Endocrinilogy)、第6巻、1023頁( Vaccine Adjuvant Background of the Invention The use of vaccines to prevent diseases in humans, farm animals, sport animals and domestic pets is a common practice and is an extension of this practice. Considerable effort has been and is being made to cover the wider range of diseases in which these patients are targeted. For example, the use of rabbit vaccines in animals is now not uncommon, and efforts are being made to obtain suitable vaccines to immunize animals against other diseases. One problem often encountered in the process of active immunization is to provide protection against subsequent attacks or to raise antibody titers sufficiently to maintain the ability to hold these levels for extended periods of time. It means that the antigen used in the vaccine is not sufficiently immunogenic. Another problem is that vaccines may have drawbacks in including cell-mediated immunity, which is the primary immune defense against bacterial and viral infections. In order to obtain a stronger humoral and / or cellular response, it is customary to administer the vaccine as a formulation containing an adjuvant, a substance that enhances the patient's response to the vaccine. The most commonly used vaccine adjuvants are oily preparations and alum. The mechanism by which such adjuvants function is not understood, and it is not possible to predict whether a particular adjuvant preparation will be sufficiently effective in certain instances. Moreover, with the advent of gene therapy, it has been reported that some success has been achieved using genes or "naked DNA" as vaccines. However, for some conventional vaccines, the immune response obtained was insufficient to generate immunization. Therefore, there is a need for more effective adjuvant preparations, generally for animals, which are suitable for activating vaccines in humans in particular. SUMMARY OF THE INVENTION The present invention relates to a composition for enhancing the immune response of an animal to an infectious disease vaccine, which composition comprises prolactin. Preferably, the composition is human prolactin and the vaccinated animal is also human. The present invention further relates to a composition for enhancing the immune response of an animal to an infectious disease vaccine, which composition comprises prolactin cDNA. Human prolactin cDNA is preferred. In another aspect, the invention relates to a method of enhancing the immune response of a subject animal to an infectious disease vaccine, comprising co-administering an effective amount of prolactin or prolactin cDNA with the vaccine. Brief Description of the Drawings Figure 1 shows the amino acid sequence for the prolactin protein. FIG. 2 shows the nucleic acid sequence for the prolactin cDNA. FIG. 3 is a graph showing bovine serum albumin (BSA) -specific antibody responses in rats immunized with BSA alone or BSA + prolactin. FIG. 4 is a graph showing a comparison of BSA-specific proliferative responses of rat PBLs at day 101 between four rats receiving BSA alone against BSA + prolactin. DETAILED DESCRIPTION OF THE INVENTION Definitions "Prolactin" as used herein is a polypeptide obtained from tissue culture or by recombinant methods as well as other methods known to those of skill in the art, which characterizes this protein. Refers to a polypeptide exhibiting an active spectrum. The term includes not only human prolactin (hPRL) but also other mammalian prolactins such as, for example, mouse, rat, rabbit, primate, pig and bovine prolactin. The amino acid sequence of recombinant hPRL is shown in FIG. In the present invention, the recombinant PRL (r-PRL) is preferable. “Recombinant prolactin”, referred to as r-PRL, is preferably human prolactin and refers to prolactin having a biological activity comparable to the intact prolactin prepared by recombinant DNA methods known to those skilled in the art. Generally, the gene encoding prolactin is excised from the intact plasmid, inserted into the cloning vector to be cloned, and used to transform a host organism. The host organism expresses the foreign gene and produces prolactin under the expression conditions. The term "adjuvant" as used herein has its conventional meaning, ie, the ability to enhance the immune response to a particular antigen. Such ability is manifested by a significant increase in immune-mediated protection. Furthermore, the term "genetic adjuvant" refers to a prolactin cDNA consisting of a sequence