한 생장 또는 유지배지로서 작용한다.
다른 태양에서 본 발명은 또한 하나 이상의 촉생 미생물을 포함하는 제1부와 담체를 포함하는 제2부로 이루어진 2부 조성물에 있으며, 여기서 담체는 변형 또는 비변형 내성 전분 또는 그것의 혼합물로 이루어지고 대장 또는 위장관의 다른 영역에서 미생물을 위한 생장 또는 유지배지로서 작용한다.
한 넓은 태양에서, 내성 전분은 촉생 미생물을 대장에 수송하는 담체로서 기능한다. 이들 미생물의 대장내로의 도입은 상기된 바와 같이 유익하다. 더욱이 내성 전분은 대장에 존재시 대장에 이미 존재하는 미생물을 위한 영양원으로서 기능할 것이다.
더 좁은 태양에서, 약간의 촉생 미생물은 그들이 내성 전분을 영양원으로 이용할 수 있도록 선택될 수 있다. 따라서 내성 전분은 담체와 촉생 미생물용 영양원으로서 둘다 기능할 것이다.
본 발명에 사용하기 적합한 다양한 촉생 미생물은Saccharomyces같은 효모와Bifidobacterium,Bacteroides,Clostridium,Fusobacterium,Propionibacterium,Streptococcus,Enteroccus,Lactococcus,Staphylococcus,Peptostreptococcus및Lactobacillus속같은 박테리아를 포함한다. 그러나 본 발명은 이들 특정 미생물에 한정되지 않는다. 당업계의 기술자는 본 발명의 조성물에 포함될 수 있는 미생물을 이해하고 인식할 수 있을 것이다.

