- 【特許請求の範囲】 1.ヒトの結腸がん腫に結合されるタンパク質抗原に特異的なモノクローナ ル抗体において、前記抗原が下記の特徴: (i) 前記モノクローナル抗体は、前記抗原のタンパク質成分のエピトープを 認識し、且つ前記抗原の炭水化物成分のエピトープを認識しない; (ii) 前記抗原は、正常なヒトの組織で検出できない; (iii)前記抗原は、結腸がん腫細胞以外のヒトのがん腫細胞で検出できない; (iv) 前記抗原は、ヒトに特異的免疫原性がある、 を有することを特徴とする抗体。 2.マウスのモノクローナル抗体 33.28であるか又は33.28によって結合さ れたそれと同一の結腸がん腫結合エピトープに特異的に結合する抗体である請求 項1記載の抗体。 3.前記結腸がん腫結合抗原が約61.1キロダルトンの分子量を有するタンパ ク質である請求項2記載の抗体。 4.マウスのモノクローナル抗体 31.1であるか又はマウスのモノクローナ ル抗体 31.1によって結合されたそれと同一の結腸がん腫結合エピトープに特異 的に結合する抗体である請求項1記載の抗体。 5.前記結腸がん腫結合抗原が約72キロダルトンの分子量を有するタンパク 質であり、且つ前記結腸がん腫結合抗原が糖タンパク質であり、そのタンパク質 成分が約61.1キロダルトンの分子量を有することを特徴とする請求項4記載の抗 体。 6.固相で免疫化される請求項1記載の抗体。 7.検出できるよう標識される請求項1記載の抗体。 8.前記の検出可能標識が放射性標識である請求項7記載の抗体。 9.細胞毒性のある放射性核種に抱合される請求項1記載の抗体。 10.細胞毒性のある薬物に抱合される請求項1記載の抗体。 11.細胞毒性のあるタンパク質に抱合される請求項1記載の抗体。 12.製薬上許容できるキャリヤーと組合せた請求項9に従う抗体を含む、ヒ トの結腸、胸部、及び卵巣がんの免疫治療用製薬組成。 13.製薬上許容できるキャリヤーと組合せた請求項10に従う治療上有効量の 抗体を含む、ヒトの結腸、胸部、及び卵巣がんの免疫治療用製薬組成。 14.製薬上許容できるキャリヤー中に請求項11に従う治療上有効量の抗体を 含む、ヒトの結腸、胸部、及び卵巣がんの免疫治療用製薬組成。 15.ヒトの結腸結合タンパク質抗原において、下記の特徴: (i) 前記抗原のタンパク質成分のエピトープは、哺乳動物の宿主に抗体応答 を誘発できるが、前記抗原の炭水化物成分のエピトープは、前記の抗体応答を誘 発できない; (b) 前記抗原を認識できるモノクローナル抗体にその試料を接触させるステ ップと; (c) その試料を免疫組織化学的染色剤で染色するステップと; (d) 染色により抗原ー抗体複合体の有無を検出するステップと を設けて成る方法。 111.ヒトの結腸がんを診断する方法において: (a) マウス/ヒトのキメラ抗体 Chi #1に結合できる抗原を帯びている結 腸がん腫を有すると思われる患者から組織学的試料を除去するステップと; (b) 前記抗原を認識できるマウスのモノクローナル抗体にその試料を接触さ せるステップと; (c) その試料を免疫組織化学的染色剤で染色するステップと; (d) 抗原ー抗体複合体の有無を検出するステップと を設けて成る方法。 112.結腸がん腫の免疫組織化学的検出に使用するキットにおいて: a) マウス/ヒトのキメラ抗体 Chi #1、及び胎児がん腫抗原用キメラ抗体 と; b) 免疫ペルオキシダーゼ用試薬及び二次的抗体と; c) 免疫ペルオキシダーゼと; d) 着色剤と を含んで成るキット。