complementary to the DNA sequence encoding the prolactin protein defined above. The sequence for the prolactin cDNA is shown in Figure 2. General Method Most commonly, a formulation containing prolactin for adjuvant purposes is administered by intramuscular or subcutaneous injection or intraperitoneal injection, although other methods of administration are possible. Standard formulations are either liquid and injectable, or solids which can be made into suitable injection liquid suspensions or solutions. Suitable excipients are, for example, water, saline, dextrose, glycerol, ethanol and the like. Non-toxic additives such as wetting agents, buffers or emulsifying agents may be added. Sustained and continuous release formulations are of great variety and can be used in the methods of the invention, as will be appreciated by those in the art. Prolactin can be administered separately from the vaccine or mixed with the vaccine. When prolactin is mixed with a vaccine, the composition administered will have an immunogen effective in inducing a specific response to a pathogen or antigen, and a pharmaceutically acceptable vaccine carrier and an immunizing effective amount. Contains prolactin. Normally, the vaccine may be administered according to the manufacturer's instructions. Other adjuvants may be administered with the vaccine or with prolactin. Typically, prolactin enhances the protection conferred by animal or human vaccines that is considered "weak" (ie, provides reduced protection in level, degree, and / or duration). Will be used for. Examples of such vaccines are, for example, bacterins such as Pseudomonas, Staphylococcal Enterotoxin, Streptococci, cytomegalovirus, HIV, bacterins of Bordetella, Escherichia coli. ) Bacterins, Haemophilus bacterins, leptospirosis vaccines, Moraxella bovis bacterins, Pasteurel la bacterins and Vibrio fetus bacterins, and bovine respiratory diseases (). Infectious bovine rhinotracheitis, parainfluenza-3, respiratory syncytial virus) vaccine, bovine viral diarrhea vaccine, equine influenza vaccine, feline leukemia vaccine, feline respiratory disease Rhinotracheitis - calicivirus pneumonia virus) vaccine, Canine Valvo virus vaccine, an attenuated or killed virus products such as transmissible gastroenteritis vaccine, pseudorabies vaccine, and rabies vaccine. Furthermore, we have shown in vitro and in vivo data that prolactin can enhance the immune response to the immunogen, thereby acting as an adjuvant, so that external administration of the prolactin gene can result in in vivo. It is believed that it causes prolactin expression and may function as an adjuvant to the immunogen, whether administered by conventional means or by gene inoculation. A "genetic adjuvant" could be made by inserting the prolactin cDNA into a DNA delivery vehicle (eg, plasmid vector, liposome, viral vector). Pellegrini I. et al., Molecular Endocrinilogy, Vol. 6, 1023 (1992), Maniatis T. et al., Molecular Cloning: A Laboratory.・ Molecular Cloning: A Laboratory Manual, 2nd Edition, Cold Spring Harbor Press (1989) and Felger P. et al., Proceedings of This could be done as described by the National Academy of Sciences (Proc.Natl.Acad.Sci.), Vol. 84, page 7413 (1991). The "genetic adjuvant" is then administered with either the cDNA encoding the immunogen or the "naked" cDNA (ie, cDNA alone) in a suitable delivery vehicle. In addition, a "genetic adjuvant" could be administered with the immunogen itself. The injection sequence will be optimized depending on the immunogen. That is, the prolactin cDNA could be co-administered with the immunogen or immunogenic cDNA, prior to their administration, or subsequent to their administration. It is believed that the prolactin cDNA can be inserted into the same delivery vehicle. Various routes of administration could be used. Example 1 Simultaneous mitogenicity of recombinant human prolactin (r-hPRL) Peripheral blood lymphocytes (PBL) were subjected to density gradient centrifugation on Ficoll Paque (Pharmacia) to normal human volunteers. Isolated from. The heparinized blood was diluted 3-fold with phosphate buffered saline (PBS) and centrifuged at 2000 rpm for 20 minutes. The soft layer of white blood cells, present on the surface of the red blood cell pellet, was collected and diluted with an equal volume of PBS. The diluted soft layer was overlaid on Ficoll Park (6 ml soft layer on 4 ml Ficoll Park in 15 ml tube) and centrifuged at 1400 rpm for 30 minutes. The PBL layer found at the