촉생 미생물은 농축물, 냉동 농축물 및 동결-건조물질을 포함하는 다양한 형태로 본 발명에 사용될 수 있다.
한 바람직한 형태에서, 촉생 미생물은 내성 전분과의 현탁물로서 동결-건조될 수 있다. 이 유형의 조성물에 대해 내성 전분은 동결방지제로서 작용할 것이다.
전형적으로 내성 전분은 2-20%w/w 레벨로 통합될 수 있다. 첨가된 내성 전분의 양에 의존하여 건조율 및 최종생성물의 밀도 및 재수화 특성이 영향받을 것이다. 총 건조고체의 약 5% 레벨이 전형적으로 사용될 것이지만, 더 높은 농도도 건괴성질을 개선하기 위해 사용될 수 있다.
다른 형태에서, 촉생 미생물은 과립생성물을 형성하는 사출공정에서 내성 전분과의 동결-건조물질로서 사용될 수 있다. 이 형태의 조성물을 제조하기 위하여는 사출온도가 미생물의 생존능에 악영향을 줄만큼 높지 않도록 보장하는 것이 중요하다. 이것을 성취하는 한 방법은 내성 전분을 고융점 지방과 함께 재료를 용융형태로 유지하기에 충분히 높은 온도로 유지된 존(zone)을 갖는 사출기내로 공급하는 것이다. 이어서 사출기의 냉각존에 촉생 미생물과 저융점 지방의 현탁물을 공급한다. 이것은 지방 및 내성 전분 배합물과의 효과적인 혼합을 초래한다. 그 다음 적당히 냉각된 다이(die)를 통해 사출이 일어난다. 용융 지방은 50℃ 이상의 융점을 가지나 약 35℃ 내지 40℃ 영역에서 높은 가소성을 가지는 야자유와 같은 경화 식물성 오일 또는 경화 및 비경화 식물성 오일의 배합물일 수 있다. 바람직하게는 오일(들)은 안정하고 비산화성이어야 한다.
사출기내로의 저융점 지방 또는 오일의 배합율은 0 내지 10%의 범위에 있을 수 있다. 식품기질을 통한 개선된 분포도와 같은 특별한 특성이 필요한 경우 더 높은 양이 사용될 수 있다.
배합물을 안정화하기 위해 다른 성분들을 첨가할 수 있다.
내성 전분 대 촉생 미생물의 혼합물은 어떤 비율로도, 바람직하게는 적어도 1:1:1 (내성전분:동결건조형태의 박테리아:오일/지방)일 수 있다. 내성전분:오일/지방의 비율을 바꿈으로써 배합물의 유동성이 개선될 수 있다.
다른 형태에서, 촉생 미생물은 내성 전분으로 미소캡슐화될 수 있다. 미소 캡슐화의 전형적 공정은 저융점 오일을 그것의 융점 이상에서 전형적으로 50℃ 미만, 바람직하게는 40℃ 미만으로 예열시키는 것을 필요로 한다. 저전단의 조건하에 10%같은 냉동건조 촉생 미생물의 양이 첨가되고 분산된다. 오일의 예컨대 20%의 양으로 내성전분이 다른 성분과 겔라틴 및 검같은 결합제와 함께 첨가된다.
그 다음 분산액이 냉각탑의 두부내로 분사되어 20-200미크론(용도에 따라) 범위내의 전형적인 평균크기를 갖는 균일입자가 형성되고 경화될 수 있도록 해준다.
최종 미소캡슐화 산물의 생존세포 카운트는 그램당 차수 108-1012미생물일 수 있다. 상기 형태 외에, 촉생 미생물은 배합, 분사냉각, 포획 및 부착을 포함하는 다른 형태로 제공될 수도 있다.
일반적으로 촉생 미생물은 내성 전분과 함께 g 내성 전분당 약 102cfu 이상, 바람직하게는 약 105이상, 가장 바람직하게는 107이상의 비율로 포함될 것이다. 최대치로서 일반적으로 g 내성 전분당 약 1012cfu 이하가 사용될 것이다.
변형 또는 비변형 내성 전분 또는 그것의 혼합물이 본 발명에 사용된다. 촉생 조성물에서 내성 전분의 이점은 그것이 대장에 도달할 때까지 소화되지 않는다는 것이다. 그러므로 그것은 그것들이 대장에 도착하는 즉시 촉생 미생물에 의한 발효용의 쉽게 구할 수 있는 기질을 제공한다. 둘다의 경우에 내성 전분의 바람직한 형태는 고아밀로스 전분, 특히 본 명세서에 참고문헌으로 포함된 WO 94/ 03049 및 WO 94/14342에 개시 및 교시된 고아밀로스 전분이다.
WO 94/03049 및 WO 94/14342에서, 내성 전분인 고아밀로스 전분이 개시되며, 그것은 50%w/w 이상, 특히 80%w/w 이상의 아밀로스 함량을 갖는 옥수수 전분; 27% w/w 이상의 아밀로스 함량을 갖는 쌀 전분; 그리고 50% 이상의 아밀로서 함량과 증가된 내성 전분 함량을 갖는 특정 과립크기 범위의 전분을 포함하며, 이들 전분은 옥수수, 보리, 밀 및 콩을 포함한다. 그러나 본 발명은 이들 형태의 내성 전분에 한정되지 않는다. 예컨대 다른 형태의 내성 전분은 바나나 및 감자와 같은 공급원에서 유래된다.
본 발명의 조성물은 촉생 미생물 및 담체가 조합되어 제시되도록 제조할 수 있다. 이와 선택적으로 촉생 미생물 및 담체는 두 개의 별도부분의 각각에 제시될 수도 있다. 이러한 형태에서, 촉생 미생물을 함유하는 부분 또는 담체를 함유하는 부분중 하나가 우선 소비되고, 그 직후 다른 부분이 소비될 수 있다.
비변형 형태에서, 내성 전분은 조성물이 대장에 도착할 때 촉생 미생물에 의한 발효용 기질로서 작용할 것이다.
또한 예컨대 과립 및/또는 과립표면의 전하밀도 또는 소수성을 변화시켜 미생물과 내성 전분 사이의 접촉융화성을 향상시키기 위해 전분을 화학적으로 변형시키는 것이 유리할 수 있다. 에테르화, 에스테르화, 산성화 등과 같은 화학적 변형은 적당한 화학처리로서 당업계에 주지되어 있다.
전분의 구성 또는 구조를 바꿈으로써 내성 전분의 효소 감수성의 정도를 변형시키는 것도 또한 유용할 수 있다. 그 예는 산 또는 효소 박형화와 2작용성 시약을 사용한 가교 결합을 포함한다.
한가지 유용한 변형은 과립의 표면에서 내부로 확장가능한 구멍 또는 침식을 특징으로 하는 전분 과립을 만드는 고아밀로스 전분의 전분분해(amylolysis)이다. 이들 구멍은 효소가 가용화된 전분과립의 더욱 효소감수성 코어내로 진입할 수 있도록 해준다.
침식된 전분 과립은 미생물 부착용 표면적을 증가시킬 뿐만 아니라, 또한 미생물의 과립내부로의 캡슐화 및 포획화를 허용한다. 후자의 성질은 미생물의 보호를 보조하여 대장으로의 수송을 용이하게 한다.
효소 처리된 내성 전분을 하기와 같이 화학적으로 변형시키는 것도 또한 본 발명의 범위내에 있다.
내성 전분에 행할 수 있는 상기 화학적, 효소적 및 효소/화학적 변형 이외에, 촉생 미생물은 2작용성 시약 또는 다작용성 시약과 같은 물질을 사용하여 내성 전분에 화학적으로 부착시킬 수 있다. 이들 유형의 시약의 예는 각각 글루테르알데히드 및 트리메타인산나트륨이다.

