Description
Claims (1)
- 【特許請求の範囲】 1.洗剤において利用するのに適当な酵素又は酵素変異体を選別する方法であ って、ここで選別すべき前記酵素変異体は細胞又はファージ粒子の表層上でそれ ぞれがディスプレーされる酵素変異体の混合物の中にあるものであり、そして i)前記混合物を流動状態の洗剤組成物の中に、前記酵素変異体の活性に対して ネガティブな影響を与える又はそのほとんどを失活させるであろう条件下で導入 する、 ii)前記混合物を、要望する特性を発揮する酵素変異体にのみ特異的に結合する キャッチャー分子と反応させ、要望の特性を示す前記細胞又はファージ・ディス プレーされた酵素と前記キャッチャーとの複合体を形成する、 iii)前記複合体を前記混合物の残部から分離する、 iv)前記複合体を解離して、キャッチャー分子のない選別すべき前記酵素変異体 をディスプレーする細胞又はファージを単離する、 v)前記ファージをそれが中で増幅するであろう宿主の中に導入する、又は va)前記細胞をその増幅を誘導する条件下で培養する、そして vi)前記細胞又はファージのゲノムから前記酵素変異体をコードするDNA 分子を 単離する、 工程を含んで成る方法。 2.前記洗剤が漂白系を含んで成る、請求項1記載の方法。 3.工程(ii)における前記条件が高温又は低温での処理を含んで成る、請求 項1又は2記載の方法。 4.工程(iii)における前記分離を表層に対する前記複合体の結合により実 施する、請求項1〜3のいづれか1項記載の方法。 5.前記結合が吸着である、請求項4記載の方法。 6.前記結合が共有的である、請求項4記載の方法。 7.前記インヒビターが前記表層に結合できるように修飾されている、請求項 4記載の方法。 8.前記表面が前記複合体の結合のために活性化されている、請求項4〜7記 載の方法。 9.前記表面がストレプトアビジンにより処理されており、そして前記インヒ ビターがビオチニル化されている、請求項8記載の方法。 10.工程(iv)における前記消化をプロテアーゼ、例えば第IX因子で実施する 、請求項1〜9のいづれか1項記載の方法。 11.工程(v)の前記宿主が細菌、例えばE.コリ、特に株DH12S である、請 求項1〜10のいづれか1項記載の方法。 12.前記酵素がプロテアーゼ、例えばスブチリシン、好ましくは高アルカリ性 スブチリシンである、請求項1〜11のいづれか1項記載の方法。 13.工程(ii)における前記キャッチャーが自殺インヒビター及び遷移状態類 似体を含んで成る群から選ばれる、請求項12記載の方法。 14.前記酵素がリパーゼである、請求項1〜11のいづれか1項記載の方法。 15.工程(ii)における前記キャッチャーが自殺インヒビター及び遷移状態類 似体を含んで成る群から選ばれる、請求項14記載の方法。 16.前記酵素がセルラーゼである、請求項1〜11のいづれか1項記載の方法 。 17.工程(ii)における前記キャッチャーが自殺インヒビター及 び遷移状態類似体を含んで成る群から選ばれる、請求項16記載の方法。 18.前記酵素がオキシドリダクターゼである、請求項1〜11のいづれか1項記 載の方法。 19.工程(ii)における前記キャッチャーが自殺インヒビター及び遷移状態類 似体を含んで成る群から選ばれる、請求項18記載の方法。 20.前記酵素がアミラーゼである、請求項1〜11のいづれか1項記載の方法。 21.工程(ii)における前記キャッチャーが自殺インヒビター及び遷移状態類 似体を含んで成る群から選ばれる、請求項20記載の方法。 22.前記酵素がリアーゼ、キシラナーゼ、ポリガラクツロナーゼ、オキシダー ゼ、ラッカーゼ、トランスグルタミナーゼ、ガラクトシダーゼ、フィターゼ又は ペルオキシダーゼである、請求項1〜11のいづれか1項記載の方法。 23.工程(ii)における前記キャッチャーが自殺インヒビター及び遷移状態類 似体を含んで成る群から選ばれる、請求項22記載の方法。 24.先の請求項のいづれか1項記載の方法の利用により選別された酵素。 25.請求項24記載の酵素を含んで成る洗浄組成物。