Ficoll-plasma interface was collected and the cells were washed 3 times with PBS. The PBLs were then resuspended in serum-free AIM-V medium from Gibco at 2x10 6 cells / ml and placed in a well of a round bottom 96-well microtiter plate in a volume of 100 μl (2x10 5 cells). PBL / well). Suboptimal doses of T cell mitogen Concanavalin A (ConA; 0.2 μg / ml) 50 μl were added together with various concentrations of r-hPRL (final concentration 0-100 ng / ml) 50 μl did. The culture was performed in 3 lines. The cells were incubated for 72 hours at 37 ° C. and 5% CO 2 , and the amount of proliferation was measured by the incorporation of tritiated thymidine. Tritiated thymidine (0.5 μCi / well) was added during the last 18 hours of incubation and cells were collected on glass fiber filters using a Skatron 96-well harvester before scintillation of radioactivity associated with the cells. It was measured by counting. The results obtained with cells from different individuals are shown in Table 1 below, showing that r-h PRL was able to enhance the proliferative response of T lymphocytes to suboptimal concentrations of ConA. . This co-mitogenic activity was best observed at r-hPRL concentrations of 1-10 ng / ml. Example 2 Enhancement of Antigen-Specific Proliferation by r-hPRL To test the ability of r-hPRL to enhance the proliferative response of human T cells to specific antigens, various concentrations of r-hPRL and most individuals were tested. PBLs were incubated with streptokinase, which is a common antigen to which is exposed. Cultures were performed in triplicate in wells of a 96-well round bottom microtiter plate, the cultures were 100 μl PBL (2 × 10 5 cells / well), 50 μl streptokinase (final concentration 25 μg / ml) and various concentrations (0-1000 ng). / Ml) of r-hPRL. Proliferation was measured by tritiated thymidine incorporation after 6 days of culture at 37 ° C, 5% CO 2 . The results shown in Table 2 indicate that r-hPRL at a concentration of 1 ng / ml significantly promoted streptokinase-induced proliferation. Example 3 Effect of Prolactin in Enhancing the Immune Response to Immunogen Twenty-four male Sprague-Dawley rats weighing 150 grams were divided into 4 groups. The control group was injected ip with 10 μg BSA mixed with alum. The other three groups were injected ip with 10 μg BSA mixed with alum along with either 180 μg prolactin, 375 μg prolactin or 750 μg prolactin. Blood was collected from the tail vein weekly for 4 weeks and serum was evaluated for antibodies to BSA by radioimmunosorbent assay (RIA). After the fourth blood draw, the animals were boosted with 10 μg BSA mixed with alum. Blood was collected from the tail vein over a period of 7 weeks to obtain serum and the serum was evaluated by RIA for the production of antibodies to BSA. To measure the effect of r-hPRL on cell responses of the immunized rats immunized rat peripheral blood lymphocytes bovine serum albumin (BSA) specific proliferation BSA in BSA ± r-hPRL, Booster Blood was collected from individual animals sacrificed 101 days after immunization. To collect peripheral blood lymphocytes (PBL), blood samples were diluted 4-fold with phosphate buffered saline (PBS) and centrifuged at 2000 rpm for 20 minutes. Soft layers were collected and Tris-ammonium chloride lysis buffer was added, followed by incubation at 37 ° C for 10 minutes to remove contaminating red blood cells. PBLs were then washed twice with PBS, 100 μg / ml penicillin, 100 μg / ml streptomycin, 20 mM Hepes buffer, 2 mM L-glutamine, 5 × 10 −5 M 2-mercaptoethanol and 5% heat-inactivated fetal calf serum. The cells were resuspended in RPMI1640 medium supplemented with 5 × 10 6 cells / ml. PBLs were added to the wells of flat-bottom 96-well microtiter plates in a volume of 100 μl (5 × 10 5 cells / well) and 1000 μg / ml of BSA added in the presence of medium alone (background control) or in a volume of 100 μl. Culture was performed in the presence. The culture was performed in 3 lines. Proliferation was measured by the uptake of tritiated thymidine after culturing at 37 ° C., 5% CO 2 for 5 days. The results show that in relative terms, PBL of rats immunized with BSA + 180 μg thPRL showed higher levels of BSA-specific proliferation than PBL from rats immunized with antigen alone. This observation suggests that r-hPRL may act to increase the cellular component of the immune response to the immunizing antigen. The results are summarized in Table 3 below and shown in FIGS. 5 and 6.