유사하게 본 발명의 조성물은 캡슐같은 투여형태로 제시될 수도 있다.
일반적으로 촉생 미생물과 조합된 내성 전분은 건조형태로 제시될 것이다. 적합한 그러나 제한적이지 않은 건조수단은 분사건조, 동결건조 및 분출상 건조를 포함한다.
(발명의 실시형태)
본 발명의 내성 전분이 취할 한 형태는 그것이 침식되거나 구멍이 파일 때이다. 그러한 전분의 한 제조예는 도 4 내지 도 7을 참조하여 지금 기술될 것이다.
고아밀로스 옥수수 전분 과립으로부터 전분분해산물의 제조
2000그램의 고아밀로스 옥수수(Starch Australasia Ltd 제 Hi-옥수수-상표) 전분(12.5% 수분) 및 물(7000mL)을 함유하는 슬러리를 수산화나트롬 용액(0.65M)을 사용하여 pH 6.0으로 조정하였다. 슬러리를 수욕에서 85℃로 가열하였다. 실험 2, 5 및 6의 이 시점에서 열안정성 세균 알파 아밀라제 Termamyl을 첨가하였다. 실험내용은 표 2에 상세히 제공된다.
60분후에 슬러리의 온도는 60℃로 감소되었고, (i) 균류 글루코아밀라제 및 세균 풀룰라나제 배합물 Dextrozyme의 첨가전에 pH를 염산(10M)을 사용하여 4.5로 조정하거나 (ii) 세균 플룰라나제 Promozyme의 첨가전에 pH를 5.0으로 조정하였다(표 2). 양 경우의 슬러리는 20분동안 60℃로 유지되었다.
그 다음 수산화나트륨 용액(0.65M)을 사용하여 pH를 pH 6.0으로 상승시키고, Whatman 54 여과지가 끼워진 Buchner 깔대기를 사용하여 불용성 부분을 회수하였다. 회수물질을 7리터의 증류수로 세척한 다음, 50℃의 팬동력 오븐에서 건조시켰다.

각 경우에 효소처리가 식이성 섬유레벨은 본질적으로 유지하면서 내성 전분 함량의 실질적 향상을 초래한다는 것이 명백하다.
그러한 효소 처리의 결과는 첨부된 도 5, 6 및 7에서, 특히 도 4의 비처리된 전분 과립과 비교시 명확히 나타난다.
이 방법으로 제조된 변형 전분은 촉생 미생물과 쉽게 조합되어 본 발명의 조성물을 형성할 수 있다.

방법
내성 전분을 50℃ 초과 융점의 용융 지방과 결합하여 즉 20kg/시간의 공급속도로 APV MPF 50.20 사출기에 공급한다. 뜨거운 물을 재순환시키고 2kg/시간의 속도로 제2 주입구에 사출기내로 전달함으로써 지방을 액체로 유지한다. 동결건조된 세균, 특히 Bifidobacterium bifidum 또는 Bif.Longum의 현탁액은 저융점 지방, 예컨대 35-37℃ 융점의 경화 옥수수유를 사용하여 제조하여서, 지방을 너무 높지 않은 융점 바로 위로 가열하고, 동결건조된 세균을 첨가하여 완전히 혼합한다. 그 다음 이 현탁액을 사출기의 존 7내로 공급하여 거기서 지방/전분 배합물과 혼합하고, 그 다음 2mm 직경구멍이 끼워진 냉각 다이를 통해 압출시킨다. 그 물질을 과립화하고 사용전까지 그 모양을 유지하기 위해 더욱 냉각시킨다.

방법
1. 안정화제가 잘 분산되도록 하면서 배양물을 제외한 모든 성분들을 조합 한다.
2. 혼합물을 88-93℃로 가열하고, 이 온도에서 30분간 방치한다.
3. 38-45℃로 냉각한다.

주의
1. 겔라틴, 겔라틴 및 검 배합물 또는 검 배합물로 구성된 다양한 이용가능한 안정화제가 있다. 추전할만한 안정화제는 a) Leiner Davis Gelatin(Australia) Co.제 겔라틴과, b) Germantown Australia Co.제 Sta-Rite 및 Stabilizer Gemcol ADQ이다.
2. 안정화제 및 MAPS 449 조합은 최종 믹스의 1% w/w이어야 한다. 더 높은 농도는 몇몇 환경하에 악영향을 줄 수 있다.
3.교반요구르트의 경우, 용기의 펌핑 및 충전은 단계 6 및 7 사이에 행한다.

(실험 1)

약간의 균주, 특히 Bifidobacterium, Bacteroides, Eubacterium 및 Clostridium 속으로부터 균주는 과립전분을 분해하는 능력을 나타냈다.
이들 시험관내 발효의 산물은 내장 및 신체의 유지 및 보호에 중요한 아세테이트, 프로피오네이트 및 부티레이트와 같은 단쇄 지방산(VFA)을 포함하였다.
각각의 세균균주가 전분 또는 화학유도체화 전분이 발효기질로서 작용할 수 있다는 것을 증명하는 개별 생장 프로필을 나타낸다는 것이 도 9에서 알 수 있다.
Bifidobacterium 균주 X8에 의해 보여진 것과 같은 약간의 경우에, 히드록시프로필화 고아밀로스 옥수수 전분을 이용하는 능력은 Bifidobacterium 균주 X8 집단이 10시간후 약 4.2log/㎖에서 6.2log/㎖로 증가할 때 배양개시후 약 8시간이 될 때까지 Bifidobacterium 균주 X8 집단의 개체군 증가에서 반영되지 않았다.
따라서 내성전분의 변형의 유형 또는 정도는 위장관에서 내성전분의 발효율 또는 발효부위를 제어하기 위한 수단으로 사용될 수 있다.
이것은 도 10에 보여진 생장능 그래프를 고려할 때 더욱 증명된다.
이것은 위장관의 한 영역에서 세균생장의 촉진을 선택적으로 표적화하는 변형 내성전분 유형을 선택할 수 있는 가능성을 허용한다.
(실험 2)

(실험 3)
내성 전분이 대장까지 통과하여 촉생 미생물의 생장을 촉진할 수 있다는 것을 증명하기 위해, 두가지 사료시험을 수행하였다.
첫 번째 시험은 어떤 천연 맹장 Bifidobacteria도 가지지 않는 것으로 알려진 마우스를 표 6에 보여진 바와 같이 Bifidobacterium 균주를 포함하는 다이어트로 공급하는 것을 포함하였다.

표 8에 보여지듯이 실험의 투여기간동안 마우스 배설물중 평균 생존 세균 카운트의 비교는 비변형된 고아밀로스 옥수수 전분 및 변형된(카르복시메틸화) 옥수수 전분의 형태로 내성 전분을 소비한 마우스에 의해 배설된 Bifidobacteria 개수의 상당한 증가를 보여주었다.
고아밀로스 옥수수 전분과 비교하여 덜 카르복시메틸화된 옥수수 전분이 배설물내 존재한다고 밝혀졌다는 것을 또한 알 수 있다. 이는 표 4와 도 9 및 10에 보여진 결과와 관련하여 언급되듯이 변형 대 비변형 내성 전분의 시험관내 차별 이용도를 더욱 증명한다.

내성 전분이 다이어트에 포함될 때 분내 단쇄 지방산(아세테이트, 프로피오네이트 및 부티레이트)의 레벨이 증가하였으나, 촉생제가 첨가될 때에는 더 증가하지 않았다. 그러나 배설물상에서 수행된 전분 분석은 큰 대조 다이어트(소화용이한 밀랍성 옥수수 전분을 함유)에서 보다 돼지에게 공급된 내성 전분에 더 큰 퍼센트의 전분이 존재한다는 것을 밝혔으나, 더 중요하게는 내성 전분이 Bif. logum과 결합되어 공급될 때 존재 전분의 퍼센트의 상당한 감소가 관찰되어서 Bif. longum이 아마도 내성 전분을 기질로서 사용하였다는 것을 가리켰다.
최종 회전의 끝에 실험 다이어트를 유지시켰다. 촉생제 보충을 중지하였다. 4일간에 걸쳐 배설물을 모니터하였을 때, Bif. longum의 레벨은 처음 2일간은 급격한 감소를 경험하였고 그 다음 평형을 이룬 것으로 나타났다. 이것은 다이어트가 장내에서 Bifidobacteria의 콜론화를 촉진할 수 있다는 것으르 제시한다. 내성 전분을 함유하지 않은 다른 보고된 실험들은 Bifidobacteria의 레벨이 실험 다이어트를 유지한 후에도 2일내 완전히 떨어진다는 것을 밝혔다.

- 하나 이상의 촉생미생물과 하나 이상의 촉생 미생물을 대장 또는 위장관의 다른 영역에 수송하는 기능을 하는 담체로 이루어진 촉생 조성물에 있어서, 그 담체는 변형 또는 비변형 내성 전분 또는 그것의 혼합물로 이루어지고 대장 또는 위장관의 다른 영역에서 미생물용 생장 또는 유지 배지로서 작용하는 것을 특징으로 하는 촉생 조성물.
